Four of 10 heifers that survived an episode of lead poisoning caused by an old broken battery in the pasture were periodically monitored from days 14 to 205 after the episode, with the aim of (i) determining the time-course of lead clearance from blood; and (ii) assessing whether the exposed cattle showed changes in haematological parameters or serum markers of hepatic and renal function (e.g. serum creatinine level) that might be useful as indicators of subclinical toxicity. On the first sampling day, lead levels in blood varied between 0.478 and 0.758 mg/l. The half-life of lead in blood was rather variable, ranging from 68 to 266 days. Despite the high blood lead levels, no significant changes were detected in haematological parameters or hepatic/renal markers, all of which remained within normal ranges. In conclusion, these cattle that survived an episode of lead poisoning may constitute a risk for human consumers, but do not appear to have suffered severe health consequences themselves.  相似文献

Fifteen Holstein dairy cattle were monitored for lymphocyte subsets and expression of adhesion molecules on cells in milk and blood at parturition and at intervals up to 21 days post-partum. Using flow cytometry, we determined percentages of T cells (CD4+, CD8+, gammadelta) and B cells from milk and blood of these cows. We also measured expression of adhesion molecules (CD62L, LFA-1, LPAM-1, and CD44) on lymphocytes in milk and blood. Significantly higher percentages of CD8+ cells were found in milk than in blood at all time points while significantly higher percentages of B cells were found in blood than in milk at all time points. There were minimal to no significant differences in percentages of CD4+ or gammadelta+ cells between milk and blood. Expression of adhesion molecules was consistently higher on all subsets of milk lymphocytes compared with blood lymphocytes. These differences were most pronounced and statistically significant at calving and in the first week following calving. CD62L, LPAM-1 and CD44 were expressed on a significantly higher percentage of lymphocytes in milk at calving than in milk at subsequent sampling times, while LFA-1 expression on lymphocytes in milk was significantly lower at calving than at subsequent times.  相似文献

Twelve dairy cows infected with Mycobacterium avium subsp. paratuberculosis were monitored for lymphocyte subsets and expression of adhesion molecules on cells in blood and milk at parturition and at intervals up to 21 days post-partum. Using fluorescent antibody labeling of cells and analysis by flow cytometry, we determined percentages of T cell subsets (CD4+, CD8+, gammadelta+) and expression of adhesion molecules (CD62L, LFA-1, LPAM-1, and CD44) on cells from blood and milk of these cows. Significantly higher percentages of CD8+ cells were found in milk than in blood at all time points; there were no significant differences in percentages of CD4+ or gammadelta+ cells. CD62L, LFA-1, and LPAM-1 were expressed on a significantly higher percentage of all T cell subsets in milk than in blood at various times after parturition. No differences were seen in expression of CD44. Increased percentages of T lymphocytes expressing adhesion molecules in milk compared to blood suggest that a migratory population of cells is being selectively recruited to the mammary gland from the circulation.  相似文献

Calcium chloride supplements such as gels and drench were studied to determine their effectiveness for increasing blood serum Ca concentrations in periparturient dairy cows. Multiparous, pregnant Holstein dairy cows (n = 36) were assigned to one of four treatments. After calving, cows in four treatments received basal diet and two doses of either control inert gel (CON), gel containing CaCl2 and vitamins (CVG), gel containing CaCl2 and minerals (CMG), or CaCl2 as drench containing vitamins (CVD). The first dose was given within 2 h after calving and the second dose 12 h after the first dose. Each dose provided .07, 54.5, 56.0, and 33.2 g of elemental Ca in CON, CVG, CMG, and CVD treatments, respectively. Blood samples were collected at 0, 15, 30, 60, 180, and 360 min after each oral dose. The blood serum Ca concentrations were 6.26, 7.56, 6.20, and 5.96 mg/dL during the pretreatment period and deviated -13.5, 7.1, 9.3, and 18.1% from pretreatment levels at 18 h after the first dose in CON, CVG, CMG, and CVD treatments, respectively. The average changes in serum P from pretreatment levels were not different among treatments. Serum Mg concentrations remained below the pretreatment levels in all four treatments. Blood serum beta-hydroxybutyrate during the first 2 wk and milk yields during the first 4 wk of lactation were the same in all treatments. Three cases of clinical milk fever were observed in CON treatment and one case in CVD treatment. The oral supplements of CaCl2 as gel or drench increased the blood Ca levels in periparturient dairy cows. Increased supply of Ca through oral supplements of CaCl2 may prevent milk fever in cows that are marginally hypocalcemic.  相似文献

An episode of zinc toxicosis in 95 veal calves caused $29,602 in losses to the producer ($315/affected calf), most of them direct, out-of-pocket losses. Increased calf mortality and increased veterinary service costs accounted for a majority (70%) of the losses. Reduction of losses may have been accomplished by removing the source and increasing the roughage content in the diet, or by shipping the animals for immediate slaughter. Given the magnitude of the financial loss, especially those associated with calf mortality, it was concluded that producers should refrain from using large amounts of mineral supplements unless a deficiency has been diagnosed.  相似文献

Adhesion molecule and homing receptor expression on blood and milk polymorphonuclear leukocytes (PMN) from periparturient dairy cattle was studied. Both percentages and the mean fluorescence intensity (MFI) of PMN expressing CD11a, CD44, CD62L, and LPAM-1 (alpha4 beta7) were evaluated at seven time points during the twenty-one day period post calving. CD11a and CD62L were expressed on 94-100% of PMN in both blood and milk and there were no significant differences in these percentages at any time point. LPAM-1 was expressed on 3-10% of the PMN in the blood and 13-45% in the milk and the percentage of cells expressing LPAM-1 in milk was significantly (P<0.05) greater than in blood at 0, 4, 10, 14, 18 and 21 days after calving. CD44 was expressed on 11-39% of the PMN in blood and 33-69% in the milk and the percentage of cells expressing CD44 in milk was significantly (P<0.05) greater than in blood at all time points. The MFI of CD11a on milk PMN was consistently higher than that of blood PMN throughout the study period and significantly (P<0.05) higher at days 4, 10 and 18 after calving.  相似文献

Lameness is one of the most painful conditions that affects dairy cattle. This study was conducted to evaluate clinical signs and plasma concentration of several pain and stress biomarkers after oligofructose-induced lameness in dairy heifers. Lameness was induced using an oligofructose overload model in 12 non-pregnant heifers. Clinical parameters and blood samples were obtained at 48 and 24 h and at 6, 12, 24, 36 and 48 h after induction of lameness. Clinical parameters included heart rate, respiratory rate, ruminal frequency and lameness score. Plasma biomarkers included cortisol, haptoglobin, norepinephrine, beta-endorphin and substance P. Differences were observed in all parameters between control and treated heifers. The plasma concentration of biomarkers increased significantly in treated animals starting 6 h after induction of lameness, reaching maximum levels at 24 h for cortisol, 48 h for haptoglobin, 6 h for norepinephrine, 12 h for substance P and at 24 h for beta-endorphin. Overall, our results confirm that lameness associated pain induced using the oligofructose model induced changes in clinical parameters and plasma biomarkers of pain and stress in dairy heifers.  相似文献

A practical protocol to study udder immune status in field conditions was planned with the aim to assess different non-specific immune parameters in milk samples from dairy heifers during the periparturient period. Five herds located in northern Italy were selected and overall 39 heifers were enrolled in the trial. Milk samples were taken at 7, 14, 21, 28, 45, 60, and 75 days after calving. The parameters assessed were N-acetyl-beta-glucosaminidase (NAGase), lysozyme, respiratory burst (RB), somatic cell counts (SCC) and serum protein profile. SCC and NAGase were higher in the first sampling after calving, while lysozyme showed large variations during the observation period without a definite trend. The levels of RB observed in the first two weeks after calving, even if lower, were not statistically different from the values observed in samples taken over the following weeks. This study confirmed that the levels of immune components in milk are different from what is observed at blood level in the same cow. A significant decrease in RB in milk polymorphonuclear leukocytes (PMN) post-calving was not observed; milk PMN from healthy cows showed low RB levels, while the values from infected quarters were significantly higher. Significant differences between healthy and infected animals were also observed for milk NAG, lactoglobulin and albumin. These data suggest that udder immune response could be influenced both by the cow immune status and by external factors such as pathogens and management. Therefore, the reduction in immune defences, particularly in heifers, is not unavoidable and methods to boost PMN activity should be explored.  相似文献

Migration of leukocytes into the mammary gland is an essential element of resistance to infection which is likely influenced by expression of adhesion molecules. The contribution of subsets to mammary gland resistance remains unclear. Mononuclear cells from milk and blood of dairy cows were examined for variation in CD4+, CD8+, and WC1+ (Workshop Cluster 1; marker for gammadelta T cells) lymphocyte phenotypes and expression of LFA-1 and L-selectin at several time points during the periparturient period and at Week 16 of lactation. Proportions of CD4+ T cells were higher (p < or = 10.05) in blood than milk at all times between Week 0 and Week 16 relative to calving; the inverse was true of CD8+ cells. Expression of L-selectin was lower (p < or = 0.05) on CD4+ cells and higher on CD8+ cells from milk. The WC1+ subset was more frequent in blood than in milk except at calving when the opposite was true. After calving, proportions of L-selectin+ WC1+ cells decreased steadily to Week 16. Expression of LFA-1 was examined on mononuclear cell populations and found to be lower on milk cells and did not vary over time. We conclude that proportions of T cells subsets differ significantly between blood and milk, particularly around calving. Corresponding variations in L-selectin expression may indicate a role for this molecule in regulating the movement of CD8+ and WC1+ T cells into the bovine mammary gland.  相似文献

Although our previous report demonstrated that adiponectin and AdipoR1 gene expressions changed among different lactation stages in the bovine mammary gland, its in vivo kinetics remain unclear in ruminant animals. In this study, we investigated the changes in circulating concentrations of adiponectin, as well as other metabolic hormones and metabolites, (i) during the periparturient period and (ii) among different lactation stages, in Holstein dairy cows. In experiment 1, serum adiponectin concentrations increased after parturition. Serum insulin concentrations were lower in the postpartum than prepartum period, whereas serum growth hormone (GH) concentrations increased in the postpartum period. Serum nonesterified fatty acids (NEFA) levels were increased during the postpartum period and were dependent on the parity. In experiment 2, there was no significant difference in plasma adiponectin concentrations among lactational stages. Plasma insulin concentrations tended to be lower in early lactation while plasma GH levels tended to be higher. Plasma NEFA concentrations were significantly lower in mid‐ and late‐lactation stages than non‐lactation stages. These findings indicate that elevation of serum adiponectin might be involved in energy metabolism just around parturition, and might exert its action through regulation of receptor expression levels in target tissues in each lactational stage in Holstein dairy cows.  相似文献

AIM: To determine the effect of iodine supplementation on milk production and iodine concentrations in milk for pasture-fed, seasonally calving dairy cows. METHODS: The study was run over two consecutive seasons on the same dairy farm. In Trial One, 294 Friesian dairy cows were either untreated or injected intramuscularly three times with iodised oil (2,370 mg iodine/dose) at the start of lactation (Day 1) and at about 100-day intervals thereafter. Iodine concentrations in milk were determined prior to each injection on Days 1, 98 and 210, and milk, fat and protein yields measured on Days 43, 98, 176 and 238. In Trial Two, 234 Friesian dairy cows were either untreated or injected intramuscularly with iodised oil (2,370 mg iodine/dose) in early lactation (Day 1) and 99 days later. Iodine concentrations in milk were determined prior to treatment on Days 1, 55 and 161. Serum thyroxine (T4) and triiodothyronine (T3) concentrations were determined in the latter two samples and milk, protein and fat yields were determined on Days 25, 119, 182 and 221. In both trials, pasture was sampled at 1- or 3-monthly intervals and iodine concentrations in herbage measured. RESULTS: Iodine concentrations in pasture averaged 0.24 mg/kg dry matter (DM) in both seasons and varied little over the experimental period. Iodine concentrations in milk from unsupplemented cows were <20 microg/L in both trials. Three iodine injections at about 100-day intervals increased iodine concentrations in milk to 58 microg/L for at least 98 days after each treatment. Two iodine injections at about 100-day intervals raised milk iodine concentrations to 160 microg/L and 211 microg/L at least 55 days after each treatment, but had no effect on serum thyroid hormone concentrations. Iodine supplementation had no significant effect on milk, milkfat or milk protein yield. CONCLUSIONS: Pastures in New Zealand that contained 0.24 mg iodine/kg DM appeared to provide an adequate iodine intake for dairy cows. Intramuscular injections of iodine resulted in significant increases in iodine concentrations in milk. CLINICAL SIGNIFICANCE: Increasing iodine concentrations in milk via intramuscular injection of iodine could provide a method for significantly increasing iodine intakes of humans, in particular, children.  相似文献

Regression analyses were performed on data from 48 Compton metabolic profile tests relating the concentrations of certain constituents in the blood of dairy cows to their milk yield, age and stage of lactation. The common partial regression coefficients for milk yield, age and stage of lactation were estimated for each blood constituent. The relationships of greatest statistical significance were between the concentrations of inorganic phosphate and globulin and age, and the concentration of albumin and milk yield.  相似文献

Lead poisoning in cattle and other food animals is of public health significance because of the potential for human exposure to lead through ingestion of contaminated meat and milk products derived from lead-poisoned animals. In Michigan, lead poisoning in livestock is a reportable disease, and positive cattle are quarantined until they test negative (<0.05 ppm blood lead). There is surprisingly little information on blood lead kinetics in cattle. The half-life has been variably reported as 9 weeks and 1-2 months. Because these data did not fit those obtained from cases received at the Michigan State University Animal Health Diagnostic Laboratory, a retrospective study was conducted to review all cases of accidental lead poisoning in cattle between 1990 and 1998. This information is needed to estimate when quarantined lead-poisoned cattle can be released. The results showed that the half-life of blood lead was quite variable and ranged from 48 to 2,507 days. The shortest half-lives (48, 56, and 57 days) were found in a lactating herd of 20-month-old heifers. The longest half-life, 2,507 days, was found in a 9-month-old castrated bull, which ingested a discarded automobile battery. Of the 24 animals monitored, only 8/24 (33%) had half-lives between 6 and 14 weeks. In conclusion, the half-life of blood lead is difficult to predict in accidental cases of cattle poisoning.  相似文献

In dairy cattle, the increase in milk yield has been accompanied by a more negative energy balance (EB) during early lactation and a decrease in fertility. As the hormone leptin is involved in regulation of nutritional status and reproductive function this hormone is an interesting protein to investigate during the periparturient period in dairy cattle. This study was performed to get insight into the function of leptin during the periparturient period and to perform an association study between polymorphisms in the bovine leptin gene and leptin receptor gene and fertility as well as production traits. Leptin concentrations in the periparturient cow undergo remarkable changes; leptin concentrations were high during late pregnancy and declined to a nadir at parturition. Genetic analysis of the leptin gene indicated that a combination of three polymorphisms located at a 135 bp region of the leptin promoter explained most of the variance in prepartum leptin concentrations. The two extreme genotype combinations could be used to investigate the function of leptin concentrations in pregnant cows. A polymorphism located on intron 2 of the leptin gene explained a significant part of the variation in milk yield. On the promoter region of the leptin gene an SNP was detected that was associated with first postpartum luteal activity (FPLA). This SNP could be a candidate marker for fertility in dairy cows. Another SNP on the leptin promoter was associated with energy balance and dry matter intake (DMI) where a higher dry matter intake occurred together with a higher energy balance. Two genotype combinations of the aforementioned three associated SNPs were defined which had a good milk yield together with a good energy balance and fertility. Calculations of an economical value per trait have to validate if one of these genotype combinations would be a possible candidate to be used in selection.  相似文献
