通过检测热休克蛋白70(heat shock protein,HSP70)在免疫器官及细胞上的分布规律,研究热应激雏鸡免疫器官中热休克蛋白70的定位及表达。结果显示:HSP70在常温饲养雏鸡的胸腺、脾脏、法氏囊中均有明显的表达,在胸腺、脾脏的细胞中主要以胞浆表达为主,而法氏囊中则以胞核表达为主,脾脏中HSP70的表达强度最为稳定。热应激时,胸腺中HSP70的IOD值随日龄增长而逐渐降低,末期显著低于对照组(P〈0.05);脾脏中HSP70的IOD值随日龄增长逐渐降低,末期稍低于对照组(P〉0.05);法氏囊中HSP70的IOD值与饲养日龄间无明显规律,末期高于对照组(P〉0.05)。结果表明,热应激对雏鸡胸腺的损伤明显,胸腺细胞通过自身合成HSP70抵抗热应激的能力较差,而对脾脏和法氏囊影响较小。 相似文献

为研究慢性热应激对昭乌达牛血清酶活性、免疫功能及热休克蛋白的影响。本试验选取日龄和体况相近、体质健康的昭乌达牛24头,根据当地牧场环境温度、湿度变化,将试验期划分为热应激期和非应激期,于各试验期最后一天清晨进行颈静脉采血并制备血清。采用ELISA方法检测血清中酶活性、免疫指标及热休克蛋白含量。结果显示:慢性热应激对西门塔尔肉牛生理指标和血清酶活性有显著影响。在慢性热应激状态下,昭乌达牛呼吸频率极显著提高(P<0.01);血清中碱性磷酸酶(AKP)活性显著降低(P<0.05),肌酸激酶(CK)活性极显著降低(P<0.01),谷丙转氨酶(ALT)和谷草转氨酶(AST)活性极显著升高(P<0.01);热应激对昭乌达牛免疫功能和热休克蛋白含量有显著影响;血清白细胞介素-1β(IL-1β)含量在慢性热应激状态下显著降低(P<0.05),白细胞介素-4(IL-4)含量和脂多糖(LPS)浓度均极显著提高(P<0.01);热休克蛋白70(HSP70)含量极显著升高(P<0.01),热休克蛋白60(HSP60)和热休克蛋白90(HSP90)含量显著提高(P<... 相似文献

本研究应用3H-Dex放射配体结合的Scatchard分析和35S-蛋氨酸体外标记法分别测定了环境温度40℃热应激时,鸡外周血淋巴细胞(PBL)的糖皮质激素受体(GR)的最大结合容量(Ro)、平衡解离常数(Kd)及其热休克蛋白的表达。结果显示在热应激处理后0.5~4h鸡PBL、GR、Ro从85.75±10.09fmol/107细胞迅速下降至15.34±2.89fmol/107细胞。仅为正常对照组Ro的17.90%(P<0.01)。与此同时细胞热休克蛋白(HSP)的合成持续增加,主要有HSP100,HSP90,HSP70和HSP25。而在热应激最初0.5h细胞总的蛋白合成急剧下降,随着应激时间的延长,HSP合成的增多,细胞总的蛋白也逐渐恢复。提示HSP在热应激过程中对细胞结构和机能的重建、维持激素与GR的亲合力,稳定受体蛋白结构,提高机体热耐受力具有重要意义。 相似文献

The effects of in utero heat stress (IUHS) range from decreased growth performance to altered behavior, but the long-term impact of IUHS on postnatal innate immune function in pigs is unknown. Therefore, the study objective was to determine the effects of early gestation IUHS on the immune, metabolic, and stress response of pigs subjected to an 8 hr lipopolysaccharide (LPS) challenge during postnatal life. Twenty-four pregnant gilts were exposed to thermoneutral (TN; n = 12; 17.5 ± 2.1 °C) or heat stress (HS; n = 12; cyclic 26 to 36 °C) conditions from days 6 to 59 of gestation, and then TN conditions (20.9 ± 2.3 °C) from day 60 of gestation to farrowing. At 12 wk of age, 16 IUHS and 16 in utero thermoneutral (IUTN) pigs were selected, balanced by sex and given an intravenous injection of LPS (2 µg/kg BW mixed with sterile saline [SAL] and injected at 2 µL/kg BW) or SAL (2 µL/kg BW). Body temperature was monitored every 30 min, and blood was obtained at 0, 1, 2, 3, 4, 6, and 8 hr following the LPS challenge. Blood samples were analyzed for glucose, insulin, non-esterified fatty acids (NEFA), cortisol, and cytokine concentrations. In addition, white blood cell counts were determined at 0 and 4 hr. Hour 0 data were used as covariates. Body temperature was increased (P < 0.01) in LPS (40.88 ± 0.08 °C) vs. SAL (39.83 ± 0.08 °C) pigs. Eosinophils tended to be decreased overall (P = 0.09; 43.9%) in IUHS vs. IUTN pigs. Glucose concentrations were reduced overall (P = 0.05; 5.9%) in IUHS vs. IUTN pigs. The NEFA concentrations tended to be greater (P = 0.07; 143.4%) in IUHS-LPS pigs compared with all other treatments, and IUTN-LPS pigs tended to have greater (127.4%) circulating NEFA concentrations compared with IUTN-SAL and IUHS-SAL pigs. Cortisol was increased (P = 0.04) in IUHS-LPS compared with IUTN-LPS pigs at 3 hr (21.5%) and 4 hr (64.3%). At 1 hr, tumor necrosis factor α was increased (P = 0.01; 115.1%) in IUHS-LPS compared with IUTN-LPS pigs. Overall, interleukin-1β (IL-1β) and interleukin-6 (IL-6) were greater (P < 0.04; 281.3% and 297.8%, respectively) in IUHS-LPS pigs compared with all other treatments, and IUTN-LPS pigs had increased IL-1β and IL-6 concentrations compared with IUTN-SAL and IUHS-SAL pigs. In summary, IUHS altered the postnatal cytokine, metabolic, and physiological stress response of pigs during postnatal life, which may have negative implications toward the innate immune response of IUHS pigs to pathogens. 相似文献

文章旨在研究日粮单独或联合使用锌和维生素C对高温条件下肉鸡生长性能和免疫反应的影响。试验选择320只1d肉仔鸡,根据体重随机分为4组,每组4个重复,每个重复20只鸡。对照组饲喂基础日粮(锌含量为40mg/kg),处理1组在基础日粮中添加60mg/kg锌,处理2组在基础日粮中添加200mg维生素C,处理3组在基础日粮中添加60mg/kg锌+300mg/kg维生素C,试验共开展35d。在试验第21~35天对鸡只进行热应激诱导,即25℃12h,25~34℃3h,34℃6h,34~25℃3h。结果显示:处理组较对照组均显著提高了肉鸡的采食量、体增重(P<0.05),显著降低了料比(P<0.05)。日粮同时添加锌和维生素C组肉鸡采食量和体增重最高(P<0.05)。与对照组相比,日粮添加60mg/kg锌、200mg/kg维生素C或锌和维生素C联合使用的肉鸡脾脏、胸腺和法氏囊重量显著提高(P<0.05)。60mg/kg锌+200mg/kg维生素C和200mg/kg维生素C组较对照组显著提高了新城疫、传染性法氏囊炎和传染性支气管炎的抗体滴度(P<0.05)。与对照组相比,60mg/kg锌+200mg/kg维生素C、200mg/kg维生素C组和60mg/kg锌组显著提高了血液白细胞总数、淋巴细胞和单核细胞占比(P<0.05)。结论:在本试验条件下,日粮单独或联合添加锌和维生素C均可显著改善热应激条件下肉鸡的生长性能和免疫状态。 相似文献

热应激是指动物机体对热应激源的非特异性防御应答的生理反应,其实质是指环境温度超过等热区中的舒适区上限温度所致的非特异性反应的总和。它通过改变禽的生理生化反应降低鸡采食量、日增重、产蛋率,降低饲料利用率,使蛋形变小、蛋壳变薄变脆,更严重者导致热衰竭,在短时间内造成鸡大批死亡,给养禽业带来极大的危害。近年来随着养鸡业集约化规模化的发展及全球性气温的普遍升高,热应激也越来越严重。因此,阐明热应激的作用机理及影响热应激的多种因素,对于探索缓解热应激的有效措施、指导生产实践有十分重要的意义。 相似文献

牦牛感染牛病毒性腹泻病毒(Bovine viral diarrhea virus,BVDV)后常发生胃肠炎等消化系统疾患和呼吸系统疾患、繁殖障碍,造成机体损伤,给牦牛产业发展造成一定影响。基于5"-UTR的差异将BVDV分为BVDV-I和BVDV-II两种基因型,牦牛源BVDV的主要基因型为BVDV-1型;根据BVDV对宿主细胞的致病性,分为致细胞病变型(Cy-topathogenic,CP)和非致细胞病变型(Non-cytopathogenic,NCP)两个生物型,牦牛源BVDV既有CP型又有NCP型,且这两种生物型能相互转化。牦牛生活的高原地区风、鸟类的迁徙等因素可能都会导致牦牛BVD传播范围广泛,牛群中大量的BVDV携带者和牦牛特殊而又复杂的生态系统导致了BVDV种间传播的巨大潜力,影响着牦牛产业的健康发展和养殖经济收益。针对本病在牦牛群中的感染流行等情况未发现相关详细的研究报道,笔者就牦牛BVD病原学、流行病学调查、实验室诊断、综合防治等方面研究进展情况进行整理,供同仁在研究和防控中参考借鉴。 相似文献

1. Hubbard x Hubbard (HH) and Shaver x Shaver (SS) chicks given a dietary supplement of either 50 mg/kg oxytetracycline (OTC) or 1 g/kg Lactobacillus culture (LC) were exposed to 36 +/- 1 degrees C for 3 h daily from day (d) 21 to 42. 2. Prior to heat treatment, body weight (d 21) and weight gain (d 1 to d 21) of OTC and LC birds were greater than those fed the control diet. Chicks given LC had the best food efficiency followed by OTC and control birds during d 1 to d 21. Body weight (d 1 and d 21) and weight gain (d 1 to d 21) were greater for HH tlhan SS chicks. 3. After 3 weeks of heat exposure, birds receiving the LC diet had greater body weight and weight gain, higher food intake and lower food efficiency than OTC and control chicks. 4. Antibody production against Newcastle discase vaccine on d 21 was not affected by strain or diet. On d 42, while diet had negligible effect on this variable among the SS broilers, HH birds fed LC had higher antibody production than those on the control diet. 5.
