Baicalein, one of the major flavones, was found to be responsible for the antioxidative activity of the traditional Chinese medicinal herb Huang-Qin ( Scutellaria baicalensis Georgi), which is widely used as an antioxidative, anti-inflammatory, and antitumor agent. The hydroxyl group of the A ring of the baicalein was alkylated at position 6 with terpenoids such as prenyl, geranyl, and farnesyl groups, and their free radical scavenging activities and glutathione (GSH) depletion capacities were examined. Their free radical scavenging activity was measured according to the 2,2'-azinobis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS(*+)) scavenging method. Baicalein and newly synthesized baicalein derivatives were found to be good free radical scavengers. Flow cytometrical method was employed to measure the intracellular antioxidative activity and GSH depletion capacity of these derivatives in human acute monocytic leukemia cell line (THP-1). It was also found that baicalein and its derivatives could decrease the levels of exogenous cumene hydroperoxide and H2O2 in THP-1 cells. These compounds also could significantly inhibit the intracellular GSH depletion induced by cumene hydroperoxide in THP-1 cells. The production of cumene hydroperoxide-induced Bax, a pro-apoptotic related protein, could also be inhibited by baicalein and its derivatives. These results suggested that baicalein and its derivatives could be beneficial to human health.  相似文献

Previously, we have found phosphopeptides (PPPs) from hen egg yolk phosvitin possess a potent antioxidative activity against oxidative stress in human intestinal epithelial cells, Caco-2. However, their biological activity at the cellular level has not yet fully understood. The objective of this study is to evaluate the regulation of glutathione (GSH) biosynthesis-associated and antioxidant enzymes against oxidative stress in Caco-2 cells using an in vitro model. Treatment of 1 mM H2O2-induced Caco-2 cells with PPPs increased cellular GSH levels, concomitant with a significant increase in gamma-glutamylcysteine synthetase (gamma-GCS) activity and the expression of gamma-GCS heavy subunit mRNA. Furthermore, intracellular glutathione reductase, glutathione S-transferase, and catalase activities were elevated by PPPs. In addition, PPPs with high content of phosphorus showed higher induction of these enzyme activities than PPPs without phosphorus. These data indicate that oligophosphopeptides from hen egg yolk phosvitin can up-regulate cellular GSH biosynthesis-associated enzymes activity and antioxidative activities, which play key roles against tissue oxidative stress in the human intestinal epithelial cells.  相似文献

While the antioxidative properties of green and black tea have been extensively studied, less attention has been given to these properties in oolong tea. The reducing powers, the 2,2-diphenyl-1-picrylhydrazyl (DPPH) scavenging activities, the amount of total phenolic compounds, the inhibitory effect on FeCl(2)/H(2)O(2) (Fenton reaction system)-induced DNA damage, and the inhibitory effect on erythrocyte hemolysis of an oolong tea water extract (OTE) were evaluated in the present study. The OTE was found to have strong antioxidative activities in all of the model systems tested. When the OTE was separated into different fractions according to molecular weight, it was found that the fractions with higher amounts of phenolic compounds (lower molecular weight) have stronger antioxidative activities. The present results support the concept that oolong tea contains several low molecular weight antioxidants that may have health promotion activities.  相似文献

The purpose of this research was to find out the effect of flour extraction rate on the antioxidative properties of traditional rye bread and then to compare the bioactive compounds content and antioxidant properties of rye breads with commercial wheat roll. Four types of rye flour with different extraction rates of 100 (whole meal dark flour), 95 (brown flour), 90 (brown flour), and 70% (light flour) originated from Warko rye cultivar were used for traditional bread baking with sourdough fermentation. Four types of the respective rye breads were analyzed for their potentially beneficial components, including tocopherols and tocotrienols, total phenolics and flavonoids, reduced glutathione, and inositol hexaphosphates. Moreover, the phenolic acids profile was provided. The Trolox equivalent antioxidant capacity (TEAC) of the breads was evaluated using free radical scavenging activities of 80% methanol extracts against ABTS*+ radical cation (ABTS radical cation decolorization method) whereas radical scavenging activity (RSA) was determined against 2,2-diphenyl-1-picrylhydrazyl radical (DPPH*). The superoxide dismutase-like activity (SOD-like activity) was evaluated as free radical scavenging activities of PBS extracts against superoxide anion radicals (O2*-). The results were compared to whole meal rye bread as well as to wheat roll taken as representative example of wheat based bakery product. The studies showed that flour extraction rates strongly affected the content of bioactive compounds and antioxidative properties of traditionally baked rye breads. The incorporation of the rye flours with extraction rates from 100 down to 70% in the formulation caused decrease in tocopherol (T), tocotrienol (T3), inositol hexaphosphate (IP6), and phenolic compound (TPC) contents in rye breads. No changes in reduced glutathione (GSH) contents were noted between each type of rye bread. A significant decrease in Trolox equivalent antioxidant capacity and radical DDPH scavenging activity was also found in bread formulated on flour with an extraction rate of 70% in comparison to the breads formulated on flour with extraction rates from 100 to 90%. The highest SOD-like activity was noted for rye bread formulated on flour with an extraction rate of 70%. The four types of rye breads showed better antioxidative properties and higher antioxidant contents when compared to wheat roll with one exception made to tocopherols and tocotrienols.  相似文献

通过水培试验研究5、10mmol·L^-1Ca^2+对不同水平镉处理下的油菜幼苗生物量、根长、丙二醛(MDA)和H2O2含量以及抗氧化酶活性的影响,利用HPLC分析外源钙对油菜幼苗镉胁迫下还原型谷胱甘肽(GSH)含量的变化情况。结果表明,镉处理浓度为150、300、450μmol·L-^1时,5和10mmol·L-1Ca^2+均能有效增加植株的生物量和根长,5mmol·L^-1Ca^2+能显著减少油菜幼苗中MDA和过氧化氢(H2O2)含量,增强超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、谷胱甘肽还原酶(GR)、谷胱甘肽过氧化物酶(GPX)和谷胱甘肽转移酶(GST)的活性及GSH的含量。镉胁迫浓度为600μmol·L-1时,施钙后油菜幼苗出现生物量低、MDA含量高、抗氧化酶活性和GSH含量均下降,与CK相比差异不显著(P〉0.05)。镉处理浓度≤450μmol·L-1时,5mmol·L^-1Ca^2+能明显缓解芥菜型油菜生长和生理所受胁迫,外源钙可作为减轻镉胁迫对油菜毒害的保护剂。  相似文献

The protective effects of hen egg yolk phosvitin phosphopeptides (PPPs) against hydrogen peroxide (H2O2)-induced oxidative stress were evaluated in an in vitro assay using human intestinal epithelial cells. Caco-2 cells were stimulated with 1 mM H2O2 for 6 h, and the secretion of IL-8, a proinflammatory mediator, was determined by ELISA as a biomarker of oxidative stress. The inhibition of H2O2-induced IL-8 secretion from Caco-2 cells was observed by pretreatment for 2 h with PPPs, but not with phosvitin. PPPs also suppressed the formation of malondialdehyde in H2O2-treated Caco-2 cells. Furthermore, intracellular glutathione levels and glutathione reductase activity were elevated by the addition of PPPs. The protective effects of PPPs against H2O2-induced oxidative stress were almost the same as that of glutathione, and PPPs with a high content of phosphorus exhibited higher protective activity than PPPs without phosphorus; however, phosphoserine itself did not show any significant antioxidative stress activity. These findings suggest that oligophosphopeptides from hen egg yolk phosvitin possess novel antioxidative activity against oxidative stress in intestinal epithelial cells and that phosphorus and peptide structure seem to have a key role in the activity.  相似文献

Compost as a soil supplement significantly enhanced levels of ascorbic acid (AsA) and glutathione (GSH) and ratios of AsA/dehydroascorbic acid (DHAsA) and GSH/oxidized glutathione (GSSG) in fruit of two strawberry (Fragaria x ananassa Duch.) cultivars, Allstar and Honeoye. The peroxyl radical (ROO(*)) as well as the superoxide radical (O(2)(*)(-)), hydrogen peroxide (H(2)O(2)), hydroxyl radical (OH(*)), and singlet oxygen ((1)O(2)) absorbance capacity in strawberries increased significantly with increasing fertilizer strength and compost use. The planting medium (compost) x fertilizer interaction for phenolics and flavonoids was significant. Fruit from plants grown in full-strength fertilizer with 50% soil plus 50% compost and 100% compost yielded fruit with the highest levels of phenolics, flavonol, and anthocyanin content. A positive relationship between antioxidant activities and contents of AsA and GSH and ratios of AsA/DHAsA and GSH/GSSG existed in fruit of both strawberry cultivars. Correlation coefficients for the content of antioxidant components versus antioxidant activity [against ROO(*), O(2)(*)(-), H(2)O(2), OH(*), or (1)O(2)] ranged from r( )()= 0.7706 for H(2)O(2) versus GSH/GSSH in cv. Allstar to r = 0.9832 for O(2)(*)(-) versus total flavonoids in cv. Allstar.  相似文献

The neuroprotective and antioxidative activities of five organosulfur compounds with a thioallyl structure (-S-CH2CH=CH2) were characterized in terms of structure-activity relationships. Among five organosulfur compounds, only S-allyl-L-cysteine (SAC) having the alanyl group (-CH2CH-NH2-COOH) and lacking the oxo (O=) group with in between molecular properties, was effective in protecting cell death induced by both oxygen glucose deprivation and global cerebral ischemia. Conversely, lipophillic organosulfur compounds including diallyl sulfide, diallyl disulfide, and diallyl trisulfide were devoid of in vitro and in vivo neuroprotective activities. Furthermore, a significant correlation was only found between the in vivo neuroprotective activity and the OH- scavenging activity (gamma = 0.55 and p = 0.032) among reactive oxygen species scavenging activities. These results indicate that the presence of the alanyl group and the absence of the oxo group are essential for the manifestation of neuroprotective activity against ischemic insults and scavenging of OH radical, with SAC surfacing as a potent neuroprotectant.  相似文献

采用蛭石栽培,在100 mmol/L NaCl 胁迫下,对耐盐性不同的两个菜用大豆[Glycine max (L.)Merr.]品种种子的过氧化氢(H2O2)含量及抗坏血酸-谷胱甘肽(AsA-GSH)循环进行了研究。结果显示,NaCl胁迫显著增加了菜用大豆种子的H2O2含量,但耐盐品种 绿领特早的增幅低于盐敏感品种理想高产95-1。NaCl胁迫期间,绿领特早种子中抗坏血酸过氧化物酶(APX)、脱氢抗坏血酸还原酶(DHAR)、谷胱甘肽还原酶(GR)活性,AsA、GSH含量以及AsA/DHA值和GSH/GSSG值的增幅高于同期的理想高产95-1,或降幅低于同期的理想高产95-1; 脱氢抗坏血酸(DHA)、氧化型谷胱甘肽(GSSG)含量的增幅低于同期的理想高产95-1。表明 绿领特早种子在胁迫期间能够保持较高的AsA-GSH循环效率,可有效地抑制H2O2的积累,这可能是其耐盐性较强的重要原因之一。  相似文献

The protective effects of amino acids against H 2O 2-induced oxidative stress were investigated in an in vitro assay using human intestinal epithelial cells. Caco-2 cells were pretreated with amino acids (1, 2, and 5 mM) for 2 h and then stimulated with 1 mM H 2O 2 for 6 h. The secretion of IL-8, a proinflammatory mediator, was determined by ELISA as an indicator of tissue oxidative stress. The inhibition of H 2O 2-induced IL-8 secretion from Caco-2 cells was observed by pretreatment with Cys, Val, Ile, Leu, Trp, His, Lys, and Ala. Cys enhanced glutathione (GSH) biosynthesis enzyme activity and increased cellular GSH levels. Branched-chain amino acids such as Val, Ile, and Leu elevated activities of GSH S-transferase (GST) and catalase. Trp, His, and Lys caused increases in GST activity. Ala enhanced GSH reductase activity. These data suggest that specific amino acids exert protective effects against tissue oxidative stress in intestinal epithelial cells based on the structure.  相似文献

为了探究H_2S对甜樱桃花器官低温伤害的缓解机理,以甜樱桃品种早大果为试材,分析了不同浓度H_2S供体NaHS对低温胁迫下甜樱桃柱头和子房AsA-GSH循环系统的影响。结果表明,喷施0.02、0.05、0.1 mmol·L~(-1)的NaHS均可降低低温胁迫下甜樱桃柱头和子房超氧阴离子(O_2~-)、过氧化氢(H_2O_2)、丙二醛(MDA)含量,显著提高还原型抗坏血酸(AsA)和还原型谷胱甘肽(GSH)含量(P0.05),降低氧化型抗坏血酸(DHA)和氧化型谷胱甘肽(GSSG)含量,使AsA/DHA和GSH/GSSG比值显著升高(P0.05),以0.05 mmol·L~(-1)NaHS效果最显著;0.02、0.05、0.1 mmol·L~(-1)的NaHS均显著提高低温胁迫下柱头和子房AsA-GSH循环中抗坏血酸过氧化物酶(APX)、脱氢抗坏血酸还原酶(DHAR)、单脱氢抗坏血酸还原酶(MDAR)、谷胱甘肽还原酶(GR)、谷胱甘肽过氧化物酶(GPX)和谷胱甘肽-S-转移酶(GST)的活性(P0.05),以0.05 mmol·L~(-1)NaHS提高上述酶活性的幅度最大。NaHS的浓度大于0.2 mmol·L~(-1)不再提高低温胁迫下AsA-GSH循环效率;低温胁迫下添加NaHS的同时添加H_2S清除剂HT可解除H_2S的效果。综上所述,喷施适量外源H_2S可有效降低低温胁迫下O_2~-、H_2O,和MDA积累,提高AsA-GSH循环效率,减轻低温对甜樱桃柱头和子房的氧化伤害。本研究结果为明确H_2S缓解甜樱桃花器官低温伤害作用机制以及生产上花期低温伤害预防提供了理论依据。  相似文献

The protective effects and the possible mechanisms of dry matter of fermented filtrate (DMF) from Antrodia camphorata in submerged culture (ACSC) on H(2)O(2)-induced cytotoxicity in HepG2 and carbon tetrachloride (CCl(4))-induced hepatotoxicity in Sprague-Dawley rats were investigated. The results showed that the inhibitory effect of DMF and its crude triterpenoids on lipid peroxidation occurred in a dose-response manner in an AAPH/linoleic acid system. When HepG2 cells were pretreated with DMF at the concentration of 0.10 mg/mL for 4 h and then induced by 1 h of treatment with H(2)O(2) (100 microM), lipid peroxidation was significantly (p < 0.05) decreased, as measured by the formation of malondialdehyde. The oral pretreatment with DMF [0.25 and 0.50 mg/kg of body weight (bw)] for 5 consecutive days prior to the administration of a single dose of 40% CCl(4) (0.10 mL/100 g of bw, ip) significantly prevented the increase in serum levels of hepatic enzyme markers (alanine and aspartate aminotransferase) and liver lipid peroxidation (p < 0.05). Histopathological evaluation of the rat liver revealed that DMF reduced the incidence of liver lesions, including neutrophil infiltration, hydropic swelling, and necrosis induced by CCl(4) in rats. Moreover, reduced glutathione (GSH)-dependent enzymes (glutathione peroxidase, glutathione reductase, and glutathione S-transferase) and the GSH/GSSG ratio were significantly improved in the oral pretreatment DMF of rats (p < 0.01). The results suggest that DMF may play a role in preventing oxidative damage in living systems by up-regulating hepatic GSH-dependent enzymes to preserve the normal GSH/GSSH ratio and scavenging free radicals formed during CCl(4) metabolism.  相似文献

1,1-Diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity of Japanese whiskey after various aging periods in oak barrels was measured to evaluate the antioxidative effects of whiskey. The activity of the whiskey increased with the aging period with high correlation. The activity of various types of whiskey was measured and shown to be correlated to the potentiation of the GABAA receptor response measured in a previous paper. However, the fragrant compounds in the whiskey which potentiated the GABAA receptor response had low DPPH radical scavenging activity, while phenol derivatives had high radical scavenging activity. The whiskey was extracted by pentane. The aqueous part showed the scavenging activity, whereas the pentane part did not. Thus, both the DPPH radical scavenging activity and the potentiation of the GABAA receptor response increased during whiskey aging in oak barrels, but were due to different components. The whiskey protected the H2O2-induced death of E. coli more than ethanol at the same concentration as that of the whiskey. The changes that occurred in the whiskey during aging may be the reason aged whiskies are so highly valued.  相似文献

Chlorophylls (Chl's) are the most abundant natural plant pigments. Four chlorophyll-related compounds (CRCs), including chlorophyllide a and b (Chlide a and b) and pheophorbide a and b (Pho a and b), were investigated for their antioxidative capacities to protect human lymphocyte DNA from hydrogen peroxide (H(2)O(2)) induced strand breaks and oxidative damage ex vivo. Lymphocytes exposed to H(2)O(2) at concentrations of 10 and 50 microM revealed an increased frequency of DNA single-strand breaks (ssb's; as measured by the comet assay) and also an increased level of oxidized nucleoside (as measured by 8-hydroxydeoxyguanosine, 8-OHdG). All Chl's reduced the level of DNA ssb's and 8-OHdG within human lymphocytes following exposure to 10 microM H(2)O(2). Only Pho a and b were able to decrease DNA ssb's and 8-OHdG following treatment of lymphocytes with 50 microM H(2)O(2), in a concentration-dependent fashion. It was demonstrated herein that Pho a and b were more antioxidative than others. We applied DPPH free-radical scavenge assays in vitro, and got similar results. Pho a and b had higher ability in scavenging capacities than others. We conclude that water-extract Chl's are able to enhance the ability of human lymphocytes to resist H(2)O(2)-induced oxidative damage, especially for Pho a and b.  相似文献

This study aimed at evaluating the antioxidative activity of crude hsian-tsao leaf gum extracted by sodium bicarbonate solutions and precipitated by 70% ethanol. The antioxidative activities, including the radical-scavenging effects, Fe(2+)-chelating ability, and reducing power as well as the inhibition of FeSO(4)-H(2)O(2)-induced malondialdehyde formation in rat tissue homogenate were studied in vitro. It was found that the antioxidative effect provided by hsian-tsao leaf gum was strongly concentration dependent. In general, the antioxidative activity increased with increasing gum concentration, to a certain extent, and then leveled off with further increase in gum concentration. A concentrtaion-dependent kinetics for the rate of change in antioxidative activity was proposed. The antioxidative activity constant (k) and the half-inhibition concentration (IC(50)) for each antioxidative reaction studied were calculated. From a comparison of the IC(50) values for different antioxidative reactions, it seemed that hsian-tsao leaf gum was more effective in scavenging superoxide radicals than chelating Fe(2+) or scavenging alpha,alpha-diphenyl-beta-picrylhydrazyl (DPPH) radicals. As compared to the commercial antioxidants, hsian-tsao leaf gum showed less scavenging effect on the DPPH radical and reducing power but better superoxide radical-scavenging effect and Fe(2+)-chelating ability than alpha-tocopherol and BHT.  相似文献

The inhibitory effects of five tea polyphenols, namely theaflavin (TF1), theaflavin-3-gallate (TF2), theaflavin-3,3'-digallate (TF3), (-)-epigallocatechin-3-gallate (EGCG), and gallic acid, and propyl gallate (PG) on xanthine oxidase (XO) were investigated. These six antioxidant compounds reduce oxidative stress. Theaflavins and EGCG inhibit XO to produce uric acid and also act as scanvengers of superoxide. TF3 acts as a competitive inhibitor and is the most potent inhibitor of XO among these compounds. Tea polyphenols and PG all have potent inhibitory effects (>50%) on PMA-stimulated superoxide production at 20 approximately 50 microM in HL-60 cells. Gallic acid (GA) showed no inhibition under the same conditions. At 10 microM, only EGCG, TF3, and PG showed significant inhibition with potency of PG > EGCG > TF3. The superoxide scavenging abilities of these six compunds are as follows: EGCG > TF2 > TF1 > GA > TF3 > PG. PG was the most potent inhibitor of PMA-stimulated H(2)O(2) production in HL-60 cells. The order of H(2)O(2) scavenging ability was TF2 > TF3 > TF1 > EGCG > PG > GA. Therefore, the antioxidative activity of tea polyphenols and PG is due not only to their ability to scavenge superoxides but also to their ability to block XO and related oxidative signal transducers.  相似文献

【目的】 叶绿体是植物体产生活性氧 (ROS)、且对盐最敏感的细胞器,本试验研究了外源壳聚糖对 NaCl 胁迫下菜用大豆[Glycine max (L.) Merr.]叶绿体内抗氧化系统的影响,以期探讨壳聚糖对 NaCl 胁迫下光合作用的调节机制。 【方法】 试验于 2014 年 4 月至 6 月在内蒙古民族大学试验基地日光温室内进行。采用蛭石栽培,选用菜用大豆盐敏感品种 ‘理想高产 95-1’ (LX)、耐盐品种‘绿领特早’ (LL)为试材。试验设 4 个处理:1) 叶面喷洒清水,根部浇灌营养液 (对照);2) 叶面喷洒壳聚糖溶液,根部浇灌营养液;3) 叶面喷洒清水,根部浇灌溶有 NaCl 的营养液;4) 叶面喷洒壳聚糖溶液,根部浇灌溶有 NaCl 的营养液。 【结果】 外源壳聚糖显著降低了 NaCl 胁迫下两品种菜用大豆叶绿体 H2O2的含量,显著提高了过氧化物酶 (POD)、抗坏血酸过氧化物酶 (APX)、单脱氢抗坏血酸还原酶 (MDHAR)、脱氢抗坏血酸还原酶(DHAR)、谷胱甘肽还原酶(GR)、谷胱甘肽过氧化物酶 (GPX)活性以及胁迫中期还原型抗坏血酸 (AsA)、还原型谷胱甘肽 (GSH)含量;与盐敏感品种 LX 相比,耐盐品种 LL 在胁迫 6~15 d 期间维持了相对较低的 H2O2含量,相对较高的 DHAR 活性及 AsA 含量,在整个胁迫期间维持了相对较高的 APX、GR、GPX 活性,在胁迫后期(12 d、15 d)维持了相对较高的 GSH 含量。 【结论】 外源壳聚糖对 NaCl 胁迫下菜用大豆叶绿体内 POD 活性及 AsA-GSH 循环产生了显著诱导作用,但对不同品种的诱导效果不同,耐盐品种 LL 的 AsA-GSH 循环维持了相对较强的活性氧清除能力,这可能是其维持较强光合能力,进而维持较旺盛生长的重要原因之一。   相似文献

In this study, the free-radical-scavenging properties of anthocyanin oligomers for 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical, alkyl radical, and hydroxyl radical were evaluated using electron spin resonance (ESR) spectroscopy. The DPPH radical, alkyl radical, and hydroxyl radical scavenging activity of anthocyanin oligomers increased in a dose-dependent manner, with the 50% inhibitory concentration (IC??) value of 13.0, 14.0, and 448.0 μg/mL, respectively. The inhibitory effect of anthocyanin oligomers on lipid peroxidation was examined with ferric thiocyanate (FTC) and thiobarbituric acid (TBA). The inhibitory activity of anthocyanin oligomers was found to be comparable to that of vitamin E. In addition, anthocyanin oligomers enhanced the activities of superoxide dismutase (SOD, EC 1.15.1.1), catalase (CAT, EC 1.11.1.6), glutathione peroxidase (GPx, EC 1.11.1.9), and glutathione-S-transferase (GST, EC 2.5.1.18) in ARPE-19 cells. In addition, anthocyanin oligomers inhibited the H?O?-induced G2/M phase arrest in ARPE-19 cells.
