AIM:To explore the effect of traditional Chinese medicine Shenshuguanxin granula on coronary circulation in a rat model of myocardial infarction (MI). METHODS:SD rats (n=50, SPF grade) were randomly divided into 5 groups (n=10):sham group, MI group, and high-dose, middle-dose and low-dose Shenshuguanxin granula treatment groups. The rat MI model was established by ligation of the coronary artery. The cardiac markers, small and medium-sized blood vessels [microvessel count (MVC) value] in the infarct zone, and platelet endothelial cell adhesion mo-lecule 1 (PECAM-1) and vascular endothelial growth factor (VEGF) expression in the infarct border zone were measured. RESULTS:After 4 weeks of coronary artery ligation, the significant increases in MVC in the infarct zone, and the expression of PECAM-1 and VEGF in the infarct border zone were detected compared with sham group (P<0.05). The differences of cardiac markers between MI group and other groups were insignificant (P>0.05). CONCLUSION:Shenshuguanxin granula improves coronary circulation in the rats with myocardial infarction by increasing the expression of PECAM-1 and VEGF, and promoting small and medium-sized angiogenesis.  相似文献

AIM: To investigate the effect of taurine on the expression of glucose transporter 1 (GLUT1) and transporter 3 (GLUT3) in rat brain with diffused brain injury (DBI).METHODS: Sixty-four male Sprague-Dawley rats were randomly divided into 4 groups: sham-operated group, DBI group, low-dose taurine group (200 mg/kg, ig) and high-dose taurine group (300 mg/kg, ig).After fed with the corresponding drugs for 7 days, the animal model of DBI was made, and the rats were executed 24 h after DBI.The expression of GLUT1 and GLUT3 in the brain was detected by the methods of immunohistochemistry and Western blotting.The pathomorphological changes of the cerebral cortex were observed under electron microscope.RESULTS: The expression of GLUT1 was detected in capillary vascular endothelial cells in each group, and cytoplasm-positive cells or the cells with buffy membrane were observed.No significant difference of the GLUT1 expression in brain tissues between DBI group and sham-operated group was detected.Compared with DBI group, the expression of GLUT1 in the brain tissues were significantly increased in low-and high-dose taurine groups (P<0.01).The expression of GLUT1 in the brain tissues in low-dose taurine group were significantly higher than that in high-dose taurine group (P<0.05).The positive staining of GLUT3 only appeared in the periphery of the third ventricle in each group in the cells with buffy membrane or positive cytoplasm.The expression of GLUT3 in the brain tissues in DBI group was significantly higher than that in sham-operated group (P<0.01).The expression of GLUT3 in the brain tissues in low-and high-dose taurine groups was significantly higher than that in DBI group (P<0.01).Compared with low dose taurine group, the expression of GLUT3 in the brain tissues were significantly increased in high-dose taurine group (P<0.01).The pathological damage of cerebral cortex in low-dose taurine group was obviously alleviated.CONCLUSION: Taurine may take part in the neuroprotective mechanisms in DBI by increasing the expression of GLUT1 and GLUT3 at protein level to maintain the energy supply in brain tissues.  相似文献

ATM: To explore the influence of Tangshen formula (TS) on endothelial function and blood rheology in diabetic nephropathy (DN) rats. METHODS: The DN rat model was established by intravenous injection of low-dose (30 mg/kg) streptozotocin (STZ) after having the high-fat/high-glucose diets for one month. The animals were divi-ded into DN model group, TS group and valsartan group. Fasting blood glucose (FBG), serum total cholesterol (TC), serum triglyceride (TG), renal cortex blood flow and hemorheology were monitored. The content of von Willebrand's factor (vWF) and plasminogen activator inhibitor-1 (PAI-1) in the serum was determined by ELISA. RESULTS: Compared with normal group, FBG,TC,TG, vWF and PAI-1 were increased in DN model group (P<0.05), and no significant difference of FBG was observed. Compared with normal group, plasma viscosity, Casson viscosity, whole blood high/medium/low-shear viscosity, erythrocyte aggregation index, erythrocyte rigidity index and erythrocyte electrophoresis time were increased, and erythrocyte deformation index was decreased in DN model group (P<0.01). Compared with DN model group, plasma viscosity, Casson viscosity, whole blood high/medium/low-shear viscosity, erythrocyte aggregation index, erythrocyte rigidity index and erythrocyte electrophoresis time were decreased (P<0.05), but there was no significant difference for erythrocyte deformation index in TS group. Compared with normal group, the renal cortex microcirculation blood flow in DN model group was significantly decreased. Compared with DN model group, the renal cortex microcirculation blood flow was significantly increased in TS group (P<0.05), and no significant change in valsartan group was found.CONCLUSION: Tangshen formula plays a protective role in the kidney of diabetic rats by improving the blood rheology and endothelial function, thus ameliorating the renal cortex microcirculation blood flow in experimental diabetic rats.  相似文献

AIM: To explore the expression and significance of fibrinogen-like protein 2 prothrombinase/fibroleukin (fgl2) in a model of rat coronary microthrombosis. METHODS: Seventy-eight Sprague-Dawley male rats were randomly assigned to model group (n=36), sham operation group (n=36) for different time (1 h, 1 d, 1week, 2 weeks, 3 weeks, 4 weeks)(n=6, respectively), and normal control group (n=6). After closing aorta, sodium laurate was injected into aortic root of the animals in model group. The animals in sham operation group were injected with normal saline for control. Microthromb were detected with HE staining and modified martius-sarlet-blue stains (MMSB). von Willebrand factor (vWF) was measured with ELISA. fgl2 mRNA and protein expressions were detected by RT-PCR and immunohistochemistry staining, respectively. RESULTS: Microthromb were observed in rat coronary microvasculature. vWF was increased in plasma. fgl2 was induced in model group compared to sham operation group (P<0.05), no significant changes were observed in sham operation group and normal control group (P>0.05). CONCLUSION: The functional disorder of endothelial cells induces the expression of fgl2 in the coronary microvasculature. fgl2 prompts the thrombosis with platelet and fibrin.  相似文献

AIM: To establish the implanted hepatic cancer models in SD rats, and to observe the effects and mechanism of Fu Zheng Kang Ai Fang on implanted hepatoma models. METHODS: Walker256 carcinoma was implanted to the liver of SD rats. The rats were treated with low-dose, medium-dose and high-dose Fu Zheng Kang Ai Fang respectively. The vital signs and the survival time were recorded. Arterial blood was collected at time points of 1 week, 2 weeks and 3 weeks after transplantation to measure the levels of lanineamino-transferases (ALT), aspartateamino-transferases (AST), aspartateamino-transferases: alanineamino-transferases (S/L), albumin (ALB), albumin/globulin (A/G), alkaline phosphatase (ALP), γ-glutamyl transferase (GGT) and alpha-L-fucosidase (AFU). RESULTS: The values of the ALT, AST in high-dose and medium-dose Fu Zheng Kang Ai Fang groups were less than those in hepatoma model group (P<0.01), and the values of ALB and A/G were more than those in hepatoma model group (P<0.05) at 3 weeks. The values of ALP, GGT and AFU in high-dose Fu Zheng Kang Ai Fang group decreased (P<0.01). The median survival rate of low-dose, medium-dose and high-dose Fu Zheng Kang Ai Fang groups increased, which were longer than that in model group. CONCLUSION: Herbal Mixture Fu Zheng Kang Ai Fang improves the quality of life and prolongs the survival time of implanted hepatoma rats. Fu Zheng Kang Ai Fang protects the liver functions, including the decrement of the ALT and AST, maintenance of the ALB level, and reduction of the hepatoma indicating enzyme like the ALP, GGT and AFU. The results show that the high-dose Fu Zheng Kang Ai Fang is the best mixture.  相似文献

AIM: To evaluate the safety and efficacy of 3,3-diindolylmethane (DIM) on neointimal proliferation of rabbit artery after balloon angioplasty. METHODS: Restenosis models of carotid artery after balloon injury was established in rabbits. 30 rabbits were randomly divided into 4 groups: sham-operated group, model group, low-dose DIM group and high-dose DIM group. DIM was given to the rabbits in low-dose treatment group (2 mg·kg-1·d-1) and high-dose treatment group (8 mg·kg-1·d-1) once a day from 3 d before operation to 4 weeks after operation. The two treatment groups were administered with intraperitoneal injection of emulsified DIM, while the other two groups with saline at the same volume. All rabbits were killed after 28 d and carotid arteries were removed. With HE staining, automatic image analysis and immunohistochemistry, artery morphology was observed and the thickness of arterial intima and media was measured. Platelet derived growth factor (PDGF) and transfer growth factor β1 (TGF-β1) were examined. The expression of 〖STBX〗bcl-2〖STBZ〗 gene was detected by in situ hybridization (ISH) with computer-assisted picture analysis system. RESULTS: No significant difference was found between low-dose DIM group and model group in intimal thickness, media thickness, luminal area and the expression of PDGF and TGF-β1 in low-dose DIM group (P>0.05). The intimal thickness was decreased in high-dose DIM group compared with model group and low-dose DIM group (P<0.01). The luminal area was significantly larger in high-dose DIM group than that in model group and low-dose DIM group (P<0.01). The expression of PDGF and TGF-β1 in low-dose DIM group was significantly reduced compared with model group and low-dose DIM group (P<0.01). CONCLUSION: The inhibition and treatment of DIM on arterial restenosis are safe and effective. The effect might be achieved by preventing neointimal proliferation and the expression of PDGF and TGF, enhancing apoptosis in the vascular smooth muscle cells.  相似文献

AIM: To study the role of injury and phenotype shift of liver sinusoidal endothelial cells in the development of portal hypertension of liver cirrhosis in rats. METHODS: The rat liver cirrhosis model was established by peritoneal injection of dimethylnitrosamine (DMN) (at a dose of 10 mg·kg-1, 3 times a week, for 4 weeks). The dynamic changes of liver cirrhosis were observed at different time points (1 day, 2 days, 3 days, 1 week, 2 weeks, 4 weeks, 6 weeks and 8 weeks). The pressure of portal vein (Ppv), the expression of CD44, von Willebrand factor (vWF), endothelin-1 (ET-1) mRNA and endothelial nitric oxide synthase (eNOS) mRNA, the serum hyaluronic acid (HA) content and liver ET-1 content were measured. RESULTS: Compared with the normal control rats, CD44 positive staining was weak in the 1 day model rats, and the numbers of fenestrae of sinusoidal endothelial cells (SECs) rapidly decreased, but serum HA content rapidly increased (P<0.05). vWF positive staining in the 2-day model rats was stronger than that in normal control rats (P<0.05). There was a positive correlation between the Ppv and the vWF expression, serum HA content in the DMN-induced liver cirrhosis rats (P<0.05). Compared with the normal control rats, ET-1 mRNA expression increased in the 2-day and 3-day model rats, and ET-1 content lightly increased. eNOS mRNA expression was stronger in the 1-day, 2-day and 3-day model rats than that in normal control rats, meanwhile eNOS always expressed at a low level. CONCLUSION: The injury and phenotype shift of SECs is a pathological basis in the development of portal hypertension of DMN-induced liver cirrhosis in rats. Imbalance of ET-1 and NO production increases intrahepatic resistance, which plays an important role in the development of portal hypertension.  相似文献

AIM:To observe the effects of methionine-induced hyperhomocysteinemia on protein C(PC),ant ithrombin-(AT-)and von willebrand factor(vWF).METHODS:Eighteen New Zealand rabbits were randomized as methionine group(group M,n=9)and control(group C,n=9),which were fed with methionine-rich diet(600 mg/d)and regular diet respectively for sixteen weeks.By the end of sixteen weeks,the serum biochemistry and PC,AT-and vWF in plasma were determined and vWF expression of endothelial cel s of aorta were examined.RESULTS:In group M,the levels of methionine(29.97±5.34 mol/L)and homocysteine(13.30±2.19mol/L)in serum were signif icantly higher than those(14.48±1.97 mol/L and 5.36 1.19 mol/L,respectively,P<0.01)of group C.The levels of AT-and PC of group M were signif icantly lower than those of group C(P<0.01).The level of vWF in plasma of group M was higher than that of group C(P<0.01).Immunohisto chemistry showed that vWF expression in endothelial cells of aorta was decreased.CONCLUSION:Methionine-induced hyperhomocysteinemia had promot ive ef ects on coagulat ion and inhibiting effects on antioagulation.  相似文献

AIM: To investigate the anti-aging effect of the stromal vascular fractions (SVF) injection in photo-aging skin of nude rats. METHODS: Six-week-old nude rats were divided into control group and photoaging group. The nude rats in photoaging group were continuously irradiated with ultraviolet B (UVB) for 8 weeks to build the photoaging skin model, and were randomly divided into placebo group, and low-dose (LD), middle-dose (MD) and high-dose (HD) SVF treatment groups. The human SVF was obtained by trypsin digestion from the adipose tissue in healthy females, and was subcutaneously injected into the back skin of photoaging nude rats in LD, MD and HD SVF treatment groups (104,105 and 106 cells in 100 μL suspension, respectively). The animals in photoaging skin model group were treated with placebo. Control group did not have any intervention. The changes in the thickness of epidermis and histology after 7 d and the thickness of the dermis and histology after 28 d were examined. RESULTS: The thickness of the epidermis was thinner in MD and HD SVF treatment groups than that in photoaging skin model group after 7 d (P<0.05). The percentage of stratum corneum and the abnormal proliferation of the epidermal basal cell layer were reduced (P<0.05). After 28 d, MD and HD SVF treatment groups showed that the content of collagen I was higher than that in photoaging skin model group (P<0.05), and the contents of collagen III and MMP3 were decreased (P<0.05). The dermis in HD group was thicker than that in photoaging skin model group (P<0.05).CONCLUSION: SVF may have anti-aging potential in photoaging skin and also has clinical application value.  相似文献

AIM:To intuitionally observe the characteristics of blood rheology in the patients with acute coronary syndrome (ACS) after percutaneous coronary intervention (PCI) for 1 year to 3 years by micro-channel array flow analyzer (MC-FAN) combined with other platelet function indexes, and to explore the correlations between the test results of MC-FAN and platelet function. METHODS:This study brought 74 patients with ACS after PCI for 1 year to 3 years into test group, and 21 healthy subjects were enrolled as normal group. The levels of platelet aggregation test (PAgT), platelet adhesiveness test (PAdT), P-selectin, platelet-derived growth factor BB (PDGF-BB) and von Willebrand factor (vWF) were detected. MC-FAN HR300 was used to detect the transiting time (MC-FAN TT) of the blood passing through the model body capillaries. The differences of the test results between the 2 groups were compared, and the correlations between the results of MC-FAN and platelet function in the patients with ACS after PCI were also explored. RESULTS:Compared with normal group, the MC-FAN TT in test group was prolonged (P<0.01), the ability of erythrocyte deformation was weakened, and the leukocyte attaching the vascular wall and platelet adhesion and aggregation relatively increased. The levels of PAgT, PAdT, P-selectin and PDGF-BB in test group were all higher than those in normal group (P<0.01). No difference of vWF between the 2 groups was observed. The intergroup correlation analysis showed that there were correlations between MC-FAN TT and platelet function, in which 10 μL MC-FAN TT and 30 μL MC-FAN TT had the most significant correlation with P-selectin (r=0601, P<0.01; r=0334, P<0.01), 60 μL MC-FAN TT had the most significant correlation with PAgT (r=0527, P<0.01), and 100 μL MC-FAN TT had the most significant correlation with PAdT (r=0. 815, P<0.05). CONCLUSION:The visualization of hemorrheologic changes and platelet function in the patients with ACS after PCI are abnormal.There are correlations between MC-FAN TT and platelet function.The results of MC-FAN can objectively evaluate the blood rheology of the patients, and provide the reference for clinical treatment.  相似文献

AIM:To explore the therapeutic effect of Qishen-Yiqi dripping pills (QS) on atherosclerosis (AS) and the mechanism. METHODS:AS rat model was established by high-fat diet, and SD rats were randomly divided into normal control group, AS model group, low-dose, middle-dose and high-dose QS groups, and positive group (n=6 each). After administration for 12 weeks, serum samples were collected to detect the serum lipid and Ca2+ levels. HE staining was used evaluated the histopathological changes of arterial tissue. The serum levels of interleukin-1β (IL-1β), IL-6 and tumor necrosis factor-α (TNF-α) were measured by ELISA. The nitric oxide (NO) level was detected by nitrate reductase method. The protein levels of transient receptor potential channel protein 1 (TRPC1), stromal interaction molecule 1 (STIM1) and endothelial NO synthase (eNOS) were determined by Western blot. RESULTS:QS significantly reduced the arterial damage via inhibiting the formation of atherosclerotic plaque and attenuated intimal thickening and vascular stenosis. Compared with AS group, the serum levels of total cholesterol (TC), triglyceride (TG) and low-density lipoprotein cholesterol (LDL-C) were decreased significantly and the levels of high-density lipoprotein cholesterol (HDL-C) were increased significantly in high-dose QS group (P<0.05). The serum levels of IL-1β, IL-6 and TNF-α in high-dose QS group were lower than those in AS group (P<0.05). Compared with AS group, the serum Ca2+ level was lowered and the arterial tissue NO level was elevated in QS groups (P<0.05). Compared with AS rats, the protein levels of TRPC1 and STIM1 were decreased significantly and the protein level of eNOS was increased significantly in the rats treated with QS (P<0.05). CONCLUSION:QS regulate calcium homeostasis via TRPC1/STIM1 pathway, increase the production of NO and inhibit the inflammatory responses, thus exerting anti-AS effect.  相似文献

AIM:To study the effects of basic fibroblast growth factor (bFGF) on brain edema, nerve function damage and autophagy related proteins in rats with head injury. METHODS:The rat model of craniocerebral injury (CI) was constructed. The rats were divided into control group, CI group, and low-, middle-and high-dose bFGF groups (n=10). The CI model was established in CI group, while the rats in control group were not given epidural impact. The rats in low-dose, middle-dose and high-dose bFGF groups were given bFGF at 2, 4 and 6 μg, respectively, by intraperitoneal injection after 30 min. The neurological function in the rats was evaluated by improved neurological function scoring. The rat brain tissues were taken, and the water content was detected. The levels of tumor necrosis factor-α (TNF-α), interleukin-6 (IL-6) and IL-1β in the brain tissue were measured by ELISA. The malondialdehyde (MDA) content was analyzed by thiobarbituric acid method. The activity of superoxide dismutase (SOD) was examined by WST-8 assay. The glutathine peroxidase (GSH-Px) activity was detected by colorimetric method. The protein levels of autophagy related proteins LC3-Ⅱ and beclin-1 in the brain tissues were determined by Western blot. RESULTS:The neurological function score was increased significantly of the rats in CI group. The rat model of craniocerebral injury was successfully constructed. Neurological function scores in the rats in low-dose, middle-dose and high-dose bFGF groups were reduced, the water content of the brain tissue was also reduced (P<0.05). The levels of TNF-α, IL-6 and IL-1 β were decreased in the brain tissues (P<0.05), the content of MDA was declined (P<0.05), the activities of SOD and GSH-Px were increased (P<0.05), the protein levels of LC3-Ⅱ and beclin-1 were decreased, compared with the untreated rats in CI group (P<0.05). CONCLUSION:bFGF improves the nerve function of the rats with craniocerebral injury, reduces the water content of the brain tissue, reduces the expression of autophagic protein LC3-Ⅱ and beclin-1.The mechanism is related to the inhibition of inflammatory reaction and oxidative damage.  相似文献

AIM:To investigate the effect of immunosuppressant FK506 on serum glucose in rats and to explore its mechanism. METHODS:Sprague-Dawley rats (n=12) were randomly divided into drug group and normal group. The rats in drug group were intraperitoneally injected with FK506 at dose of 1 mg·kg-1·d-1 and the rats in normal group received saline (1 mL·kg-1·d-1, ip) for 14 d. The fasting weight and fasting glucose were regularly measured every 2 d. Visceral fat was isolated from the rats at the end of experiment. The mRNA expression of adiponectin, leptin, visfatin, resistin, retinol-binding protein 4 (RBP4) and peroxisome proliferator-activated receptors γ (PPAR-γ) was determined by real-time fluorescence quantitative PCR. The protein expression of PPAR-γ and adiponectin was measured by Western blotting. RESULTS:Compared with normal group, the concentration of fasting blood glucose in model group was significantly increased from the 10th day (P<0.05). At day 14, the fasting blood glucose of the model group increased from (5.10±062) mmol/L to (7.73 ± 0.73) mmol/L. No significant change of blood glucose in normal group between the 10th day and the 14th day [from (4.66 ± 0.32) mmol/L to (5.80±0.10) mmol/L] was observed. Compared with normal group, the mRNA expression of PPAR-γ, adiponectin and leptin in the adipose tissue of model group was significantly decreased (P<001), whereas the expression of visfatin, resistin and RBP4 was significantly increased (P<005). Compared with normal group, the expression of PPAR-γ and adiponectin in model group was decreased (P<001). CONCLUSION:FK506 may decrease the expression of PPAR-γ to change the expression of adipocytokines and induce hyperglycemia in rats.  相似文献

AIM: To establish a microthrombus model by carrageenan (Ca)/ lipopolysaccharides (LPS) intraperitoneal injection in rats with hyperhomocysteinemia (HHcy) and endothelial dysfunction induced by L-methionine intake.METHODS: ① Male Sprague Dawley rats were randomly divided into 2 groups: control and endothelial dysfunction (HHcy) groups. L-methionine was administered by gavage in HHcy group for total 4 weeks.Purified water was administered by gavage in control rats.Plasma Hcy,NO and vWF were examined and the thoracic aorta were excised after 4 weeks of L-methionine treatment to evaluate endothelial function.② Male Sprague Dawley rats were randomly divided into 3 groups to establish a microthrombus formation model with Ca/ LPS: control,microthrombus formation (Ca/LPS) and endothelial dysfunction plus mitoarothrombus formation (HHcy+Ca/LPS) groups.Control rats were injected with normal saline (NS).Ca/LPS rats were intraperitoneally injected with carrageenan (Ca) and followed by lipopolysaccharides (LPS) 16 h later.HHcy+Ca/LPS rats were intragastric gavaged by L-methionine for total 4 weeks,and then were injected with Ca/LPS in the same way as Ca/LPS group.Cruor parameters and platelet count were detected at 20 h after LPS or NS injection and the mesentery microcirculation was monitored.Plasma NO and vWF were also detected at 24 h after LPS or NS injection.RESULTS: ① Plasma Hcy concentrations and vWF level were significantly increased in HHcy group,while plasma NO content was significantly decreased compared with that in control group.Endothelial dependent relaxation (EDR) of aortic rings was significantly decreased in HHcy group,suggesting endothelial damage/dysfunction was induced by HHcy.② Mesentery capillary was obviously blocked by microthrombus in Ca/LPS rats and was blocked more seriously in HHcy+Ca/LPS rats.Cruor parameter results suggested that Ca/LPS rats were in hypercoagulable phase and HHcy+Ca/LPS rats were in hypocoagulable phase at 20 h after LPS injection.Platelet count and plasma NO content in HHcy+Ca/LPS group were significantly decreased,while plasma vWF level was significantly increased compared with Ca/LPS group.CONCLUSION: L-methionine intake induces severe HHcy and causes endothelial dysfunction in rats.Microcirculation dysfunction and microthrombosis can be caused by Ca/LPS intraperitoneal injection and may be aggravated by endothelial dysfunction.  相似文献

AIM:To investigate effects of propolis-ethanol extract on platelet activity in flowing blood. METHODS:Intima-injury model was made by in vitro perfusion. Coverslips was coated by using fibrinogen and human Ⅲ type collagen to detect platelet adhesive rate when blood went through fibrinogen and collagen surface in 1 000/s shear rate. Three experimental groups were set up: negative control group (24% ethanol), experimental group (0.1 g/L, 24% propolis-ethanol extract) and positive control group (0.1 g/L, ferulic acid). RESULTS:Intima-injury model was made. On fibrinogen and collagen, platelet adhesion on area of experimental group was lower than that in negative control group (P<0.01). There was no significant difference between experimental group and positive control group (P>0.05). Propolis-ethanol extract decreased more obviously the platelet adhesion area on fibrinogen surface than that in collagen surface (P<0.01). CONCLUSION:Propolis-ethanol extract inhibits platelet activation and reduces its adhesion on fibrinogen and collagen surface, especially in fibrinogen surface.  相似文献

AIM:To evaluate the safety and feasibility of using vascular endothelial growth factor (VEGF) in the establishment of Walker-256 transplanted liver cancer model. METHODS:SD rats (n=45) were divided into 3 groups:via the caudal vein, the rats in normal saline (NS) group were injected with 0.9% sodium chloride (0.1 mL/d), the rats in 20 mg/L VEGF group were injected with 20 mg/L VEGF (0.1 mL/d), and the rats in 40 mg/L VEGF group were injected with 40 mg/L VEGF (0.1 mL/d). All the injection began 1 week before transplantation of liver cancer, and stopped on the day the cancer model was established. Prepared tumor tissue was transplanted into the subcapsular space of the liver. Three days, 1 week and 2 weeks after the transplantation, magnetic resonance imaging (MRI) was performed for analyzing the tumor growth and the characteristics. The overall survival of the rats was also recorded. RESULTS:Successful establishment of Walker-256 transplanted liver cancer model was achieved. Among 45 rats, 1 rat died 1 d after implanting the tumor both in NS group and 20 mg/L VEGF group, while 3 rats died in 40 mg/L VEGF group 1 week after building the model, mainly because of the progression of tumors. Three days after modeling,the numbers of the rats in which the tumor was positively detected by MRI in 3 groups were 0, 7 and 10, respectively; 1 week after modeling, those were 3, 13 and 13, respectively; 2 weeks after modeling,those were 12, 13 and 10, respectively. Between NS group and 20 mg/L VEGF group, the statistical significance existed in the number of the rats in which the tumor was positively detected by MRI after 3 d of implanting, so did the NS group and 40 mg/L VEGF group. No statistical significance in the overall survival time between NS group and 20 mg/L VEGF group (P>0.05) was observed, but the significance existed between 40 mg/L VEGF group and NS group (P<0.01). CfONCLUSION:The application of VEGF at dose of 20 mg/L and 0.1 mL/d shortens the time to establish the transplanted liver cancer model without influence on the overall survival, which is a safe, feasible and efficient way, and is more suitable for anti-VEGF drug investigation.  相似文献

AIM:To investigate the effects of Maxing-Shigan decoction on airway remodeling and expression of matrix metalloproteinase-9 (MMP-9) and tissue inhibitor of metalloproteinase-1 (TIMP-1) in the lung tissues of asthmatic mice, and to explore its possible mechanism in treatment of asthma. METHODS:The BALB/c mice were divided into blank control group, model group, low-dose Maxing-Shigan decoction group, middle-dose Maxing-Shigan decoction group, high-dose Maxing-Shigan decoction group and positive control group. The mice were sensitized and challenged with ovalbumin to establish asthma model. The mice in blank control group and model group were given saline by oral administration before 30 min of suscitation. The mice in low-dose, middle-dose and high-dose Maxing-Shigan decoction groups were given Maxing-Shigan decoction at 5.0 g/kg, 10.0 g/kg and 20.0 g/kg, respectively, by oral administration before 30 min of suscitation. The mice in positive control group was given dexamethasone at 0.005 g/kg by oral administration before 30 min of suscitation. After consecutive administration for 7 d, the variations of airway responsiveness, the percentage of the goblet cells, the collagen deposition, and the eosinophil (EOS) counts in bronchoalveolar lavage fluid (BALF) of each group were observed. The protein levels of MMP-9 and TIMP-1 in the lung tissues were determined by ELISA and Western blot. The mRNA expression of MMP-9 and TIMP-1 was detected by RT-qPCR. RESULTS:Compared with blank control group, the airway responsiveness, the goblet cell percentage, the collagen deposition, the EOS counts in BALF, the protein levels of MMP-9 and TIMP-1, and the mRNA expression of MMP-9 and TIMP-1 were significantly increased in model group (P<0.01). Compared with model group, all of the indexes were reversed in low-dose, middle-dose and high-dose Maxing-Shigan decoction groups and positive control group (P<0.05 or P<0.01). CONCLUSION:Maxing-Shigan decoction improves airway remodeling in asthma model mice by down-regulating the expression of MMP-9 and TIMP-1.  相似文献

AIM: To observe the effects of vascular endothelial growth factor (VEGF) on the proliferation and differentiation of neural stem cells (NSCs) of rats in vitro.METHODS: NSCs isolated from the hippocampal gyrus of SD rats were primary cultured and subcultured,and then divided into two groups: (1) the cells in VEGF group were treated with 150 μg/L VEGF in the culture system,and VEGF was removed at the 7 th day;(2) control group (without VEGF treatment).The cellular morphology of two groups was observed by contrast phase microscope.Nestin and NF-200 expressing cells were detected via immunofluorescence method.The percentages of the immunostaining positive cells in each group at the 7 th day and at the 11 th day were determined.RESULTS: At the 7 th day,the percentage of nestin positive cells in VEGF group was 52.19%±7.95%,vs 29.26%±4.12% in control group (P<0.01).The percentage of NF positive cells in VEGF group was 22.33%±4.13%,vs 38.62%±5.31% in control group (P<0.01).At the 3 th day after VEGF was removed,the percentage of NF positive cells in VEGF group was 43.10%±3.70%,vs 30.56%±4.16% in control group (P<0.01).CONCLUSION: VEGF stimulates the proliferation of neural stem cells and inhibits their differentiation.  相似文献

AIM: To study the effects of Yangyu Tuji (YYTJ) on delayed healing wound of diabetic rats caused by streptozotocin (STZ). METHODS: SD male rats were randomly divided into control group (control), model group (model); and 3 different dose groups of YYTJ. 55 mg/kg STZ were given by intraperitoneal injection except for control group. After 30 days, a round skin of 1.6 cm diametre was excised on all dorsal back of rats. The healing time and healing rate were observed according to re-epithelization. The content of collagenⅠ and Ⅲ was observed by Picric acid-Sirius red staining , Matrix metalloproteinase-1, 13 (MMP-1, -13), tissue inhibitor of metalloproteinases-1 (TIMP-1) by immuno-histochemistry assay. All data were analyzed by IPP software. RESULTS: The healing time in each group treated with YYTJ was shorter than that in model group (P<0.01), and the healing rate was increased (P<0.01, P<0.05). Content of type I collagen, ratio of type Ⅰ and Ⅲ collagen of high and mid dose group were significantly higher than that in model group (P<0.01) at 3rd, 7th, 11th day. The expression of MMP-1, -13 of each groups were higher than that in model group at 7th day (P<0.01, P<0.05), and MMP-1 trend to equal with model group at 11th day. MMP-13 was significantly lower than that in model group at 11th day (P<0.01, P<0.05). TIMP-1 of each group of wound was higher than that in model group at 3rd, 7th, 11th day (P<0.01, P<0.05). The ratio of type Ⅰ and Ⅲ collagens in each group was lower than that in model group at 11th day (P<0.01). Ratio of MMP-13 and TIMP-1 of high dose group and mid dose group were higher than that in model group at 3rd and 7th day (P<0.01). The ratio of each group was lower than that in model group at 11th day (P<0.01). Meanwhile, ratio of MMP-13 and TIMP-1 of high dose group and mid dose group were lower than that of lower dose group (P<0.05). CONCLUSION: It is possible that YYTJ accelerates wound healing by increasing collagen content of type Ⅰ and Ⅲ, especially type Ⅰ, as well as improves collagen deposition by regulating the balance of MMP and TIMP.  相似文献
