 研究表明,旁路氧化作用能够降低辣椒炭疽菌对嘧菌酯的敏感性。离体培养条件下,旁路氧化在辣椒炭疽菌野生菌株的孢子萌发中提供大于95倍生理功能的补偿作用;在菌丝生长中提供20~35倍的补偿作用。但在接种前用嘧菌酯处理辣椒,旁路氧化作用并不明显,嘧菌酯对辣椒炭疽病有良好的保护、治疗、铲除和抗产孢作用。用嘧菌酯处理过的病斑产生的分生孢子萌发活力和致病力下降。炭疽菌离体下的菌丝耗氧率测定表明,10μg/mL嘧菌酯处理1 h对菌丝呼吸耗氧有良好的抑制作用,其抑制效果随药剂浓度提高而略有增加。随处理时间延长,药剂对菌丝耗氧的抑制作用下降,但仍然抑制菌丝生长。研究证明:延长处理时间,呼吸作用对药剂的敏感性下降的机理不是因为旁路氧化途径增强,也不是因为基质中药剂效力的下降,而是存在其它机制。  相似文献

 Inhibitors of respiration with high affinity to the Qo site of cytochrome b constitute a major class of modern agricultural fungicides. Many fungal organisms, including plant pathogens, can circumvent this inhibition site by expressing alternative respiration, a pathway dependent on alternative oxidase and other unidentified gene products. The restriction enzyme-mediated insertion (REMI) technique was employed in this study to disrupt genes involved in the expression of fully functional alternative respiration of Magnaporthe grisea. In one of the REMI mutants obtained, the rescue response mediated by alternative respiration was completely abolished. In two other mutants, the response was diminished but not entirely silenced. For all three mutants, phenotype changes were not explained by an altered structure of the alternative oxidase gene AOXMg or by a decreased level of gene expression in response to the Qo inhibitor azoxystrobin. The gene potentially affected in one of the REMI mutants was a homolog of the ABC1 family of genes encoding chaperone-like proteins with roles in the optimal assembly of mitochondrial membrane complexes. Field isolates of Mycosphaerella fijiensis, causing black Sigatoka of banana, were characterised for their sensitivity to different inhibitors of the cytochrome bc1 enzyme complex (Qo respiration inhibitors, strobilurin fungicides), using physiological, biochemical and molecular genetic methods. Strobilurin‐resistant isolates exhibited very high resistance factors both in mycelial growth inhibition and NADH consumption assays. Cross‐resistance was observed among all Qo inhibitors, including trifloxystrobin, azoxystrobin, famoxadone, strobilurin B and myxothiazol. However, the Qi and the cytochrome aa3 inhibitors, antimycin A and potassium cyanide, respectively, were not cross‐resistant to Qo inhibitors. In sensitive but not in resistant isolates, mixtures of Qo inhibitors and SHAM, an inhibitor of the alternative oxidase (AOX), were more active than the components alone, indicating that the alternative pathway is essential in metabolism, but not causal for resistance. In the cell‐free NADH‐consumption assay, the Qo inhibitors affected the sensitive but not the resistant isolates, suggesting that AOX was not active in sub‐mitochondrial particles. In whole cells, however, the AOX has a basic expression level and is probably not inducible by trifloxystrobin. Sequencing of the cytochrome b gene of sensitive and resistant M fijiensis isolates revealed a difference in the nucleotide sequence leading to a single amino acid change from glycine to alanine at position 143 in the resistant isolate. This change is known to occur also in the naturally tolerant basidiomycete Mycena galopoda. It is suggested that the field isolates of M fijiensis can acquire resistance to Qo inhibitors due to a target site alteration with a single base pair change. Resistant isolates do not seem to contain a mixture of mutated and non‐mutated DNA, indicating a complete selection of resistant mitochondria and a maternally donated mode of resistance. © 2000 Society of Chemical Industry  相似文献

采用菌丝生长速率法和孢子萌发法分别测定了嘧菌酯对番茄早疫病菌Alternaria solani菌丝生长的抑制作用及其对分生孢子萌发的影响,同时测定了其对菌丝呼吸速率的影响。结果表明,嘧菌酯抑制番茄早疫病菌分生孢子萌发的作用强于对菌丝生长的抑制作用。抑制菌丝生长的剂量-反应曲线与抑制菌丝呼吸的剂量-反应曲线趋势相似,但菌丝在经嘧菌酯处理5~7 h后呼吸速率恢复到对照水平,相比之下,嘧菌酯却能一直抑制菌丝的生长。因此,采用孢子萌发法测定番茄早疫病菌对嘧菌酯的敏感性比用菌丝生长速率法测定更为适宜。水杨肟酸与嘧菌酯在抑制番茄早疫病菌菌丝生长和分生孢子萌发方面有协同作用,对菌丝生长的平均协同系数为7.24。随处理时间延长,药剂对菌丝耗氧的抑制作用下降,但仍抑制菌丝生长。研究表明:呼吸作用对药剂的敏感性随着处理的延长而下降的机理不是因为旁路氧化途径增强, 也不是因为基质中药剂效力的下降,而是存在其它机制。  相似文献

Mutants of Botrytis cinerea with moderate and high resistance to pyraclostrobin, a Qo inhibitor of mitochondrial electron transport at the cytochrome bc 1 complex, were isolated at a high mutation frequency, after nitrosoguanidine mutagenesis and selection on medium containing pyraclostrobin and salicylhydroxamate (SHAM), a specific inhibitor of cyanide-resistant (alternative) respiration. Oxygen uptake in whole cells was strongly inhibited in the wild-type strain by pyraclostrobin and SHAM, but not in the mutant isolates. Cross-resistance studies with other Qo and Qi inhibitors (QoIs and QiIs) of cytochrome bc 1 complex of mitochondrial respiration showed that the mutation(s) for resistance to pyraclostrobin also reduced the sensitivity of mutant strains to other QoIs as azoxystrobin, fluoxastrobin, trifloxystrobin and picoxystrobin, but not to famoxadone and to the QiIs cyazofamid and antimycin-A. An increased sensitivity of pyraclostrobin-resistant strains to the carboxamide boscalid, an inhibitor of complex II, and to the anilinopyrimidine cyprodinil, a methionine biosynthesis inhibitor, was observed. Moreover, no effect of pyraclostrobin resistance mutation(s) on fungitoxicity of the hydroxyanilide fenhexamid, the phenylpyrrole fludioxonil, the benzimidazole benomyl, and to the phenylpyridinamine fluazinam, which affect other cellular pathways, was observed. Study of fitness parameters in the wild-type and pyraclostrobin-resistant mutants of B. cinerea showed that most mutants had a significant reduction in the sporulation, conidial germination and sclerotia production. Experiments on the stability of the pyraclostrobin-resistant phenotype showed a reduction of resistance, mainly in moderate resistant strains, when the mutants were grown on inhibitor-free medium. However, a rapid recovery of the resistance level was observed after the mutants were returned to a selective medium. Studies on the competitive ability of mutant isolates against the wild-type parent strain, by applications of a mixed conidial population, showed that, in vitro, all mutants were less competitive than the wild-type strain. However, the competitive ability of high resistant mutants was higher than the moderate ones. Pathogenicity tests on cucumber seedlings showed that all mutant strains tested exhibited an infection ability similar with the wild-type parent strain. Preventive applications of the commercial product of F-500 25EC (pyraclostrobin) were effective against lesion development on cotyledons by the wild-type, but ineffective, even at high concentrations, against disease caused by the pyraclostrobin-resistant isolates. Boscalid (F-510 50WG) was found equally effective against the disease caused by the wild-type or pyraclostrobin-resistant mutants. This is the first report indicating the appearance of B. cinerea strains resistant to QoI fungicides by the biochemical mechanism of site modification and the risk for field resistance.  相似文献

The resistance to strobilurin-related fungicides and its molecular basis in laboratory mutant isolates of Cercospora beticola was investigated. After ultraviolet mutagenesis, mutants with high, moderate or low resistance levels to pyraclostrobin were isolated from a wild-type strain of C. beticola. Fungitoxicity tests on the response of resistant isolates on medium containing pyraclostrobin and salicylhydroxamate (SHAM), a specific inhibitor of cyanide-resistant (alternative) respiration, indicated that the biochemical mechanism of alternative oxidase was not responsible for the reduced sensitivity to pyraclostrobin for half of the mutants. Cross-resistance studies with other inhibitors of the cytochrome bc 1 complex of the mitochondrial respiratory chain showed that the mutation(s) for resistance to pyraclostrobin also reduced the sensitivity of mutant strains to other Qo inhibitors such as azoxystrobin and fenamidone, but not to the Qi inhibitor cyazofamid. No effect of pyraclostrobin-resistant mutation(s) on fungitoxicity of the carboxamide boscalid, the triazoles epoxiconazole and flutriafol and to the benzimidazole benomyl, which affect other cellular pathways or other steps of the respiratory chain, was observed. Study of fitness parameters showed that most mutants had a significant reduction in sporulation and pathogenicity compared to the wild-type parental isolate. However, experiments on the stability of the resistant phenotype did not show a significant reduction of the resistance for half of the mutants when grown for at least four generations on pyraclostrobin-free medium. Molecular analysis of cytochrome b cDNA, isolated from the wild-type and the pyraclostrobin-resistant mutant isolates, revealed two novel amino acid replacements at positions involved in Qo resistance in other species. The glycine (GGT) to serine (AGT) replacement at position 143 (G143S) was found in the isolate with the highly resistant phenotype. The second amino acid change was the replacement of phenylalanine (TTC) by valine (GTC) at position 129 (F129V), which was found in a mutant strain with the moderately resistant phenotype. Four additional mutations located in conserved regions of the mitochondrial cytochrome b gene (I154L, N250D, E256G and V261D) were detected in some mutant isolates of C. beticola but their possible role in Qo-resistance needs further investigation. This is the first study reporting C. beticola strains resistant to Qo inhibitor fungicides due to the biochemical mechanism of target-site modification, resulting from amino acid changes in the mitochondrial cytochrome b␣gene.  相似文献

浙江省果蔬灰霉病菌对嘧菌酯的抗药性研究   总被引:3,自引:2,他引:1  
采用菌丝生长速率法,连续监测了2010—2012年间浙江省果蔬灰霉病菌对QoI类杀菌剂嘧菌酯的敏感性变化。 结果表明:病菌群体中的低敏感性亚群体的比例明显上升,EC50值>5 mg/L 菌株的比例分别为12.5%、15.8%和28.3%;在菌丝生长阶段和孢子萌发阶段,旁路氧化在灰霉病菌对嘧菌酯敏感性中的平均相对贡献值(F)分别为2.91±0.89和5.72±2.82;嘧菌酯抗药性菌株的菌丝生长速率、产孢量、产菌核数和致病力与敏感菌株相比无显著差异。抗药性分子机制研究表明,灰霉病菌中存在2种类型的cyt b基因:Ⅰ型cyt b基因在第143位密码子后紧跟内含子;Ⅱ型cyt b基因在第143位密码子后没有紧跟内含子。大多数的灰霉病菌菌株属于Ⅱ型。Ⅰ型菌株均为嘧菌酯敏感菌株,Ⅱ型菌株为嘧菌酯敏感菌株或抗性菌株。抗性菌株的cyt b 基因的第143位密码子由甘氨酸(GGC)突变为了丙氨酸(GCC),抗药性机制为G143A。  相似文献

A new class of agricultural fungicides derived from strobilurins act as respiration inhibitors by binding to mitochondrial cytochrome b. The effects of the strobilurin, kresoxim-methyl, on conidia germination, mycelial growth and the protection of apple leaves from scab development were investigated for two isolates of Venturia inaequalis randomly selected from a culture collection. Inhibition of mycelial growth required relatively high doses of kresoxim-methyl (ED50=1 μg ml-1) for both isolates. In comparison, germination of conidia was highly sensitive for one of the isolates (ED50=0·005 μg ml-1), while the level of inhibition achieved for the second isolate was 60-fold less (ED50=0·3 μg ml-1). As deduced from identical sequences of cytochrome b cDNAs prepared from both isolates, the different responses of germinating conidia to kresoxim-methyl were not caused by differences in the sequence of cytochrome b as the target site for strobilurin action. Strong synergistic effects of salicylhydroxamic acid on kresoxim-methyl inhibitory potency suggested that the reduced kresoxim-methyl sensitivity observed for germinating conidia was caused by interference of the alternative respiratory pathway with inhibitor action. However, this mechanism of target site circumvention in germinating conidia had no adverse effects on the protection of apple leaves from scab infection by kresoxim-methyl. © 1998 Society of Chemical Industry  相似文献

为评价烟草赤星病致病菌链格孢Alternaria alternata对嘧菌酯的抗性风险,以敏感菌株J6为试材,通过菌丝药剂驯化和分生孢子紫外诱变诱导抗性突变体,并对抗性突变体的生物学特性进行了研究,同时对抗性突变体与敏感菌株线粒体的细胞色素b基因 (cyt b) cDNA序列全长进行了测序分析。结果表明:经药剂驯化未获得抗性突变体,而紫外诱变共获得7株抗性突变体,突变频率约为0.007%,抗性水平分别为5.27、8.28、25.28、12.82、6.14、9.28和52.91倍。适合度研究表明,抗性突变体与敏感菌株的分生孢子萌发能力及致病力相当,但分生孢子产生量均高于敏感菌株,菌丝生长速率除突变体6-1外均快于敏感菌株。cyt b基因cDNA序列分析表明:有4株抗性突变体在不同位点上发生了核苷酸突变,其中突变体6-7 cyt b的249位和871位碱基由T突变为C,但其编码的氨基酸未发生突变;突变体6-8 cyt b的734位碱基由T突变为C,引起所编码的245位丙氨酸突变为缬氨酸 (V245A);突变体6-9 cyt b的510位碱基由T突变为A,所编码的170位由精氨酸替代了丝氨酸 (S170R);突变体6-11 cyt b的732位碱基由T突变为A,所编码的244位由苯丙氨酸替代了亮氨酸 (L244F),其776位碱基由T突变为C,所编码的259位由丙氨酸替代了缬氨酸(V259A),其1 156位碱基由A突变为G,所编码的氨基酸未发生变化。研究结果初步表明,烟草赤星病菌对嘧菌酯存在潜在的抗药性风险,其cyt b基因的点突变与其对嘧菌酯的抗药性有关。  相似文献

The sensitivity of Venturia inaequalis field isolates to inhibitors of the cytochrome bc1 complex at the Qo site (QoIs) was characterised at the molecular, biochemical and physiological level, and compared to other respiration inhibitors. Comparison of a sensitive and a QoI-resistant isolate revealed very high resistance factors both in mycelium growth and spore germination assays. Cross-resistance was observed among QoIs such as trifloxystrobin, azoxystrobin, famoxadone, strobilurin B and myxothiazol. In the mycelium growth assay, antimycin A, an inhibitor of the cytochrome bc1 complex at the Qi site, was less active against the QoI-resistant than against the sensitive isolate. The mixture of QoIs with salicylhydroxamic acid (SHAM), an inhibitor of the alternative oxidase, exerted synergistic effects in the spore germination but not in the mycelium growth assay. Thus, the cytochrome and the alternative respiration pathways are assumed to play different roles, depending on the developmental stage of the fungus. Induction of alternative oxidase (AOX) by trifloxystrobin was observed in mycelium cells at the molecular level for the sensitive but not the resistant isolate. Following QoI treatment, respiration parameters such as oxygen consumption, ATP level, membrane potential and succinate dehydrogenase activity were only slightly reduced in Qo-resistant mycelium cells, and remained at much higher levels than in sensitive cells. In contrast, no difference was observed between sensitive and resistant isolates when NADH consumption was measured. Comparison of the cytochrome b (cyt b) gene of the sensitive and resistant isolates did not reveal any point mutations as is known to occur in resistant isolates of other plant pathogens. It is assumed that QoI resistance in V inaequalis may be based on a compensation of the energy deficiency following QoI application upstream of the NADH dehydrogenase of the respiratory chain.  相似文献

Strobilurin类杀菌剂是在具有杀菌活性的天然β-甲氧基丙烯酸酯衍生物的基础上研发出来的,具有广谱、高效、安全的特点。主要以粘噻唑为例综述了该类杀菌剂作用靶标及作用位点的研究过程及最新进展。生化作用机理的研究明确了粘噻唑作用于线粒体呼吸链bc1复合物中的细胞色素b及铁硫蛋白,阻止了从细胞色素b到细胞色素c1的电子传递,从而抑制线粒体的呼吸作用;蛋白质晶体学研究明确了粘噻唑的作用位点(Qo位点)是通过氢键与铁硫蛋白及细胞色素b的残基结合的。其他strobilurin类杀菌剂和粘噻唑具有相同的靶标,但由于各化合物之间存在一定的结构差异,其具体作用位点亦有一定的差别。  相似文献

QoI fungicides, inhibitors of mitochondrial respiration at the Qo site of cytochrome b in the mitochondrial bc(1) enzyme complex, are commonly applied in vineyards against Plasmopara viticola (Berk. & MA Curtis) Berl. & De Toni. Numerous treatments per year with QoI fungicides can lead to the selection of resistant strains in the pathogen population owing to the very specific and efficient mode of action. In order to evaluate the resistance risk and its development, two different methods, biological and molecular, were applied to measure the sensitivity of oospores differentiated in vineyards, both treated and untreated with azoxystrobin, from 2000 to 2004. Assays using oospores have the advantage of analysing the sensitivity of bulked samples randomly collected in vineyards, describing accurately the status of resistance at the end of the grapevine growing season. Both methods correlated well in describing the resistance situation in vineyards. QoI resistance was not observed in one vineyard never treated with QoI fungicides. In the vineyard where azoxystrobin had been used in mixture with folpet, the selection of QoI-resistant strains was lower, compared with using solely QoI. In vineyards where QoI treatments have been stopped, a decrease in resistance was generally observed.  相似文献

测定了苯并咪唑类杀菌剂敏感-乙霉威抗性(BenS-DieR)、苯并咪唑类杀菌剂抗性-乙霉威敏感(BenR-DieS)和苯并咪唑类杀菌剂抗性-乙霉威抗性(BenR-DieR)3种类型的番茄叶霉病菌Cladosporium fulvum菌株对不同类型药剂的敏感性。结果表明,蕃茄叶霉病菌对供试药剂的敏感性与其对苯并咪唑类杀菌剂及乙霉威的敏感性无关。根据药剂对3类菌株EC50值的平均值, 16种杀菌剂抑制菌丝生长的活性依次为腐霉利>乙烯菌核利>异菌脲>戊唑醇>百菌清>嘧霉胺>醚菌酯>代森锰锌>8-羟基喹啉铜>丙环唑>苯醚甲环唑>嘧菌酯>灭锈胺>烯酰吗啉>烟酰胺>三唑酮;抑制孢子萌发的活性依次为醚菌酯>腐霉利>百菌清>乙烯菌核利>灭锈胺>8-羟基喹啉铜>异菌脲>代森锰锌>嘧菌酯>烟酰胺>嘧霉胺>戊唑醇>丙环唑>苯醚甲环唑>三唑酮>烯酰吗啉。通过紫外诱变共获得17株抗异菌脲突变体,突变频率为4.5×10-7。其中低抗、中抗和高抗菌株分别占 17.65%、70.59%和11.75%。这些突变体对腐霉利和乙烯菌核利表现交互抗性,对苯并咪唑类、脱甲基抑制剂(DMIs)、QoIs等药剂的敏感性与亲本菌株之间没有显著性差异,与亲本菌株在生长、产孢、致病能力等方面也无显著差异,但对渗透胁迫的敏感性要显著高于亲本。  相似文献

Mutants of Ustilago maydis (DC) Corda with high resistance to azoxystrobin (RF 164 to 4714, based on EC50 values), an inhibitor of mitochondrial electron transport at the cytochrome bc1 complex, were isolated in a mutation frequency of 2.3 x 10(-7) after nitrosoguanidine mutagenesis and selection on media containing 1 microgram ml-1 azoxystrobin in addition to 0.5 mM salicylhydroxamate (SHAM), a specific inhibitor of cyanide-resistant (alternative) respiration. Oxygen uptake in whole cells was strongly inhibited in the wild-type strains by azoxystrobin (1.5 micrograms ml-1) in addition to SHAM (1 mM), but not in the mutant isolates. Genetic analysis with nine such mutant isolates resulted in progeny phenotypes which did not follow Mendelian segregation, but satisfied the criteria of non-Mendelian (cytoplasmic) heredity. In crosses between three mutant isolates with the compatible wild-type strains, the sensitivity was inherited by progeny maternally from the wild-type parent strain (criterion of uniparental inheritance). In crosses between wild-type strains and remaining mutant isolates, a continuous distribution of sensitivity in the progeny was found (criterion of vegetative segregation). The third criterion of cytoplasmic resistance (criterion of intracellular selection) was fulfilled by experiments on the stability of resistance phenotypes. With two exceptions, a reduction of resistance was observed in the mutant strains when they were grown on inhibitor-free medium. Recovery of the high resistance level was observed after they were returned to the selection medium. Cross-resistance studies with other fungicides, which also inhibit electron transport through complex III of respiratory chain, showed that mutations for resistance to azoxystrobin were also responsible for reduced sensitivity to kresoxim-methyl (RF 18 to 1199) and to antimycin-A (RF 20 to 305), which act at the Qo and Qi sites of the cytochrome bc1 complex, respectively. Studies of the fitness of azoxystrobin-resistant isolates showed that these mutations appeared to be pleiotropic, having significant adverse effects on growth in liquid culture and pathogenicity on young corn plants.  相似文献

The ability to develop fungicide resistance was assessed in Monilinia fructicola isolates with different fungicide sensitivity phenotypes by adapting mycelium and conidia to increasing concentrations of selective fungicides and UV mutagenesis. Results showed that adaptation to Quinone outside inhibitor (QoI) fungicide azoxystrobin and sterol demethylation inhibitor (DMI) fungicide propiconazole was more effective in conidial-transfer experiments compared to mycelial-transfer experiments. DMI-resistant (DMI-R) isolates adapted to significantly higher doses of azoxystrobin in both, mycelial- and conidial-transfer experiments compared to benzimidazole-resistant (BZI-R) and sensitive (S) isolates. Adaptation to propiconazole in conidial-transfer experiments was accelerated in BZI-R isolates when a stable, nonlethal dose of 50 microg/ml thiophanate-methyl was added to the selection medium. One of two azoxystrobin-resistant mutants from DMI-R isolates did not show any fitness penalties; the other isolate expired before further tests could be carried out. The viable mutant caused larger lesions on detached peach fruit sprayed with azoxystrobin compared to the parental isolate. The azoxystrobin sensitivity of the viable mutant returned to baseline levels after the mutant was transferred to unamended medium. However, azoxystrobin resistance recovered quicker in the mutant compared to the corresponding parental isolate after renewed subculturing on medium amended with 0.2 and 1 microg/ml azoxystrobin; only the mutant but not the parental isolate was able to adapt to 5 microg/ml azoxystrobin. In UV mutagenesis experiments, the DMI-R isolates produced significantly more mutants compared to S isolates. All of the UV-induced mutants showed stable fungicide resistance with little fitness penalty. This study indicates the potential for QoI fungicide resistance development in M. fructicola in the absence of a mutagen and provides evidence for increased mutability and predisposition to accelerated adaptation to azoxystrobin in M. fructicola isolates resistant to DMI fungicides.  相似文献

番茄叶霉病菌对嘧菌酯的抗性检测及抗性风险评估   总被引:3,自引:1,他引:2  
利用孢子萌发法测定了从河北、浙江、江苏、山东、四川等省田间采集的97个番茄叶霉病菌菌株对嘧菌酯的敏感性。结果表明,田间番茄叶霉病菌已对嘧菌酯产生不同程度的抗药性。97个菌株可分为敏感菌株(EC50值50值5.4~54 μg/mL),中抗菌株(EC50值54~270 μg/mL)、高抗菌株(EC50值270~1 350 μg/mL)、特高抗菌株(EC50值>1 350 μg/mL) 5种类型,分别占55.67%、14.43%、11.34%、10.31%和8.25%。其中从未用过甲氧基丙烯酸酯类杀菌剂的27个菌株的EC50平均值为(0.544±0.349) μg/mL,可作为敏感基残;高抗和特高抗菌株集中在常施用甲氧基丙烯酸酯类杀菌剂的河北定州。通过紫外线诱导和药剂驯化的方法获得了番茄叶霉病菌5个抗嘧菌酯突变体,其抗药性不稳定,在不含药PDA平板上继代培养后抗药性水平下降,甚至恢复至敏感状态;除突变体cf 21M1的产孢量高于其亲本外,抗药性突变体的菌落生长速率、产孢量、孢子萌发率均显著低于其亲本菌株,所有抗嘧菌酯突变体及其亲本菌株接种于番茄离体叶片后,突变体病情指数与亲本菌株之间没有显著差异。嘧菌酯与多菌灵、苯醚甲环唑之间不存在交互抗药性。田间抗药性检测及室内抗药性风险评估结果表明,番茄叶霉病菌对嘧菌酯具有较高的抗药性风险。  相似文献

The molecular profile and the biological response of isolates of Pyricularia oryzae Cavara obtained from ctenanthe to two strobilurins (azoxystrobin, kresoxim-methyl) and the phenylpyridinamine fungicide fluazinam were characterized, and compared with isolates from rice plants. Five different isozymes (alpha-esterase, lactate, malate, isocitrate and sorbitol dehydrogenases) and five random decamer primers for RAPD-PCR were used to generate molecular markers. Using unweighted pair-group with arithmetic average analysis, ctenanthe isolates were found to form a separate group distinct from that of the rice isolates for both sets of markers. Amplified polymorphic sequences of mitochondrial cytochrome b that were digested with Fnu4HI or StyI revealed no differences among Pyricularia isolates at amino acid positions 143 or 129 which confer resistance to strobilurins in several fungi. In absence of the alternative respiration inhibitor salicylhydroxamic acid (SHAM) the three fungicides showed inferior and variable efficacy, with a trend toward the rice isolate being less sensitive. The addition of SHAM enhanced the effectiveness of all fungicides against isolates regardless of their origin. Appressorium formation was the most vulnerable target of action of the respiration inhibitors and azoxystrobin the most effective. This is the first report of a comparison between the molecular profiles and sensitivities to respiration inhibitors for Pyricularia oryzae isolates from a non-gramineous host and from rice.  相似文献

The necrotrophic fungal phytopathogen Sclerotinia sclerotiorum (Lib.) de Bary has a broad host range and frequently causes destructive diseases. The extensive use of common fungicides to control these diseases has selected for resistance in populations of S. sclerotiorum. In this study, 105 isolates of S. sclerotiorum from different geographical regions in Jiangsu Province of China were characterized for baseline sensitivity to azoxystrobin, and the average EC50 value was 0.2932 μg/mL for mycelial growth. Of the mixtures of the fungicides thiram and azoxystrobin that were tested using an in vitro mycelial growth assay, the 1:4 ratio provided the greatest inhibition of S. sclerotiorum. When tested against nine isolates, the 1:4 mixture resulted in a mean synergy ratio of 2.31, indicating synergistic inhibition. Mycelial respiration was inhibited for about 2 h by azoxystrobin alone but for 48 h by the mixture of thiram and azoxystrobin. Salicylhydroxamic acid (SHAM, a known inhibitor of alternative respiration) also increased the inhibition of mycelial growth and respiration caused by azoxystrobin. These results suggest the need for further study of effects of combinations of azoxystrobin with thiram or SHAM in planta to evaluate their potential for management of diseases caused by S. sclerotiorum.  相似文献

由立枯病丝核菌引起的烟草立枯病是我国烟草苗床上危害最严重的病害之一。本研究评价了5种杀菌剂 (嘧菌酯、啶酰菌胺、氟啶胺、丙环唑和嘧霉胺) 对立枯丝核菌菌丝生长、菌核形成和萌发的影响,以及其对烟草立枯病的防治效果。结果表明:立枯丝核菌菌丝对氟啶胺和嘧菌酯的敏感性高于丙环唑和啶酰菌胺,而对嘧霉胺的敏感性较低;嘧菌酯对菌核形成的抑制作用强于丙环唑、氟啶胺、啶酰菌胺和嘧霉胺;5种杀菌剂对立枯丝核菌菌核萌发均无抑制作用。在离体烟叶的保护活性方面,12.5和50 mg/L的嘧菌酯和啶酰菌胺对立枯病的保护作用优于氟啶胺、丙环唑和嘧霉胺;在治疗活性方面,50和200 mg/L的嘧菌酯的治疗作用优于其他4种杀菌剂。因此,供试的5种杀菌剂中嘧菌酯最适合用于烟草立枯病的防治。  相似文献
