Germinated brown rice (GBR) recently has received renewed attention due to its enhanced nutritional value. Pasting properties and in vitro starch digestibility of GBR were examined before and after hydrothermal treatments. Steeping in water (30 °C, 24 h) raised the moisture content and germination percentage of brown rice. Pasting viscosity was substantially decreased but gelatinization temperatures and enthalpy were decreased only marginally by germination (30 °C, 48 h). However, annealing (50 °C, 24 h) and heat-moisture treatment (100 °C, 1 h at 30% moisture) after germination resulted in increased pasting viscosity and gelatinization temperatures. The hydrothermal treatments, however, induced browning reactions to darken the flour of GBR. The digestibility of starch in brown rice was increased by germination. The contents of rapidly digestible starch (RDS), slowly digestible starch (SDS) and resistant starch (RS) in the cooked brown rice were 47.3%, 40.8%, and 11.9%, respectively, but changed to 57.7%, 39.1%, and 3.2%, respectively upon germination. The hydrothermal treatments, however, decreased the digestibility of starch in GBR. The heat-moisture treatment decreased the RDS content in GBR near to that of native brown rice. The digestibility and physical properties of brown rice can be controlled by germination and hydrothermal treatments.  相似文献

Improvement of rice storage proteins is important in rice breeding for high nutritional quality. Seventy-one recombinant inbred lines (RIL) derived from a cross between japonica variety Asominori and indica variety IR24 were used to study the inheritance of crude protein and protein fraction contents in rice. A total of 16 QTL were identified and mapped on eight chromosomes. Several QTL affecting contents of different protein fractions were mapped in the same chromosomal region. In particular, two QTL with a significant contribution were identified to simultaneously affect prolamin and glutelin contents. One QTL denoted as qCP-12 affecting crude protein content (CP) was located in the same region as QTL qGLT-12 affecting glutelin content, in agreement with the positive correlation between glutelin level and protein content. QTL with larger genetic effects were further confirmed using two sets of chromosome segment substitution lines (CSSL), where Asominori and IR24 were used as the recurrent parents. By QTL comparative analysis, two QTL for CP, three for globulin content and one for prolamin content were located in the vicinity of CP QTL previously identified in polished rice. Application of these results in rice breeding is discussed.  相似文献

Although dozens of transgenic Bacillus thuringiensis (Bt) rice lines have been developed, none of them has been released to farmers. Under field conditions, we evaluated the influence of a hybrid Bt rice on the primary rice insect pests and rice yield in 2005 and 2006. Four treatments were evaluated, including Bt and non-Bt rice treated with insecticides when necessary, and unprotected Bt and non-Bt rice. Unprotected Bt rice exhibited stable and high control of the three primary lepidopteran pests, Chilo suppressalis Walker, Tryporyza incertulas Walker and Cnaphalocrocis edinalis Güenée. Under unprotected conditions, larval densities of these three pests in Bt plots decreased by 87.5–100% compared to those in non-Bt plots, and percentages of damaged stems and leaves remained less than 0.6% during the entire rice growing season. In early rice growth stages, populations of two important planthoppers, Nilaparvata lugens Stål and Sogatella furcifera Hovarth, were significantly affected only by protection level (protected vs unprotected). However, in late rice growth stages (filling and maturing), densities of planthoppers were significantly affected both by protection level and by rice type (Bt vs non-Bt), and densities of N. lugens were significantly higher in Bt plots than in non-Bt plots under unprotected conditions. Pesticide sprays were reduced by 60 and 50% in protected Bt vs protected non-Bt plots in 2005 and 2006, respectively. Yield of unprotected Bt rice increased by 60–65% compared to unprotected non-Bt rice, but decreased by 28–36% compared to protected Bt rice. These results show that Bt rice increased yield greatly, but still required pesticide sprays to avoid losses caused by non-target insect pests.  相似文献

The aphids Macrosiphum euphorbiae (Thomas) and Myzus persicae (Sulzer) (Hemiptera: Aphididae) are responsible for yield reduction in potato (Solanum tuberosum) production by direct phloem feeding and by spreading viruses. Breeding resistant traits from Solanum chomatophilum into the potato germplasm provides alternative means to control aphid infestations. Integrated pest management strategy, using plant resistance, benefits from the characterization of the resistance and of its impact on aphid biology. Our objective was to characterize the resistance of S. chomatophilum by assessing the effects of accessions, plant parts on aphid performance, and by assessing the impact of the resistance factors on different aphid developmental stages and on alate morph production. Detailed aphid performance was obtained by measuring fecundity, survival, percentage of nymphs that reached adult moult, and population growth using whole plant and clip cage experimental designs. Accession and plant physiological age, but not aphid developmental stage, influenced all life-history parameters, except for alate morph production which was not induced on the resistant accessions. Plant part influence was independent of plant species and accession. Both experimental designs resulted in congruent resistance levels at the accession level for each of the two aphid species, supporting the use of any of them in S. chomatophilum resistance screening. PI243340 was resistant to both aphid species, while PI365324 and PI310990 were also resistant to M. euphorbiae and M. persicae, respectively.  相似文献

Transgenic corn hybrids that express toxins from Bacillus thuringiensis (Bt) are highly effective against the European corn borer, Ostrinia nubilalis (Hübner), and the closely related Asian corn borer, Ostrinia furnacalis (Guenée). Since the registration of Bt corn hybrids in the U.S. in 1996, there has been a great deal of information generated on O. nubilalis. However, relatively little information exists for O. furnacalis. To help determine whether the information generated for O. nubilalis can be leveraged for decisions regarding the use of transgenic Bt corn against O. furnacalis, experiments were designed to determine whether the pattern of sensitivity to various Bt Cry1 toxins is similar between the two species. Test insects included laboratory-reared O. furnacalis originating from Malaysia, a Bt-susceptible laboratory colony of O. nubilalis maintained at the University of Nebraska-Lincoln (UNL) and an out-group consisting of the sugarcane borer, Diatraea saccharalis (F.), from Louisiana which represents a different genus from the same family. O. furnacalis and O. nubilalis exhibited a similar pattern of susceptibility to all the Cry1 toxins and were highly susceptible to the range of Bt toxins tested including Cry1Aa, Cry1Ab, Cry1Ac and Cry1F. Both of the Ostrinia species were more tolerant to Cry1Ba compared with D. saccharalis, although sensitivity of O. furnacalis was intermediate and did not differ significantly from that of O. nubilalis and D. saccharalis. D. saccharalis was also susceptible to the range of toxins tested but unlike the two Ostrinia species, was more tolerant to Cry1F and more susceptible to Cry1Ba. These results indicate that both of the Ostrinia corn borer species are similar in sensitivity to the Cry1Aa, Cry1Ab, Cry1Ac, Cry1Ba and Cry1F toxins, thus suggesting shared toxin receptors and mechanisms of toxicity for the two species.  相似文献

A major esterase (designated OsEST1) showing high activity using 1-naphthyl acetate as a substrate was identified from rice bran and purified approximately 239-fold to near-homogeniety. The purified enzyme migrated as a single polypeptide band on native and SDS-polyacrylamide gels and had a molecular mass of 25 kDa under denaturing conditions. Analysis of its tryptic peptides by MALDI-TOF-MS and subsequent data mining identified a corresponding cDNA OsEST1 consisting of 714 nucleotides and encoding a 238 amino acid protein. Analysis of its primary sequence indicated that OsEST1 is a GDSL-motif carboxylester hydrolase belonging to the SGNH protein subfamily in containing the putative catalytic triad of Ser11, Asp187, and His190. OsEST1 showed the highest catalytic activity at approximately pH 8.0–8.5 and at 45 °C with Km and Vmax values for 1-naphthyl acetate of 172 μM and 63.7 μmol/min/mg protein, respectively. However, OsEST1 showed no activity with triacylglycerol. Alignment of the primary sequence of OsEST1 and other rice GDSL-motif esterases/lipases showed that OsEST1 aligns with a specific family of plant SGNH esterases involved in response to dehydration and cuticle formation. These results suggest that OsEST1 is not a lipase but an esterase activity which has some other function in rice, especially during seed development.  相似文献

Characterization of rice storage proteins by SE-HPLC and micro z-arm mixer   总被引:2,自引:0,他引:2  
While the effect of protein content and composition on the functional properties of wheat flour is well studied, our knowledge on the same properties of rice flour is limited. This work was conducted to study the relationship between the dough mixing properties of flour from different rice cultivars and protein content and composition. An efficient sonication-based two-step extraction procedure was applied to isolate rice flour proteins. The size-exclusion HPLC (SE-HPLC) method, originally developed for separating wheat proteins, was applied with some minor modifications in order to study the size distribution of rice flour proteins. Four fractions were distinguished on the SE-HPLC profile and were further characterized by SDS-PAGE. Fractions I–III consisted of glutelins, while fraction IV contained albumin, globulin and prolamin proteins. When rice dough was characterized on the basis of mixing parameters in a micro z-arm mixer, significant differences were observed depending on the protein composition of the flour. Statistical analysis results indicated that the functional properties of the flour from different rice cultivars were associated with the amount of polymeric proteins and their size distribution.  相似文献

Glutelin, a major protein in rice grains, is encoded by a multigene family. However, its protein composition is not well characterised. Here, we identified and characterised two novel glutelin subunits, GluBX and GluC. The individual glutelin subunits of japonica cv. Nipponbare and indica cv. 93-11 rice were analysed using 2-dimensional gel electrophoresis, LC–MS/MS, and Western blotting. Comparison of the glutelin profiles between three japonica and three indica cultivars indicated two distinct subunits (GluA-1 and GluA-3 isomers) and a distinction in the subunit composition (notably GluA-3 and Lys-rich GluB-1 components) of these two subspecies. Sequence alignment revealed different nutritional (Lys residues) and functional (Cys residues) characteristics between the type-A and type-B glutelin subfamilies. We also analysed amino acid and total protein contents of the grains in thirty-five cultivars, and we demonstrated that the Lys-rich glutelin composition of indica cultivars is superior to that of japonica cultivars. The Lys-rich and Cys-poor GluBX subunit is a native protein and is a high nutritional protein in grains. Our combined approaches for the identification of glutelin subunits have revealed the nutritional characteristics of individual subunits in rice, and this knowledge will provide new insights for improving grain quality during rice breeding.  相似文献

II-32B is a key maintainer line used for hybrid rice breeding in China. However, it is of poor quality for most Chinese consumers because of its high apparent amylose content (AAC), high gelatinization temperature (GT) and non-fragrance. It is well known that the AAC, GT and fragrance traits are largely controlled by the Wx, starch synthase IIa (SSIIa), and fragrance (fgr) genes, respectively. With the aid of functional markers, we improved the quality of II-32B by introgressing the Wx, SSIIa, and fgr genes from Yixiang B, a fragrant maintainer line that has low AAC and low GT. The microsatellite allele (CT)17 of Wx, the contiguous single nucleotide polymorphism TT allele of SSIIa and the 8-bp deletion allele of fgr were transferred to II-32B by two backcrosses and three selfings. Molecular marker assisted selection was applied in the series to select for individuals carrying Wx-(CT)17, SSIIa-TT, and fgr-deletion alleles. According to the marker genotypes, seventeen homozygous lines for Wx-(CT)17, SSIIa-TT, and fgr gene markers were finally selected. The improved II-32B lines were fragrant with reduced AAC and GT.  相似文献

Longer-term partial stagnant flooding, particularly when it occurs following transient complete submergence causes severe damage to modern rice varieties. Progress was made in developing varieties tolerant of complete submergence through transfer of the Submergence-1 (SUB1) gene into popular varieties. However, SUB1 may not be effective under partial stagnant flooding (SF), as the new varieties may not elongate and continue growth when fully or partially submerged because of the SUB1-mediated suppression of elongation. We tested a set of rice genotypes, including a pair of near-isogenic lines (NILs), Swarna and Swarna-Sub1, under either SF or SF following complete submergence of 12 d. Swarna-Sub1 had higher survival and yield than Swarna following 12 d of submergence, but survival and grain yield of all lines decreased substantially when SF of 15-30 cm followed complete submergence, with the sensitive lines experiencing greater reductions in growth and yield. This suggests the importance of combining SUB1 with tolerance of SF in areas where both stresses are expected during the season. Swarna and Swarna-Sub1 are more sensitive to long-term partial SF than IR49830 and IR42 because of their short stature, and Swarna-Sub1 showed slightly higher reduction in tillering than did Swarna when subjected to deeper SF alone, possibly because of further inhibition of elongation by SUB1 if induced in submerged tissue. The results suggest that tolerance of these genotypes to SF depends less on SUB1 introgression and more so on the genetic background of the recipient genotype, with better performance of the genotypes that are inherently taller, such as IR42 and IR49830. The SUB1 donor landrace FR13A and its derivative breeding line IR49830 had better survival and relatively less reduction in grain yield under SF following complete submergence and under longer-term partial SF, indicating that these genotypes may have genes other than SUB1 for submergence and SF tolerance. For better adaptation to prolonged SF, SUB1 should be introgressed into genotypes that tolerate partial SF through better tillering ability and taller shoots, because SUB1 may not be effective in shorter genotypes, as it promotes survival of submerged plants by hindering shoot elongation to conserve energy reserves. Varieties combining tolerance of prolonged SF with SUB1 will have broader adaptation in flood-prone areas and greater impacts on yield stability.  相似文献

Biocontrol capacity of two plant growth-promoting rhizobacteria (PGPR) strains, against blast disease in rice paddy fields in Southern Spain was studied in three cropping seasons. Both strains (Pseudomonas fluorescens Aur 6 and Chryseobacterium balustinum Aur 9) had already shown biocontrol capacity against pathogens, ability to induce systemic resistance against leaf pathogens and against salt stress in different plant species. Bacterial treatments were carried out on seeds and/or on leaves. Strains were inoculated individually and in combination. Protection against natural disease incidence was evaluated, and rice production and quality measured in 2005 and 2006 trials. In 2004, natural disease incidence was low (between 0.1% and 0.35% of damaged leaf surface) due to environmental conditions; under these conditions, both strains significantly protected plants against rice blast. In 2005, disease incidence was higher than in 2004, reaching higher values of affected leaf surface in controls. In these conditions, each strain individually protected rice against rice blast, although the combination of both strains was the most effective treatment. All three treatments (Aur 6, Aur 9 and Aur 6 + Aur 9) reached 50% protection in panicles, with Aur 9 being the most effective. In 2006, the most effective treatment was the combination of both strains on leaves in three physiological stages, suggesting a biocontrol mediated protection. On the other hand, when bacteria were applied to seeds, disease incidence decreased up to 50%, suggesting induction of systemic resistance. Finally, a direct relation between protection mediated by the PGPR and the increase in rice productivity (mT/ha) and quality (weight of 1000 seeds and number of intact grains after milling) was found.  相似文献

In rice grains, high amylose content (AC) is correlated with poor grain quality, particularly in indica hybrid rice. To obtain indica hybrid rice with improved cooking and eating qualities, we introduced the antisense Waxy (Wx) gene into 2 elite parental lines of indica hybrid rice by using co-transformation methods. Subsequently, we selected several elite homozygous transgenic lines that did not contain the selectable marker. The expression of the endogenous Wx gene of the selected transgenic lines was significantly downregulated, resulting in low AC in the mature seeds; moreover, the AC in some lines reduced to the level observed in glutinous rice. With the decrease in AC, the gel consistency of the transgenic rice became softer, and the gelatinization temperature tended to be higher than those of the wild types, especially in the case of the Longtefu-derived transformants. We also analyzed the pasting properties of the selected transgenic low-AC lines, and we noted an improvement in the pasting properties of the transgenic rice lines. The results from a field trial indicated that the grain weights of the transgenic lines with lower AC exhibit remarkable reduction compared with those of the wild types.  相似文献

Stevia rebaudiana is a valuable medicinal plant species and it is being used for the treatment of diabetes. Currently, there is a high demand for raw material of this medicinal herb due to ever increasing diabetes disorder among the population. In order to meet the increased demand an efficient in vitro propagation of S. rebaudiana was established. Nodal explants collected from the field were cultured on MS basal medium fortified with different concentrations of BAP (0.5-3.0 mg/l) and KIN (0.5-3.0 mg/l) individually for shoot bud induction. In vitro derived nodal buds were cultured on MS medium supplemented with different concentrations (0.5-3.0 mg/l) of BAP and KIN for multiple shoot bud regeneration. In the second experiment, in vitro derived buds were placed on MS medium supplemented with different concentrations of BAP (0.5-3.0 mg/l) in combination with 0.5 mg/l IAA or IBA or NAA for shoot bud multiplication. The highest frequency (94.50%) of multiple shoot regeneration with maximum number of shoots (15.69 shoots/explant) was noticed on MS medium supplemented with 1.0 mg/l BAP. For large scale plant production, in vitro derived nodal bud explants were cultured on MS medium fortified with 1.0 mg/l BAP, in which about 123 shoots/explant were obtained after three subcultures on the same media composition. Elongated shoots (>2 cm) dissected out from the in vitro proliferated shoot clumps were cultured on half-strength MS medium containing different concentrations of NAA (0.1-0.5 mg/l) and/or MS medium fortified with various concentrations (0.5-2.0 mg/l) of auxins (NAA, IAA and IBA) for root induction. Highest frequency of rooting (96%) was noticed on half-strength MS medium augmented with 0.4 mg/l NAA. The rooted plantlets were successfully transferred into plastic cups containing sand and soil in the ratio of 1:2 and subsequently established in the greenhouse. The present in vitro propagation protocol would facilitate an alternative method for rapid and large-scale production of this important antidiabetic medicinal plant.  相似文献

The starch and protein properties of rice grain are important factors for sake brewing and these properties are reported to be influenced by temperatures during grain ripening. Amylose content, nitrogen content, protein composition, pasting properties measured by a Rapid Visco Analyser (RVA), and their relationship to temperatures during ripening were investigated in a rice cultivar, Yamadanishiki, which was grown under various conditions in the same experimental field. The average temperature after heading was significantly correlated with amylose content and RVA properties, but not with nitrogen content. Under high temperatures during ripening, a decrease in prolamin, which accumulated in type I protein body (PB-I), and an increase in glutelin, which deposits in type II PB (PB-II), were recognized. The ratio of PB-II/PB-I and RVA pasting temperatures were distinctly increased as the temperature became higher. High temperatures during grain ripening would lead to difficulties in digesting steamed rice grains by Aspergillus oryzae, together with ease in digesting rice protein. The average temperature of 11–20 days after heading showed a higher correlation coefficient than that of 1–10 or 21–30 days, implying that temperatures during the middle stage of grain development would be important in determining the rice component that relates to brewing properties.  相似文献

Sclerotinia rot caused by the fungus Sclerotinia sclerotiorum is one of the most serious and damaging diseases of oilseed rape and there is keen worldwide interest to identify Brassica genotypes with resistance to this pathogen. Complete resistance against this pathogen has not been reported in the field, with only partial resistance being observed in some Brassica genotypes. Introgression lines were developed following hybridization of three wild crucifers (viz. Erucastrum cardaminoides, Diplotaxis tenuisiliqua and E. abyssinicum) with B. napus or B. juncea. Their resistance responses were characterized by using a stem inoculation test. Seed of 54 lines of B. napus and B. juncea obtained from Australia, India and China through an Australian Centre for International Agricultural Research (ACIAR) collaboration programme were used as susceptible check comparisons. Introgression lines derived from E. cardaminoides, D. tenuisiliqua and E. abyssinicum had much higher levels (P < 0.001) of resistance compared with the ACIAR germplasm. Median values of stem lesion length of introgression lines derived from the wild species were 1.2, 1.7 and 2.0 cm, respectively, as compared with the ACIAR germplasm where the median value for stem lesion length was 8.7 cm. This is the first report of high levels of resistance against S. sclerotiorum in introgression lines derived from E. cardaminoides, D. tenuisiliqua and E. abyssinicum. The novel sources of resistance identified in this study are a highly valuable resource that can be used in oilseed Brassica breeding programmes to enhance resistance in future B. napus and B. juncea cultivars against Sclerotinia stem rot.  相似文献

Grain protein concentration (GPC) affects wheat nutritional value and several critical parameters for bread and pasta quality. A gene designated Gpc-B1, which is not functional in common and durum wheat cultivars, was recently identified in Triticum turgidum ssp. dicoccoides. The functional allele of Gpc-B1 improves nitrogen remobilization from the straw increasing GPC, but also shortens the grain filling period resulting in reduced grain weight in some genetic backgrounds. We developed isogenic lines for the Gpc-B1 introgression in six hexaploid and two tetraploid wheat genotypes to evaluate its effects on bread-making and pasta quality. In common wheat, the functional Gpc-B1 introgression was associated with significantly higher GPC, water absorption, mixing time and loaf volume, whereas in durum wheat, the introgression resulted in significant increases in GPC, wet gluten, mixing time, and spaghetti firmness, as well as a decrease in cooking loss. On the negative side, the functional Gpc-B1 introgression was associated in some varieties with a significant reduction in grain weight, test weight, and flour yield and significant increases in ash concentration. Significant gene × environment and gene × genotype interactions for most traits stress the need for evaluating the effect of this introgression in particular genotypes and environments.  相似文献

Two purified proteins G-6 and G-4-2 were obtained from Arca subcrenata Lischke using the homogenization, salting-out with ammonium sulfate, ion-exchange chromatography and gel filtration chromatography techniques. The purity of G-6 and G-4-2 was over 96%, as measured by RP-HPLC. G-6 and G-4-2 were measured by SDS-PAGE and IEF-PAGE to have molecular weights of 8.2 kDa and 16.0 kDa, and isoelectric points of 6.6 and 6.1, respectively. The amino acid constituents of G-6 and G-4-2 were also determined. The existence of saccharides in G-6 was demonstrated by the phenol-sulfuric acid method. G-6 and G-4-2 inhibited the proliferation of human tumor cells in vitro. By MTT assay, the IC50 values of G-4-2 were 22.9 μg/mL, 46.1 μg/mL and 57.7 μg/mL against Hela, HL-60 and KB cell lines, respectively, and the IC50 value of G-6 against HL-60 cell line was measured to be 123.2 μg/mL.  相似文献

The influences of the insecticides quinalphos, chlorpyriphos, methyl parathion, endosulfan, imidacloprid and deltamethrin applied three times at 10 days intervals at half of their recommended field concentrations to potted plants of a planthopper-susceptible and a planthopper-resistant rice variety, on reproduction and survival of whitebacked planthopper, Sogatella furcifera (Horvath), and on the chemistry of its host plant, were investigated. Methyl parathion, deltamethrin and quinalphos enhanced the fecundity of the hopper (164-211 vs. 131 for the control) and consequently the resurgence ratio (increased up to 1.75 fold). Methyl parathion and deltamethrin significantly increased nymphal survival (59.3 vs. 52.2% for the control) and the growth index (4.8 vs. 4.2 for the control) of the hopper. The sex ratios of adults emerging on methyl parathion- and deltamethrin-treated rice plants increased in favour of females (1.51 vs. 1.15 for the control) on the susceptible variety but was not altered on the resistant variety. Biochemical analyses of the rice leaves revealed significantly higher quantities of reducing sugars, proteins and amino acids, but lower amounts of total phenols in leaf sheaths and blades of methyl parathion-, deltamethrin- and quinalphos-treated plants of the two varieties. The leaf sheaths and blades of the resistant variety contained more than twice the total phenols present in those of the susceptible variety. The results showed a positive correlation between increased levels of reducing sugars, amino acids and proteins in the rice plants and the fecundity of S. furcifera and a negative correlation between phenol contents of the rice plants and fecundity.  相似文献

Randomly amplified polymorphic DNA markers (RAPD) were employed to assess the level of genetic stability of long term micropropagated prickly pear (Opuntia ficus-indica) plantlets.Thirteen micropropagated plantlets were chosen from a clonal collection of shoots that originated from a single mother shoot. This clonal collection had been maintained under in vitro culture conditions for at least 5 years, as achieved for the time by axillary branch multiplication in Opuntia ficus-indica.Twenty arbitrary primers were used to compare RAPD patterns between in vitro raised material and the mother plant. Only 11 primers were found to yield distinct and reproducible amplification products resulting in a total of 87 amplified products, out of which 82 bands were monomorphic across all the plantlets and 5 showed polymorphisms.Cluster analysis performed on the basis of similarity indices indicated that all micropropagated plantlets and their mother plant grouped together in one major cluster with a 91% level of similarity.Low level of genetic variation has been detected, as polymorphic bands accounted for just 2.79% of the total genetic variation. This very low level of genetic variation, despite more than 5 years of in vitro culture, demonstrates the genetic stability of Opuntia ficus-indica and indicates that the axillary branch multiplication method is highly reliable for the multiplication of genetically true-to-type plant material.The high degree of clonal fidelity detected here, recommend the use of axillary-branching micropropagation technique for the safe in vitro conservation of prickly pear interesting genetic resources.  相似文献
