The purpose of this trial was to determine whether an addition of folic acid to a commercial diet would affect serum Zn, Fe and Cu status in sows between weaning and 30 d of gestation. At weaning, 162 sows were assigned randomly to six groups and housed in individual cages fitted on a slatted floor. There were six treatments according to a 2 X 3 factorial arrangement: two levels of supplementary folic acid (0 and 5 mg/kg of diet) and three treatments to stimulate ovulation (none, flushing and pregnant mare serum gonadotropin [PMSG] i.m. injection). Control groups were fed a commercial-type diet, and folic acid-treated groups were fed the same diet supplemented with 5 mg/kg of pteroylglutamic acid. All sows were mated twice within 7 d after weaning. Of the 162 animals originally selected, 123 sows were pregnant and used in this trial. Serum folates, Zn, Cu and Fe were measured at weaning, mating and 30 d of gestation. Serum Cu, Zn and folates increased between weaning and mating, and then decreased to 30 d of gestation. Supplementing the commercial diet with folic acid elevated serum folates between weaning and d 30 of gestation (P less than .001). Folic acid supplementation also was associated with a higher level of serum Zn at 30 d of gestation. Supplemental folic acid had no effect on the pattern of serum Cu and Fe throughout the experiment.(ABSTRACT TRUNCATED AT 250 WORDS)

At weaning, 162 sows were assigned randomly to six treatments (27 in each treatment) according to a 2 X 3 factorial arrangement: two levels of supplementary folic acid (0 and 5 mg/kg of diet) and three treatments to stimulate ovulation (none, flushing and pregnant mare serum gonadotropin [PMSG] injection). All sows were mated twice within 7 d after weaning. Of the 162 animals originally selected, 123 sows were pregnant and used in this trial. The flushing treatment consisted of allowing sows ad libitum access to feed from the day after weaning through the 1st day of behavioral estrus, whereas control animals received 2.4 kg of feed daily. The hormonal treatment consisted of one i.m. injection of 1,250 IU of PMSG the day after weaning. The commercial-type diet used as the control was computed to contain .6 mg folates per kilogram. Folic acid supplementation elevated (P less than .001) serum folates between weaning and 30 d of gestation. Fetuses of sows fed the diet supplemented with folic acid had a higher (P less than .05) total protein concentration than fetuses of control sows, whereas RNA and DNA concentrations and protein:DNA ratio were not affected. The PMSG treatment elevated (P less than .05) ovulation rate, whereas the flushing or folic acid treatments had no effect on this trait. The addition of 5 mg/kg folic acid to the commercial-type diet improved (P less than .05) the survival rate of fetuses during early gestation and tended (P = .096) to increase the number of fetuses presumably living at 30 d of gestation when this treatment was associated with high ovulation rate.(ABSTRACT TRUNCATED AT 250 WORDS)

The study was conducted to determine the response of sows to oxidized and reduced forms of supplemental folic acid in the diet. Gilts were mated and fed a standard corn-soybean meal diet with no supplemental folic acid. On d 105 of gestation, gilts were randomly assigned to one of four dietary treatments for the remainder of the study. Treatments were: 1) diet with no supplemental folate (control), 2) diet with 2.1 ppm (calculated) of added folate supplied by a synthetic pteroylmonoglutamate form (MG), 3) diet with 2.1 ppm (calculated) of added folate supplied by N5-formyl-5,6,7,8-tetrahydrofolic acid (THFA), or 4) a commercial bacterial cell powder source (Aj-PG) rich in reduced folates. Blood samples for high-performance liquid chromotography determination of reduced plasma folates were collected from gilts on d 105 of gestation, at weaning, at mating, and when the females were slaughtered on d 45 after mating for the second parity. There were 19, 18, 18, and 22 sows for the control, MG, THFA, and Aj-PG treatments, respectively. Supplementing folacin just before farrowing and during lactation had no effect on sow and litter performance during parity 1 (P > 0.10). Live fetuses at d 45 of gestation in Parity 2 were 10.06, 12.23, 10.87, and 11.07 for the control, MG, THFA, and Aj-PG treatments, respectively, and did not differ (P > 0.10). Fetal survival and placental size and protein content were generally unaffected by folate treatment. Concentration of reduced folates in sow plasma was 13.50, 13.58, 22.50, and 17.79 nM at weaning and 12.55, 19.29, 18.96, and 21.88 nM at mating for the control, MG, THFA, and Aj-PG treatments, respectively, with the THFA treatment elevated above the controls at weaning (P < 0.05) and the Aj-PG treatment greater than controls at mating (P < 0.05). At weaning, the reduced sources of supplemental folate (THFA and Aj-PG) were more effective in elevating plasma reduced folates than the oxidized folate supplement (MG; P < 0.05). Nonetheless, folate supplementation did not significantly improve sow reproductive performance in the subsequent parity, and there was no indication that reduced folate sources were superior to the oxidized pteroylmonoglutamate form as folate supplements for sows.

Sows at their second parity were randomly distributed in five groups of seven animals each to determine the dietary concentration of folic acid that optimizes the metabolic utilization of the vitamin during gestation. The groups differed by dietary supplement of folic acid: 0, 5, 10, 15, or 20 ppm. Sows were fed 2.5 kg of diet each day. The response of serum folates and folate binding capacity to treatments and the excretion of urinary folates after an i.v. injection of folic acid were measured. The total daily excretion of urinary folates was corrected according to the response to one i.v. injection of saline on the day preceding the i.v. injection of folic acid. The decrease of total serum folates throughout gestation was less pronounced in the groups fed 15 and 20 ppm of dietary folic acid (supplement x period interaction, P<.06) than it was in the other three treatments. The proportion of i.v. folic acid not recovered in sow urine (injected - excreted) decreased as the amount of dietary folic acid increased to reach a minimum, which differed according to the period (supplement x period interaction, P<.02); it was 15 ppm during wk 1 of gestation and 10 ppm for the other periods studied. The unrecovered folates increased over a dietary concentration of 15 ppm. These minimum values correspond to the most appropriate feed concentration that covered the whole body utilization (tissue and cell metabolism, catabolism, and storage) of folates by the sows and could be interpreted as a reliable index of the requirement.

Folic acid and reproductive performances of sows
Two-hundred nine sows were used in a 2 X 2 split-plot unbalanced design to measure the effect of folic acid against control, and flushing against a normal level of feeding, between weaning and mating on the following variables: serum folates at weaning and at 60 d of gestation, blood hemoglobin (Hb) and hematocrit (Ht) after 15 wk of gestation and reproductive performance at farrowing. Folic acid was administered im according to a schedule that maintained serum folates at approximately the same level between weaning and 60 d of gestation. The treatments had no effect on Hb or Ht after 15 wk of gestation. Average live litter size was 12.0 piglets/litter for sows receiving the folic acid and flushing treatments as compared with 10.5 for sows without any treatment; the main effect of folic acid was significant (P less than or equal to .04). Intralitter variation in birth weight and total litter weight tended to be increased by folic acid administration. Results showed that the administration of folic acid during gestation could appreciably improve the reproductive performance of sows.

A cooperative experiment to evaluate biotin addition to sow diets was conducted at three research stations using 303 litters. Primiparous and multiparous sows (overall average parity 2.8) were fed a 14% CP corn-soybean meal diet (140 micrograms/kg biotin), with or without supplemental biotin (330 micrograms added biotin per kg feed), throughout gestation and lactation. As many sows as possible were fed their respective diets through three successive parities. During gestation, sows were given from 1.82 to 2.27 kg of feed per day, depending on environmental conditions; during lactation sows had ad libitum access to feed. Supplemental biotin had no effect (P greater than .35) on sow weights at breeding, at d 109 of gestation, at farrowing or at weaning. No differences were found in litter size at birth (P greater than .18), but at d 21 of lactation, sows fed the diet containing supplemental biotin had larger litters than sows fed the unsupplemented diet (9.4 vs 8.7 pigs, respectively; P = .01). Pig weights at birth and d 21 of lactation were not affected (P greater than .20) by dietary treatment. Biotin supplementation did not affect (P greater than .28) the length of the interval from weaning to estrus. No evidence was found that feet cracks or bruises were reduced by biotin supplementation. The results indicate that biotin supplementation of a corn-soybean meal diet during gestation and lactation increased the number of pigs at d 21 of lactation, but it did not decrease the incidence of foot lesions.

Serum folates during the reproductive cycle of sows
In a first trial, serum folates concentration was measured in 105 sows randomly distributed into seven groups of 15 animals each. Each group represented one time period of the reproductive cycle: weaning, mating, 15, 30, 60, 90 and 110 d of gestation. The average serum folates concentrations for the different groups of sows were, expressed as ng/ml +/- SE, 99.5 +/- 8.9, 68.5 +/- 9.6, 63.9 +/- 7.4, 59.5 +/- 5.7, 39.6 +/- 5.7, 45.3 +/- 5.8 and 49.1 +/- 6.3, respectively. Results showed a biphasic decrease in serum folates levels, first at mating and then at 60 d of gestation. These results indicate a possible deficiency in serum folates during mid-gestation. In a second trial, 20 sows were assigned randomly to four groups of five animals each, representing four time periods of the reproductive cycle: weaning, mating, 30 and 60 d of gestation. One intramuscular injection of 15 mg folic acid was administered to all sows and serum folates concentration was measured before the injection and at 2-d intervals during a period of 14 d after the injection. An increase in serum folates was noted in sows of all groups. The time response curves observed after the injection are presented and discussed. The decrease in folates levels observed during early and mid-gestation could be moderated by im administration of this vitamin.

In three experiments, the influence of insulin administered after weaning was examined in primiparous sows given extra feed or in primiparous compared to multiparous sows. In Exp. 1, 171 primiparous and 231 multiparous crossbred sows on a commercial farm were injected with 0.4 IU/kg BW insulin (Eli Lilly Lente Iletin II) or saline for 4 d beginning the day after weaning (d 0) and were fed 2.3 kg/d until mating. In Exp. 2, 153 primiparous sows from the same farm as those in Exp. 1 were injected with insulin or saline as in Exp. 1 and were fed 2.7 or 3.6 kg/d until mating. In Exp. 3, 63 primiparous crossbred sows were injected with insulin or saline as described above and fed either 2.3 or 4.5 kg/d for 5 d after weaning and were remated. On the commercial farm (Exp. 1 and 2), insulin administration increased percentage in estrus for primiparous sows compared to multiparous sows (treatment x parity interaction, P < 0.02) but tended to lower litter size in primiparous sows (treatment x parity interaction, P < 0.06). In Exp. 2, insulin combined with extra feed increased (P < 0.05) litter size by two pigs but tended (P < 0.07) to decrease farrowing rate in that group (treatment x feed interaction). Weaning-to-estrus interval, pregnancy rate, ovulation rate, and embryo survival were not influenced by treatment or feeding level (Exp. 3); however, postweaning intake and embryo survival were negatively related for saline-treated sows only (r = -0.55; P < 0.01), and backfat depth at weaning and embryo survival were positively related for insulin-treated sows only (r = 0.44; P < 0.05). Overall, insulin administration differentially influenced reproduction in primiparous sows and may have interacted with metabolic or nutritional state of the animal.

Previous studies showed that supplementation of sows' diets with L-carnitine increases body weights of their piglets at birth. This study was performed to investigate whether piglets of sows treated with L-carnitine differ in their growth potential from that of piglets of untreated control sows after weaning. It was also investigated whether supplementation of piglets' diets with L-carnitine improves their growth after weaning. In two trials, piglets of the first litters of primiparous sows (trial 1) and the second litters of the same sows (trial 2) were divided into four groups: group 1, piglets of control sows, fed a control diet; group 2, piglets of control sows fed a diet supplemented with 30 mg L-carnitine/kg; group 3, piglets of L-carnitine-treated sows, fed a control diet; group 4, piglets of L-carnitine-treated sows fed a diet supplemented with 30 mg L-carnitine/kg. Mean initial body weights of the piglets of the four groups were identical. They were 8.5 kg in trial 1 and 12.5 kg in trial 2. Diets were fed ad libitum over a period of 35 days. Piglets from sows treated with L-carnitine did not differ in body weight gains, feed intake and gain : feed ratio from those of control sows. In trial 1, piglets supplemented with L-carnitine had higher body weight gains (p < 0.005) and showed a tendency towards a higher gain : feed ratio (p = 0.09) than piglets fed the control diets. In trial 2, no significant difference in these parameters emerged between piglets fed the diet supplemented with L-carnitine and those fed the control diet. In conclusion, this study shows that dietary L-carnitine treatment of sows does not improve the growth potential of their piglets after weaning under the conditions of equal initial body weights. The study also shows that L-carnitine supplementation of their diets improves the growth performance in light piglets of primiparous sows.

In order to determine the effect of folic acid on serum and milk folates in lactating sows as well as on serum folates and growth rate of the piglets, sows (n = 25) received either saline or 15 mg folic acid i.m. each week from d 2 after parturition to weaning, 26 d later. Blood samples were drawn from all sows at 110 d of gestation and every week during lactation. Milk samples were taken at d 7 and 21 of lactation. Piglets were weighed and blood samples were collected weekly during lactation. Serum folates of sows increased during lactation. The rate of increase was more pronounced (P less than .0002) after folic acid injections. Milk folates concentrations decreased (P less than .0007) from d 7 to 21 of lactation but were higher (P less than .0001) in treated sows (11.8 +/- .7 ng/ml) than in control sows (7.9 +/- .4 ng/ml). Serum folates of piglets in control litters increased from 55.0 +/- 2.2 ng/ml at 2 d of age to a peak value of 86.3 +/- 3.1 ng/ml 2 wk later, and then gradually decreased. In piglets from treated dams, the time response curve was similar to that of the controls, but values were about 15% higher (P less than .01). The growth rate of piglets until 8 wk of age was not changed (P greater than .47) by folic acid injection of sows. More studies are needed to evaluate the practical importance of changes in folates status in establishing the folic acid requirements of lactating sows.

Two experiments were conducted to evaluate whether administration of recombinant porcine somatotropin (pST) to sows (Hampshire-Yorkshire) enhanced lactational performance. In Exp. 1, sows (n = 84) were fed a corn-soybean meal diet (17.8% CP), or a similar diet with 8% added fat, from d 108 of gestation to d 28 of lactation. Half of the sows fed each diet were injected with 6 mg/d of pST from d 108 of gestation to d 24 of lactation. Diets were fed at 2.27 kg/d from d 108 of gestation until farrowing and then were self-fed during lactation. By d 3 of lactation, litter size was standardized at 8 to 10 pigs per litter. Treating sows with pST resulted in a 10-fold increase (P less than .001) in serum somatotropin at 4 h postinjection. Serum glucose was increased (P less than .01) and serum triglycerides, creatinine, and urea N were decreased (P less than .01) by pST. During the summer, apparent heat stress occurred in pST-treated sows, resulting in 14 deaths. Most (10) of the deaths occurred just before, during, or shortly after farrowing. Fewer (P less than .08) deaths occurred when pST-treated sows were fed the diet with added fat. Sows treated with pST consumed less feed (P less than .10) and lost more backfat (P less than .10) during lactation than controls. Increasing the dietary fat did not prevent these changes. Weaning weights of pigs and milk yield of sows (estimated by deuterium oxide dilution) were not affected by pST treatment. In Exp. 2, sows (n = 42) were injected weekly with 0 or 70 mg of pST on d 3, 10, 17, and 24 of lactation. Litters were standardized by d 3 at 8 to 10 pigs, and sows were fed the same control (low fat) diet as in Exp. 1. Sows treated with pST consumed less feed and lost more weight and backfat during lactation than untreated sows. Litter size, average pig weaning weights, and milk yield were not influenced by pST treatment. These data indicate that a 6-mg daily injection of pST from 6 d prepartum to d 24 of lactation or a 70-mg weekly injection of pST from 3 d postpartum to d 24 of lactation does not increase milk production in lactating sows.

The effect of L-carnitine supplementation during pregnancy and lactation on performance parameters of sows was studied. The trial comprised a total of 127 sows (40 gilts, 87 mature sows) which were divided into a control and a treatment group. All animals were fed individually and received basic feed mixtures for pregnancy and lactation with low carnitine concentrations (gestation diet: 4.7 mg/kg feed, lactation diet: 12.5 mg/kg feed). The rations of the sows in the treated group were supplemented with 125 mg L -carnitine per head and day during pregnancy and 250 mg L -carnitine per head and day during lactation. The animals of the control group received identical feed mixtures in identical amounts, but without the L -carnitine supplement. L -carnitine supplementation resulted in higher sow liveweight gains between day 1 and day 85 of pregnancy. The number of piglets per litter and the number born alive did not differ between the control sows and those treated with L -carnitine. However, the L -carnitine-supplemented sows produced only half as many non-viable piglets as the control animals. Moreover, litter weight and mean birth weight of piglets from L -carnitine-treated sows were higher than in the control sows. This effect was more marked in gilts (+8% higher litter weight, +9% higher piglet weight) than in sows (+7% and +6%, respectively). Piglets from sows whose ration was supplemented with L -carnitine showed higher liveweight gains during the suckling period (+12% for gilts, +4% for sows), which is why litter weights post weaning were also higher among the sows treated with L -carnitine than in the control sows (+14% for gilts, +10% for sows). Overall, the study shows that dietary supplementation with L -carnitine during pregnancy and lactation improves the reproductive performance of sows.

The present work aimed to determine if different levels of prolificacy either by parity or by genetic origin are linked to folate metabolism. Nulliparous Yorkshire-Landrace (YL) and multiparous YL, and multiparous Meishan-Landrace (ML) sows were randomly assigned to two treatments: 0 ppm or 15 ppm folic acid+0.6% glycine. Supplements were given from the estrus before mating until slaughter on d 25 of gestation. At slaughter, embryo and endometrial tissues were collected to determine concentrations of DNA, protein, and homocysteine. Allantoic fluid samples were also collected to determine concentrations of folates, vitamin B12 and amino acids. Blood samples were taken at first estrus, at mating, and on d 8, 16, and 25 of gestation to determine serum concentrations of folates, vitamin B12, and relative total folate binding capacity (TFBC). Over the entire experiment, multiparous YL sows had higher average serum concentrations of folates than nulliparous YL sows (P < 0.05) but had similar serum concentrations of relative TFBC. Concentrations of folates and relative TFBC averaged higher in ML measured over the entire experiment than in multiparous YL sows (P < 0.05). Concentrations of serum vitamin B12 were higher in multiparous YL than in ML sows or YL nulliparous sows (P < 0.05) over the entire experiment. In allantoic fluid, folates, vitamin B12, and essential amino acids contents were significantly lower in ML than in YL multiparous sows (P < 0.05). The folic acid+glycine supplement increased concentrations of serum folates, but the increase was more marked in nulliparous YL sows (nulliparous x folic acid+glycine, P < 0.05). The folic acid+glycine supplement had no effect on litter size and embryo survival, but it tended to increase embryo DNA in multiparous YL sows (P = 0.06) but not in ML and nulliparous YL sows. Homocysteine was decreased by folic acid+glycine supplement in embryos from all sows, but in endometrium, the folic acid+glycine effect was dependent on parity (nulliparous x folic acid+glycine, P < 0.05). The effects of folic acid+glycine on litter size and embryo development and survival and some aspects of folate metabolism suggest that the basal dietary content of folic acid+glycine was adequate for ML and nulliparous YL sows but not to optimize embryo development in YL multiparous sows.

The aim of this study was to determine the effect of dietary Forsythia suspensa extract (FSE) supplementation to lactating sows and nursery pigs on post-weaning performance, antioxidant capacity, immunoglobulins, and intestinal health. Based on backfat, body weight (BW), and parity, 24 gestating sows (Landrace × Yorkshire) with average parity of 3.38 ± 0.61 and BW of 234 ± 6.81 kg were allotted into two dietary treatments (control vs. 100 mg/kg FSE) with 12 sows per treatment from day 107 of gestation to day 21 of lactation. After weaning, based on the initial BW and source litter, 192 nursery pigs (Duroc × [Landrace × Yorkshire], average BW of 6.98 ± 0.32 kg, weaned at day 21) were allotted into four dietary treatments with eight replicate pens per treatment, six pigs per pen for a 4-wk study. The treatments included the following: 1) CC (sows and their piglets both fed control diet); 2) CF (sows fed control diet and their piglets fed FSE diet [containing 100 mg/kg FSE]); 3) FC (sows fed FSE diet and their piglets fed control diet); and 4) FF (sows and their piglets both fed FSE diet). The MIXED procedures of SAS for a split-plot arrangement with sow diet as the whole plot and nursery diet as split plot were used to analyze the data. After weaning, piglets from FSE-fed sows had improved (P < 0.05) average daily gain and feed efficiency, and lower (P < 0.05) diarrhea rate in overall (day 1 to 28) compared with those from sows fed control diet. Piglets from FSE-fed sows also had higher (P < 0.05) contents of immunoglobulin G (IgG), growth hormone, superoxide dismutase (SOD), total antioxidant capacity in serum, villus height in ileum, and villus height to crypt depth ratio in jejunum, as well as lower (P < 0.05) content of malondialdehyde (MDA) in serum and crypt depth in ileum compared with those from sows fed control diet. Piglets fed FSE during nursery had increased (P < 0.05) concentrations of IgG, SOD, and catalase, and decreased (P < 0.05) MDA and tumor nuclear factor-α levels in serum compared with those fed control diet during nursery. Piglets from FC group had increased (P < 0.05) protein expression of occludin in jejunal mucosa and relative abundance of Lactobacillus on genus level in colon compared with those from CC group. In conclusion, for the performance and intestinal health, diets supplemented with FSE during lactation phase seemed more efficient to alleviate weaning stress than the nursery phase. In terms of the antioxidant status and immunoglobulins, FSE supplemented in both phases were efficient for nursery pigs.

One hundred fifty-three gilts were maintained in three breeding groups and fed gestation-lactation diets supplemented with either 0 (control), 1.65 or 6.62 mg of supplemental folic acid/kg of diet for two consecutive parities. Serum folate concentrations of sows were linearly (P less than .05) increased by dietary additions of folic acid during both gestation and lactation, but serum glucose and urea concentrations were unaffected by treatment. Serum folate concentrations decreased from breeding to d 60 and 90 of gestation and then increased through lactation for all treatments. Number of pigs born and live pigs at birth, d 14 and d 21 were quadratically (P less than .05) increased by folic acid additions. Average pig weights were similar among treatments (P greater than .10) on both d 0 and 14 of lactation but were less (P less than .01) than the other treatment groups on d 21 for pigs from sows fed the 1.65 mg/kg treatment. Litter weights were quadratically (P less than .01) increased on d 0 and d 14 by folic acid supplementation. Sow weight gain and backfat thickness loss were unaffected by treatment during gestation (P greater than .06); sow weight loss and backfat thickness loss increased quadratically with increasing level of folic acid during lactation (P less than .06 and .05, respectively). More control sows exhibited estrus by d 7 postweaning than sows receiving folic acid supplementation in parity I (P less than .05); however, no differences (P greater than .10) were detected among treatments by d 14, nor were any differences observed by d 7 in parity II. Conception rate was unaffected by folic acid additions. Dietary folic acid supplementation improved sow reproductive performance by increasing the number of pigs born alive.

An experiment comprising 19 German Landrace sows was established to evaluate the effect of folic acid supplements (10 mg/kg concentrate) on homocysteine and folic acid concentration in plasma and serum, respectively, of highly pregnant sows as compared to an unsupplemented control (basal diet contained 0.62 mg folic acid/kg concentrate). Blood samples were taken between day 75 and 110 of gestation for homocysteine analysis and on day 100 of gestation for folic acid determination. Due to the folic acid supplements serum folic acid concentration increased significantly (104 nmol/l in controls and 140 nmol/l in supplemented sows). In contrast, homocysteine concentration in the plasma was not significantly influenced by folic acid supplements (16.6 mumol/l in controls and 15.2 mumol/l in supplemented sows). Further investigations seem to be necessary to clarify the physiology of homocysteine metabolism in swine.
