Diploid wheat, Triticum monococcum s.l., is a host for the wheat leaf rust fungus, Puccinia triticina. Some accessions have been reported to show a high degree of prehaustorial resistance. This is non-hypersensitivity resistance, which acts before the formation of haustoria by the pathogen. To assess the frequency of prehaustorial resistance 598 accessions of diploid wheat were inoculated with the wheat leaf rust isolate Felix. Most T. monococcum s.s. accessions (84%) were resistant whereas all T. urartu and all but three T. boeoticumaccessions were susceptible. Histological components analysis revealed that a high percentage of prehaustorial resistance to P. triticina was found in only three T. monococcum accessions. No haustoria were observed in such infection units confirming the prehaustorial nature of the resistance. Prehaustorial abortion of certain infection units in an accession always coincided with posthaustorial abortion of the other infection units. The bottleneck restricting introgression of useful genes directly from diploid into hexaploid wheats is the low number of BC1F1 seeds obtained. In crosses between hexaploid wheat (Triticum aestivum L.; AABBDD) and Aegilops squarrosa L. (DD) or T. urartu Thum. (AA), this bottleneck may be overcome simply by pollinating a sufficient number of F1 spikes. However, hybrids between hexaploid wheat cultivars (T. aestivum) and T. monococcum L. (AA) generally are highly female-sterile, often having no pistils. One T. monococcum accession, PI 355520, when crossed with T. aestivum, produced hybrids with female fertility in the same range as that of T. aestivum/A. squarrosa or T. aestivum/T. urartu hybrids, ca. 0.5 to 1.0 backcross seed per spike. We found that female fertility was controlled by two duplicate genes in PI 355520, and that this accession can be used as a bridging parent to introgress genes from other T. monococcum accessions into hexaploid wheat. Pairing of homologous chromosomes was less frequent and weaker in such crosses than in T. aestivum/A. squarrosa crosses, but homoeologous bivalents occurred at a rate of almost 0.5 II per cell. Restitution division was detected in crosses involving all three diploid species and was confirmed cytologically in crosses with PI 355520. Chromosome numbers of BC1F1 plants ranged from 35 to 67; plants with 49 or more chromosomes occurred at frequencies of 0.09 to 0.21 among progeny of A. squarrosa and T. urartu and 0.29 in progeny of T. aestivum/T. monococcum crosses involving PI 355520. These results are consistent with those of previous studies, demonstrating the potential of direct Hexaploid/diploid crosses for rapidly introgressing useful genes into Hexaploid wheat with minimum disturbance of the background genotype.  相似文献

A total of 259 accessions of wild Triticum species originating from different countries, along with 91 triticale (6×)× bread wheat true-breeding derivatives, two bread wheat, and three triticale cultivars were screened for resistance to the Russian wheat aphid, a serious insect pest of the wheat crop. Twenty-four entries with low damage ratings on the basis of amount of leaf rolling and leaf chlorosis were retested along with resistant and susceptible controls. On the basis of leaf roll damage ratings, eight entries including four Triticum monococcum var. boeoticum (T. boeoticum), one T. monococcum var. monococcum (T. monococcum), two T. timopheevii var. araraticum (T. araraticum), and one triticale cultivar were significantly superior to ‘Karl’ (susceptible control) wheat. Among these, four accessions — three T. boeoticum and one T. araraticum— were significantly superior to all other entries and were equal to the resistant control (PI 372129) in resistance rating based on leaf rolling and leaf chlorosis (except T. boeoticum TA 202). The leaf chlorosis damage rating of all accessions were significantly lower than that of the susceptible check.  相似文献

Seed germination, seedling growth, mitotic and nucleolar activity were investigated as indicators of metabolic activity in a diploid wheat, Triticum monococcum ssp. monococcum cv. Einkorn. Two developmental phases were identified, an initial lag phase during which reactivation of NOR's and the initiation of mitotic activity (10 h) took place prior to the appearance of the primary root (16 h). Dominance interactions were observed between the NOR's of chromosomes 1A and 5A with the NOR's of the one pair activated before the other and forming therefore larger nucleoli. The total number of nucleoli per 500 cells also varied according to the stages of the cell cycle over time. The mitotic activity had an initial exponential rise, but showed signs of flattening out after 10 h of mitosis with opposite levels of activity in the primary and first pair of lateral roots thereafter. Secondly, an exponential phase in root and shoot growth, mobilization of reserve food and respiration rate was observed during seedling growth. Comparisons were done on work from a previous investigation on two polyploid wheat accessions belonging toT. turgidum ssp. durum and T. aestivum species. T. monococcum displayed a higher embryo growth than the polyploids, that may be explained by having a lighter embryo and caryopsis weight that may influence the uptake of water and reactivation of metabolic activity. During the seedling growth period, the two polyploid species displayed a faster growth rate than the diploid probably due to the differences in the number of genomes, the hybrid nature of the polyploids and a more complex gene control in the polyploids. Summary The increasing popularity of organic agriculture and health food products has led to a renewed interest in hulled wheat species such as emmer (Triticum turgidum ssp. dicoccumSchubler). Knowledge on agronomic and quality traits is required for effective and efficient use of germplasm collections in breeding programs. The objective of this study was to estimate agronomic and grain quality traits of emmer wheat cultivated in Italy. A total of 20 emmer accessions consisting of landraces, breeding lines or cultivars selected from landraces and modern cultivars were examined under low input conditions. The study was conducted for three successive years (2002–2004) at one location of Southern Italy (Foggia). The entries were characterized for agronomic and qualitative traits [grain yield (t ha−1), thousand grain weight (g), test weight (kg hl−1), grain protein content (%), HMWG composition, dry gluten content (%), gluten index and yellow index, alveograph indices and Total Organic Matter (TOM) on cooked pasta]. The results showed a large genetic variability for most of the traits measured and, even if most of the accessions showed inferior bread- and pasta-making performance, modern cultivars exhibited improved quality traits with some potential to perform healthy and tasty food.  相似文献

Summary Digitaria eriantha is a subtropical grass which is widely sown as a pasture species in South Africa and cultivars have recently been released in Australia. The species is variable in seed production and in other agronomic attributes. This paper describes a breeding program designed to combine the seed production of cv. Premier with the high productivity in winter-spring of an almost sterile genotype.Hybrids between the two accessions were either triploid and sterile, or diploid with low fertility. The F2 to F5 generations were selected intensively for fertility and, at two contrasting sites, for spring productivity, with both maternal and paternal selection. The F6 generation was grown as replicated swards and yield in spring, and fertility, were used for selection of elite F5 plants to comprise the parents of a new cultivar (cv. Advance).In evaluation trials, spring dry matter production of Advance averaged 40% higher than that of Premier. On average, seed production of Advance was 11% lower than that of Premier and botanical fertility was consistently about half that of Premier. Experimental yields of cleaned seed of Advance were up to 138 kg/ha. In commerce, seed yields of Advance are likely to be lower than those of Premier unless higher inflorescence densities than Premier can be achieved.  相似文献

Near-isogenic Rht lines of ten modern bread wheat (Triticumaestivum L.) and six durum wheat (T. turgidum L.) cultivars weredeveloped and evaluated in replicated trials under three soil moisturetreatments for two years in northwestern Mexico. The three soil moisturetreatments were created by providing one, two or six irrigations during eachcrop season. Grain yield and other traits were measured for each line ineach trial. Mean grain yields of short and tall T. aestivum or T.turgidum isolines were similar in the lowest yielding environment whenmean grain yields (0% grain moisture) of T. aestivum and T.turgidum were 2,232 and 1,870 kg ha-1, respectively. Mean grainyield of dwarf T. aestivum was significantly higher than that of tallgenotypes in another five trials with moderate to high yields. Theperformance of dwarf and tall T. turgidum isolines was unpredictablein moderate yielding trials, and the dwarf isolines yielded significantly morein trials that received six irrigations. Given that the tall isolines producedsignificantly more straw than their shorter counterparts, cultivation of tallwheats may be beneficial in semiarid environments where farmers' yields areclose to 2.5 t ha-1 or lower, and straw has value.  相似文献

Flanking sequences of the puroindoline a(pinA) and puroindoline b (pinB) genes from Triticum monococcum, T. urartu, Aegilops speltoides and A. tauschii were obtained by inverse PCR. Two accessions from each of these related diploid taxa were sequenced, and the sequences compared to those of the soft hexaploid wheat cultivar `Penawawa'. As expected from the origin of the D-genome in hexaploid wheat, the highest sequence identity was observed for the A. tauschii sequences, whereas the sequences for A.speltoides were the most divergent. The promoter sequences were further analyzed for putative regulatory elements, and the possible significance of several highly conserved sequence motifs is discussed. Western blots of Triton X-114 extracted proteins separated by SDS-PAGE confirmed the accumulation of pinA and pinB gene products in the endosperm of all the germplasm lines studied. The deduced amino acid sequences for the related diploid taxa do not imply any major structural changes as compared to the wild-type T. aestivum puroindolines, and is therefore in agreement with the soft endosperm texture of these species. Summary Susceptibility to drought stress is a key factor in dry land lentil (Lens culinaris Medikus) production in the Mediterranean region of West Asia and North Africa. This study examined the response to drought stress of 121 accessions representing all subspecies of the genus Lens; cultivated, and the wild L. culinaris ssp. orientalis (Boiss.) Ponert, L. culinaris ssp. odemensis (Ladiz.), L. nigricans M.B. Godr. ssp. nigricans Godr. and L. nigricans ssp. ervoides (Brign.) Ladiz. for their potential use in breeding for dry land conditions. Accessions were grown under two moisture regimes (dry land and dry land plus supplemental irrigation) at Breda, Syria during the 1990–91 and 1991–92 seasons. The cultivated lentil had markedly superior seed and straw production than did the wild Lens species. Time to flowering accounted for less than 10% of the variation in yield of wild accessions under rainfed conditions in the two seasons, showing that, in contrast to the cultivated germplasm, drought escape was relatively unimportant in wild lentil. Performance under drought in wild lentil, measured in terms of dry land seed yield or drought susceptibility index (S), was randomly distributed among collection locations with little relation to collection site aridity. Direct selection of wild lentil germplasm for biomass yield under dry conditions is of little value and an evaluation of wild accessions in hybrid combination is needed.  相似文献

This study was conducted to determine if Brassica germplasm bulks created and maintained by the USDA-ARS North Central Plant Introduction Station (NCRPIS) were made with genetically indistinguishable component accessions and to examine newly identified putative duplicate accessions to determine if they can be bulked. Using ten microsatellite primer pairs, we genotyped two bulks of B. rapa L. ssp. dichotoma (Roxb.) Hanelt comprising four accessions and three bulks of B. rapa L. ssp. trilocularis (Roxb.) Hanelt comprising fourteen accessions, as well as four pairs of putatively duplicate accessions of B.␣napus L. Assignment tests on ten individual plants per accession were conducted using a model-based clustering method to arrive at probabilities of likelihood of accession assignment. The assignment tests indicated that one of the two bulks of B. rapa ssp. dichotoma involves genetically heterogeneous accessions. It was observed in the B. rapa ssp. trilocularis bulks that the component accessions could be differentiated into groups, with misassignments observed most frequent within groups. In B. napus, only one of the four pairs of putative duplicates showed significant genetic differentiation. The other three pairs of putative duplicates lack differences and support the creation of bulks. The results of the assignment tests were in agreement with cluster analyses and tests of population differentiation. Implications of these results in terms of germplasm management include the maintenance and/or re-creation of some Brassica germplasm bulks by excluding those accessions identified as being unique in this study.  相似文献

Seed dormancy is one of the most important parameters affecting the malting process and pre-harvest sprouting in barley (Hordeum vulgare L.). Variation of seed dormancy in 4365 cultivated and 177 wild barley (ssp. spontaneum) accessions derived from different regions of the world was investigated in Okayama University, Kurashiki, Japan. Seed dormancy of each accession was estimated from their germination percentages at 0, 5, 10 and 15 weeks post-harvest after-ripening periods. All of the wild barley accessions showed less than 10% germination at 0 week after-ripening period. Level of seed dormancy in 4365 cultivated barley accessions showed a clear geographical differentiation. Seventy seven percent of Ethiopian accessions showed high germination percentages, while 86% of Japanese, Turkish and North African accessions showed low germination percentages at 0 week after-ripening period. A half diallel cross using eleven barley accessions with different level of dormancy revealed that seed dormancy was predominately controlled by additive gene effects. These results suggest that large genetic diversity for seed dormancy in barley is explained as different levels of additive accumulation of genetic factors. Barley varieties showing appropriate dormancy could be developed by crossing among barley germplasm accessions used in the present study.  相似文献

Summary A geographically representative selection of germplasm of Beta vulgaris, section Beta has been assessed for characteristics important in sugarbeet breeding, including downy mildew resistance, resistance to aphid colonisation and infection by the beet virus yellow complex. The occurrence of maintainer lines for cytoplasmic male-steriles was also investigated. Desirable qualities were found in some accessions, including nothern European wild vulgaris ssp. maritima and some old multigerm cultivars of fodder beets.  相似文献

Granule-bound starch synthase (GBSS) is the primary enzymeresponsible for the synthesis of amylose in amyloplasts of cereal endospermcells. In bread wheat there are three structural genes (Wx-A1, Wx-B1,and Wx-D1) encoding for isoforms of GBSS. The loss of one or moreGBSS isoforms results in the reduction (partial-waxy) or absence (waxy) of amylose in the starch. Waxy wheats may find application inthe production of modified food starch and their flour may be used toextend the shelf life of baked products. In order to breed high qualitywheats able to produce bread with delayed staling, the genetic variabilityfor the waxy trait in our germplasm has been investigated. Weanalysed 288 cultivars of bread wheat, 139 cultivars of durum wheat andabout 200 accessions from other Triticum species. Gel electrophoresisshowed 63 bread wheats deficient in the Wx-B1, one in the Wx-A1 and one in the Wx-D1 protein isoforms, as well as one Triticum dicoccum lacking the Wx-A1 isoform. None of the analysedTriticum monococcum, Triticum durum, Triticum speltaand Triticum timopheevi accessions showed mutations at the Wxloci. The wheat accessions with Wx mutations were evaluated with aRapid Visco Analyser (RVA) to investigate starch properties. All theanalysed cultivars showed Peak Viscosity and Final Viscosity different fromthe normal wheat. Other analyses to evaluate the rheological characteristicsof the partial-waxy genotypes are under way and a breedingprogramme to select new waxy wheat varieties is in progress  相似文献

Summary Seven spelt wheat accessions of different origin were hybridized with the susceptible bread wheat cultivar Taichung 29 in order to study the genetics of their resistance to yellow rust (Puccinia striiformis Westend. f. sp. tritici). One Iranian and five European accessions were found to carry Yr5 of Triticum aestivum ssp. spelta var. album, whereas a factor for resistance in the Iranian accession 415 was confirmed to be genetically distinct from Yr5. The alleles for resistance in each of the accessions studied showed a monogenic dominant mode of inheritance. Twenty-eight spelt wheat accessions, including those studied for their resistance to yellow rust, were subjected to polyacrylamide-gel-electrophoresis to study variation for gliadin storage protein patterns. Thirteen distinct patterns were revealed, implying the presence of duplicates within the studied spelt wheat collection.  相似文献

Summary The characterization of 39 Italian ecotypes and cultivars of Triticum turgidum L. spp. dicoccum Shrank ex Schübler (emmer wheat) was performed utilizing agro-morphological and molecular tools. Emmer wheat is a hulled species which grows wild in the Near East and is still cultivated in the Mediterranean Basin. Due to its characteristics, in Italy it is cultivated mainly in marginal lands of central and southern Italy, where local varieties, adapted to the natural environment where they originated, are used. Emmer wheat cultivation has been drastically reduced during the last century as a consequence of its low yield. Nevertheless, more recently, its agronomic and nutritive values, together with its use in health food products, made its cultivation economically viable in the marginal lands with a parallel increase of the cultivated area which is now more than 2000 ha. In the present paper the results of morphological evaluation, carried out in an experimental field in central Italy using a randomized block design with three replications, and molecular characterization are reported. The analysed material showed distinctive molecular traits and the existence of a huge amount of diversity not only between varieties, but also within them. When the accessions were clustered utilizing their genetic distance, the clusters were not always in agreement with the accessions origins. The obtained results gave information that can be useful for: (i) future registration of material, (ii) germplasm conservation and (iii) use of this valuable source of emmer germplasm for future breeding programmes.  相似文献

Summary Mycorrhizal colonization and growth dependency were studied at a single rate of phosphorous application in wild and cultivated primitive and modern wheats, inoculated with Glomus intraradices Schenck & Smith. Mycorrhizal colonization found in Triticum timopheevii var. araraticum (AAGG) was higher than that found in the other tetraploid wheats (AABB). Mycorrhizal dependency was higher in representatives of the D genome donor — Aegilops squarrosa, compared with representatives of the A and possible B genome donors T. monococcum and Ae. sharonensis, Ae. longissima and Ae. speltoides, respectively. The nature of response to VAM in hexaploid wheat was controlled by factors of the A and B genomes which are epistatic over those located in the D genome. The high mycorrhizal colonization and dependency which was found in T. timopheevii var. araraticum may indicate special genomic affinity possessed by the G genome of wheat in VAM interaction. Based on the 27 wheat lines and species tested in this study only low correlation between G. intraradices colonization and its contribution to plant growth can be suggested.  相似文献

Granule‐bound starch synthase of ancient wheat and related species was examined by sodium dodecyl sulphate polyacrylamide gel. A total of 13 different alleles were revealed in a collection of three accessions of diploid wheat, six accessions of tetraploid wheat, 49 accessions of spelt wheat, nine accessions of Sitopsis and two accessions of Aegilops tauschii. A new allele named Wx‐A1a′ appeared in four spelt wheat accessions. The tetraploid wheat accessions evaluated did not show any polymorphism; nevertheless the tetraploid accessions of Sitopsis section revealed three novel alleles. The novel allele Wx‐Ddn1g was found in two accessions of A. ventricosa and the Wx‐Ddcm1h and Wx‐Ddcm1i in two accessions of A. crassa. A novel allele named Wx‐Au1g was found in Triticum urartu, which is different from the also new Wx‐Am1h allele of T. monococcum. The diploid‐related species accessions revealed two novel alleles named Wx‐Bsl1h and Wx‐Bs1g found, respectively, in A. longissima and A. speltoides. The amylose content was measured for the different alleles found in all evaluated species and no significant effects of the allele composition on the amylose content were detected.  相似文献

Although western wheatgrass [Pascopyrum smithii (Rydb.)] is an important perennial grass species for agriculture and conservation management in central and western North America, its lack of adequate seed production and seedling vigor limits its effectiveness. To address the weaknesses a study was conducted to assess rhizome spread, seed production, seed weight, germination percentage, and emergence rate of seed produced from 48 western wheatgrass cultivars and germplasm accessions at a field site near Nephi, UT, USA during 2007 and 2008. The western wheatgrass cultivars had approximately two times higher seed production than the germplasm accessions during both 2007 and 2008 and also had higher seed weight in 2007 and emergence rate in 2008. The germplasm accessions had higher seed weight in 2008. For the remaining traits there were no differences among the different germplasm sources. Based on principle component analysis a subset of cultivars and germplasm accessions with high seed production and emergence rate were identified that could be used to produce improved cultivars and germplasms. There was little evidence of strong relationship between geographic, genetic, and phenotypic distances among the various lines examined. Additionally, based on genetic marker data, a subset of lines was grouped into three populations. Based on these results, selection among lines could occur to maximize agricultural performance regardless of site of origin, or within population selection could be practiced to meet conservation goals of minimizing hybridization among populations.  相似文献
