Samples of blood and milk were taken from clinically ketotic cows at 12-hour intervals before and after treatment The concentration of beta-hydroxybutyrate was determined in plasma and milk, and the concentration of acetone was determined in milk. Measurement of milk acetone had the optimal combination of sensitivity, specificity and positive predictive value to identify ketotic cows and was subsequently used in a comparison of the fertility of ketotic and normal cows. Two samples of milk were taken approximately two weeks apart, between 12 and 60 days after calving, from 410 cows on three farms and the concentration of acetone in the milk was measured. Thirty cows with milk acetone concentrations of at least 0.4 mmol/litre were compared with paired control cows with milk acetone concentrations less than 0.3 mmol/litre. The ketotic cows had a significantly longer calving-to-conception interval and a significantly higher culling rate due to a failure to conceive (P<0.05).  相似文献

The mean urea nitrogen concentration in vitreous humor (VUN) in 97 healthy steers after death was 15.7 mg/dl. The mean serum urea nitrogen concentration in the same cattle was 20.0 mg/dl. The mean vitreous creatinine (VC) concentration was 0.7 mg/dl, and the serum creatinine value was 1.5 mg/dl. The VUN and VC were both significantly lower than serum urea nitrogen and serum creatinine, respectively, but varied in a consistent, predictable manner. In tests with 10 cows, postmortem intervals as long as 36 hours and ambient temperatures up to 30 C had no significant effects on VUN and VC. Evaluation of 8 animals with uremia confirmed that the vitreous humor is a stable fluid compartment after death, usable as an indicator of serum urea nitrogen and creatinine.  相似文献

Four groups of four healthy mid-lactation Friesian cows were fed a compound feeding stuff containing either 2, 10 or 250 mg sulphamethazine/kg, corresponding to 0, 2, 10 and 250 per cent of the therapeutic inclusion rate in rations for pigs, at a flat rate of 3 kg twice daily for 21 days, followed by a seven-day withdrawal period. The cows were machine-milked twice daily and pooled milk samples from each cow were analysed by a commercially available microbiological assay with a sensitivity of 100 micrograms/litre and by a high performance liquid chromatography (HPLC) procedure with a limit of detection of 10 micrograms/litre. No sulphamethazine was detected by HPLC in the milk samples taken from any of the cows fed the concentrate containing 2 or 10 mg/kg. The milk samples from all four cows fed the highest concentration of sulphamethazine contained from 21 to 120 micrograms/litre while they were being fed the contaminated concentrate. The cow with the highest concentrations of sulphamethazine was the only one which repeatedly tested positive by the microbiological assay. The concentration of sulphamethazine declined rapidly during the withdrawal period and the drug was not detectable by either method in samples taken from two days after the contaminated feed was withdrawn.  相似文献

2 experimental cows received isobutylidenedi urea added to a natural diet in amounts of 175 g (I) and 730 g (II) per day for a period of several weeks before the trial was started. On the 1st day of experiment the morning dose was labelled with 5.05 g of excess 15N. 8 hrs after the beginning of the trial of 15N level in the TCE soluble portion of blood plasma (TCE=trichloroacetic acid) increased and remained at an elevated level until the 36th hour of experiment. Similarly, the values for maximum urinary 15N concentrations were maintained for a prolonged period of time. Isobutylidenedi urea was excreted with the urine in rates related to its solubility. Only small percentages of the 15N intake were excreted in the TCE soluble portion of the milk (cow I: 0.03%; cow II: 0.05%). The 15N-labelling of milk protein provides evidence for the fact that nitrogen from IBDU is utilized for the synthesis of milk in the cows. The amount of urea in milk averaged 400 mg per litre. None of the milk samples tested contained IBDU.  相似文献

Three groups of four lactating cows received a subcutaneous injection of 0 . 05, 0 . 10 and 0 . 15 mg Se/kg body weighty respectively administered as sodium selenate. A fourth group was injected with saline. In all the cows injected with sodium selenate, the concentration of Se in blood increased rapidly and was significantly higher than in control cows for two days in the group receiving the lowest dose and for 182 days (the duration of the experiment) in the two other groups. The activity of glutathione peroxidase in blood increased slowly in all cows injected with sodium selenate and was significantly greater than in control cows after 15, 22 and 29 days respectively, and remained significantly greater for 63, 91 and 182 days respectively. In a second experiment a single subcutaneous injection of 0 . 15 mg Se/kg body weight had no effect on the mean milk yield of 37 animals (19 . 1 kg/day) compared with the milk yield of a similar group of control animals (19 . 1 kg/day) during 70 days. The concentration of Se in milk was significantly higher on the first (168 microgram/litre) and second (69 microgram/litre) day after injection than in control animals (mean 26 microgram/litre).  相似文献

Subclinical ketosis of dry cows was studied as to its effect on the composition of colostrum and on the health condition and selected clinico-biochemical parameters in the new-born calves of these cows. The experimental group consisted of eight cows with ketonuria and their calves. The control group consisted of six cows without ketonuria and their new-born calves. Calves of both groups were reared in similar conditions. Colostrum samples were taken from the first milking and then from the milking 24 hours after parturition. The blood of the calves was sampled before the intake of colostrum and 24 hours after birth. The concentration of ketone bodies, particularly beta-hydroxybutyrate, was much higher in the colostrum of both groups of cows than in the blood (62.6 mg total ketone bodies per 1 litre of the first colostrum of the cows of the experimental group). The concentration of ketone bodies slightly increased after parturition. The cows with ketonuria secreted into colostrum a larger amount of oxidated ketone bodies. Colostrum quality was good in both groups. Decreased viability was not recorded during the clinical examination of the calves after birth. The level of total ketone bodies in the blood of pre-colostral calves in the experimental group was 10.4 mg per litre (a much lower concentration than in blood of their dams), and exhibited no appreciable change after taking in the colostrum. The blood of calves born to cows with ketonuria contained significantly higher amounts of oxidated ketone bodies. No correlation was demonstrated between the contents of ketone bodies in the blood of cows, in the blood of calves, and in the colostrum. The calves of the experimental group were found to have a significantly higher AST activity in plasma, as compared with the control group. Twenty-four hours after birth, the quantity of immunoglobulins in the blood serum was significantly lower in the calves of the experimental group. The effect of serum glucose, ASAT and urea on reproductive performance was studied in 45 Finnish dairy cows from 8 different herds. Blood samples were taken a month before calving as well as 2 weeks and 2 months after calving. Serum urea, ASAT and glucose were used as indicators of metabolic and energy balance. Glucose and ASAT concentrations were significantly correlated with fertility 2 weeks after calving, but not before calving or 2 months after calving. Antepartum, 60% of the cows had a low urea level; at 2 weeks and at 2 months postpartum 89% and 65%, respectively had low levels of urea. The cows were divided into groups as follows: low or high (greater than or equal to 2.9 mmol/l) glucose level and low or high (greater than or equal to 2.5 mmol/l) urea level. Low levels of glucose and/or urea in puerperium were associated with poor fertility. The intervals from calving to first insemination and conception were shorter in the group with high glucose and high urea than in the group with low values. The cows with the shortest intervals from calving to first insemination and conception showed adequate metabolic balance (high levels of urea and glucose). Clinical ketosis had an adverse affect on intervals from calving to first insemination and on conception. Intervals from calving to first insemination and to conception were significantly (p less than 0.01) longer in ketotic cows than in non-ketotic ones. Liver damage, reflected as elevated activity of ASAT, was associated with poor fertility. If shorter calving intervals are desired, adequate energy balance in puerperium is essential.  相似文献

On a farm where the ration of cows contained 88.0 g of butyric acid, an experimental group of cows (n = 8), producing milk containing 7.9 mg or more acetone per litre, was studied for 14 days for the effect of a 250 g supplement of sodium acetate to the ration (combined with single i.m. administration of vitamins A, D2 and E) on selected metabolism parameters and on milk production. As distinct from the control group of cows (n = 8) from the same farm which produced milk containing 3.9 mg or less acetone per litre and which were fed without sodium acetone supplements, a tendency of increased alkaemia of the organism was suggested in the experimental cows. This tendency manifested itself during the trial in increased pH values, increased base excess (BE) and standard bicarbonate (SB) in the blood, and in an increase in the pH value and net acido basic secretion in urine. A decrease was recorded in the concentration of the acetone + acetacetic acid sum, the same as beta-hydroxybutyric acid in blood and the sum of acetone and acetacetic acid in milk (P less than 0.01). An insignificant increase of the activity of gammaglutamyl transpeptidase (GMT) was recorded in the blood serum of the experimental cows and a significant increase occurred in the content of potassium (up to P less than 0.01) and urea (up to P less than 0.01) in urine. Starea, an intimate mixture of gelatinized wheat starch and urea, was tested again urea with regard to the ammonia release in the rumen, the amount and composition of the nitrogen fraction reaching the intestine, the nitrogen and energy balances, and the performance of lactating cows. Maize silage was the basal feed in all experiments. The ammonia concentration in the rumen liquor was measured in two cows fitted with a permanent rumen cannula. The curve of the ammonia concentration was about the same with both nitrogen supplements, and peaked at about 250 mg/l (14.7 mmol/l) two hours postfeeding. Two dry cows fitted with a re-entrant cannula at 10 cm from the pylorus were fed with maize silage supplemented with increasing amounts of either urea or starea. The duodenal nitrogen flow rapidly levelled off with both supplements and no significant difference between them could be detected. The amount of ammonia in the duodenal digesta remained very low. Nitrogen digestibility measured in lactating cows was not noticeably affected when starea instead of urea was added to maize silage. However, nitrogen excretion in the urine tended to decrease, while nitrogen secretion in milk was augmented with starea. The digestibility of the rations and their metabolizable energy supply were not significantly affected, and milk production was quite the same with both supplements, apart from the higher amount of milk protein obtained with starea compared with urea. As a general conclusion from the whole work conducted on the nutritive value of urea for lactating cows, it can be stated that the decline in performance of the animals after substitution of urea for conventional protein is mainly due to a decrease of the metabolizable energy concentration in the diets. None of the attempts tested in an effort to improve the nutritive value of urea was able to overcome this difficulty.  相似文献

The concentration of urea in the milk of 510 dairy cows in 10 herds was determined at regular intervals for a year. The herds contained approximately equal numbers of Swedish Red and White, and Swedish Holstein cows. The mean ± sd concentration in the samples from individual cows was 5.32 ± 1.13 mmol/1, and the mean concentration in bulk milk was 5.39 ± 0.96 mmol/1. These values indicated that on average the herds were fed too much protein relative to their intake of energy throughout the year.Herd factors had a strong influence on the milk urea concentration. The concentration was lower during the first month of lactation than later in the lactation, and lower when the cows were housed during the winter than when they were grazing. There was a weak positive relationship between the daily milk yield and urea concentration, particularly during late lactation, but there was no relationship with either breed or age. Bulk milk urea was a reliable guide to the average urea concentration of a herd.  相似文献

The stimuli that elicit thirst were studied in four ponies. Nineteen hours of water deprivation produced an increase in plasma protein from 67 +/- 0.1 g/litre to 72 +/- 2 g/litre, a mean (+/- se) increase in plasma sodium from 139 +/- 3 to 145 +/- 2 mmol/litre and an increase in plasma osmolality from 297 +/- 1 to 306 +/- 2 mosmol/litre. Undeprived ponies drank 1.5 +/- 0.9 kg/30 mins; 19 h deprived ponies drank 10.2 +/- 2.5 kg/30 mins and corrected the deficits in plasma protein, plasma sodium and plasma osmolality as well as compensating for the water they would have drunk during the deprivation period. In order to determine if an increase in plasma osmolality would stimulate thirst, 250 ml of 15 per cent sodium chloride was infused intravenously. The ponies drank when osmolality increased 3 per cent and when plasma sodium rose from 136 +/- 3 mmol/litre to 143 +/- 3 mmol/litre. Ponies infused with 15 per cent sodium chloride drank 2.9 +/- 0.7 kg; those infused with 0.9 per cent sodium chloride drank 0.7 +/- 0.5 kg. In order to determine if a decrease in plasma volume would stimulate thirst, ponies were injected with 1 or 2 mg/kg bodyweight (bwt) frusemide. The study was performed in eight high-pregnant cows in which medium to high ketonuria was repeatedly detected. The control group consisted of six cows free from ketonuria before parturition. Blood and urine were sampled from the cows two to eight weeks before parturition, on the day of parturition, 24 hours, three to five days and two to six weeks after parturition. The concentrations of oxidated ketone bodies and beta-hydroxybutyrate were determined by the distilling method. Further, the values of clinico-biochemical parameters in the urine and blood were determined within the range of the preventive diagnosis system. No marked changes in the health condition were found by the current clinical examination. The average concentration of total ketone bodies in the blood of the cows two to eight weeks before parturition was 29.2 mg per litre; the respective values recorded in the control cows were lower by about a half. The concentration of ketone bodies in the blood of the cows in the experimental group decreased on the day of parturition. The decrease was followed by a gradual rise of the total ketone bodies content in the blood up to 50.6 mg per litre two to six weeks after parturition. Alterations testifying to a disorder in the function of liver were observed in the cows of both groups throughout the period under study. The disorder was due to excessive administration of crude protein to the high-pregnant cows combined with the use of low-quality silage. After parturition the feed ration was not differentiated according to performance and was deficient in the basic nutrients and vitamins. Ketonuria was obviously due to the simultaneous intensification of hepatal and alimentary ketogenesis, caused by hepatopathy in combination with the intake of large amounts of poor-quality silage containing butyric acid.  相似文献

Hypomagnesaemic tetany was induced in non-lactating and lactating ewes by feeding them semi-synthetic low magnesium diets containing additional potassium chloride and citric acid. Aqueous and vitreous humour were sampled from one eye at the time of death (fresh) and from the second eye after the head had been stored at ambient temperature for 24 hours (24-hour). There were significant relationships between the concentrations of magnesium in cerebrospinal fluid and plasma and its concentrations in fresh aqueous humour and fresh vitreous humour. Magnesium concentrations of < 0.33 mmol/litre in fresh aqueous humour and < 0.50 mmol/litre in 24-hour aqueous humour were associated with severe hypomagnesaemia and tetany. However, the concentration of magnesium in aqueous humour is relatively unstable and, unless the time of death was known accurately, its interpretation would be difficult. Magnesium concentrations of < 0.60 mmol/litre in fresh vitreous humour and < 0.65 mmol/litre in 24-hour vitreous humour were associated with severe hypomagnesaemia and tetany in adult sheep. The concentration of magnesium in vitreous humour was relatively stable for up to 48 hours postmortem.  相似文献

Over a period of seven years, 262 recumbent dairy cows were investigated, and serum samples were analysed for creatine phosphokinase (CPK), lactate dehydrogenase (LDH) and aspartate aminotransferase (AST) activities. The results from cows for which all three measurements were available, were analysed statistically by the non-parametric receiver operating characteristic (ROC) method, to evaluate the value of the serum enzyme activities for predicting a failure to recover. The sensitivity and specificity of the activity levels were calculated over all cut-off points, and ROC curves were created by plotting sensitivity as a function of 1--specificity at each cut-off point for samples grouped by the interval after the cows became recumbent. The predictive values of the tests were compared by calculating the areas under the curves, and the positive and negative predictive values of the tests were calculated and plotted against the prevalence of a failure to recover. The results from each of the enzymes were significantly better than from a random test with no discriminatory ability. The optimal cut-off points maximising the sensitivity and specificity of the tests were 2330, 2225 and 171 U/litre for CPK, LDH and AST, respectively. The predictive value of AST was significantly better than that of CPK or LDH, and measures made on the second and third day of recumbency were significantly better with optimal cut-off points of 128 and 189 U/litre, respectively.  相似文献

Severe hypomagnesaemia and tetany were induced in 10 lactating cows by feeding them semi-synthetic low magnesium diets and the animals were used to study the stability of postmortem markers of hypomagnesaemic tetany. There were significant relationships between the concentrations of magnesium in either cerebrospinal fluid (CSF) or plasma and either aqueous or vitreous humour. The onset of hypomagnesamic tetany was also associated with low magnesium concentrations in plasma, CSF and aqueous and vitreous humour. Magnesium concentrations less than 0.25 mmol/litre in fresh aqueous humour may be indicative of severe hypomagnesaemia and possible tetany in lactating cows, but the concentration of magnesium in aqueous humour was unstable postmortem. The concentration of magnesium in vitreous humour was relatively stable and a concentration of less than 0.55 mmol/litre could be used as a diagnostic marker of tetany in cattle for up to at least 48 hours postmortem, at ambient temperatures typical of Northern Ireland.  相似文献

Electrolytes, metabolites, cortisol and reproductive hormones were measured in maternal plasma taken at least twice daily from three cases of bovine hydrops before, during and after parturition induced by dexamethasone or prostaglandin. Caesarean operations were required for two of the cases. Maternal plasma electrolytes remained within the normal range, but average potassium and creatinine concentrations were higher (9.2 and 0.68 mmol/litre, respectively) than normal (4.7 and 0.42 mmol/litre) in samples of amniotic fluid obtained at calving. Sodium (100 mmol/litre) and chloride (67 mmol/litre) in allantoic fluid were also higher than normal (53 and 20 mmol/litre, respectively). Conversely, creatinine concentrations were lower than normal in allantoic fluid (2.2 vs 13.8 mmol/litre). Oestradiol concentrations were lower than normal in maternal plasma (ranges: less than 20 to 140 pg/ml vs 30 to 440 pg/ml); maximum prostaglandin F metabolite (PGFM) concentrations were slightly elevated (ranges 1.1 to 2.0 ng/ml vs 0.4 to 0.9 ng/ml). Progesterone and cortisol concentrations remained within the normal range; the latter hormone increased markedly in parallel with raised PGFM concentrations. In two cases, the concentrations of reproductive hormones tended to be lower in the amniotic fluid than in the allantoic fluid. For example, progesterone concentrations were 42.8 and 14.9 ng/ml in the amniotic fluids vs 64.2 and 29.8 ng/ml in the allantoic fluids of the two cows; PGFM concentrations were 27.7 and 4.3 ng/ml vs 34.6 and 5.0 ng/ml, and oestradiol concentrations were 1.5 and 3.5 ng/ml vs 1.1 and 6.4 ng/ml in the two fluids, respectively.  相似文献

Two comparative trials were performed, each with 16 cows which in the period of 2-6 weeks after parturition had 7.9 mg and more acetone in 1 litre of milk. The cows, crossbreds of the Czech Red-Pied cattle with the Holstein cattle, were divided into control and test groups, eight in each using the system of pairs. The cows of test groups were given for a fortnight feed rations containing a meal of milk thistle (Silybum marianum, L., Gaert.) seeds, at a rate of 0.3 kg per head/day with the contents of 2.34% silybin and silydianin (substances of the so called silymarin complex of the flavonolignane group). In comparison with the control cows, in the blood and milk of the former ones a decrease was demonstrated in the sum of acetone + acetoacetic acid (up to P less than 0.01) and beta-hydroxybutyric acid in the blood (up to P less than 0.05). The ketonuria degree dropped remarkably. Although there were not observed any differences in the parameters of acid-base metabolism in the blood (pH, PCO2, BE, SB, BB), the pH values and net acid-base output in urine were higher in these cows. Milk production in the cows of control groups was decreasing during the trial (up to P 0.01), but in the test cows it was higher by 7.7% (trial 1) and by 3.4% (trial 2), in comparison with the milk yield at the beginning of the trials. Differences in metabolism parameters and milk production in favour of the cows which were given milk thistle in their feed rations were observed even in a fortnight after the diet stopped to contain this ingredient.  相似文献

Transfer of chlortetracycline from contaminated feedingstuff to cows' milk   总被引:2,自引:0,他引:2  
Three groups of four Friesian cows in mid-lactation were fed a compound feedingstuff contaminated with 2, 10 or 300 mg chlortetracycline/kg for 21 days, and were then fed an uncontaminated diet for seven days. A fourth group of four cows was fed an uncontaminated diet throughout the study. Daily pooled milk samples from each cow were analysed by high performance liquid chromatography (HPLC) with a detection limit of 50 microg chlortetracycline/litre. Chlortetracycline was detected in only two milk samples taken from one of the animals fed feed containing 300 mg 300 mg chlortetracycline/kg, and both contained less than the maximum residue limit (MRL) specified by the European Union (100 microg/litre). All the milk samples were also analysed by the Delvotest SP microbiological assay, which has a detection limit of 300 microg chlortetracycline/litre. During the treatment period, this method gave four presumptive false-positive results, because they were not confirmed by HPLC. Selected daily pooled samples from each treatment group were also analysed by the semi-quantitative Charm II radioreceptor assay with a detection limit of 10 microg chlortetracycline/litre. Immunoreactive chlortetracycline was detected only in the animals fed feed containing 300 mg chlortetracycline/kg and several of the results exceeded the EU MRL during the treatment period. No significant treatment effects on animal performance were observed. However, there was a trend towards a higher milk fat concentration (P<0.09) and a lower milk protein concentration (P<0.07) with increasing concentration of chlortetracycline in the diet.  相似文献

The relationship between milk urea concentration and the fertility of dairy cows was examined in two studies. The first examined the relationship between bulk milk urea concentration and overall herd fertility, using data collected from 250 herds in the UK. There was no relationship either between bulk milk urea concentration and fertility, or between changes in bulk milk urea concentration and fertility. The second study compared the relationship between the milk urea concentration five days after service, and the fertility of individual cows on 11 UK dairy farms. There was no significant difference between the milk urea concentration of the cows that became pregnant and those that did not.  相似文献

Milk urea determination is being used as a broad indicator of protein/energy imbalance in dairy herds. The main purpose of this study was to compare blood and bulk milk urea values in grazing herds, to evaluate their seasonal variation under South Chilean conditions, and to examine their potential relationships with herd fertility. The association between herd blood urea concentration (mean of seven lactating cows) and bulk milk urea concentration (tank containing milk from the previous 24 h) was determined in 21 diary herds. Reference values, seasonal and herd variance, and the frequency of herds with values outside a range of 2.5 to 7.3 mmol/l were determined in bulk milk samples obtained monthly for a period of one year from 82 suppliers at two creameries located in southern Chile. Finally, bulk milk urea was measured every two weeks in samples from 24 herds, and the first service conception rate (FSCR) from 2153 dairy cows was determined. Mean bulk urea concentration was highly correlated with mean herd blood urea concentration (r = 0.95; p < 0.01). Mean urea concentration in the bulk milk samples obtained during one year from 82 herds was 4.9 +/- 1.2 mmol/l, with a range of 1.5 to 11.6 mmol/l. The highest values were found during spring and the lowest values during the summer. There was a high seasonal variation (CV = 13-47%) and between-herd variation (CV = 20-31%). Out of a total of 984 samples, 5.4% had urea values > 7.3 mmol/l and 3.8% had values < 2.5 mmol/l. Of the 82 herds, 27% had values outside the reference interval (2.5-7.3 mmol/l) on two or more occasions. FSCR was lower in herds when the bulk milk urea was > 7.3 mmol/l (50.7%) than in cows, where the urea concentration was < 5.0 mmol/l (73.8%) at the time of insemination. The study concluded that bulk milk urea concentrations provided information similar to herd blood urea concentrations in local grazing dairy herds. There was a high frequency of herds with abnormal values, with large variations between herds and between seasons. Increased milk urea concentrations during spring were associated with lower conception rates.  相似文献
