We studied the growth properties of three different hemoglobin genotypes of juvenile turbot (Scophthalmus maximus) reared at 10, 14, 18 and 22 °C. The genotype Hb-I(2/2) displayed the overall highest growth rate in the temperature range 14–22 °C, whereas no differences were found at sub-optimal temperature (10 °C). The differences in growth were largest at optimal temperatures where the Hb-I(2/2) genotype displayed 13% higher growth than the two other genotypes. Optimal temperature for growth (T opt.G) varied between the genotypes with the genotype Hb-I(2/2) displaying the highest T opt.G (23.0 °C) and Hb-I(1/1) the lowest (19.0 °C). The biological significance of this link between biochemical genetic variation and physiological properties might be influences on growth pattern, ultimate size and age at first maturity. This is the first reported quantitative trait locus (QTL) for this species.  相似文献

The aim of this study was to investigate the oxygen binding properties of the turbot (Scophthalmus maximus) hemoglobin polymorphism with special references to pH and temperature. Hemolysate samples from the three hemoglobin genotypes Hb- I(1/1), Hb-I(1/2) and Hb-I(2/2), were tested at 10, 16 and 19 °C and at pH values 7.2, 7.5 and 7.8 at 100 mm NaCl respectively. Hb-I(2/2) had the highest oxygen affinity at all three temperatures followed by Hb- I(1/2) and Hb-I(1/1). There was a significant decrease in oxygen affinity with increasing temperature and increasing pH in the range 10 to 19 °C for all three genotypes. The genotype Hb-I(1/1) had the highest Bohr effect followed by genotype Hb-I(2/2) and Hb-I(1/2). The effect was highest at 10 °C and decreased with temperature. Temperature sensitivity of the O2 binding for turbot hemoglobin was low and increased in general with increasing pH. It is hypothesised that the low sensitivity hemoglobins may be an adaptation to variable temperature conditions in the distribution area of the species.  相似文献

Four groups of 1+ year-old Atlantic salmon, Salmo salar, pre-smolts were reared under various temperature regimes: constant 10 °C from November onwards; ambient temperature until either 15 December or 1 February, then 10 °C; or ambient temperature throughout (control; 2–3 °C November–March). From 20 May onwards, temperature in all groups was ambient, increasing from 10 °C to 17 °C in late July. Rearing temperature had no significant effect on either the timing of completion of smolting, or the duration of the smolt-window. Mean gill Na+-K+ ATPase activity was not significantly affected by temperature regime; it increased in all groups from < 2.0 mol Pi mg protein–1 h–1 (units) in January to 5–7 units in mid-April, then back to < 2.0 units in July. Survival in 96 h, 37) salinity (S) tolerance tests was similar in all groups, increasing from < 10% in early March, to > 90% from mid-April to mid-June, then decreasing to < 20% by early July. Increased winter temperature significantly increased growth and condition factor compared with the control, but during April–May all four groups exhibited similar temporary decreases in condition factor in association with the completion of smolting.  相似文献

Theeffect of temperature on scope for growth was determined in juveniles of thenorthern scallop Argopecten purpuratus (Lamarck, 1819).This economically important species, has been introduced into the south ofChilegiven that large-scale cultivation within the natural distribution zone islimited. Nevertheless, the lower water temperatures that exist in the southcould restrict the growth rate. No difference in the scope for growth wasdetected in specimens kept at 10 ± 1 °C and 18 ±1 °C and fed with a mixed diet 1:1 of Chaetocerosgracilis and Tetraselmis suecica. Temperaturedid not affect the quantity of energy derived from food ingested, but energyloss through respiration, faeces and excretion was greater at 10 ± 1°C than at 18 ± 1 °C. The resultsindicate that the growth rates of A. purpuratus aremaintained between 10 ± 1 °C and 18 ± 1°C, which would favour large-scale cultivation over a widelatitudinal range.  相似文献

An alkaline deoxyribonuclease (DNase) from cod pancreatic tissue has been characterized. The enzyme is a DNase I type endonuclease and hydrolyzes effectively both native and denatured DNA. Monomeric actin inhibits the enzyme reaction. The enzyme obeys Michaelis-Menten kinetics and the apparent Km value for native linear duplex DNA is 33 µg/ml. The cod DNase opens supercoiled plasmid DNA, by introducing adjacent nicks in both strands, possibly separated by 5–10 nucleotides. DNA hydrolyzed by cod DNase functions as substrates both for DNA polymerase and ligase, and the nicks therefore contain 5-phosphoryl and 3-hydroxyl groups. Optimum concentrations of divalent cations are 5 mM Mg2+, 0.63 mM Mn2+ and 0.075 mM Ca2+. However, Ca2+ is apparently not essential for the enzymatic functions. The enzyme has a narrow temperature optimum at 42°C and is thermolabile above 50°C; however, Mn2+ shifts the optimum slightly to 45°C by causing increased temperature stability. The cod DNase reaction is inhibited by the DNA intercalating compounds actinomycin D and ethidium bromide. Histidine-modifying reagents such as tosyl phenylalanyl chloromethylketone and diethyl pyrocarbonate inhibit the enzyme activity, but the cod DNase is insensitive to disulfide-reducing agents.  相似文献

The present paper describes the growth properties of juvenile Atlantic cod (Gadus morhua) reared at 7, 10, 13 and 16 °C, and a group reared under “temperature steps” i.e. with temperature reduced successively from 16 to 13 and 10 °C. Growth rate and feed conversion efficiency of juvenile Atlantic cod were significantly influenced by the interaction of temperature and fish size. Overall growth was highest in the 13 °C and the T-step groups but for different reasons, as the fish at 13 °C had 10% higher overall feeding intake compared to the T-step group, whereas the T-step had 8% higher feeding efficiency. After termination of the laboratory study the fish were reared in sea pens at ambient conditions for 17 months. The groups performed differently when reared at ambient conditions in the sea as the T-step group was 11.6, 11.5, 5.3 and 7.5% larger than 7, 10, 13 and 16 °C, respectively in June 2005. Optimal temperature for growth and feed conversion efficiency decreased with size, indicating an ontogenetic reduction in optimum temperature for growth with increasing size. The results suggest an optimum temperature for growth of juvenile Atlantic cod in the size range 5–50 g dropping from 14.7 °C for 5–10 g juvenile to 12.4 °C for 40–50 g juvenile. Moreover, a broader parabolic regression curve between growth, feed conversion efficiency and temperature as size increases, indicate increased temperature tolerance with size. The study confirms that juvenile cod exhibits ontogenetic variation in temperature optimum, which might partly explain different spatial distribution of juvenile and adult cod in ocean waters. Our study also indicates a physiological mechanism that might be linked to cod migrations as cod may maximize their feeding efficiency by active thermoregulation.  相似文献

The changes in proximate composition, amino acid (total and free) and fatty acid content of artificially propagated trout cod, Maccullochella macquariensis larvae from five mothers hatched, weaned and reared separately, each in two groups, one fed with Artemia naupli and the other starved, for 15 days (after yolk resorption), are presented. There was no significant change in the proximate composition of fed larvae with devlopment, but in starved larvae the protein (linearly) and lipid (curvi-linearly) content decreased significantly as starvation progressed. The essential amino acids (EAA) and non- essential amino acids (NEAA) found in highest amounts in trout cod larvae were lysine, leucine, threonine and arginine, and alanine, serine and glutamic acid, respectively. In fed larvae the total amino acid (TAA), TEAA and TNEAA content did not vary significantly as development progressed. In starved larvae the TAA, EAA and NEAA content, as well as all the individual amino acids decreased significantly (P<0.05) from the levels in day of hatch and/or yolk-sac resorbed larvae. The greatest decrease occurred in the TEAA content (7.38±0.76 at day of hatch to 1.96±0.09 15 day starved in moles larva–1; approximately a 74% decrease), whereas the decrease in TNEAA was about 38%. Unlike in the case of TAA distinct changes in the free amino acid (FAA) pool were discernible, from day of hatch and onwards, in both fed and starved trout cod larvae. In both groups of larvae the most noticeable being the decrease of % FEAA in TFAA, but not the % FAA in TAA. Four fatty acids together, accounted for more than 50% of the total in each of the major fatty acid categories in all larvae sampled; 16: 0, 18:1n-9, 22: 6n-3 and 20: 4n-6, amongst saturates, monoenes, n-3 PUFA and n-6 PUFA, respectively. Twelve fatty acids either decreased (14: 0, 16: 1n-7, 20: 1n-9, 20: 4n-6, 20: 5n-3, 22: 5n-3 and 22: 6n-3) or increased (18: 2n-6, 18: 3n-3, 18: 3n-6, 18: 4n-3 and 20: 3n-3) in quantity, after 15 days of feeding, from the base level in day of hatch and/ or yolk- sac resorbed larvae. The greatest increase occurred in 18: 3n-3 from 6.4±0.1 to 106.2±13.1 g mg lipid–1 larva–1, and the greatest decrease occurred in 22: 6n-3 (181.2±12.4 to 81.4±6.2 g mg lipid–1 larva–1). In starved larvae, at the end of 15 days, all the fatty acids, except 18: 0, 20: 3n-3 and 20: 4n-6, decreased significantly (P<0.05) from the levels in day of hatch and/or yolk- sac resorbed larvae.  相似文献

Hypothalamic dopamine (DA), noradrenaline (NA) and 5-hydroxytryptamine (5-HT) levels exhibited marked day-night variations under ambient photoperiod and temperature (12L12D; 17±1°C) with peak values at mid-light phase. The 16L8D; 22±1°C treatment reversed the diurnal rhythm of 5-HT, but not that of DA and NA. However, there was an overall increase in the levels of the catecholamines on exposure to the long photoperiod and high temperature. The administration of melatonin in the fish held on 16L8D; 22±1°C regime restored the 5-HT rhythm to that of the 12L12D; 17±1°C control group, but with elevated mid-photophase value. However, there was no effect of the indole treatment on the 5-HT rhythm under the 12L12D; 17±1°C regime. Melatonin causes a significant reduction of NA level in both the groups, while DA level did not change in either group.  相似文献

Variations in growth and survival of hatchery-reared post-metamorphicjuveniles of great scallop Pecten maximus prompted anexamination of settlement and postlarval development. The effects ofseawater flow and temperature on great scallop metamorphosis andpostlarvae were studied over a 4–5 week period. In allexperiments, and regardless of environmental conditions, great scallopmetamorphosed after a 2–3 week period with values of 35 to70%. Subsequently, spat numbers increased slightly. Spatmortality generally occurred from the third week onward and reachedlevels as high as 30% by the fifth week under standardconditions. At 20 °C, however, 60% mortality levels wererecorded. Differences in spat growth rate, ranging from 37 to 45 mday–1, were noticed at different seawater flow ratesbut no clear tendency could be discerned. Temperature affected spatgrowth with an increase in size from 24 m day–1 at15 °C to 35 m day–1 at 18 °C. Conversely,growth was suppressed at 20 °C (14 m day–1).For optimal metamorphosis and postlarval development in great scallop, aseawater flow of 4.3 L h–1 per sieve and a temperatureof 15 °C are recommended.  相似文献

Four size groups of juvenile farmed Celtic Sea cod (2.7–41.8 g) were reared at a range of constant temperatures (8–19°C). The optimum temperature for growth (Topt.G) decreased from 15.1°C for ~3 g fish to 12.5°C for ~42 g fish. A comparison of these results with those published for a more northerly population (Icelandic) suggests that there is no significant difference in the optimal temperature for growth of cod stocks within the size range studied. In contrast, the growth rates of Celtic Sea cod were lower than those derived from these established models (for a northerly stock) for small juveniles (<5 g), but similar for larger fish (>40 g). Thus, while Topt.G appears fixed across the range, there may be high plasticity in local growth performance throughout the Holarctic distribution. Some possible explanations for these differences are considered.  相似文献

The endemic, anadromous cyprinidChalcalburnus tarichi is the only fish species known to occur in alkaline Lake Van (Eastern Anatolia, Turkey). EightC. tarichi were maintained individually in Lake Van water (17 – 19°C; pH 9.8; 153 mEq·I–1 total alkalinity; 22 total salinity) and tank water samples analyzed for 24 h in 2 to 4 h intervals. At zero time, < 1µM ammonia was present and urea was undetectable in the tank water; at 24 h, total ammonia and urea made up 114±32 and 35±25µM, respectively. Over the experimental period, ammonia-N and urea-N excretion averaged 1041±494 and 607±169moles·kg–1 fish·h–1, respectively. The extent of urea excretion was highly variable between specimens. Uric acid excretion was not detectable.Urea was present at high concentrations in all tissues and plasma (25 – 35moles·g–1·ml–1) of freshly caughtC. tarichi; total ammonia content of the tissues was by a factor of 1.9 (liver) to 3.0 (brain) lower. High arginase activity (2.4±0.2 U·min–1·g–1) was detected in the liver ofC. tarichi but ornithine carbamoylphosphate transferase, a key enzyme of the ornithine-urea-cycle, was absent. Ureagenesis is likely through degradation of arginine and/or uricolysis. High glutamine synthetase activity (11±0.6 U·min–1·g–1) and low ammonia content in brain suggest that, like other teleosts,C. tarichi has an efficient ammonia detoxification in the brain, but in no other tissue.Nitrogenous waste excretion at alkaline pH is discussed. The ability ofC. tarichi to excrete high levels of ammonia at extremely alkaline pH is unique among teleosts studied so far. The mechanism of ammonia excretion under Lake Van conditions remains to be elucidated.  相似文献

To further examine the concept of egg quality and the physiology of stored salmonid eggs, we investigated the effects of different oxygen tensions on the adenosine triphosphate (ATP) levels of unfertilized, activated, and fertilized chinook salmon (Oncorhynchus tshawytscha) eggs. The ATP levels of unfertilized chinook salmon eggs were 2.61±0.14 nmol ATP per egg (17.6±0.9 mol l–1 relative to cell water) and ranged from 1.98 to 3.63 nmol ATP per egg. The ATP content of unfertilized eggs maintained at 10 °C under 100% O2, 21% O2, and 100% N2 remained unaltered throughout a 120 h storage period. Storing eggs under identical conditions at 20 °C (in an effort to speed egg metabolism and ATP turnover) resulted in significant O2-independent decreases in ATP levels. However, ATP levels of unfertilized eggs exposed to 1 mmol l–1 potassium cyanide (a potent inhibitor of oxidative phosphorylation) at 10 °C were significantly decreased after 24 h and continued to decline throughout the 120 h maintenance period to about 30% of time=0 values. Maintenance with exogenous nutrients (5 mmol l–1 acetate plus 5 mmol l–1 pyruvate) over 120 h at 10 °C did not alter the ATP content of unfertilized eggs. Eggs activated by exposing them to 10 °C water for a few minutes showed a rapid decrease in ATP values, regardless of whether the eggs were fertilized or not. Following an initial 25% drop after fertilization, the ATP levels remained stable for the remainder (5 d) of the incubation period in eggs maintained in 10 °C water. Therefore, unfertilized chinook salmon egg ATP levels appear to be relatively stable and maintained by a low, cyanide-inhibitable metabolism. The stability of egg ATP levels may be one reason that salmonid eggs can be stored for several days while eggs from other fishes cannot.  相似文献

Newly hatched juvenile Buccinum undatum can be reared under laboratory conditions. Good was growth is achieved when juveniles were fed on combined diets (blue mussel, cod, and fish pellets). Juveniles reached shell heights of 33.0 ± 4.2 mm, 26.9 ± 3.8 ± mm, 23.2 ± 2.2 mm, and 20.1 ± 1.6 mm, after 14 months of fedding on a combined diet, blue mussel, cod, and fish pellets, respectively under ambient sea temperature and salinity. After 14 months juveniles fed blue mussel had the highest survival rates (67%) followed by those fed a combination of all other experimental diets (61%), cod waste (53%) and fish-feed pellets (46%). High mortalities were recorded in most treatments during the summer months between June and September. This species appears to have an aquaculture potential, as juveniles readily feed on artificial diets at an early age, show high survival rates and could potentially reach market size in 2 years or less. The major constraint in realising this potential at present, is the relatively low value of the species; if market values increased as a result of serious depletion of natural populations, hatchery production of juveniles for intensive aquaculture or restocking could become economically viable.  相似文献

Fast and slow muscle fibers were isolated from the myotomes of atlantic cod (Gadus morhua L.) and sculpin (Myoxocephalus scorpius L.). Epinephrine was found to have no effect on twitch or sub-tetanic contractions in fast muscle fibres. Isoprenaline (10–6M) had no effect on the contractility of slow muscle fibres. In contrast, epinephrine elicited a dose-dependent decrease in the half-time for twitch relaxation (t1/2r), and in most cases a decrease in twitch amplitude. The maximum decrease in t1/2r was around 5–20% of control values (at 10–6M epinephrine), with a half maximal response at about 30 nmol l–1. Responses to epinephrine were unaffected by propranolol and reversed by phentolamine, consistent with the stimulation of -adrenoreceptors. 10–6M epinephrine produced a rise in cAMP levels from 1.8 to 3.1 pmol mg dry wt–1 in cod slow fibres. However, the cellular mechanism underlying the action of epinephrine is unclear since forskolin, a potent activator of adenylate cyclase activity, where it has been investigated, was found to increase not decrease twitch duration and amplitude. The responses of fast and slow fibres to epinephrine and its antagonists were similar in summer (13°C) and winter acclimatized (5–6°C) sculpin.It is suggested that epinephrine may act to modulate the active state of slow muscle fibres at high cruising speeds and thereby increase swimming performance.  相似文献

The influence of temperature on growth and biomass composition of two species of Spirulina, S. maxima and S. platensis used for food was studied. A 4L fermenter with temperature and agitation control was used to cultivate both species. Under continuous light, maximum cell production of 2.4 g l–1 was verified for both cultures studied at temperatures above 25 °C: S. maxima (30 °C and 35 °C) and S. platensis (25 °C and 30 °C). An accentuated lag phase was observed for all cultures at lower temperatures (15–20 °C), and a maximum biomass production of 1.5 g l–1 was achieved. It was also observed that an increase of temperature caused a marked decrease in protein content, while carbohydrate synthesis was stimulated. The concentration of -linolenic acid varied from 11–16% for S. maxima and from 12–14% for S. platensis, at the optimum growth temperatures. Greater culture volumes were also studied in order to compare the performance of glass and plastic containers. At optimum growth temperature, S. maxima produced the same cell growth and similar final biomass composition.  相似文献

Gastric acid secretion from isolated cod stomach mucosa was measured using a pH-static titration method. A basal acid secretion rate (BASR) of 6.0±0.6 nEqH+min–1cm–1 was measured when using 0.9% NaCl as luminal solution. There was a dose-dependent increase in response to histamine between 0.12 and 0.20 M (EC50=0.15 M), above which gastric acid secretion plateaued at 13.5±1.8 nEqH+min–1cm–1. Ranitidine, a H2-receptor antagonist, completely blocked the stimulatory effect of histamine and reduced the BASR. The H1-receptor antagonist, clemastine, did not inhibit the response to histamine. Acid secretion rates decreased significantly when the pH of the luminal side of the mucosa was lowered from pH 5.75 to pH 4.50, indicating that a negative feedback mechanism was operating. Histological staining showed that oxynticopeptic cells were uniformly distributed throughout the cardiac stomach.It is concluded that the acid secretion in the isolated stomach mucosa of cod can be measuredin vitro with a pH-static titration method. The method was used to demonstrate that the BASR is downregulated by a decrease in pH. Furthermore, we conclude that the histamine receptor in the cod stomach mucosa resembles the mammalian H2-receptor and that histamine is secreted under basal conditions.  相似文献

Thisstudy investigated the effects of shelter surface area (SSA) on the feeding,growth and survival of the donkey-ear abalone, Haliotisasinina reared in mesh cages (0.38×0.38×0.28m) suspended in flow-through tanks (water volume = 6m3). Cages had sections of polyvinylchloride (PVC) thatprovided shelters with surface area of 0.22 m2, 0.44m2 and 0.66 m2.Hatchery-produced abalone with initial shell length of 32 ± 1mm and wet weight of 7.5 g were stocked at 50individuals cage–1 that corresponded to stocking densities ofca. 227, 113 and 75 abalone m–2 of SSA. The ratios of sheltersurface area to cage volume (SSA:CV) were 5.5, 11 and 16.5. Abalones wereprovided an excess red seaweed Gracilariopsis bailinae(= Gracilaria heteroclada) at weekly intervals overa 270-day culture period. Feeding rates (18–20% of wet weight), foodconversion ratio (26–27) and percent survival (88–92%) did notdiffer significantly among treatments (p > 0.05). Body size at harvest rangedfrom 56 to 59 mm SL and 52 to 57 g wet body weightwith significant differences between abalone reared at SSA 0.22m2 and 0.66 m2 (p < 0.05).Abalone reared in cages with 0.66 m2 SSA grewsignificantly faster at average daily growth rates of 132 m and188 mg day–1. Stocking densities of 75–113m–2 SSA in mesh cages suspended in flow-throughtanks resulted in better growth of abalone fed red seaweed.  相似文献

The objective of this study was to determine the potential sites of maximal growth limitation in Atlantic cod (Gadus morhua). Forty cod were reared in ten sea water tanks. Fish were randomly divided into 5 groups, a control group (injected with saline solution) and 4 experimental groups that received different levels of recombinant bovine somatotropin (rbST: 1.0, 2.0, 4.0 and 10.0 g g fish–1 2 weeks–1) by injection in the abdominal cavity. Fish were fed ad libitum 3 times a week during four weeks. We measured individual body mass, growth rate, food intake and food conversion efficiency. At the end of the experiment, we measured the activity of acid proteases in the stomach; trypsin and chymotrypsin in the pyloric caeca; alkaline phosphatase (ALP) and glutamyltransferase (GGT) in the intestine. No significant differences were observed in growth rate, food intake or food conversion efficiency among the five groups. Trypsin, chymotrypsin, ALP and GGT activities when expressed in U g fish–1 were correlated with growth rate. These enzymes were also correlated with food ingestion except for trypsin when expressed in U mg protein–1. Trypsin was the only enzyme that showed a significant correlation with food conversion efficiency. Our conclusion is that, at the level of digestion, trypsin is the only enzyme measured that could be suspected to potentially limit growth rate in cod.  相似文献

In order to examine the effects of temperature on benzo[a]pyrene (BaP) metabolism and adduct formation in the absence of the effects of temperature on uptake, gulf toadfish,Opsanus beta, were given a dose of 0.05 mg/kg3H-BaPvia caudal vein cannulae at their acclimation temperatures. (18 or 28°C) or following an acute temperature change (18 to 28°C or 28 to 18°C). After 72h, BaP-derived radioactivity was detected in all tissues examined and, as in otherin vivo studies of fish, the highest levels were found in the bile, the liver and the kidney. Temperature did not affect the total amount of BaP metabolized and excreted to the bile, but there were significant quantitative differences between temperature treatments in the classes of Phase I metabolites accumulated. Fish acclimated to high temperature accumulated more BaP triols and tetrols (breakdown products of highly carcinogenic BaP diol epoxides) than fish acclimated at low temperature regardless of exposure temperature: the proportion of biliary metabolites as tetrols and triols in each of the four temperature treatments (acclimation: exposure temperature), 28:18, 28:28, 18:18 and 18:28°C were 21.3±3.6, 58.1±6.1, 14.2±1.8 and 20.9±3.2% (mean±SEM, n=4), respectively. Significant quantities of BaP-DNA and BaP-hemoglobin adducts were detected; however, only the amounts of BaP-DNA adducts showed sensitivity to temperature. As predicted from our metabolite data, high acclimation or exposure temperature led to a significant increase in the amount of BaP-DNA adducts formed: adduct formation in the temperature treatments, 28:18, 28:28, 18:18 and 18:28°C were 342±52, 526±51, 155±42 and 252±55 fg BaP/ µg DNA (mean±SEM, n=4), respectively. These results are discussed in the context of mechanisms of high temperature-enhancement of carcinogenesis in fish.  相似文献

Feed intake, specific growth rate and changes in body composition were studied in juvenile (140-170 g) Baltic salmon, Salmo salar, reared at three temperatures (2, 4 and 6 °C) under continuous light conditions. Feed intake increased from 20.4 kJ kg-1 day-1 at 2 °C to 63.8 kJ kg-1 day-1 at 6 °C, and growth rate increased from 0.10% day-1 to 0.37% day-1 over the same temperature interval. The estimated lower temperature limits for feeding and growth were approximately 0.35 °C and 0.6 °C, respectively. Amongst fish reared at 2 °C the majority (86%) of the weight gain consisted of water and protein, and these fish deposited very little lipid. Lipid deposition increased amongst the groups of fish held at the higher temperatures, and amongst the salmon reared at 6 °C lipid accounted for 43% of the body energy gain.  相似文献
