A general approach was developed to quantify hydroxycinnamic acid derivatives and the glycosides of flavonols and flavones using UV molar relative response factors (MRRFs). More than 90 standards were analyzed by LC-MS and divided into five groups based on the λ(max) of their band I absorbance profiles. For each group, a commercially available standard was chosen as the group reference standard. Response factors were determined for each standard in each group as purchased (MRRF) and, when possible, after vacuum drying (MRRF(D)). The MRRF(D) values for 17 compounds whose λ(max) values fell within ±2 nm of the group reference standard were 1.01 ± 0.03. MRRF values for compounds whose λ(max) values fell within ±10 nm of the group reference standard were 0.96 ± 0.13. Group reference standards were used to quantify 44 compounds in Chinese lettuce, red onion, and white tea. This approach allows quantitation of numerous compounds for which there are no standards.  相似文献

When standard solutions of polynuclear aromatic hydrocarbons (PAHs) were analyzed by capillary column gas chromatography using splitless injections, response factors were observed to be dependent on the solvent used to prepare the standard. This report presents the response factors for 16 individual PAHs in 5 commonly used solvents: acetonitrile, methanol, toluene, isooctane, and cyclohexane. To minimize quantitation errors due to differences in transfer efficiency, samples and standards of PAHs should be prepared in the same solvent.  相似文献

Analytical difficulties in the rapid and accurate determination of diacetyl (DA), an important flavor compound in wine, at low concentrations have been overcome by the use of solid-phase microextraction (SPME) with deuterated diacetyl-d(6) (d6-DA) as an internal standard followed by gas chromatography-mass spectrometry (GC-MS). The GC-MS analyses showed that the values of the ion response ratio of DA to d6-DA were consistent regardless of the conditions of SPME headspace and were not influenced by the presence of sulfur dioxide in wine. The quantitation value of DA was represented as the concentration of free plus bound with sulfur dioxide forms of DA. The detection limit of DA in wine was as low as 0.01 microg/mL with linearity through to 10 microg/mL.  相似文献

Recent changes in European legislation require monitoring of 24 volatile compounds in perfumes as they might elicit skin sensitization. This paper reports a GC-MS quantitation procedure for their determination in fragrance concentrates. GC and MS conditions were optimized for a routine use: analysis within 30 min, solvent and internal standard selection, and stock solution stability. Calibration curves were linear in the range of 2-100 mg/L with coefficients of determination in excess of 0.99. The method was tested using real perfumes spiked with known amounts of reference compounds.  相似文献

Cinnamyl alcohol dehydrogenase (CAD) activity is deficient in loblolly pine (Pinus taeda L.) harboring a mutated allele of the cad gene (cad-n1). We compared lignin structure of CAD-deficient and wild-type pines, both types segregating within full-sib families obtained by controlled crosses. The type and frequency of lignin building units and distribution of interunit bonds were determined from the GC-MS analysis of thioacidolysis monomers and dimers. While the lignin content was only slightly reduced, the lignin structure was dramatically modified by the mutation in both mature and juvenile trees. Lignins from CAD-deficient pine displayed unusually high levels of coniferaldehyde and dihydroconiferyl alcohol. In addition, biphenyl and biphenyl ether bonds were in large excess in these abnormal lignins. These results suggest that the CAD-deficient pines efficiently compensate for the shortage in normal lignin precursors by utilizing nontraditional wall phenolics to construct unusual lignins particularly enriched in resistant interunit bonds.  相似文献

An experimental design has been used to optimize the extraction of volatile compounds from summer truffle aroma (Tuber aestivum) by using headspace solid phase microextraction. The extracted compounds have been analyzed by gas chromatography with a flame ionization detector and by gas chromatography-mass spectrometry (GC-MS). In an attempt to develop an objective method to fully characterize truffle aroma, a fiber of medium polarity (for flavors) was used to avoid discrimination toward very nonpolar and polar volatile compounds. To optimize the extraction conditions, a response surface experimental design was applied considering three factors such as extraction temperature, equilibrium time, and extraction time. From the statistical analysis of the experimental design, it was possible to determine that the most important factor influencing the abundance of aroma compounds was the extraction temperature. Optimal extraction temperature was established at approximately 50 degrees C. By using GC-MS, it was possible to identify 37 compounds, most of them previously described as responsible for truffle aroma.  相似文献

The structures of two previously unknown sesquiterpene alcohols of the potato (Solanum tuberosum) were assigned. The potato alcohols were obtained by steam-distillation, preparative column chromatography, and separation into fractions by HPLC on a silica gel column. The fractions were studied by GC-FID, GC-MS, and NMR spectroscopy. The potato sesquiterpene alcohols were identified as kunzeaol (6-alpha-hydroxygermacra-1(10),4-diene) and ledol. These two compounds were used in feeding tests with larvae and beetles of the Colorado potato beetle (Leptinotarsa decemlineata Say). In a bioassay, kunzeaol was found to act as a feeding attractant for the beetles.  相似文献

Ethyl esters (FAEE) and trideuterium-labeled methyl esters (d3-FAME) of fatty acids were prepared and investigated regarding their suitability as internal standards (IS) for the determination of fatty acids as methyl esters (FAME). On CP-Sil 88, ethyl esters of odd-numbered fatty acids eluted approximately 0.5 min after the respective FAME, and only coelutions with minor FAME were observed. Depending on the problem, one or even many FAEE can be added as IS for the quantification of FAME by both GC-FID and GC-MS. By contrast, d3-FAME coeluted with FAME on the polar GC column, and the use of the former as IS requires application of GC-MS. In the SIM mode, m/z 77 and 90 are suggested for d3-methyl esters of saturated fatty acids, whereas m/z 88 and 101 are recommended for ethyl esters of saturated fatty acids. These m/z values give either no or very low response for FAME and can thus be used for the analysis of FAME in food by GC-MS in the SIM mode. Fatty acids in sunflower oil and mozzarella cheese were quantified using five saturated FAEE as IS. Gravimetric studies showed that the transesterification procedure could be carried out without of loss of fatty acids. GC-EI/MS full scan analysis was suitable for the quantitative determination of all unsaturated fatty acids in both food samples, whereas GC-EI/MS in the SIM mode was particularly valuable for quantifying minor fatty acids. The novel GC-EI/MS/SIM method using fatty acid ethyl esters as internal standards can be used to quantify individual fatty acids only, that is, without determination of all fatty acids (the common 100% method), although this is present. This was demonstrated by the exclusive quantification of selected fatty acids including methyl-branched fatty acids, erucic acid (18:1n-9trans), and polyunsaturated fatty acids in cod liver oil and goat's milk fat.  相似文献

A sensitive method for quantitating the pharmacologically active polyacetylenes panaxynol and panaxydol in Radix Ginseng was developed using a capillary gas chromatography-mass spectrometric (GC-MS) method. The detection mode of selected ion monitoring (SIM) allowed sensitive and selective quantitation of the two compounds in ginseng. Method validation showed that the GC-MS method has much lower detection and quantitation limits than the high-performance liquid chromatography (HPLC)-UV method. This indicates that GC-MS is particularly useful for the analysis of polyacetylene compounds, which have relatively low abundances compared with ginsenosides in ginseng. Based on the quantitative results of different types of ginseng herbs, it was found that the panaxydol and panaxynol contents were higher in forest ginseng than in cultivated ginseng. This method was further applied to the quantitative analyses of panaxynol and panaxydol in Radix Notoginseng and American ginseng. The ratio of panaxydol to panaxynol can be utilized as a marker for differentiating ginseng, notoginseng, and American ginseng. This study introduces the first GC-MS method for the quantitative analysis of polyacetylenes in herbs of the Panax genus.  相似文献

Headspace solid-phase microextraction (SPME), followed by gas chromatography (GC)-mass spectrometry (MS) determination, has been used for the analysis of honey volatiles. Two SPME fibers were employed to study the composition of volatiles from various types of Spanish honeys. The best results were obtained with the Carboxen/PDMS fiber, using a homogenization time of 1 h at 70 degrees C and a sampling period of 30 min. A total of 35 compounds were detected, most of them identified by GC-MS and quantified using external standards. Differences in the composition of honey volatiles were obtained, and these results allowed the differentiation of honeys. However, further studies are necessary to confirm the utility of this technique as an alternative tool for the characterization of the floral origin of honeys.  相似文献

Milled wood and milled wood lignin (MWL) samples were subjected to DFRC and thioacidolysis. Despite the fact that both methods selectively cleave aryl ether bonds, substantial differences in results were obtained. Lignin thioacidolysis gave total molar yields of degradation monomer products in the range of 3.5-7 mol % higher than DFRC. GPC analysis showed that the thioacidolysis-treated lignin was degraded to a lower average molecular weight than that treated by DFRC. Contrary to results reported for lignin model compounds, these results indicate that the DFRC method does not completely or efficiently degrade the lignin polymer. In fact, the DFRC-degraded lignin retained much of the characteristics of the original MWL. Elemental analysis revealed the presence of bromine in the DFRC-treated lignin, and two-dimensional (1)H-(13)C HMQC NMR spectroscopy showed the presence of beta-O-4 linkages in the DFRC-treated lignin. No beta-O-4 interunit linkages were detected in the thioacidolysis-treated lignin. These results are consistent with the lower monomer yields and the higher average molecular weight of the DFRC-treated lignin and indicate inefficiency in the chemistry of the method, probably due to steric constraints of the polymeric nature of lignin.  相似文献

A sensitive gas chromatographic method for the quantitative analysis of zearalenone in blood serum is described. Zearalenone is eluted from blood serum by column chromatography followed by base-acid extraction with dichloromethane as the organic phase. After epicoprostanol (internal standard) is added, the sample is evaporated to dryness, derivatized, and injected onto the gas chromatographic column. A number of silylating agents and reaction conditions were investigated. Derivatizing zearalenone with N-methyl-N-trimethylsilyltrifluoroacetamide in the presence of acetone at room temperature for at least 2 hr gave best results. Sensitivity limit is < 0.5 ng injected, equivalent to 100 ng zearalenone/mL blood serum. A linear standard curve is observed when 0.5-30 ng zearalenone derivative is injected onto the Perkin-Elmer gas chromatograph. For quantitation, a standard curve is prepared by plotting amounts of zearalenone (ng) injected vs. ratios for peak areas of zearalenone and epicoprostanol derivatives. The internal standard procedure improves the precision by minimizing variations in sample injections and detector response. Percent recovery from blood serum is 68-75 in the range of 1.6-8.0 micrograms zearalenone/mL blood.  相似文献

Fresh juices of four Italian cultivars of lemons (Citrus limon Burm) have been analyzed by headspace solid phase microextraction coupled to gas chromatography and gas chromatography-mass spectrometry (GC-MS). The best results have been obtained with the 2 cm 50/30 microm divinylbenzene/carboxen on poly(dimethylsiloxane) fiber, using a homogenization time of 1 h at 40 degrees C and a sampling period of 30 min. A total of 35 volatile compounds have been identified by GC-MS, and their relative amounts have been calculated by adding internal standard to the samples. Differences in composition of lemon juices volatile components have been observed. Verdello Siracusano lemon juice has the highest amount of volatile compounds (50.28 mg/L), followed by Interdonato (8.39 mg/L), Primo Fiore Capo d'Orlando (5.75 mg/L), and Femminello Siracusano (2.62 mg/L) juices. Volatile compounds mainly consist of mono- and sesquiterpene hydrocarbons and oxygenated molecules (aldheydes, monoterpene alcohols, and monoterpene esters). Headspace solid phase microextraction coupled to a gas chromatograph equipped with a specific sulfur detector, a sulfur chemiluminescence detector, let us detect and quantify dimethyl sulfide compound at the microgram/liter level in lemon juices.  相似文献

The present paper describes a liquid chromatographic (LC) method for purification of crude swine tissue extracts before gas chromatographic/mass spectrometric (GC/MS) quantitation and confirmation of sulfamethazine at low ppb levels. Fractions corresponding to sulfamethazine were collected, evaporated to dryness, N-methylated with diazomethane, concentrated, and analyzed by GC/MS. A mass spectrometer was set to selected ion monitoring (SIM) mode. Ions 233, 227, 228, and 92 m/z were detected. Ratio 227/233 m/z (sulfamethazine/internal standard, [phenyl 13C6] sulfamethazine) was used for quantitation, while ratios 228/227 and 92/227 m/z, respectively, were used for confirmation. Quantitation in spiked blank muscle tissue was tested from 100 to 1 ppb and found acceptable at all concentrations studied; coefficients of variations ranged from 4.9 to 14.4%. Similar results were obtained for liver tissue from 5 to 20 ppb; coefficients of variation ranged from 1.2 to 9.1%.  相似文献

A capillary gas chromatographic (GC) method has been developed for the separation and determination of policosanol components extracted from rice bran wax. A Varian CP-sil 8 CB column was employed, and an oven temperature was programmed. Gas chromatography-mass spectrometry (GC-MS) was used to identify the composition of policosanol. Quantitative analysis was carried out by means of hydrogen flame ionization detector (FID) with dinonyl phthalate (DNP) as internal standard. The results indicated that the extract obtained by dry saponification has the highest contents of octacosanol and triacontanol among extracts by all used extraction methods including dry saponification, saponification in alcohol, saponification in water (neutralized and non-neutralized), and transesterification. Meanwhile, the GC-MS fingerprint of policosanol extracted by dry saponification has been established. Euclidean distance similarity calculation showed remarkable consistency of compositions and contents among 12 batches of policosanol from a rice bran wax variety. This protocol provided a rapid and feasible method for quality control of policosanol products.  相似文献

Dynamic headspace sampling was used to collect aroma compounds from raw samples of four carrot (Daucus carota L.) cultivars (Brasilia, Duke, Fancy, and Cortez). The collected volatiles were analyzed by capillary GC-FID and GC-MS using large-volume cool on-column injection (LVI-COC). Of the 36 compounds identified, 6 had not been previously detected in carrots. Significant differences between the carrot cultivars were found for 31 of the identified volatiles as well as for total monoterpenes, sesquiterpenes, and total volatile content. Mono- and sesquiterpenes accounted for about 98% of the total volatile mass in all cultivars. LVI-COC injection was used to determine the loss of carrot volatiles during concentration of headspace samples under a stream of nitrogen. The loss among major monoterpenes in the concentrated samples varied from 16% for p-cymene to >40% for alpha-pinene as compared to nonconcentrated samples. The loss among high-boiling sesquiterpenes varied from not detectable (beta-caryophyllene, alpha-humulene, and caryophyllene oxide) to approximately 7% for (E)- and (Z)-gamma-bisabolene.  相似文献

The volatile compounds of guava wine were isolated by continuous solvent extraction and analyzed by GC-FID and GC-MS. A total of 124 volatile constituents were detected, and 102 of them were positively identified. The composition of guava wine included 52 esters, 24 alcohols, 11 ketones, 7 acids, 6 aldehydes, 6 terpenes, 4 phenols and derivatives, 4 lactones, 4 sulfur-compounds, and 5 miscellaneous compounds. The aroma-active areas in the gas chromatogram were screened by application of the aroma extract dilution analysis and by odor activity values. Twelve odorants were considered as odor-active volatiles: (E)-β-damascenone, ethyl octanoate, ethyl 3-phenylpropanoate, ethyl hexanoate, 3-methylbutyl acetate, 2-methyltetrahydrothiophen-3-one, 2,5-dimethyl-4-methoxy-3(2H)-furanone, ethyl (E)-cinnamate, ethyl butanoate, (E)-cinnamyl acetate, 3-phenylpropyl acetate, and ethyl 2-methylpropanoate.  相似文献

We investigated the coupling of abundance of bacteria, phytoplankton and ciliates with hydrocarbons in the surface water and sediments of five interconnected ponds in the arid Sfax solar salterns. This study aimed at determining the potential sources of hydrocarbons and the effects of salinity gradients on microorganism metabolism. Hydrocarbon analysis was performed by gas chromatography (GC-FID) and gas chromatography coupled with mass spectrometry (GC-MS). The GC-FID allowed the detection of aliphatic hydrocarbons and n-alkanes ranging from n-C13 to n-C30. Total aliphatic hydrocarbon concentrations varied from 92.5 mg. l?1 in the first pond (having marine characteristics) to 661.1 mg. l?1 in the last pond (crystallizer) (316.8?±?120.1 mg. l?1) for water samples and from 26.7 to 127.8 μg. g?1 dry weight for sediment samples. The GC-MS enabled us to detect halogenated hydrocarbons (bromoalkanes and chloroalkanes) and n-alkenes. The distribution of n-alkanes indices coupled to several environmental factors suggests that a major fraction of hydrocarbons resulted from both prokaryotic (bacteria) and eukaryotic (protists) developments. A low hydrocarbon fraction might be petrogenic.  相似文献

Two stable isotope dilution assays were developed for the quantitation of acrolein in fats and oils using [(13)C(3)]-acrolein as the internal standard. First, a direct GC-MS headspace method, followed by an indirect GC-MS method using derivatization with pentafluorophenyl hydrazine, was established. Analysis of six different types of oils varying in their pattern of fatty acids showed significant differences in the amounts of acrolein formed after heating at various temperatures and for various times. For example, after 24 h at 140 °C, coconut oil contained 6.7 mg/kg, whereas linseed oil was highest with 242.3 mg/kg. A comparison of the results showed that the extent of acrolein formation seemed to be correlated with the amount of linolenic acid in the oils. Although the acrolein concentrations were lowered in all six oils after frying of potato crisps, linseed and rapeseed oil still contained the highest amounts of acrolein after frying. By applying both methods on different thermally treated fats and oils, nearly identical quantitative data were obtained.  相似文献
