烟草品种资源对根结线虫病抗病性鉴定   总被引:7,自引:0,他引:7  
 在1996和1997年,在温室采用卵接法和病土接种法,测定65份烟草种质资源对南方根结线虫1号小种和爪哇根结线虫的抗病性。其中,16份抗南方根结线虫1号小种,25份中抗南方根结线虫1号小种;仅有Va102中抗爪哇根结线虫;经田间试验测定10份烟草种质资源对花生根结线虫2号小种的抗病性,CF954对花生根结线虫2号小种表现中抗。本文还研究了卵接法和病土接种法对测定品种抗性的差别,结果表明二者无显著差异。  相似文献

南方、爪哇和花生根结线虫的快速灵敏的PCR鉴定方法   总被引:7,自引:0,他引:7  
 为了研制南方、爪哇和花生根结线虫快速灵敏的检测和鉴定方法,分别分离了4个南方根结线虫和3个爪哇根结线虫特异性的随机扩增多态性DNA (RAPD)片段。在这些RAPD标记DNA序列的基础上,设计了多对SCAR PCR引物,并用源于国内外的南方、爪哇、花生、北方和象耳豆根结线虫群体验证其扩增特异性和灵敏度。最终确定了3对高效扩增的SCAR引物,它们组合使用可以可靠灵敏地鉴定南方、爪哇和花生根结线虫。3对引物的扩增灵敏度达1/3条的二龄幼虫、雄虫或雌虫,这表明本研究研制的PCR鉴定法可用于生产实践中土样和根样中3种根结线虫快速灵敏的鉴定。  相似文献

Cylindrocladium black rot (CBR), caused by Cylindrocladium parasiticum , and root-knot nematode, Meloidogyne arenaria , both infect and cause damage to the roots of peanut. Greenhouse and microplot experiments were conducted with the runner type peanut genotypes C724-19-15, C724-19-25 and Georgia-02C with different levels of resistance to nematode and CBR to better understand the interactions between the two pathogens. In the greenhouse, inoculation of 500–3000 eggs per plant of M. arenaria did not affect the level of root rot induced by 1·0 to 5·0 microsclerotia of C. parasiticum per g soil. In microplots, the root rot ratings from Georgia-02C and C724-19-25 were higher in plots infested with M. arenaria (0·4–2·0 eggs per cm3 soil) and C. parasiticum than in plots with C. parasiticum alone; however, M. arenaria did not increase the root rot ratings on the nematode resistant C724-19-15. This was inconsistent with results in the greenhouse. Gall indices were not affected by C. parasiticum inoculations in the greenhouse or microplots. In both 2006 and 2007, a significant interaction between C. parasiticum inoculum densities and nematode level was observed on plant mortality. CBR inoculum greatly increased mortality on C724-19-25 and Georgia-02C, but not on C724-19-15, in the presence of M. arenaria . The mortality increase was more apparent at lower inoculum levels of both pathogens, but on the nematode-susceptible cultivars plant mortality was more with co-inoculations of the two pathogens than from either alone. Simultaneous inoculation with the two pathogens decreased yield of C724-19-25 and Georgia-02C as C. parasiticum inoculum levels increased, but even the largest inoculum of M. arenaria (2·0 eggs per cm3 soil) did not decrease yield of C724-19-15.  相似文献

为了解根结线虫在中国南方稻区的分布和种类, 从15个省(直辖市/自治区)采集1 181份土样, 经生物测定发现312份土样中种植的水稻或番茄幼苗根系会产生根结, 阳性总检出率为26.42%。利用线虫特异性引物从171份阳性样品中鉴定出6种根结线虫:拟禾本科根结线虫Meloidogyne graminicola、北方根结线虫M.hapla、花生根结线虫M.arenaria、爪哇根结线虫M.javanica、象耳豆根结线虫M.enterolobii和南方根结线虫M.incognita, 分别占鉴定总样品数的88.89%、41.52%、29.82%、10.53%、4.09%和3.51%。被1、2、3、4、5、6种根结线虫混合侵染的土样比例分别为42.11%、41.52%、12.87%、2.92%、0.58%和0, 其中最常见的为拟禾本科根结线虫单独侵染(34.50%)、拟禾本科根结线虫与北方根结线虫(22.81%)或花生根结线虫(13.45%)混合侵染。本研究结果将为制定相应的传播阻断策略和防控措施提供参考。  相似文献

ABSTRACT Root-knot nematodes of the genus Meloidogyne are economically important pathogens of a wide range of crops. The tomato resistance gene Mi typically confers resistance to the three major species, M. incognita, M. javanica, and M. arenaria. However, virulent populations completely overcoming the Mi resistance still occur. In an attempt to develop molecular markers for virulence against Mi and gain insights into the genetic relationships among virulent populations of different species and origins, random amplified polymorphic DNA (RAPD) analyses of laboratory-selected virulent, field virulent, and avirulent populations of M. incognita, M. javanica, and M. arenaria were carried out. A RAPD marker, specific for selected virulent populations, was identified, and subsequently, converted to a sequence characterized amplified region (SCAR). Sequence characterization of the SCAR locus showed that alleles from laboratory- and field-selected virulent populations were highly similar to each other and clearly different from alleles from natural virulent and avirulent populations. This result suggests that the genetic mechanism for virulence against Mi may be similar among selected virulent populations of the three Meloidogyne spp., but different between selected and natural virulent populations. Based on the nucleotide polymorphisms at the SCAR locus, codominant and dominant polymerase chain reaction-based markers were developed enabling rapid diagnosis of selected virulent genotypes in M. incognita, M. javanica, and M. arenaria.  相似文献

ABSTRACT Root-knot nematodes (Meloidogyne spp.) are sedentary, obligate endoparasites in plants, where they induce specialized feeding sites. The feeding sites act as strong metabolic sinks to which photosynthates are mobilized. The histopathological modifications in the nematode-induced feeding sites of artificially inoculated chickpea cv. UC 27 were qualitatively and quantitatively compared using five isolates of M. artiellia and one isolate each of M. arenaria, M. incognita, and M. javanica. All Meloidogyne isolates infected chickpea plants, but root gall thickening was significantly less for M. artiellia isolates than for the other Meloidogyne species. Nevertheless, neither the number of giant cells in the feeding site (averaging four to six) nor the area of individual giant cells was influenced by nematode species or isolate. However, the number of nuclei per giant cell was significantly smaller, and the maximum diameters of nuclei and nucleoli were significantly greater, in giant cells induced by M. artiellia isolates than in those induced by M. arenaria, M. incognita, or M. javanica. In a second experiment, M. artiellia-induced giant cells in faba bean and rapeseed also contained a small number of large nuclei.  相似文献

 应用瑞典Pharmacia Biotech.公司开发的全自动快速水平电泳仪(PhastSystem)进行酯酶和苹果酸脱氢酶电泳,对云南省烟草和观赏植物一串红上的根结线虫进行快速鉴定,得出4种根结线虫:花生根结线虫(Meloidogyne arenaria)、爪哇根结线虫(M.javanica)、南方根结线虫(M.incognita)和北方根结线虫(M.hapla)。文章提供电泳方法,认为PhastSystem电泳仪是根结线虫种类快速鉴定的理想设备。  相似文献

近年来,随着我国荞麦面积的迅速增加,根结线虫病呈逐年加重趋势,严重影响了荞麦的产量与品质。本研究于2014年对西南10个地区17个乡镇秋播苦荞麦根结线虫的分布、发生种类以及危害程度进行了调查。结果表明,西南地区危害荞麦的根结线虫种类有南方根结线虫(Meloidogyne incognita)、爪哇根结线虫(M.javanica)和花生根结线虫(M.arenaria)3种,其中南方根结线虫为优势种群。田间根结线虫种群大多数为单一种群,23.5%的样品为南方根结线虫与爪哇根结线虫(或花生根结线虫)组成的混合种群。调查发现前作为烟草或马铃薯的地块,荞麦根结线虫发生危害严重,平均被害株率为6.0%~77.5%,病情指数为1.4~26.1,而前作为玉米的荞麦根结线虫则较轻,被害株率最高为16.0%,相应病情指数为2.3。  相似文献

成都平原水旱轮作田常见根结线虫调查   总被引:1,自引:0,他引:1  
调查四川省成都平原地区水旱轮作田根结线虫发生情况及种类分布,为水旱轮作田根结线虫的防治提供依据。对成都平原15个县(市、区)28个乡(镇)共159块水旱轮作田的土样进行根结线虫普遍率调查,利用线虫ITS通用引物和8对根结线虫特异性引物对其中检出率高的地区重新采集的土样进行根结线虫种类分子鉴定。结果表明,成都平原水旱轮作田根结线虫平均检出率为40.25%。重新采集的土样共鉴定出5种根结线虫:拟禾谷根结线虫Meloidogyne graminicola、花生根结线虫M. arenaria、南方根结线虫M. incognita、北方根结线虫M. hapla和爪哇根结线虫M. javanica,各线虫的检出率分别为40.91%、34.09%、13.64%、13.64%和2.27%,其中29.55%的样品由2种以上根结线虫混合侵染。拟禾谷根结线虫和花生根结线虫为该地区优势种群。  相似文献

Sorption and desorption of ‘Pegosperse’ 100-O(PEG 100-O; diethylene glycol monooleate, containing 15% diester) surfactant by unaltered (CM) and dewaxed (DCM) adaxial cuticle membranes isolated from apple (Malus pumila M.) leaves were studied. The aim of this study was to understand interactions between surfactants and cuticles. Enzymatically isolated cuticles were soaked in buffer or PEG 100-O solution (pH 7–0). and the weight changes of cuticles were measured to determine the amount of surfactant sorbed or desorbed by the cuticles. For very low surfactant concentrations, sorption was measured by changes in the surface tension of the solutions. PEG 100-O sorption by both the CMs and the DCMs occurred mainly in the first three hours and was concentration-dependent. The DCMs always sorbed more surfactant than the CMs. Desorption of PEG 100-O from both CMs and DCMs was rapid in the first few hours and then decreased to a relatively low rate until the surfactant was totally desorbed from the cuticles after about two months. The sorption and complete desorption of the surfactant by both CMs and DCMs show that PEG 100-O interacts with both cutins and waxes of the cuticles and the interactions are reversible.  相似文献

以石羊河中游荒漠河岸植被优势种群沙蒿(Artemisia arenaria)为研究对象,采用点格局分析法,对垂直于河道不同距离梯度上沙蒿种群的空间格局和空间关联性进行了分析。结果表明:① 随着垂直于河道方向距离的递增,沙蒿种群由增长型逐渐演变为衰退型;② 垂直于河道不同距离梯度上沙蒿种群的格局特征为,在相对小的尺度上聚集分布特征明显,随尺度的增加聚集性减弱并逐渐表现出随机分布的格局特征,但距离河道近的沙蒿种群聚集强度大于距离河道远的种群;不同距离梯度上,沙蒿种群均表现出幼体聚集分布,成体随机分布的格局特征;③ 垂直于河道不同距离梯度上沙蒿种群不同龄级之间的关联性表现出异质性。距离河道近的沙蒿种群中,形体差异大的植株之间具有正关联性,反映了互利的生态关系;距离河道较远的沙蒿种群中,形体差异小的植株之间具有负关联性,反映了互斥的生态关系;其他龄级植株之间没有表现出明显的关联性。  相似文献

为明确上海地区稻田新优势杂草耳叶水苋的发生规律,对上海地区稻田中耳叶水苋的发生情况进行了田间调查和定点观察,并在室内研究了其生物学特性。结果表明,耳叶水苋发生周期长,有两个明显的发生高峰,分别为水稻播种后第1周和第3周。水稻播种后45 d,耳叶水苋种群数量达到最大,随后,种群数量逐渐下降。耳叶水苋的种子在23~27 ℃条件下发芽最好。作为一种水生杂草,其种子在土壤含水量40%~60%的湿润或饱和状态下,发芽率较高,且耳叶水苋的种子对水层具有较好的适应能力,即使处于10 cm水层下也可以正常萌发。耳叶水苋种子极小,其顶土能力较差,覆盖0.5 cm土层能有效抑制其出苗。  相似文献

 象耳豆根结线虫是一种在中国具有潜在经济重要性的农作物病原物。为提供有助于控制象耳豆根结线虫传播扩散的方法,研制了该线虫的快速PCR鉴定和检测法。该方法PCR引物的扩增目标为rDNA-IGS2区域,其设计依据象耳豆根结线虫与南方、爪哇、花生和北方根结线虫在该区域核酸序列的差异。通过对6种近似根结线虫的不同地理群体及自然土壤线虫群体的测试,验证了设计的PCR引物针对象耳豆根结线虫的特异性和可靠性。本方法具有快速灵敏的特点,可用于象耳豆根结线虫单条线虫的直接鉴定以及混合土壤线虫群体中象耳豆根结线虫的检测。  相似文献

运用比较形态学、同工酶和 mtDNA-PCR 对采自海南岛9个市县的南药根结线虫进行种类鉴定,结果发现有南方根结线虫、象耳豆根结线虫、爪哇根结线虫、花生根结线虫和西班牙根结线虫等5种根结线虫危害海南岛南药。5种根结线虫均能在海巴戟上寄生与危害,而南方根结线虫寄主非常广泛,可危害大多数调查的药用植物。  相似文献

研究表明,我国红麻上的根结线虫有南方根结线虫(Meloidogyne incognita)1号和2号小种,爪哇根结线虫(M.javanica)和花生根结线虫(M.arenaria)2号小种,南方根结线虫是优势种。它们都不为害棉花、花生、小麦、芝麻和苎麻,对高粱和玉米为害极轻。丝瓜、西瓜、番茄和空心莲子草为其共同寄主。南方根结线虫还侵染大豆、豇豆、苦瓜、菜瓜、南瓜、茄子、辣椒、烟草、黄麻、甘薯、黄瓜、白菜、莴苣、菜豆和扁豆等作物。红麻根结线虫的二龄幼虫主要分布在0~40cm 土层内,砂壤土中的线虫密度大于黄壤土。病原线虫以卵和二龄幼虫主要在病株残体中越冬。实行轮作、清除病残和药剂处理土壤是防治本病的有效措施。  相似文献

A molecular protocol is presented for distinguishing seven of the most common and economically important Meloidogyne spp. DNA was extracted from individual second-stage juvenile (J2) nematodes of Meloidogyne spp. and amplified by PCR (polymerase chain reaction). Fifteen PCRs including amplification of rDNA, specific SCAR (sequence characterized amplified region) and RAPD (random amplified polymorphic DNA) fragments were possible from the extracted DNA. This enabled a molecular diagnostic key for M. incognita , M. javanica , M. arenaria , M. mayaguensis , M. hapla , M. chitwoodi and M. fallax to be designed. The key unifies published methods into a single logical schematic using primer combinations that were previously validated and shown to work reliably and specifically. The protocol can be used with single juvenile or adult nematodes and the schematic can readily be expanded to accommodate more species. The use of RAPD amplification to assist with identification of samples which do not yield diagnostic amplification products after the first three steps of the molecular key is also described.  相似文献

Four species of root-knot nematodes, namely Meloidogyne incognita (Kofoid et White) Chitwood, M. javanica (Treub) Chitwood, M. arenaria (Neal) Chitwood, and M. hapla Chitwood have a wide host-range and are widely distributed throughout the world. For each of the species there are certain non-hosts and these are different for each species, thus making it possible, through the use of differential hosts, to identify the species as well as detect pathogenic variations within a species. A knowledge of the nature and extent of this variation is needed for the development of varieties which possess a broad base for resistance.  相似文献
