The decrease of genetic variation in crops like tomato (Solanum lycopersicum L.) due to domestication, breeding, and population bottle necks for fresh market consumptions has increased the need for this cash crop’s conservation and characterizations. To study the genetic variation of this valuable genetic resource in the National Plant Gene Bank of Iran, a collection of 589 tomato accessions from different regions was characterized using semi-automatic and high-throughput techniques. Based on the fruit shape, the studied accessions were classified into ten groups and significant differences for all the fruit characteristics studied indicated an abundant diversity within the germplasm collected from different regions. Studied tomato germplasm was grouped into nine main clusters based on hierarchical cluster analysis. More than 83% of data variation was explained by seven components in which the first two components explained for 50% of the variation where fruit shape index and proximal/distal fruit end shape showed a high contribution in the variation of the first component. This unique genetically divergent germplasm could be utilized to select tomato breeding lines of interest. Also, accessions belonged to the heterotic clusters could be selected for hybridization and conservation purpose as well as enrichment of pre-breeding programs of the tomato germplasm.

A newly developed nongenetically modified purple tomato V118 was investigated for its phytochemical compositions and antioxidant activities. A highly efficient and sensitive UPLC method was developed for both the phenolics and carotenoids, which showed that in addition to the phytochemicals commonly known for tomatoes, V118 had a unique composition of anthocyanins. The total carotenoid content of V118 was 234.78 μg/g dry weight (DW), and the total phenolic content was 659.11 mg GAE/100 g DW. The antioxidant activities of the lipophilic extract as measured by the PCL and ORAC-L assays were 30.11 μmol TE/g DW and 11.97 μmol TE/g DW, respectively, while the hydrophilic extracts as determined by the ORAC-H and FRAP assays were 323.23 μmol TE/g DW and 54.95 μmol AAE/g DW, respectively. The LC-MS study showed three major anthocyanins, which were mainly acylglycosides of petunidin and malvidin. This study showed that purple tomatoes such as V118 possess additional phytochemicals like anthocyanins, which can potentially have added health benefits.  相似文献

This study aims to evaluate the effects of nitrogen (N) and phosphorus (P) on tomato leaves at the microscale in order to propose new methods to detect N and P conditions of plants. N and P solutions composed of three N levels (N0.25, 1.75; N1, 7; and N1.5, 10.5 mmol L?1) and three P levels (P0.25, 0.17; P1, 0.67; and P1.5, 1.01 mmol L?1) with ten replications. Results showed a significant decrease in the leaf thickness among plants treated with N0.25, N1.5N and P0.25, whereas showed an increase among plants treated with P1.5 (p < 0.05). Stomatal density was reduced in P0.25- and N0.25-treated plants, whereas increased in N1.5- and P1.5-treated plants (p < 0.05). Moreover, N0.25-treated plants showed reduced trichome density, whereas N1.5-treated plants exhibited the highestamount of trichome density (p < 0.05). The densest veins occurred in the leaves of P1.5P-treated plants, then density reduced in plants with N1.5, N1P1, N0.25, P0.25 treatments, in a decreasing order (p < 0.05).  相似文献

Sink and source activity in two tomato (Solanum lycopersicum L.) genotypes that vary in fruit Brix were investigated to identify differences that potentially underscore this trait. Solara (Brix 9%) accumulated almost twice the glucose, fructose, and sucrose in ripe fruit and had a higher horticultural yield (25% greater) compared to Moneymaker (Brix 5%). 14C-glucose feeding suggested large disparities in sucrose metabolism in ripe fruit between genotypes. Biochemical pathways in the leaf adjacent to a fruiting truss at night were also analyzed since in many species, this is the period when leaf reserves are mobilized to feed the plant. Surprisingly, leaf metabolism, i.e., starch and sugar content, the levels of polar metabolites assayed by GC-TOF MS and 14CO2-pulse-chase fluxes in detached leaves, did not change between the day and night in either genotype. Solara has a higher morphological source-to-sink ratio, and this may contribute to higher Brix in that genotype.  相似文献

Induction of abiotic stress in tomato plants has been proposed as a mechanism for improving the nutritional quality of fruits. However, the occurrence of biotic stress can interfere with normal abiotic stress responses. In this study, the combined effect of water stress and infection with plant-parasitic nematodes on the nutritional quality of tomato was investigated. Plants were exposed to one or both stresses, and the levels of phenolic compounds, carotenoids, and sugars in fruits were analyzed as well as physiological responses. Levels of carotenoids lycopene and β-carotene were lower in water-stressed tomatoes but exhibited a different response pattern under combined stress. Nematode stress was associated with increased flavonoid levels, albeit with reduced yields, while chlorogenic acid was increased by nematodes, water stress, and the combined stress. Sugar levels were higher only in tomatoes exposed to both stresses. These results emphasize the importance of studying plant stress factors in combination.  相似文献

The aim of this study was to investigate the impact of application of calcium silicate and salinity singly, on plant growth and nutritional behavior and photosynthetic pigments of tomato. Application of sodium chloride (NaCl) induced significant reduction in plant development and growth parameters. Salt stress also led to an accumulation of sodium (Na+) and a decrease in potassium (K+) concentration. Reduction of chlorophyll and carotenoid in leaves were amongst other symptoms in salt-affected plants in 2 cultivars. Rio Grande was qualified as salt sensitive and Moneymaker as the salt tolerant genotype. Application of Silicon (Si) only improved plant behaviour as compared to control. Furthermore, Si induced ameliorative effects on the growth potential of NaCl stressed plants. This Si-ameliorative effect on plant varied depending on the considered cultivar and Si concentration. Based on these results, application of calcium silicate was suggested as an alternative way to ameliorate the harmful effects of salinity on tomato.  相似文献

Produced waters are considered a byproduct from the oil industry that may contain mineral elements essential for plant nutrition, which enables them for irrigation purposes. Due to the growing demand for water for agricultural use in the world, the aim of this study was to determine the feasibility of using dilutions of fresh water and produced water derived from the gas-producing zone of Sabinas-Piedras Negras, in northern México and their effect on mineral content and allocation, growth, and yield of tomato plants. The results showed that the dilutions containing produced water reduced fruit, stem, and leaf fresh weight, and that the uptake and accumulation of minerals in plant tissues, except for sodium, was also decreased. However, plants irrigated with dilutions of produced and fresh water showed increased translocation of essential minerals from the leaves to the fruit, which partially contributed to alleviate the negative effects on fruit yield.  相似文献

Abstract

Phosphorus (Pi) is one of the most limiting factors in plant nutrition as it is the least mobile and available nutrient to plants in most soil conditions. The management of Pi fertilization in agriculture raises ecological, economic, and social issues, since phosphate rock minerals are the only significant global resources of Pi and they will be rapidly depleted. Eggshell waste is a big problem for food companies producing different types of egg products, since the eggshell waste is very often simply discarded and disposed at landfills, with high costs related to their disposal. The aim of this work was the characterization of eggshells as a Pi source for plants, using tomato (Solanum lycopersicum L cv Marmande) as a model species. Plants were grown hydroponically being exposed to adequate and limited Pi availability, with or without eggshell powder. Plant growth performance was characterized by analyzing changes in fresh weight, protein, chlorophyll concentration, carotenoid content, and measuring the plant’s capability to accumulate phosphate. The addition of eggshell powder to the nutrient solution significantly improved plant growth and increased protein and chlorophyll concentration, not only with respect to P-deficient control, but also with P-sufficient ones. Furthermore, eggshell powder significantly increased Pi accumulation in P-deficient plants, suggesting that eggshell waste could be a suitable material as Pi source for tomato plants, thus contributing to the environmentally friendly disposal of this waste.  相似文献

The labeling of products containing genetically modified organisms (GMO) is linked to their quantification since a threshold for the presence of fortuitous GMOs in food has been established. This threshold is calculated from a combination of two absolute quantification values: one for the specific GMO target and the second for an endogenous reference gene specific to the taxon. Thus, the development of reliable methods to quantify GMOs using endogenous reference genes in complex matrixes such as food and feed is needed. Plant identification can be difficult in the case of closely related taxa, which moreover are subject to introgression events. Based on the homology of beta-fructosidase sequences obtained from public databases, two couples of consensus primers were designed for the detection, quantification, and differentiation of four Solanaceae: potato (Solanum tuberosum), tomato (Solanum lycopersicum), pepper (Capsicum annuum), and eggplant (Solanum melongena). Sequence variability was studied first using lines and cultivars (intraspecies sequence variability), then using taxa involved in gene introgressions, and finally, using taxonomically close taxa (interspecies sequence variability). This study allowed us to design four highly specific TaqMan-MGB probes. A duplex real time PCR assay was developed for simultaneous quantification of tomato and potato. For eggplant and pepper, only simplex real time PCR tests were developed. The results demonstrated the high specificity and sensitivity of the assays. We therefore conclude that beta-fructosidase can be used as an endogenous reference gene for GMO analysis.  相似文献

A new source to produce a significant quantity of a naturally occurring polyphenol, piceatannol, was investigated in this study. Both resveratrol and piceatannol are recognized as important ingredients in functional foods due to their beneficial health effects. However, unlike resveratrol, the piceatannol concentration in plants is very low. Thus, calluses of peanuts, an easily obtainable source, were chosen as the material to induce piceatannol production under controlled conditions. To induce resveratrol and piceatannol, calluses were exposed to the ultraviolet (UV) irradiation. Significant quantities of resveratrol and piceatannol were produced by calluses upon UV irradiation in both static and suspension culture conditions. The amounts of piceatannol and resveratrol produced in 1 g of calluses ranged from 2.17 to 5.31 microg and from 0.25 to 11.97 microg, respectively, in static culture. In suspension culture, the amounts of induced piceatannol and resveratrol were somewhat lower. The quantities of induced piceatannol and resveratrol reached a maximum at 18 h after UV irradiation treatment in static culture. In contrast, the levels of resveratrol and piceatannol remained almost constant throughout the experiments in suspension culture. The piceatannol produced by calluses in all studies was much higher than the values reported in the literature, whereas the resveratrol produced was comparable to reported values.  相似文献

Purpose: Due to environmental concerns, efforts are made to replace the use of peat in horticultural growth media by organic wastes. Four growth media were prepared with the purpose of achieving adequate physical and chemical properties for plant production. Materials and methods: Growth media prepared from mixtures of coir (C) and paper sludge (P), respectively, with two biogas digestates from food waste (D1 and D2), were tested. These mixtures, 20% D1 or D2?+?80% C or P (v/v), were evaluated as growth media for tomato (Solanum lycopersicum L.) and lettuce (Lactuca sativa L.). Results and conclusion: The growth media were all physically stable during the growing period, provided all the macronutrients and most of the micronutrients necessary for plant growth, adequate pH conditions, as well as an adequate electrical conductivity. The mixture of D2 and P produced the highest biomass compared to a mineral fertilised peat (control), with a biomass production of 76% of the control for lettuce and 54% for tomato. Causes for the biomass reduction relative to the control may be related to ammonium toxicity effects, and/or limited plant-available water. The digestates, particularly D1, seemed also to have a phytotoxic effect on the germination.  相似文献

The effect of a foliar spray of selenium on potatoes was investigated for 2 years. Amounts of 0, 50, and 150 g of Se ha(-)(1) were applied both as sodium selenate and as sodium selenite in water, either pure or with the addition of 0.15% of soluble leonardite as a source of humic acids (pH 7). Tuber selenium concentration increased with the application levels, both with sodium selenate and with sodium selenite, when only aqueous solutions were used. When humic acids were added, the tuber selenium level rose more markedly after the application of sodium selenate as compared to the case of the aqueous solutions; however, in the case of sodium selenite, the level showed a large increase only after the application of 50 g of Se ha(-)(1). Kinetics showed that humic acids raised the selenate availability, but no differences were found in the distribution of selenium in the tuber fractions. Foliar application of selenium with humic acids was proven to be a good way to increase the selenium content of potatoes, but the assimilation process of selenium was simpler with selenate than with selenite.  相似文献

A multicommutated flow system was developed for the determination of aluminum in crystallized fruit samples. Spectrophotometric determination is based on the reaction of aluminum with chrome azurol S. The binary sampling technique was implemented to improve mixing conditions and to minimize reagent consumption. Three different working zones were established (0.5-5.0, 5.0-25.0, and 10.0-100 ppm) using the zone sampling approach, allowing us to adapt the extent of the in-line dilution. The influence of the chemical and physical parameters on the performance of the system was studied. Detection limits of 0.1, 0.6, and 0.8 ppm were obtained for the lowest, the medium, and the highest dispersion system, respectively. The procedure was applied to the determination of aluminum in crystallized fruit extracts. The results were in agreement with those obtained by the reference flame atomic absorption procedure at a 95% confidence level. Repeatability (RSD) was better than 2.4% in all of the three application zones.  相似文献

A total of 106 samples taken from natural Argentinean populations of the weedy Solanum elaeagnifolium (subgenus Leptostemonum) were studied cytologically to understand the impact of the different ploidy levels in its distribution and origin. Classical Feulgen staining was employed to determine mitotic chromosome numbers in all samples. 2C nuclear DNA content was determined by means of PI flow cytometry in eight samples of different ploidy levels. Principal component analysis and GIS tools were employed to compare altitude, annual precipitation and annual mean temperature among accessions. Three cytotypes were found: diploid (2n?=?24) which is widespread, tetraploid (2n?=?48) centered in western and southern Argentina, and hexaploid (2n?=?72) which predominates in central Argentina extending as well to the east. The annual precipitation is significantly different between tetraploids and hexaploids. Cx-values ranged from 1.231 to 1.275?pg, with statistical differences (of about 24.5–50.9?Mbp, p?≤?0.05) among accessions. Diploids are the most widespread cytotype and have adapted to a number of very different habitats. Tetraploids live in arid or semi-arid regions with a mean annual rainfall less than 500?mm. Hexaploids are successful in colonizing wetter areas, where no tetraploids were found. Thus, the distribution of cytotypes may be associated with habitat differences, particularly soil moisture. The observed cytotype pattern and the differences in DNA content suggest multiple places of origin for the polyploidy of S. elaeagnifolium in Argentina.  相似文献

For the first time, a cytosolic carotenoid cleavage enzyme isolated from quince (Cydonia oblonga) fruit is described. The enzyme was partially purified by using centrifugation, acetone precipitation, ultrafiltration (300 kD, 50 kD), isoelectric focusing (pH 3-10), and sodium dodecyl sulfate polyacrylamide gel electrophoresis (7.5%). In this way, an enzymatically active protein fraction was obtained that contained three similar proteins, all exhibiting molecular weights in the range of 20 kD. Using beta-carotene as substrate, the enzyme activity was detected spectrophotometrically at a wavelength of 505 nm. The time constant of the reaction was 8.2 min, the Michaelis constant (K(m)) was 11.0 micromol x L(-1), and the maximum velocity (v(max)) was 0.083 micromol x L(-1) x min(-1) x mg(protein)(-1). The optimum temperature was above 50 degrees C.  相似文献

Tomato pulp waste, a byproduct obtained during the processing of tomato juice, has been shown to be a rich source of lycopene. The objectives of this study were to use gelatin and poly(gamma-glutamic acid) (gamma-PGA) as coating materials for the encapsulation of lycopene extract from tomato pulp waste. Initially, lycopene was extracted with supercritical carbon dioxide, followed by microencapsulation using an emulsion system consisting of 4.5% gelatin, 10% gamma-PGA, and 4.8% lycopene extract. Analysis of differential scanning calorimetry revealed that the thermal stability of the coating material could be up to 120 degrees C, with a mean particle size of 38.7 microm based on Coulter counter analysis. The total weight of microencapsulated powder was 617 microg with the yield of lycopene being 76.5%, indicating a 23.5% loss during freeze drying. During storage of microencapsulated powder, the concentrations of cis-, trans-, and total lycopene decreased along with increasing time and temperature. A fast release of lycopene in the powder occurred at pH 5.5 and 7.0, while no lycopene was released at pH 2.0 and 3.5.  相似文献

Bioactivity-guided fractionation of cranberry fruit was used to determine the identity of triterpenoid esters from Vaccinium macrocarpon, which inhibit tumor cell growth and may play a role in cancer prevention. In our previous study, a fraction from whole fruit exhibited tumor cell growth inhibition in vitro. The major components of this fraction were isolated by chromatographic separation of ethyl acetate extracts, purified by semipreparative HPLC, and identified by NMR as cis- (1) and trans- (2) isomers of 3-O-p-hydroxycinnamoyl ursolic acid. These triterpenoid esters have not been previously reported in Vaccinium fruit. Bioassay of the purified triterpene cinnamates in tumor cell lines in vitro showed slightly greater activity of compound 1 in most cell lines, with GI(50) values of approximately 20 microM in MCF-7 breast, ME180 cervical and PC3 prostate tumor cell lines. Quercetin was slightly less active than 1, while cyanidin-3-galactoside exhibited much lower cytotoxicity, with GI(50) greater than 250 microM in all cell lines. Phenylboronic acid (3) was also isolated from the fruit but showed insignificant antitumor activity.  相似文献
