Phelipanche ramosa (Branched broomrape) is an obligate root parasitic plant that is a major pest of oilseed rape in France. Knowledge on seed viability and dormancy under field conditions is crucial to understand how to control P. ramosa, but is as yet unknown. Our study aimed to quantify these processes with a 2‐year seed burial experiment. Two genetically distinct populations of P. ramosa were studied, collected on winter oilseed rape (population O) and hemp (population H). Seed mortality was very low in both populations (4–7% per year). Although obligate parasitic seeds are assumed to germinate only after exposure to germination stimulants from host root exudates, a high proportion of population H seeds germinated spontaneously (up to 90%). Seeds of both populations displayed seasonal dormancy, with timing and magnitude depending on the population. Dormancy was low at the time each native host crop is usually sown. Populations differed in germination dynamics, with seeds of population H germinating faster. The difference in behaviour that we observed between populations is consistent with reported adaptations of pathovars to their preferred hosts. The results indicate that the parasitic plant management requires targeting at the populations concerned. For example, delayed sowing is more promising against population O than against population H.  相似文献

The effects of calcium hypochlorite [Ca(OCl)2] and gibberellic acid (GA), both in combination and separately, were studied on the germination of Orobanche crenata seeds. Hypochlorite was applied either before or after the conditioning phase for periods up to 150 min. The seeds were exposed to GA during the conditioning phase. After conditioning the seeds were exposed to either the synthetic germination stimulant GR 7, flax root exudate (FRE) or distilled water. GR 7 exploited the germination potential of O. crenata better than FRE and this in turn better than water. In the presence of GR 7 or FRE the number of germinating seeds generally increased when the seeds were briefly (5–20 min) exposed to hypochlorite. In the absence of GR 7 and FRE, hypochlorite only increased germination if the seeds were treated during medium-long periods (20–60 min) after conditioning. If GR 7 or FRE were not applied the germination percentage was usually significantly higher when conditioning was done in the presence of GA.  相似文献

BACKGROUND: Orobanche and Phelipanche species (the broomrapes) are root parasitic plants, some of which represent serious weed problems causing severe yield losses on important crops. Control strategies have largely focused on agronomic practices, resistant crop varieties and herbicides, albeit with marginal success. An alternative control method is the induction of suicidal seed germination with natural substances isolated from root exudates of host and non‐host plants. RESULTS: Soyasapogenol B [olean‐12‐ene‐3,22,24‐triol(3β,4β,22β)] and trans‐22‐dehydrocampesterol [(ergosta‐5,22‐dien‐3‐ol, (3β,22E,24S)] were isolated from Vicia sativa root exudates. They were identified by comparing their spectroscopic and optical properties with those reported in the literature. Soyasapogenol B was very specific, stimulating the germination of O. minor seeds only, whereas trans‐22‐dehydrocampesterol stimulated P. aegyptiaca, O. crenata, O. foetida and O. minor. CONCLUSION: Soyasapogenol B and trans‐22‐deydrocampesterol were isolated for the first time from Vicia sativa root exudates, and their biological activity as stimulants of Orobanche or Phelipanche sp. seed germination was reported. Seed dormancy is a common strategy of many plants to survive in natural and agricultural ecosystems. This study examined the effects of some chemical factors, including gibberellic acid and potassium nitrate, the prechilling temperature, duration and conditions and the light regimes on the seed dormancy‐breaking of Papaver rhoeas L. and Papaver dubium L. The results showed that all the tested seed dormancy‐breaking treatments significantly stimulated the germination of the two Papaver species. The seed germination was significantly influenced in relation to the seeds that were treated with 0–1250 p.p.m. of gibberellic acid. The maximum germination was observed in P. rhoeas L. at 750 p.p.m. and in Pa. dubium L. at 500 p.p.m. in the light/dark regime. There was a significant increase in the germination with an increase in the potassium nitrate concentration. The greatest germination was achieved at the lowest potassium nitrate concentration (0.5 g L?1) in the light/dark regime. The wet prechilling was more effective than the dry prechilling in the dormancy‐breaking of both P. rhoeas L. and Pa. dubium L. The highest germination percentage was found in the wet prechilling condition after 45 days' prechilling duration. The combination of gibberellic acid and potassium nitrate was more successful than their separate application in the stimulation of germination in both Papaver species. Among the combined treatments of prechilling and gibberellic acid, the highest germination of P. rhoeas L. was recorded in the seeds that were treated with wet prechilling for 45 days with 750 p.p.m. gibberellic acid, while in Pa. dubium L., the maximum germination was recorded with wet prechilling for 45 days with 500 p.p.m. of gibberellic acid.  相似文献

Various Orobanche species are weedy and cause severe reduction in the yields of many important crops. The seeds of these parasitic weeds may remain dormant in the soil for many years until germination is stimulated by the release of a chemical signal from a host plant. In order to determine the effects of fenugreek root exudate on the induction of Orobanche crenata, Orobanche ramosa and Orobanche foetida seed germination, root exudate was collected from hydroponically grown fenugreek seedlings. Fractionation patterns obtained from column and thin layer chromatography of the fenugreek root exudate showed a set of metabolites differing in their polarity with stimulatory activity on Orobanche seed germination. The crude root exudate stimulated both O. ramosa and O. crenata seed germination to the same level caused by the synthetic germination stimulant GR24 at 10 mg L?1. It also stimulated O. foetida seed germination which did not respond to GR24. Active fractions of root exudate stimulated the germination of Orobanche species differentially.  相似文献

Plant-induced germination of Plasmodiophora brassicae resting spores was studied in a laboratory experiment. Spore reaction was analysed in nutrient solution with exudates from growing roots of different plant species – one host plant (Brassica rapa var. pekinensis) and four non-host plants (Lolium perenne, Allium porrum, Secale cereale and Trifolium pratense) – and in controls with distilled water and nutrient solution. It was found that root exudates from L. perenne stimulated spore germination more than exudates from the other plants, including those from the host plant. The effect could not be explained by differences in the nutritional composition of the solutions due to differential uptake of the plant species, or by differences in root activity, measured as exudation of soluble sugars. This is the first time such a separation of factors has been done in analysing the influence of plants on P. brassicae germination. Although stimulation of P. brassicae resting spore germination is not restricted to the presence of host plants, it seems to vary depending on the plant species.  相似文献

The response of microconidia from pathogenic and non-pathogenic Fusarium oxysporum to root exudates from tomato plants inoculated with different pathogenic and non-pathogenic F. oxysporum strains was studied. Root exudates from non-inoculated tomatoes highly stimulated the microconidial germination of the two tomato pathogens, F. oxysporum f.sp. lycopersici strain Fol 007 and F. oxysporum f.sp. radicis-lycopersici strain Forl 101587. In root exudates from tomato plants challenged with the pathogen Fol 007 the microconidial germination of Fol 007 was increased, whereas in root exudates from plants challenged with Forl 101587 the microconidial germination of Fol 007 was reduced. Root exudates of tomato plants challenged with the non-pathogenic unspecific F. oxysporum strain Fo 135 and the biocontrol strain Fo 47 clearly reduced microconidial germination of the pathogenic strain Forl 101587. Moreover, the microconidial germination rate of the biocontrol strain Fo 47 was increased in the presence of root exudates of tomato plants challenged with the tomato wilt pathogen Fol 007. These results indicate that pathogenic and non-pathogenic F. oxysporum strains alter the root exudation of tomato plants differently and consequently the fungal propagation of pathogenic and non-pathogenic F. oxysporum strains in the rhizosphere is affected differently.  相似文献

Orobanche spp. (broomrapes) are holoparasites that subsist on the roots of many important crops and can considerably reduce yield. The control of Orobanche spp. includes physical, chemical and biological methods. Interactions between parasitic angiosperms and their hosts first occur at the level of parasite seed germination. The seeds of all Orobanchaceae germinate in soil under natural conditions only in response to specific chemical exudates from the host plant. This study describes the influence of different plant growth regulators and host plant root exudates on germination and development of calli from Orobanche seeds in vitro . The effect of indole-3-acetic acid, gibberellic acid and kinetin on the germination of Orobanche seeds varied with concentration. These plant growth regulators also affected the period of germination and the structure of calli and protrusions. An in vitro system for the collection of tobacco root exudates was established. Compounds released from the host roots of three different tobacco cultivars were found to provoke high levels of germination of the Orobanche seeds without any period of pre-conditioning. This study developed methods for the investigation of host–parasite interactions and the effect of germination stimulants in Orobanche spp.  相似文献

The seed germination behaviour of four populations each of Diplotaxis erucoides (L.) DC. and D. virgata (Cav.) DC, two weedy Brassicaceae species widely distributed in the Mediterranean region, was studied under controlled light, temperature and moisture conditions. Germination rates in D. virgata were generally higher at alternating (25/15^C) and low (15^C) temperatures, whereas in D. erucoides optimal germination rates were achieved at higher temperatures (25^C). No correlation between seed weight and germination was found in either species. In D. virgata, the geographic origin of the seed had an important effect on germination percentage. Great germination variability was found among the populations and even among seeds belonging to the same population. In this species, the application of gibberellic acid stimulated germination, especially in the populations with higher dormancy levels. Dormancy was found in mature D. virgata seeds but not in those of D. erucoides. In both species, germination percentages decreased after a certain period of storage at low temperatures. This period varied depending on the species and the population considered.  相似文献

Parasitic weeds of the genera Striga and Orobanche spp. cause severe yield losses in agriculture, especially in developing countries and the Mediterranean. Seeds of these weeds germinate by a chemical signal exuded by the roots of host plants. The radicle thus produced attaches to the root of the host plant, which can then supply nutrients to the parasite. There is an urgent need to control these weeds to ensure better agricultural production. The naturally occurring chemical signals are strigolactones (SLs), e.g. strigol and orobanchol. One option to control these weeds involves the use of SLs as suicidal germination agents, where germination takes place in the absence of a host. Owing to the lack of nutrients, the germinated seeds will die. The structure of natural SLs is too complex to allow multigram synthesis. Therefore, SL analogues are developed for this purpose. Examples are GR24 and Nijmegen‐1. In this paper, the SL analogues Nijmegen‐1 and Nijmegen‐1 Me were applied in the field as suicidal germination agents. Both SL analogues were formulated using an appropriate EC‐approved emulsifier (polyoxyethylene sorbitol hexaoleate) and applied to tobacco (Nicotiana tabacum L.) fields infested by Orobanche ramosa L. (hemp broomrape), following a strict protocol. Four out of 12 trials showed a reduction in broomrape of ≥95%, two trials were negative, two showed a moderate result, one was unclear and in three cases there was no Orobanche problem in the year of the trials. The trial plots were ca 2000 m2; half of that area was treated with stimulant emulsion, the other half was not treated. The optimal amount of stimulant was 6.25 g ha?1. A preconditioning prior to the treatment was a prerequisite for a successful trial. In conclusion, the suicidal germination approach to reducing O. ramosa in tobacco fields using formulated SL analogues was successful. Two other options for weed control are discussed: deactivation of stimulants prior to action and biocontrol by Fusarium oxysporum. © 2016 Society of Chemical Industry  相似文献

Broomrapes (Orobanche spp.) are parasitic weeds that cause significant losses of crop yield. Experiments were conducted to investigate the seed response to the artificial germination stimulant GR24 in three species of Orobanche subjected to preconditioning under various temperatures, water potentials and with plant growth regulators. The highest germination percentages were observed in Orobanche ramosa, Orobanche aegyptiaca and Orobanche minor seeds conditioned at 18°C for 7 days followed by germination stimulation at 18°C. With the increase of the conditioning period (7, 14, 21 and 28 days), the germination percentage of O. ramosa and O. aegyptiaca progressively decreased. When conditioned at −2 MPa, the germination percentage was lower than at 0 and −1 MPa, especially at 13 and 28°C. Orobanche minor seeds could retain relatively high germination if conditioned at 18, 23 or 28°C, even after significantly extended conditioning periods (up to 84 days). GA3 (30–100 mg L−1), norflurazon and fluridone (10–100 mg L−1), and brassinolide (0.5–1.0 mg L−1) increased seed germination, while 0.01 mg L−1 uniconazole significantly reduced germination rates of all three Orobanche spp. The promotional effects of GA3 and norflurazon and the inhibitory effect of uniconazole were evident, even when they were treated for 3 days. Germination of Orobanche seeds was much lower when the unconditioned seeds were directly exposed to GR24 at 10−6 m . This early GR24-induced inhibition was however alleviated or even eliminated by the inclusion of GA3 or norflurazon (10–50 mg L−1) in the conditioning medium. On the contrary, the inclusion of uniconazole increased the inhibitory effect of GR24, particularly in the case of O. ramosa.  相似文献

In guava decline, Fusarium solani-immune guava trees become susceptible to extensive root rot caused by this fungus after parasitism by Meloidogyne enterolobii. To understand the mechanisms involved in this disease, root exudates were collected from nematode-inoculated (NI) or uninoculated (UN) guava plants cultivated in sand. After filtration through a Millipore® membrane, NI and UN exudates were used: i) to prepare media to assess their effect on mycelial growth and production of propagules of F. solani isolate UENF/CF 163, and ii) to incubate macro- and microconidia to assess their effect on germination. NI exudates promoted (P?<?0.05) more mycelial growth and production of propagules than UN exudates or water. NI and UN exudates were used to water guava seedlings laid over seed germination paper inside plastic boxes. Half of the seedlings had an agar plug colonized by the fungus positioned in the collar region. Upon watering with NI exudates the fungus caused (P?<?0.05) extensive rotting of the seedlings’ rootlets. NI and UN exudates, either unlyophilized or lyophilized and re-suspended to the original concentration, were used to water guava seedlings grown in sterile sand before being inoculated (or left uninoculated) as described before. Solely upon watering with NI exudates, in its unlyophilized form or after lyophilization, the fungus caused a reduction (P?<?0.05) of shoot and root biomass associated with rotting of roots. These results suggest that M. enterolobii induces chemical changes in the root exudates of guava trees, which are necessary for root invasion causing root rotting by F. solani.  相似文献

Salinity and Orobanche or Phelipanche spp. infection are important crop stress factors in agricultural areas. In this study, we investigated the effect of salt stress on Phelipanche ramosa seed germination and its attachment onto Arabidopsis thaliana roots. We also evaluated the effect of both stresses on the expression of genes regulated by abiotic and biotic stresses. According to our results, high concentration of NaCl delayed P. ramosa seed germination in the presence of a strigolactone analogue (GR24). A similar pattern was observed in the presence of A. thaliana plants. Furthermore, we found that salt‐treated A. thaliana seedlings were more sensitive to P. ramosa attachment compared with the untreated plants, indicating that there was a positive correlation between salt sensitivity and the ability of P. ramosa to infect A. thaliana plants. At the molecular level, a synergystic effect of both salt and P. ramosa stresses was observed on the cold‐regulated (COR) gene expression profile of treated A. thaliana seedlings. Our data clarify the interaction between parasitic plants and their hosts under abiotic stress conditions.  相似文献

In studies on competition between crops and weeds it may be desirable to manipulate time of emergence, density and spatial arrangement of weeds. This requires a method to obtain a fast and uniform emergence of weeds. Two planting methods were compared that differed in the medium (agar or blotting paper) on which seeds were germinated, and in the method of transplanting the seeds (with or without the medium). In wet soil, emergence started earlier when seeds were germinated on agar and subsequently planted with agar. Root tips of seeds germinated on blotting paper may have been damaged to a greater extent during transplanting, because root tips tended to grow into the blotting paper. The duration, heterogeneity and final level of emergence were not affected by the planting method. In dry soil in a greenhouse experiment, the final level of emergence was much less when seeds were planted using the blotting paper method than when they were planted using the agar method. The agar may have provided a small source of water which reduced dehydration of emerging plants. The results were similar for the three species studies, namely, Chenopodium album L., Senecio vulgaris L. and Solanum nigrum L.  相似文献

Buthidazole (3-[5-(1,1-dimethylethyl)-1,3.4-thiadiazol-2-yl]-4-hydroxy-l-methyl-2-imidazolidinone) at concentrations of 10?6-10?4M did not affect germination of corn (Zea mays L.,‘Pioneer 3780’), redroot pigweed (Amaranlhus retroflexus L.), alfalfa (Medicago saliva L., ‘Vernal’), and quackgrass (Agropyron repens(L.) Beauv.) seeds. Stressing the seeds obtained from mature corn plants treated either pre-emergence or pre- plant incorporated with buthidazole at several rates by accelerated ageing and cold treatments further indicated that this herbicide did not affect germination. Total photosynthesis and dark respiration of corn plants 12 days after pre-emergence application and of redroot pigweed, alfalfa, and quackgrass plants after postemergence application of buthidazole at several rates were measured with an infrared CO2 analyser. The results suggested that buthidazole was a rapid inhibitor of photosynthesis of the sensitive redroot pigweed and quackgrass plants, with less effect on corn and alfalfa. Buthidazole did not affect respiration of the examined species except for a transitory increase in corn and alfalfa 12 days after pre-emergence or 4 h after postemergence treatment with buthidazole at 0.56 or 1.12 and 2.24 kg/ha, respectively. A long-term inhibition of quackgrass respiration 96 h after treatment with buthidazole at 1.12 and 224 kg/ha was also evident.  相似文献

Abstract

Striga asiatica (L.) Kuntze, a root parasite, causes severe loss of yield in sorghum and several other crops. The seeds of the parasite are induced to germinate by a stimulant in the host root exudate. Presowing hardening of the host with vanillic acid, caffeic acid and ferulic acid (25 ppm) reduces the induction of seed germination in the parasite by the host root exudate. The treatment causes a slight improvement in the dry matter production in the host and in addition, increases the phenolics level in the host root exudate. The latter effect might be responsible for reducing germination in Striga. If the treatment remains effective under field conditions also, it reduces significantly the incidence of Striga in cultivated fields.  相似文献

通过水培法收集韭菜、大葱、野薄荷、藿香和薰衣草的根系分泌物,研究其对大丽轮枝菌菌丝生长、孢子萌发的影响,并通过室内盆栽试验研究了植物根系分泌物对大丽轮枝菌微菌核萌发的影响。结果表明,5种植物根系分泌物对大丽轮枝菌菌丝生长和孢子萌发均表现为抑制作用;随着浓度的升高,抑制作用增强,而在同一浓度下,随着处理时间的延长,根系分泌物对孢子萌发的抑制作用逐渐减弱。盆栽试验表明,植物根系分泌物对大丽轮枝菌微菌核的萌发均表现为抑制作用,且微菌核对韭菜根系分泌物最为敏感。5种植物中,韭菜根系分泌物的抑制作用最强,野薄荷次之,藿香最弱。  相似文献

Fusarium wilt caused by Fusarium oxysporum f. sp. pisi (Fop) is one of the major constraints of pea worldwide. Its control is difficult and is mainly based on the use of resistant cultivars. This study aimed to identify and characterize resistance mechanisms interfering with Fop spore germination, as an additional pre‐penetration resistance mechanism little explored so far. For this, root exudates were collected from 12 pea accessions with differential responses to the disease, from resistant to susceptible, and their effects on Fop germination and growth were determined. While root exudates from most accessions stimulated Fop germination, the root exudates of three accessions, JI 1412, JI 2480 and P42, did not stimulate, or even inhibited, Fop germination. Although some additional compounds might be involved, the analysis showed that the most active metabolite was the pea phytoalexin pisatin. Pisatin was identified in the active fraction of pea root exudate extracts and its amount in the root exudates was negatively correlated with the extent of Fop germination. This suggests an important role of pisatin in the constitutive defence of pea against F. oxysporum.  相似文献

Germination behaviour of seeds of Amaranthus retroflexus L, harvested under natural conditions The influence of time of emergence of seed-bearing plants and of the climate at the time of dissemination on dormancy of seeds of Amaranthus retroflexus L. formed solely on the main inflorescence was studied. Under conditions favourable to germination (30 C) the seeds harvested latest (November) are at shedding the least dormant and those produced very early (August) the most dormant. But after dry storage for 60 days, the seeds had a germinative capacity near 100% whatever their origin. Under conditions less favourable to germination (20 C) the seeds harvested from the plants which had developed very late (June-July) germinated better than those developed on plants which appeared very early (March April). These differences were not overcome by dry storage. In the light of these facts, the authors discuss the elTects of interactions between climatic factors and the physiological state of the plants on the germinability of naturally-disseminated seeds.  相似文献

Studies have been carried out on the herbicidal action of asulam [methyl (4-aminophenylsulphonyl)carbamate] and sulphanilamide, alone or in association either with 4-aminobenzoic acid (4ABA) or 4, 6-diamino-1-(3, 4-dichlorophenyl)-1, 2-dihydro-2, 2-dimethyl-1,3,5-triazine (DCDT). The soaking of wheat seeds (Triticum estivum L.) for 12 h at 30°C in asulam and DCDT in a 10:1 ratio doubled the inhibition of root growth produced by soaking in asulam alone; the addition of 4ABA partially reversed the activity of asulam. Foliar applications of a mixture of asulam + DCDT (1.1 + 0.55 kg ha?1) markedly increased the activity of asulam in susceptible wheat, wild oat (Avena fatua L.), tolerant flax (Linum usitatissimum L.), and in Stellaria media L. The activity of asulam at 1.1 kg ha?1 was reversed by 4ABA at 2.2 kg ha?1 by about 50% in wheat and wild oat, 82% in flax and 100% in S. media. The results indicate that asulam and sulphanilamide act by similar mechanisms in apparently inhibiting the biosynthesis of folic acid.  相似文献
