ABSTRACT

In temperate regions, post-larvae freshwater prawn, Macrobrachium rosenbergii, are grown to more advanced sizes in tanks prior to pond stocking. This intermediate stage of culture is referred to as the nursery period. Little research has been conducted on different management practices on juvenile prawn growth and survival during this 30-60 day period. Survival during the nursery stage has been highly variable and may be related to the cannibalistic behavior of juvenile freshwater prawn when cultured at high densities in the nursery. The objective of this study was to evaluate the effect of stocking density, relative to the provision of artificial substrate (number of prawns/m2 of substrate), on growth, survival, and economic variables for freshwater prawn juveniles during nursery production. Post-larvae (0.01%0.00 g, n = 300) were stocked into nine 1900 L tanks, each provided with 20.5 m2 of artificial substrate in the form of horizontal layers of black plastic mesh (10 mm) spaced 5 cm apart. Tanks were randomly assigned one of three prawn densities (215, 430, or 860 post-larvae/m2 of substrate), which equated to 2.3,4.6 and 9.2 prawn/L, respectively. Juvenile prawn were fed a commercial trout diet (42% protein) at a percentage of body weight according to a feed rate table. Water quality was maintained using a flow rate of 8 L/min in each tank from a reservoir pond. Temperature was maintained at approximately 28°C using heat pumps. After 56 days there was no significant difference (P >0.05) in average weight of juvenile prawn stocked at the three densities (0 = 0.58%0.12 g, n = 9). Survival was significantly lower (P <0.05) for prawn stocked at 860 m2 (62%) than in those stocked at 430/m2 (78%) and 215/m2 (94%), which were not statistically different (P >0.05). Even with reduced survival, the highest stocking density produced the greatest number of nursed juveniles based on both tank volume (5.5/l) and surface area (530/m2), at the lowest average cost.  相似文献

ABSTRACT

Dopamine at concentrations of 0.15, 1.5, and 15.0 mg/kg was included in two different formulated diets that were fed to recently metamorphosed postlarvae of the freshwater prawn, Macrobrachium rosenbergii, for 60 days. Dopamine, in the form of dopamine hydrochlo-ride, was incorporated into the formulated diets by either solubilizing it in alcohol, followed by submersion of diet and evaporation, or inclusion in a lipid premix ingredient. The alcohol evaporation method at 0.15, 1.5, and 15.0 mg/kg significantly increased the percentage of individuals without external male characteristics. This research is the first reported use of dietary administration of dopamine to crustaceans, and the first documentation of the apparent dietary control of gonadal development and sex differentiation in crustaceans. Replication of results, a histological examination of a large sample of individuals, and continued monitoring of those individuals fed the dietary dopamine are needed to verify the apparent effect.  相似文献

Abstract

Prawns (Macrobrachium rosenbergii) were subjected to conditions, which simulated domestic and export handling practices. Prawns were mixed with crushed ice to maintain the prawn samples temperature at 0°C and were stored at room temperature (25 ±2°C) for 48 hr for the domestic handling trial, while for export handling, the samples were kept in a cold room at (0 ±1°C) for 72 hr. Both processes showed sensory, chemical and instrumental changes but the magnitude of the changes for domestic handling was greater than for those of the export trial. Maximum load (firmness), energy to the breaking point (chewiness) and percent of cooking loss increased as storage time increased up to 12 hr, after which prolonged storage resulted in a decrease in these parameters in the cooked samples. Expressible juice decreased during the first 12 hr, later increased gradually. Other chemical parameters such as the soluble-insoluble collagen ratio, proteolysis index (PI) and total volatile nitrogen (TVN) of cooked prawn increased with the increase in storage time. Good correlations among instrumental, chemical and sensory evaluation results were obtained for texture and taste-odor attributes of samples from both of the handling trials. Blanching and deheading of the prawns were found to maintain qualities very well, while samples, which underwent proper handling procedures, exhibited some quality changes after 48 hr.  相似文献

Abstract

After harvest, adult prawn are often transported for sales to live markets, such as ethnic Asian outlets, in major urban centers. Poor survival during transportation has hindered development and expansion of these markets. Methodologies to increase survival during transport could contribute to industry viability. Three independent trials were conducted. In the first trial, three biomass densities (25, 50 and 100 g/L) were evaluated in 100-L, open plastic containers aerated with pure oxygen and compressed air. Water quality analyses were performed prior to stocking. After 24 hours in the model transport containers, water quality analysis was conducted and all prawn were removed, determined to be alive or dead, and each group weighed and counted. There was no significant difference (P > 0.05) for prawn survival (overall mean 98%) among the three densities. Concentrations of dissolved oxygen and nitrite were not effected by hauling density (P < 0.05). Total ammonia-nitrogen and un-ionized ammonia-nitrogen concentrations increased (P < 0.05) as biomass densities increased, though values remained within what are considered tolerable ranges. In the second trial, the effect of added substrate and temperature on transport survival was evaluated with prawns stocked at the high density (100 g/L). Two water temperatures (21°C and 26°C) with and without substrate were evaluated in a 2 × 2 factorial with three replicate, 100-L model transport containers per treatment combination (12 containers total). Factorial analysis indicated no significant statistical interaction (P > 0.05) between the presence of substrate and water temperature on any measured variable. The main effects of substrate and temperature were then analyzed separately. The presence or absence of substrate had no significant impact (P > 0.05) on prawn survival. However, temperature had a highly significantly impact (P < 0.01) on survival; prawn survival at 21°C averaged 97% compared to 24% at 26°C. The third trial was a commercial verification trial in which 500 kg of live prawn were transported to New York from Kentucky. These data indicate prawn can be successfully transported at 100 g/L for 24 hours when temperatures are maintained near 21°C. Adding substrate to the transport tank appears to provide no benefit.  相似文献

ABSTRACT

Recent developments in prawn production technologies (i.e., added substrate, increased stocking densities, size grading, and increased feed rates) have increased production rates from 900-1,000 kg/ha to over 2,500 kg/ha. While prawn can receive substantial nutritional benefit from natural foods at the lower biomass densities, at higher production rates prawn are likely to be more dependent on prepared diets. To ensure that maximum production is being achieved by these new production technologies, production rates must not be nutritionally constrained. This study was conducted to compare the current recommended technology of phase feeding of different quality feedstuffs to prawns of different sizes with the feeding of a high-quality penaeid diet throughout the production season. Two treatments were evaluated: Treatment 1 was phase feeding (current technology-control) where prawns were fed unpelleted distillers' grains with solubles (DDGS) for the first four weeks, then a 28%-protein prawn diet for weeks 5-12, and finally a 40%-protein penaeid diet for weeks 13-16. In Treatment 2, prawn were fed the 40%-protein penaeid diet throughout the entire production period. Feeding rates in both treatments were based upon a feeding table. Three 0.04 ha ponds were used for each treatment. All ponds were stocked at 59,280 juveniles/ha and were provided with artificial substrate in the form of a polyethylene “safety fence” oriented vertically to increase available surface area by 50%. After 106 culture days, no significant differences (P >0.05) were observed between treatments in terms of yield, average individual weight, food conversion ratio (FCR), or survival, which averaged 2,575 kg/ha, 46 g, 2.3, and 94%, respectively, overall. Due to the higher cost of the penaeid diet (US$0.84/kg), feeding costs for the penaeid diet treatment (Treatment 2) were 38% higher than those for Treatment 1. No benefit to using higher protein diets during the first 12 weeks of prawn pond production was observed.  相似文献

ABSTRACT

The freshwater prawn, Macrobrachium rosenbergii, commonly called ‘scampi,’ is the most important culturable freshwater species of prawn with a fairly high growth rate, wide range of temperature (15-35°C) and salinity tolerance. The nutrients required by this species for growth and associated physiological functions are similar to those required by other crustaceans. Current data suggest that a digestible protein level of above 30 is required for maximum growth and protein efficiency. Ingredients such as mussel meat meal, squid meal, and shrimp meal serve as potential sources of protein in formulated diets. The complete quantitative requirements for all the essential amino acids for this prawn species have not yet been worked out. With respect to non-protein energy supply in the form of carbohydrate, studies indicated that scampi is capable of utilizing various carbohydrate sources efficiently. Dietary carbohydrate in the form of complex polysaccharides appears to be more effective as an energy source. Like other crustaceans, scampi also has limited ability to synthesize sterol from acetate and mevalonic acid. Quantitative estimates of the essential fatty acid requirements have been made in post-larval scampi. Data on vitamin and mineral requirements are scarce. Recent developments in diet formulation, practical feeding practices and the aquaculture potential in India have been indicated with emphasis on sustainable production.  相似文献

利用对虾育苗场的设计进行罗氏沼虾工厂化育苗试验,于136m^3水体中投放状幼体1438万尾,育出商品仔虾964万尾,幼体培育成活率为67%,平均每尾亲虾出苗4725尾,单位水体平均出苗量为7.09万尾,最高为10.2万尾/m^3。从4月上旬到5月下旬,罗氏沼虾雌性亲虾可抑卵三次以上,前后抱卵批次所孵出的幼体质量没有明显差异。试验还证明,幼体培育池内氨氮浓度高达1056.4μg/l对成活率没有多大曩  相似文献

采用生物化学方法分别测定了罗氏沼虾胚胎发育期5种消化酶:胃蛋白酶、类胰蛋白酶、淀粉酶、纤维素酶和脂肪酶的比活力,测定了6种同工酶:酯酶(EST)、苹果酸脱氢酶(MDH)、谷氨酸脱氢酶(GDH)、碱性磷酸酶(ALP)、乳酸脱氢酶(LDH)和淀粉酶(AMY)在成熟卵细胞和胚胎发育时期的表达特征。结果显示:5种消化酶的活力表现出两种变化趋势,即胃蛋白酶、类胰蛋白酶、淀粉酶和纤维素酶的酶活力呈现出“高—低—高”的变化趋势,而脂肪酶活力则呈现出下降的变化趋势。6种同工酶酶谱则随胚胎发育渐趋复杂,酶活性随之增强。研究表明,所测定的消化酶以及同工酶在罗氏沼虾受精卵期均已表现出酶活性,此阶段为卵源性的酶;原肠期是胚胎发育中一个较为重要的时期,胚胎内酶基因表达能力开始逐渐增强,开始合成一些酶类,为胚胎发育后期利用卵黄物质提供了保证。消化酶以及同工酶对卵黄物质的利用和胚胎发育的物质代谢起重要作用。  相似文献

通过非离子氨氮对罗氏沼虾幼体的急性致毒试验表明:Ⅴ期幼体96h的半致死浓度(LC50)值为1.14mg/L,安全浓度(SC)值为0.11mg/L;Ⅸ期幼体96h的LC50值为1.40mg/L,SC值为0.14mg/L,建议在育苗期间加强水质管理,有效地控制水体中非离子氨氮的浓度。  相似文献

为探索罗氏沼虾(Macrobrachium rosenbergii)铁虾综合征(IPS)的分子机制,采用高通量测序平台(Illumina Hiseq-2500)分别对患IPS罗氏沼虾(IPS虾)和正常罗氏沼虾开展转录组测序,进行生物信息学分析。结果显示,高通量测序共获得56.42 G高质量数据,拼接后得到221 901条单基因序列(unigene),长度范围为201~30 985 bp,平均长度为1572 bp,N50长度为2867 bp,N90长度为646 bp。将单基因序列分别在Nr、Nt、Swissprot、KEGG、KOG、GO、PFAM数据库进行序列比对及功能注释,103 570条得到注释,其中,GO数据库注释到的单基因序列最多。差异表达分析显示,2003个基因在IPS虾眼柄中差异表达,包括1209个上调基因和794个下调基因,516个基因被注释到242条KEGG通路中,翻译、信号转导和免疫系统富集的差异基因数目最多。催乳素、雌激素、胰岛素、促性腺激素释放激素、胰高血糖素、催产素、谷氨酸能突触、血清素能突触等与生殖调控相关激素的代谢过程在IPS虾与正常虾眼柄之间存在差异。此外,一些已被证明在免疫反应中起重要作用的基因在IPS虾眼柄中显著上调,如血管内皮生长因子受体1、丝氨酸蛋白酶抑制剂6、C型凝集素、芳基硫酸酯酶B、酚氧化酶原激活酶2a、组织蛋白酶B、组织蛋白酶L、甲壳类抗菌肽4等。同时,注释到溶酶体、吞噬体、抗原处理与呈递、细胞凋亡、内吞作用等多条与免疫相关的途径,支持近期研究得出的罗氏沼虾IPS与病原感染相关的结论。本研究为解析罗氏沼虾IPS的成因和分子机制提供了数据支撑。  相似文献

Abstract

The effect of nitrate concentration on giant river prawn, Macrobrachium rosenbergii, larvae was investigated. Survival rate, weight gain, and larval development were evaluated for different concentrations of nitrate in three experiments. The experiments were divided in to two phases. In the first phase, larvae from stages I through VIII were analysed, while in the second phase larvae from stage VIII through post-larvae metamorphosis were analysed. Oxygen consumption was also determined for zoea I, II, and VIII exposed to 0, 700, and 1,000 mg/L of nitrate-N. No effect was observed for concentrations up to 180 mg/L NO3-N (experiments I and II), and nitrate levels as 1,000 mg/L NO3-N did not affect survival in the first phase of the third experiment. On the other hand, larval stage index (LSI) and weight gain decreased as nitrate-N concentration increased from 0 to 1,000 mg/L. In the second phase, survival and metamorphosis rate decreased as nitrate concentration increased, according to a linear model. The effect of nitrate levels on weight gain followed a curvilinear pattern. Larval respiration decreased in the water where nitrate was added, but only during stage II. The results demonstrated that nitrate presents extremely low toxicity for giant river prawn larvae, and data were related to the levels of nitrate that usually occur in larviculture systems also discussed. Therefore, nitrate is not a limiting factor for giant river prawn larviculture.  相似文献

从濒死的罗氏沼虾体内分离到菌株M rM0602,经形态学观察及API 20E鉴定系统鉴定为嗜水气单胞菌。人工感染健康罗氏沼虾,100%致死剂量为1.0×104cfu/m l,半致死剂量(LD50)为3.16×103cfu/m l。测定菌株M rM0602的胞外产物对小鼠和新西兰大白兔红细胞的溶血活性,表明胞外产物对小鼠红细胞有微弱的溶血活性,对新西兰兔红细胞无溶血活性。胞外蛋白酶活力测定显示,菌株M rM0602胞外产物具有很高的蛋白酶活力,且耐热性很强,65℃仍保持较高活性。常规药物敏感性试验表明,菌株M rM0602对环丙沙星和恩诺沙星高度敏感。  相似文献

Long‐term cryopreservation of the giant freshwater prawn, Macrobrachium rosenbergii, spermatophores using glycerol (Gly) and ethylene glycol (EG) as cryoprotective agents (CPAs) was studied. The tolerance of sperm to cryopreservation was evaluated on the basis of sperm survival and fertilizing ability. The survival of the sperm was determined by trypan blue staining, while the fertilizing ability was assessed from artificial insemination of the cryopreserved spermatophores. The rates of embryo survival on day 5 after spawning and of spermatophores capable of producing embryos survived to hatching were determined. Storage of spermatophores at ?20°C without CPA for a short period of up of 1–5 days decreased the sperm survival significantly and did not preserve fertilizing ability. Preservation at ?20°C in the presence of 10% or 20% Gly or of 10% or 20% EG offered a simple and efficient short‐term storage up to 10 days. For a long‐term storage, cryopreservation in the presence of 20% EG at ?196°C was more efficient than at ?20°C. High sperm survival rates and high fertilizing ability were recorded from those cryopreserved at ?196°C for up to 150 days. High sperm survival rates with moderate levels of fertilizing ability were obtained from those cryopreserved at ?20°C for not more than 30 days. The results indicate that preservation at ?196°C with 20% EG is a suitable procedure for long‐term storage of the giant freshwater prawn spermatophores.  相似文献

The giant freshwater prawn, Macrobrachium rosenbergii, is a species with a high commercial value in aquaculture. Two experiments were performed to determine the effects of salinities on the osmoregulation, growth and molting cycles of M. rosenbergii during growout. The first experiment was designed to determine whether these animals are capable of adapting to the changes in salinity seen in salinity intrusions in tropical deltas, with an incremental increase in salinity of 3‰ per day from 0‰ to 30‰ Haemolymph osmolality was rapidly regulated up to salinities of 15‰ , whereas animals conformed at higher salinities. The second experiment determined the growth, moulting cycle, osmolality, muscle water content and mortality during a 4‐month experiment at 0‰, 15‰ or 25‰ salinity. The weight gains in 0‰ and 15‰ were not significantly different and were comparable to the growth rates achieved in production farms with body mass increases of 2.6 and 2.3‐fold their initial body mass, respectively, after 4 months. The 25‰ group suffered from low growth, high mortality and a significantly lower moulting frequency. These data show that this species can be reared in brackish water up to 15‰, allowing for farming in the large areas impacted by salt water intrusions in tropical deltas.  相似文献

甲壳动物十足目的大多数种类,其受精卵自产出后就附着于亲体腹部,并完成胚胎发育过程直至孵化,这对于胚胎的发育、保护和幼体散布有重要的生物学意义。胚胎的附着机制较为复杂,与刚毛、卵柄、卵索、卵膜和黏液腺有关:卵柄和卵索的形成决定了胚胎发育的顺利进行;卵膜的变化影响着胚胎的附着;黏液腺在胚胎附着过程中形成卵柄和外层卵膜,胚胎是依靠卵柄或卵索附着于腹肢的刚毛上,进而完成胚胎发育过程。本研究采用常规组织学和扫描电镜的方法研究了罗氏沼虾(Macrobrachium rosenbergii)胚胎及其附着结构。结果表明,胚胎附着与母体腹肢的结构、黏液腺分泌的黏液相关,也与胚胎表面结构、次级卵膜的形态和结构密切相关。原肢底节和内肢刚毛密而长,基部有细小的分支,有利于胚胎的附着,外肢刚毛羽状则有利于胚胎的托附和保护。抱卵雌虾腹肢的原肢和内肢上布满腹肢黏液腺分泌的黏液;携卵刚毛中空、壁厚,刚毛外壁由微绒毛状结构组成,扩大了携卵刚毛的表面积,以利胚胎附着。胚胎外表面由初级卵膜和次级卵膜组成,次级卵膜主要由卵巢黏液腺分泌的黏液参与形成。排卵时,受精卵依靠坚韧的次级卵膜相互粘连,同时,部分受精卵在刚毛上滑动、旋转,并借助次级卵膜逐渐形成了卵柄或卵索固着在腹肢刚毛上。从受精卵到胚胎孵化,胚胎之间最主要的连接方式是依靠次级卵膜的连接。胚胎的表面有许多褶皱和黏液,利于胚胎的附着以及胚胎对溶氧和小分子物质等的通透。以上结构与附着相适应,保证了胚胎发育的顺利进行和幼体的正常孵化。本研究旨为进一步认识甲壳动物胚胎附着机制、胚胎发育和人工育苗的研究提供参考。[中国水产科学,2007,14(1):67-73]  相似文献

罗氏沼虾肌肉白浊病(whitish muscle disease of Macrobrachium rosenbergii)是一种发生在罗氏沼虾苗种阶段的流行病,发病虾苗出现肌肉白浊、白斑或白尾症状,死亡率高达60%以上,作者所在实验室在确定其病原是罗氏沼虾诺达病毒(Macrobrachiium rosenbergii Nodavirus,MrNV)的基础上,用MrNV免疫BALB/c小鼠,取小鼠脾细胞与SP2/0鼠骨髓瘤细胞融合,用间接ELISA筛选阳性孔,经有限稀释法克隆,得到12株能特异分泌抗MrNV的单兜隆抗体(Mab)的杂交瘤细胞。注射小鼠,制备腹水单克隆抗体,ELISA效价为1:10^5~10^6。亚型鉴定结果表明,有6株单抗为IgG1型,4株甲抗为IgG2a型,2株单抗为IgG2b型。12株单抗与对虾白斑综合征病毒(WSSV)和桃拉综合征病毒(TSV)均无交叉反应:挑选效价高的腹水单抗2B5,建立了检测罗氏沼虾诺达病毒的三抗体夹心酶联免疫吸附测定(TAS—ELISA)法,该方法检测灵敏度达0.98ng左右。Wesrtem blot分析表明,2B5能与MrNV 43kD的外膜蛋白特异性结合。  相似文献

Freshwater prawn (Macrobrachium rosenbergii) culture in the Western Hemisphere is primarily, if not entirely, derived from 36 individual prawns originally introduced to Hawaii from Malaysia in 1965 and 1966. Little information is available regarding genetic variation within and among cultured prawn stocks worldwide. The goal of the current study was to characterize genetic diversity in various prawn populations with emphasis on those cultured in North America. Five microsatellite loci were screened to estimate genetic diversity in two wild (Myanmar and India‐wild) and seven cultured (Hawaii‐1, Hawaii‐2, India‐cultured, Israel, Kentucky, Mississippi and Texas) populations. Average allelic richness ranged from 3.96 (Israel) to 20.45 (Myanmar). Average expected heterozygosity ranged from 0.580 (Israel) to 0.935 (Myanmar). Many of the cultured populations exhibited reduced genetic diversity when compared with the Myanmar and the India‐cultured populations. Significant deficiency in heterozygotes was detected in the India‐cultured, Mississippi and Kentucky populations (overall Fis estimated of 0.053, 0.067 and 0.108 respectively) reflecting moderate levels of inbreeding. Overall estimate of fixation index (Fst = 0.1569) revealed moderately high levels of differentiation among the populations. Outcome of this study provide a baseline assessment of genetic diversity in some available strains that will be useful for the development of breeding programmes.  相似文献
