China was one of the first countries to use artificial insemination (AI) in equids, and it achieved international recognition for its widespread application of AI to commercial horse breeding. This article reviews the history of equine AI in China. The technique originated from the high demand for horses to be used in agriculture, transportation, and the military. Artificial insemination was identified as an ideal tool for Chinese horse breeding to improve the productivity of native horses, especially during 1950-1970. Presently, AI is still practiced commonly in China, and it includes the use of fresh semen and transported cooled semen. The use of frozen semen has also been resumed to broaden the range of elite Sporthorse stallions and to preserve threatened or endangered native breeds. Accurate prediction of the optimum time for insemination depended mainly on transrectal palpation of the mare's ovaries. In addition to controlled methods of insemination, factors like volume and number of spermatozoa in the inseminate and timing and frequency of insemination were optimized to accomplish high fertility rates in the field. Production of hinnies and mules for agricultural labor and donkeys for meat and hide gelatin stimulated a nationwide upsurge of research into the reproductive physiology of and AI technology in donkeys. In the future, there will be further increases in the use of traditional and modern AI techniques in the breeding of Chinese equids.  相似文献

人工授精技术是一种较先进的辅助生殖技术,可提高繁殖育种的效益,但因猫类较独特的生殖特点,其人工授精技术发展受限。近年来,科研人员和临床工作者借鉴其他物种相对成熟的人工授精技术,结合猫类生殖特点,对猫人工授精过程中精液采集、精液品质评估、精液保存、刺激母猫排卵方式及具体的人工输精等相关技术进行了优化和创新,如开发了新颖实用的尿导管法采精技术以采集猫精液;在传统精液品质评估的基础上,配合使用荧光染色技术、流式细胞技术及体外受精技术对猫精液品质进行评估;探索不同冷冻液配方及冷冻程序对猫精液保存效果的影响,并尝试使用超快速冷冻法保存公猫遗传物质;研究使用不同种类及不同剂量的激素对诱导母猫发情和排卵效果的差异,以及不同输精部位、方法及时机对母猫妊娠结果的影响等。尽管其中一些新技术与方法已在试验动物中彰显出了其实际应用潜力,但当下依旧还存在操作相对复杂、成本较高、效果低于预期等问题,导致目前猫人工授精技术的应用较局限,不如在犬或其他动物中应用广泛,未来还需更进一步研究和探索才能满足临床实际应用的需要。文章就上述猫人工授精技术中各个方面的研究进展进行了综述,以期为该技术的进一步研究和应用提供参考。  相似文献

Artificial insemination (AI) in pigs has been established for about four decades but ejaculates are still used insufficiently. Higher demand of semen for AI and new techniques that involve low sperm concentration require the optimization of insemination protocols. Based on the knowledge of the physiology of sperm transportation and events in the female genital tract prior to fertilization, new strategies are under development to minimize sperm losses. One goal is to deposit the semen into the uterine horn rather than into the proximal cervix. It was shown that the minimal number of spermatozoa necessary for surgical AI at the utero‐tubal junction (UTJ) were at least 1 × 106 diluted in 0.5 ml of a special extender. Artificial insemination into the distal part of the uterine horn required about 1 × 107 million sperm in 20 ml of extender. Meanwhile, first insemination devices for non‐surgical intra‐uterine AI are commercially available. Using similar sperm concentrations as for surgical AI, non‐surgical uterine insemination did not differ significantly from control inseminations in terms of pregnancy rate and litter size. With respect to the fertilizing capacities of their ejaculates, boars have to be selected more strictly for sperm quality parameters as most of the compensatory effects of sperm cells disappear in maximally extended semen samples.  相似文献

The liberalization of European animal breeding legislation and an increasing diversity of equestrian sports have led to a constant rise in the number of horse breeds and breed registries. In addition to the trend towards more and smaller breed registries, there is another trend towards an international expansion of the bigger established sport horse breeds. Regional breeds, at least in smaller countries, may no longer be able to run an independent breeding programme. The typical horse breeder, in the future, will be a female and qualified in equestrian sports. Artificial insemination (AI) mainly with fresh or cooled-transported semen has become a major breeding tool, allowing breeders all over Europe to benefit from the best stallions of most breeds. New AI techniques such as low-dose insemination may remain restricted to individual stallions and also the interest of breeding programmes in sex determination of foals via semen sorting is limited. Embryo transfer and associated techniques, although allowed by most breeds, have not contributed significantly to genetic progress in European sport horses so far. A potential use of cloning may be to produce gonad-intact copies from geldings that have performed to a superior level. With a more open and international structure of horse breeding and increased use of AI, equine reproduction and biotechnology should be emphasized by veterinary curricula and continuing professional education programmes.  相似文献

Important early studies on mammalian artificial insemination (AI) were carried out in equids, and at the end of the 19th century, the first AI programs were set up in horses. At that time, the most systematic research on equine AI was performed in Russia. After World War I, AI research shifted to cattle and sheep. This time saw major advances such as the development of artificial vaginas and phantoms for semen collection. Semen dilution counteracted the detrimental effect of seminal plasma, allowed semen storage, and increased the volume of an ejaculate for insemination of more mares. In the late 1930s, techniques for cooled semen AI as used today were in principle available. After World War II, the number of mares inseminated decreased, but with a new role of the horse as a partner in equestrian sports, new interest in equine AI was raised. In contrast to the situation in cattle, frozen semen has not replaced cooled semen AI in the horse. Recent advances in insemination of horses are the sexing of sperm, low-dose deep intrauterine insemination, and intracytoplasmic sperm injection.  相似文献

Benign prostatic hyperplasia was diagnosed in an American Staffordshire Terrier of high breeding value presenting concurrent haematuria. Castration as a treatment was synchronized with the oestrus cycle of a bitch selected for insemination. After castration the cauda epididymis was flushed with Gent semen extender and collected spermatozoa were filtered and analysed by Hamilton Thorn computer assisted sperm analysis. A total of 7 ml semen containing 742 x 10(6) spermatozoa with 76.5% mean motility was used for insemination. Intravaginal insemination of the bitch was performed with an insemination catheter for dogs (Kruuse, Marslev, Denmark) on the day when plasma progesterone levels reached 9.9 ng/ml. Normal pregnancy without complications resulted in eight live-born puppies 63 days after insemination. This is the first report of a normal pregnancy and birth of puppies from a bitch inseminated with epididymal semen obtained from a dog affected by benign prostate hyperplasia.  相似文献

REASONS FOR PERFORMING STUDY: Historically, artificial insemination (AI) using frozen semen has been perceived to have poorer success rates and be more labour intensive than using chilled semen. A retrospective study was therefore conducted to compare the conception rate achieved by AI between chilled and frozen semen, using fixed time insemination protocols over 2 breeding seasons. HYPOTHESIS: Artificial insemination using chilled semen produces a higher conception rate than that achieved with frozen semen. METHOD: Mares (n = 251) were inseminated with either chilled (n = 112) or frozen (n = 139) semen in the 2006 and 2007 northern hemisphere breeding season. Per rectum ultrasonography of the mare's reproductive tract determined the timing of insemination, and deslorelin acetate was used to induce ovulation. Chilled semen insemination was performed using a single preovulatory dose delivered into the uterine body. Frozen semen was administered as 2 doses (pre- and post ovulation) using a deep uterine insemination technique. Pregnancy was detected ultrasonographically at 15 days post insemination. Conception rates were compared using a Chi-squared test. RESULTS: Insemination with frozen semen produced a significantly (P = 0.022) higher seasonal conception rate (82.0%) than that achieved with chilled semen (69.6%). CONCLUSIONS AND POTENTIAL RELEVANCE: Insemination with frozen semen can achieve conception rates equal to those with chilled semen, enabling the mare owner a greater selection of stallions.  相似文献

There has been increased interest recently in the conservation of wild felids and preservation of valuable cat breeds. Assisted reproduction, by means of artificial insemination (AI), is an important tool for developing breeding programs for conservation. Optimal use of AI requires accurate data on semen conservation protocols and its long-term storage/survival. In this paper, semen cooling and freezing processes are described, with special emphasis on the results obtained in experiments performed in the domestic cat. SUMMARY The financial costs and benefits associated with the use of artificial insemination (AI) in commercial flocks are evaluated. Benefits are calculated in terms of net present values after summing the discounted value of benefits over 20 years. Two breeding strategies are evaluated. With the first, AI is used to produce flock ewes and wethers. The method is unlikely to be profitable unless high breeding value rams are available for AI programs with fresh semen. With the second, AI is used to produce home-bred rams, which in turn sire flock ewes and wethers. This approach is more likely to be profitable. The cost of AI per lamb weaned from laparoscopic AI programs is about $100. Benefits exceed this cost for rams of very high merit when wool prices are moderate or higher. Flock structure has a significant effect on the benefits. Flocks with low wether retention rates have benefits half that of flocks that retain most wethers to 6 years of age. AI with purchased semen also provides benefits to risk management for owners of commercial flocks who wish to breed their own replacement flock rams.  相似文献

Artificial insemination is an effective technique for improving utilization of stallions in breeding programs. When proper semen handling and insemination procedures are used, optimal pregnancy rates are attainable. When AI techniques are employed for mares and stallions with marginal fertility, pregnancy rates may be improved in comparison with natural mating. Preservation of stallion semen in the liquid or frozen state reduces the costs and potential health hazards incurred by transporting mares and provides easier access to genetic material that may otherwise be unavailable. Acceptable pregnancy rates are consistently obtained with cooled semen. Conversely, techniques for cryopreservation of stallion semen will require more refinement before the procedure can be considered commercially viable on a wide scale.  相似文献

Efficient artificial insemination (AI) is essential for future challenges in the pig industry. Knowledge on the exact relation between semen quality characteristics and fertility can have a major impact on both the genetic merit of future animals and the efficiency of AI. Variation in fertility is caused not only by farm- or sow-related parameters but also by boar- and semen-related parameters. In pig AI there is no gold standard concerning semen quality assessment. Assessing semen quality characteristics objectively and relating them to large field fertility datasets leads to an efficient production of insemination doses, which results in an efficient dissemination/descent of the breeding program required genes. Overall, this contributes to the development of semen quality assessments, which improves the prediction of porcine male fertility. Knowing which semen characteristics, and to what extent, contribute to male fertility and makes the field fertility more predictable.  相似文献

Genetic parameters were estimated for semen production traits collected in an Austrian AI centre in the years 2000-2004. In total, 12,746 ejaculates from 301 Austrian dual-purpose Simmental (Fleckvieh) AI bulls were examined considering different effects on ejaculate volume, sperm concentration, percentage of viable spermatozoa in the ejaculate, total spermatozoa per ejaculate and motility. The model for genetic parameter estimation included the fixed effects age of bull, collection interval, number of collections on collection day, bull handler, semen collector, year and month of collection, a random additive genetic component and a permanent environmental effect. Correlations between estimated breeding values for semen traits and male fertility from the routine evaluation were calculated. The fertility trait considered in the routine evaluation is non-return rate 90 for the first insemination. All semen production traits were moderately heritable. Heritabilities for volume, concentration, percentage of viable spermatozoa, total number of spermatozoa and motility were 0.18, 0.14, 0.10, 0.22 and 0.04, respectively. Correlations between breeding values for semen quality traits and routinely estimated breeding values for male fertility were low and ranged from 0.08 to 0.17 indicating that semen production traits are rather poor predictors of male fertility.  相似文献

The common piping guan (Pipile cumanensis cumanensis) is serving as a model for assisted reproductive techniques (ARTs) in Cracidae (Aves: Galliformes). Manual collection methods used in domestic birds and other exotics were modified to optimize quality and quantity of fresh semen samples. In addition, a method to subjectively assess semen quality on the basis of its color, clarity, and consistency was developed to serve as a tool when laboratory equipment is not available. In a species where only two eggs per clutch is typical, predictability of the date of the first egg is critical to ensure adequate inseminations before oviposition. An increase in both the distance between the female's pubic bones and degree of abdominal swelling indicated optimal time to initiate artificial insemination (AI) and allowed the number of semen samples used to fertilize the first egg in the clutch to be minimized while ensuring its fertility. In three separate AI trials, a total of four fertile eggs were produced, a first for cracids. Two of the four fertile eggs hatched successfully, and the chicks' parentages were confirmed using DNA fingerprinting. This study represents the first published account of the use of ARTs in Cracidae and forms the basis for future developments (e.g., AI of frozen-thawed semen) aimed at optimizing genetic management of cracids in ex situ breeding programs.  相似文献

The extenders and freezing rates from three different freezing protocols were combined and compared to each other in order to study the post-thawing acrosome integrity and fertility of frozen dog sperm. A commercial bovine TRIS-base extender (TRILADYL) and two self-made canine semen extenders (Norwegian and Dutch) were combined with a conventional bovine and two canine freezing regimes, and acrosome integrity of frozen/thawed spermatozoa was assessed by fluorescein isothiocyanate conjugated peanut agglutinin staining (FITC-PNA). Differences between freezing/thawing protocols were reflected in the proportion of cells with acrosomal damage and not based on motility results. It was concluded that during dog semen cryopreservation extenders had less influence on the post-thawing sperm quality than did the freezing rates. The optimal extender/freezing rate combination (TRILADYL/Norwegian) was used in the clinical practice to evaluate the fertility of frozen sperm administered by intrauterine insemination using a surgical approach. The pregnancy rate was 57% (4/7), but the average litter size was low (2.8). This may have been due to the insufficient sperm numbers contained in an insemination dose and/or to the incorrect timing of artificial insemination (AI). The final conclusion is that the commercial bovine extender is useful for freezing dog semen, and the TRILADYL/Norwegian freezing protocol is recommended as the most advantageous combination for the freezing of canine semen in the clinical practice.  相似文献

An opportunity for study of the potential role of semen in the transmission of bovine leukosis virus (BLV) was provided when a Jersey herd was found to be BLV-seronegative. This was a closed herd; new genetic material had been introduced by artificial insemination (AI), using semen collected and processed at 7 AI centers in the United States. Of 66 donor bulls from which semen had been collected for AI use in the herd during the 5 years the herd remained seronegative, 24 were consistently BLV-seropositive and 2 became seropositive for BLV during the study. Semen collected from the BLV-seropositive bulls accounted for 1,019 semen units, representing 48.3% of the semen purchased. The maintenance of BLV seronegativity in this herd for 5 years, when semen from BLV-seropositive bulls was used for AI, provided evidence for the lack of infectivity of BLV in bovine semen.  相似文献

Artificial insemination (AI) can undoubtedly be regarded as the oldest and most widely used assisted reproductive technique/technology (ART) applied in livestock production and it is one of the most important ARTs. The three cornerstones of its application are that it is simple, economical and successful. Artificial insemination offers many well-known benefits for producers. Fresh, fresh + diluted + chilled and frozen semen can be used for AI in small ruminants. To ensure its successful use, the AI technique must be selected on the basis of the type of semen planned to be used. This review paper gives a detailed overview of semen processing and its effects on semen quality, as well as of the AI techniques applied in small ruminants and their success rates.  相似文献

Our previous report indicated that addition of Orvus ES Paste (OEP) to the extender of frozen canine semen protected acrosomes and maintained sperm motility after thawing. In this study, artificial insemination (AI) using the frozen semen was carried out. The frozen semen was prepared using egg yolk Tris-fructose citrate, and the final concentrations of glycerol and OEP were 7% (v/v) and 0.75% (v/v), respectively. AI was performed during the optimal mating period predicted from the peripheral plasma progesterone level. In intrauterine insemination (IUI), the bitches were laparotomized and 1 x 10(8) spermatozoa were infused into one of the uterine horns. In insemination of non-OEP supplemented semen, 3 x 10(8) spermatozoa were inseminated. In intravaginal insemination (IVI), 10-40 x 10(8) spermatozoa were inseminated. Conception was obtained in nine of 10 bitches (90.0%) that underwent IUI. The number of newborns was from 1 to 7 (mean 3.6 +/- 0.9). The mean ratio of the number of puppies to the number of ovulations in the inseminated uterine horn was 71.8%. The number of puppies did not exceed the number of ovulation in the inseminated uterine horn. Conception using non-OEP supplemented frozen semen was unsuccessful in all four bitches. In IVI, conception was not obtained in any of the six bitches that received insemination of 10 x 10(8) or 40 x 10(8) spermatozoa, but two of three bitches that received insemination of 20 x 10(8) spermatozoa were fertilized. It was shown that a high conception rate can be obtained by IUI using OEP-supplemented frozen canine semen. Developmenmt of a non-surgical method of IUI and a method of freezing canine sperm applicable to IVI is necessary.  相似文献

The objectives of this study were to determine associations between low farrowing rate and various management factors in sow herds. In 30 sow herds, a management survey, breeding observations, semen evaluation, and semen storage temperature monitoring were completed. Herds with an average farrowing rate of < 85% were classified as low farrowing rate herds while those with an average farrowing rate of ≥ 85% were classified as good farrowing rate herds. Low farrowing rate herds were more likely than good farrowing rate herds to move boars into gilt pens for estrus detection, breed a high proportion of sows by artificial insemination (AI) only, start heat detection 3 d post-weaning, wipe the vulva prior to breeding, and use “hands-free” AI devices.  相似文献
