At the IPK Gatersleben a series of 85 bread wheat (T. aestivum)/goatgrass (Aegilops tauschii) introgression lines was developed recently. Based on the knowledge that chromosome 7D of this particular Ae. tauschii is a donor of resistance to septoria tritici blotch (Mycosphaerella graminicola), a sub-set of thirteen chromosome 7D introgression lines was investigated along with the susceptible recipient variety ‘Chinese Spring’ (CS) and the resistant donor line ‘CS (Syn 7D)’. The material was inoculated with two Argentinian isolates of the pathogen (IPO 92067 and IPO 93014) at both the seedlings (two leaf) and adult (tillering) stages at two locations over 2 years (2003, 2004). The resistance was effective against both isolates and at both developmental stages, and the resistance locus maps to the centromeric region of chromosome arm 7DS. On the basis of its relationship with the microsatellite marker Xgwm44, it is likely that the gene involved is Stb5. Stb5 is therefore apparently effective against M. graminicola isolates originating from both Europe and South America.  相似文献

Devastating maize grain yield and quality losses are caused by Aspergillus flavus, Fusarium verticillioides and Stenocarpella maydis ear rots especially in tropical countries. Therefore, combining ability of tropical maize populations for ear rot severity and ear rot-related traits was investigated. Ten full-sib progenies, comprising one resistant and one susceptible from each of the five populations, were selected for mating in a 10 × 10 full diallel. The full-sib progeny crosses were evaluated across two environments with two replications in Zambia. To determine resistance across three ear rots that occur together in Zambia, the crosses were artificially inoculated with a mixture of Aspergillus flavus, Fusarium verticillioides and Stenocarpella maydis isolates. There were marked differences between environment main effects and their interaction with GCA and SCA effects were highly significant, suggesting observation of genotype × environment interaction effects. Both additive and non-additive gene effects were significant for ear rot severity. Highly significant reciprocal differences were also revealed, suggesting that cytoplasmic gene effects and their interaction with nuclear genes were responsible in modifying resistance across the three ear rot diseases in the full-sib progenies that were derived from the five tropical maize populations.  相似文献

Summary Twenty Finnish isolates of Rhynchosporium secalis (Oud.) J.J. Davis, the causal agent of scald, were taken from infected barley (Hordeum vulgare L.) plants and inoculated on to seedlings of a differential series of barley containing a range of major genes for resistance to the fungus, as well as on to six Nordic 6-row spring barleys and three winter ryes (Secale cereale L.). These fungal isolates derived from four sites and three host varieties. Disease development was monitored on two leaves of seedlings in the greenhouse employing a standard scale, and on adult plants in the field by assessing the diseased area on the three uppermost leaves. A comparison was also made between the pathogenicity and virulence of ten Finnish and ten Canadian R. secalis isolates. The Finnish isolates varied in virulence, but with the exception of Algerian (CI 1179) seedlings and adult La Mesita (CI 7565) all seedlings and adult plants of the entire differential series were resistant to all isolates. Canadian isolates were, on average, less virulent than Finnish isolates. All the Nordic checks were susceptible to all Finnish and seven Canadian isolates, but differences in the degree of susceptibility were evident. Isolates of R. secalis from barley were non-pathogenic on rye, isolates from Elymus repens L. were non-pathogenic on barley and rye, and isolates from rye were only pathogenic on rye. Finnish R. secalis isolates contain no redundant pathogenic diversity. The differential series represents a useful, but as yet untapped, source of resistance to R. secalis for Finnish barley breeders.  相似文献

Summary Fifty-six landraces of Portuguese coles (Brassica oleracea l.), representing fourteen morphotypes, were screened for resistance to Leptosphaeria maculans at the cotyledon stage. Three isolates of the pathogen belonging to three pathogenicity groups were used in the screening. The mean disease interaction phenotype of the B. oleracea landraces with the L. maculans isolates confirmed their differences in pathogenicity. When compared to pathogenicity grouping using a B. napus differential set, a different classification was observed on B. oleracea for the three L. maculans isolates, suggesting possible differences in the genetic interaction between the two plant species and the pathogen. Most of the Portuguese cole landraces were found to be susceptible to all three isolates of L. maculans, although a few have shown intermediate interaction phenotype. Some accessions, on the other hand, elicit a differential reaction with the three isolates tested, and can be further selected to be used in host differential sets or in breeding for resistance to black leg.Fifty-four landraces of Portuguese coles, representing eleven morphotypes, were also screened in the field for resistance to Xanthomonas campestris pv. campestris. A good level of resistance to the bacterium was found in several of the morphotypes.Accessions of the Penca type, particularly, were identified as highly resistant. Landraces belonging to the same Portuguese cole group presented a similar range of mean interaction phenotype with the bacterium. Some Portuguese landraces of B. oleracea are potential sources for use in breeding programs for black rot resistance.  相似文献

Summary Variation in the responses of a wide range of accessions of four native Australian species of Glycine (viz. G. canescens, G. clandestina, G. tabacina and G. tomentella) to infection by eight Australian isolates of Phakopsora pachyrhizi was analyzed.Differences in the infection type responses of the various wild Glycine species were sufficient to recognize six different virulence combinations amongst the eight pathogen isolates.A set of differential hosts useful in the identification of different races of the pathogen is presented to facilitate further examination of the virulence structure of the pathogen population.  相似文献

Co-evolution of wheat and its devastating pathogen Mycosphaerella graminicola (anamorph Septoria tritici), the causal agent of septoria tritici blotch, a foliar disease of wheat, is suggested to occur in Fertile Crescent as their center of origin and, thus, interaction between pathogen virulence and host resistance is important subject to be addressed. We have investigated resistance spectra of 54 wheat genotypes including a set of differentials carrying known resistance genes and virulence patterns of 14 M. graminicola isolates at seedling stage under controlled environmental conditions. The isolates were collected in Iran from five provinces. Diversity in virulence and aggressiveness was observed among the isolates from four provinces. Isolates collected from Golestan province were virulent to all wheat genotypes from germplasm of Iran, while specific resistances were identified to the isolates from other provinces. Among wheat genotypes, cvs. Chamran, Morvarid and Hirmand had the greatest number of specific resistances as well as partial resistance. Wheat genotypes of the differential set also differed in their reactions to the isolates. Arina, Flame and TE 9111 were specifically resistant to the greatest number of isolates from different provinces. Most isolates were virulent to the other differentials such as cvs. Shafir, Estanzuela federal and Courtot indicating that extensive adaption of virulence to most of the known resistance genes (Stb) has occurred in these regions. The new sources of resistance to highly virulent isolates from Iran may also be utilized in wheat breeding programs to develop resistant cultivars against pathogen populations in other countries.  相似文献

Summary Seedlings of a differential barley (Hordeum vulgare L.) series (21 genotypes) and of six check genotypes were used in the greenhouse to assess variation in virulence among 20 single-spore isolates of the net blotch pathogen. Pyrenophora teres Drechs. f.teres Smedeg., collected from various sites in Finland. The experiment was run twice and symptom expression was recorded on the first three leaves. Analysis of second leaf symptom scores from Run 1 indicated differences in virulence between isolates, all of which were pathogenic, and differential resistance among the barleys. The virulence of P. teres isolates appeared to be conditioned by the host barley from which the isolate derived; the average virulence of isolates collected from a susceptible host was greater than that of isolates collected from a less susceptible host. Results from Run 2 were similar regarding resistance within the barleys, but variation in virulence among the P. teres isolates was not consistent with that from Run 1. CI 9819 caries duplicate genes for resistance to P. teres (Rpt1b and Rpt1c), and CI 7548 possesses Rpt3d. Both genotypes were highly resistant to all isolates. The Rpt1a gene of Tifang (CI4407) conferred resistance to all isolates in Run 2, but only to about half of the isolates in Run 1. The checks, including two of the symptomatically most resistant Nordic barley genotypes, were universally susceptible during these stringent tests. No selective pressure has been placed on Finnish isolates of P. teres through previous deployment of major resistance genes, and it is speculated that any variation in virulence among isolates is likely to be due to a combination of evolutionary forces including, natural selection, random genetic drift and gene flow.  相似文献

Summary Pathogenicity patterns of 42Mycosphaerella graminicola (Septoria tritici) isolates secured from the major wheat growing regions in Israel, were assessed on seedlings of 16 bread and durum wheats. The spring bread wheat cultivar Titan (CI12615), the winter bread wheats Bezostaya 1, Kavkaz, NE7060 and the durum wheats Nursit 163 and Zenati Bouteille, all exhibited a high level of resistance. Significant cultivar × isolate interactions were recorded for isolates from the different regions in relation to the response of the 16 cultivars organized in 6 reponse classes to the test isolate ISR8036. Number of genes for resistance in the 16 cultivars was estimated, based on the assumption of a gene-for-gene relationship. Twelve complementary genes were hypothesized in the 42M. graminicola isolates × 16 wheat cultivars matrix. There was considerable variation in virulence frequencies between regions and between locations within the same region. The overall frequency of Bezostaya-Kavkaz virulence was low throughout the country (5–7%). The frequency of designated hypothesized virulence genes VST OLAF, VST COLOTANA, VST IAS 20 in Israel, was high in most locations. The reported analytical approach enables the identification of virulence hot-spots and assists in designing breeding for resistance strategies.  相似文献

Summary Previously identified segregant lines of oats with levels of adult plant resistance to powdery mildew (Erysiphe graminis f.sp. avenae) better than the resistant parent maintained this high level of resistance in field nurseries over two growing seasons. This enhancement was not expressed on inoculated detached leaves under laboratory conditions where no differences between the more resistant parent cv. Maldwyn and the most resistant segregants was detected. Reduced resistance was not detected in tests where leaf segments of the transgressive lines were inoculated with trained isolates. Problems associated with selecting for adult plant resistance under both field and laboratory conditions are discussed.  相似文献

Summary Twenty-one accessions of 3 wild Lactuca species which could be hybridised with L. sativa, the cultivated lettuce, were inoculated at different stages of plant development with 3 multivirulent isolates of Bremia lactucae. Nineteen sources of resistance to B. lactucae, not attributable to the previously recognised resistance factors 1–11 were identified. Two lines of L. serriola showed similar resistance patterns as lines carrying R11. The resistance of some accession was incomplete particularly at the seedling stage and this phenomenon may be race specific.Tests on segregating F2 populations of crosses between 2 different L. serriola accessions and L. sativa cultivars showed that the resistance in one line (LSE/18) appears to be inherited as a single dominant gene, which is sometimes incomplete in expression and allelic to either Dm6 or R7. The segregation patterns for resistance in PI 281876 did not give readily interpretable ratios.To assess the frequency of occurrence in B. lactucae populations of virulence factors to overcome this novel resistance, 11 of the novel sources of resistance were inoculated with numerous collections of the pathogen from the UK, Czechoslovakia and elsewhere and found to show a high level of resistance.  相似文献

Summary Grain-yield data obtained from 49 varieties of maize (Zea mays L.), grown at each of five sites for two years, were analyzed with the objective of identifying two or three sites where mean performance adequately predicted the mean performance of all five sites combined. Regression of overall varietal mean on site or site-combination means was done and the predictive potential of a site or combination of sites was judged by four criteria: (i) r20.9, (ii) b1.0, (iii) high yield (i.e., yield greater than or equal to overall mean yield) and (iv) consistent yield (i.e., yield did not fall below the mean yield by more than 10%). In addition, a trial differentiation index, D, was used to compare the site combinations selected on the basis of the above criteria with the differentiation potential of all five sites combined. None of the five individual sites and the ten two-site combinations satisfactorily predicted overall mean yield. However, five three-sites combinations had values of D similar to that of all the five sites. Rank-correlation coefficients of the three-site combinations with all five sites ranged from 0.90 to 0.79. It was concluded that certain three-site combinations could be satisfactorily used for preliminary maize yield trials in the rainforest ecological zone of S.W. Nigeria.  相似文献

This experiment was carried out to investigate whether and how much field resistance to late blight, caused by Phytophthora infestans, is present in the local cultivated potato germplasm. In total 36 entries were compared in a field experiment in an area highly conducive to late blight development. Of the 36 cultivars 32 were local cultivars belonging to five Solanum species, S. tuberosum (1 accession), S. andigena (18), S. juzepczukii (2), S. stenotomum (9) and S. ajanhuiri (2). The other four cultivars were derived from breeding programmes, one being the Dutch cultivar Alpha used as a highly susceptible control. The 36 cultivars were planted according to a simple 6 × 6 lattice design with three replicates. Each replicate was divided in six incomplete blocks each with six cultivars. The disease severity was assessed weekly during 9 weeks starting 48 days after planting. The area under the disease progress curve (AUDPC) was used as a measure of the field resistance. Nine isolates from surrounding potato fields were tested for their virulence to the resistance genes R1–R11 using 22 differential cultivars. The components of the field resistance of 19 of these cultivars were compared in the greenhouse using a local isolate with virulence to all known R-genes, except to R9. The nine isolates represented seven races with a race complexity varying from 7 to 10 virulence factors. All isolates carried virulence against R1, R2, R3, R7, R10 and R11, while virulence against R9 was absent. The AUDPC among the 32 local cultivars ranged from very large, significantly larger than that of ‘Alpha’ to very small. The AUDPC from S. stenotomum accessions ranged from very large to intermediate, those from S. andigena accessions from large to very small. Especially among the S. andigena accessions interesting levels of field resistance were found. Four components of field resistance were assessed, latency period (LP), lesion size (LS), lesion growth rate (LGR) and relative sporulation area (RSA). All four showed a considerable variation among the cultivars. The LP ranged from 3½ to 6 days. The LS ranged from 225 mm2 to 20 mm2. The LGR varied about six-fold, the RSA more than 10-fold. The components tended to vary in association with one another. LP and LGR were well associated with each other and had a significant correlation with the AUDPC.  相似文献

Summary Isolates of three species of rust (Puccinia allii, P. mixta and Uromyces ambiguus) were each inoculated onto a range of potential host plants, selected to include the major European crop Allium species together with the commoner wild species in the section Allium. Measurements of incubation period, lesion type, latent period and pustule quantity were made in a single environment. Each isolate was able to sporulate on a range of Allium species. There was evidence of specialisation to particular sections of the genus; all the leek isolates performed well on the majority of hosts in section Allium, whilst the chive isolate grew poorly on most members of that section. Exceptions to this are exemplified by A. fistulosum being attacked by all three rust species and A. sativum being attacked by both leek and chive isolates. A continuum of reaction types was found which, for the leek isolates, ranged from the low levels of quantitative resistance in leek cultivars, through higher quantitative levels within the A. ampeloprasum complex, to qualitative resistance in several interactions in non-host sections of Allium, where the pathogen was able to infect and form colonies of some size without sporulating.  相似文献

Seventy-five barley landraces from Morocco were tested for resistance to powdery mildew and a number of different resistance genes were detected. Thirty-five isolates of Blumeria graminis f. sp. hordei and the Pallas isoline differential set were used. Isolates used in the experiment had virulences corresponding to all major resistance genes used in Europe. Forty-four of the tested landraces showed resistant reactions. From each of these landraces, one to five resistant plants were selected and 92 single plant lines were created. Six lines selected from 3landraces were assumed to carry the mlo gene but they were discarded after microscopic investigation. Seventeen lines were tested in the seedling stage with 17isolates and another 69 lines were tested with 23 differential isolates. These lines showed 71 reaction spectra to isolates of powdery mildew. Eight lines (9%), 255-3-3, 282-3-4, 286-1-1, 294-2-3,294-2-4, 295-1-2, 308-1-2 and 327-2-1, selected from 7 landraces showed resistance to all isolates. Seventy-eight lines (90%) showed a resistant infection type 2with more than 50% of the isolates used. In most of the selected lines (86%) unknown genes, alone or in combination with known specific resistance genes, were detected. Four different resistance alleles (Mlat, Mla6, Mla14 and Mla1) were postulated to be present in the tested lines. The most common was Mlat, which was postulated in 35 (41%) lines. The use of newly identified sources of powdery mildew resistance in barley breeding is discussed. Sources of resistance to Fusarium spp. are needed to develop maize hybrids resistant to the accumulation of fungal mycotoxins in the grain. In a search for resistant germplasm in 1999 and 2000, a set of Argentinian maize populations was evaluated in Ottawa, Canada, for resistance to ear rots after inoculation with local isolates of Fusarium verticillioides and F. graminearum. Sixteen of these populations, varying in observed resistance levels, were re-evaluated in 2003 and 2004 in Pergamino, Argentina, using local isolates of the same fungi. Conidial suspensions of each fungal species were inoculated into the silk channel of primary ears. Disease severity was assessed after physiological maturity using a scale based on the percentage of visibly infected kernels. Genotype effect was more important than genotype-by-fungal species or genotype-by-fungal species-by-environment interaction effects. In addition, disease severity levels associated with each fungal species were positively correlated (P < 0.05) (r = 0.90, r = 0.81, r = 0.87 and r = 0.53, in Ottawa 1999 and 2000, and Pergamino 2003 and 2004, respectively). Populations ARZM 01107, ARZM 07138, ARZM 10041, ARZM 13031, ARZM 16002 and Pora INTA exhibited the highest and most stable resistance to both species. Considering that disease resistance exhibited low specificity to the environment and to the fungal species in evaluations conducted in a wide range of environments and with fungal isolates collected from different hemispheres, the most resistant populations are potential sources of genes for stable resistance to these Fusarium spp.  相似文献

A total of 240 kale, 38 cabbage and 126 winter cauliflower French landraces from the B. oleracea genepool of INRA were assessed for resistance to clubroot caused by Plasmodiophora brassicaeWoron. Two French isolates of the pathogen (K and SJ) were used in the experiments under controlled conditions. The reaction of the 126 cauliflower accessions to naturally occurring clubroot was also evaluated in field trials. Kales exhibited considerable variation for expression of disease resistance and high levels of resistance were found in several accessions. In this group, single resistant plants were observed in most of the morphological types and from quite different geographical origins. Cabbage accessions were moderately to highly susceptible to both isolates. All cauliflower populations proved to be highly susceptible to K isolate and moderately susceptible to SJ isolate. In field trials, cauliflowers were also severely infected. Two lines selected from a resistant kale population were highly resistant against a large range of pathotypes of the pathogen. These lines presented a sufficient level of resistance to be directly useful in the breeding program in order to develop cauliflower and broccoli hybrids resistant to clubroot. Summary The Rp1 locus of maize is a complex rust resistance locus where multiple resistance genes are clustered. Rare recombination events between Rp1 genes or alleles can produce two or more detectable genes linked in coupling phase. Such compound genes can then be manipulated as a single gene in breeding programs. Several compound Rp1 genes, each carrying two or three tightly linked resistance genes, were constructed to test their utility in controlling common rust. While none of the lines carrying single Rp1 genes were resistant to all of the characterized North American P. sorghi biotypes, most of the two component and all of the three component Rp1 complexes were resistant. The potential for utilization of compound resistance genes in other crop species is discussed.  相似文献

Summary Head blight caused by Fusarium culmorum and F. graminearum is damaging in all winter rye (Secale cereale L.) growing areas. For hybrid breeding, the relative magnitude of general (GCA) and specific combining ability (SCA) is a crucial parameter for developing appropriate selection procedures. Forty single-cross hybrids were produced by crossing six and seven inbred lines of the Petkus and Carsten gene pool, respectively, in a factorial design. Hybrids were evaluated in two years with artificial F. culmorum inoculation. Resistance traits were head blight rating and grain weight relative to the non-inoculated control. Both resistance traits were closely correlated across both years (r-0.8, P=0.01). Significant genotypic variation was found for both traits with medium to high estimates of heritability (h2=0.6-0.8). Components of variance for GCA were, across years, 10 and 6 times larger than those for SCA for head blight rating and relative grain weight, respectively. Significant SCA effects were found for 15 to 20% of all cross combinations across both traits in each year. SCA effects were, however, inconsistent over years leading to a high SCA-year interaction. In conclusion, resistance to Fusarium head blight among the interpool hybrids tested was conditioned mainly by additive gene action that could be utilized by recurrent selection in multi-environment trials.Abbreviations GCA general combining ability - SCA specific combining ability  相似文献

Summary Genotypes of eastern cottonwood, Populus deltoides, from Kentucky or Mississippi were inoculated using a leaf disk assay with isolates of poplar leaf rust, Melampsora medusae f. sp. deltoidae (Mmd), from both locations to determine if pathogenic races differ at these locations. Disease severity measurements, including infection probability (number of uredia produced per uredospore), sporulation (number of uredospores/cm2), and the progeny/parent ratio (number of daughter uredia produced per mother uredium) were significantly higher when trees of southern origin were inoculated with isolates from southern locations as compared to isolates from northern locations. In addition, larger variation in disease severity measurements attributable to specific interactions, i.e. the variance due to isolates adjusted for virulence on a universal suscept, were found in northern than in southern isolates. These results suggest that isolates of southern origin have broader adaptation and are more virulent on local southern host genotypes than isolates of northern origin. Summary Accessions of Hordeum vulgare subsp. spontaneum, the progenitor of cultivated barley, were screened in field and glasshouse trials for resistance to the aphid Rhopalosiphum padi. A few selected lines were furthermore hybridized with a modern barley variety and the resulting populations evaluated. High levels of resistance were found among some of the spontaneum accessions resulting in lower aphid growth rates (maximum reduction 57%). Segregation patterns among siblings in F2 populations were continuous, indicating the presence of several genes with possibly additive effects. The usefulness of H. vulgare subsp. spontaneum for breeding aphid resistant barley is discussed.  相似文献
