Effect of selenium on sheep lymphocyte responses to mitogens   总被引:1,自引:0,他引:1  
The effect of selenium (Se) on sheep lymphocyte response to mitogens was studied. In an indoor experiment lambs were fed a basal diet containing 0.13 mg Se kg-1, and supplemented with, respectively, 0.1 or 0.5 mg Se kg-1, either as sodium selenite or as selenomethionine. Enhancement of the proliferative response of lymphocytes to phytohaemagglutinin (PHA), pokeweed mitogen (PWM) and concanavalin A was found in lambs following selenium supplementation at the lower levels. The highest dietary selenium content, however, induced decreased mitogen response. Transient increases in lymphocyte response to PHA and PWM by ewes supplemented with selenium was demonstrated in one field study and a combined effect of selenium and vitamin E was seen in another. There was no stimulatory effect on the mitogen response of lymphocytes from sheep supplemented with dietary vitamin E alone.  相似文献

Pregnant ewes were supplemented with dl-alpha-tocopheryl acetate, either as a single intramuscular dose (500 mg two weeks before lambing) or perorally (150 mg daily during 3-4 weeks before lambing). Ewes without such a supplementation were controls. The vitamin E supplemented ewes had nearly twice as high vitamin E (alpha-tocopherol) concentrations as the unsupplemented control ewes at lambing both in serum and in colostrum. The vitamin E concentration in colostrum was 5-11 higher than in milk 1 week after lambing. Both supplementations somewhat increased the vitamin E serum concentration of the newborn lambs, but the increase was negligible in comparison with the effect produced by the consumption of colostrum. All lambs had very low serum concentrations at birth. The lambs from the supplemented ewes had significantly higher serum values than the control lambs 24 h after birth. The ewes had somewhat higher selenium status at birth than their offsprings when evaluated by glutathione peroxidase (GSH-Px) in the erythrocytes. It seems reasonable that nutritional muscular degeneration may arise in newborn lambs with a normal selenium status if their vitamin E status is critical, either because of an inadequate consumption of colostrum or because of a vitamin E deficient diet during pregnancy with a low vitamin concentration of colostrum as a consequence.  相似文献

Twenty-one 6 months old female lambs were divided into 7 groups and fed a basal diet containing 0.13 mg Se/kg. The basal diet was further supplemented with 0, 0.1, 0.5 or 1.0 mg Se/kg either as sodium selenite or as selenomethionine, and was fed for 10 weeks. Both feed additives produced an increase in the selenium concentration in the tissues analysed. Significant correlations were found between the concentrations of selenomethionine or sodium selenite added to the feed and the subsequent tissue levels. However, the selenium levels seemed to plateau at approximately 0.5 mg Se/kg of supplemented sodium selenite. The total glutathione peroxidase (GSH-Px) activity of the tissues increased when the selenium supplementation increased from 0 to 0.1 mg/kg for both selenium compounds. With further increase in selenium supplementation the GSH-Px activity in the tissues plateaued except in the blood where the activity continued to rise with increasing selenomethionine supplementation. The selenium dependent GSH-Px activity in the liver rose with increasing selenomethionine supplementation, but approached a plateau when 0.1 mg Se/kg as sodium selenite was added to the feed. The selenium concentration in whole blood responded more rapidly to the selenium supplementation than did GSH-Px activity. The experiment indicates that the optimal selenium concentration in the feed is considerably higher than 0.1 mg Se/kg, and that selenium levels of 1.0 mg/kg in the feed do not result in any risk for the animals or the consumers of the products.Key words: dietary selenium, lambs, selenium concentrations, glutathione peroxidase activities, tissues  相似文献

Nineteen multiparous late-pregnant dairy cows were divided into an experimental group (n = 10) and a control group (n = 9). Animals in the experimental group were fed a diet supplemented with chromium chelate admixed into wheat meal at 5 mg per animal per day. The supplementation was started 4 weeks before calving and stopped by the end of week 3 after calving. All the cows were vaccinated with tetanus toxoid in the fifth and sixth week of lactation. Monitoring of the characteristics of non-specific immunity did not show any significant between-group differences in total and differential leucocyte counts, percentages of lymphocyte subpopulations, activities of lectin-stimulated lymphocytes, phagocytic activities, and the contents of total immunoglobulins and lysozyme in blood sera. Tetanus toxoid-specific antibody titres, those of the IgG2 isotype in particular, were higher in the experimental group than in the control animals. These results indicate that chromium supplementation at the level used in this experiment modulated the regulation of functions of the immune system.  相似文献

A 2 x 2 factorial arrangement of treatments with 78 mature ewes was used to evaluate the effects of supplementing the pregnant ewe's diet with high levels of minerals and vitamin E on immunoglobulin G (IgG) absorption by the lamb and whether any altered efficacy of IgG absorption was due to the colostrum or to the lamb. The ewes were estrus-synchronized in October and housed in wk 10 of gestation. In the final 7 wk of gestation, a grass silage-based diet, offered ad libitum, was supplemented with 500 g of a 19% CP concentrate, and from 1 wk later until lambing, half the ewes was offered 48 g of a mineral/vitamin supplement containing 6.5 g of Ca, 4.9 g of P, 5.9 g of Mg, 4.0 g of Na, 790 mg of Zn, 3.5 mg of Se, 40 mg of I, 200 mg of Mn, 20 mg of Co, and 40 IU of vitamin E. At birth, the lambs were allocated to one of four treatments in a 2 x 2 factorial arrangement, with lamb origin and colostrum origin as the two factors. The lambs born to ewes not offered the mineral supplement were fed colostrum obtained from their own dams or from ewes in the mineral-supplemented treatment, whereas lambs born to ewes given supplemental minerals were fed colostrum obtained either from their dams or from ewes in the control treatment. The ewes were milked at 1, 10, and 18 h postpartum and the lambs were fed using a stomach tube. A 5-mL blood sample was taken from each lamb at 24 h postpartum for IgG analysis. The level of fecal adhesion to the upper tail/rump area of the lamb was subjectively scored at 72 h postpartum. There was no difference in gestation length, lamb birth weight, colostrum yield, or IgG production (P = 0.16 to 0.82). When ewes were fed supplemental minerals, the serum IgG content of the progeny was lower than in their control counterparts (6.8 vs. 16.1 g/L; P < 0.001), regardless of whether the lamb received colostrum from ewes with or without access to supplementary minerals. The difference in serum IgG concentrations at 24 h postpartum was a direct reflection of a compromised efficiency in IgG absorption. The progeny of ewes with access to minerals had higher (P < 0.05) levels of fecal adhesion, which was not related to the origin of the colostrum, indicating altered digestive function in these lambs. We conclude, using the sheep as a model, that high mineral intakes in late pregnancy not only lower serum IgG concentrations in the lamb, but also that high mineral intakes result in the neonate being preprogrammed at birth so that it is born with a compromised ability to absorb colostral IgG.  相似文献

AIM: To determine concurrent changes in serum methylmalonic acid (MMA) and vitamin B12 concentrations of ewes and their lambs on cobalt-deficient properties, subsequent to cobalt supplementation. METHODS: Three experiments were carried out on two farms. Groups of ewes (n=25-50) were either supplemented with cobalt bullets during late pregnancy, 23-47 days before the mean lambing date, or left unsupplemented. In two experiments, lambs from within each group were supplemented directly by vitamin B12 injection at 3-weekly intervals from birth, and in the third experiment by injection with micro-encapsulated vitamin B12 at tailing and 3 months later. Pasture samples were obtained for analysis of cobalt content at each sampling time. Blood samples were obtained and liveweight recorded from ewes and lambs at approximately monthly intervals. On one farm (two experiments), liver and milk samples were obtained from ewes and liver samples from lambs. RESULTS: Serum vitamin B12 concentrations in unsupplemented ewes fell below 250 pmol/L during early lactation in all experiments and mean concentrations as low as 100 pmol/L were recorded. MMA concentration was maintained below 2 micromol/L in serum from supplemented ewes but increased to mean concentrations ranging from 7 to 14 micromol/L at the nadir of serum vitamin B12 concentration during peak lactation. A significant liveweight response to supplementation was recorded in ewes on one property, and the vitamin B12 concentration in the ewes' milk and in the livers of their lambs more than doubled. No liveweight-gain response to supplementation was observed in lambs on this property. Mean serum MMA concentrations in lambs ranged from <2 in supplemented, to 19.2 micromol/L in unsupplemented lambs, and the latter had concurrent serum vitamin B12 concentrations of >300 pmol/L. Pasture cobalt concentration was lowest at 0.04-0.09 microg/kg dry matter (DM) on the property on which responses in lambs occurred but considerably higher (>0.09 microg/kg DM) on the property on which responses in ewes occurred. On the second property, serum vitamin B12 concentrations in lambs at tailing were extremely low (100 pmol/L), irrespective of supplementation of dams with cobalt. Mean serum MMA concentration was increased to 20 and 42 micromol/L in lambs from supplemented and non-supplemented ewes, respectively. Weight-gain response to direct supplementation of lambs with vitamin B12 occurred during suckling in the latter, but not the former. Lambs from ewes supplemented with vitamin B12 showed a much bigger increase in serum vitamin B12 concentrations a month after supplementation than did lambs from unsupplemented ewes (+1,400 pmol/L vs + 650 pmol/L). CONCLUSIONS: Serum MMA concentration gave a more precise indication of responsiveness to vitamin B12 or cobalt supplementation than serum vitamin B12 concentrations in ewes and lambs. Neither very low serum vitamin B12 nor elevated MMA concentrations were necessarily indicative of responsiveness to supplementation in suckling lambs, but the latter gave an early indication of impending responsiveness. Supplementation of the ewe with a cobalt bullet appeared to protect the growth performance of the lamb for 90 days and influence the subsequent serum vitamin B12 response in the lamb to vitamin B12 supplementation. CLINICAL SIGNIFICANCE: Supplementing ewes with cobalt bullets in late pregnancy can improve the vitamin B12 status of their lambs, and modify their response to vitamin B12 supplementation.  相似文献

AIM: To determine the effect of increasing doses of long-acting injectable vitamin B12 plus selenium (Se) given pre-mating on the vitamin B12 and Se status of ewes and their lambs from birth to weaning. METHODS: Four groups of 24 Poll Dorset ewes each were injected 4 weeks pre-mating with different doses of a long-acting vitamin B12 + Se product, containing 3 mg vitamin B12 and 12 mg Se per ml. The treatment groups received 5 ml (15 mg vitamin B12 + 60 mg Se), 4 ml (12 mg vitamin B12 + 48 mg Se), 3 ml (9 mg vitamin B12 + 36 mg Se), or no vitamin B12 or Se (control). Twelve of the twin-bearing ewes per group were selected for the study. Efficacy of the product was evaluated from changes in the concentrations of vitamin B12 in serum and liver, and of Se in blood, liver and milk in the ewes during gestation and lactation, and in their lambs from birth to weaning. Pasture samples in paddocks grazed by the ewes and lambs were collected at about 2-monthly intervals from 200-m transects. RESULTS: The flock was Se-deficient, as the mean initial concentration of Se in the blood of ewes was 182 (SE 20.3) nmol/L. Compared with untreated controls, all doses significantly (p < 0.01) increased concentrations of Se in the blood of ewes for at least 300 days. Selenium concentrations in milk were likewise increased throughout lactation, as were those in the blood and liver of lambs. The mean concentration of vitamin B12 in the serum of ewes was initially > 1,000 pmol/L, but this decreased within 28 days to < 460 pmol/L. Treatment with the 5-ml and 4-ml doses raised serum vitamin B12 concentrations of ewes for at least 176 days (p < 0.01), while their lambs had significantly greater concentrations of vitamin B12 in serum and liver for less than 37 days after birth. Tissue concentrations and duration of elevation of both vitamin B12 and Se were proportional to the dose administered. The mean concentrations of Se and cobalt (Co) in the pastures were 32 and 74 microg/kg dry matter (DM), respectively. CONCLUSIONS: Injecting ewes from a Se-deficient flock 4 weeks prior to mating with 48 or 60 mg Se and 12 or 15 mg vitamin B12 increased and maintained the Se status of ewes for at least 300 days, and of their lambs from birth to weaning. The vitamin B12 status of ewes was increased for at least 176 days and that of their lambs for less than 37 days. Due to the proportional nature of the response to increasing dosage, the dose rate of the formulation tested can be adjusted according to the severity of Se and Co deficiency in a flock. CLINICAL SIGNIFICANCE: A single subcutaneous injection of vitamin B12 + Se administered pre-mating to Se-deficient flocks is likely to prevent Se deficiency in ewes and their lambs until weaning, as well as increase the vitamin B12 status of ewes and their lambs until 5 weeks after lambing.  相似文献

This study was conducted to determine the effect of dietary selenium (Se) and vitamin E (Vit E) on various blood characteristics and the primary and secondary humoral immune response of lambs challenged with parainfluenza3 virus (PI3 V). Treatments included: 1) +Se/+Vit E, 2) -Se/+Vit E, 3) +Se/-Vit E and 4) -Se/-Vit E. The basal diet (-Se/-Vit E) was deficient in Se and Vit E. Sodium selenite (.2 mg Se/kg diet) and alpha-tocopherol acetate (20 mg Vit E/kg diet) were added to +Se and +Vit E diets, respectively, to provide adequate levels of each according to NRC recommendations. Following a 10-wk dietary adaptation and depletion period, lambs in all treatment groups were intratracheally inoculated with PI3 V on d 0 and 35 of the 70-d study. Prior to inoculation, whole blood and plasma glutathione peroxidase (GSH-Px) activities were higher (P less than .01) for +Se lambs. Whole blood and plasma GSH-Px increased (P less than .01) after primary viral inoculation in +Se lambs but not in -Se lambs. Serum immunoglobulin M (IgM) concentrations were enhanced (P less than .05) by Se supplementation on d 14, 35 and 49 of the study. Selenium and (or) Vit E did not affect serum immunoglobulin G (IgG) levels. Serum PI3 V antibody titers increased after inoculation on d 0 and 35 in all treatment groups. In trials conducted in 1984 and 1985, the effects of selenium injections on the antibody responses of lambs marginally deficient in selenium to Salmonella dublin were assessed. Control lambs (mean glutathione peroxidase [GSHPx] less than 20 iu/ml) responded well to the vaccine. No difference was observed in the antibody titres of animals treated twice with 5 mg selenium as sodium selenate. During the following season animals given 50 mg selenium as barium selenate showed slightly higher titres than controls. The results show that marginally selenium deficient lambs are able to elicit strong antibody titres to a bacterial antigen and that supplementation with selenium produces, at best, a marginal enhancement of the responses observed.  相似文献

1. The effects of selenium and vitamin E supplementation on some immune parameters were investigated in broilers. 2. Broiler chicks were fed on maize-soybean diets with different concentrations of vitamin E (0-200 mg/kg) and selenium (0-0.2 mg/kg diet) either alone or in combinations from 1 to 42 d of age. 3. Chicks were immunised against Newcastle disease virus (NDV) vaccine at 21 d of age and haemagglutination inhibition (HI) titres were determined after 10 d. 4. Chicks receiving supplements of 200 mg vitamin E/kg and 0.2 mg selenium/kg produced significantly higher HI antibody titres. This was associated with an increased serum concentration of total immunoglobulins and circulatory immune complexes. 5. The chicks given 200 mg vitamin E/kg and 0.2 mg selenium/kg had significantly heavier spleen and bursa. 6. These results suggested that vitamin E and selenium have synergistic effects on immune responses.  相似文献

1. The effect of increasing dietary selenium (Se) on production performance and immune responses in growing (0 to 6 weeks) Japanese quail was investigated. 2. One-day-old chicks (240) were randomly selected and divided into 12 groups with 20 chicks in each group (3 dietary treatments x 4 replicates). The basal diet contained 0.2 mg Se/kg and the two experimental diets were supplemented with 0.5 and 1.0 mg Se/kg. 3. Body weight gain, food intake and food conversion ratio and mortality were not affected by Se supplementation. 4. On d 28, antibody responses to inoculated sheep red blood cells were determined. Antibody titres were significantly higher after feeding the two Se-supplemented diets. 5. During week 4, the response to intradermally injected phytohaemagglutinin, an index of the in vivo cell-mediated immune response, was shown to be increased in the groups fed on the Se-supplemented diets. 6. After 6 weeks, the relative weights of the bursa of Fabricius and thymus were greater in the chicks given the Se-supplemented diets but there was no effect on the relative weight of spleen and liver. 7. It is concluded that supplementing the diet with Se has a beneficial effect on immune responses but does not affect production performance in growing Japanese quail.  相似文献

The main purpose of the study was to investigate whether the feeding system applied has any effect on the status of blood selenium (Se) and vitamins A and E in dairy sheep. In total 200 dairy sheep from 10 flocks were used in the study (20 animals per flock). Group A consisted of 100 sheep (five flocks) reared under the intensive feeding system and group B of 100 sheep (five flocks) reared under the semi-intensive feeding system. The 100 sheep of each group consisted of 25 lambs aged 3-6 months, 25 ewes 1-3 years, 25 ewes more than 3 years and 25 non-lactating ewes in late gestation. Another purpose was to evaluate the potential effect of the age and the reproductive stage of the animals on these parameters. To determine the effect of age, 150 of these animals were divided into three subgroups: 50 lambs, 50 non-pregnant lactating ewes aged 1-3 years and 50 non-pregnant lactating ewes aged more than 3 years. For the evaluation of the effect of the reproductive stage the 50 non-lactating ewes in late gestation and the 100 non-pregnant lactating ewes were used. Blood samplings were performed once, between December and January for non-lactating ewes in late gestation and March to May for lambs and lactating ewes. Whole blood Se and vitamin E and A serum concentrations were determined. The main conclusion is that the feeding system significantly affects Se and serum vitamin A concentration, as they were higher in the intensive one. It was secondly concluded that age affects the serum concentrations of vitamin A.  相似文献

1. This study compared the effect of dietary supplementation with organic or inorganic selenium (Se) sources plus control amounts or large amounts of vitamin E (alpha-tocopherol acetate) in broilers raised at control (20 to 24 degrees C) or low (14.5 to 16.8 degrees C) temperatures after 2 weeks of age. 2. The following dietary treatments were used from one day old. Diet 1, the control diet, comprised a commercial diet containing 0.15 mg/kg inorganic Se and 50 mg vitamin E/kg feed. Diet 2 was the same as diet 1, supplemented with 0.15 mg/kg inorganic Se. Diet 3 was the same as diet 2 but was supplemented with 200 mg/kg vitamin E. Diet 4 was the same as diet 1, but inorganic Se was replaced with 0.30 mg/kg organic Se. Diet 5 was the same as diet 4, supplemented with 200 mg/kg vitamin E. 3. Low temperature reduced the growth rate of broilers; however, at 6 weeks, there were no differences in the body weights of birds fed on organic Se supplemented diets housed at low or control temperature. The feed conversion ratio was significantly affected by low temperature but not by diet. The heterophil/lymphocyte ratio was higher in chicks after one week in the cold, indicating mild stress. Blood triiodothyronine levels were significantly higher in birds after 1 and 4 weeks in the cold but thyroxin was not affected. 4. Organic Se supplementation increased relative lung weight at the control temperature, which might lead to greater respiratory capacity. Relative spleen weight significantly decreased in broilers fed diets supplemented with inorganic Se under cold conditions, a possible indication of chronic oxidative stress. 5. At the low temperature, supplementation with organic Se alone, or with inorganic Se and vitamin E increased glutathione peroxidase (GSHPx) activity and glutathione (GSH) concentration in the liver of broilers, which may indicate increased activity of birds' antioxidant defence against suboptimal environments.  相似文献

The effect of dietary vitamin E, selenium (Se) and their different combinations on body weight gain, food consumption, food conversion efficiency, leukocyte migration inhibition and antibody production was determined in broilers. 2. Chicks were fed on maize-soya bean based diets with concentrations of supplemental vitamin E varying from 0 to 300 IU/kg and selenium concentrations varying from 0 to 1 mg/kg either alone or in combination from 1 to 42 d of age. 3. The chicks were immunised for Newcastle Disease Virus (NDV) vaccine at 21 d. Per cent leukocyte migration inhibition (LMI) was studied on 42 d. Antibodies to NDV in serum were determined at 10 and 21 d post immunisation (PI). 4. Chicks receiving Se, 1 mg/kg and vitamin E 300 IU/kg had significantly higher cellular immune responses in terms of per cent LMI. 5. Maximum body weight gain and best efficiency of food utilisation were obtained in chicks fed diets containing 0.50 mg/kg Se and 300 IU/kg vitamin E. 6. Significantly higher antibody titres (HI and ELISA) at 10 d PI were attributed to 0.06 mg/kg and 150 IU/kg Se and vitamin E, respectively. 7. These data suggest that optimum growth and immune response may be achieved at supplemental level of Se of 0.06 mg/kg and vitamin E at 150 IU/kg. The vitamin E level is higher than that recommended by NRC (1984, 1994).  相似文献

Fifteen horses used for serum production were maintained on low vitamin E and selenium diets. They were divided into four groups receiving: Group 1 no supplements, Group 2 vitamin E, Group 3 selenium and Group 4 both vitamin E and selenium. The humoral immune response to novel antigens, such as tetanus toxoid and equine influenza virus, was increased in groups receiving either vitamin E or selenium/vitamin E. No effects were recorded on the titres against Escherichia coli or the levels of immunoglobulin G.  相似文献

To investigate the influence of maternal Se supply and plane of nutrition on lamb morbidity, mortality, and passive transfer of IgG, pregnant ewe lambs were used in 2 experiments with 2 × 3 factorial treatment arrangements. Supplementation of Se began at breeding and was either adequate Se (ASe, 9.5 μg/kg of BW) or high Se (HSe, 81.8 μg/kg of BW) in Exp. 1 or ASe (11.5 μg/kg of BW) or HSe (77.0 μg/kg of BW) in Exp. 2. On d 50 or 40 of gestation for Exp. 1 or 2, respectively, ewes were assigned randomly to 1 of 3 nutritional planes: 60% (RES), 100% (control, CON), or 140% (HI) of NRC requirements. This resulted in the following treatments: ASe-RES, ASe-CON, ASe-HI, HSe-RES, HSe-CON, and HSe-HI. Upon parturition, lambs were separated from their dams and serum samples obtained. Lambs were fed artificial colostrum for the first 20 h and then placed on milk replacer and grain pellets until completion of the study (Exp. 1, 57 d; Exp. 2, 21 d). Twenty-four hours after parturition, lamb serum samples were collected for IgG analysis. All lambs were reared similarly and morbidity and mortality assessed. Main effects were considered significant when P ≤ 0.05. In Exp. 1, there was a Se × plane of nutrition interaction (P ≤ 0.01) for lamb morbidity from birth to weaning and for 24-h IgG concentration. Lambs from ASe-RES and HSe-HI ewes were treated more frequently (P < 0.01) for respiratory and gastrointestinal disease, and lambs from HSe-HI ewes had the smallest (P < 0.01) 24-h serum IgG concentration. In Exp. 1, lambs from HI ewes also had the greatest (P < 0.01) mortality rates from birth to weaning compared with lambs from CON and RES ewes. In Exp. 2, there was an effect (P < 0.01) of maternal plane of nutrition with lambs from RES ewes having increased 24-h IgG compared with lambs from CON and HI ewes. There was no effect of maternal Se supplementation on lamb 24-h IgG in Exp. 2; however, there was a Se × plane of nutrition interaction (P < 0.01) for morbidity. From birth to 21 d of age, lambs from ASe-CON ewes had fewer (P < 0.01) treatment days compared with lambs from any of the other treatment groups. There also tended (P = 0.08) to be an effect of maternal Se supplementation on lamb mortality with increased mortality observed in lambs from HSe ewes. Results from the studies show a restricted maternal plane of nutrition can increase lamb serum IgG concentration. Selenium results were not consistent between the 2 experiments and may be due to differences in maternal Se.  相似文献

Forty-eight Blackbelly X Dorset, 27 Finnish, 26 Finnish X Dorset, 28 Rambouillet and 8 Dorset Suffolk-sired lambs were used in this experiment. Three weeks before lambing, one-half of the ewes received a selenium emulsion (Se-E) containing .05 mg selenium and 3.7 IU of vitamin E/kg body weight (BW). A 2 X 3 X 2 factorial arrangement was used; lambs from either treated or nontreated ewes were randomly assigned irrespective of breed to one of six treatment combinations consisting of 0 or .025 mg/kg BW selenium (Se) injected at birth or two .025 mg/kg BW Se injections, one at birth and one 2 to 3 wk later, and two levels of injectable Vitamin E (E; 0 and 100 IU) given at birth. Both lambs and ewes were provided access to 75% concentrate diets supplemented with Se and E at recommended NRC levels. Plasma activity of creatine phosphokinase (CPK) was highest at 1 d of age and exhibited decreases (P less than .001) over time. In lambs, the E injection tended to decrease plasma activity of CPK. Plasma glutathione peroxidase activity was lowest at 1 d of age and increased over the course of the experiment but was unaffected by treatments (P less than .05). Plasma tocopherol concentration decreased (P less than .01) with time, with E therapy tending to increase tocopherol concentration. Differences in mean plasma tocopherol concentrations among breeds were also observed (P less than .01). Selenium concentration increased over time and with the E injection (P less than .01). An interaction between ewe and lamb Se-E treatments also was observed (P less than .10), with nontreated lambs from nontreated ewes exhibiting lower Se concentrations than treated lambs from injected ewes. An increase in lamb plasma Se concentration was noted in response to Se-E treatments (P less than .001). In the ewes, plasma tocopherol concentration was lower while Se concentration was higher at 18 d than at 1 d postpartum (P less than .01 and P less than .001, respectively). Milk Se concentration was lower at 18 d than at 1 d (P less than .001) and was higher (P less than .10) in Se-E-treated ewes.  相似文献

An experiment was conducted on 30 male Nellore lambs with average body weight (BW) of 15.45 +/- 0.06 kg to determine the level of the copper (Cu) supplementation in diet from inorganic and organic sources required for optimum immunity and its effect on copper dependent enzymes by allotting them randomly to five groups in completely randomized design. The dietary treatments were viz., basal diet (no Cu supplementation, BD), other four groups were offered BD supplemented with 7 or 14 ppm Cu from copper sulphate (CuSO(4)) and Cu-proteinate, respectively. The lambs were fed the respective diets at 3.5 per cent BW to meet the requirements except Cu for 180 days. The humoral immune response against Brucella abortus and chicken RBC was assessed after 90 days of feeding. The in vivo delayed type hyper sensitivity reaction against PHA-P and in vitro lymphocyte proliferation against Con A indicative of cell mediated immune response (CMI) was assayed at 180 days of feeding. At the end of experiment four lambs from each group were slaughtered for estimation of liver superoxide dismutase activity (SOD). The ceruloplasmin and RBC-SOD activities were higher (P < 0.05) in 14 ppm Cu supplemented lambs from Cu-proteinate at 90 and 180 days, while the liver SOD activity was higher (P < 0.05) in lambs fed 14 ppm Cu from CuSO(4). The STAT titres against B. abortus were higher in Cu supplemented lambs, with no effect of dose of supplementation. Lambs supplemented with Cu-proteinate had higher titers than CuSO(4) on 7 and 14 days of post sensitization. The total immunoglobulin concentration and the CMI response against PHA-P and Con-A was higher (P < 0.05) in lambs fed 14 ppm Cu-proteinae diet. The IgM level was though high in Cu supplemented lambs, no dose or source effect were observed. The study indicated that Cu dependent enzymes activity and immune response were highest and respond better against stress in lambs on 14 ppm supplemented Cu from Cu-proteinate.  相似文献

The effects of dietary restriction of vitamin E (Vit E) and selenium (Se) on lymphocyte proliferation, natural killer (NK) cell activity, antibody-dependent cell-mediated cytotoxicity (ADCC), and on burst respiratory response of stimulated granulocytes as measured by chemiluminescence (CL) were studied in pigs. Six male weanling pigs were maintained for 25 d on a torula yeast-based diet containing no measurable amount of alpha-tocopherol and less than .02 mg of Se per kilogram of feed. Six others received the same basal diet supplemented with 33 IU of DL-alpha-tocopheryl acetate and .2 mg of Se per kilogram of feed. All pigs were inoculated with Salmonella typhisuis on d 21 of the feeding period and killed on d 25. Tests to measure cellular immune functions were performed on cells isolated from blood samples taken on d 21 and 25. After 21 d of feeding, lymphocyte blastogenesis responses to phytohemagglutinin, concanavalin A, and pokeweed mitogen in pigs fed the Vit E- and Se-deficient diet were normal compared with the response in pigs fed the supplemented diet. Moreover, the cytotoxic activity of NK cells, the ADCC response, and the CL response of granulocytes were not affected. After 25 d, a marked suppression of lymphocyte response to mitogens occurred in pigs fed the Vit E- and Se-deficient diet when the cells were cultured in the presence of autologous serum. When fetal bovine serum replaced autologous serum in the cultures, no suppression was observed. No effect on NK activity and ADCC was observed, whereas the CL peak response of granulocytes tended to be higher in pigs fed the deficient diet.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Twenty-four 7-mo-old beef heifers (Charolais Simmental cross), weighing 213 kg, were used to determine the effect of vitamin E (VitE) and(or) selenium (Se) supplementation on the humoral response to a standard dose of Brucella abortus strain 19 vaccine and on the levels of naturally occurring immunoglobulins (Ig) to several antigens. The treatments were as follows: Group 1, no supplement; Group 2, supplementation with 6 g of elemental Se; Group 3, supplementation with 1,400 IU/d of VitE; and Group 4, Se and VitE supplements combined. There were no significant differences in anti-B. abortus IgG1, IgG2, or IgM antibody levels due to Se, VitE or Se/VitE treatments; the concentrations of IgA antibody were too low to be measured with the ELISA test used. Statistical analysis revealed that the levels of total and IgM natural antibody to Salmonella typhimurium were higher in Group 3. Perhaps VitE supplementation given in conjunction with B. abortus vaccine enhanced the production of antibody to S. typhimurium in several animals whose humoral system had been activated by previous exposure to this organism.  相似文献
