Reliable characterization of the variation among wild and cultivated yams in Nigeria is essential for improved management and efficient utilization of yam genetic resources. RAPD and double stringency PCR (DS-PCR) analyses were used to investigate genetic relationships and the extent of redundancy among 30 accessions of two cultivated, and 35 accessions of four wild yam species collected from Nigeria. Twenty-five selected random decamer and two microsatellite primers were used individually and in combination to generate DNA profiles for each accession of the six Dioscorea species. The number of amplified fragments varied from 7 to 18 fragments per primer/primer combination. Different levels of intraspecific genetic diversity were found, with Dioscorea rotundata Poir. being the most variable. Based on identical profiles for the RAPD and DS-PCR primers, 12 duplication groups consisting of a total number of 37 accessions were observed in the present study. An UPGMA analysis grouped the majority of plants according to the species. Cultivated yams belonging to the D. cayenensis–rotundata species complex, which were classified into seven morphotypes/varietal groups, could be clearly separated into two major groups corresponding to D. rotundata Poir. and D. cayenensis Lam. D. cayenensis cultivars exhibited a low level of intraspecific variation and were genetically close to the wild species Dioscorea burkilliana J. Miège. D. rotundata cultivars classified into six varietal groups showed a high degree of DNA polymorphism and were separated into two major groups that appeared most closely related to Dioscorea praehensilis Benth. and Dioscorea liebrechtsiana de Wild. We propose, based on these results, that cultivars classified into D. cayenensis should be considered as a taxon separate from D. rotundata. The implications of intraspecific variability for the ex situ conservation of wild and cultivated yam germplasm in Nigeria are discussed.  相似文献

Lupinus angustifolius L. is a Mediterranean species, domesticated in the 20th century, representing an important grain legume crop in Australia and other countries. This work is focused on the collection of wild germplasm and on the characterisation of morphological and molecular diversity of germplasm accessions. It reports the collection of 81 wild L. angustifolius accessions from the South and Centre of Portugal, available at the ‘Instituto Superior de Agronomia Gene Bank’, with subsequent morphological and molecular characterisation of a selection of these and other accessions. A multivariate analysis of morphological traits on 88 L. angustifolius accessions (including 59 wild Portuguese accessions, 15 cultivars and 14 breeding lines) showed a cline of variation on wild germplasm, with plants from Southern Portugal characterised by earlier flowering, higher vegetative development and larger seeds. AFLP and ISSR molecular markers grouped modern cultivars as sub-clusters within the wider diversity of wild germplasm, revealing the narrow pool of genetic diversity on which domesticated accessions are based. The importance of preserving, characterising and using wild genetic resources for L. angustifolius crop improvement is outlined by the results obtained.  相似文献

In this paper, we report on morphological diversity of Amygdalus L. species native to Lebanon, following a countrywide survey of almond germplasm whereby a total of 149 accessions were collected throughout the country and characterized by thirteen quantitative and four qualitative traits. The results indicated that the genetic diversity of Amygdalus communis L., Amygdalus korshinskyi Hand.-Mazz., and Amygdalus orientalis Duh. in Lebanon is high. Principal component analysis revealed that nut weight, nut volume, nut width, kernel volume and shell strength had highest loading in the first component that accounted for 38.7% and 46.7% of total variation in A. communis and A. orientalis, respectively. In contrast, leaf traits were present in the second component which accounted for 18% and 23.2% of total variation in each species, respectively. No significant correlations were detected between leaf parameters and fruit traits in both species. The results indicated that quantitative leaf characters for all three species were determined by rainfall and not altitude whereby adjacent accessions located in drier areas had smaller leaf sizes than those located in more humid regions. Quantitative fruit characters did not seem to vary accordingly. Qualitative leaf traits in all three species reflected a variability which was independent of rainfall. A. communis populations showed high variability, suggesting that they could be a valuable source in almond improvement programs.  相似文献

Two introduced wild species Helianthus annuus L. and H. petiolaris Nutt. have become widespread in central Argentina and overlap the sunflower crop region. Intermediate off-type plants between the wild and cultivated species are often found, which is of concern because of the recent release of imidazolinone resistant varieties and the likely use of genetically modified sunflower cultivars. The progeny of 33 off-type plants obtained from 14 representative sites of the diffusion area were studied to confirm hybrid origin. Germination, survival, morphological traits and days to flowering confirmed hybridization between crop and both wild species, when compared to eight accessions of typical wild plants. Some progenies were presumably crop–wild H. annuus hybrids, some originated from the cross of cultivated plants and H. petiolaris, and two were the advanced generation of a cultivated hybrid. Hence, morphological traits are a good clue for the identification of spontaneous hybrid plants at field. The results indicate that crop–wild hybridization and introgression occur at various places in central Argentina. This fact may represent a way to herbicide resistance escape and future transgene escape if GM sunflower cultivars are released for commercial use.  相似文献

The wild lettuce, Lactuca aculeata Boiss. et Ky., is closely related and fully interfertile with cultivated lettuce, Lactuca sativa L. Altogether, 155 accessions of this wild species, which is rare conserved within the world’s germplasm collections, were collected from 12 localities throughout northeastern Israel. Ten of these localities represent different sites across the Golan Heights, while two, high-density populations were suprisingly found at the Hula Plain (first report of this species in this region). Lactuca aculeata was recorded at various elevations (222–968 m a.s.l.) and habitats. The taxonomic status of 30 L. aculeata accessions was morphologically validated during ex situ seed regeneration of 31 random accessions representing all 12 localities. Characterization of 12 traits showed that the Israeli populations of L. aculeata do not exhibit broad morphological variability, but more great levels of variation were obtained for developmental traits. Relatively little variability was observed in the morphology of cauline leaves or in the distribution of anthocyanin pigmentation in bracteae. In two cases, L. aculeata plants lacked trichomes in the inflorescence, a typical feature in the majority of plants. These unique collections of wild lettuce may carry novel sources of genetic variation for a wide range of traits and, thus, should be of interest for careful evaluation and exploitation in lettuce breeding.  相似文献

Variation for 24 morphological traits measured in a greenhouse environment and 36 randomly amplified polymorphic DNA (RAPD) markers was assessed among 318 wild octoploid strawberry (Fragariaspp.) genotypes from diverse habitats across the northern USA. RAPD marker frequencies and certain leaf and flower morphology traits (petiole color, leaf mass/area ratio, leaflet length and width, flower and receptacle diameter, petal width, flowers/inflorescence) were significantly different between the F. chiloensis-platypetala and F. virginiana-glauca species complexes. The proportion of variation accounted for by provenance effects was lower for the RAPD markers than for most morphological traits, especially in the F. virginiana-glauca species complex. Morphological traits of potential adaptive importance group the collection into provenances within each species-complex, and reflect the significant habitat and geographic differences across the region from which the germplasm was collected. Variation among populations within provenances was low for the molecular and most morphological traits, with a much larger amount of variability among plants within populations. Most of the variation for the presumably more selectively-neutral RAPD data was among plants within populations and populations within provenances rather than among the provenances that were recognized based on morphological traits, especially in the F. virginiana-glauca complex. Patterns of diversity for morphological traits must be considered, along with more selectively-neutral molecular characters such as RAPDs, to formulate effective sampling strategies and to properly estimate the quantity and apportionment of diversity within this germplasm.  相似文献

Exploitation of wild gene pool for breeding is a common practice in an increasing number of cultivated plants. The cultivated lentil could not attain the substantial improvement in the yield potential due to loss of genes for higher productivity and lack of resistance against biotic and abiotic stresses. The absence of evaluation data of wild lentils for characters of economic importance, besides biotic and abiotic stresses, is one of the constraints in their use in lentil breeding programme. In the present study, 70 wild accessions from four wild Lens subsp./sp. (L. culinaris subsp. orientalis, L. odomensis, L. ervoides and L. nigricans) along with 3 checks (Precoz, PL-406 and PL-639) were evaluated for phenological and agro-morphological characters, for their reaction to three fungal diseases (wilt, powdery mildew and rust) and screened for tolerance to moisture stress. The wild accessions showed higher performance for branches/plant as compared to cultivated genotypes. Similarly, a few accessions of L. culinaris subsp. orientalis were earlier to flower and had higher seeds and seed yield/plant as compared to cultivated lentil. However, some were comparable with cultivated genotypes for flowers/peduncle, peduncle length and plant height. The mean performance for flowers per peduncle, leaflets per leaf, plant height, seeds and seed yield per plant increased, while decreased for days to flowering and maturity, and branches per plant during the evolution of cultivated lentil from the wild Lens taxa. Of Lens taxa, L. nigricans had the maximum resistant accessions for biotic and tolerance to abiotic stresses. The valuable variation existing among wild accessions can be exploited following introgression with cultivated lentils. It will help in the flow of useful genes from wild to cultivated lentil for generating wide spectrum of variability and its subsequent use in genetic restructuring of lentil.  相似文献

Seven Brassica species were evaluated for their resistance to the cabbage aphid, Brevicoryne brassicae, in a series of field experiments. Four wild Brassica species, two 8 chromosome species with similarities to the B genome of Brassica nigra (Brassica fruticulosa and Brassica spinescens) and two 9 chromosome species containing the C genome (Brassica incana and Brassica villosa) were identified as possessing consistently high levels of antibiosis mediated resistance to B. brassicae. None of the species were shown to possess consistently high levels of antixenosis resistance. In more detailed glasshouse experiments one B-like genome species, B. fruticulosa, showed considerable variation between accessions collected from different sites for resistance to B. brassicae. In addition, individual accessions of one A genome species (Brassica rapa) and one C genome species (Brassica alboglabra) were shown to be highly variable in their resistance to B. brassicae, some plants of each accession being highly resistant and others very susceptible. The implications of the variability in resistance to B. brassicae within wild Brassica species for exploitation in Brassica breeding programmes are discussed.  相似文献

A total of 359 accessions of oil palm (Elaeis guineensis Jacq.) originating from 11 African countries (Nigeria, Cameroon, Congo DR, Tanzania, Angola, Senegal, Sierra Leone, Guinea, Ghana, Madagascar and Gambia) were characterized using the RFLP method using the standard Deli dura as the check. Genomic DNA from each sample was digested using five restriction enzymes and hybridized with four oil palm cDNA probes. Data were analyzed using Biosys-1 computer software to calculate the genetic variability parameters. In general, all the collections exhibited higher levels of diversity than the standard variety, Deli dura. The standard variety, Deli dura, lost 36 alleles as compared to the natural populations indicating a reduction in genetic variability. Material from Nigeria showed the highest mean number of alleles per locus (1.9) and percentage of polymorphic loci (67.2%). These findings, combined with others, suggest that Nigeria may be the center of diversity of wild oil palm. It further suggests that oil palm natural populations maybe possessing adequate genetic variability that are potentially useful for improvement programs.  相似文献

The genus Asparagus (Asparagaceae) encompasses 100–300 species, including the important vegetable crop, A. officinalis (garden asparagus). Previous attempts to hybridize garden asparagus with A. densiflorus (Kunth) Jessop, with the aim of introducing disease resistance, were unsuccessful because of the failure of endosperm development. In the present study, reciprocal interspecific hybrids between garden asparagus and A. schoberioides Kunth were generated by hand pollination. The F1 hybrids were analyzed by using both morphological and molecular techniques. This is the first report describing the production of an interspecific hybrid between garden asparagus at the diploid level (2n = 2x = 20) and its diploid (2n = 2x = 20) wild relative, A. schoberioides. Morphological characteristics of candidate hybrids were a mixture of those found in the parents, and cytological and RFLP analyses confirmed that morphologically intermediate plants were indeed diploid hybrids of those two species. In other words, post-zygotic isolation is not complete between phylogenetically distinct these two species. Our results suggest that other Asparagus species may be capable of hybridizing with A. officinalis and that introducing wild characters through interspecific hybridization may offer advantages for breeding for novel traits. Appropriate planting patterns ensure sustained soil fertility and higher productivity for farmers. This study assessed the intercropping crop combinations and information sources of kola farmers in Osun State, Nigeria. Sixty respondents were selected using multistage and purposive random sampling techniques. Data were collected with a structured questionnaire. The arable crops commonly intercropped with kola were plantain/banana, yam, cassava, and maize. Tree crops combined with kola included cocoa, citrus, and oil palm. Co-farmers/friends and radio served as popular sources of information for farmers. Farm size was significantly related to respondents’ information sources on kola intercropping. The authors recommend that the Cocoa Research Institute of Nigeria's extension personnel make greater use of information sources most frequently used by respondents in order to ensure adequate cropping systems.  相似文献

Summary Leaf miner (Liriomyza cicerina Rond.) causes considerable damage to chickpea (Cicer arietinum L.) in West Asia, North Africa and Southern Europe. Use of resistant cultivar is the economical method of control. Screening of over 7000 germplasm accessions did not result in identification of highly resistant accessions to this insect. Therefore, 200 accessions of eight wild Cicer species were evaluated in search of resistance to leaf miner between 1988 and 1991 at ICARDA, Syria. Accessions of Cicer species were screened under natural insect infestation in the field during spring (March–June), along with a susceptible-cum-indicator line. Two accessions of C. cuneatum (ILWC-40 and -187) and 10 accessions of C. judaicum (ILWC-44, -46, -56, -57, -58, -95, -103, -196, -206, and -207) were rated 2 on a 1–9 scale, where 1 = free from any damage and 9 = maximum damage. Another 18 lines of C. judaicum, four lines of C. pinnatidum and one line of C. reticulatum were rated 3 (resistant). Genetic variability of wild populations, closely related to domesticated species, constitute important genetic resources for plant breeding programs. In this paper, we analysed the variation of eight wild populations of Solanum lycopersicum var. cerasiforme in a common garden experiment for levels of plant infestation by whitefly, leaf trichome density as a defensive character preventing infestation by whitefly, and the effect of whitefly incidence into vegetative and reproductive plant characters. Number of adults of whitefly was recorded in the eight wild populations of S. lycopersicum var. cerasiforme, one population of the wild species S. habrochaites (C-360), and one of a cultivated variety of S. lycopersicum (Rio Grande). There were significant differences among the wild populations of S. lycopersicum var. cerasiforme in the average level of whitefly incidence and trichome density. Cultivated tomatoes had the higher incidence of whiteflies ( = 7.50 ± 0.14) followed by plants of S. lycopersicum var. cerasiforme ( = 2.02 ± 0.92) and plants of S. habrochaites with the lowest incidence ( = 0.36 ± 0.35). Whitefly incidence was negatively correlated with trichome density (r = − 0.38, p < 0.0001), suggesting that trichomes deter or limit the establishment of whiteflies. Additionally, a significant negative effect of whitefly incidence along the growing season upon plant growth rate (number of branches and height) and fruit production was detected.  相似文献
