Culicoides species (Diptera: Ceratopogonidae) belonging to the Obsoletus and Pulicaris groups are considered to be the main vectors of bluetongue virus (BTV) in non Mediterranean Europe. Selected terrestrial microhabitats (n = 17) on a farm in northern Spain were sampled repeatedly over a year-long period and characterized for use by Culicoides species for immature development. Concurrent use of CDC light traps showed the presence of 37 species and 66,575 specimens of adult Culicoides. A total of 28 species and 11,396 individuals emerged from laboratory-maintained soil samples. Culicoides obsoletus and Culicoides scoticus (pooled as Obsoletus complex) were particularly abundant (comprising 58.6% and 74.5% of the total collections in light traps and emergence traps respectively). Potential key vectors of animal viruses (such as BTV) were found in two main terrestrial types of microhabitats. In the case of C. obsoletus, different types of manure (old and composted manure, manure mixed with organic matter, and fresh manure) produced most of the specimens. In contrast, larvae of C. scoticus and Culicoides lupicaris were associated with soil substantially comprised of rotting leaf litter that included the parasitic plant Lathraea clandestina. Several species, Culicoides festivipennis, Culicoides punctatus and Culicoides brunnicans, were very common in mud at pond margins. Indeed, pond microhabitats and runoff below barn rooflines supported the greatest species richness. In the pond habitat, 49.4% of Culicoides specimens emerged from mud at the water edge, as opposed to 50 cm above (32.4%) and 1 meter above waterline (18%). Similar species richness, but statistically significant differences in abundance, were observed among the four pond microhabitats. Overall, the majority of the specimens were found in the upper layer (0–3 cm), except in manure, where they preferred deeper layers (>6 cm). Maximum peaks of abundance occurred in both light traps and soil samples in summer months, whereas increased captures in autumn were noticed only in light traps. Both trapping systems failed to collect adult Culicoides midges in the coldest months of December, January and February. The literature on immature habitats of species suspected in BTV transmission in Europe, the Pulicaris group and particularly the Obsoletus group, is briefly reviewed.  相似文献

Bluetongue (BT) is an important disease of ruminants which exhibits its most severe clinical signs on cattle and especially on certain breeds of sheep. The known vectors of BT are small insects of the genus Culicoides (Diptera; Ceratopogonidae). Two species from this genus – Culicoides imicola and Culicoides obsoletus – play the major role in the transmission of the disease in Europe. Several prophylactic methods are used to avoid transmission; however, an easy and cost-effective preventive technique would be very useful for the control of the Culicoides populations near the animals. In the present study, the insecticide effect of cypermethrin treated nets on a Culicoides population was evaluated. A polyethylene net sprayed with 1 L cypermethrin solution (1%) surrounding a UV light suction trap was placed at a cattle farm in Majorca (Balearic Islands). Collections of Culicoides and other fauna from the trap and floor around the net were compared with a control. Results showed no significant differences in the collection of Culicoides midges between the insecticide-treated net and the control. However, significant differences were observed in the collection of the non-target fauna between the treated net and the control, indicating that the dose used in the present trial was enough to kill most of the arthropods that contacted the net. The reasons for these equivocal findings and means to improve this technique for the control of Culicoides midges are discussed.  相似文献

Interleukin-3 is a growth and differentiation factor for various hematopoietic cells. IL-3 also enhances stimulus-dependent release of mediators and cytokine production by mature basophils. Function of IL-3 has not been studied in horses because of lack of horse-specific reagents. Our aim was to produce recombinant equine IL-3 and test its effect on sulfidoleukotriene and cytokine production by equine peripheral blood leukocytes (PBL).Equine IL-3 was cloned, expressed in E. coli and purified. PBL of 19 healthy and 20 insect bite hypersensitivity (IBH)-affected horses were stimulated with Culicoides nubeculosus extract with or without IL-3. Sulfidoleukotriene (sLT) production was measured in supernatants by ELISA and mRNA expression of IL-4, IL-13 and thymic stromal lymphopoietin (TSLP) assessed in cell lysate by quantitative real-time PCR.Recombinant equine IL-3 (req-IL-3) had a dose dependent effect on sLT production by stimulated equine PBL and significantly increased IL-4, IL-13 and TSLP expression compared to non-primed cells.IL-3 priming significantly increased Culicoides-induced sLT production in IBH-affected but not in non-affected horses and was particularly effective in young IBH-affected horses (≤3 years).A functionally active recombinant equine IL-3 has been produced which will be useful for future immunological studies in horses. It will also allow improving the sensitivity of cellular in vitro tests for allergy diagnosis in horses.  相似文献

Horses with insect bite hypersensitivity (IBH) have difficulty in completely avoiding allergens, so effective treatment options are required. A randomised, placebo controlled and double blinded field study was conducted to determine the pharmacokinetics and efficacy in reducing dermatitis of the antihistamine cetirizine given orally at 0.4 mg/kg twice daily for 3 weeks. The influence of protection blankets and stabling were also investigated.The estimated maximum plasma concentration (Cmax) and trough plasma concentration of cetirizine were 135 ng/mL and 18 ng/mL, respectively. There was no difference in dermatitis reduction between the treatment and placebo groups (P = 0.77). The findings indicated that cetirizine was of no apparent benefit in treating IBH at the dose rate tested. The use of blankets and stabling were shown to have favourable influence on the dermatitis (P < 0.05) and may be the preferred options to prevent this condition.  相似文献

Insect bite hypersensitivity (IBH) is an allergic dermatitis of horses caused by IgE-mediated reactions to bites of Culicoides and sometimes Simulium spp. The allergens causing IBH are probably salivary gland proteins from these insects, but they have not yet been identified. The aim of our study was to identify the number and molecular weight of salivary gland extract (SGE) proteins derived from Culicoides nubeculosus which are able to bind IgE antibodies (ab) from the sera of IBH-affected horses. Additionally, we sought to investigate the IgG subclass (IgGa, IgGb and IgGT) reactivity to these proteins. Individual IgE and IgG subclass responses to proteins of C. nubeculosus SGE were evaluated by immunoblot in 42 IBH-affected and 26 healthy horses belonging to different groups (Icelandic horses born in Iceland, Icelandic horses and horses from different breeds born in mainland Europe). Additionally, the specific antibody response was studied before exposure to bites of Culicoides spp. and over a period of 3 years in a cohort of 10 Icelandic horses born in Iceland and imported to Switzerland. Ten IgE-binding protein bands with approximate molecular weights of 75, 66, 52, 48, 47, 32, 22/21, 19, 15, 13/12 kDa were found in the SGE. Five of these bands bound IgE from 50% or more of the horse sera. Thirty-nine of the 42 IBH-affected horses but only 2 of the 26 healthy horses showed IgE-binding to the SGE (p<0.000001). Similarly, more IBH-affected than healthy horses had IgGa ab binding to the Culicoides SGE (19/22 and 9/22, respectively, p<0.01). Sera of IBH-affected horses contained IgE, IgGa and IgGT but not IgGb ab against significantly more protein bands than the sera of the healthy horses. The cohort of 10 Icelandic horses confirmed these results and showed that Culicoides SGE specific IgE correlates with onset of IBH. IBH-affected horses that were born in Iceland had IgGa and IgGT ab (p< or =0.01) as well as IgE ab (p=0.06) against a significantly higher number of SGE proteins than IBH-affected horses born in mainland Europe. The present study shows that Culicoides SGE contains at least 10 potential allergens for IBH and that IBH-affected horses show a large variety of IgE-binding patterns in immunoblots. These findings are important for the future development of a specific immunotherapy with recombinant salivary gland allergens.  相似文献

In this work the clinical evolution and the specific serum IgG and IgE antibody responses in sheep after primary (n = 10) and secondary (n = 4) experimental challenges with the mange mite Sarcoptes scabiei var. ovis were studied. The primary infection was characterized by the development of mange lesions in all sheep, a detection of live S. scabiei mites in 70% skin scrapings taken in week 10 post-challenge (PC), strongly raised and sustained specific IgG levels and a more moderate but continuous rise in specific IgE levels. Seroconversion was detected for IgG and IgE by ELISA in 90% and 60% of the sheep in week 8 PC, respectively. By Western-blotting (WB), ten IgG-reactive bands (36–120 kDa) and four IgE-reactive bands (90–180 kDa) were observed in week 8 PC. Following the secondary challenge the ewes developed a smaller area of mange lesion than that seen following primary challenge and live S. scabiei mites were not detected in skin scrapings collected in week 8 PC, suggesting that sheep had developed immunity to re-infection. Compared to primary infection, the specific IgG secondary antibody levels were transient, but in contrast there was an anamnestic IgE response, resulting in an elicitation of specific serum IgE levels in week 2 PC significantly higher than those demonstrated after primary infection. WB analysis revealed one additional IgG-reactive band (180 kDa) and no additional IgE-reactive bands. Determining the immunodiagnostic or vaccination value of the IgG-reactive antigens and IgE-reactive allergens detected requires further studies.  相似文献

Vitamin D is essential in calcium and phosphorus regulation, bone physiology, cell proliferation and epithelial integrity. Literature on vitamin D in growing horses is sparse, and the effect of age on vitamin D has not been evaluated in equids in the United States or in tropical countries. The goal of this study was to determine if there was an effect of age on serum 25(OH)D3 concentrations in equids in the US (Ohio/Kentucky) and Thailand (Chiang Rai and Kanchanaburi) during the same time of the year. Blood samples were collected from healthy ponies (n = 21) and Thoroughbred foals (n = 13), yearlings (n = 10), and horses (n = 20) in Thailand and from Thoroughbred foals (n = 10) and horses (n = 17) in the US. Serum concentrations of 25(OH)D3, calcium and phosphorus were measured.In both countries, serum 25(OH)D3 concentrations were lower in foals than in yearlings and adult horses. Serum 25(OH)D3 concentrations were higher in horses than in ponies in Thailand, but were not different between horses from either country. Calcium concentrations were not different between groups or location. In both countries, phosphorus concentrations were higher in foals than in older groups; however, were not different between ponies and horses. This study shows that independent of geography there are age-related differences in 25(OH)D3 concentrations in horses and further confirms that 25(OH)D3 concentrations are lower in horses compared to other species. The information will serve as the basis for future clinical studies and to help understand better the pathophysiology of equine disorders associated with calcium and phosphorus dysregulation.  相似文献

Recurrent airway obstruction (RAO) is a common condition in stabled horses characterised by small airway inflammation, airway neutrophilia and obstruction following exposure of susceptible horses to mouldy hay and straw and is thus regarded as a hypersensitivity reaction to mould spores. However, the role of IgE-mediated reactions in RAO remains unclear.The aim of the study was to investigate with a serological IgE ELISA test (Allercept?), an in vitro sulfidoleukotriene (sLT) release assay (CAST®) and with intradermal testing (IDT) whether serum IgE and IgE-mediated reactions against various mould, mite and pollen extracts are associated with RAO. IDT reactions were evaluated at different times in order to detect IgE-mediated immediate type reactions (type I hypersensitivity reactions, 0.5–1 h), immune complex-mediated late type reactions (type III reactions, 4–10 h) and cell-mediated delayed type reactions (type IV hypersensitivity reactions 24–48 h).In the serological test, overall the control horses displayed more positive reactions than the RAO-affected horses but the difference was not significant. Comparison of the measured IgE levels showed that the RAO-affected horses had slightly higher IgE levels against Aspergillus fumigatus than controls (35 and 16 AU, respectively, p < 0.05), but all values were below the cut off (150 AU) of the test. In the sLT release assay, seven positive reactions were observed in the RAO-affected horses and four in the controls but this difference was not significant.A significantly higher proportion of late type IDT reactions was observed in RAO-affected horses compared to controls (25 of 238 possible reactions versus 12 of 238 possible reactions, respectively, p < 0.05). Interestingly, four RAO-affected but none of the control horses reacted with the recombinant mould allergen A. fumigatus 8 (rAsp f 8, p < 0.05), but only late phase and delayed type reactions were observed.In all three tests the majority of the positive reactions was observed with the mite extracts (64%, 74% and 88% of all positive reactions, respectively) but none of the tests showed a significant difference between RAO-affected and control animals. Our findings do not support that IgE-mediated reactions are important in the pathogenesis of RAO. Further studies are needed to investigate whether sensitisation to mite allergens is of clinical relevance in the horse and to understand the role of immune reactions against rAsp f 8.  相似文献

Subtle lameness in horses may be difficult to diagnose and methods to evaluate lameness objectively are useful when equine clinicians fail to reach a consensus. The aim of this study was to determine whether equine pressure plate measurements are repeatable when used to calculate forelimb loading (peak vertical pressure [PVP], peak vertical force [PVF], vertical impulse [VI]) and symmetry ratios, and to establish if these data are similar to the ‘gold standard’ force plate values. Since plate dimensions are relatively small, ponies were used to enable recordings to be taken from both forelimbs in one trial. Five sound ponies were walked and trotted over a pressure plate which was embedded in a custom-made runway. For each pony, five valid trials were recorded during two different days. A trial was considered valid if complete prints of both fore hooves were recorded consecutively and if velocity was within a preset range.The PVP, PVF and VI values showed an acceptable variability (CV ? 16%), with PVF (130% of bodyweight [BW], n = 5) similar to previously reported force plate data (128% BW, n = 48). Mean symmetry ratios appeared to be high (>95%) and showed a low variability (CV < 5%). The stand-alone pressure plate permitted adequate registration of forelimb PVF, VI and limb symmetry with a high level of precision in sound ponies.  相似文献

A comparison of animal gains and vegetation trends was made from 2002–2008 between a continuous season-long stocking (SLS) system and a modified intensive–early stocking system (IES) with late-season grazing (IES 1.6× + 1; 1.6 times the number of animals of the SLS system from May 1 to July 15, and 1 times the number of animals of SLS from July 15 to October 1) on shortgrass native rangeland of western Kansas. The continuous season-long stocked system placed animals at a density of 1.37 ha · steer?1 from May through October, or 2.63 animal unit months (AUM) · ha?1, whereas the intensive–early stocked system with late-season grazing (3.33 AUM · ha?1) stocked pastures at 0.85 ha · steer?1 from May through the middle of July, and then stocked pastures at 1.37 ha · steer?1 for the remainder of the grazing season by removing the heaviest animals mid-July each yr. Average daily gains (0.78 vs. 0.70 kg · d?1, P = 0.039) and total animal gain (58 vs. 52 kg, P = 0.042) were different between the continuous season-long stocked and the intensive–early stocked animals during the first half of the grazing season. No difference was found between average daily gain (0.61 vs. 0.62 kg · d?1, P = 0.726) and total animal gain (48 vs. 49 kg, P = 0.711) for the continuous season-long stocked and intensive–early stocked with late-season grazing animals during the last half of the season. Total individual animal gain (106 vs. 101 kg, P = 0.154) and average daily gain (0.70 vs. 0.66 kg · d?1, P = 0.152) was not different between the continuous season-long stocked and the intensive–early stocked system animals that were on pasture the entire grazing season. Total beef gain on a land-area basis (96 vs. 77 kg · ha?1, P = 0.008) was greater for the modified intensive–early stocked system with late-season grazing with greater animal densities. Changes in residual biomass and most key vegetation components at the end of the grazing season were not different between the two systems.  相似文献

Johne's disease is an enteric mycobacterial infection of ruminants that has significant global economic impact. The classic host reaction is one of an early T-cell mediate immune response, with predominant interferon gamma (IFNγ) activity; there is subsequent lowering of this response as animals reach the terminal stages of disease. Interleukin (IL)-10, which can suppress Th1-type and enhance Th2-type cytokine production, is considered to play a role in the later stages of Johne's disease. To determine the role of IL-10 throughout the course of Johne's disease we studied groups of sheep with either no Johne's disease (n = 10), natural infection (n = 30) or experimental infection (n = 58). Disease status of the animals was comprehensively assessed by culture of Mycobacterium avium subsp. paratuberculosis (Mptb), histopathology and serology. Antigen-specific IL-10 secretion in peripheral blood of sheep exposed to Mptb was significantly higher than in control animals (P < 0.001) as early as 4 months post-inoculation, and increased progressively. In ileal and jejunal lymph node cells, IL-10 secretion was also significantly higher in animals that were exposed to Mptb compared to controls (P < 0.05). The early IL-10 response seen in peripheral blood cells may be a reflection of early responses at sites of Mptb infection. IL-10 secretion from ileal and jejunal lymph node cells was significantly higher in exposed animals with no lesions or with paucibacillary lesions when compared to animals with multibacillary lesions. These novel findings demonstrate that increased IL-10 activity commences soon after exposure to the causative mycobacterium and may play a role in determining disease outcome.  相似文献

Athletic performance is both a stress factor and an adaptive response to exercise that may be modulated by training, reduce inflammation and help prevent disease. Studies on the endocrinology of exercise and training have demonstrated the existence of an integrated metabolic network of hormone and cytokine regulation. Subsequent molecular studies have shown that repeated bouts of exercise may establish new basal levels of gene expression at rest.The Thoroughbred horse may be a useful ‘exercise model’ for inter-individual comparisons between subjects with homogeneous genetic and environmental backgrounds and similar exercise management practices. In this study, the effects of training and acute effort on gene expression were evaluated with a real time PCR approach in athletic (n = 10) and sedentary horses (n = 9), using a previously characterised panel of genes known to be highly modulated during effort (CXCL2, TLR4, IL1β, IL8, IL1RII, IL18, IL6 and CEBPβ). A ‘rest comparison’ was performed to evaluate a training effect in both groups while a ‘race comparison’ was performed in athletic horses only (before, immediately after, and 12 h after racing) to determine the effect of acute effort.The results indicated that many of the investigated genes (TLR4, IL1β, IL1RII, IL18, IL6 and CEBPβ) were expressed to a greater extent in athletic horses compared to sedentary animals when both were at rest. However, a time-course comparison in the athletic horses revealed that genes exhibiting the highest levels of expression at rest did not show significant changes after the race. The findings suggested that training may exert a conditioning on gene expression at rest leading to a more prompt response to exercise-induced stress in Thoroughbreds.  相似文献

Insect bite hypersensitivity (IBH) is an equine skin allergy caused by bites of Culicoides spp. and impacts on the welfare of affected horses. The aim of this study was to identify and quantify risk factors for IBH. Data from 3453 Friesian horse mares and 7074 Shetland pony mares scored for IBH by inspectors during obligatory foal inspections were analysed using breed-specific multivariable logistic regression models. The combined effect of month and year of scoring, Province and inspector were significantly associated with IBH in both breeds. In Shetland pony mares, withers height and coat colour were also significantly associated with IBH, while body condition had a nearly significant effect. The outcomes from this study on risk factors might contribute to the development of more efficient measures to reduce the prevalence of IBH.  相似文献

Feline obesity generally results in aberrations to plasma metabolite levels, such as lipid concentrations and lipoprotein composition. This study sought to investigate the resultant effect of obesity on cholesterol lipoprotein composition and circulating adiponectin concentrations in cats. Plasma glucose, lipids (triglyceride, cholesterol and free fatty acid), insulin and adiponectin concentrations, and cholesterol lipoprotein composition were measured and compared between body condition score (BCS) determined normal healthy control and obese cats. Although the obese group demonstrated higher levels of plasma cholesterol, glucose, and triglycerides, as compared to healthy controls, the difference was insignificant thus indicating that the BCS determined obese cats may have been overweight and not morbidly obese. Plasma insulin levels were significantly higher (25–30%) versus healthy control animals thereby possibly hinting at the ensuing emergence of obesity induced insulin resistance. However, the BCS determined obese cat demonstrated a significant reduction (p < 0.05) in plasma adiponectin concentration and a significant increase (p < 0.05) in LDL-cholesterol % as compared to age matched healthy control animals. This would indicate that changes in plasma adiponectin concentration and cholesterol lipoprotein composition may be good early indicators of obesity in cats.  相似文献

We conducted a study on how Acacia sieberiana respond to repeated burning in the Kidepo National Park in northeastern Uganda. The study was conducted to understand effects of common burning regimes (early dry season, late dry season, and no burn [control]) in the area on Acacia sieberiana. The three treatments were applied for three consecutive years to 14 replicate blocks in a randomized block design. All A. sieberiana trees were number tagged and monitored for height and girth (diameter at breast height) growth. All fires were set as head-fires and attained intensity ranging between 422 and 5693 kW · m−1. Both early and late dry season burning increased the number of small (< 49 cm) A. sieberiana trees after 2 yr. Burning did not affect the growth rates. Although the number of trees < 49 cm increased after 2 yr, the mortality in this height class was also increased by the late dry season burning, and after 3 yr of consecutive burning there were no statistical treatment differences in the height class < 49 cm. Late dry season burning also led to high mortality among trees > 250 cm in the third year. Mortality attributed to elephant browsing was important in all treatments but a substantial portion of mortality could not be attributed to any particular cause. In the late burn, fire was the most important mortality factor. Thus, 2 yr of burning may be used as a tool to stimulate recruitment of A. sieberiana, but additional years of late dry season burning will increase the mortality of older trees.  相似文献

In this preliminary study, a novel DNA microarray system was tested for the diagnosis of bovine piroplasmosis and anaplasmosis in comparison with microscopy and PCR assay results. In the Dakahlia Governorate, Egypt, 164 cattle were investigated for the presence of piroplasms and Anaplasma species. All investigated cattle were clinically examined. Blood samples were screened for the presence of blood parasites using microscopy and PCR assays. Seventy-one animals were acutely ill, whereas 93 were apparently healthy. In acutely ill cattle, Babesia/Theileria species (n = 11) and Anaplasma marginale (n = 10) were detected. Mixed infections with Babesia/Theileria spp. and A. marginale were present in two further cases. A. marginale infections were also detected in apparently healthy subjects (n = 23). The results of PCR assays were confirmed by DNA sequencing. All samples that were positive by PCR for Babesia/Theileria spp. gave also positive results in the microarray analysis. The microarray chips identified Babesia bovis (n = 12) and Babesia bigemina (n = 2). Cattle with babesiosis were likely to have hemoglobinuria and nervous signs when compared to those with anaplasmosis that frequently had bloody feces. We conclude that clinical examination in combination with microscopy are still very useful in diagnosing acute cases of babesiosis and anaplasmosis, but a combination of molecular biological diagnostic assays will detect even asymptomatic carriers. In perspective, parallel detection of Babesia/Theileria spp. and A. marginale infections using a single microarray system will be a valuable improvement.  相似文献
