Using the thiocholine method and histological techniques the myenteric plexuses of pigeon and hen were studied. Investigations revealed the presence of a nervous network in the wall of the small intestine of both animals. It consists of many nerve fibres crossing each other and creating meshes in a variety of shapes. The density of the network was different according to the species and to the parts of intestine. The myenteric plexus from the pigeon's duodenum is thicker (3.7-fold) than in the remaining part of the small intestine; in the hen this is approximately 2.2-fold thicker. The meshes of the duodenum in both species are smaller than in the jejunum and ileum. The results of histological investigations showed different localization of myenteric plexuses; in pigeon in the space between the circular and longitudinal muscle layers, and in hen within the circular muscle.  相似文献

Development of mechanistic models of digestion is a means to account for interactions between different nutrients, and between nutrients and the animal. Although some digestion models for pigs have been proposed, quantitative information concerning the underlying processes is extremely limiting. The aim of this study was to determine the effect of dietary fibre on the transit of the solid and liquid phases of digesta in different segments of the gastro-intestinal tract (GIT) in growing pigs. Six barrows (initial BW 30 kg and fitted with two simple T-cannulas at the proximal duodenum and distal ileum) were used in a double 3 × 3 Latin square design. Pigs were offered diets differing in total dietary fibre content (17, 22 and 27%) at 4 h intervals. A single meal marked with YbO2 and Cr-EDTA was used to determine the passage kinetics of respectively the solid and liquid phases of digesta in different segments of the GIT.

The mean retention time (MRT) averaged 1 h from the mouth to the proximal duodenum (stomach), 4 h from the proximal duodenum to the distal ileum (small intestine), and 35 h from the distal ileum to the faecal excretion (large intestine). Increase in dietary fibre content had no effect on MRT in the stomach, tended to decrease MRT of solid phase in the small intestine (P = 0.07), and decreased MRT of both phases in the large intestine (43 to 29 h, P = 0.06). For all diets, transit of the liquid phase in the large intestine was 4–8 h less than transit of the solid phase.  相似文献

The effects of 5-hydroxytryptamine (5-HT), HTF 919, a new 5-HT(4) agonist, and the antagonists SB 203-186 (5-HT(4)) and tropisetron (5-HT(3)) on intestinal motility were tested in vitro on isolated preparations of horse ileum and pelvic flexure. Concentration-response curves were created by cumulative application of the agonists with or without preincubation of the antagonists. The 5-HT preparation induced a concentration-dependent contraction in equine ileum and pelvic flexure. The results indicate that 5-HT receptors are present in all parts of equine intestine investigated in this study. Tropisetron was found to act as a noncompetitive antagonist in all locations of the equine intestine. SB 203-106 was confirmed as an antagonist to 5-HT in the equine ileum circular muscle, in pelvic flexure circular and longitudinal muscle. Nevertheless, a discernible increase of smooth muscle contractions caused by HTF 919 could only be observed in pelvic flexure. In accordance with an earlier study in the guinea pig, in the equine gut HTF 919 acted as a partial agonist for the 5-HT(4) receptor with an affinity constant in the nanomolar range. It is concluded that 5-HT receptors, and especially their subtypes, may represent a promising target for the treatment and prevention of gastrointestinal (GI) motility disorders in horses.  相似文献

The histology of the canine intestine has not been accurately defined. To establish the precise thickness of its different layers, whole wall samples of the small intestine were removed from 41 cadavers at five standardised sampling sites (duodenum, proximal jejunum, distal jejunum, proximal ileum and distal ileum). The total thickness was estimated by morphometry, as was the thickness of the mucosa, muscularis mucosae, submucosa and muscularis externa. In addition, the size of the lymphoid aggregates in the submucosa and the thickness of the circular and longitudinal layers within both the muscularis mucosae and the muscularis externa were estimated. The total intestinal thickness depended very much upon the thickness of the mucosa and submucosa. The mucosa decreased progressively from proximal to distal parts of the small intestine (47% reduction). The thickness of the submucosa, however, changed little from the duodenum to the distal jejunum, but increased significantly in the ileum; this change was positively correlated with the amount of lymphoid tissue. Sex influenced the thickness of the intestinal wall, with males displaying higher thickness values along the small intestine. Conversely, no correlation between bodyweight and intestinal thickness was found for any of the five sampling sites. This study gives absolute and relative values for the thickness of the layers of the dog intestine which might help in the diagnosis of small intestinal pathology from postmortem samples and/or endoscopic biopsies.  相似文献

In order to explore the influence of temperature changes on animal digestive tract smooth muscle,the contractility of isolated rabbit small intestine (duodenum,jejunum and ileum) were measured at different temperatures (39→32℃,32→39℃,39→42℃ and 42→39℃) using the biological signal processing system.The results showed that the contraction frequency of duodenum and jejunum was positively correlated with temperature changes between 39 and 32℃,while the amplitude and tension of duodenum were negatively correlated with temperature changes.Only the contraction frequency of ileum was positively correlated with temperature changes between 39 and 32℃.Changes of frequency,amplitude and tension of duodenum between 39 and 42℃ were similar with those changes between 39 and 32℃.However,the contraction of jejunum and ileum was irregular.When the temperature was decreased from 42 to 39℃,the contraction of small intestine could not return to the condition in 39℃.The results suggested that the optimal bath temperature was 37℃ in contraction test of rabbit small intestine in vitro,and the present results could be referenced to explore the disorder of gastrointestinal movement due to the change of environment temperature in animals.  相似文献

为了探究温度变化对动物消化道平滑肌运动的影响,本试验用生物信号处理系统测定了浴槽不同温度(39→32℃、32→39℃、39→42℃和42→39℃)对家兔离体小肠各段(十二指肠、空肠、回肠)平滑肌运动的影响。结果显示,当温度在39和32℃之间变化时家兔离体十二指肠和空肠运动的收缩频率同温度变化呈正相关,而十二指肠收缩幅度和收缩张力同温度变化呈负相关,但回肠仅表现在收缩频率同温度变化呈正相关;当温度从39℃升高到42℃时,十二指肠的收缩频率、收缩幅度和收缩张力变化与温度在39和32℃之间变化情况相同,但是空肠和回肠的规律性较差;当温度从42℃高温降低到39℃时,3段离体小肠运动不能恢复到39℃时的运动状态,家兔小肠离体试验浴槽最适温度为37℃。本研究结果提示,在进行动物小肠离体试验时应注意浴槽温度对小肠运动的影响,同时本研究结果也为探究由于环境温度变化导致的动物胃肠运动异常的机理提供参考。  相似文献

We report here the interesting case of a 5-year-old male cynomolgus monkey with goblet cell hyperplasia and thickening of the muscular layer throughout the small intestine without exhibiting any clinical symptoms. Necropsy examination showed diffuse thickening of the intestinal wall from the jejunum to the ileum, with an appearance likened to a rubber tube. Histopathologically, marked thickening was observed in both the mucosal and muscular layers in the jejunum and ileum, and slight thickening was observed in the duodenum. Goblet cell hyperplasia with extension of the circular folds and villi was prominently observed. The mucosal surface was covered with a thick mucus layer containing desquamated mucosal epithelial cells, and both the inner and outer muscular layers were markedly thickened due to smooth muscle hypertrophy. Neither macroscopic nor histopathological examination identified any causative factors, such as infection, enteritis and intestinal stenosis, or obstruction that may have caused development of this lesion. Given these observations, this case may simply be considered of spontaneous goblet cell hyperplasia and muscular layer thickening in the small intestine of a cynomolgus monkey.  相似文献

REASONS FOR PERFORMING STUDY: Prokinetic drugs used to treat gastrointestinal ileus in man have equivocal results in horses. In man, prokinetic drugs have 5-hydroxytryptamine4(5-HT4) receptors as their target, but little is known about the 5-HT-receptor subtypes in the equine small intestine. OBJECTIVE: Functional and immunohistochemical identification of the serotonin receptor subtype(s) responsible for the 5-HT induced contractile response in the equine circular jejunum. METHODS: Isometric organ-bath recordings were carried out to assess spontaneous and drug-evoked contractile activity of equine circular jejunum. Histological investigations by immunofluorescence analyses were performed to check for presence and localisation of this functionally identified 5-HT receptor subtype. RESULTS: Tonic contractions were induced by 5-HT in horse jejunal circular muscle. Tetrodotoxin, atropine and NG-nitro L-arginine did not modify this response. A set of 5-HT receptor subtype selective antagonists excluded interaction with 5-HT1B, 1D, 2A, 3, 4 and 7 receptors. The selective 5-HT1A receptor antagonists WAY 100635 and NAN 190 caused a clear rightward shift of the concentration-response curve to 5-HT. The contractile effect of 5-CT, that can interact with 5-HT1A, 1B, 1D, 5 and 7 receptors was also antagonised by WAY 100635, identifying the targeted 5-HT receptor as a 5-HT1A-like receptor. Immunohistology performed with rabbit polyclonal anti-5-HT1A receptor antibodies confirmed the presence of muscular 5-HT1A receptors in the muscularis mucosae, and both longitudinal and circular smooth muscle layers of the equine jejunum. CONCLUSIONS: Contractile responses in equine jejunal circular smooth muscle induced by 5-HT involves 5-HT1A-like receptors.  相似文献

REASON FOR PERFORMING STUDY: Tachykinins have profound effects on equine intestinal motility, but the distribution of the neurokinin receptors (NKRs) through which they act is unknown. This study reports the distribution of one of these receptors, the neurokinin-1 receptor (NK1R), in smooth muscle throughout the equine intestinal tract. OBJECTIVES: To quantify the distribution of the NK1R, based upon mRNA expression, in smooth muscle of different regions of the equine intestinal tract. METHODS: Nine regions of the intestinal tract were sampled in 5 mature horses. Total RNA was isolated from smooth muscle and reverse transcribed; NK1R mRNA was then quantified using real-time PCR. RESULTS: NK1R mRNA was found at all levels of the sampled intestinal tract. The smooth muscle of the proximal small intestine and the ventral colon exhibited the highest level of NK1R mRNA expression in the equine intestinal tract. CONCLUSIONS: Tachykinins probably affect intestinal contractility and propulsion in the proximal small intestine and in the ventral colon.  相似文献

试验旨在研究表达耐热肠毒素(STa)的重组大肠杆菌对7日龄仔猪肠道形态结构及抗氧化功能的影响。选取24头7日龄仔猪,随机分在4个日粮处理组,分别为对照组(人工乳),STa组(人工乳+2×109 CFU重组菌LMG194-pBAD-STa),LMG194组(人工乳+2×109 CFU大肠杆菌LMG194),K88组(人工乳+2×109 CFU大肠杆菌K88)。试验第5天进行攻毒,第7天屠宰取样,测量小肠黏膜的绒毛高度和隐窝深度,检测空肠、回肠、结肠及血清中的过氧化氢酶(CAT)、总一氧化氮合成酶(TNOS)与诱导型一氧化氮合酶(iNOS)的活力及血清中丙二醛(MDA)与过氧化氢(H2O2)的含量。试验结果表明,与对照组相比,STa组仔猪各肠段绒毛高度均显著降低(P<0.05),十二指肠隐窝深度及绒毛高度与隐窝深度的比值显著降低(P<0.05),十二指肠、空肠、回肠绒毛表面积显著降低(P<0.05);同时,STa组回肠、结肠CAT活力显著降低,血清、空肠、回肠和结肠中的TNOS活力显著降低(P<0.05),STa组结肠iNOS活力显著降低(P<0.05),STa组血清中的MDA与H2O2含量显著提高(P<0.05)。结果显示,表达STa的重组大肠杆菌可导致7日龄仔猪肠道结构损伤和抗氧化能力下降。  相似文献

This experiment was conducted to investigate the effects of recombinant E.coli expressing heat-stable enterotoxin(STa) on intestinal absorption and barrier function, intestinal morphology and antioxidant capacity of 7 days old piglets. Twenty-four 7 days old piglets were allotted to four treatments:control group (artificial milk), STa group (artificial milk +2×109 CFU E.coli LMG194-pBAD-STa), LMG194 group (artificial milk +2×109 CFU E.coli LMG194), and K88 group (artificial milk +2×109 CFU E.coli K88).The pigs were treated with E.coli on the 5th day and slaughtered on the 7th day. The results showed that, compared with the control group, villus height in jejunum, ileum and duodenum, crypt depth and villus height/crypt depth in duodenum, and small intestine villi surface area were significantly decreased in STa group (P<0.05). The activities of catalase (CAT) in ileum and colon, total nitric oxide synthase (TNOS) in serum, ileum, jejunum and colon, inducible nitric oxide synthase (iNOS) in colon were significantly decreased in STa group (P<0.05). STa group also had a higher levels of malondialdehyde (MDA) and hydrogen peroxide (H2O2) in serum (P<0.05). These results suggested that recombinant E.coli expressing STa could lead to intestinal injury and oxidative stress of 7 days old piglets.  相似文献

The effect of medetomidine, a potent and highly selective α2-adrenoceptor agonist, on the motility of the gastric antrum, duodenum, mid-jejunum and ileum was investigated in ten dogs. Its effect on the release of gastrin was also determined. Administration of medetomidine intramuscularly (i.m.) at a dose of 40 μg/kg inhibited the motility of the gastric antrum, duodenum, mid-jejunum and ileum significantly, in comparison to administration of xylazine intramuscularly at a dose of 2.0 mg/kg. The release of gastrin was also significantly decreased in dogs receiving medetomidine. It was found to inhibit the motility in the gastric antrum and duodenum longer than in the mid-jejunum and ileum, presumably by acting specifically on α2-adrenoceptors, likely at the peripheral level. Medetomidine also inhibited the gastric contraction associated with gastrin secretion.  相似文献

【目的】 在从腹泻仔猪粪便中分离鉴定猪A群轮状病毒(Rotavirus group A, RVA)并了解其致病性。【方法】 应用MA-104细胞从仔猪腹泻粪便中分离鉴定RVA, 将其经口感染健康初生仔猪, 观察其临床症状, 并利用实时荧光定量PCR检测排毒、HE染色观察病理变化、免疫组织化学(immunohistochemistry, IHC)试验了解病毒分布。【结果】 分离到1株可引起MA-104细胞明显细胞病变的G9P[23] RVA, 命名为RVA/Pig-tc/CHN/SCJY-13/2017/G9P[23](简称SCJY-13株), 病毒滴度为105.5 TCID50/100 μL。经口感染SCJY-13株的仔猪在感染后11 h出现腹泻, 持续到感染后165 h完全恢复, 其发病率为100%(7/7), 病死率为28.57%(2/7)。感染后8 h可从仔猪肛拭子检测到病毒核酸, 直至感染后192 h, 峰值出现在感染后24 h。感染后54 h各段小肠病毒载量达到最高, 其中回肠中病毒载量最高, 显著高于十二指肠、空肠(P<0.05);HE染色和IHC检测结果显示, SCJY-13在感染仔猪小肠的绒毛及隐窝中大量聚集, 尤其是回肠段; SCJY-13株感染引起十二指肠、空肠和回肠绒毛固有层大量淋巴细胞浸润、黏膜上皮细胞和肠绒毛尖端空泡变性、柱状细胞增多、绒毛断裂脱落等, 以回肠段较严重。【结论】 SCJY-13株能引起新生仔猪100%发病, 其主要靶位区在回肠, 感染后排毒时间长。本试验结果为猪RVA的致病性研究提供了一定的参考依据。  相似文献

The aim of the present study was to clarify the participation of endogenous arachidonic acid (AA) metabolites in regulating porcine basilar, coronary, pulmonary and mesenteric arterial tones in vitro . A cyclooxygenase inhibitor, indomethacin, relaxed basilar artery but not other arteries examined. Quinacrine (a phospholipase A2 inhibitor), OKY-046 (a thromboxane (TX) A2 synthetase inhibitor) and ONO-3708 (a TXA2/prostaglandin H2 receptor antagonist) produced relaxation in basilar arteries with intact endothelium. Nordihydroguaiaretic acid (a lipoxygenase inhibitor) had no effect on the tone. The amount of TXB2 (a stable metabolite of TXA2) spontaneously released from porcine basilar arteries was 6–10 fold more than those from other arteries. Indomethacin and OKY-046 mostly inhibited the production of TXB2. Endothelial denudation decreased indomethacin-induced relaxation and the amount of TXB2. These results suggest that a vasoconstricting substance(s) is released from endothelial cells and possibly smooth muscle cells in porcine basilar arteries in vitro . The main constricting substance is proposed to be TXA2. On the other hand, several arteries from peripheral vascular beds did not release this vasoconstricting substance.  相似文献

旨在探讨猪德尔塔冠状病毒(PDCoV)感染仔猪对小肠杯状细胞(goblet cell,GC)的影响,将6头未吃初乳的初生仔猪静养2 d后随机分为感染组(n=3)和对照组(n=3),感染组仔猪口服接种5 mL 1×105 TCID50·mL-1 PDCoV-CHN-HG-2017毒株,对照组仔猪口服5 mL DMEM培养基。感染组仔猪分别在口服接种后22、39和70 h出现明显腹泻、脱水和嗜睡等临床症状,及时实施安乐死并采集小肠组织样品;对应时间点分别选取对照组1头仔猪实施安乐死并采集小肠组织样品。采用HE染色、PAS染色和AB染色观察小肠组织病理学变化和GC数量变化。采用荧光定量PCR和ELISA分别检测初生仔猪小肠组织中发状分裂相关增强子1(Hes1)和黏蛋白2(MUC2)的转录和含量的变化。结果显示,感染组仔猪与对照组仔猪相比小肠各段黏膜结构均有不同程度损伤,绒毛长度降低且差异显著(P<0.05),VH:CD的比值降低且差异显著(P<0.05)。PAS染色与AB染色结果一致,小肠各段GC数量均有不同程度减少且差异显著(P<0.05、P<0.01或P<0.001)。感染组仔猪与对照组仔猪相比小肠各段Hes1 mRNA的转录量和含量均升高,在空肠和回肠中Hes1 mRNA转录量差异显著(P<0.01或P<0.05),在十二指肠和回肠中Hes1含量差异显著(P<0.01)。感染组仔猪与对照组仔猪相比小肠各段MUC2 mRNA的转录量和含量均降低,在十二指肠和回肠中差异显著(P<0.05、P<0.01或P<0.001)。结果表明,PDCoV感染初生仔猪后引起小肠GC数量明显减少。PDCoV感染仔猪可能通过激活肠道Notch信号通路下游靶基因Hes1表达来阻碍小肠中GC形成和分泌,导致GC数量减少和MUC2表达降低。  相似文献

OBJECTIVES: To determine the in vitro effect of prostaglandin E2 (PGE2), PGF2alpha, PGI2; and nonsteroidal anti-inflammatory drugs (NSAID; ie, flunixin meglumine, ketoprofen, carprofen, and phenylbutazone) on contractile activity of the equine dorsal colon, ventral colon, and pelvic flexure circular and longitudinal smooth muscle. ANIMALS: 26 healthy horses. PROCEDURE: Tissue collected from the ventral colon, dorsal colon, and pelvic flexure was cut into strips and mounted in a tissue bath system where contractile strength was determined. Incremental doses of PGE2, PGF2alpha,, PGI2, flunixin meglumine, carprofen, ketoprofen, and phenylbutazone were added to the baths, and the contractile activity was recorded for each location and orientation of smooth muscle. RESULTS: In substance P-stimulated tissues, PGE2 and PGF2alpha enhanced contractility in the longitudinal smooth muscle with a decrease or no effect on circular smooth muscle activity. Prostaglandin I2 inhibited the circular smooth muscle response with no effect on the longitudinal muscle. The activity of NSAID was predominantly inhibitory regardless of location or muscle orientation. CONCLUSIONS AND CLINICAL RELEVANCE: In the equine large intestine, exogenous prostaglandins had a variable effect on contractile activity, depending on the location in the colon and orientation of the smooth muscle. The administration of NSAID inhibited contractility, with flunixin meglumine generally inducing the most profound inhibition relative to the other NSAID evaluated in substance P-stimulated smooth muscle of the large intestine. The results of this study indicate that prolonged use of NSAID may potentially predispose horses to develop gastrointestinal tract stasis and subsequent impaction.  相似文献

In horses, gastrointestinal (GI) disorders occur frequently and cause a considerable demand for efficient medication. 5-Hydroxytryptamine receptors (5-HT) have been reported to be involved in GI tract motility and thus, are potential targets for treating functional bowel disorders. Our studies extend current knowledge on the 5-HT7 receptor in equine duodenum, ileum and pelvic flexure by studying its expression throughout the intestine and its role in modulating contractility in vitro by immunofluorescence and organ bath experiments, respectively.5-HT7 immunoreactivity was demonstrated in both smooth muscle layers, particularly in the circular one, and within the myenteric plexus. Interstitial cells of Cajal (ICC), identified by c-Kit labeling, show a staining pattern similar to that of 5-HT7 immunoreactivity.The selective 5-HT7 receptor antagonist SB-269970 increased the amplitude of contractions in spontaneous contracting specimens of the ileum and in electrical field-stimulated specimens of the pelvic flexure concentration-dependently.Our in vitro experiments suggest an involvement of the 5-HT7 receptor subtype in contractility of equine intestine. While the 5-HT7 receptor has been established to be constitutively active and inhibits smooth muscle contractility, our experiments demonstrate an increase in contractility by the 5-HT7 receptor ligand SB-269970, suggesting it exerting inverse agonist properties.  相似文献

Prostaglandin F receptors (PGF Rs) were measured in bovine corpus luteum and myometrial cell membranes using a radiometric method. The inhibition of labelled PGF binding exerted by d-cloprostenol, dl-cloprostenol, PGF and PGE1 (l0–11 M to 10–4 M) was evaluated in vitro. Results strongly suggest that cloprostenol binding to PGFRs is stereospecific. d-Cloprostenol and PGF were equipotent, about 150 times more potent than d-cloprostenol (P < 0.05) and approximately 280 times more potent than PGE, (P < 0.05) in inhibiting [3H]PGF binding to corpus luteum cell membranes. Such differences were less evident in myometrial cell membranes, where d-cloprostenol and PGF were about 10 times more potent than d-cloprostenol (P < 0.05) and approximately 95 times more potent than PGEl (P < 0.05).  相似文献

Isolated sheep lung parenchymal strips responded to histamine > carbachol > PGF2a > 5-HT with contractions, and to isoproterenol (Isop), and to large doses of epinephrine (E), norepinephrine (NE) and phenylephrine (PE) with relaxations. PGF2a-contracted lung strip responded to PGE1 and PGE2 with relaxation. The strips which were partially contracted to histamine, PGF2a, 5-HT and carbachol also responded to isop, E and NE with relaxations. Histamine responses were not modified by metiamide (an H2-receptor antagonist). Mepyramine and atropine selectively antagonized contractions to histamine and carbachol, respectively. After β-blockade with propranolol, lung strips responded to NE > E > PE > isop with contractions, which were inhibited or reversed by phentolamine and dibenzyline. It is concluded that H1 receptors are present in sheep peripheral airway smooth muscles, and that a- and β-adrenoceptors mediate contraction and relaxation, respectively, in sheep lung strips.  相似文献

对小尾寒羊羔羊小肠不同部位(十二指肠、空肠和回肠)pH及淀粉酶、胰蛋白酶、糜蛋白酶、脂肪酶和乳糖酶的活性进行了测定,结果表明:羔羊小肠不同部位内容物pH不同,回肠pH显著高于空肠,空肠和回肠pH显著高于十二指肠,羔羊1月龄后,小肠各段pH变化不大;小肠不同部位淀粉酶和胰蛋白酶、糜蛋白酶及脂肪酶活性不同,空肠段淀粉酶、胰蛋白酶、糜蛋白酶及脂肪酶活性高于十二指肠和回肠段,随着羔羊日龄的增长,小肠淀粉酶、胰蛋白酶、糜蛋白酶及脂肪酶活性增加,3月龄后各消化酶活性变化差异不显著(P<O.05);羔羊小肠不同部位乳糖酶活性不同,空肠乳糖酶活性显著高于十二指肠和回肠,随着羔羊日龄的增长,小肠乳糖酶活性逐渐降低。  相似文献
