Summary Based on maximization of selection response, Peek (1986) proposed methodology for determining the optimum number of crosses and progeny per cross in breeding self-fertilizing crops. By this method the total selection response is obtained by adding the individual responses from the two steps of selection: selection between crosses and selection within crosses. In this paper, Peek's approach has been generalized for finite or nonnormal populations. Optimum numbers of crosses are determined by testing capacity, number of selected crosses, number of selected lines from each selected cross, and heritability.In the case selection of the best line from the best cross, the optimum number of crosses increases monotonically with increasing testing capacity (for any given fixed heritability). For increasing heritabilities, however, the optimum number of crosses exhibits relatively flat maxima (for any given fixed testing capacity). These maxima are located at intermediate or sub-intermediate heritabilities.For varying numbers of selected crosses and selected lines from each selected cross the main results are: The optimum number of crosses i) increases with increasing testing capacity, ii) increases with increasing number of selected crosses and iii) decreases with increasing number of selected lines from each selected cross.  相似文献

Summary The response to selection is determined by the genetic and nongenetic variance, the selection intensity, and the size of the experiments. If, in pure line breeding, selection starts in segregating generations, the gain depends on the relation of additive to epistatic variance. The gain can be increased if an index is constructed using the information from relatives. From animal breeding this kind of selection is known as combined selection. In this paper the optimal number of families and subfamilies is determined that will maximise the selection response by combined selection for a fixed total size of experiments. The composition of the genetic variance has a imited influence on the optimal size of the progenies. If epistatic variance is important, then the number of F2 families has to be reduced and the number of F3 families and F4 families must be increased. The same is true if the nongenetic variance is increased.  相似文献

Crosses of plants from the same I0.1 line can be used for studying linkage relations between incompatibility loci and marker genes. Only one of the two incompatibility loci of rye has so far been localized, on chromosome 1R. In order to locate the second locus, crosses were performed with plants possessing known isozyme markers of the rye genome. Linkage was found with the β-glucosidase locus on chromosome 2R. By combined analysis a recombination value of C.144 was determined.  相似文献

本文以高油品种哈交97-5404-1为母本,以哈交99-5448-4为父本,建立重组自交系群体.应用SSR技术,对不同世代F2∶3,F2∶6、F2∶9遗传群体中的油份QTL定位进行了分析,不同世代大豆分离遗传群体中油份含量均接近于正态分布,油份含量性状表达偏向于母本哈交97-5404-1,F2∶3代获得了对油份贡献率较高的QTL,F2∶9代则获多个与油份相关的QTL,大豆F2∶3和F2∶9代的遗传群体适宜用做品质性状的QTL定位,不同世代定位的油份QTL,均与SSR位点Satt193有关,通过对来自全国不同品种的SSR进行了分析和方差分析证实,Satt193在第一等位变异下(即DNA片段长度为270 bp)做为油份的筛选标记具有实用性,而在第三等位变异下(即DNA片段长度为220 bp)做为蛋白质材料的筛选标记具有应用性.  相似文献

Summary The number and size of crosses in a breeding programme were discussed to the conclusion that the number of crosses rather than the size of a cross should be increased with a given total population size. The advantage due to this manipulation, however, is relatively small when such genotypes as improved for many (twenty-five or more) loci are to be obtained, or when a high proportion (around 0.1 or more) of available combinations have the potentiality of releasing the objective genotypes. In such a case, the ease with which crossing and cultivation can be carried out becomes the decisive factor.  相似文献

Summary Genetics of parthenocarpy in line RP 75/59 was tested under natural low temperature conditions, under which only seedless fruits were produced. Results were consistent with the hypothesis that three recessive genes with additive effects are responsible for parthenocarpy. Linkage studies, using 40 morphological marker genes located among all tomato chromosomes except chromosome seven, revealed linkage of one gene to diageotropica (dgt) located on chromosome 1 L site 152, and a second gene to yellow verescent (yv) located on chromosome 6 L site 34. Location of the third gene involved in parthenocarpy could not be determined. To calculate the power of the linkage tests, a simulation was carried out for three genetic models; the results are presented graphically in two figures.  相似文献

The interspecific genetic introgression approach has been shown to facilitate the detection and dissection of quantitative trait loci (QTL). A population consisting of 121 F6 recombinant inbred lines was developed by crossing Gossypium hirsutum cv. ‘Handan 208’ and G. barbadense cv. ‘Pima 90’ via single‐seed descent. Genotyping indicated that the mean ‘Pima 90’ allele frequency at each locus was 21%. Phenotyping and phenotypic distribution indicated a trend of return of individual lines’ characters to ‘Handan 208’ coupled with a wide variance for each trait. Significant loci influencing fibre quality were detected by one‐way analysis of variance (anova; P < 0.005) and association analysis [?log10(P) ≥ 3]: five and three markers for fibre length, four and one marker(s) for uniformity, two and one marker(s) for micronaire, 13 and 15 markers for strength, six and 10 markers for elongation, respectively. Two‐way anova based on genotypes of all marker loci combination showed that 807 two‐locus combinations were significant, and two‐way anova based on marker genotypes of QTL markers combination showed five significant digenic interactions (P < 0.01).  相似文献

Pre-harvest sprouting resistance in white-seeded wheat, Triticum aestivum L. is a genetically complex trait that varies with environmental conditions. Such variation causes difficulty in phenotypic characterization of populations to study inheritance or develop suitable DNA markers. To minimize random environmental effects, we evaluated controlled environments to measure dormancy. A population of 380 doubled haploid lines, AC Karma/SC8021V2, was evaluated in the glasshouse where the developing grains would not be exposed to moisture and greater consistency in temperature could be achieved. AC Karma is sprouting susceptible and SC8021-V2 is sprouting resistant. The population plus eight checks were seeded in early spring so the plants would reach physiological maturity under long days, requiring less supplemental light, and when the external temperature would be low enough that it would not cause difficulty in cooling the glasshouse. An alpha-lattice in a randomized complete block design with three replications was used. The blocks were arranged to minimize the environmental sources of variability in the glasshouse within each block. A sub-set of this population was tested in six field environments. Dormancy was characterized by germination of seed harvested near physiological maturity, from which a germination resistance index was calculated. The dormancy expressed in the glasshouse was significantly correlated with five of six field environments and highly significant in two of these. There was significant bidirectional transgressive segregation in both field and glasshouse environments. We are currently repeating this glasshouse experiment to confirm the results.  相似文献

Seed dormancy in the grain legumes may interfere with germination in some situations, yet is not well described. Dormancy in faba bean (Vicia faba L.) was investigated to determine the mode of inheritance and whether the gene or genes controlling this trait are linked to other genes. Seeds produced by 235 recombinant inbred lines at the F6 generation were used for this study. The parents were the cultivar ‘Optica’ and 172, an Afghanistan landrace which possesses extended seed dormancy. Germination was scored on a seed sample from each recombinant inbred line. Segregation patterns indicated that the trait is monogenic. This gene, named doz, is linked to a gene controlling anthocyanin and proanthocyanidin synthesis, sp-b, with the genetic distance estimated to be about 25 cM. No associations were detected between seed dormancy and pod dehiscence, seed lustre or mean seed weight. The implications of the persistence of a gene for seed dormancy in the gene pool of V. faba for the introgression of traits from landraces and cultivars possessing dormancy and for the origins of domesticated V. faba are discussed.  相似文献

Summary This short paper discusses the optimum number of crosses and size of progenies for a given total population size to minimize the risk of not finding any favourable genotype in the breeding material. It is shown that the number of crosses must generally be as large as possible. 本研究利用L16(54)正交试验设计研究了影响冬瓜SSR-PCR的五个因子(10×Buffer(含Mg2+),d NTP,DNA,引物,r Taq),研究结果表明Buffer(含Mg2+)和r Taq对PCR影响最明显,最终筛选出最佳反应体系:10×Buffer 2.0μL(1×Buffer),d NTP 2.0μL(200μmol/μL),DNA 0.6μL(6 ng/μL),引物0.4μL(0.2μmol/L)。体系体积为20μL,循环数为35。引物筛选程序为:94℃预变性3 min,94℃变性45 s,58℃~68℃复性30 s,5个循环,每个循环退火温度降2℃,72℃延伸1 min,94℃变性30 s,50℃~58℃复性1 min,8个循环,每个循环退火温度降低1℃,72℃延伸1 min,94℃变性30 s,50℃复性30 s,72℃延伸1 min,共30个循环,72℃延伸7 min。本研究建立了适合冬瓜的SSR反应体系和引物多态性筛选程序。  相似文献

Several species of wheat stem sawflies (WSS) are pests of wheat, including Cephus cinctus Norton in North America. Larvae feed inside the stems and cut the stem near plant maturity. The primary means of control is resistance due to solid stems, largely controlled by a locus on chromosome 3B (Qss.msub‐3BL). Cultivars that differed for WSS resistance, but with similar stem solidness, were crossed to determine the genetic basis for the differences. The cultivar ‘Scholar’ is susceptible, while ‘Conan’ shows resistance. Scholar and Conan possessed different alleles at Qss.msub‐3BL. Both alleles conferred solidness, yet the Conan allele conferred higher WSS resistance. An allele from Conan on chromosome 4A also decreased infestation and stem cutting. The 3B and 4A alleles from Conan acted in an additive fashion to provide increased WSS resistance without increasing stem solidness. Stem solidness has long been used by breeders as a proxy for WSS resistance because of its simplicity. Our results suggest that other resistance mechanisms may complement solid stems.  相似文献

应用L16(44)正交设计对影响鸭茅SSR-PCR的主要参数进行优化,建立适于鸭茅的SSR反应体系和扩增程序.在15μL体系中各反应的最适合含量为:50ng模板DNA,240μmol/L dNTP,0.4μmol/L SSR引物,1.0 U Taq DNA聚合酶,1.5μL 10×PCR Buffer,2.5mmol/L MgC12.PCR适宜扩增程序为:94℃预变性4min,94℃变性30 s,52℃复性30 s,72℃延伸1 min,共35个循环,72℃延伸10 min,4℃保存.并对引物最适合退火温度进行优化,最终确定引物退火温度为48~52℃.同时选用100对鸭茅引物对4份材料进行扩增,筛选出条带清晰,多态性好的引物30对.用于鸭茅SSR标记的进一步研究.  相似文献

The study on the genetic basis of heterosis has received significant attention in recent years. In this study, using a set of introgression lines (ILs) and corresponding testcross F1 populations, we investigated heterotic loci (HL) associated with six yield-related traits in both Oryza sativa L. subsp. indica and japonica. A total of 41 HL were detected on the basis of mid-parent heterosis values with single-point analysis. The F1 test-cross population showed superiority in most yield-related traits and was characterized by a high frequency of overdominant HL. Thirty-eight of the 41 HL were overdominant, and in the absence of epistasis, three HL were dominant, suggesting that heterotic effects at the single-locus level mainly appeared to be overdominant in rice. Twenty-four HL had a real positive effect, suggesting that they are viable candidates for the improvement of rice yield potential. Compared with the quantitative trait loci (QTLs) detected in the ILs, only six out of the 41 (14.6%) HL were detected in QTL analysis under the same statistical threshold, indicating that heterosis and trait performance may be conditioned by different sets of loci.  相似文献

The stem solidness trait in wheat has been the most effective mechanism for management of the wheat stem sawfly (WSS) for six decades. However, recent results have shown that in certain genotypes, the degree of stem solidness is not a useful indicator of WSS resistance. A morphological characterization of solidness expression indicated that in the genotype ‘Conan’, very solid pith undergoes rapid retraction during stem maturation, resulting in significantly less solidness at maturity. In other solid‐stemmed genotypes, including the standard WSS‐resistant cultivar ‘Choteau’, dense pith in the stem remains nearly unchanged throughout plant development. In cage trials, ‘Conan’ plants were less preferred for oviposition by the WSS when paired with ‘Choteau’ plants. Field bioassays using near‐isogenic lines differing for alleles at Qss.msub‐3BL showed that the Conan allele provides higher levels of early stem solidness and rapid pith retraction during stem maturation. These results suggest that the traditional approach for increasing WSS resistance by selecting for increasing stem solidness needs to be modified to consider temporal variations in pith expression associated with alleles at Qss.msub‐3BL.  相似文献

高产是甘蓝型油菜育种的重要目标之一,产量是多基因控制的数量性状。本文通过QTL作图分析了产量及其相关性状的数量性状位点,以甘蓝型油菜中油821和保604 F1代小孢子培养获得的DH系为作图群体,构建了由20个连锁群组成的,包括251个分子标记( 2个RFLP标记,72个RAPD标记,91个SSR标记,86个SRAP标记)的遗传连锁图(10个标记没有分配到连锁群中)。图谱的平均图距为6.96 cM,共覆盖油菜基因组1 746.5 cM。在此图谱基础上采取复合区间作图法,检测到与油菜产量及其相关性状有关的QTL共17个。其中控制株高的3个分别位于第4、第9和第10连锁群上,对性状的解释率为9.42%~17.58%;与分枝部位有关的4个分别位于第4、第6和第7连锁群上,其中Bp1 和Bp2 均位于第4连锁群,对性状的解释率为8.13%~15.20%;与主花序有效长有关的3个分别位于第4、第10和第16连锁群上,对性状的解释率为7.49%~23.36%;与一次有效分枝有关的2个分别位于第1、第4连锁群上,对性状的解释率为15.29%~19.58%;与角果总数和千粒重有关的分别位于第4连锁群和第9连锁群上,贡献率分别为17.42%和7.64%;与单株产量有关的3个分别位于第3、第4和第15连锁群,共解释26.60%的表型变异。部分性状的QTL在连锁群上成簇分布,对性状贡献率很大,表现主效QTLs的特点,相应的性状之间也呈显著相关,这表明一因多效或者相关的QTLs之间紧密连锁是性状相关的遗传基础。本研究中与主效QTLs连锁的标记可用于油菜产量性状的分子标记辅助选择。  相似文献
