Three methods not reported previously to determine flooding tolerance, and a visual scoring system developed for this study, were utilized to evaluate the effects of soil waterlogging upon 3 bean genotypes empirically differing in tolerance. These methods consisted of: (1) Triphenyl Tetrazolium Chloride Reduction Method (TTC), which measures flooding tolerance by determining the rate of root respiration during stress; (2) Electrical Conductivity Method (EC), in which the degree of root cell membrane integrity is determined; and (3) Pressure Chamber Method (PC), by which the degree of plant desiccation is found by measuring the water potential of the xylem. A visual scoring method based on the external appearance of the plant was also used. The agreement among methods was generally good. The TTC and EC methods were more time-consuming and results more variable, especially from the latter. Results of the PC and visual scoring methods were highly correlated (r=0.85). However, TTC and EC required only 7-day-old seedlings, thus allowing the screening of large numbers of samples and the efficient use of space. The F1 and F2 progeny from crosses between the most tolerant genotype (P074) and the most sensitive genotype (P149) indicated that differences in reaction to flooding are probably heritable.  相似文献

To evaluate the effect of maturity and storage temperature on the development of the physiological disorder peteca, lemons cv. Eureka were harvested from a grove with northern exposure located in Mallarauco (33°45′ Latitude South) Metropolitan Region of Chile, at 2 levels of maturity (silver or yellow). After storage for 60 d at 3 or 7 °C, under 90% R.H., fruits were evaluated visually for incidence of epidermal and subepidermal peteca. At that time, the concentration of galacturonic acid and degree of methylation were measured in the albedo, and calcium, hydrogen peroxides and total oxalate concentrations, peroxidase and polyphenoloxidase (PPO) activities were determined in the juice and albedo. Yellow lemons at 3 °C developed more epidermal peteca than fruit stored at 7 °C, and that silver fruit stored at 3 and 7 °C, with these last lemons presenting a higher pectin quality than fruit stored at 3 °C. Maturity and temperature interacted significantly on the concentrations of peroxide and calcium in the albedo, which in average were much higher than in the juice, while for PPO and peroxidases, this interaction was highly significant both in juice and albedo, with higher activity in the albedo. These biochemical evaluations suggest that peteca is a result of some kind of stress on the fruit. It is concluded that yellow lemons are more prone to develop peteca than silver fruit. They will develop greater incidence of the disorder when stored at temperatures as low as 3 °C for 60 d, and that this condition affects the degree of methylation in the albedo, which would end up altering the quality of pectins.  相似文献

The Fraxinus genus consists of ca. 65 deciduous, rarely evergreen trees found mainly throughout Europe, Asia and North America. Fraxinus species possess the aesthetic characteristics desired of urban trees (autumn colour, attractive bark and flowers); however, no scientifically based tolerance ranking exists for many genotypes within this genera. Aims of this study were to identify whether differences in drought tolerance exist within the Fraxinus genus and thereby provide information as to their usefulness for planting in urban landscapes where water stress poses a threat to transplant survival. At the cessation of a 2-week drought stress and subsequent recovery period, marked differences in drought tolerance among genotypes were distinguished based on net photosynthesis, leaf chlorophyll fluorescence and chlorophyll concentration. In addition, measurement of light absorbance, light trapping, electron transport and dissipation fluxes per leaf cross section of photosystem II at the cessation of the drought period provided an insight into mechanisms of drought damage and resilience among the genotypes tested. The results of this investigation indicate that F. excelsior ‘Aurea Pendula’, F. nigra Marsh., F. ornus L., F. angustifolia ‘Raywood’, F. excelsior L. and F. excelsior ‘Jaspidea’ should be considered for urban planting sites, where water supply is limited. Care should be taken when planting F. americana L., F. americana ‘Autumn Purple’ and F. velutina Torr., into urban environments as results indicated these species are sensitive to drought.  相似文献

Leaf structural adaptations for the reduction of water loss were examined in two olive (Olea europaea L.) cultivars (Chemlali and Chétoui) growing under water stress conditions. Leaf measurements included leaf tissue thickness, stomatal density, trichome density, specific leaf area, leaf density, water relations, and gas exchange. We found considerable genotypic differences between the two cultivars. Chemlali exhibited more tolerance to water stress, with a thicker palisade parenchyma, and a higher stomatal and trichome density. Chemlali leaves also revealed lower specific leaf area and had higher density of foliar tissue and lower reduction in net CO2 assimilation rate. The mechanisms employed by these two cultivars to cope with water deficit are discussed at the morpho-structural level. The morphological and structural characteristics of the leaves are in accordance with physiological observations and contribute to the interpretation of why the olive cv. Chemlali is more drought-tolerant than cv. Chetoui. Furthermore, from the behaviour of Chemlali plants we consider this cultivar very promising for cultivation in semi-arid areas.  相似文献

Relatively few studies have investigated mistletoe infestation of tree species in urban settings, and even fewer considered infestation of alien tree species. Here, mistletoe infestation incidence and intensity were studied in Durban, South Africa. The relationship of infestation to tree CBH and height was examined in detail for the mistletoe species Erianthemum dregei. Although E. dregei is known to have a wide range of host taxa, we report E. dregei infestation on 27 new host species for the first time. E. dregei occurred on 30 host species with a preference for the alien host tree Melia azedarach in our study area. The relationship between infestation and CBH indicated that the trees (irrespective of species) of ≥200 cm were more susceptible to the mistletoe attack. The implications of this pattern, as well as those of high host diversity and especially of infestation of alien trees are discussed in a conservation perspective.  相似文献

A protocol was developed for direct shoot and plantlet regeneration from in vitro regenerated leaf explants of male Pistacia vera L. cv. ‘Atl?’. Leaves excised from axenic shoot cultures of pistachio were used to induce organogenesis on a Murashige and Skoog (MS) medium with Gamborg vitamins supplemented with combinations of different concentrations of BAP and IAA. The highest adventitious shoot regeneration in 35% of the explants, with the number of shoots ranging from 2 to 3 per explant, occurred in the explants cultured during the establishment phase in the medium with 1 mg l−1 IAA and 2 mg l−1 BAP. For shoot multiplication, the highest number of new microshoot/explants (5.76) was obtained in a culture medium supplemented with 1 mg l−1 BAP, but it was not significantly different from the number obtained at 2 mg l−1 BAP. A high rooting frequency (84%) for microshoots was recorded on a medium supplemented with 2 mg l−1 IBA. In vitro rooted plantlets were transferred to pots filled with a mixture of soil, sand and peat (1:1:1). They were weaned in a growth room and finally moved to a greenhouse. This protocol could be utilized for in vitro clonal propagation of this economically important plant.  相似文献

The response of 3-year-old grapevines (Vitis vinifera L. cultivar ‘Thompson Seedless’) to furrow and drip irrigation was quantified in terms of water status, growth, and water use efficiency (WUE). Drip irrigation was applied daily according to best estimates of vineyard evapotranspiration while furrow irrigations were applied when 50% of the plant available soilwater content had been depleted. Drip and furrow irrigated vines showed similar water status (midday leaf water potential, Ψ1) and shoot growth patterns throughout the season. Dry weight partitioning was not significantly different between treatments but root mass was somewhat larger for the furrow than drip irrigated vines. Nitrogen concentrations of the fruit and roots were significantly (P < 0.05) less for the drip irrigated vines when compared with the furrow treatment. Similar WUE (kg water kg−1 fresh fruit wt.) were obtained for both treatments indicating that furrow irrigation was as efficient as drip irrigation under the conditions of this study. The data indicate that drip irrigation may increase the potential for control of vine growth by making vines more dependent on irrigation and N fertilization than furrow irrigation.  相似文献

The application of de-icing salts for winter road maintenance is recognized as a major contributor to the decline of urban trees. We conducted a long-term monitoring program across several locations in the City of Edmonton (Alberta, Canada) to evaluate the impact of roadway salt application on tree species widely planted in boulevards and right-of-ways: Ulmus americana, Fraxinus pennsylvanica, Pinus contorta, and Picea glauca. Soil and leaf samples were collected from a total of 16 sites over six years. There were four sites selected for each tree species: three mid- to high- traffic roadside sites that received regular winter maintenance and one non-serviced site (control). Sampling was performed three times per year from late spring to late summer. Airborne salinity was assessed in four locations at different distances from the road. In 50% of the roadside sites, soil electrical conductivity (EC) values exceeded 2 dS m−1. Soil pH in all of the roadside sites was also significantly higher than in the control sites, with values ranging from 7.6 to 8.5. In all four species, trees growing in sites with high soil EC had increased leaf Na concentrations and reduced leaf chlorophyll concentrations. Among the airborne monitoring sites, Na deposition in high traffic locations was over four-fold higher than those measured in the control location. Furthermore, Na levels remained relatively high at 20–50 m from the main road. Our data suggest that while soil salinity is among the main stressors affecting roadside trees in Edmonton, salt spray deposition may also have a significant impact on trees located close to high vehicle traffic areas and dense road networks. Our study highlights the importance of collecting data over several years and from multiple locations to account for the spatial and temporal heterogeneity of the urban environments in order to better evaluate the impact of road salt application on urban trees.  相似文献

Variations in morpho-anatomy, chemical composition, gas exchange and stem water potential (Ψstem) were studied in leaves of Prunus avium L. cultivars with different growth habit: Burlat and Summit (upright, dense canopies), and Van (spreading, open canopy). Light transmittance was highest in Van canopies. Therefore, Van allowed an easier penetration of the visible radiation through the canopy, which improves photosynthetic rates, especially at harvest. Conversely, no differences in Ψstem were found among cultivars. Van showed the highest leaf mass per unit area and variations in this parameter were mainly associated with alterations in leaf thickness than in leaf density. Leaves of Summit and Van were thicker than those of Burlat, mainly due to increased palisade and spongy parenchyma thickness. However, Burlat leaves had the highest palisade/spongy ratio. Total chlorophyll concentration per area and the chlorophyll a/b ratio were always higher in Van leaves than in the other two cultivars. In contrast, Van leaves had the lowest concentrations of soluble carbohydrates and starch. In dense canopies of Summit and Burlat, light reduction caused a notable increase in the relative concentration of total phenols. The above results provide useful information about the biological behaviour of sweet cherry cultivars with open and dense canopies in order to help the agronomical decisions regarding the local environmental conditions and the consumers’ preference. In particular, it is important to manipulate factors such as orchard exposure, row orientation, plant spacing, training system and pruning, which are able to reduce shading in the canopy.  相似文献

Non-destructive and mathematical approaches of modeling can be very convenient and useful for plant growth estimation. To predict individual leaf area, fresh weight, and dry weight of a cucumber (Cucumis sativus L.), models were developed using leaf length, leaf width, SPAD value, and different combinations of these variables. Eight regression equations, commonly used for developing growth models, were compared for accuracy and adaptability. The three nonlinear models developed were as follows: individual leaf area (LA) = −210.61 + 13.358W + 0.5356LW (R2 = 0.980***), fresh weight (FW) = −2.72 + 0.0135LW + 0.00022LWS (R2 = 0.956***), and dry weight (DW) = 0.25 − 0.00102LS + 0.000077LWS (R2 = 0.956***), where L is the leaf length, W the leaf width, S the SPAD value, and LWS = L × W × S. For validation of the model, estimated values for individual leaf area, fresh weight, and dry weight showed strong agreement with the measured values, respectively. Leaf dry weight, especially, was estimated with a higher degree of accuracy through the use of a SPAD value, as well as leaf length and width. Therefore, it is concluded that models presented herein may be useful for the estimation of the individual leaf area, fresh weight, and dry weight of a cucumber with a high degree of accuracy.  相似文献

The temperature induced changes in chlorophyll fluorescence induction parameters (F0, Fm, Fv and their ratios) measured by a PEA analyzer were used to screen for heat-tolerance 12 cultivars and lines of common bean (Phaseolus vulgaris L.) available in the germplasm collections of Maritsa Vegetable Crops Research Institute, Plovdiv, and Institute of Plant Genetic Resources, Sadovo, Bulgaria. The experiments were carried out in a selection field of Maritsa Institute. The measurements were made on six different dates during the bean reproductive period (in June–July) when the temperatures were approximately 26 °C in the morning to and 42 °C at noon. Cucumber chilling-resistant cultivar Changchun mici and -sensitive cultivar Beijing jietou were used in this study to investigate the effects of exogenous PAs on protection against chilling injury as well as on changes of physiological features, and the fluctuation of free PAs content in the leaves under chilling stress. Upon chilling treatment, free spermidine (Spd), spermine (Spm) and putrescine (Put) were remarkably induced in the leaves of cv. Changchun mici 1 day after treatment. The induction of Put declined thereafter, whereas Spd and Spm levels increased steadily. In the leaves of cv. Beijing jietou, Put content was increased only at 1 day after chilling while Spd content decreased significantly upon chilling treatment. Chilling reduced soluble protein content, and decreased the activities of antioxidant enzymes, including superoxidase dismutase (SOD), peroxidase (POD), catalase (CAT) and ascorbate peroxidase (APX) only in Beijing jietou. However, these changes could be renovated by exogenous application of Put and Spd. It was also found that pretreatment with Put and Spd diminished the increased electrolyte leakage and malondialdehyde (MDA) content caused by chilling in the leaves of both cultivars. Pretreatment of methyglyoxal-bis-(guanylhydrazone) (MGBG), the PAs biosynthetic inhibitor cancelled the effects of PAs in most of the treatments. Moreover, histochemical staining and quantitative measurements showed that exogenous application of Put and Spd eliminated but MGBG exaggerated the hydrogen peroxide (H2O2) accumulation caused by chilling stress, especially in leaves of Beijing jietou. Interestingly, Changchun mici was found to contain higher endogenous free PAs contents compared to Beijing jietou. While no significant difference of SOD, POD and CAT activities was found between non-chilling Changchun mici and Beijing jietou seedlings, the former exhibited higher APX activity than the latter. These results suggest that PAs play important roles in the tolerance of cucumber against chilling stress, which is most likely achieved by acting as oxidative machinery against chilling injury.  相似文献

Six enzyme systems were surveyed in 12 peach cultivars using polyacrylamide gel electrophoresis of leaf extracts. Variation between cultivars was observed in four enzymes: peroxidase, isocitrate dehydrogenase, acid phosphatase and alkaline phosphatase. Genetic variation parameters revealed that the maximum proportion of polymorphic loci was 46.1%. Out of 15 loci studied, 9 were monomorphic and 6 were polymorphic for 20 alleles. A dendrogram showed the grouping of cultivars on the basis of genetic distance values. The use of isozyme electrophoresis to find the inter-relationship of cultivars is discussed.  相似文献

There has been recent market interest in “baby” salad greens. However, little information exists on the nutritional differences between immature “baby” greens and produce traditionally sold at the fully mature stage. Kale (Brassica oleracea L. var. acephala D.C.) contains high levels of lutein and β-carotene, which possess important human health properties. Kale was grown in a controlled environment and pigments were measured in young (<1 week), immature (1–2 weeks), mature (2–3 weeks), fully developed (3–4 weeks) and senescing (>4 weeks) leaves using high-performance liquid chromatography (HPLC). Significant differences were observed for all pigments during leaf development. Pigment accumulation followed a quadratic trend, with maximums occurring between the 1st and 3rd week of leaf age. The highest concentrations of lutein measured 15.1 mg/100 g fresh mass and occurred in 1–2 week old leaves. The remaining pigments reached maximum levels at 2–3 weeks (β-carotene at 11.6 mg/100 g; chlorophyll a at 251.4 mg/100 g; and chlorophyll b at 56.9 mg/100 g fresh mass). Mature fully expanded kale leaves accumulated higher carotenoid concentrations than immature or “baby” leaves, with senescent leaves having the lowest carotenoid concentrations. Harvesting kale leaves at a mature stage of development resulted in maximum carotenoid values. Cultural management practices that increase carotenoid concentrations would be expected to improve nutritional quality for fresh markets.  相似文献

Three nectarine (Prunus persica var. Nectarina Maxim.) cultivars grown under solar-heated greenhouse and open-field in northwest China, were tested to evaluate their photosynthetic and chlorophyll fluorescence response to both growth conditions, and whether nectarine plants acclimate to the solar-heated greenhouse growth condition. Comparisons of light-saturated photosynthetic capacity (Amax) and CO2-saturated photosynthetic capacity (RuBPmax) indicated that each cultivar (Z, Zao-Hongzhu; H, Hua-Guang; Y, Yan-Guang) maintained similar rates of light-saturated and CO2-saturated carbon assimilation when grown in both conditions. The curve of diurnal variation of net photosynthetic (PN) rate showed double peaks in open-field but single when grown in greenhouse. Compared with open-field-grown plants, a significant increase of daily average PN was found in Z but decreased in Y in greenhouse. The diurnal variation of Fv/Fm indicate that plants grown in greenhouse experience less photoinhibition than in open-field condition. A reduction in chlorophyll (chl) a/b ratio in leaves of greenhouse grown plants with significant increase in chlorophyll (chl) b content were obtained. The results suggest that some nectarine cultivars have the ability to acclimate to the solar-heated greenhouse growth condition.  相似文献

In vitro development of isolated embryos and axillary bud proliferation were studied in Pistacia vera L. Different regulator-free nutrient media were compared to determine the effects of the mineral solution, agar and sucrose concentrations on seedling development from mature embryos. Nutrient-rich MS [Murashige, T., Skoog, F., 1962. A revised medium for rapid growth and bioassays with tabacco tissue cultures. Physiol. Plant. 15, 473–479] and DKW [Driver, J.A., Kuniyuki, A.M., 1984. In vitro propagation of Paradox walnut rootstock. HortScience 19, 507–509] mineral solutions were more efficient for the development of aerial parts than nutrient-poor KN [Knop, W., 1884. Bereitung einer concentrierten nährstofflosung für pflanzen. Landwersuhssat 30, 292–294] and WT [Withe, P.R., 1936. Plant tissue cultures. Bot. Rev. 2, 419–437] solutions. Reducing the agar concentration enhanced fresh matter production and balanced seedling development. When tested at different concentrations, sucrose was found to orient mature embryo development, with the best results obtained at concentrations of 2–4%, whereas high concentrations (6 and 12%) mainly inhibited elongation of the aerial parts. Plantlets obtained previously from in vitro cultured embryos were propagated by axillary budding. High bud proliferation (six shoots per explant) was achieved when using 17.8 μM benzyladenine (BA) combined with 0.5 μM indole-3-butyric acid (IBA). The addition of 2.9 μM gibberellic acid (GA3) to the propagation medium did not improve axillary shoot yields and on average, media with GA3 produced 2.3–2.6 elongated stems per cultured explant. Shoots were rooted in vitro in half-strength MS medium containing 12.3 μM IBA.  相似文献

The changes in chlorophyll a fluorescence, caused by high temperature (HT), have been used to study heat-tolerance in nine common bean (Phaseolus vulgaris L.) accessions (3 cultivars and 6 lines) available in the gene banks of Maritsa Vegetable Crops Research Institute and Institute for Plant Genetic Resources. The plants were grown in controlled greenhouse conditions until the blossoming stage, treated with HT (45° C) for 2 h and returned for recovery for 4 h at 23° C. The obtained results allowed us to differentiate four groups, among the studied bean plants, by their response to HT. The first group, includes a single cultivar – ‘Secuntsa’. The accession was considered heat sensitive, because it showed a decrease of its total performance index (PItotal), calculated by JIP-test, during HT stress and did not recover to the initial values. The second group, comprised of lines RH13, BBSR17, BBSR28, and ‘Starozagorski cher’, expressed an increase in PItotal during heat stress, which however was followed by a decline in PItotal values during recovery after HT treatment. The third group – RRR46 displayed a decrease in various JIP-test parameters during HT treatment followed by full recovery after returning the plants to 23° C. The accessions from the fourth group RH26D, ‘Ranit’, similarly to the control heat tolerant line 83201007 did not show significant alteration in PItotal. We assumed that the accessions from the fourth group are heat-tolerant. The line RRR46 is also promising in terms of heat tolerance because of its flexible response to HT treatment. These genotypes will be used in our further breeding program.  相似文献

Horse chestnut (Aesculus hippocastanum L.) is a common urban tree species in Ljubljana, the capital of Slovenia. This area is forecast to experience a general reduction in precipitation and an increase in temperature, which increases water demand in plants. Because A. hippocastanum is known for its drought vulnerability, the question of the future suitability of this urban tree species in Ljubljana has arisen. To investigate how climate has influenced A. hippocastanum radial growth and how trees responded to extreme climatic events, standardized precipitation-evapotranspiration index (SPEI) was used as a proxy for water demand. Climatic signal and its stability through time were calculated using Pearson’s correlation coefficient. Additionally, to investigate whether the trees had a common response to extreme climatic events, pointer years were calculated using Cropper values. We sampled 19 trees that were growing in Tivoli Park in Ljubljana. After successful cross-dating of 15 trees, the ring count showed that the trees had up to 201 tree-rings and had 130 on average. Climate-tree growth analysis showed that in July, 3-month SPEI had the strongest influence on radial growth, but its influence on radial growth decreased over time, possibly due to the die-off process of trees. The narrowest tree-rings were a result of unusually dry periods at the time of cambium activity and/or new cell growth. With the forecast of longer, more frequent summer drought periods in Ljubljana, soil moisture stress will increase, and as a result, a decrease in radial tree growth of A. hippocastanum trees from Tivoli Park is expected.  相似文献

用色差计和叶绿素仪对白菜亚种3个变种(普通白菜、乌塌菜和薹菜)19个品种的叶片颜色和叶绿素含量进行测定,并对色泽参数进行了相关分析和聚类分析。结果表明,供试品种间叶绿素含量及色泽参数差异显著;色泽比(h)和饱和度(C)与叶绿素含量呈一定相关性。聚类分析将供试品种分为叶色较浅和叶色较深两类;绿秀、PAK、矮脚黄、塌地乌3号、精品塌地乌与日本品种夏帝、华冠、冬赏味颜色相近。用色差计检测白菜叶片颜色方法可行,并具有操作简单、成本低、时间短、无损伤等优点。  相似文献
