A large number of Fusarium species are associated with Fusarium head blight of wheat and other small-grain cereals as well as seedling blight and brown foot rot. Different Fusarium species tend to predominate under different environmental conditions and in different regions. In addition to causing disease, these fungi are of particular significance because they produce a number of mycotoxins including the trichothecenes and enniatins that contaminate infected grain. The nature and amount of the mycotoxins that accumulate will alter according to the species or even the particular isolates involved in the infection. It is highly desirable to be able to analyse such complex infections to determine which species and, preferably, which chemotypes are present, in order to understand the factors that affect the pathogenicity of each species and to evaluate the potential risk for contamination of grain with mycotoxins. This paper reports the development of molecular methods, based upon the polymerase chain reaction (PCR), for the detection of mycotoxigenic fungi. Several of the Fusarium species involved are closely related, making the development of specific assays problematic. We describe the development of primers specific to individual species and discuss how this work provides insight into fungal populations and relates to taxonomic studies. In some instances, it is desirable to detect the presence of potential mycotoxin producers rather than individual fungal species. Generic assays have been produced for several genes involved in trichothecene biosynthesis and for enniatin synthetase in order to permit the detection of species able to produce the associated mycotoxins. Additional work is under way to refine assays to enable detection related to the class of trichothecene and chemotype of isolate because of the potential risk posed to human and animal consumers by different trichothecenes.  相似文献

Fungi produce low molecular weight secondary metabolites such as antibiotics and mycotoxins. Antibiotics cure diseases whereas mycotoxins cause diseases in plants, animals and human beings. Species such as Aspergillus, Fusarium, Penicillium and Stachybotrys are known to produce mycotoxins that accumulate in processed foods and feeds, although the incidence of infection occurs before processing, during the active growth of the organism. Among the mycotoxins, aflatoxins produced by Aspergillus flavus and A. parasiticus have been extensively studied at the molecular level. A complex biosynthetic pathway involving sixteen steps is mediated by individual major genes. These fungi have eight linkage groups, but the aflatoxin/sterigmatocystin (AF/ST) metabolic pathway genes have been mapped to only three linkage groups; ten of them belong to linkage group VII, and one of each to linkage group II and VIII. These genes are involved in both the regulatory and biosynthetic pathways and are clustered on the respective chromosomes. Clustering of genes in fungi indicates an evolutionary trend among genes that orchestrate gene function. Being linked together they segregate as a unit, thereby conferring a selective advantage to the organism. The evolution of gene clusters takes place through vertical or horizontal gene transfer. In fungi, horizontal gene transfer is most effective. Functionally, the mechanism of evolution of mycotoxin gene clusters in fungi seems to be similar to the evolution of a super-gene. The possible implications of evolutionary parallelism of gene clusters and super-genes is briefly explored.  相似文献

This review describes the naturally occurring mechanisms in cereals that lead to a reduction of Fusarium trichothecene mycotoxin accumulation in grains. A reduction in mycotoxin contamination in grains could also limit fungal infection, as trichothecenes have been reported to act as virulence factors. The mechanisms explaining the low toxin accumulation trait, generally referred to as type V resistance to Fusarium, can be subdivided into two classes. Class 1 includes mechanisms by which the plants chemically transform the trichothecenes, leading to their degradation or detoxification. Among the detoxification strategies, glycosylation of trichothecenes is a natural process already reported in wheat. According to the structure and the toxicity of trichothecenes, two other detoxification processes, acetylation and de-epoxidation, can be expressed, at least in transgenic plants. Class 2 comprises mechanisms that lead to reduced mycotoxin accumulation by inhibition of their biosynthesis through the action of plant endogenous compounds. These include both grain constitutive compounds and compounds induced in response to pathogen infection. There are already many compounds with antioxidant properties, like phenolic compounds, peptides or carotenoids, and with prooxidant properties, like hydrogen peroxide or linoleic acid-derived hydroperoxides, that have been described as ‘modulators’ of mycotoxin biosynthesis. This review addresses for the first time different studies reporting specific in vitro effects of such compounds on the biosynthesis of Fusarium mycotoxins. A better understanding of the natural processes limiting accumulation of trichothecenes in the plant will open the way to the development of novel breeding varieties with reduced ‘mycotoxin risk’.  相似文献

Although Aspergillus species are not usually considered as serious plant pathogens, Aspergilli are frequently encountered in plant products. The most important consequence of their presence is mycotoxin contamination. The main mycotoxins produced by Aspergilli are the aflatoxins, ochratoxin A and patulin, which are produced by a variety of Aspergillus species in different plant commodities. Phylogenetic analysis of sequences of the ribosomal RNA gene cluster is useful for clarifying taxonomic relationships among toxigenic Aspergilli causing pre- and postharvest contamination of agricultural products. Molecular data has enabled us to clarify the taxonomy of black Aspergilli, A. flavus and its relatives, and sections Circumdati and Clavati, which include ochratoxin and patulin-producing species. Phylogenetically unrelated species were found to produce the same mycotoxins, indicating that mycotoxin-producing abilities of the isolates have been lost (or gained) several times during the evolution of the genus. The data also indicate that biosynthetic gene-based probes are necessary for molecular detection of these mycotoxin-producing organisms. The organisation of the biosynthetic genes of patulin and ochratoxins is unknown, although experiments are in progress in several laboratories to clarify the genetic background of biosynthesis of these mycotoxins. Identification of biosynthetic genes responsible for mycotoxin production is essential for clarifying the evolution of mycotoxin biosynthesis in Aspergilli, and to develop specific gene probes for the detection of mycotoxin-producing Aspergilli in agricultural products.  相似文献

Epidemiology of Fusarium Diseases and their Mycotoxins in Maize Ears   总被引:1,自引:0,他引:1  
Fusarium species cause two distinct diseases on ears of maize, Fusarium ear rot (or pink ear rot) and Gibberella ear rot (or red ear rot), both of which can result in mycotoxin contamination of maize grain. The primary causal agent for Fusarium ear rot is Fusarium verticillioides, but F. subglutinans and F. proliferatum are also important. Gibberella ear rot is caused primarily by F. graminearum, but F. culmorum can also be important, especially in Europe. Aspects of the epidemiology of both diseases have been studied for decades, but only recently have efforts been made to synthesize this information into comprehensive models of disease development. Much of the work on F. graminearum has focused on Fusarium head blight of small-grain crops, but some of the results obtained are also relevant to maize. The primary mycotoxins produced by these fungi, fumonisins and deoxynivalenol, have differing roles in the disease-cycle, and these roles are not completely understood, especially in the case of fumonisins. Progress is being made toward accurate models for risk assessment of both diseases, but key challenges remain in terms of integrating models of pre- and post-infection events, quantifying the roles of insects in these diseases, and characterizing interactions among competing fungi and the environment.  相似文献

Fusarium head blight (FHB), caused by fungi belonging to the Fusarium genus, is a widespread disease of wheat (Triticum aestivum) and other small-grain cereal crops. The main causal agent of FHB, Fusarium graminearum, produces mycotoxins mainly belonging to type B trichothecenes, such as deoxynivalenol (DON), that can negatively affect humans, animals and plants. DON detoxification, mainly through glucosylation into DON-3-O-glucose, has been correlated with resistance to FHB. A UDP-glucosyltransferase from the model cereal species Brachypodium distachyon has been shown to confer resistance both to initial infection and to spike colonization (type I and type II resistances, respectively). Here, the functional characterization of transgenic wheat lines expressing the Bradi5g03300 UGT gene are described. The results show that, following inoculation with the fungal pathogen, these lines exhibit a high level of type II resistance and a strong reduction of mycotoxin content. In contrast, type I resistance was only weakly observed, although previously seen in B. distachyon, suggesting the involvement of additional host-specific characteristics in type I resistance. This study contributes to the understanding of the functional relationship between DON glucosylation and FHB resistance in wheat.  相似文献

作用模式得到明确的具有除草活性的植物病原真菌毒素为数不多,主要是危害农作物的病原真菌产生的植物毒素,目前只有少数几个具有除草活性的杂草病原真菌植物毒素的作用模式得到鉴定。而近些年来,新获得的绝大多数具有除草活性的杂草病原真菌植物毒素的作用模式尚未得到鉴定。在作用模式已经明确的具有除草活性的植物病原真菌毒素中,除了极个别与有些化学合成除草剂共享相同作用模式,绝大多数具有与现有化学合成除草剂完全不同的作用模式和独特的分子靶标位点,很有希望开发成为新型除草剂。本文重点介绍了目前已经得到鉴定的具有除草活性的植物病原真菌毒素的作用模式。  相似文献

Fusarium head blight of small-grain cereals, ear rot of maize, seedling blight and foot rot of cereals are important diseases throughout the world. Fusarium graminearum, F. culmorum, F. poae, F. avenaceum and Microdochium nivale (formerly known as F. nivale) predominantly cause Fusarium diseases of small-grain cereals. Maize is predominantly attacked by F. graminearum, F. moniliforme, F. proliferatum and F. subglutinans. These species differ in their climatic distribution and in the optimum climatic conditions required for their persistence. This review deals with the influence of climate on the production and dispersal of inocula, growth, competition, mycotoxin production and pathogenicity. Most species produce inocula, grow best, and are most pathogenic to cereal heads at warm temperatures and under humid conditions. However, the optimal conditions for F. moniliforme and F. proliferatum maize ear rot tend to be hot and dry and M. nivale head blight, seedling blight and foot rot of small-grain cereals tend to occur under cooler conditions. Seedling blight and foot rot caused by other species are favoured by warm dry weather. Between them, these fungi produce four important classes of mycotoxins: trichothecenes, zearalenone, fumonisins and moniliformin. Conditions favourable for in vitro growth are also generally the most favourable for mycotoxin production on cereal grains. These fungi rarely exist in isolation, but occur as a complex with each other and with other Fusaria and other fungal genera. Climatic conditions will influence competition between, and the predominance of, different fungi within this complex.  相似文献

The distribution and co‐occurrence of four Fusarium species and their mycotoxins were investigated in maize samples from two susceptible cultivars collected at 14 localities in South Africa during 2008 and 2009. Real‐time PCR was used to quantify the respective Fusarium species in maize grain, and mycotoxins were quantified by multi‐toxin analysis using HPLC‐MS. In 2008, F. graminearum was the predominant species associated with maize ear rot in the eastern Free State, Mpumalanga and KwaZulu‐Natal provinces, while F. verticillioides was predominant in the Northwest, the western Free State and the Northern Cape provinces. In 2009, maize ear rot infection was higher and F. graminearum became the predominant species found in the Northwest province. Fusarium subglutinans was associated with maize ear rot in both years at most of the localities, while F. proliferatum was not detected from any of the localities. Type B trichothecenes, especially deoxynivalenol, and zearalenone were well correlated with the amount of F. graminearum, fumonisins with F. verticillioides, and moniliformin and beauvericin with F. subglutinans. This information is of great importance to aid understanding of the distribution and epidemiology of Fusarium species in South Africa, and for predicting mycotoxin contamination risks and implementing preventative disease management strategies.  相似文献

The spread of fungicide resistant and/or tolerant phytopathogenic fungi is an important factor affecting crop protection. However, the mechanisms of fungal response to fungicide application are not entirely characterised. In particular, the contribution of previously known resistance factors and the final influence of fungicide treatments on metabolism of surviving mycelia (e.g. mycotoxin increased release and biosynthesis potentially causing contamination of the crops) merit investigation, in order to improve future molecular diagnostics of fungicide resistant strains. The performed experiments have shown distinct expression changes for homologs of a known fungicide resistance factor Flr1 (yeast; DHA1 family of major facilitator superfamily transporters) after azole application in cultured fusaria. Two distantly related homologs of that gene were selected, based on the unsupervised clustering and phylogenetic analysis of transporter sequences. One of these (FGSG_02865), was found to occur across the Fusarium sambucinum complex (F. graminearum, F. culmorum, F. cerealis) and was upregulated starting 24 h after fungicide treatments. This delayed response may point to possible involvement of DHA1 antiporters in a generalised response to stress resulting from fungicide treatment. Additional expression profiling was conducted for the mycotoxin biosynthetic genes (trichothecene and zearalenone gene clusters) in strains of Fusarium sambucinum complex cereal pathogens. The expression changes, when subjected to treatment with the fungicides (flusilazole, carbendazim), show that even an effective treatment (in this study, the benzimidazole fungicide carbendazim) applied to the grown mycelium, can result in enhanced activation of mycotoxin biosynthetic genes in fungal cells which survive the treatment. Our results suggest that increased mycotoxin contamination can be strongly influenced not only by the amount or the type of antifungal compound, but also the timing of fungicide exposition (stage of infection).  相似文献

High year-to-year variability in the incidence of Fusarium spp. and mycotoxin contamination was observed in a two-year survey investigating the impact of maize ear rot in 84 field samples from Germany. Fusarium verticillioides, F. graminearum, and F. proliferatum were the predominant species infecting maize kernels in 2006, whereas in 2007 the most frequently isolated species were F. graminearum, F. cerealis and F. subglutinans. Fourteen Fusarium-related mycotoxins were detected as contaminants of maize kernels analyzed by a multi-mycotoxin determination method. In 2006, a growth season characterized by high temperature and low rainfall during anthesis and early grain filling, 75% of the maize samples were contaminated with deoxynivalenol, 34% with fumonisins and 27% with zearalenone. In 2007, characterized by moderate temperatures and frequent rainfall during the entire growth season, none of the 40 maize samples had quantifiable levels of fumonisins while deoxynivalenol and zearalenone were detected in 90% and 93% of the fields, respectively. In addition, 3-acetyldeoxynivalenol, 15-acetyldeoxnivalenol, moniliformin, beauvericin, nivalenol and enniatin B were detected as common contaminants produced in both growing seasons. The results demonstrate a significant mycotoxin contamination associated with maize ear rots in Germany and indicate, with regard to anticipated climate change, that fumonisins-producing species already present in German maize production may become more important.  相似文献

ABSTRACT Fusarium head blight (FHB), or scab, is a destructive disease of small grains caused by members of the Fusarium graminearum species complex, comprised of at least nine distinct, cryptic species. Members of this complex are known to produce mycotoxins including the trichothecenes deoxynivalenol (DON) along with its acetylated derivatives and nivalenol (NIV). In this study, 31 strains, belonging to eight species of this complex and originating from diverse hosts or substrates, were tested for differences in aggressiveness and mycotoxin production. Large variation among strains, both in terms of their aggressiveness and the ability to produce trichothecenes on a susceptible cultivar of wheat was found; variation appears to be a strain-specific rather than species-specific characteristic. While pathogenicity was not influenced by the type of mycotoxin produced, a significant correlation was observed between the amount of the dominant trichothecene (DON and its acetylated forms or NIV) produced by each strain and its level of aggressiveness on wheat. Some isolates also were tested for their ability to infect rice cv. M201, commonly grown in the United States. While tested strains were capable of infecting rice under greenhouse conditions and causing significant amount of disease, no trichothecenes could be detected from the infected rice florets.  相似文献

Fungal incidence and mycotoxin contamination of farm-stored maize were assessed and compared in grain samples from three villages each in two agroecological zones over time. Maize samples were collected at 2 and 4 months after stocking from 72 farmers’ stores in 1996 and 1997 in the Humid Forest (HF) and Western Highlands (WHL) of Cameroon. Mycological assays of these samples revealed several fungal species.Nigrospora spp. were the most prevalent fungi in HF (32%) and WHL (30%) in 1996,Fusarium verticillioides (22%) andF. graminearum (27%) were also isolated from these samples. In the WHL in 1996, no significant difference in fungal incidence was found among villages for samples collected 2 months after harvest, but at 4 months incidence was significantly higherP<0.05). In 1997 the levels of fungal contamination were lower than in 1996. The incidence ofAspergillus spp. was low in general, ranging from 0.0 to 5.9% infected kernels. Analysis with thin layer chromatography detected low levels of aflatoxins in a few samples.F. verticillioides mycotoxin fumonisin Bi (300-26,000 ng/g) andF. graminearum metabolites deoxynivalenol (<100–l,300 ng/g) and zearalenone (<50–110 ng/g) were determined by means of polyclonal antibody competitive direct enzyme-linked immunosorbent assay. A significant correlation (r=0.72; P=0.0001) was found between the incidence ofF. graminearum and the contamination with deoxynivalenol. Storage time (2vs 4 months after stocking) had a significant positive effect (r=0.39; P=0.013) on the level of fumonisin B1. This is the first report of the natural occurrence of these mycotoxins in maize in Cameroon.  相似文献

The development of new maize hybrids with resistance to Fusarium infection is an effective means of minimizing the risk of mycotoxin contamination. Several maize hybrids have been investigated for Fusarium ear rot and accumulation of fumonisin B1 (FB1), fumonisin B2 (FB2), beauvericin (BEA) and fusaproliferin (FP) after artificial inoculation in the field with toxigenic strains of Fusarium verticillioides and Fusarium proliferatum. The year of inoculation had a significant influence on the disease severity and mycotoxin accumulation in maize kernels. Of all the hybrids tested, only Mona exhibited resistance to ear rot caused by F. verticillioides and produced low levels of fumonisins during three years of experiments. In Fusarium-damaged kernels (FDK), fumonisin B1, fumonisin B2, beauvericin and fusaproliferin were detected at concentrations much higher (up to 10–20 times) than in healthy-looking kernels (HLK). Animal and human exposure to these mycotoxins can be drastically reduced by removing mouldy and visibly damaged kernels from the commodity.  相似文献

Thirty-two fungal species belonging to 17 genera were recorded on 45 seed samples of lentil (Lens esculenta) collected from different governorates in Egypt. The prevalent genera wereAlternaria, Aspergillus, Cladosporium, Fusarium andPenicillium. Chitosan enhanced suppression of seedborne mycoflora, as estimated using either agar plates or a blotter test of lentil seed samples. These results suggested an alternative non-toxic means for controlling seedborne fungi. Furthermore, pretreatment of lentil seeds with chitosan significantly reduced the natural contamination with mycotoxins — aflatoxins, ochratoxin A, citrinin and zearalenone — under storage conditions for 6 months.  相似文献

Fusarium head blight (FHB) is a devastating disease of important cereal crops resulting in significant yield loss and mycotoxin contamination. Persistent outbreaks of FHB in Europe and North America have led to various efforts to understand the mechanisms of resistance to this disease and mycotoxin biosynthesis. Recent data on the epidemiology of the common mycotoxigenic species of Fusarium, Alternaria, Aspergillus and Penicillium in infected or colonized plants, and in stored or processed plant products from the Mediterranean area are reviewed. Emphasis is placed on the toxigenicity of the causal fungal species and the natural occurrence of well known mycotoxins (aflatoxins, ochratoxins, fumonisins, trichothecenes, zearalenone, patulin, Alternaria-toxins and moniliformin), as well as some more recently described compounds (fusaproliferin, beauvericin) whose toxigenic potential is not yet well understood. Several Fusarium species reported from throughout the Mediterranean area are responsible of the formation of mycotoxins in infected plants and in plant products, including: Fusarium graminearum, F. culmorum, F. cerealis, F. avenaceum, F. sporotrichioides and F. poae, which produce deoxynivalenol, nivalenol, fusarenone, zearalenone, moniliformin, and T-2 toxin derivatives in wheat and other small grains affected by head blight or scab, and in maize affected by red ear rot. Moreover, strains of F. verticillioides, F. proliferatum, and F. subglutinans, that form fumonisins, beauvericin, fusaproliferin, and moniliformin, are commonly associated with maize affected by ear rot. Fumonisins, were also associated with Fusarium crown and root rot of asparagus and Fusarium endosepsis of figs, caused primarily by F. proliferatum. Toxigenic A. alternata strains and associated tenuazonic acid and alternariols were commonly found in black mould of tomato, black rot of olive and citrus, black point of small cereals, and black mould of several vegetables. Toxigenic strains of A. carbonarius and ochratoxin A were often found associated with black rot of grapes, whereas toxigenic strains of A. flavus and/or P. verrucosum, forming aflatoxins and ochratoxin A, respectively, were found in moulded plant products from small cereals, peanuts, figs, pea, oilseed rape, sunflower seeds, sesame seeds, pistachios, and almonds. Finally, toxigenic strains of P. expansum and patulin were frequently found in apple, pear and other fresh fruits affected by blue mould rot, as well as in derived juices and jams.  相似文献
