The effect of catalase on the toxicity of cadmium (Cd) in Cd-resistant Staphylococcus aureus 3719+ and its plasmid-negative Cd-sensitive variant 3719– was studied. Catalase on a solid medium increased the recovery of Gd-stressed S. aureus 3719– cells, and the addition of catalase into a liquid growth medium resulted in a shortened lag phase of growth especially in S. aureus 3719–. The catalase activity of S. aureus 3719+ cell suspensions was greater than the corresponding activity of S. aureus 3719– cell suspensions. Cd did not influence the activity of beef liver catalase or the catalase production of the bacterial cells.Catalase reduced the toxicity of Cd especially for S. aureus 3719–. The greater catalase production of S. aureus 3719+ might be one factor in its resistance against the toxic effects of Cd. It is suggested that Cd together with hydrogen peroxide may induce oxidative damage to cells if there is not sufficient catalase available to decompose all the hydrogen peroxide formed.  相似文献

Cysteine was shown to have a protective effect on the killing rate caused by cadmium (Cd) but no effect on the uptake of Cd in S. aureus 3719-. The addition of 254 × 10−3 mmol/l of sodium selenite or more into the liquid growth medium increased the lag phase of growth of S. aureus 3719-. Sodium selenite had no protective effect on the toxicity of Cd in S. aureus 3719- as evaluated by the length of the lag phase of growth in the presence of Cd and varying amounts of sodium selenite. 534 × 10−6 mmol/l of sodium selenite in the growth medium had no effect on the uptake of Cd in S. aureus.  相似文献

The accumulation and distribution of cadmium (Cd) between cellular subfractions were studied in Cd-resistant Staphylococcus 3719 + and its plasmid-negative Cd-sensitive variant 3719–, grown in media containing 0.01, 0.1 or 50 mg of Cd/litre. In the media containing 0.01 and 0.1 mg of Cd/1 the uptake of Cd by the Gd-resistant S. aureus was significantly less than that by Cd-sensitive S. aureus, both in toto and in terms of the cell wall fractions. Only variant 3719 + grew in the medium containing 50 mg of Cd/1. The finding of a lag phase in the growth of both variants is interpreted to suggest that the resistance is of inducible character, even in the sensitive S. aureus. The mechanism of resistance to Cd could consist of both barrier and clearance functions.  相似文献

Vitamin E deficiencies can impact normal growth and development in humans and animals, and assessment of circulating levels of vitamin E and its metabolites may be an important endpoint for evaluation. Development of a sensitive method to detect and quantify low concentrations of vitamin E and metabolites in biological specimens allows for a proper diagnosis for patients and animals that are deficient. We developed a method to simultaneously extract, detect, and quantify the vitamin E compounds alpha-tocopherol (α-TP), gamma-tocopherol (γ-TP), alpha-tocotrienol (α-TT), and gamma-tocotrienol (γ-TT), and the corresponding metabolites formed after β-oxidation of α-TP and γ-TP, alpha-carboxymethylbutyl hydroxychroman (α-CMBHC) and alpha- or gamma-carboxyethyl hydroxychroman (α- or γ-CEHC), respectively, from equine plasma and serum. Quantification was achieved through liquid chromatography–tandem mass spectrometry. We applied a 96-well high-throughput format using a Phenomenex Phree plate to analyze plasma and serum. Compounds were separated by using a Waters ACQUITY UPLC BEH C18 column with a reverse-phase gradient. The limits of detection for the metabolites and vitamin E compounds were 8–330 pg/mL. To validate the method, intra-day and inter-day accuracy and precision were evaluated along with limits of detection and quantification. The method was then applied to determine concentrations of these analytes in plasma and serum of horses. Alpha-TP levels were 3–6 µg/mL of matrix; the metabolites were found at much lower levels, 0.2–1.0 ng/mL of matrix.  相似文献

Four dogs with poor semen quality, low seminal plasma superoxide dismutase (SOD) activity and low blood plasma testosterone (T) levels were orally administered one vitamin E tablet containing 50 mg α-tocopheryl acetate per dog daily for 4 weeks. The mean values of semen quality were temporarily improved after the start of vitamin E treatment and the values of 4, and 5 weeks after that were significantly different from those before the treatment (P<0.05–0.001). The mean blood plasma T and seminal plasma SOD activity values slightly increased in the 4 dogs after the treatment. The results of the present study indicate that poor semen quality in dogs with low seminal plasma SOD can be improved by vitamin E treatment.  相似文献

A randomized blocked factorial experiment was conducted with 90 young pigs. From 4 to 11 weeks of age the pigs were kept in individual pens and fed a selenium supplemented basal diet consisting mostly of propionic acid treated barley, soybean meal and dried skim milk, and containing < 0.5 mg vitamin E per kg. The treatment factors during this period were 3 dietary levels of added vitamin E (nil, 10 and 30 mg/kg) and a 6 % supplement of fresh or oxidized fat (2/3 lard and 1/3 herring oil). From 11 weeks of age until slaughter at 90 kg the pigs received the vitamin E supplements but no fat or dried skim milk. The basal diet for this later experimental period was based on untreated dry barley. Blood samples collected during the period of investigation were examined for vitamin E and for resistance against erythrocyte lipid peroxidation (ELP) in order to evaluate the antioxidant status.Analysis of variance and Student’s t-test on least squares means showed the ELP to be influenced independently by the vitamin E supplement and, during the fat feeding period, by the quality of the fat supplement, with the highest peroxidation resistance (low ELP) in the groups fed fresh fat and a high level of vitamin E. Blood vitamin E level was only influenced — positively — by the vitamin E supplement although variations in the feed vitamin E level below 10–15 mg vitamin E per kg did not result in corresponding variations in measurable blood vitamin E concentrations. In the same low range of vitamin E in the feed there was a statistically significant difference in ELP values between the different vitamin E treatment groups. No clinical manifestations of selenium-vitamin E deficiency were observed in the pigs.The ELP and the plasma vitamin E levels observed would seem to suggest that a total of 15 mg vitamin E per kg barley-based feed will not always be sufficient for growing pigs.  相似文献

As a result of research conducted in the US, recommendations for dry cow vitamin E intakes have increased seven fold there, however there has been no change to recommendations in the UK. As part of a larger study comparing the impact of existing UK and new US recommended vitamin E intakes on the health and fertility of commercial dairy cows in the UK, a study was set up to investigate the effect of route of supplementation and stage of lactation, over a 21 day period, on the response to mega-supplementation of cattle receiving supposedly adequate vitamin E. The study assessed the response of dry, peak lactation and mid lactation cows to in-feed or parenteral vitamin E supplementation (7 animals per treatment/lactation stage group) by measuring plasma and milk vitamin E concentrations, blood glutathione peroxidase (GSH-Px) activity and milk yields over a 21 day period. Plasma vitamin E concentrations were significantly influenced by a time, stage and treatment interaction (P = 0.046). Both dry and lactating animals had significantly higher plasma vitamin E concentrations at some time points in the parenteral supplemented cows compared to the in-feed supplementated animals (P ≤ 0.011 and P < 0.01, respectively). Milk vitamin E concentrations did not significantly differ between lactation stages but treatment had a significant effect on concentrations (P < 0.008) when lactation stage was removed from the model. There was no significant difference in milk yield between treatment groups. A significant relationship between plasma and milk vitamin E concentrations was only found in the parenterally supplemented cows (r = 0.435, P < 0.001). In cattle with intakes greater than the ARC recommendations, measurement of plasma vitamin E concentration may be of limited value in determining whether there has been a response to supplementation. The relationship between plasma and milk vitamin E concentrations is too poor for milk vitamin E concentrations to be used as a proxy for plasma vitamin E.  相似文献

An increase in the pH of Plate Count Agar led to increased toxicity of cadmium (Cd) for Micrococcus luteus ATCC 9341, Staphylococcus aureus ATCC 12600, Clostridium perfringens ATCC 8798, Escherichia coli ATCC 11775 and Pseudomonas aeruginosa ATCC 10145. The effect of the pH on toxicity was not clearly observed with Streptococcus bovis ATCC 9809 and was absent with Bacillus subtilis ATCC 8473.When potassium phosphate buffer was added to the growth medium, the Cd toxicity for M. luteus and B. subtilis was enhanced. The toxicity of Cd for S. bovis decreased when potassium phosphate buffer was added to the medium. When the pH increased at differing phosphate concentrations, the sensitivity of M. luteus to Cd decreased.In the authors’ opinion, the effect of pH on Cd toxicity for bacteria is due to a continuously increasing negative charge towards an alkaline value by most bacteria which increases the affinity of cations towards the cell wall.  相似文献

The management of diabetes mellitus mandates measurement of blood glucose. Saliva offers an alternative to blood sampling, but measurement of the salivary glucose concentration is difficult, and the blood-to-saliva glucose time lag is uncertain. We aimed to determine the serum–saliva glucose time lag in the saliva of healthy dogs. The combined duct of the mandibular and sublingual salivary glands of 6 dogs was cannulated to collect saliva and prevent glucose degradation by oral bacteria. Following a 0.25 g/kg IV bolus of dextrose, paired serum–saliva samples were collected at baseline and in twelve 5-min blocks over 60 min. Serum and salivary glucose levels were analyzed with a linear mixed model for repeated measures with a compound symmetry error structure. Mean (±SD) saliva production was 10.3 ± 2.9 µL/kg/min, and the area under the curve (AUCglucose)saliva/serum ratio was 0.006, which highlights the magnitude of the large difference in glucose concentration between the 2 compartments. The supplementation of vitamin E on broiler chicken diets is essential to the prevention of lipid oxidation reactions in the meat and improvement of meat quality. The objective of this study was to assess the effect of different doses of dietary vitamin E on breast meat quality of broiler chickens in the finishing period. Five doses of vitamin E were used (30, 90, 150, 210, and 270 mg/kg feed) in broilers'' diets from 42 to 54 d of age. A completely randomized design was conducted, followed by a split-plot, where the vitamin E dose was considered as the whole plot, and broilers’ age at slaughter was the subplot. Breast meat quality was assessed at 4 different ages (45, 48, 51, and 54 d old), using 50 birds per age, totaling 200 birds. Meat quality characteristics evaluated were: pH at 24 h post mortem, color (brightness, redness, and yellowness), water holding capacity, cooking loss, shear force, and lipid peroxidation. There was no interaction between age and dose of vitamin E for meat quality characteristics (P > 0.05). The age at slaughter had a quadratic effect (P < 0.05) on pH, brightness, redness, and water holding capacity. Although pH values were higher in the breast meat of older birds (51 and 52 d old), breast meat of younger birds (48 d) had a more reddish aspect. Shear force value was higher in breast meat of birds slaughtered at later ages (P < 0.01), as a linear age-effect was observed. Brightness increased linearly (P < 0.05) with higher vitamin doses, whereas treatments did not alter yellowness, cooking loss, and lipid peroxidation. In this study, increasing vitamin E doses in the finishing period increased the brightness of broiler breast meat, whereas slaughtering at later ages resulted in greater meat pH and shear force value.  相似文献

In order to analyze the effect of vitamin E on Th1 and Th2 cytokine production, porcine peripheral blood mononuclear cells (PBMC) were isolated from healthy pigs (n = 8) and cultured with either 0, 10, 50, or 100 μM of vitamin E (α-tocopherol). PBMC were stimulated with PHA for either, 24 h to determine: (a) the concentration of tocopherol incorporated into the cell membrane, (b) cytokine production and (c) Th1 and Th2 regulators gene expression; or 72 h to determine the proliferation of PBMC. Vitamin E was incorporated into the PBMC in a dose dependent manner, giving as a result a high proliferation of cells irrespective of the dose of vitamin E used. Regarding cytokine production, vitamin E consistently decreases the mRNA expression and the percentage of cells producing IL-10. Vitamin E did not influence the production of IFN-γ but the lowest level of vitamin E (10 μM) was sufficient to maximally increase the proportion of cells producing IL-2, to diminish IL-4, and discreetly increase the mRNA expression of TBX21 vs. GATA3. In conclusion, our results revealed that vitamin E is able to suppress IL-10 production and to influence the production of IL-2, IL-4, and maybe TBX21. Vitamin E clearly has immunomodulatory effects, though further work in vivo to determine the physiological nature of these effects is warranted.  相似文献

Calcium (Ca) requirement increase rapidly by pregnant ewes at late gestation as a result of high fetal growth. Furthermore, Ca deficiency results in poor growth and bone development in growing newborn lambs. The aim of this study was to evaluate the effect of calcium supplementation (Calcium carbonate–CC) and vitamin AD3E injection (75,000 IU/shot) to pregnant Awassi ewes at late gestation (− 45 days prepartum) on their mineral levels and performance of their newborn lambs. Forty pregnant Awassi ewes at late gestation (65 ± 6.1 kg) were selected before parturition (− 45 days; using hormonal sponges for estrus synchronization) and randomly divided to 4 dietary treatments. The treatments were: group 1 Control (1.4% CC); group 2 (2.4% CC); group 3 (1.4% CC + 2 shots vitamin AD3E at − 45 and − 21 days prepartum); group 4 (2.4% CC + 2 shots vitamin AD3E at − 45 and − 21 days prepartum). Blood samples were collected from ewes (− 45, 0 days prepartum: 30 and 60 days postpartum) and lambs (0, 30 and 60 days old). Average milk yield and samples were collected. Body weights of lambs were taken at birth, 30 and 60 days old. Blood serum and milk samples were prepared and analyzed by AAS for Ca, Mg, Co, Mn and Cu concentrations.Calcium concentrations in bloods serum of ewes, 0 and 60 days postpartum, and lambs at weaning (60 days old) from group 4 were significantly (P < 0.05) higher compared to other groups. No significant effect (P > 0.05) of treatment on Co, Mg and Cu in blood serum of ewes and lambs except for Cu concentration in ewes. Moreover, a significant increase (P < 0.01) in milk inorganic% (30 and 60 days samples) in ewes from group 2 when compared with other groups. The average milk yield (AMY) of ewes from group 2 and 4 at late stage of lactation (45–60 days) were significantly higher (P < 0.05) compared to groups 1 and 3. In addition, Ca% in colostrums samples from ewes in groups 2 and 4 were significantly higher (P < 0.01). The average daily gain (ADG) and total body gain for lambs up to weaning (60 days old) were significantly higher (P < 0.05) in groups 1 and 2 compared with the other groups.In conclusion, increasing level of Ca intake in feed of Awassi ewes at late gestation, with or without vitamin AD3E injections, improve the calcium concentration in colostrums and blood serum of ewes and their lambs without negative effect in their health and performance.  相似文献

The antibacterial effect of lactoferrin (Lf) was tested on isolates of Escherichia coli (E. coli), Staphylococcus aureus (S. aureus), and coagulase-negative staphylococci (CNS) as well as on Pseudomonas aeruginosa (P. aeruginosa) and Klebsiella pneumoniae (K. pneumoniae), originally isolated from bovine mastitis. Concentrations of Lf used were 0.67 mg/ml, 1.67 mg/ml, and 2.67 mg/ml. Growth of udder pathogens was monitored by turbidometry either in broth culture or in whey prepared from normal milk. We focused on 3 different growth variables: lag time, slope, and maximum absorbance of bacterial growth curves. Growth inhibition was seen in the broth but hardly at all in whey. The isolates of E. coli and CNS did not grow sufficiently well in whey to draw any conclusions. The most effective inhibitory activity of Lf was seen against E. coli and P. aeruginosa. All 5 E. coil isolates had similar growth patterns. Inhibition of growth by Lf was concentration-dependent. The concentration of 0.67 mg/ml in broth and whey was generally too low for a significant inhibitory effect.  相似文献

Blood plasma analyses for vitamins A, E and selenium were performed from calving to five weeks of lactation in 29 cows. Twelve of the 29 cows had fat cow syndrome. The healthy cows had significantly higher (P<0.01) plasma vitamin A (40 μg/dL) and vitamin E (5 μg/mL) levels than the cows with fat cow syndrome (29 μg vitamin A/dL and 3 μg vitamin E/mL). At parturition, vitamin A level in plasma was low (25 μg/dL) but increased progressively thereafter (up to 51 μg/dL) in healthy cows, whereas cows with fat cow syndrome had lower levels of vitamin A, bordering on deficiency. The possible role of vitamin E in the alleviation of fat cow syndrome by affecting oxidation-reduction reactions in the liver is discussed. Significant (P<0.01) difference was not observed in selenium blood plasma level (35 ng/mL) between the two groups of cows or in another random group of 12 cows clinically affected by fat cow syndrome.  相似文献

A randomized, blocked 23 factorial experiment was conducted with 48 pigs from sows fed a diet low in selenium and vitamin E. From 3 to 12 weeks of age the piglets were kept in single pens and fed a basic diet consisting mostly of barley, dried skim milk, soybean meal and dried yeast, and containing 55 µg selenium and 3 mg vitamin E per kg. The treatment factors — i.e. feed supplements — were 2 levels of Se (nil, 60 µg/kg), 2 levels of vitamin E (nil, 50 mg/kg), and 2 levels of the feed antioxidant ethoxyquin (nil, 150 mg/kg). Blood samples, collected at termination of the experiment, were examined for glutathione peroxidase activity (GSH-Px) and resistance against erythrocyte lipid peroxidation (ELP) to evaluate Se and vitamin E status, respectively. Analysis of variance showed the GSH-Px activity to be litter-dependent (P < 0.001) and influenced by selenium supplementation (P < 0.001) but not by the other supplements or by interactions between supplements. Resistance against ELP was influenced only by vitamin E supplementation (P < 0.001). GSH-Px and ELP thus seem to be valuable and simple methods for evaluating, respectively, Se status and vitamin E status in growing pigs.  相似文献

Very virulent infectious bursal disease virus (vvIBDV) causes high mortality in chickens but measures to reduce the mortality have not been explored. Chickens (8–9 weeks) were treated with 3 agents before and during vvIBDV inoculation. Dexamethasone treatment reduced the mortality of infected chickens (40.7% vs. 3.7%; p < 0.001), but treatment with aspirin or vitamin E plus selenium did not affect the mortality. The bursa of Fabricius appeared to have shrunk in both dead and surviving chickens (p < 0.01). The results indicate that dexamethasone can reduce mortality in vvIBDV-infected chickens and may provide therapeutic clues for saving individual birds infected by the virus.  相似文献

We investigated the effect of supplemental vitamin E on the peripheral blood leukocyte population in Japanese Black calves. Twenty-six calves kept at the same farm were studied. They were divided into two groups; thirteen calves received 300 IU/day of vitamin E orally from 1 to 3 months of age (VE group), and the other thirteen calves did not receive the vitamin E supplement (control group). The VE group showed a higher serum vitamin E concentration at 2 and 3 months of age compared with the control group (P<0.01). The numbers of CD3+ cells and CD4+ cells were higher in the VE group than in the control group, and the difference was statistically significant at 3 months of age (P<0.05). The numbers of CD21+ cells were higher in the VE group than in the control group, and the difference was statistically significant at 2 months of age (P<0.05). The numbers of CD335+ cells tended to be higher in the VE group than in the control group. The numbers of CD8+ cells and CD14+ cells tended to be higher in the VE group than in the control group at 3 and 4 months of age. This study demonstrated that the supplementation of suckling Japanese Black calves with vitamin E might affect the numbers of some immune cell types in the peripheral blood.  相似文献

This experiment aimed to examine the effect of periodical application of bioactive peptides derived from cottonseed (BPC) in comparison with using sub-therapeutic doses of lincomycin and the excessive inclusion of vitamin E on performance, immunity, total antioxidant capacity of serum and intestinal morphology of broiler chickens. A total of 240 one-d-old male broiler chicks with similar initial weight (Ross strain) were randomly assigned to 6 groups (8 chicks/pen): non-treated group (basal diet), basal diet supplemented with 2 mg/kg lincomycin, basal diet supplemented with 50 IU vitamin E, basal diet supplemented with 6 g BPC/kg in starter period, basal diet supplemented with 6 g BPC/kg in starter and grower periods and basal diet supplemented with 6 g BPC/kg throughout the whole experiment. The highest final body weight was obtained in the group supplemented with BPC in starter and grower periods. In the finisher phase, broilers fed the diet containing BPC in the starter period and in the whole trial had significantly (P < 0.05) better feed conversion ratios (FCR). Jejunal villus height was significantly elevated in broilers supplemented with antibiotic (P < 0.001), furthermore it tended to be greater in broilers fed BPC in the starter period. The jejunal villus height-to-crypt depth ratio was significantly (P < 0.01) higher in broilers fed the diet containing antibiotic in comparison to other groups. Humoral immune response against Newcastle disease vaccine tended to be elevated in broilers fed the diet containing BPC in the whole trial (P > 0.05). Broilers supplemented with BPC in starter and grower, and in the whole trial had significantly (P < 0.05) higher antibody titers against sheep red blood cells (SRBC). The highest total antioxidant capacity was obtained in broilers supplemented with the excessive level of vitamin E, furthermore it tended to improve in broilers fed the diet containing BPC in the whole trial. In summary, the results of the study indicated that addition of BPC in broiler diets in the whole trial could improve FCR, immune responses and total antioxidant activity of serum, and BPC could be used in broiler diets as an alternative to in-feed antibiotics.  相似文献

Using high pressure liquid chromatography the serum concentration of vitamin E was measured in dairy cows fed either hay or silage as their main roughage, in calves fed milk-replacer, and in young intensively fed bulls. The concentrates fed to the cows, calves and bulls were supplemented with 5–10, 25 and 5–10 mg DL-α-tocopheryl acetate per kg, respectively, and the milk-replacer for the calves was supplemented with 50 mg DL-α-tooopheryl acetate per kg powder. Cows fed silage as their main roughage had higher serum vitamin E concentrations (: 3.8–5.2 mg/l) than cows fed only hay (: 2.5–4.1 mg/1). Lactating cows had higher vitamin E concentrations than dry cows (: 4.1–5.2 and 2.5–3.8 mg/l, respectively) and calves and bulls had much lower vitamin E concentrations (: 1.4 and 1.2 mg/l, respectively) than cows. Thirty per cent of the calves and 41 % of the bulls had serum vitamin E concentrations less than 1.0 mg/l, suggesting that in these animals the conventional level of supplementation of feeds with DL-α-tocopheryl acetate in Sweden is probably inadequate for the prevention of nutritional muscular degeneration and other negative effects.  相似文献

The effect of varying levels of dietary vitamin E on selenium status was determined using 40 Wistar rats with similar initial body weight. The rats were equally divided into four groups and fed the following dietary treatments (mg vitamin E/kg DM): 18 (control; C‐diet), 0 (0E‐diet), 9 (0.5E‐diet) and 36 (2E‐diet) for either 4‐week (Phase 1) or 8‐week (Phase 2) period. At the end of experiment, animals were slaughtered to measure vitamin E and selenium levels in the brain, liver, spleen, kidney, muscles and blood tissues. Dietary vitamin E levels did not affect feed and water intake and body weight. But whole‐blood selenium concentration in rats fed 0E‐diet was higher than in rats fed 2E‐diet after 4 weeks. Selenium level in muscle, spleen and brain was also higher (p < 0.05) in rats fed the 0E‐diet than in rats fed C‐diet after 4 weeks of feeding. On the other hand, selenium level in the muscle was lower (p < 0.05) in rats fed 0.5E‐diet than in those fed 0E‐diet after 4 weeks of feeding, but not after 8 weeks of feeding. Increasing dietary vitamin E level directly influenced selenium content of the spleen and brain after 8 weeks of feeding 2E‐diet. Moreover, the twofold increase in vitamin E intake resulted in a tendency to reduce whole‐blood selenium level and total selenium in the liver and kidney after 4 and 8 weeks. The results showed that the increasing dietary vitamin E level resulted in a tendency to reduce Se contents in some vital organs of the body such as the liver and kidney, suggesting their close compensatory interrelationship. Therefore, dietary vitamin E level directly influenced selenium metabolism in the animal body.  相似文献
