A 12‐week feeding trial was conducted to evaluate the effects of fish oil replacement by soybean oil, on lipid distribution and liver histology of two commercially important finfish species: rainbow trout (Oncorhynchus mykiss) and European sea bass (Dicentrarchus labrax). Sea bass (16.2 ± 0.5 g; mean ± SD) and rainbow trout (52.1 ± 0.5 g) juveniles were fed one of three isonitrogenous (500 g kg?1 CP) and isoenergetic (19 kJ g?1) diets, containing 0% (control, diet A), 25% (diet B) and 50% (diet C) soybean oil. At the end of the experiment, lipid deposition was evaluated in muscle, liver and viscera. Cholesterol and triglycerides levels were also determined in plasma. Tissue total, neutral and polar lipid composition (g kg?1 total lipids) showed no significant differences within species, regardless the dietary treatment. The same trend was observed for plasma parameters (P > 0.05). Viscera were the preferential tissue of lipid deposition, with 252–276 and 469–513 g kg?1 total lipid content in trout and sea bass, respectively. Dietary fish oil replacement had no effect on either hepatic lipid droplets accumulation or degree and pattern of vacuolization in the observed liver sections. These data suggest that both sea bass and trout can be fed diets containing up to 50% soybean oil without adverse effects on tissue lipid composition or liver histology.  相似文献

This study evaluated the effects of diets containing 0, 25, 50, 75 and 100 g kg?1 Spirulina platensis on proximate composition, fatty acid profile and lipid peroxidation of rainbow trout (Oncorhynchus mykiss). Supplementation of S. platensis did not change moisture and protein contents, but fish fed 50 and 100 g kg?1 S. platensis had lower muscle lipid content than those fed control diet (< 0.05). Fish fed 100 g kg?1 of S. platensis contained lower percentages of saturated and monounsaturated fatty acid and a higher percentage of polyunsaturated fatty acid than those fed control diet (< 0.05). The n‐3/n‐6 ratio of the fatty acid increased and muscle atherogenic and thrombogenic indices were significantly decreased as the dietary supplement of S. platensis increased. Furthermore, lipid peroxidation of the fillet significantly decreased with increasing dietary S. platensis at 4 °C and at ?20 °C (< 0.05). The results of this study show that supplementation of S. platensis to the diet improves muscle quality of the rainbow trout.  相似文献

Rainbow trout (Oncorhynchus mykiss) skin cell cultures were obtained by trypsinization of the tissue and grown in Leibovitz L-15 medium. Lipid class compositions, and fatty acid profiles of total lipids and individual phospholipid classes were determined at different times of culture. The metabolism of polyunsaturated fatty acids (PUFA) was investigated by incubating primary cultures after 7 and 14 days with [1-14C]18:2n-6 and [1-14C-]18:3n-3. The change in morphology between epithelial-like primary cultures and fibroblastic-like secondary subcultures was accompanied by alterations in the lipid composition. Polar lipids became predominant by 14 days in culture. The relative proportions of phosphatidylcholine (PC), the most abundant phospholipid, phosphatidylinositol and cholesterol increased significantly, while sphingomyelin decreased. Saturated fatty acids, 18:1n-9, n-6 and n-9PUFA were more abundant in total lipid in cultures at 14 days and 4 months than in cells initially isolated which contained higher percentages of longer chain monoenes and n-3PUFA. The changes in fatty acid composition with time in culture were observed in all the major phospholipid classes. Rainbow trout skin cells in culture desaturated and elongated both 18:2n-6 and 18:3n-3, with 20:4n-6 and 20:5n-3 being the most abundant products, respectively. PC presented the highest incorporation of radioactivity, especially following incubation with 18:3n-3. Lipid metabolism in general increased with the age of primary cultures, with both the amount of C18 PUFA incorporated and metabolized by desaturation/elongation significantly increased in 14 day cultures compared to 7 day cultures. Product/precursor ratios calculated for both n-6 and n-3 fatty acids showed that, while 6 desaturase activity was increased significantly with cell age, 5 desaturase activity was more affected by the fatty acid series, with 18:3n-3 being more readily transformed to 20:5n-3 than 18:2n-6 to 20:4n-6. Further desaturation of 20:5n-3 to hexaenes was low. Overall, the data suggested that the trout skin cell cultures were more similar to mammalian skin fibroblasts than mammalian epidermal/keratinocyte cultures.  相似文献

This study investigates whether tissue free amino acid (FAA) pools in rainbow trout,Oncorhynchus mykiss (Walbaum), are altered following feeding and the relationships between the amount of food consumed and the FAA pool size. Trout were starved for 7 days to provide baseline data and then refed on day 8. Individual food intake was measured by radiography and the consumption of amino acids (AA) calculated from dietary protein consumption. Total FAA concentrations in the stomach, liver and white muscle were little changed at various times after the meal and this pattern was repeated for the majority of individual FAA. Overall, the most notable change was a reduction in essential FAA concentrations (principally in valine, leucine and isoleucine) in the white muscle following feeding. However, in the caeca total FAA, total essential FAA and a number of individual FAA were significantly elevated at 4, 9 and 15h following feeding. There were few significant correlations between dietary amino acid consumption and total tissue FAA and essential FAA concentration in the stomach, caecum and white muscle; correlations were stronger in the liver. In order to explain the relative constancy of total FAA concentrations in the tissues following food intake (which represents over 100% of the total FAA pool) a model is presented that quantifies the AA flux through the free pools and considers the role of protein turnover in regulating FAA pool size.  相似文献

Effects of feed ration levels (RLs) and age on sensory characteristics, fillet lipid content (LC) and fatty acid composition (FA) of rainbow trout, Oncorhynchus mykiss (Walbaum), fillet were investigated in a longitudinal feeding experiment. Daily RLs were administered as a percentage of the ration size necessary for expected maximum growth (RL100). Fish on RL50, RL75, RL100 and RL200 were sampled at 1.4, 2.0 and 2.4 years of age. Analyses of LC and FA were carried out on a fillet cross-section. Sensory analyses were performed by an expert panel using ranking and conventional profiling. RL had no effect on the intensity of fresh smell or any sensory characteristic of taste or consistency of fish on RLs over 75%, irrespective of age; fish on RL50, however, scored significantly lower for fresh taste and firmness. Age had no effect on the intensity of total taste, whereas fresh taste increased and rancidity decreased with increasing age irrespective of RL. LC related strongly to RL and age, and increased up to 2.0 years of age. An increase in LC resulted in a higher level of monounsaturated FA and a lower level of polyunsaturated FA. A restriction to RL75 had no negative effects on sensory characteristics, LC or FA of the fillet.  相似文献

ABSTRACT:   This study investigated the effect of conjugated linolenic acid (CLNA) on the growth and lipid composition of fingerling rainbow trout Oncorhynchus mykiss . Thirty individuals in each tank with three dietary treatments and duplicate groups were subjected to three dietary levels (0:4, 2:2, 4:0) of CLNA and linolenic acid (LNA) at 16°C in the laboratory for a period of 9 weeks. Punicic acid ( cis -9, trans -11, cis -13–18:3) and linseed oil were used as sources of CLNA and LNA, respectively. The hepatosomatic index and body lipid content of the 2% CLNA dietary treatment increased significantly ( P  < 0.05) with respect to the control (LNA treatment). But the specific growth rate and percent gain did not improve significantly. After 6 weeks' observation, the 4% CLNA dietary treatment exhibited higher mortality and liver weight but lower weight gains (1.1–1.9 g). No significant differences were observed in the results of the feed efficiency and moisture as well as the protein content of the whole body among the treatments. Incorporation of CLNA into fish tissues was at a distinctly higher level in neutral lipids than in polar lipids. Increased levels of 22:6n-3 were found in the 4% CLNA treatment but decreased levels were found in the 2% CLNA treatment in both neutral and polar fractions. In the CLNA treatments, saturated fatty acid was relatively unaffected, the level of 18:1 was increased and the level of 18:2n-6 was decreased. These results indicate that the lower level (2%) of CLNA had no significant effect on the growth and the higher level (4%) of CLNA caused higher mortality as well as attenuating the growth of the fish. Dietary CLNA accumulated in a greater amount in the neutral lipids and modulated the fatty acid composition in the whole body of rainbow trout fingerlings.  相似文献

Rainbow trout Oncorhynchus mykiss (Walbaum) were studied for 98 days for their response to the administration of a 1% rapeseed lecithin supplement in dry pellets. Lecithin tended to stimulate erythropoiesis, which manifested itself as a higher erythrocyte count, a higher haematocrit level and higher haemoglobin concentration during the course of the trial, compared with the control group. The significant differences that were found in the haematological profile, in the parameters of nitrogen metabolism (total protein, blood nitrogen urea, uric acid, creatinine) and mineral metabolism (inorganic phosphate, total calcium) and in the activity of catalytic enzymes (alanine aminotransferase), in certain periods of the trial do not suggest any adverse effects on the state of health of the fish; the levels of haematological parameters remained within the range of reference values. The catalytic activity of the aspartate aminotransferase, alkaline phosphatase, lactate dehydrogenase and α‐hydroxybutyryl dehydrogenase as well as the hepatosomatic index and condition factors calculated after Fulton and after Clark were unchanged. Lecithin supplement to the diet caused no clinical changes in the state of health of the fish, nor were any changes ascribable to the administration of lecithin observed on the basis of the patho‐anatomic and histological examination. Considering this and the good results in terms of the supporting antioxidant properties and the effect on growth capacity, recorded in previous experiments, rapeseed lecithin at the tested concentration level can be recommended as a supplement to dry pellets for rainbow trout.  相似文献

The aim of this study was to determine the effects of replacing fish oil (FO) with laurel seed oil (LSO), as an alternative plant lipid source in diets on the growth and fatty acid composition of rainbow trout (Oncorhynchus mykiss; 111.47 ± 0.2 g mean individual weight). At the end of the feeding trial, survival was 100% in all treatments. No significant differences were seen in growth between the dietary groups (P > 0.05). The protein, lipid and ash contents were not significantly different among the groups (P > 0.05); however, there was a significant difference in protein and ash content between the treatment groups and the initial, and between the 50LSO group and the initial group, respectively (P < 0.05). The viscerosomatic index (VSI) and hepatosomatic index (HSI) values were not affected by increasing LSO percentages in the diets. The n‐6 polyunsaturated fatty acid (PUFA) concentration increased with increasing LSO levels in the diets. In contrast, the n‐3 PUFA levels decreased with increasing LSO levels in the diets. The liver and muscle were used for the analysis of fatty acids. The highest level of eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) concentrations was recorded in fillet of fish fed the FO diet and the lowest in those fed the 50LSO diet. However, EPA and DHA ratios in the liver of fish fed the 75LSO diet were higher than those in fillet of fish fed the FO and 50LSO diets. No significant differences were seen in fatty acid composition between the dietary groups (P > 0.05). Based on the results of growth performance and fatty acid composition of the experimental fish in this study, it can be concluded that the 75% concentration of laurel seed oil performed best among the diets tested in the experiment.  相似文献

The application of natural and innocuous compounds has potential in aquaculture as an alternative to antibiotics. We evaluated the effect of diet supplementation with Thymus vulgaris essential oil (TVEO) on the allochthonous microbial composition of rainbow trout. DNA was extracted directly from the intestinal contents, and the V3‐V4 regions of the 16S rRNA genes were amplified by PCR. The bacterial composition was analysed using temporal temperature gradient electrophoresis (TTGE). No significant changes (P>0.05) were detected in the TTGE profiles of TVEO‐treated trout compared with the controls. The Dice similarity index revealed a high stability (Cs >70%) of the intestinal microbiota in both groups during the 5‐week period. Sequence analyses of the TTGE bands revealed the same bacterial composition in both groups, with most bacteria belonging to the Proteobacteria and Firmicutes phyla. The in vitro antibacterial activity of TVEO was assessed using a range of normal intestinal isolates and fish pathogens. The inhibitory concentrations for all the tested bacteria were higher than the TVEO levels used in trout, which may explain the in vivo results.  相似文献

Two levels (40 or 80 mg kg?1) of avoparcin were tested as a growth promoter in rainbow trout, Oncorhynchus mykiss (Walbaum), fed for 14 weeks. At 80 mg kg-1 the weight gain was higher (P <0.05) than in the unsupplemented control or at the 40 mg kg-1 dose. At either dose, the feed conversion rate and the protein efficiency ratio were more favourable (P <0.05) than in the control. Inclusion of the antibiotic significantly improved (P < 0.05) the apparent digestibility of lipids. The lipid content of flesh was significantly increased by inclusion of avoparcin in the diet; moreover, at the high level of inclusion the content of some unsaturated fatty acids (18:3ω3 and 20:5ω3) appeared to be significantly increased. No avoparcin residues were detected in the flesh.  相似文献

The present study investigated the effect of arginine on seminal plasma composition in rainbow trout. Male rainbow trout broodstocks (2500 ± 200 g) were fed five practical diets (each consisting of three triplicates) supplemented with Arginine at 0.50%, 1.50% and 2.00%. The control group were fed without arginine. Broodstock feeding lasted for 90 days, and then fish semen was sampled. Results indicated no significant differences in LDH, ALP, Fe2+ and phosphorous content among the different treatments. The lowest levels of AST and ALT and the highest levels of Ca2+ and Mg2+ ions were observed in the treatment fed with 1.50% arginine, which showed significant differences from other treatments (P < 0.05). Moreover, the amount of Cl?, Na+ and K+ ions was significantly increased in the seminal plasma in fish fed diets containing arginine in comparison with the control. As the amount of arginine was increased, the levels of uric acid became significantly greater in contrast to urea and glucose levels. The highest amounts of cholesterol, fructose and total protein were observed in treatments fed on 2.00%, 0.50% and 1.00% arginine, respectively, showing significant differences from other treatments (P < 0.05). The highest pH value was assayed in the 1.50% arginine treatment. Results indicated that arginine had a potential efficacy on semen quality in rainbow trout broodstocks.  相似文献

For the first time, pre‐ and post‐hepatic plasma lipid profiles were monitored following a single meal in a free‐swimming, non‐anaesthetized fish. Rainbow trout (Oncorhynchus mykiss; 700–1500 g; 10 °C) were equipped with cannulae in the dorsal aorta (DA) and hepatic portal vein (HPV). Simultaneous blood samples, taken from both cannulae at 0, 3, 6, 12, 24 and 48 h postprandial, revealed the time course of the plasma lipid profiles following a single meal (1% of body mass). Primarily monounsaturated fatty acids with the exception of 18:1n ? 9, increased significantly from baseline by 12 h postprandial without greatly affecting total plasma lipid concentrations. Total plasma lipids then showed a small peak at 24 h postprandial, coinciding with a peak in triacylglycerols. We conclude that assimilation of lipids from the digest into the plasma is slower than reported for proteins and carbohydrates in the same species. Furthermore, as there were no significant differences between the HPV and DA, no measurable effect of hepatic passage on plasma lipid levels was resolved. Therefore, we also conclude that, in contrast to that in higher vertebrates, hepatic passage does not seem to have a major role in rainbow trout for modulating the postprandial plasma profile of lipids.  相似文献

The effects of sub-lethal doses of dichlorvos and formalin, antimicrobial/parasitic agents used in aquaculture, on lipid composition and metabolism of rainbow trout skin cells in primary culture were investigated. [1-14C]Stearic (18:0), [1-14C]lin 18:2n-6) and [1-14C]linolenic (18:3n-3) acids were used as tracers to determine effects on fatty acid incorporation and metabolism. Formalin increased cell numbers and reduced the lipid content of the cells and the incorporation of radioactive fatty acids. The effects of dichlorvos were qualitatively similar but quantitatively less. Formalin induced relatively small but significant changes in lipid class composition including a decreased proportion of phosphatidycholine with increased proportions of sphatidylethanolamine and phosphatidylserine. Dichlorvos had no significant effect on lipid class compositions. The trout primary skin cells expressed substantial 9, 6 and 5 fatty acyl desaturase activities. Although, as expected, the cells were m active towards [1-14C]18:3n-3, the cells were unusually active towards [1-14C]18:2n-6. Both dichlorvos and, especially, formalin appeared to significantly inhibit 9 and 6 desaturation. Changes in the distribution of radioactivity between individual spholipid classes was also influenced by formalin and dichlorvos, and this may be related to changes in desaturase activity. This study has shown that topically active agents used in aquaculture, formalin and dichlorvos, had a range of effects on the rainbow trout skin cell cultures that may affect cell proliferation and lipid and fatty acid metabolism. Both agents significantly inhibited desaturation of fatty acids, particularly of 18:2n-6 to 20:4n-6 and, as 20:4n-6 is a major eicosanoid precursor ish and considering the importance of eicosanoids in the biochemistry of skin, it is suggested that these agents may have direct effects on fish skin that could have important consequences for fish health in general.  相似文献

To study the intestinal fatty acid absorption in fish in vitro, enterocytes were isolated from the intestine of rainbow trout and incubated with an equimolar mixture of seven fatty acids [16:0, 18:1n-9, 18:2n-6, 18:3n-3, 20:4n-6, 20:5n-3 and 22:6n-3] in which the component carrying a radioactive label was varied. The fatty acid mixture was presented in the form of micelles formed by sonication with sodium taurocholate. Control studies showed that the presence of sodium taurocholate in the incubation medium caused an immediate 23% increase in the mortality of the cells but the remaining cells were viable. The effect of the bile salt on cellular permeability was evident at longer exposure periods. The proportions of 14C-labelled 20:5n-3 and 22:6n-3 present as monomeric fatty acids in the micellar solutions were higher than those of 16:0, 18:1n-9, 18:2n-6, 18:3n-3 and 20:4n-6. The rates of uptake of 14C-labelled 20:4n-6, 20:5n-3 and 22:6n-3 by enterocytes were significantly lower than those of the other fatty acid substrates over the first minute of incubation but no significant differences in uptake rate between fatty acids were obvious over a 15 min incubation period. Notable differences were observed between substrates in their distribution pattern in enterocyte lipid classes. Although most of the radioactivity from all radiolabelled substrates was recovered in triacylglycerols, the amounts of 14C- labelled 16:0, 20:4n-6, 20:5n-3 and 22:6n-3 recovered in the polar lipid fraction were higher than those of 14C-labelled C18 substrates, particularly after 15 min. Conversely, the initial esterification rates of 18:1n-9, 18:2n-6 and 18:3n-3 into triacylglycerols were significantly higher than those of 20:4n-6, 20:5n-3 and 22:6n-3. It is concluded that isolated enterocytes can be used for the study of the mechanism of intestinal fatty acid absorption in fish.  相似文献

As a marine carnivore exhibiting exceptionally high growth rates, cobia are considered a species for which fish oil (FO) replacement may be difficult. However, partial, if not complete, FO replacement is necessary to ensure sustainability. We evaluated the effects of graded substitution of dietary FO with soybean oil (SO) in cobia culture. Feeds contained FO (100% FO), SO (0% FO) or blends of the two (67% FO, 33% FO) as the supplemental lipid source. Production performance was largely unaffected by partial replacement of FO with SO: feed intake and final weight were reduced only in the 0% FO dietary treatment. Fillet total lipid fatty acid (FA) composition differed among the dietary treatments, closely approximating dietary FA profile. As increasing amounts of FO were replaced, SO‐associated FA became enriched within the fillet lipid at the expense of FO‐associated FA. Fillet lipid classes were associated with a particular FA signature, regardless of dietary FA profile. SO can replace a substantial amount of dietary FO; however, juvenile cobia appear to exhibit a nominal requirement for intact long‐chain polyunsaturated FA. Therefore, aggressive FO replacement may result in essential fatty acid deficiencies unless the feeds can be amended with alternative sources of these essential nutrients.  相似文献

Clove oil has been demonstrated to be an effective, inexpensive anaesthetic and euthanizing agent for a number of fish species, including rainbow trout, used in aquaculture and fisheries research. However, the potential for clove oil to cause perturbations in important plasma hormone concentrations has not been investigated. The effect of anaesthesia and euthanasia in trout with eugenol (the active ingredient in clove oil) on plasma cortisol, glucose, growth hormone (GH) and two thyroid hormones [tri‐iodothyronine (T3) and thyroxine (T4)] was compared with tricaine methanesulfonate (MS‐222) anaesthesia, and stunning by cranial concussion in two experiments. Effects on blood chemistry were different when comparing the particular anaesthetic method being used. Stunning fish significantly increased plasma cortisol and glucose levels (both P<0.05), while euthanizing fish using either clove oil or MS‐222 had no effect on these hormone levels. In contrast, the levels of GH, T3 and T4 hormones were unaffected regardless of whether fish were euthanized by stunning, MS‐222 or clove oil. Variation in effects between hormones were observed using clove oil eugenol. In fish sampled 10 min after anaesthetizing with 150 mg L?1 of eugenol, cortisol levels were significantly decreased (P<0.03), while there were no differences in either glucose or GH levels. Tri‐iodothyronine and T4 also showed significantly elevated levels (P<0.05) after 10‐min exposure to eugenol. These results highlight the importance of investigating the potential effects of any new anaesthetic or euthanizing compounds on blood plasma parameters, prior to using them in a research setting, or when comparing results to other studies which have utilized alternative anaesthetic compounds.  相似文献

Processed soybean meal ingredients have become an emerging plant‐based protein used in aquafeed formulations and have been shown to influence gastrointestinal health and microbiota in cultured species. Two 60‐day feeding trials with juvenile rainbow trout (Oncorhynchus mykiss, Walbaum) were conducted to compare the effects of fishmeal (FMC), defatted soybean meal (SBM), bioprocessed soybean meal (BSBM) and commercial soy protein concentrate (CSPC) ingredients on intestinal histology, innate immunity and microbiota profiles. Results indicated no significant differences in intestinal immunoglobulin concentrations (p = .41) or gut leukocyte phagocytosis at day 15 samplings (p = .41). Intestinal lysozyme content was found to be highest at day 60 in the BSBM treatment group (p < .01). No significant differences across treatments were observed in histological profiles at day 0 (p = .75), day 15 (p = .08) and day 60 (p = .22). ANOSIM analysis for microbiota beta diversity showed differences among all treatment groups, with the exception of the high and low levels of BSBM (p = .40). There were also differences observed in the alpha diversity ratios, with significant differences in gut communities among dietary groups as demonstrated by Chao‐1 (p < .01) and Shannon indices (p = .02). The incorporation of processed soy‐based proteins alters the microbial community composition within the distal intestine, and modulates lysozyme concentrations within the distal intestinal tissues.  相似文献

This study aimed to investigate the effects of dietary crude palm oil (CPO) on fatty acid metabolism in liver and intestine of rainbow trout. Triplicate groups of rainbow trout for 10 weeks at 13 °C were fed on diets in which CPO replaced fish oil (FO) in a graded manner (0–100%). At the end of the trial, fatty acid compositions of flesh, liver and pyloric caeca were determined and highly unsaturated fatty acid (HUFA) synthesis and fatty acid oxidation were estimated in isolated hepatocytes and caecal enterocytes using [1‐14C]18:3n‐3 as substrate. Growth performance and feed efficiency were unaffected by dietary CPO. Fatty acid compositions of selected tissues reflected the dietary fatty acid composition with increasing CPO resulting in increased proportions of 18:1n‐9 and 18:2n‐6 and decreased proportions of n‐3HUFA, 20:5n‐3 and 22:6n‐3. Palmitic acid, 16:0, was also increased in flesh and pyloric caeca, but not in liver. The capacity of HUFA synthesis from 18:3n‐3 increased by up to threefold in both hepatocytes and enterocytes in response to graded increases in dietary CPO. In contrast, oxidation of 18:3n‐3 was unaffected by dietary CPO in hepatocytes and reduced by high levels of dietary CPO in enterocytes. The results of this study suggest that CPO can be used at least to partially replace FO in diets for rainbow trout in terms of permitting similar growth and feed conversion, and having no major detrimental effects on lipid and fatty acid metabolism, although flesh fatty acid compositions are significantly affected at an inclusion level above 50%, with n‐3HUFA reduced by up to 40%.  相似文献
