Mating behaviour and vegetative compatibility were studied in Spanish populations of Botryotinia fuckeliana. Fifty-seven isolates out of the 61 tested were sexually fertile with one or more of the reference strains of known mating type (MAT1-1 or MAT1-2). Thirty-nine isolates were heterothallic, giving fertile crosses when mated with the MAT1-1 (24 isolates) or the MAT1-2 (15 isolates) reference strain. Eighteen isolates crossed successfully with both MAT1-1 and MAT1-2 reference strains, and were referred to as homothallic or MAT1-1/2. Both mating types were widespread, being represented in isolates from two regions, from the same and different greenhouses, from different hosts, and from different years of isolation. Isolates were paired on Malt Extract Agar + NaCl to evaluate vegetative compatibility. Most of the paired isolates were unable to fuse and showed a different reaction of incompatibility. Nitrate-non-utilising (nit) mutants were selected by growth on a medium amended with 30–50gl–1 potassium chlorate. Over 600 chlorate-resistant sectors were recovered from 40 isolates at a mean frequency of 0.15–2.39 sectors per colony, but only 11% were identified as nit mutants by their thin growth with no aerial mycelium on minimal medium. However, most of these nit mutants reverted to wild type during six months of storage on chlorate-amended medium. Genetic complementation between nit mutants occurred only in two cases between mutants from the same isolate.  相似文献

Cyprodinil is a representative of the new class of broad-spectrum anilinopyrimidine fungicides. The effect of cyprodinil on mycelial growth of Botryotinia fuckeliana on solid agar medium depends on the composition of the medium and on the age of the mycelium to be used for the bioassay. An in-vitro method was developed to study the sensitivity distribution to cyprodinil in two wild-type populations of B. fuckeliana from fruits of strawberry with grey mould. To validate the applicability of the method, sensitivities to cyprodinil, mepanipyrim and pyrimethanil were monitored in populations of B. fuckeliana from grapes with grey mould from different vineyards, including one trial vineyard where reduced performance of cyprodinil had been encountered. The monitoring procedure was based on the inhibition of the mycelial growth on a synthetic medium containing L -asparagine (asp-agar) amended with the active ingredients. The mycelium was grown on asp-agar discs, starting from a spore suspension, for 17 h prior to the beginning of the test. This procedure proved to be efficient. The two wild-type populations from different sampling sites showed similar distributions of the sensitivity to cyprodinil. Some isolates from the trial site with reduced performance of anilinopyrimidines showed reduced sensitivities to cyprodinil, mepanipyrim and pyrimethanil, demonstrating cross-resistance between these anilinopyrimidines.  相似文献

为了明确设施蔬菜灰霉病菌Botryotinia fuckeliana的抗药性现状,采用菌丝生长速率法检测了20052006年采自浙江、江苏、山东和辽宁4省的144个菌株对6种常用防治药剂的敏感性。结果表明,灰霉病菌对百菌清已经产生了低水平的抗性,频率为5.56%,其对多菌灵的抗性非常严重,总的抗性频率为43.05%,高抗(HR)频率为27.08%;乙霉威的EC50值在0.137728.9μg/mL之间,平均为40.06μg/mL。其中多菌灵-乙霉威双抗频率为36.11%,且首次检测到了两种新的双抗类型。二甲酰亚胺类杀菌剂在生产上已经应用近20年,但灰霉病菌对异菌脲和腐霉利只有频率为20%左右的低水平抗性,没有检测到高抗菌株;苯胺嘧啶类杀菌剂嘧霉胺虽然只应用几年时间,但已经产生了抗性,其抗性菌株频率为4.16%。研究表明,设施蔬菜灰霉病菌对常用的6种防治药剂均产生了不同程度的抗性。  相似文献

Nitrate non-utilising (Nit) mutants of six field strains and three single-ascospore strains of Botryotinia fuckeliana were selected by plating mycelial plugs onto a nitrate-containing minimal medium amended with chlorate. Mutants were characterised by growth responses on minimal medium amended with various sole nitrogen sources. For each parental strain two mutants were produced: nit1, defective in nitrate reductase apoenzyme; and NitM, defective in the molybdenum cofactor pathway. Complementation on nitrate minimal medium was observed between nit1 and NitM mutants from the same parent in all instances. However, complementation was not observed between any such combinations when the mutants were derived from different parents. It is concluded that a vegetative incompatibility system operates in B. fuckeliana resulting in multiple vegetative compatibility groups. One of the single-ascospore strains was derived from a cross between two of the field strains, indicating that new compatibility groups can be generated during sexual reproduction. Mycelial interaction zones were formed between all parental strains when they were paired on NaCl-amended medium, indicating congruence between mycelial incompatibility and vegetative incompatibility.  相似文献

Fenhexamid, a recently developed botryticide, is shown here to inhibit sterol biosynthesis. When the fungus Botryotinia fuckeliana was grown in the presence of fenhexamid, the ergosterol content was reduced, and three 3‐keto compounds, 4α‐methylfecosterone, fecosterone and episterone, accumulated, suggesting an inhibition of the 3‐keto reductase involved in C‐4 demethylation. Thus, fenhexamid belongs to a new, promising class of sterol biosynthesis inhibitors not previously used in agriculture or in medicine. © 2001 Society of Chemical Industry  相似文献

BACKGROUND: QoI fungicides, inhibitors of mitochondrial respiration, are considered to be at high risk of resistance development. In several phytopathogenic fungi, resistance is caused by mutations (most frequently G143A) in the mitochondrial cytochrome b (cytb) gene. The genetic and molecular basis of QoI resistance were investigated in laboratory and field mutants of Botryotinia fuckeliana (de Bary) Whetz. exhibiting in vitro reduced sensitivity to trifloxystrobin. RESULTS: B. fuckeliana mutants highly resistant to trifloxystrobin were obtained in the laboratory by spontaneous mutations in wild‐type strains, or from naturally infected plants on a medium amended with 1–3 mg L?1 trifloxystrobin and 2 mM salicylhydroxamic acid, an inhibitor of alternative oxidase. No point mutations were detected, either in the complete nucleotide sequences of the cytb gene or in those of the aox and Rieske protein genes of laboratory mutants, whereas all field mutants carried the G143A mutation in the mitochondrial cytb gene. QoI resistance was always maternally inherited in ascospore progeny of sexual crosses of field mutants with sensitive reference strains. CONCLUSIONS: The G143A mutation in cytb gene is confirmed to be responsible for field resistance to QoIs in B. fuckeliana. Maternal inheritance of resistance to QoIs in progeny of sexual crosses confirmed that it is caused by extranuclear genetic determinants. In laboratory mutants the heteroplasmic state of mutated mitochondria could likely hamper the G143A detection, otherwise other gene(s) underlying different mechanisms of resistance could be involved. ABSTRACT DNA sequence polymorphisms in the putative two-component histidine protein kinase encoded by the Daf1 gene have been identified within a sample of 5 sensitive and 27 dicarboximide-resistant field strains of Botryotinia fuckeliana (anamorph Botrytis cinerea). The gene of 3948 bp is predicted to encode a 1315-amino acid protein comprising an N-terminal region, an amino acid repeat region, which has been hypothesized to be the binding site for dicarboximide fungicide, and a C-terminal region encompassing kinase and response regulator domains. Two amino acid variants were distinguished among the sensitive strains characterized by alanine (group 1), or threonine (group 2), at position 1259 in the C-terminal region. All resistant strains could be classified into either group 1 or group 2 but, in addition, all showed changes in the second amino acid repeat region. On the basis of the differences in this repeat region, four classes of resistant strains were recognized; class 1 characterized by an isoleucine to serine mutation, class 2 by an isoleucine to asparagine mutation, class 3 by an isoleucine to arginine mutation (all at position 365), and class 4 by an isoleucine to serine mutation (position 365) as well as a glutamine to proline mutation (position 369). All classes showed similar low levels of resistance to iprodione and to vinclozolin, except for class 3 and class 4 strains, which show low resistance to iprodione but moderate (class 3) or high (class 4) resistance to vinclozolin. The classes as a group did not differ from sensitive strains in osmotic sensitivity measured as mycelial growth response, but some class 1 strains showed an abnormal morphology on osmotically amended medium. The evolution of the amino acid differences is discussed in relation to field observations. It is proposed that class 1 and class 2 strains arose by single mutations within the sensitive population, whereas classes 3 and 4 arose by single mutations within a resistant population.  相似文献

‘Aliette’, a formulation of the fungicide fosetyl-aluminium, is moderately active in vitro against the ascomycete Botryotinia fuckeliana (Botrytis cinerea). In contrast, fosetyl-aluminium and related chemicals, such as phosphonic acid and sodium phosphonate, have low efficacy, although it is enhanced by media with low phosphate content. This indicates that the effectiveness of ‘Aliette’ against B. fuckeliana is not due to fosetyl-aluminium but to adjuvants included in the formulation. The genetic basis of differences in sensitivity to ‘Aliette’ was investigated in field and laboratory isolates of B. fuckeliana. Genetic analysis showed that reduced sensitivity to ‘Aliette’ always co-segregated with resistance to benzimidazole fungicides in meiotic progeny of sexual crosses. This indicates that differences in sensitivity to ‘Aliette’ in B. fuckeliana are due to the major gene Mbcl of resistance to benzimidazole fungicides and active adjuvant(s) present in ‘Aliette’ are linked to benzimidazoles by cross-resistance.  相似文献

An interactive relationship between vitiviruses and grapevine leafroll viruses was characterized in grapevine. Grapevine viruses A and B (GVA and GVB) were found more frequently in the presence of co-infecting Grapevine leafroll associated viruses (GLRaV-1, ?2 or ?3) than in their absence. The titers of the vitiviruses in co-infection with leafroll viruses were found to be higher than were their titers in the absence of leafroll virus infection. The occurrence of vitivirus-associated stem-pitting symptoms was correlated with leafroll virus co-infection. Specific pairing associations on the species level were found between different viti- and leafroll virus species: GVB was associated preferentially with GLRaV-2; GVA was associated preferentially with GLRaV-1 and GLRaV-3. In contrast to the increase in vitivirus titer seen with leafroll virus co-infection, the incidence and titer of grapevine leafroll virus appeared to be unaltered by vitivirus co-infection. The potential for a synergistic enhancement of grapevine disease in co-infected vines is discussed.  相似文献

Non-aggressive mutants were isolated from a single-ascospore strain ofBotryotinia fuckelianafollowing ultraviolet and chemical mutagenesis of conidia and screening on French bean leaves (Phaseolus vulgaris). Crosses with reference strain SAS56 revealed one ultraviolet induced mutant (Mp97) in which the non-aggressive phenotype segregated 1:1 indicating control by a single gene with a major effect on pathogenicity. Further crosses showed that this gene, designatedPat1, is not linked toMbc1(benzimidazole resistance),Daf1(dicarboximide resistance),nit1(nitrate non-utilising), orSel1(sodium selenate resistance). The pathogenicity ofPat1strains was compared with various wild-type strains. Mutant strains produced small, restricted lesions on leaves of French bean and soybean (Glycine max) and slowly spreading lesions on rose (Rosaspp.) flowers. The mutant was essentially non-pathogenic on tomato stems at 20 and 25 °C but produced a few invasive infections at 10 and 15 °C. Tests of radial growth rate, sporulation and sclerotial production showed thatPat1strains exhibit no gross unfitness or unusual nutrient requirement. Althoughin vivostudies showed substantially less total polygalacturonase activity inPat1lesions compared to those produced by wild-type strains, differences in polygalacturonase isozyme profiles were not correlated with the presence ofPat1.Pat1strains showed relatively low acid production in pH indicator medium. Microscopic examination showed that lesions induced by the Mp97 mutant on French and soybean leaves (4 days old), but not by its parent, were surrounded by a distinct staining zone of mesophyll cells suggesting a difference in host response.
