Seven serum samples of 101 samples from nondomestic, captive and free-ranging felids from the United States were indirect fluorescent antibody positive for antibodies to Neospora caninum, whereas 44 samples were positive for antibodies to T. gondii. Although none of the captive animals displayed clinical signs of disease, nondomestic felids in the United States have been exposed to, and are likely infected with, N. caninum and T. gondii. This may have serious implications for zoological gardens exhibiting susceptible animals, such as kangaroos, close to felids. 相似文献

This study was undertaken to determine whether Haemobartonella felis (Mycoplasma haemofelis), the causative bacterial agent of feline infectious anemia, infects nondomestic cats. Routine complete blood count and polymerase chain reaction (PCR) were performed to detect the gene for 16S ribosomal RNA for the organism. Sixty-four blood samples were collected from 54 nondomestic cats, including tigers (Panthera tigris), cheetahs (Acinonyx jubatus), lions (P. leo), mountain lions (Felis concolor), snow leopards (P. unica), and a jaguar (P. onca). Some cats were sampled on two or three different dates. Two tigers were positive for H. felis by PCR analysis. As previously described in domestic cats, the parasitemia appears to be intermittent in nondomestic cats. 相似文献

The purpose of the present study was to determine experimental conditions to stimulate secretion of thyroid hormones (T 3 and T 4) with thyrotropin-releasing hormone (TRH) injections in suckling piglets during the first weeks of postnatal life. Three consecutive experiments were conducted. Four 10–20 d old piglets were i.m. injected with 0, 20, 100, 500 μg (experiment 1) or 0, 4, 20, 100 μg TRH/kg BW (experiment 2) according to a 4 × 4 latin square design involving different litters in each experiment. Blood samples were taken −15, −1, 15, 30, 45, 60, 90, 120 180 and 300 min after TRH injection in experiment 1, and −.25, −.08, .25, .5, 1, 2, 4, 6, 8, 12, 24, 30, 36, 48, 60 and 72 hr after TRH injection in experiment 2. T 3 and T 4 levels were significantly (P<.01) increased as soon as 30 and 45 min after TRH injection, respectively. Maximal levels of T 3 and T 4 were obtained 2 and 4 hr after the injection of 100 μg TRH. T 3 and T 4 returned to basal levels within 6 and 8 hr post injection, respectively. Plasma pGH levels were significantly (P<.001) increased 15 min after TRH injection in piglets injected with 500 μg. In experiment 3, 100 μg TRH/kg BW were injected i.m. either daily or every other day from .0 to 23 days of age. Results showed that T 4 response to TRH did not decrease after repeated injections. These results indicate that daily i.m. injections of 100 μg TRH/kg BW can be used to increase thyroid hormone levels for at least 13 d in the young suckling piglet. 相似文献

Sera from 68 nondomestic captive and free-ranging felids from southern Africa were tested for antibodies to Neospora caninum and Toxoplasma gondii by the indirect fluorescent antibody test. Four of the 68 (5.9%) serum samples were positive for antibodies to N. caninum, with titers ranging from 1:50 to 1:200. All other animals were negative for antibodies to N. caninum at a dilution of 1:50. Fifty of the 68 (74%) serum samples tested positive for antibodies to T. gondii, with titers ranging from 1:50 to 1:26,500. Four animals tested positive for antibodies to both N. caninum and T. gondii. None of these animals displayed clinical signs of disease. Results of this study indicate that nondomestic felids in southern Africa have been exposed to, and are likely infected with, N. caninum and T. gondii. 相似文献

Seroprevalence of Toxoplasma gondii was determined in 865 captive neotropical felids from 20 states from Brazil, sampled from September 1995 to April 1997. Sera were tested by the modified agglutination test (MAT) using formalin-fixed whole tachyzoites and mercaptoethanol. Antibodies (MAT> or =1:20) to T. gondii were found in 472 of 865 (54.6%) cats: in 45 of 99 (45.9%) jaguarundis (Herpailurus yagouaroundi), in 97 of 168 (57.7%) ocelots (Leopardus pardalis), in 68 of 131 (51.9%) oncillas (L. tigrinus), in 35 of 63 (55.5%) margays (L. wiedii), in 1 of 8 (12.5%) Pampas-cat (Oncifelis colocolo), in 9 of 12 (75.0%) Geoffroys-cat (O. geoffroyi), in 134 of 212 (63.2%) jaguars (Panthera onca), and in 83 of 172 (48.2%) pumas (Puma concolor). Antibody titers were: 1:20 in 27 felids, 1:25 in 142 felids, 1:40 in 6 felids, 1:50 in 292 felids, and > or =1:500 in 5 felids. The high seroprevalence of T. gondii antibodies found in the present study suggested a widespread exposure of neotropical cats to T. gondii in zoos in Brazil. The results warrant an investigation on the mode of exposure and oocyst shedding by neotropical cats. 相似文献

OBJECTIVE: To determine the usefulness of measuring serum free thyroxine (T4) concentration as a diagnostic test for hyperthyroidism in cats, and to determine the influence of nonthyroidal disease on free T4 concentration in cats without hyperthyroidism. DESIGN: Prospective case series. ANIMALS: 917 cats with untreated hyperthyroidism, 221 cats with nonthyroidal disease, and 172 clinically normal cats. PROCEDURE: Serum free T4, total T4, and total triiodothyronine (T3) concentrations were measured in cats with untreated hyperthyroidism and cats with nonthyroidal disease. Serum total T4 and T3 concentrations were determined by use of radioimmunoassay, and free T4 concentration was measured by use of direct equilibrium dialysis. Reference ranges for hormone concentrations were established on the basis of results from the 172 clinically normal cats. RESULTS: Sensitivity of serum free T4 concentration as a diagnostic test for hyperthyroidism was significantly higher than the test sensitivity of either total T4 or T3 concentration. Of the 221 cats with nonthyroidal disease, 14 had a high free T4 concentration (ie, false-positive result). Therefore, calculated specificity of measuring serum free T4 concentration as a diagnostic test for hyperthyroidism was significantly lower than test specificity of measuring either the total T4 or T3 concentration. CONCLUSIONS AND CLINICAL RELEVANCE: Results indicate that determination of free T4 concentration is useful in the diagnosis of hyperthyroidism, especially in cats in which hyperthyroidism is suspected but total T4 and T3 concentrations are within reference ranges. However, because some cats with nonthyroidal disease have high serum free T4 concentrations, hyperthyroidism should not be diagnosed solely on the finding of high free T4 concentration. 相似文献

Changes in thyroxine (T4), triiodothyronine (T3), and cortisol during a combined adrenal (dexamethasone suppression/adrenocorticotrophic hormone response test) and thyroid function tests (thyroid-stimulating hormone [TSH] response test) were determined in 20 healthy hospitalized pet dogs. The effect of dexamethasone on T4 and T3 changes was evaluated during a simultaneous TSH response/dexamethasone suppression adrenocorticotrophic hormone response test. Greater ranges in basal cortisol concentrations and slower changes after dexamethasone was administered were observed in healthy pet dogs kenneled in a hospital setting than those reported for conditioned laboratory dogs. Pet dogs were observed to demonstrate cortisol suppression more reliably at 4 hours than at 2 hours after dexamethasone was administered. Dexamethasone had no effect on the response to TSH as assessed by T4 and T3 assays, thus supporting the validity of combining adrenal and thyroid response tests in a 5-hour period. 相似文献

OBJECTIVE: To determine whether administration of phenobarbital, potassium bromide, or both drugs concurrently was associated with abnormalities in baseline serum total thyroxine (T4), triiodothyronine (T3), free T4, or thyrotropin (thyroid-stimulating hormone; TSH) concentrations in epileptic dogs. DESIGN: Prospective case series. ANIMALS: 78 dogs with seizure disorders that did not have any evidence of a thyroid disorder (55 treated with phenobarbital alone, 15 treated with phenobarbital and bromide, and 8 treated with bromide alone) and 150 clinically normal dogs that were not receiving any medication. PROCEDURE: Serum total T4, total T3, free T4, and TSH concentrations, as well as serum concentrations of anticonvulsant drugs, were measured in the 78 dogs with seizure disorders. Reference ranges for hormone concentrations were established on the basis of results from the 150 clinically normal dogs. RESULTS: Total and free T4 concentrations were significantly lower in dogs receiving phenobarbital (alone or with bromide), compared with concentrations in clinically normal dogs. Administration of bromide alone was not associated with low total or free T4 concentration. Total T3 and TSH concentrations did not differ among groups of dogs. CLINICAL IMPLICATIONS: Results indicate that serum total and free T4 concentrations may be low (i.e., in the range typical for dogs with hypothyroidism) in dogs treated with phenobarbital. Serum total T3 and TSH concentrations were not changed significantly in association with phenobarbital administration. ObjectiveTo determine the effectiveness of yohimbine as an antagonist of ketamine-xylazine anaesthesia in captive Asiatic lions ( Panthera leo persica), tigers ( Panthera tigris) and leopards ( Panthera pardus). Study designProspective clinical trial. AnimalsFifty-two healthy adult lions, 55 adult leopards and 16 adult male tigers. MethodsCaptive wild felids in Indian zoos were anaesthetized with a combination of ketamine (2.2-2.6 mg kg ?1) and xylazine (1.1-1.3 mg kg ?1) using a dart propelled from a blowpipe. Time to onset of anaesthesia, lateral recumbency and induction time were measured, and physiological variables (respiration, heart rate and rectal temperature) were recorded once after the onset of complete anaesthesia. Anaesthesia was antagonized at various time periods with an intravenous administration of either 0.1 or 0.15 mg kg ?1 yohimbine. Onset of arousal and time to complete anaesthetic recovery were recorded. ResultsA total of 123 immobilizations were conducted between 2000 and 2005. Anaesthetic induction was achieved in 15-25 minutes in all animals. Incidents of sudden recovery or life-threatening effects associated with immobilizations were not observed. Yohimbine effectively antagonized anaesthesia in all animals within 10 minutes without any excitatory behaviour compared to control animals. No adverse reactions/side effects to yohimbine were recorded except that a few leopards exhibited seizure-like signs for a short period immediately after yohimbine administration. The duration of anaesthesia had no significant effect on the recovery time in any of the animals. Conclusion and clinical relevanceYohimbine antagonized the xylazine portion of ketamine-xylazine anaesthesia and thereby hastened recovery from anaesthesia in Asiatic lions, tigers and leopards. 相似文献

Our objective was to examine the ability of thyroid releasing hormone (TRH) to stimulate not only the release of the thyroid hormones, but also prolactin (PRL) in the female pig. An experiment was conducted to determine the effect of dose and route of administration of TRH on the concentration of PRL and thyroxine (T4) in cyclic gilts. Six gilts were injected with 0, 5, 25, 125, and 625 micrograms TRH and fed 0, 5, 2.5, 12.5 and 62.5 mg TRH. Gilts received TRH once daily. During the 10-day treatment period, route of TRH administration alternated between i.v. injection and feeding. The dose of TRH progressed from the lowest to the highest. Blood samples were taken prior to TRH injection and thereafter at 15-min intervals for 3 hr. Sampling continued for an additional 3 hr at 30-min intervals when TRH was fed. Concentrations of PRL and T4 were determined by radioimmunoassay. Intravenous injection of gilts with 125 and 625 micrograms TRH resulted in an increase in PRL from 0 to 15 min (P less than .05). All doses of TRH given i.v. elevated T4 over a 2-hr period (P less than .01). TRH failed to increase PRL when TRH was fed (P greater than .5). The feeding of 62.5 mg TRH elevated T4 from 0 to 6 hr (P less than .01). Thus, TRH injection increased PRL rapidly and T4 gradually. When TRH was fed, only a gradual elevation in T4 was observed. We conclude that TRH can elicit the release of both PRL and T4 in the cyclic gilt, but magnitude and duration of the PRL and T4 response depends on the dose and route of TRH administration. 相似文献

Two frozen, raw horse meat-based diets fed to captive exotic felids at Brookfield Zoo were irradiated to determine the extent of microbial destruction and whether radiation treatment would affect consumption and/or fecal consistency in exotic cats. Fifteen cats, two African lions (Panthera leo), two Amur tigers (Panthera tigris altaica), one Amur leopard (Panthera pardus orientalis), two clouded leopards (Neofelis nebulosa), two caracals (Felis caracal), one bobcat (Felis rufus), and five fishing cats (Felis viverrinus), housed at Brookfield Zoo were fed nonirradiated and irradiated raw diets containing horse meat with cereal products and fortified with nutrients: Nebraska Brand Feline and/or Canine Diet (Animal Spectrum, North Platte, Nebraska 69103, USA). Baseline data were obtained during a 2-wk control period (nonirradiated diets), which was followed by a 4-wk period of feeding comparable irradiated diets. Feed intake and fecal consistency data were collected. An estimated radiation dose range of 0.5-3.9 kilograys reduced most microbial populations, depending on specific diet and microbe type. Irradiation had no overall effect on either feed consumption or fecal consistency in captive exotic cats, regardless of species, age, sex, or body mass. Data indicate that irradiation of frozen horse meat-based diets (packaged in 2.2-kg portions) result in microbial destruction in these products but that product storage time between irradiation and sampling may also affect microbial reduction. However, irradiation would be an appropriate method for reducing potentially pathologic bacteria in raw meat fed to exotic cats. 相似文献

Enzyme immunoassays (EIAs) for the determination of autoanti-bodies (AA) to thyroid antigens in canine serum were developed. Streptavidin (SA) was immobilized as capture molecule on microtitreplates (MTP). Thyroglobulin (Tg) purified from canine thyroids and the thyroid hormones thyroxine and triiodothyronine (T3 and T4) were conjugated to biotin labelling reagents and attached to the MTP over the SA-biotin bridge. Bound AA were detected with anti-dog-immunoglobulin G (IgG) labelled with horseradish peroxidase. Serum samples from dogs which were allotted to four groups were analysed: A (n = 31), biochemical evidence of hypothyroidism; B (n = 76), clinical signs of hypothyroidism; C (n = 47), euthyroid with non-thyroidal disease; D (n = 186), clinically healthy. The validity of the assays was tested with two different methods. After thiophilic absorption chromatography of positive sera, a positive reaction in the EIA was only detected in those fractions which coeluted with the canine IgG standard. Furthermore, the positive reaction was blocked by the addition of the corresponding antigen. In 55% of the hypothyroid dogs AA to Tg and/or T3 and T4, respectively, were found (up to a titre of 1 : 1600). In group B 34% of the dogs were diagnosed positive, but the titre was lower (up to 1 : 400). In the groups C and D the number of dogs with AA and their titre was significantly lower. Two different methods for distinguishing positive and negative test results were compared in order to increase the specificity of the tests without decreasing the sensitivity. The EIAs are precise and based on high agreement with previous reported assays able to discriminate dogs with thyroiditis from healthy ones. The effect of malabsorption syndrome (stunting or runting syndrome) on the thyroid function of broilers was investigated in control and inoculated broilers from 1 to 29 days of age. The broilers were infected at 1 day of age with intestinal homogenates from chickens naturally suffering from this syndrome. The body weight of inoculated broilers was significantly (P less than 0.05) lower 1 week after inoculation than that of controls. The level of thyroxine in the serum of inoculated birds was lower (P less than 0.05) from day 6 through the remainder of the trial. The level of triiodothyronine of inoculated birds was depressed (P less than 0.05) on day 4, but 1 week later it returned to normal. The earliest phenomenon indicative of disturbance of thyroid function was the significant depression of 5'-deiodination in liver homogenates of inoculated broilers as early as day 2. It is concluded that thyroid function is one of the earliest targets of this syndrome. 相似文献

A high prevalence of fecal Salmonella shedding in a collection of healthy exotic felids precipitated a change to two new commercially available feline diets. One year after initiation of the new diets, 18 fecal samples from individual felines, their exhibits, and representative samples of the diets were cultured for Salmonella spp. Only one culture grew a Salmonella sp. Salmonella uganda was cultured from the feces of one snow leopard (Felis uncia). Feeding a diet with minimal to no Salmonella contamination lowered Salmonella shedding rates in this collection of captive exotic felids. 相似文献

BackgroundExposure to xenoestrogens in humans and animals has gained increasing attention due to the effects of these compounds on reproduction. The present study was undertaken to investigate the influence of low-dose dietary phytoestrogen exposure, i.e. a mixture of genistein, daidzein, biochanin A and formononetin, on the establishment of testosterone production during puberty in male goat kids. MethodsGoat kids at the age of 3 months received either a standard diet or a diet supplemented with phytoestrogens (3 - 4 mg/kg/day) for ~3 months. Plasma testosterone and total and free triiodothyronine (T 3) concentrations were determined weekly. Testicular levels of testosterone and cAMP were measured at the end of the experiment. Repeated measurement analysis of variance using the MIXED procedure on the generated averages, according to the Statistical Analysis System program package (Release 6.12, 1996, SAS Institute Inc., Cary, NC, USA) was carried out. ResultsNo significant difference in plasma testosterone concentration between the groups was detected during the first 7 weeks. However, at the age of 5 months (i.e. October 1, week 8) phytoestrogen-treated animals showed significantly higher testosterone concentrations than control animals (37.5 nmol/l vs 19.1 nmol/l). This elevation was preceded by a rise in plasma total T 3 that occurred on September 17 (week 6). A slightly higher concentration of free T 3 was detected in the phytoestrogen group at the same time point, but it was not until October 8 and 15 (week 9 and 10) that a significant difference was found between the groups. At the termination of the experiment, testicular cAMP levels were significantly lower in goats fed a phytoestrogen-supplemented diet. Phytoestrogen-fed animals also had lower plasma and testicular testosterone concentrations, but these differences were not statistically significant. ConclusionOur findings suggest that phytoestrogens can stimulate testosterone synthesis during puberty in male goats by increasing the secretion of T 3; a hormone known to stimulate Leydig cell steroidogenesis. It is possible that feedback signalling underlies the tendency towards decreased steroid production at the end of the experiment. 相似文献

1. In the studies reported here, broiler lines divergently selected for susceptibility to ascites under low temperature conditions were tested for their sensitivity to 3,3’,5‐triiodothyronine (T3) with respect to growth rate, rate of mortality, plasma concentrations of T3, right ventricular hypertrophy (RVH) and incidence of ascites. 2. Mean body weight of the ascites‐susceptible line (BC‐line) was higher than that of the ascites‐resistant line (A‐line). Adding 0.5 mg T3/kg of the diet depressed growth rate to the same extent in both lines. The effect of T3 on growth was more pronounced for males than for females. 3. T3‐supplementation increased the relative weight of the heart and the incidence of RVH to the same extent in both lines. More of the T3‐treated BC‐line chickens had fluid accumulation in the abdominal cavity than the T3‐treated A‐line chickens. 4. Dietary T3‐treatment depressed the plasma concentration of growth hormone (GH) profoundly and insulin‐like growth factor (IGF‐I) slightly but to the same extent in both lines. The coefficient of variation of GH concentrations indicate that T3 treatment mainly decreased GH‐pulsatility in young growing broilers. 5. Higher doses of dietary T3 (1 and 2 mg/kg) increased mortality in a dose‐dependent manner. With 2 mg T3/kg, mortality in the BC‐line was almost double that in the A‐line. 6. These studies indicate that the development of ascites could be linked with thyroid function. Moreover, dietary T3 supplementation could be used to help identify ascites‐inducing factors or genetic lines with differential sensitivity for ascites. 相似文献
