Post mortem storage is a necessary process for removal of pin bones without destruction of fillets, thereby avoiding volume and economic loss. However, the enzymes involved in loosening pin bones during storage have not been studied to a great extent. In this study, the activities and localization of MMPs in the connective tissue (CT) of pin bones dissected from fillet of salmon and cod were investigated. Interestingly, the enzyme activity profile in these two species was different during post mortem storage of fish fillets. Adding MMP inhibitor (GM6001) and serine protease inhibitor (Pefabloc) revealed different effects in the two species, suggesting different regulations in salmon and cod. In situ zymography with the same inhibitors verified MMP and serine protease activity in CT close to pin bone at early post mortem (6 h) in salmon. However, MMP inhibition was not evident in cod in this area at that time point. Immunohistochemistry further revealed MMP9 and MMP13 were located more to the outer rim of CT, facing the pin bone and adipose tissue, while MMP7 was more randomly distributed within CT in salmon. In contrast, all these three MMPs were randomly distributed in CT in cod. In summary, our study reveals different MMP enzyme profiles in salmon and cod in the pin bone area, influenced by serine proteases, and suggests that MMPs and serine proteases must be taken in consideration when studying the conditions for early pin bone removal.  相似文献

Trimethylamine oxide (TMAO) is found at high levels in wild Atlantic cod. Trimethylamine oxide is reduced to trimethylamine (TMA) during iced storage. In this study, the levels of TMAO and TMA in wild and farmed cod and the impact of storage, gender and season on these levels were investigated. Wild cod had higher levels of TMAO than farmed cod. Wild cod also had higher levels of TMA after 15 days of iced storage. Farmed cod were found to have levels of TMA‐N <5 mg/100 g after 15 days of iced storage, which is below the maximum permitted level for fresh fish. Therefore, TMA appears to be an unsuitable measure of freshness in farmed cod. Female farmed cod had higher levels of TMAO than male farmed cod; no such difference between genders was found in wild cod. Female and male farmed cod had similar levels of TMA after post‐mortem iced storage. The hepatosomatic index (HSI) of female farmed cod was higher than that seen in male cod and this may be the underlying reason for the differences in muscle TMAO. Levels of TMAO in male farmed cod were higher during the autumn and were correlated with a larger HSI during the same period.  相似文献

The effects of different egg white protein coatings on Atlantic salmon quality preservation after 4 months of frozen storage were evaluated. These coatings increased the yield of the fillets and protected them from lipid oxidation, providing better protection than glazing. Egg white protein coatings were better options than glazing to preserve salmon color during frozen storage. The best results were obtained with the application of coatings before freezing. No clear effects of the type and concentration of plasticizer in most quality parameters were found. Egg white protein + glycerol 2:1 applied before freezing was the best coating for frozen Atlantic salmon protection.  相似文献

Abstract

The shelf life of iced redfish is known to be 16–19 days. This study evaluated the effects of modified atmosphere (CO2/N2: 60/40) in bulk storage of redfish and subsequent modified atmosphere packaging (MAP) of fillets. Fish were evaluated by sensory, microbiological and chemical methods. Modified atmosphere (MA) bulk storage of whole fish for more than 5 to 10 days did not improve their sensory quality or significantly increase shelf life. In fact, the study showed negative effects in texture and overall appearance. MAP of fillets processed from 10-day MA bulk stored fish showed a modest increase in shelf life, but negatively affected their texture. Sensory qualities, i.e., tenderness, that decreased under MA bulk storage were improved upon aerobic storage for fish stored under MA for 5 to 10 days, but not 14 days. Lower microbial levels were found in MA bulk stored fish and MAP fillets compared to traditionally iced fish, while higher TMA levels were found in MAP fillets.  相似文献

Improved slaughtering procedures of farmed fish may provide sufficient time so that filleting can be performed pre‐rigor while the muscle pH is still high. Such filleting not only reduces fillet gaping but also lowers the transportation costs and makes fresh fillets available to the markets at an earlier stage. The aim of our work was to determine the weight reduction of the fillets due to liquid loss and to study the proteins and enzymes in the drip. After 11 days of cold storage, fillets of farmed Atlantic cod produced pre‐rigor had a weight loss of 10% and a fillet contraction of 19% while for fillets produced post‐rigor the values were 5% and 4% respectively. At the same time, approximately twice the amount of proteins had been lost from the pre‐rigor‐produced fillets. Sodium dodecyl sulphate polyacrylamide gel electrophoresis analysis showed that the proteins in the drip were similar to the sarcoplasmatic proteins extracted from the muscle. Specific analysis of proteolytic enzymes indicated that they are less stable in the expelled liquid than in the flesh during storage. The extensive loss of weight and proteins from pre‐rigor‐produced fillets during subsequent storage must be taken into account if such processing is considered for farmed cod.  相似文献

ABSTRACT

In order to evaluate the effect of chilled (T1), frozen (T2), and freeze-chilled (T3) treatment on the flavor of grass carp fillets and soups, adenosine triphosphate (ATP)-related compounds, umami-taste amino acids (UTAA), thiobarbituric acid (TBA), peroxide value (PV), and soup sensory score were analyzed. Results showed that the sum of ATP, ADP, and AMP contents in T2 fillets were significantly higher (p < 0.05) than others, but the sum of hypoxanthine riboside (HxR) and hypoxanthine (Hx) contents for T2 fillets and soups were significantly lower (p < 0.05) than others during storage. The T3 fillets and soups had lower inosine 5-monophosphate (IMP) contents than T1 after Day 2, but higher UTAA contents than T1 during storage. The T3 fillets had the highest TBA and PV values among the three groups during storage. According to the total aerobic counts, K value, and sensory score, all the fillets and soups kept good quality during storage. Therefore, freeze-chilled treatment not only can extend fish shelf life but also retain excellent fish flavor.  相似文献

ABSTRACT

Livers of Atlantic cod (Gadus morhua) are traditionally used in cod liver oil production or consumed cooked or canned. The farming of cod is a relatively new industry in Norway. The aim of this study was to determine quality and shelf life of fresh liver from farmed cod during chilled storage on ice by hydrolysis and oxidation state and sensory quality and the influence on canned liver. In two experiments, livers from farmed cod were stored chilled and sampled from Days 0 to 13, respectively. Quality, measured as hydrolytic and oxidation degradation, was reduced after 7 days of storage, while sensory quality was reduced after 4 days. Free fatty acids increased from Day 7 in both experiments, while peroxide value and anisidine value showed no change when the livers were single wrapped. Rancid odor was the first sign of oxidation and was registered after three to four days of storage. Canning within 2 days of storage prevented leakage of oil from the canned livers. Sensory analyses of oxidation are recommended as a sensitive and rapid method to detect oxidation of chilled cod liver.  相似文献

Abstract

Coho salmon (Oncorhynchus kisutch) fillets were processed using five different methods (smoking, canning, freezing, acidifying, and salting) to evaluate the effect of preservation choice on the quality of polyunsaturated fatty acids (PUFA). Salmon preserved by smoking, canning, or freezing retained higher values of total fatty acids, including n-3 PUFAs such as eicosapentaenoic acid and docosahexaenoic acid. Salting and acidifying (pickling) treatments resulted in a significant decrease in PUFAs. The results of this study are intended to provide direction for handling and storage of salmon to retain the maximum levels of high-value n-3 PUFAs.  相似文献

ABSTRACT

Soluble gas stabilization (SGS) is a relatively recent methodology of active packaging that has been proposed to extend the shelf life of packaged fish. The aim of the present work was to study the effect of SGS applied at different extents (2 bars: 30 and 60 min) on the shelf life of sea bream and sea bass fillets packed in air during chilled storage. Quality changes were evaluated by sensory assessment, microbiological analysis (TVC), TBARs, pH value, TVB-N, and TMA-N. No significant extension of sensory shelf life was visible as a function of SGS treatment. The treatment of fillets in CO2 at 2 bars during 60 min had a positive effect on the microbiological shelf life of both species. On the other hand, TMA-N and TVB-N showed similar changes during storage period and were not affected by the CO2 treatment. Sea bream treated with SGS always presented higher TBARs than control samples. Nevertheless, oxidation of sea bream and in particular, of sea bass fillets, did not appear to be a significant problem during chilled storage.  相似文献

The effect of gamma irradiation (0, 1.8, and 3.3 kGy) on the microbiological, chemical, and color characteristics of marinated (7% acetic acid and 10% NaCl) and vacuum-packed anchovy fillets was analyzed during 20 months of refrigerated storage (4 ± 1°C). Acidity, pH, water activity (aw), total volatile basic nitrogen (TVBN), lipid oxidation, and color parameters were determined. Mesophilic and psychrotrophic bacteria, sulphite-reducing clostridia, total and fecal coliforms, Staphylococcus spp., yeasts, and molds were investigated. Gamma irradiation reduced the initial mesophilic bacterial counts and inhibited mesophils growth during 20 months. As a result, the production of TVBN during storage was lower in irradiated samples than in control. Also, lipid oxidation was lower in irradiated samples than in nonirradiated. The color of anchovy fillets was not affected by the irradiation treatment. Even if nonirradiated anchovy fillets presented a high stability in comparison with the traditional product (in flasks with vegetable oil and spices), gamma irradiation improved the microbiological and chemical quality of anchovy fillet marinades without inducing changes on its characteristic color for 20 months.  相似文献

Escapes of cod (Gadus morhua) from sea cages represent an economic problem for farmers and a potential environmental problem. We estimate that 0–6% of cod held in sea‐cage farms in Norway were reported to have escaped each year from 2000 to 2005, which is a high proportion compared with salmon. We interviewed employees at 19 coastal sea‐cage cod farms in Norway to investigate both how and why cultured cod escape and to document cage handling and management strategies that were effective in minimizing escapes. Based on the interviews, we describe five working hypotheses that may explain why a greater proportion of cod than salmon escape: (1) cod are more willing to escape than salmon; (2) cod bite the net cage and create wear and tear; (3) net cages have insufficient technical standards for cod culture; (4) cod are placed in sea cages at considerably smaller sizes than salmon; and (5) cod are more popular feed for predators. Preliminary testing of the hypothesis that cod bite netting and create holes was done by placing pre‐damaged net panels with cut twines and control panels inside sea cages. Holes in the pre‐damaged net panels increased in size over a period of 3 months. The type of damage indicated that biting of netting twines was the likely cause.  相似文献

为研究臭氧减菌化处理罗非鱼片冰温贮藏过程中蛋白质生化特性变化规律,本研究对冰温贮藏过程中经臭氧处理罗非鱼片肌原纤维蛋白盐溶性、Ca2+-ATPase活性、表面疏水性、巯基及羰基含量变化进行了评价。结果显示:在冰温贮藏过程中,臭氧处理组和对照组罗非鱼片蛋白质均发生了不同程度的变性,表现为:随贮藏时间的延长,肌原纤维蛋白盐溶性、Ca2 -ATPase活性及巯基含量均呈下降趋势,而肌动球蛋白表面疏水性和羰基含量则呈上升趋势;冰温贮藏20 d后,臭氧处理组罗非鱼片肌原纤维蛋白盐溶性、Ca2 -ATPase活性和巯基含量分别下降了76.57%、89.76%和66.72%,而对照组则分别下降了69.05%、86.45%和62.29%;另一方面,臭氧处理组罗非鱼片肌动球蛋白表面疏水性和羰基含量分别上升了111.75%和76.46%,对照组则分别上升了104.77%和58.23%。在同一贮藏时间臭氧处理组罗非鱼片肌原纤维蛋白盐溶性、Ca2 -ATPase活性及巯基含量较对照组低,而肌动球蛋白表面疏水性和羰基含量较对照组高。研究表明:臭氧水的氧化性及处理过程中产生的ROS加速了冰温贮藏过程中罗非鱼肌肉蛋白质的变性。  相似文献

为了改善鱼肉的品质,文章通过测定样品的挥发性盐基氮(TVB—N)、Ca^2+-ATP酶活性以及鱼片色泽和质构指标随时间的变化情况分别评价了4种不同致死方式(I.鳃部放血;II.冰激致死;Ⅲ.敲击头部致死;Ⅳ.敲击头部后放血)对罗非鱼(Oreochromissp.)鱼肉品质的影响。结果显示,贮藏期间4个组的TVB—N显著增大,但Ⅱ组和Ⅲ组的TVB—N显著低于I组和Ⅳ组,有利于鱼片冰藏期的保鲜;4个组鱼片的Ca^2+-ATP酶活性显著下降,Ⅲ和IV2种宰杀方式有利于保持Ca^2+-ATP酶活性;4个组的亮度(L^*)、红绿色值(a^*)和黄蓝色值(b^*)都随着贮藏时间的延长而增加,I和Ⅳ2种宰杀方法使鱼片L^*较大而a^*较小,但4组之间b^*无显著差异;鱼片的硬度、咀嚼性和胶着性随贮藏时间的延长呈下降的趋势,其中Ⅲ组的质构品质保持得最好。综合以上所有指标,可以认为Ⅲ组最有利于罗非鱼片的保鲜,其次为Ⅱ组。  相似文献

High pressure (HP) treatment of 250 MPa pressure, 6-min holding time, and 400 MPa/min ramp rate at 25°C was applied to headless Indian white prawn (Fenneropenaeus indicus) to investigate the significant modifications on the functional properties of protein during chilled storage. Muscle fibers were shrunk and extracellular space apparently reduced after HP treatment. Myofibrillar proteins denatured and sarcoplasmic proteins aggregated and were found to be stable in HP-treated sample during chilled storage. Water-holding capacity, solubility, viscosity, and Ca2+ ATPase activity of protein were diminished, whereas foam expansion, foam volume stability, and turbidity of proteins improved with HP treatment and storage period (p < .05). Turbidity of the protein was inversely proportional to viscosity, solubility, and Ca2+ ATPase activity in HP-treated samples.  相似文献

Antioxidants have been widely used as additives to provide protection against oxidative degradation of foods by free radicals. The effect of thyme essence (Zataria multiflora Boiss) on the rancidity development in cobia (Rachycentron canadum) fillets during frozen storage was studied. Cobia fillets were treated with thyme essence (250 and 500 ppm) and then stored at??18°C for up to 6 months. Rancidity development was measured by several biochemical indices including free fatty acids (FFA), peroxide value (PV), thiobarbituric acid (TBA), and complemented by sensory analysis (flesh odor, consistency, and appearance). Also, pH and expressible moisture were measured during 6-month storage. Proximate composition was also determined in the first day. TBA, PV, and FFA levels increased in all treatments due to lipid oxidation. Thyme essence showed an antioxidative effect in cobia fillets during frozen storage as indicated by TBA, PV, and FFA levels. Results showed that FFA, primary and secondary oxidation products, expressible moisture (EM), and pH of thyme essence treated samples were significantly lower than those of the control samples (p < 0.05). Thyme essence retarded oxidative changes in frozen cobia fillets, and the best oxidation inhibition was obtained using thyme essence at 500 ppm.  相似文献

The biogenic amine contents in fillets of three freshwater fish of kutum (Rutilus frisii), common carp (Cyprinus carpio), and Caspian salmon (Salmon caspius) were monitored during storage at 4°C for 20 days. The amounts of biogenic amines were determined at regular intervals using high performance liquid chromatography after derivatization with benzoyl chloride. Significant differences in the levels of selected biogenic amines among the fish samples were found (p < 0.05) during storage times. While the amount of putrescine, cadaverine, histamine, and tyramine in the fish fillets increased during the storage time, the changes in spermine and spermidine contents of the fish species were minor and both showed low and inverse correlations with storage time. For each of the three fish species, the sensory acceptability limit was found to be 4–8 days. The quality and biogenic amine indices of fish samples showed high correlation with storage time (r > 0.9). The principal component analysis of the results indicated that the formation of biogenic amines displays a certain behavioral pattern in all fish samples.  相似文献

ABSTRACT

This study assessed effects of high pressure processing on physicochemical quality and endogenous enzyme activities of grass carp fillets stored at 4 ºC for 21-day storage. Fresh fillets were vacuum-packed and subjected to high pressure processing (0.1, 200, 300, 400, 500, and 600 MPa at room temperature for 15 min). Results showed that high pressure processing significantly delayed microbial growth, and reduced total volatile basic nitrogen (TVB-N) and thiobarbituric acid reactive substances (TBARS) levels of grass carp fillets, especially for the 600 MPa group. On the 21st day, the corresponding increases of 6%, 35%, 45%, 43%, and 53% in hardness were observed for 200 MPa, 300 MPa, 400 MPa, 500 MPa, and 600 MPa samples compared with control samples, respectively. In addition, enzyme activities in different groups decreased during storage. Among them, calpain, and myofibril-bound serine proteinases (MBSP) activities were immediately reduced after high pressure treatments, while the activities of collagenase, cathepsin B, and cathepsin D were significantly (< 0.05) inhibited when the pressure exceeded 400 MPa. However, the pressure treatments activated cathepsin L initially, and then the activity gradually decreased in treated samples. Generally, high pressure processing reduced activities of most of the endogenous enzymes and improved the quality and extended the shelf life by at least 4 ~ 6 days of refrigerated grass carp fillets.  相似文献

Abstract

The effects of light salting by brine injection and brine immersion on physicochemical and textural properties of cod fillets were evaluated. Light salting significantly increased yield and water holding capacity. Adding brine injection to the process can be used to shorten the process time needed to obtain the desired salt content and increase yield. The effects on chemical composition were most significant in salt content, but water and protein content were also affected. Water content decreased from the tail to head direction of the fillets, both in unsalted and light salted fillets. Changes in protein content were in opposite proportion to changes in water content. This could be seen in the fresh fish and also after freezing, both in unsalted and light salted fillets. Texture of the fish was not significantly affected by salting and freezing.  相似文献
