To investigate potential use of dietary β-1,3-glucan for health management of hybrid striped bass, juvenile fish were fed diets supplemented with yeast glucan (MacroGuard® ) at 0.05%, 0.1% or 0.2% of diet for 4 weeks, followed by immune response assays and a bath challenge with Streptococcus iniae . Dietary glucan significantly ( P <0.05) enhanced neutrophil oxidative radical production, and fish fed 0.1% glucan had a significant ( P <0.05) reduction in mortality (10%) after bacterial challenge compared with fish fed the control diet (46.7%). However, accumulative mortality of fish fed 0.2% glucan was not significantly different from that of fish fed the control diet. To further elucidate this observation, macrophages from sub-adult hybrid striped bass were isolated and cultured in L-15 medium with 10% foetal calf serum and penicillin/streptomycin supplemented with 0.2, 0.5, 1, 2, 5, 20 and 100 μg soluble glucan (MacroGuard® ) mL−1 for 24 and 48 h. Intracellular superoxide anion production was significantly ( P <0.001) increased by 0.5 μg glucan mL−1 , but significantly ( P <0.001) suppressed by doses >5 μg glucan mL−1 . It is concluded that dietary yeast glucan has potential for use in diet formulations of hybrid striped bass to limit the adverse effects of S. iniae , but dosage should be an important consideration in administration. 相似文献

Estimating the quantitative essential amino acid requirements of striped bass Morone saxatilis, using fillet A/E ratios 总被引:1,自引:0,他引:1
In order to determine the essential amino acid requirements (EAA) of striped bass Morone saxatilis , fillets were analysed to ascertain the relative amino acid concentrations for determining A/E ratios ((EAA/total EAA) × 1000)). Analysis of the striped bass fillets yielded the following concentrations of essential amino acids (g kg–1 ) and A/E ratios, respectively: arginine, 12.5, 115; histidine, 5.1, 47; isoleucine, 8.0, 74; leucine, 17.1, 157; lysine, 20.2, 186; methionine + cysteine, 9.2, 85; phenylalanine + tyrosine, 16.0, 147; threonine, 9.8, 90; tryptophan, 1.9, 18; and valine, 9.1, 84. In two experiments, diets with graded levels of EAA were fed to striped bass weighing 111 ± 3 g and 790 ± 122 g per fish, respectively. In both experiments, the dietary A/E ratios were maintained in the same relative concentrations as determined in the striped bass fillets. Statistical analysis of weight gains, feed conversions and nitrogen balance indicated significant differences ( P < 0.05) between treatments. Non-linear regression analysis of the response criteria pooled from both experiments yielded the following estimates of dietary EAA requirements (g kg–1 dry diet) when digestible energy equalled 13.39 MJ kg–1 diet: arginine, 14; histidine, 6; isoleucine, 9; leucine, 19; lysine, 22; methionine + cysteine, 10; phenylalanine + tyrosine, 17; threonine, 11; tryptophan, 3; and valine, 10. The use of fillet A/E ratios allows for the rapid estimation of quantitative EAA requirements and the development of species specific diets for new aquaculture species. The data presented here are the first to simultaneously describe all the dietary EAA requirements for M. saxatilis. 相似文献

Abstract The ability of consumers and fishermen to distinguish-between wild striped bass, Morone saxatilis (Walbaum), and its hybrids may be important to marketing farm-raised fish and managing wild stocks. The ability of randomly selected persons to distinguish between wild and hybrid specimens was examined and it was found that a significant improvement in recognition occurred with only minimal training. Average correct response of 70% without training rose to nearly 90% after training. Moreover, the respondents' sex and age were important factors in determining successful identification. 相似文献

Three experiments were conducted that were designed to evaluate our ability to predict essential amino acid (EAA) needs of hybrid striped bass using the quantified lysine requirement and whole‐body amino acid concentrations. In the first experiment, six diets containing various amino acid profiles were fed to triplicate groups of fish initially weighing 7.7 g per fish. At the end of the 8‐week experiment, no significant differences were detected in growth rates or feed efficiencies (FE) between fish fed a practical diet containing 510 g kg?1 herring fish meal (FM) and fish fed a purified diet containing the amino acid profile of herring fish meal (CAA‐FM). Growth responses of fish fed purified diets containing 100 (HSB), 110 (HSB110), 120 (HSB120) or 140 g 100 g?1 (HSB140) of the amino acid profile of hybrid striped bass whole‐bodies were significantly lower than those of fish fed diet FM. In the second experiment, triplicate groups of fish (5.6 g per fish) were fed diets containing various energy : protein (E : P) ratios (34.8, 41.2, 47.5 and 53.9 kJ g?1 protein) and one of two amino acid profiles (CAA‐FM and HSB120) in a 4 × 2 factorial design. Carbohydrate concentration was varied to achieve the desired energy concentrations. At the end of the 8‐week experiment, weight gain and FE were significantly higher in fish fed diets formulated to simulate the amino acid profile of herring fish meal (CAA‐FM) compared with fish fed diets formulated to contain 120 g 100 g?1 of the amino acid profile of hybrid striped bass whole‐bodies (HSB120). Weight gain, FE and survival data indicated the optimum dietary E : P was 41.2 kJ g?1 protein. Dietary treatments in the final experiment included three amino acid profiles and four levels of lipid in a 3 × 4 incomplete factorial design. Dietary amino acid treatments included the amino acid profile of herring fish meal (CAA‐FM) or 120 g 100 g?1 of the predicted EAA requirement profile for hybrid striped bass (HSB120). The amino acid profile of the remaining dietary treatment (PRED+) was similar to that of the HSB120 treatment, but contained additional threonine, isoleucine and tryptophan. Diets CAA‐FM and HSB120 contained either 90, 130, 170 or 210 g kg?1 lipid, whereas diet PRED+ contained 130 g kg?1 lipid. Dietary treatments were fed for 10 weeks to triplicate groups of fish initially weighing 81.0 g per fish. Weight gain and FE were not significantly affected by dietary amino acid profile. Feed efficiency was significantly reduced in fish fed diets containing 210 g kg?1 lipid compared with fish fed diets containing 90–170 g kg?1 lipid. Intraperitoneal fat (IPF) ratio and hepatosomatic index (HSI) values generally increased as dietary lipid concentrations increased. Total liver lipid concentrations were significantly reduced in fish fed diets containing 210 g kg?1 lipid compared with those of fish fed 90–130 g kg?1 lipid. Results of this study indicate an appropriate dietary amino acid profile can be predicted for hybrid striped bass using the quantified lysine requirement and whole‐body amino acid concentrations. This study was conducted to determine the dietary vitamin E requirement of juvenile hybrid striped bass ( Morone chrysops female × Morone saxatilis male). Semi-purified diets supplemented with 0.2 mg Se kg−1 from Na2 SeO3 and either 0 (basal), 10, 20, 40, 60, or 80 mg vitamin E kg−1 as DL -α-tocopheryl acetate were fed to hybrid striped bass initially averaging 1.8 ± 0.1 g (mean ± SD) for 12 weeks. Fish fed the basal diet, which contained 5.8 mg α-tocopherol kg−1 dry weight, were darker in colour and had reduced weight gain, as well as generally reduced haematocrit values compared with fish fed diets supplemented with vitamin E. In addition, fish fed diets containing less than 20 mg supplemental vitamin E kg−1 had significantly ( P < 0.05) reduced weight gain and feed efficiency compared with those fed diets supplemented with vitamin E at 20–80 mg kg−1 . Dietary supplementation of vitamin E caused incremental increases in the concentration of α-tocopherol in both plasma and liver tissues. However, hybrid striped bass fed graded levels of vitamin E did not exhibit a dose response in terms of ascorbic acid-stimulated lipid peroxidation of hepatic microsomes. Regression analysis of weight gain data using the broken-line model indicated a minimum vitamin E requirement ( ± SE) of 28 ( ± 3) mg kg−1 dry diet. Based on these data, the dietary vitamin E requirement of hybrid striped bass appears to be similar to that determined for other fish species. 相似文献

Two growth studies were conducted to determine the dietary threonine requirement of reciprocal cross hybrid striped (sunshine) bass. Semipurified diets were prepared with crystalline amino acids and lyophilized fish muscle to supply 350 g crude protein kg−1 diet. The basal diet contained 4.9 g threonine kg−1 from fish muscle, and test diets were supplemented with graded levels of L-threonine. In the first experiment, fish initially averaging ≊ 9.8 g each were fed diets containing threonine levels of 4.9, 7.5, 10.0, 12.5, 15.0 and 17.5 g kg−1 dry diet for 7 weeks. Weight gain, feed efficiency and protein efficiency ratio (PER) were significantly ( P < 0.01) influenced by dietary threonine level. Based on weight-gain responses, a threonine requirement (± SE) of 8.4 (± 0.8) g kg−1 dry diet was determined, and dietary threonine levels of 10.0 g kg−1 diet or greater resulted in the highest levels of free threonine in plasma.
Based on the results of the first experiment, a second feeding trial was conducted with diets containing threonine levels of 4.9, 6.5, 8.0, 9.5, 11.0 and 12.5 g kg−1 dry diet. Fish initially averaging ≊ 3.0 g each were fed each diet for 8 weeks. Weight gain, feed efficiency and PER values of fish were markedly improved, with increases in dietary threonine up to 8.0 g kg−1 dry diet. Regression analysis of weight gain, feed efficiency and PER data using the broken-line model resulted in threonine requirement estimates of 9.7, 8.5 and 8.6 g kg−1 dry diet, respectively. Based on these data, the threonine requirement of juvenile sunshine bass was determined to be ≊ 9.0 g kg−1 dry diet or 26 g kg−1 of dietary protein. 相似文献

The hybrid striped bass (HSB) farming industry has considerable potential for growth into domestic retail markets, but expansion of this industry is limited by high production costs that dictate high prices for HSB. It is widely recognized within the industry that selective breeding of an improved HSB will be required to increase production efficiency and reduce market prices. A National Program of Genetic Improvement and Selective Breeding for the HSB Industry has been initiated and some progress has been made toward domestication of the parent species of the hybrid. However, uncertainty remains as to which breeding procedures will most rapidly yield sustainable genetic gains in key production traits. This paper consolidates and reviews general information on the biology of temperate basses (genus Morone) relevant to selective breeding of improved HSB. The topics covered include control of reproduction, geographic distribution of stocks and population genetic variation. This is followed by a brief review of the current application of selective breeding techniques, including those based on molecular markers. Finally, we discuss potential avenues for genetic improvement of HSB in a selective breeding program. 相似文献

Requirements for six of the 10 indispensable amino acids (IAA) have not been quantified for hybrid striped bass. In this study, we estimate the requirement for l ‐tryptophan by nonlinear regression analysis of several growth indicators. Fifteen isocaloric, isonitrogenous diets were formulated to contain 1.0, 1.3, 1.6, 1.9, 2.2, 2.5, 2.8, 3.1, 3.4, 3.7, 4.0, 5.0, 6.0, 10.0 or 14 g l ‐tryptophan kg?1 diet and fed to reciprocal cross hybrid striped bass for 7 weeks. After 5 weeks, survival of fish receiving the basal diet was 40% while surviving fish fed this diet were in poor health. Survival of fish receiving 1.3 g Trp kg?1 diet declined to 70% by termination of the trial. Survival in other treatments was 100%. Fish weight gain was 100% or greater for fish receiving 1.6 g Trp kg?1 diet or more. Hepatosomatic index, muscle ratio and intraperitoneal fat (IPF) ratio also responded to dietary tryptophan concentration. IPF was inversely related to dietary tryptophan concentration. The dietary tryptophan requirement was estimated to be between 2.1 and 2.5 g kg?1 diet (6–7 g kg?1 protein), depending on response variable, using four‐ and five‐parameter saturation kinetics models. This study was conducted to determine the effect of phytase on apparent dry matter (DM), crude protein and phosphorus (P) digestibility of four plant feedstuffs (isolated soya protein, ISP; soyabean meal, SBM; corn gluten meal, CGM and wheat middlings, WM) fed to striped bass Morone saxatilis . One reference diet was formulated based solely on animal feed ingredients. Four test diets were prepared by mixing 60–75% of the reference diet with 40–25% of the test ingredient in order to reach similar levels of total P (0.67–0.73%) and to have a minimum of 35% protein (36.1–53.0%). Chromic oxide was added at 0.5% of the diet and used as an indigestible marker. A phytase solution was sprayed postpelleting on the test diets at a concentration of 1000 phytase units kg−1 (PU kg−1 ) dry diet. The test diets (with or without phytase supplementation) and the reference diet (nine treatments total) were fed to 3-year-old striped bass and were replicated in time ( n =3) during the 4-week period. Digestibility of DM was significantly lower ( P < 0.001) for SBM and WM when compared with CGM and ISP and was not influenced by phytase supplementation. Crude protein digestibility was also not affected by phytase supplementation and was significantly lower ( P < 0.09) for SBM when compared with ISP. Phosphorus digestibility was improved by approximately 23% with the addition of phytase for all four feedstuffs tested ( P < 0.001). Furthermore, WM had significantly lower ( P < 0.0001) P digestibility when compared with the other feedstuffs, irrespective of phytase supplementation. 相似文献

This study was carried out to test the effect of triiodothyronine (T3) on the growth and survival of larval striped bass (Morone saxatilis). Growth and survival of striped bass held in 5 ppt seawater and treated with various doses of T3 were measured beginning at 5 and 16 days after hatching. Body content of T3 was measured by radioimmunoassay. T3 dissolved in the 5 ppt seawater was taken up by larval striped bass in a dose-dependent manner, and affected the growth and survival of the fish. At 5 days after hatching, T3 at 100 ng ml–1 and 50 ng ml–1 retarded the growth of larval striped bass and caused a lower survival rate than T3 at 25 ng ml–1 or the control treatment. At 16 days after hatching, T3 at 100 ng ml–1 retarded the growth of larval fish and caused a higher mortality. T3 at 10 ng ml–1 and 1 ng ml–1 did not show any effect on either survival or growth. Body content of T3 returns to control levels within days following end of treatment. The results indicate that exogenous T3 can be detrimental to the growth and survival of larval striped bass. 相似文献

The egg yolk precursor, vitellogenin (VTG), was purified from blood plasma of striped bass by chromatography on hydroxylapatite
or DEAE-agarose. The fish were first implanted with estradiol-17β (E2), which induced vitellogenesis. A rabbit antiserum (a-FSPP) raised against plasma from mature female striped bass, and then
adsorbed with mature male plasma, was used to detect female-specific plasma protein (FSPP) in the chromatography fractions.
Striped bass VTG (s-VTG) was collected from the peak fraction that was induced by E2, reacted with a-FSPP, and contained all detectable phosphoprotein. It appeared as a single band (Mr ≂ 170,000) in SDS-PAGE or Western blots using a-FSPP, and as a pair of closely-spaced phospholipoprotein bands in native
gradient-PAGE, suggesting that there is more than one circulating form of s-VTG. The relationship of s-VTG to the yolk proteins
was verified using a-FSPP. The antiserum reacted with the main peak from gel filtration of saline ovary extracts, and it specifically
immunostained the two main bands in Western blots of the extracts and the yolk granules of mature oocytes. The amino acid
composition of s-VTG was similar to that of VTG from other fish and Xenopus. A radial immunodiffusion assay for s-VTG was developed using a-FSPP and purified s-VTG as standard. The s-VTG was not detected
in blood plasma of males, immature females, or regressed adult females, but plasma s-VTG levels were highly correlated with
plasma E2 and testosterone levels, and oocyte growth, in maturing females. The results indicate that the maturational status of female

Several experiments were conducted to determine the effects of phytase on dietary phosphorus (P) utilization by striped bass Morone saxatilis fed high phytate diets. The experiments were designed to determine the effectiveness of various dietary levels of a dry or liquid phytase concentrate incorporated in diets to improve the P utilization of striped bass. Fish were fed various basal diets containing over 700 g kg?1 plant feed ingredients and 4.9–7.1 g kg?1 P and 1.5–1.7 g kg?1 non-phytin P. A diet supplemented with potassium monophosphate (PMP) and containing 9 g kg?1 total P and 6 g kg?1 non-phytin P with no added phytase was the positive control. The dietary treatments were assigned to duplicate tanks and diets were fed to juvenile striped bass for up to 14 weeks. The effectiveness of the phytase treatments was determined by measuring weight gain, feed conversion, serum, scale and vertebral calcium and P, as well as P absorption. Apparent P absorption was determined using 5 g kg?1 chromic oxide as an indigestible marker in the diet. In experiment one, significant improvements (P < 0.05) were found in scale and vertebral phosphorus concentrations with 2400 Phytase Units (PU) kg?1 (PU is the quantity of enzyme which liberates one micromole of inorganic P per minute from 0.015 tool L?1 sodium phytate at 37°C and pH 5.5) added to the diet. In experiment two, significant differences were observed between serum phosphorus in the phytase and no-phytase groups, while there were no differences between the positive control (PMP supplemented) and the phytase-treated fish. It was concluded that 2400 PU kg?1 of enzyme resulted in bone mineralization and serum phosphorus concentrations equal to that observed with 13 g kg?1 dietary PMP addition (9 g kg?1 total P). 相似文献

The refinement of hybrid striped bass feed formulations has been progressing rapidly. There is still, however, limited information available regarding nutrient digestibility and amino acid availability from common feedstuffs. A pair of experiments was conducted to determine the protein digestibility and amino acid availability to sunshine bass from an assortment of commercially available animal protein feedstuffs, blended animal products and plant protein feedstuffs in extruded diets. The feedstuffs tested were blood meal, poultry by‐product meal, fish solubles, Pro‐Pak 60, Pro‐Pak 65, ProCon 65RDB, and 60FMC for the animal protein feedstuffs and brewer's yeast, canola meal, peanut meal, and sunflower meal for the plant protein feedstuffs. Test diets consisted of a mixture of nutritionally complete reference diet and test ingredient. Triplicate tanks of fish were fed their respective diets for 7 days prior to collection of faeces by stripping. Apparent digestibility coefficients of protein (ADC‐CP) in the animal products ranged from 47% for 60FMC to a high of 70% for fish solubles. ADC‐CPs for animal products were not different across products. Blood meal, poultry by‐product meal, Pro‐Pak 60, Pro‐Pak 65, and ProCon had intermediate ADC‐CPs of 63, 55, 63, 57 and 52%, respectively. ADC‐CP in plant feedstuffs ranged from 43% for canola meal to 80% for peanut meal. Brewer's yeast, canola meal, and sunflower meal had intermediate ADC‐CPs at 54, 43 and 69%, respectively. Apparent amino acid availability coefficients were variable across animal products and did not necessarily correlate to the ADC‐CPs for a given feedstuff. Isoleucine availability was low in blood meal at 38% compared with 59% or better for the remaining amino acids. Lysine, tyrosine and phenylalanine availability from fish solubles was low at 31, 35 and 44%, respectively. Amino acid availability from Pro‐Pak 60 was consistently higher across all amino acids for the animal products and blends tested. Of the plant products tested, peanut meal was the best performing feedstuff relative to amino acid availability. 相似文献

Performance traits and body composition of juvenile hybrid striped bass (Morone chrysops × M. saxatilis) in response to hypoxia were evaluated in replicate tanks maintained at constant dissolved oxygen concentrations that averaged 23.0 ± 2.3%, 39.7 ± 3.0% and 105.5 ± 9.5% dissolved oxygen saturation. Fish were fed a commercially formulated feed daily to apparent satiation. Total feed intake and fish growth and yield increased linearly in response to increased dissolved oxygen concentration. Nutrient utilization was reduced significantly only at the greatest level of hypoxia. With the exception of whole body protein content, whole body compositional indices and nutrient retention efficiencies were linearly related to dissolved oxygen concentration. Results demonstrate that as hypoxia becomes more severe, juvenile hybrid striped bass feed intake is reduced, which affects growth and nutrient retention. 相似文献

Whole-animal thyroxine (T4) and 3,5,3′-triiodothyronine (T3) levels were measured in larval and juvenile striped bass, Morone saxatilis, reared for 10 days at one of three levels of salinity (equivalent to fresh water (FW), one-third seawater (1/3 SW), and
seawater (SW) and two temperatures (15°C and 20°C). The striped bass were pre-metamorphic larvae, metamorphic larvae or juveniles.
The short-term effects of seawater on plasma T4 levels of juvenile striped bass were also measured. Higher salinities increased T4 levels in premetamorphic larvae. In metamorphic larvae, SW and 1/3 SW increased T4 levels and SW increased T3 levels at 20°C. This response was eliminated in those at 15°C. Whole-animal thyroid hormone content was unaffected by salinity
or temperature in juvenile striped bass, although significant fluctuations in plasma T4 levels occurred in those transferred to 1/3 SW and SW. The thyroid axis of striped bass responds to salinity and temperature
as early as in the pre-metamorphic stage. Thyroid hormones may mediate the beneficial effects of salinity on larval striped

A study was conducted with market‐size (867 g) hybrid striped bass to determine the nutrient digestibility and apparent amino acid availability of six common feedstuffs. The animal‐protein feedstuffs tested were menhaden fish meal (MEN), anchovy fish meal (ANCH), pet‐food grade poultry by‐product meal (PBM‐pet), and feed‐grade poultry by‐product meal (PBM‐feed), while the plant‐protein feedstuffs were dehulled solvent extracted soybean meal (SBM) and distiller’s dried grains with solubles (DDGS). Test diets consisted of a 70 : 30 mixture of reference diet to test ingredient with chromic oxide (10 g kg?1%) as the inert marker. Diets were randomly assigned to triplicate tanks of fish that were fed their respective diets for 7 days prior to fecal collection by stripping. Two feeding trials were conducted sequentially to determine the digestibility of the six test ingredients. In trial 1, the three ingredients evaluated were MEN, PBM‐feed, and PBM‐pet. In trial 2, the three ingredients evaluated were ANCH, SBM, and DDGS. Apparent digestibility coefficients of protein (ADC‐CP) were significantly (P < 0.05) different among test ingredients in trial 1 as protein digestibility of MEN (86%) was significantly higher (P < 0.05) than that of PBM‐feed (75%), but was not significantly different from that of PBM‐pet (78%). Protein digestibilities in trial 2 were not significantly different among test ingredients and averaged 76% for ANCH, SBM, and DDGS. Some apparent amino acid availability coefficients differed among feedstuffs for both trial 1 and trial 2. MEN provided higher amino acid availabilities for alanine, aspartic acid, glutamic acid, lysine, valine, and tryptophan (99%, 98%, 94%, 96%, 99%, and 108%, respectively) when compared to PBM‐feed (73%, 50%, 69%, 80%, 77%, and 91%, respectively) and PBM‐pet (79%, 66%, 81%, 81%, 78%, and 99% respectively). Glycine, histidine, leucine, and proline availabilities in MEN (95%, 96%, 100%, and 98%, respectively) were significantly (P < 0.05) higher than those of PBM‐feed (64%, 82%, 82%, and 57%, respectively), but were not significantly different from PBM‐pet (85%, 92%, 89%, and 80%, respectively). For trial 2, apparent amino acid availabilities for cystine, isoleucine, lysine, and tyrosine were significantly higher (P < 0.05) among treatments fed SBM (100%, 87%, 93%, and 97%, respectively) and ANCH (37%, 95%, 92%, and 84%, respectively) compared to treatments fed DDGS (?13%, 52%, 62% and 62%, respectively). Overall, amino acid availability in SBM and the two PBM’s appear comparable to MEN and ANCH and corroborate their high value as potential replacements for fish meal in sunshine bass diets. Wild striped bass,Morone saxatilis, were collected from coastal waters and spawning areas to describe the endocrine correlates of oocyte development in non-captive, migratory fish. The fish were classified according to their most advanced oocytes. Serum levels of estradiol (E2), testosterone (T) and 17-20-dihydroxyprogesterone (DHP) were measured by radioimmunoassay (RIA). Females in the primary growth phase and early secondary growth phase (pre-vitellogenic) had low levels of plasma steroids, ovarian lipid content and gonadosomatic indices (GSIs). Significant increases in E2, T, ovarian lipid content and GSIs occurred during the vitellogenic phase. Maximum levels of all reproductive parameters were found in prespawning fish sampled in the Hudson River. Mean levels of E2, T, ovarian lipids and GSIs for these fish were 2.0±0.5 ng/ml, 3.0±0.3 ng/ml, 24±1 mg/g, and 5.6±0.3% (mean±SEM), respectively. In fish induced to spawn with human chorionic gonadotropin (HCG), DHP levels (1.9±0.4 ng/ml) were significantly elevated. Similar levels were found in two fish captured during the spawning season, suggesting that DHP may serve as the maturation-inducing steroid in this species. 相似文献

A previously developed monoclonal antibody (mAb) specific for the heavy chain of hybrid striped bass (HSB) Morone chrysops×M. saxatilis immunoglobulin was used in an assay to detect the humoral response to antigens of Streptococcus iniae. In order to validate this assay, an anti‐S. iniae antibody was produced in HSB by immunization with formalin‐killed cells of S. iniae mixed with Freund's complete adjuvant (FCA). After boosting with cells mixed with Freund's incomplete adjuvant (FIA), fish were challenged with live S. iniae by i.p. injection. This resulted in mortalities of 100%, 13% and 7% for non‐immunized, immunized and non‐challenged fish respectively. Live S. iniae were recovered only from moribund non‐immunized challenged fish, indicating that the immunization conferred protection against S. iniae. Sera were taken at 28 and 42 days post challenge and anti‐S. iniae antibody was measured by both agglutination and indirect ELISA. Titres of anti‐S. iniae antibody increased only in the immunized group as measured by agglutination and ELISA. Serum complement measured in the final bleeding was significantly higher in the immunized group. Western blotting using immune HSB serum indicated that the predominant antigen in this case was the high molecular weight polysaccharide of S. iniae. 相似文献

Two concurrent 12-week feeding trials were conducted to evaluate the bioavailability of inorganic sodium selenite and organic seleno-DL-methionine and to investigate the potential interaction between selenium and vitamin E in juvenile hybrid striped bass. In experiment 1, purified diets utilizing casein, gelatin and an amino acid premix as protein sources with a basal selenium concentration of 0.11 mg Se kg−1 were supplemented with either Na2 SeO3 to provide selenium concentrations of 1.19, 2.00, 5.17 and 21.23 mg Se kg−1 or with seleno-DL-methionine to provide 0.90, 1.26 and 2.55 mg Se kg−1 and fed to juvenile hybrid striped bass in aquaria. A second experiment evaluated potential interactions by feeding these purified diets with or without supplemental vitamin E or sodium selenite, singularly or in combination. No overt selenium deficiency signs were exhibited by fish in either of the experiments; however, signs of selenium toxicity including retarded weight gain (WG), reduced feed intake and feed efficiency ratio (FER) as well as increased mortality, were observed in fish fed the diet containing more than 20 mg Se kg−1 . Whole-body selenium and whole-body selenium retention were linearly influenced by sodium selenite and selenomethionine. However, there was no significant effect of dietary selenium, vitamin E or their interaction on WG, FER and survival. Slope-ratio analysis showed that bioavailability of seleno-DL-methionine as a selenium source for juvenile hybrid striped bass was significantly ( P < 0.01) higher (3.3-fold) than sodium selenite. 相似文献
