Fresh aerial tissue and roots of 14 wild plants in Okinawa prefecture were investigated for their antimicrobial activity against Ralstonia solanacearum , which causes bacterial wilt of potato. A 70% aqueous ethanol extract of fresh aerial tissue of Geranium carolinianum L. showed strong antimicrobial activity against R. solanacearum . This extract also showed antimicrobial activity against the pathogens causing common scab of potato and soil rot of sweet potato. The antimicrobial substance could be extracted with hot water, and was effective against R. solanacearum in soil. In the field test, a treatment combining incorporation of dried aerial tissue into the soil and solarization was highly effective for control of bacterial wilt of potato. These findings suggest that G. carolinianum L. could be used as a biological agent for the control of bacterial wilt of potato.  相似文献

We isolated an antimicrobial compound from the aerial tissue of Geranium carolinianum and identified it as ethyl 3, 4, 5-trihydroxy benzoate (ethyl gallate) by 1H-NMR and 13C-NMR and gas chromatography–mass spectrometry. The antimicrobial activity of ethyl gallate against three potato pathogens was assayed by the paper disk method. The activity against Ralstonia solanacerum , Streptomyces scabies , and Streptomyces acidiscabies was observed at concentrations >200, >300, and >300 µg disk−1, respectively. These results suggest that the antimicrobial activity of Geranium carolinianum against soil-borne plant disease pathogens is partly related to ethyl gallate.  相似文献

Host–parasite relationships and pathogenicity of Meloidogyne javanica on potatoes (newly recorded from Malta) were studied under glasshouse and natural conditions. Potato cvs Cara and Spunta showed a typical susceptible reaction to M. javanica under natural and artificial infections, respectively. In potato tubers, M. javanica induced feeding sites that consisted of three to four hypertrophied giant cells per adult female. Infection of feeder roots by the nematode resulted in mature large galls which usually contained at least one mature female and egg mass. In both tubers and roots, feeding sites were characterized by giant cells containing granular cytoplasm and many hypertrophied nuclei. Cytoplasm in giant cells was aggregated alongside the thickened cell walls. Stelar tissues within galls appeared disorganized. The relationship between initial nematode population density ( P ) [0–64 eggs + second-stage juveniles (J2s) per cm3 soil] and growth of cv. Spunta potato seedlings was tested under glasshouse conditions. A Seinhorst model [ y = m  + (1 −  m ) z ( P − T )] was fitted to fresh shoot weight and shoot height data of nematode-inoculated and control plants. Tolerance limits ( T ) for fresh shoot weight and shoot height of cv. Spunta plants infected with M. javanica were 0·50 and 0·64 eggs + J2s per cm3 soil, respectively. The m parameter in that model (i.e. the minimum possible y -values) for fresh shoot weight and shoot height were 0·60 and 0·20, respectively, at P  = 64 eggs + J2s per cm3 soil. Root galling was proportional to the initial nematode population density. Maximum nematode reproduction rate was 51·2 at a moderate initial population density ( P  = 4 eggs + J2s per cm3 soil).  相似文献

Benzothiadiazole (BTH), as Bion WG50, and acetylsalicylic acid (ASA) treatments of potato foliage of field- and glasshouse-grown potato plants were compared for control of two foliar diseases, early blight ( Alternaria solani ) and powdery mildew ( Erysiphe cichoracearum ). The effect of these treatments on harvested tubers wound-inoculated with the dry rot fungus ( Fusarium semitectum ) was also evaluated. BTH (50 mg a.i. L−1) gave almost complete control of both foliar pathogens on inoculated glasshouse-grown plants and reduced the severity of leaf spotting diseases (mainly early blight) in the field. BTH (100 mg a.i. L−1) and ASA (400 mg a.i. L−1) reduced the severity of dry rot in field-grown tubers in some post-harvest wound-inoculated treatments but not others and a similar reduction occurred with tubers inoculated post-harvest from BTH-treated plants grown under glasshouse conditions. BTH treatment increased β-1,3-glucanase activity in leaves > stem > tubers > stolons but not in roots. Increased enzyme activity was recorded for up to 45 days post-treatment.  相似文献

The relationship between initial nematode population density ( P i) in a series from 0–128 eggs and second-stage juveniles per cm3 soil and growth of spinach cv. Symphony F1 seedlings was tested under glasshouse conditions. A Seinhorst model [ y = m  + (1 −  m ) z P–T ] was fitted to fresh top- and total plant-weight data for inoculated and control plants. Tolerance limits ( T ) of spinach cv. Symphony F1 to M. incognita race 1 for fresh top and total plant weights were 0·25 and 0·5 eggs and second-stage juveniles per cm3 soil, respectively. The minimum relative values for fresh top and total plant weights were zero in both cases at P i ≥ 32 eggs and second-stage juveniles per cm3 soil. Root galling was least at low initial population densities and greatest at 16 eggs and second-stage juveniles per cm3 soil. Maximum nematode reproduction rate was 33·1-fold at the lowest P i.  相似文献

Rhamnolipids, extracellular metabolites of Pseudomonas aeruginosa with surfactant properties, proved to be very effective in controlling the spread of brown root rot disease caused by Phytophthora cryptogea in the hydroponic forcing system of witloof chicory ( Cichorium intybus var. foliosum ). The biosurfactant was applied as the product PRO1, a formulation of 25% rhamnolipids in oil. Both an in vitro screening and in vivo experiments in a mini-hydroponic system demonstrated the ability of PRO1 to control brown root rot. A 25  µ g mL−1 rhamnolipids nutrient solution was enough to obtain good control of an artificial infection with a zoospore suspension of P. cryptogea . The biosurfactant PRO1 performed well in a semicommercial system under growers' conditions. A treatment of 25  µ g mL−1 rhamnolipids (100  µ g mL−1 PRO1) reduced the disease incidence significantly in two independent experiments. However, PRO1 was not effective when a mycelial suspension was used as inoculum. Rhamnolipids have good potential to limit the spread of P. cryptogea in the hydroponic forcing system of witloof chicory, and can be used as a preventive measure against brown root rot.  相似文献

Pot experiments were conducted to evaluate the level of imazamox tolerance in five red rice ( Oryza sativa L.) and four barnyardgrass (three Echinochloa crus-galli (L.) Beauv. and one Echinochloa oryzoides (Ard.) Fritch) morphologically distinct biotypes collected from rice fields in northern Greece. The susceptibility of barnyardgrass biotypes to propanil was also studied. Red rice biotypes were not controlled by imazamox applied at 40 g ha−1. In contrast, 80 g imazamox ha−1 provided 56–84% red rice control (averaged across shoot number and fresh weight reduction). Not all barnyardgrass biotypes were susceptible to imazamox applied postemergence. However, propanil applied at 2.6 kg ha −1 controlled the E. crus-galli biotypes well, but propanil applied at rates of 2.6 and 5.2 kg ha −1 was not effective in reducing the shoot number and fresh weight of the E. oryzoides biotype. Propanil applied at 10.4 kg ha −1 reduced the shoot number and fresh weight of this biotype by 78 and 85%, respectively. In most cases, a linear equation ( y  = % of control, x  = g ha−1) provided the best fit for regressions between red rice or barnyardgrass shoot number or fresh weight and imazamox rates. The results of this study suggest that postemergence application of imazamox is not effective against all red rice and barnyardgrass biotypes found in the rice fields of Greece and that significant variability regarding herbicide efficacy among biotypes might exist.  相似文献

Bacterial black spot of corn-salad ( Valerianella locusta ) caused by Acidovorax valerianellae appeared in France in the beginning of the 1990s. The disease is now widespread in the main area of corn-salad production and is responsible for high depreciation and significant economic losses every year. A semiselective medium is described for the detection of the bacterium in various environments and to identify primary sources of inoculum. The medium TSAV (tryptic soy broth, 3 g L−1, agar 15 g L−1, 5-fluorouracyl 5 mg L−1, novobiocin 5 mg L−1, propiconazole 5 mg L−1) increased chances of obtaining cultures of A. valerianellae from naturally infested soil and from root-debris. Using TSAV, the pathogen was detected in root-debris up to 39 days after the harvest of a diseased crop. As only a few days usually separates harvest from the next sowing date, soil is likely to be an important source of inoculum for the next crop.  相似文献

An isolate of Gliocladium roseum proved highly antagonistic to Botrytis cinerea . Sporulation of B. cinerea on chickpea seed naturally infected or inoculated with B. cinerea was suppressed by seed treatment with conidial suspensions of G. roseum at 107 and 108 conidia/mL, respectively. Establishment of healthy seedlings in punnets (small trays) 5 weeks after sowing with inoculated seed was increased from 29.2% to 59.7% by treatment with G. roseum at 3×107 conidia/mL, and from 1.4% to 69.4% with G. roseum at 3×108 conidia/mL, the latter being equivalent to disease control by Thiram. There was no significant effect of Rhizobium on disease suppression by G. roseum , and treatment with G. roseum at 108 conidia/mL did not reduce nodulation. Amendment with culture filtrates of G. roseum did not affect the growth rate of B. cinerea on potato dextrose agar, indicating that constitutive production of an antibiotic is not involved in biocontrol. A selective medium was developed to enumerate propagules of G. roseum on seed recovered from soil. There was no significant change in the population of G. roseum on seed after incubation for 4 weeks in soil to which the isolate of G. roseum was indigenous.  相似文献

The take-all fungus, Gaeumannomyces graminis var. tritici , was highly sensitive to fluquinconazole ( in-vitro EC50 0·016–0·018 mg L−1), a fungicide developed for use as a seed treatment to control take-all, and to prochloraz (EC50 0·006 mg L−1). Fungi of other genera that were commonly isolated from cereal roots were sensitive in varying degrees to prochloraz but were relatively insensitive (e.g. Fusarium culmorum , EC50 > 20 mg L−1) or slightly sensitive (e.g. Epicoccum purpurascens , EC50 0·514 mg L−1) to fluquinconazole. Gaeumannomyces graminis var. graminis and G. cylindrosporus , weak parasites that can protect roots against take-all, and an unnamed Phialophora sp., all closely related to the take-all fungus, were highly or moderately sensitive to fluquinconazole. Alternaria infectoria and E. purpurascens were most consistently effective in suppressing development of take-all on pot-grown wheat plants dual-inoculated with G. graminis var. tritici and the nonpathogen. Take-all was decreased more on dual-inoculated wheat plants grown from seed treated with fluquinconazole or fluquinconazole plus prochloraz than when only an antagonistic fungus ( A. infectoria , E. purpurascens , Fusarium culmorum or Idriella bolleyi ) or a seed treatment was applied. These fungi were less effective in combination with seed treatments on barley. Gaeumannomyces graminis var. graminis and G. cylindrosporus , tested on wheat, suppressed take-all only in the absence of fungicides. It is suggested that the performance of seed treatment containing fluquinconazole against take-all may in some circumstances be enhanced by its partial specificity for the take-all fungus.  相似文献

Chemical and biological agents were evaluated for their ability to suppress root rot, caused by Phytophthora cactorum , in field-grown radiata pine seedlings in New Zealand. Trials were conducted over two seasons in an area of a forest nursery with a natural infestation of P. cactorum , and a history of root rot. In each season, symptoms of root rot developed during April, one month after root pruning, when seedlings were approximately six months old. In trial one, root rot incidence by mid July 2007 was 9·1% in untreated plots and 8·4% in plots that had been treated with metalaxyl-M/mancozeb (14 kg ha−1) at seedling emergence. Disease incidence was lowest (2·1%) in plots that received seven monthly applications of phosphorous acid (6·5 L ha−1). Other treatments, including seed coating with thiram or Trichoderma spp., and foliar applications of methyl jasmonate, did not control disease. In trial two, effects of treatment timing relative to root pruning were investigated. By late June 2008, three months after root pruning, root rot incidence was 22·2% in the untreated plots. Phosphorous acid was the most effective treatment and almost completely suppressed disease (0·1% incidence) when applied fortnightly from February until May (seven applications). Metalaxyl-M/mancozeb (15 kg ha−1) was not effective (21·4% incidence) when applied five months before root pruning. However, disease incidence was reduced when the chemical was applied one week after root pruning (14·9% incidence) and greater control was achieved (8·2% incidence) when the application rate was increased to 50 kg ha−1.  相似文献

Isolates of Phytophthora erythroseptica , causal agent of pink rot of potato, were obtained from diseased tubers collected in Prince Edward Island (PEI), Canada. In an in vitro assay of field and single-zoospore isolates, all isolates of the pathogen from PEI were sensitive to metalaxyl (EC50 < 0·5  µ g mL−1), unlike moderately or highly resistant reference isolates obtained from Maine, USA. Allozyme-banding patterns at the glucose-6-phosphate isomerase ( Gpi ) locus were identical (91/91) for all isolates of P. erythroseptica examined from PEI and Maine, but could be used to distinguish isolates of P. erythroseptica from local isolates of Phytophthora infestans . Inoculation of potato tubers from plants treated with foliar applications of metalaxyl in the field indicated that compounds inhibitory to metalaxyl-sensitive isolates of P. erythroseptica were present in the periderm, even after 4 months' storage. By contrast, moderately or highly resistant isolates of the pathogen caused significantly ( P  ≤ 0·05) more disease than sensitive isolates in tubers from plants treated with metalaxyl. The effectiveness of metalaxyl in controlling pink rot in a particular region will depend on resistance levels of local populations of P. erythroseptica . Preventing the development and spread of resistant strains of P. erythroseptica will be critical in maintaining metalaxyl as an effective chemical for control of this pathogen.  相似文献

Two bacterial isolates, Bacillus megaterium (c96) and Burkholderia cepacia (c91), demonstrated to be antagonistic against Fusarium oxysporum f.sp. radicis-lycopersici , the causal organism of fusarium crown and root rot of tomato, were evaluated as biocontrol agents alone and when integrated with the fungicide carbendazim. In an initial screening, these isolates reduced disease incidence by 75 and 88%, respectively. In vitro , both biocontrol agents were highly tolerant to the fungicide carbendazim, commonly used to control fusarium diseases. Carbendazim reduced disease symptoms by over 50% when used at > 50  µ g mL−1, but had little effect at lower concentrations. Combination of the bacterial isolates and carbendazim gave significant ( P  ≤ 0·05) control of the disease when plants were artificially inoculated with the pathogen. Application of carbendazim at a low concentration (1  µ g mL−1) in combination with B. cepacia c91 reduced disease symptoms by 46%, compared with a reduction of 20% obtained with the bacterium alone and no control with the chemical treatment alone. A combination of B. megaterium c96 with an increased application rate of 10  µ g mL−1 carbendazim significantly reduced disease symptoms by 84% compared with inoculated controls and by 77% compared with carbendazim treatment alone. In this experiment, the integrated treatment also slightly outperformed application of 100  µ g mL−1 carbendazim, and bacteria applied without fungicide also provided good disease control.  相似文献

The population of Phytophthora infestans in Brazil consists of two clonal lineages, US-1 associated with tomatoes and BR-1 associated with potatoes. To assess whether host specificity in these lineages resulted from differences in aggressiveness to potato and tomato, six aggressiveness-related epidemiological components – infection frequency (IF), incubation period (IP), latent period (LP), lesion area (LA), lesion expansion rate (LER) and sporulation at several lesion ages (SSLA) – were measured on detached leaflets of late blight-susceptible potato and tomato plants. Infection frequency of US-1 was similar on potato and tomato leaflets, but IF of BR-1 was somewhat reduced on tomato. Incubation period was longer on both hosts with US-1, although this apparent lineage affect was not significant. Overall there was no host effect on IP. On potato, BR-1 had a shorter LP (110·3 h) and a larger LA (6·5 cm2) than US-1 (LP = 162·0 h; LA = 2·8 cm2). The highest LER resulted when isolates of BR-1 (0·121 cm2 h−1) and US-1 (0·053 cm2 h−1) were inoculated on potato and tomato leaflets, respectively. The highest values of the area under the sporulation capacity curve (AUSC) were obtained for isolates of US-1 inoculated on tomato leaflets (6146) and for isolates of BR-1 on potato leaflets (3775). In general, higher values of LA, LER, SSLA and AUSC, and shorter values of LP were measured when isolates of a clonal lineage were inoculated on their original host than with the opposite combinations. There is evidence that there are quantitative differences in aggressiveness components between isolates of US-1 and BR-1 clonal lineages that probably contribute to host specificity of P. infestans populations in Brazil.  相似文献

The effects of different levels of inoculum of Aspergillus carbonarius and time of inoculation on berry infection and the development of aspergillus bunch rot on grapevines (cv. Sultana) were studied under field conditions. Inflorescences at full bloom were inoculated with aqueous spore suspensions of A. carbonarius containing 0 or 1 × 106 spores mL−1 in 2004/05 and 0, 1 × 102 or 1 × 105 spores mL−1 in 2005/06. In both years, the incidence of infection in inoculated berries was significantly higher than in uninoculated berries. Incidence of infection in berries from veraison until harvest was higher than at earlier stages of bunch development (berry set to berries that were still hard and green). Inoculation of bunches at veraison did not significantly increase A. carbonarius infection prior to harvest, at harvest, 6 days after harvest or when berries were over-ripe. Bunches inoculated at harvest did not significantly increase infection 6 days after harvest or when berries were over-ripe. Aspergillus carbonarius was isolated more frequently from the pedicel end (53·1%) than from the middle section (37·5%) and distal end (35·0%) of berries that were inoculated with 105 spores mL−1.  相似文献

Over a period of 3 years, five agronomically distinct crops of winter wheat were grown in plots in which straw (1 kg m−2), manure (4 kg m−2) or nothing were incorporated into the soil. Plant establishment and height, but not leaf area per tiller, were lower in straw-treated plots. Fertilizer regimes differed between years. Soil and leaf nitrogen were recorded; there was no obvious link between N and any disease or soil amendment. The numbers of leaf layers scorable for disease were similar in all treatments. At the end of the season, plants from straw-treated plots had consistently reduced septoria tritici blotch (caused by Mycosphaerella graminicola ), powdery mildew [caused by Erysiphe ( Blumeria ) graminis ], brown rust (caused by Puccinia recondita ) and foot rot (caused by Fusarium spp.). Early on, M. graminicola was worse in straw-treated plots. In manure-treated plots, P. recondita was reduced but effects on other diseases were inconsistent and slight. A fungicide, chlorothalonil, was applied in one crop; its effects did not interact with those of other treatments. Mycosphaerella graminicola was not suppressed by straw in outdoor pot experiments. Late in the season, straw-treated plants had significantly higher leaf silica ( P  < 0·01). In a glasshouse experiment, plants supplied with silicon had less E. graminis infection ( P  < 0·001) and higher leaf silica, but effects on M. graminicola were inconsistent. A prior inoculation of M. graminicola primed plant defences against a subsequent attack of E. graminis , but only in the presence of adequate Si. It is postulated that straw acts in the field by increasing Si availability.  相似文献

A reliable, sensitive, low-cost and easy-to-use technique is described for the detection of Ralstonia solanacearum (the causal organism of bacterial wilt, BW) in soil. A total of 273 potato isolates belonging to five different biovars (Bv), originating from 33 countries worldwide, were tested and successfully detected by antibodies produced at the International Potato Center (CIP). Isolates of R. solanacearum belonging to Bv1 and Bv2A were successfully detected by double antibody sandwich–enzyme-linked immunosorbent assay (DAS–ELISA) at low population levels after incubation of soil suspensions for 48 h at 30°C in a new semiselective broth containing a potato tuber infusion. Detection thresholds of 20 and 200 CFU g−1 inoculated soil were obtained for Bv1 and Bv2A, respectively. Sensitivity of detection of Bv2A was similar or even higher in five different inoculated soil types. No cross-reactions were obtained in DAS–ELISA after enrichment of soil suspensions (i) prepared from 23 different soils sampled in BW-free areas in six departments of Peru; and (ii) inoculated with 10 identified bacteria and 136 unknown isolates of soil microbiota isolated from eight different locations. Only the blood disease bacterium gave a low-level reaction after enrichment. In naturally infested soils, average sensitivities of 97·6 (SE 14·8) and 100·9 (SE 22·6) CFU g−1 were obtained for biovars 1 and 2A, respectively. By making serial dilutions of the soil suspension before enrichment, densities of R. solanacearum could be determined in a semiquantitative way. Results also showed that composite samples of five soils could be analysed to assess field soil populations without reducing detection sensitivity.  相似文献

Imperata cylindrica (L.) Raeuschel is a dominant and infamous grass weed in the savannah of West Africa. Research to reduce the weed to non-damaging levels is a priority activity at many agricultural institutions. The successful development and implementation of long-term I. cylindrica management strategies depend on the ability to predict changes in weed composition after I. cylindrica has been controlled effectively. The weed flora and soil seedbank were assessed from 329 fields dominated by this species in the fringes of the humid forest (HFF), coastal/derived savannah (CDS) and in the southern Guinea savannah (SGS) in 1996 and 1997. The objectives of the study were to correlate species composition of the weed flora with that of the soil seedbank and to determine the effect of management factors and soil properties on the composition of the weed flora. Species richness in the weed flora and in the weed seedbank was higher in the SGS than in the CDS and HFF. Mean weed density per field was generally higher in the HFF (156 ± 25.0 weeds m–2) than in the CDS (108 ± 8.1 weeds m–2) and in the SGS (92 ± 6.3 weeds m–2). Weed composition varied with agroecological zone as well as with management factors and soil properties. Sørenson's index of similarity was low (mean=0.20) in all zones, indicating poor similarity between the weed flora above-ground and the soil seedbank.  相似文献

The liana, hiptage ( Hiptage benghalensis ), is currently invading the wet tropics of northern Queensland and remnant bushland in south-eastern Queensland, Australia. Trials using seven herbicides and three application methods (foliar, basal bark, and cut stump) were undertaken at a site in north Queensland (158 700 hiptage plants ha−1). The foliar-applied herbicides were only effective in controlling the hiptage seedlings. Of the foliar herbicides trialed, dicamba, fluroxypyr, and triclopyr/picloram controlled >75% of the treated seedlings. On the larger plants, the cut stump applications were more effective than the basal bark treatments. Kills of >95% were obtained when the plants were cut close to ground level (5 cm) and treated with herbicides that were mixed with diesel (fluroxypyr and triclopyr/picloram), with water (glyphosate), or were applied neat (picloram). The costings for the cut stump treatment of a hiptage infestation (85 000 plants ha−1), excluding labor, would be $A14 324 ha−1 using picloram and $A5294 ha−1 and $A2676 ha−1, respectively, using glyphosate and fluroxypyr. Foliar application using dicamba for seedling control would cost $A1830 ha−1. The costs range from 2–17 cents per plant depending on the treatment. A lack of hiptage seeds below the soil surface, a high germinability (>98%) of the viable seeds, a low viability (0%) of 2 year old, laboratory-stored fruit, and a seedling density of 0.1 seedlings m−2 12 months after a control program indicate that hiptage might have a short-term seed bank. Protracted recolonization from the seed bank would therefore be unlikely after established seed-producing plants have been controlled.  相似文献
