根据已报道的14个cry1Ca基因,设计能够扩增cry1Ca全长基因的简并引物。利用该引物对本实验室分离的472株Bt菌株进行筛选。PCR扩增发现22株菌含有cry1Ca基因。对22株菌株进行cry基因多样性分析,结果获得5种类型酶切图谱,表明这些菌株分为5种类型。SDS-PAGE结果显示22株菌均表达约130ku的蛋白。Western Blotting结果证实这些株菌中cry1Ca基因均正常表达。提取菌株的晶体蛋白,经胰蛋白酶活化,对甜菜夜蛾[Spodoptera exigua(Hübner)]进行初步生测,结果表明22株菌中活化的Cry蛋白对甜菜夜蛾均表现出很强的毒杀作用。进一步从5个类型的菌中各挑选一株毒力较高的菌株进行LC_(50)的测定,结果证实它们均具有很高毒力,其中,T1-E12(0.087μg/g)、B16-C8(0.103μg/g)、T1-B8(0.202μg/g)的活性与阳性对照菌株G03(0.090μg/g)相当,具有生产应用潜力。本研究为新型高效Bt菌株的挖掘奠定了基础。  相似文献

利用生物活性测定方法从本实验室已分离保存的Bt菌株中筛选出了5株对甜菜夜蛾具有较高杀虫毒力的菌株。该5株Bt菌株对甜菜夜蛾初孵幼虫的LC50值为0.556~0.914mg/mL胞晶混合物,其中GS3菌株杀虫活性最高,LC50值为0.556mg/mL胞晶混合物;包含晶体形态主要是球形、大菱形、小菱形等;所含基因类型主要是cry1Aacry1Abcry1Lacry1Iacry1Ibcry2Abcry2Adcry2Ahcry7Abcry9Ba等;5株菌株均表达约60kD和130~150kD蛋白,其中2株还表达较弱的80kD蛋白。从GS3菌株中克隆得到一种新的cry2A基因,GenBank登录号为KT692984,由Bt基因国际命名委员会命名为cry2Ah5。该基因开放阅读框为1899bp,编码632个氨基酸,编码蛋白分子量为70.8kD,等电点pI为8.18,与其他Cry2Ah蛋白序列相似性为97%~99%。该基因在大肠杆菌BL21(DE3)中表达约70kD蛋白,与预测分子量相符。本研究筛选得到5株对甜菜夜蛾具有较高杀虫毒力的Bt菌株,克隆表达了一种新的cry2A基因cry2Ah5,为甜菜夜蛾的生物防治提供了新的菌株及基因资源。  相似文献

The noctuid moth Spodoptera littoralis (Boisduval) is an important pest of many cultivated plants worldwide and five different geographical Nucleopolyhedrovirus (NPV) strains of this pest have been isolated to date. Two of these, a plaque-purified variant of the S. littoralis NPV from Morocco (SpliNPV-M2) and a SpliNPV isolated from field-infected S. littoralis larvae found in Turkey (SpliNPV-TR1), were compared biologically in terms of infectiveness (median lethal dose, LD50) for third instars and in terms of virulence (median lethal time, LT50) for neonates and third-instar S. littoralis larvae. The LD50 values of SpliNPV-TR1 and SpliNPV-M2 were 20.73 and 185.21 occlusion bodies (OBs)/larva, respectively, with non-overlapping confidence limits indicating they were significantly different. Thus, SpliNPV-M2 was found to be significantly less infective (about nine times higher LD50) than SpliNPV-TR1. The LT50 values of neonates for SpliNPV-M2 and SpliNPV-TR1 were 37 and 43.9 h at a concentration of 10(6) OBs ml(-1), respectively. For these same isolates, the LT50 values at a concentration of 3 x 10(6) OBs ml(-1) were calculated as 35.6 and 41.7 h, respectively. The LT(50) values of third instars for SpliNPV-M2 and SpliNPV-TR1 were 147.4 and 160.5 h, respectively, at a dose of 3000 OBs/larva and 145.4 and 152.4 h, respectively, for the same isolates at a dose of 20,000 OBs/larva. On the other hand, Helicoverpa armigera (Hübner) and Agrotis ipsilon (Hufnagel) revealed a lack of lethality of the SpliNPV-TR1 isolate.  相似文献

Baculoviruses can alter the development and physiological status of their insect hosts. In the present study, two Spodoptera littoralis nucleopolyhedroviruses (SpliNPV-TR1 and SpliNPV-M2) were examined in terms of their effect on the stage development of S. littoralis at two doses/concentrations by inoculation of neonates or third instars. Both isolates had an acute pathological effect on neonates. However, larval development was prolonged (6-36 h) by either infection when neonates were infected. When third-instar larvae were inoculated, no mortality occurred until insects moulted to later stages. When the isolates were compared, more mortality occurred in the fifth instar with SpliNPV-M2 than with SpliNPV-TR1, whereas less mortality occurred in the sixth instar with SpliNPV-M2 than with SpliNPV-TR1. However, none of the isolates demonstrated a dose effect. Larvae died at 108-198 h post-inoculation at all infections, and none of them pupated. Larval development was retarded (0-126 h) for each stage when third-instar larvae were inoculated. Consequently, SpliNPV-M2 would be a more effective control agent than SpliNPV-TR1 for control of S. littoralis since it kills larvae at earlier developmental stages.  相似文献

Application of Bacillus thuringiensis tenebrionis (Bt) and expression of the Bt protein Cry3Aa in genetically modified crops are used for targeted control of the Colorado potato beetle Leptinotarsa decemlineata (Say). The Cry3A proteins are selectively toxic for the beetles but the present study describes effects of Cry3Aa on the Egyptian armyworm, Spodoptera littoralis (Boisduval). Cry3Aa expressed in potatoes or added to an agar-base wheat-germ diet reduced the growth of S. littoralis caterpillars and the fertility of adults. The effect of 1.4 mg kg(-1) Cry3Aa in potato leaves was comparable with that of 3.3 mg kg(-1) in the diet. This difference in activity was correlated with better digestibility and higher conversion efficiency of the diet that also supported higher reproduction rate: S. littoralis grown on the potatoes reached a similar size to those on the diet but laid only 702 instead of 1077 eggs per female. Cry3Aa consumption reduced body growth as a consequence of lower food intake without significantly affecting food digestibility and the conversion efficiency of nutrients. The 11% and 5% body weight reductions caused by 1.4 mg kg(-1) Cry3Aa in potatoes and 3.3 mg kg(-1) in the diet, respectively, were associated with 74% and 65% reduction in the number of progeny; S littoralis grown on a diet with 9.1 mg kg(-1) Cry3Aa were 10% smaller and produced no viable progeny. These data suggest that the curtailment of reproduction was not caused by a general shortage of nutrient reserves but by a more direct Cry3Aa effect on the reproduction process.  相似文献

The effects of DPX-MP062 [methyl 7-chloro-2,3,4a,5-tetrahydro-2-[methoxycarbonyl(4-trifluoromethoxyphenyl)carbamoyl] indeno[1,2-e][1,3,4] oxadiazine-4a-carboxylate] a broad-spectrum insecticide with a novel mode of action, on the Egyptian cotton leafworm, Spodoptera littoralis, were studied in laboratory experiments. Egg hatch was affected by high concentrations (125 mg AI litre-1) of DPX-MP062. Larvae that hatched from treated eggs were significantly affected at concentrations of 12·5 mg AI litre-1 and greater. Larvae were fed castor bean leaves treated with DPX-MP062; 1st-instar larvae were the most susceptible development stage. Pupation and adult formation were determined in assays with 5th-instar larvae. There was strong suppression of adult formation; 65 and 91% at 0·5 and 0·75 mg AI litre-1, respectively. Highly affected larvae died before pupation; slightly affected ones reached pupation 2–4 days later, were smaller than larvae in the untreated control, and were sometimes unable to develop into normal adults. Comparatively high concentrations (50 and 100 mg AI litre-1) of the test compound were necessary to affect adults by ingestion, but no effects from contact application could be determined at a concentration of 100 mg AI litre-1. © 1998 Society of Chemical Industry  相似文献

Compared with an unselected susceptible population, a cotton bollworm, Helicoverpa armigera (Hübner), population selected for 22 generations with transgenic cotton leaves (modified Cry1A) in the laboratory developed 11.0-fold resistance to Cry1Ac (one single-protein product MVPII). Resistance to Bacillus thuringiensis Berliner subsp kurstaki (Btk) was selected for 22 generations with a 5.2-fold increase in LC50. The estimated realized heritabilities (h2) of resistance for transgenic-cotton- and Btk-selected populations were 0.1008 and 0.2341, respectively. This reflects the higher phenotypic variation in response to Cry1Ac in the transgenic-cotton-selected population. This variation may have been caused by differences in protein toxin levels expressed in different growth stages of the transgenic cotton. Because of the different slopes of the probit regression lines between Cry1Ac and Btk, the estimated realized h2 cannot be used visually to compare resistance development to Cry1Ac and Btk in H armigera. Thus, the response quotient (Q) of resistance was also estimated. The Q values of resistance for transgenic-cotton- and Btk-selected populations were 0.0763 and 0.0836, respectively. This showed that the rate of resistance development would be similar in both selection populations. This result indicates that the selection of resistance using transgenic cotton is different from that selected using the single toxin. Resistance risk to transgenic cotton and Btk in field populations was assessed assuming different pressures of selection by using the estimated h2. Assuming the h2 of resistance in a field population was half of the estimated h2, and the population received prolonged and uniform exposure to transgenic cotton or Btk causing >70% mortality in each generation, we predicted that resistance would increase 10-fold after <23 generations for Cry1Ac in transgenic cotton-selected-populations and after <21 generations for Btk in Btk-selected populations. Cross-resistance would be expected after <48 generations for Btk in transgenic-cotton-selected populations and after <21 generations for Cry1Ac in Btk-selected population. The results show that the potential to evolve resistance is similar in both transgenic-cotton- and Btk-selected populations, but that cross-resistance development to Btk is slower in transgenic-cotton-selected populations than cross-resistance development to Cry1Ac in Btk-selected populations.  相似文献

BACKGROUND: Spodoptera litura (F.) is an important pest worldwide, with over 112 host plants, and is exposed to insecticides throughout the year, resulting in the rapid development of resistance. Insecticide mixtures can delay the development of resistance more effectively than sequences or rotations. Cypermethrin, deltamethrin, profenofos, chlorpyrifos and fipronil were assessed separately and in mixtures against laboratory susceptible S. litura and two field‐collected populations. RESULTS: The field‐collected population from Khanewal (KWL) was significantly more resistant to cypermethrin, deltamethrin, chlorpyrifos and profenofos than one collected from Muzaffar Garh (MGH). Mixtures of cypermethrin + chlorpyrifos or profenofos and of deltamethrin + chlorpyrifos or profenofos at 1:1, 1:10 and 1:20 ratios significantly increased (P < 0.01) toxicity to cypermethrin and deltamethrin in field populations. The combination indices of cypermethrin + chlorpyrifos at 1:1 and 1:10 ratios and cypermethrin + fipronil at 1:1, 1:10 and 1:20 ratios for the KWL strain and of cypermethrin + profenofos or fipronil at 1:1, 1:10 and 1:20 ratios for MGH were significantly below 1, suggesting synergistic interactions. The inhibitors DEF and PBO largely overcame resistance to deltamethrin, cypermethrin and profenofos, suggesting that resistance to the insecticides was associated with esterase and monooxygenase detoxification respectively. CONCLUSION: Chlorpyrifos, profenofos and fipronil could be used in mixtures to restore cypermethrin and deltamethrin susceptibility. 球孢白僵菌对斜纹夜蛾高毒菌株筛选与制剂的研发   总被引:1,自引:0,他引:1  
斜纹夜蛾Spodoptera litura是一种危害极大的农业害虫,为了探索1条可持续、无污染的新型防治方法,我们通过标准喷塔法从12株球孢白僵菌Beauveria bassiana中筛选出1株对斜纹夜蛾2龄幼虫高毒力的菌株--球孢白僵菌Bb2860菌株.在喷雾浓度为(2592±275)孢子·mm-2的高浓度下,菌株Bb2860对斜纹夜蛾2龄幼虫在第7d的校正死亡率达82.43%±6.03%.不同制剂的室内生测试验表明,孢子水悬液和2种油剂(分别为不含稳定剂和含有稳定剂)在中浓度319~529孢子·mm-2的喷雾浓度下,7d后对斜纹夜蛾2龄幼虫的累计死亡率分别为54.8%、70.0%和76.7%;菌药协同作用为86.8%.另外,于2009年9月在安徽马鞍山进行自然条件下对花椰菜叶上斜纹夜蛾幼虫的田间小区药效试验.处理9d后,纯菌剂的相对防效为77.64%;低剂量和常规用量5%氟啶脲(抑太保)的相对防效分别为49.02%和89.70%;菌药混用的相对防效为91.13%.本研究显示出利用球孢白僵菌制剂及其与低剂量抑太保混用防治斜纹夜蛾的良好应用前景.  相似文献

BACKGROUND: The cyclodepsipeptide destruxin produced by the entomopathogen Metarhizium anisopliae (Metch.) was administered by different methods, topical application, ingestion and a combination of the two, in an attempt to minimize the mycotoxin dose for efficient management of the insect pest Spodoptera litura (Fab.).RESULTS: The insecticidal activity of destruxin on the larval stages of S. litura showed an ascending trend in LD(50) values with increasing age. The value for 12-day-old larvae in the combined application assay was as low as 0.045 microg g(-1) body weight of crude destruxin from M-19 strain when compared with the corresponding values of 0.17 microg g(-1) body weight in the ingestion assay and 0.237 microg g(-1) body weight in the topical application assay. On the other hand, values were higher in the treatments with crude destruxin from the low-virulence M-10 strain of M. anisopliae showing the least quantities of A and E components of destruxin.CONCLUSION: Laboratory bioevaluation showed the combination assay of ingestion and topical application of crude destruxin to be efficient in enhancing its insecticidal properties. The adopted combination assay apparently simulates application of the insecticide at field level. Quantitative differences between destruxins from low- and high-virulence strains of M. anisopliae are in accordance with its presumed role in virulence.  相似文献
