Congenital bilateral renal hypoplasia in Large White pigs. 总被引:1,自引:0,他引:1
Bilateral renal hypoplasia and/or agenesis was detected in 6 of 26 pure-bred Large White piglets in 2 consecutive litters. The affected piglets were all dead at birth. The defect was characterised by a variable nephrogenic zone beneath the renal capsule and by a greatly reduced number of glomeruli which were hypertrophied. The piglets were born to the same sow but they were sired by different boars. Both boars and the sow were related through their pedigree. A genetic basis for the defect, probably involving a recessive gene defect inherited in a simple Mendelian manner, seems likely. 相似文献

Neuropeptide Y (NPY) provides an important hypothalamic link between nutritional status and neuroendocrine mechanisms regulating growth and reproduction. The objective of the following series of experiments was to determine the effects of single or continuous administration of NPY on secretion of luteinizing hormone (LH) and (or) growth hormone (GH). In experiment 1, four ovariectomized (OVX) ewes and four OVX + estrogen-treated ewes each received, in a 4 x 4 Latin Square arrangement of treatments, a single injection of 0, 0.5, 5, or 50 microg NPY via an intracerebroventricular (i.c.v.) cannulae to determine the effects on secretion of GH. NPY significantly elevated serum GH at the 50 microg dose regardless of estrogen exposure (P = 0.003). In experiment 2, eight OVX ewes were infused i.c.v. with NPY or saline (n = 4/trmt) continuously for 20 h in a linearly increasing dose, ending at 50 microg/h NPY. Blood samples were collected via jugular cannulae every 10 min during hour -4-0 (interval 1, pre-treatment), hour 6-10 (interval 2) and hour 16-20 (interval 3) relative to the initiation of infusion (0 h). Mean LH and LH pulse frequency were lower in NPY- versus saline-infused ewes during intervals 2 and 3 (P < 0.01), but NPY had no discernable effect on serum GH (P > 0.10). In experiment 3, four OVX ewes were continuously infused with NPY as in experiment 2, except that the maximum 50 microg/h dose was achieved after only 10 h of infusion. Blood samples were collected every 10 min, beginning 4 h before and continuing until 4h after the NPY infusion. Mean serum LH changed significantly over time (P = 0.0001), decreasing below pre-treatment levels by hour 3 of NPY infusion (P < 0.01), and returning to pre-treatment concentrations following the end of infusion (P > 0.15). Serum GH also changed significantly over time (P < 0.001). Mean GH levels tended to be greater than pre-treatment levels by hour 2 of infusion (P < 0.08), but thereafter returned to basal levels. Serum GH also increased following the end of NPY infusion (P < 0.03). From these data we conclude that NPY exerts a persistent inhibitory effect on secretion of LH, and may stimulate the secretion of GH during the initiation and cessation of infusion of NPY. These observations support a role for NPY in mediating the effects of undernutrition on both LH and GH, and also provide evidence for potential mechanisms by which leptin, acting through NPY, may stimulate the secretion of GH. 相似文献

The effect of intramuscular injection of naloxone, an antagonist of opiatergic actions, on serum concentrations of luteinizing hormone (LH) and ovarian activity in prepuberal ewes was investigated. Naloxone stimulated release of LH over a period of approximately 3 h. Subsequently, serum concentrations of LH did not differ from controls. Serum concentrations of estradiol-17β were elevated 8 h after treatment with naloxone. Ewe lambs did not ovulate. Endogenous opioid peptides may be involved in the regulation of secretion of LH in immature female sheep. 相似文献

Experiments were conducted to characterize the ability of the antiestrogen enclomiphene (ENC) to block the effects of estradiol on secretion of LH in ovariectomized ewes. To determine whether ENC could block an estradiol-induced LH surge, ewes (n = 4/group) were administered 10 to 250 mg ENC followed 30 min later by 25 micrograms estradiol. Ten or 25 mg ENC suppressed the estradiol-induced LH surge in one of four ewes, whereas 100- or 250-mg doses suppressed the LH surge in three and four of four ewes, respectively. In ewes that received a single treatment of 100 mg ENC plus 25 micrograms estradiol, serum concentrations of LH remained below 1 ng/ml for 3 wk. Compared with untreated ewes, the number of pituitary GnRH receptors was elevated (P less than .05) at 12 d and 28 d, but pituitary content of LH had decreased (P less than .05) by 28 d in ewes treated with 100 mg ENC. To determine whether ENC could block the inhibitory effects of estradiol on serum concentrations of LH, ewes received injections of .03, .1, 1 or 10 mg ENC every 4 d. Half the ewes treated with each dose also received estradiol implants. Injection of .03, .1 or 1 mg ENC alone did not affect serum concentrations of LH, whereas the 10-mg dose decreased serum concentrations of LH below 1 ng/ml by wk 1 of treatment. No dose prevented the inhibition of serum concentrations of LH caused by estradiol implants. The secretion of luteinizing hormone in ewes of Finnish Landrace during estrus. Acta vet. scand. 1979, 20, 216–223. — Luteinizing hormone immunoreactivity was measured in the venous plasma of four cycling Finnish Landrace sheep during the breeding season in connection with one synchronized estrus and the subsequent one. The ewes were slaughtered after the second estrus to establish the number of ovulations. To determine the LH concentration, a heterologous method of assay was used; this was based on the cross reaction of sheep plasma LH in a human LH radioimmunoassay system.As a result of the investigation, it was found that the peaks of LH were lower during the time of synchronized estrus and that these peaks occurred earlier than in the subsequent estrus. However, the differences were not statistically significant. On account of the limited material, the effect of the occurrence of the LH peak on the number of ovulations could not be established. 相似文献

Ovariectomized (OVX) ewes were injected with 5 ml of either bovine serum, charcoalextracted bovine follicular fluid (FF), or whole bovine FF. Five hours after this pretreatment, ewes on each pretreatment were injected with either 0, 1, or 5 μg of GnRH. Ewes that were pretreated with either type of FF had decreased concentrations of FSH regardless of dose of GnRH when compared to ewes pretreated with bovine serum. There was no effect of charcoal extraction. There were no differences among the pretreatment groups in LH response to GnRH. In a second experiment, OVX ewes were pretreated (4 ml) with either bovine serum or bovine FF 5 hr prior to GnRH or with bovine FF 42, 30 and 18 hr prior to GnRH. Ewes were injected with either 0 or 5 μg of GnRH. Pretreatment with FF for 5 or 42 hr prior to GnRH resulted in significantly decreased concentrations of FSH both at the time of GnRH treatment and during the following 2 hr. Concentrations of LH did not differ among pretreatment groups. In a third experiment, OVX ewes were pretreated with either bovine serum or bovine FF 30, 18 and 5 hr prior to GnRH. Ewes were injected with either 0, 5 or 50 μg of GnRH. Pretreatment with FF resulted in decreased concentrations of FSH both at the time of GnRH treatment and during the following 2 hr. A possible role for endogenous opioid peptides (EOP) in the control of luteinizing hormone (LH) and prolactin (PRL) secretion was studied by injecting the opioid antagonist, naloxone (NAL), into postpartum ewes and cows. Twelve ewes that lambed during the fall breeding season and nursed their lambs were injected iv with NAL (1.0 mg/kg) on d 10, 14, 18, 22 and 26 postpartum. Blood samples were collected at 15-min intervals from 2 h before to 2 h after NAL, and serum concentrations of LH and PRL were quantified. Following treatment on d 10, suckling lambs were removed from 6 of the 12 ewes, creating non-suckled (NS) and suckled (S) treatment groups for subsequent study on d 14 through 26. On d 10, NAL treatment increased LH (P less than .01) but concentrations of PRL were not affected. When averaged across d 14 to 26, post-NAL concentrations of LH were greater (P less than .001) than pre-NAL concentrations (6.5 +/- .7 vs 1.9 +/- .4 ng/ml). In contrast, concentrations of PRL in the post-NAL period were lower (P less than .001) than pre-NAL concentrations (129 +/- 15 vs 89 +/- 10 ng/ml). Compared with S ewes over d 14 to 26, those in the NS group had similar pre-NAL concentrations of LH, tendencies for higher (P less than .10) post-NAL concentrations of LH, lower (P less than .001) mean serum concentrations of PRL (pre- and post-NAL) and similar pre-NAL vs post-NAL differences in serum PRL. Six suckled beef cows on d 24 to 35 were injected iv with either saline or NAL (.5 mg/kg) in a replicated crossover design. Injections of NAL increased serum concentrations of LH (P less than .05), when averaged over all 12 injections in the six cows, but serum PRL was not changed. However, three of six cows did not respond to NAL with increases in serum LH. These non-responding cows were similar to the responding cows in their pre-injection concentrations of LH and PRL, but they tended (P = .10) to have higher serum concentrations of cortisol than responding cows. 相似文献

In 1984, 31 per cent of 437 ram lambs reared on one property were found to have either bilateral or unilateral testicular hypoplasia. Similar numbers of ram lambs were reared in the next three years and 13, 2 and 0.5 per cent were affected. Severely abnormal testicles typically produced no spermatozoa and presented a 'Sertoli cell only' picture. Sixty-six of 200 ewe lambs born in 1984 were not marked by a harnessed teaser before mating; 12 of these were examined further and eight were found to have bilateral ovarian hypoplasia. The ovaries were very small and contained no oocytes or follicles. Plasma gonadotrophin levels were higher than normal in bilaterally affected rams and ewes but plasma testosterone and inhibin concentrations in rams with bilateral hypoplasia were not significantly different from those in normal rams. A genetic cause of the gonadal hypoplasia was considered unlikely in view of the pattern of occurrence and an examination of the flock pedigrees. No environmental cause was found. Two environmental sources of toxins were considered worthy of further investigation; onion grass (Romulea species) with its leaf spot fungus Helminthosporium biseptatum and the water supply which could have contained high concentrations of arsenic or other toxins during the period when the mothers of the affected lambs were in early pregnancy. 相似文献

Recent observations in seasonal-breeding mammals indicate that the hypothalamus is programmed to become leptin resistant during long days (LD) and leptin sensitive during short days (SD). These observations support the possibility that photoperiod mediates at least part of its effects on melatonin secretion through changes in leptin sensitivity. Herein we examined the interaction of season and recombinant ovine leptin (oleptin) on melatonin secretion by pineal explants in short-term culture. Glands were collected after sunset from eight ewes during LD (March, April, May, June) and from an additional eight ewes during SD (September, October, November, December). Glands were transected saggitally and coronally into quarters, with each equilibrated in 2.5ml of DMEM for 120min, followed by a 3h incubation in medium containing either 0 or 50ng/ml of oleptin. Treatment with oleptin reduced (P<0.001) melatonin secretion compared to controls during LD by approximately 22% at 2, 2.5 and 3h of culture. However, in cultures from glands collected during SD, oleptin stimulated (P<0.078) melatonin secretion approximately 50% compared to control. These effects were consistent throughout each respective season. We conclude that the secretion of melatonin from the ovine pineal gland is negatively responsive to leptin during LD, whereas leptin may stimulate melatonin secretion during SD. 相似文献

Thirty-two mature crossbred (Fine Wool Synthetic, FWS) sheep developed for fine wool production in India were treated for superovulation and oestrus synchronization in spring season. The ewes were randomly allocated to four treatment groups in a factorially designed experiment. For induction of superovulation, pregnant mare serum gonadotrophin (PMSG) was administered alone (group 1), in combination with gonadotrophin releasing hormone (GnRH) (group 2) or with follicle stimulating hormone (FSH) (group 3) and with both (group 4). Oestrus was synchronized in all the ewes by two injections of prostaglandin F2 (PGF, 10 mg each) administered at an interval of 10 days. Superovulation treatment started 48 h prior to the second PGF injection. The proportion of ewes in oestrus did not differ significantly (p>0.05) in the four groups. The use of GnRH set the ewes into oestrus earlier than the ewes in the other groups. Treatment with PMSG (800 IU) in conjunction with 4 g of Buserilin (GnRH) increased the ovulation rate (9.1±2.6 corpora lutea (CL)) above that observed when PMSG was used alone (3.0±0.7 CL). The use of FSH (0.5 U ovagen) in conjunction with PMSG was characterized by a decrease in the proportion of ewes with 2 CL (4/8 vs 7/8; p<0.05) and in the number of ovulations, i.e. CL observed (2.4±0.6 vs 9.1±2.6), and a nonsignificant increase in the incidence of large follicles (LF) (4.6±1.28 vs 3.25±0.6; p>0.05). The interaction between treatments of FSH and GnRH was not significant (p>0.05).It is concluded that use of GnRH, in conjunction with either PMSG alone or PMSG plus FSH treatment, advanced the onset of oestrus and increased the ovulation rate in FWS sheep. 相似文献

A dose of 6 X 10(4) Sarcocystis ovicanis sporocysts produced premature parturition in ewes and eventually proved fatal. The main pathological findings were myositis, myocarditis and encephalitis. Dose rates over the range of 2.5 X 10(3) to 6 X 10(4) depressed the haematocrit levels of pregnant ewes during the period 5-9 weeks after inoculation. Serum calcium, magnesium and phosphate values of ewes recently affected by vaginal prolapse were compared with unaffected ewes in four flocks. Subclinical hypocalcaemia was demonstrated in some affected and unaffected ewes in three flocks. Magnesium and phosphate values were normal. In two flocks the body condition of ewes recently affected by vaginal prolapse was variable and reflected the variation in condition found in the flock. In a third flock affected ewes had significantly lower body condition scores than unaffected ewes (P less than 0.001). Analysis of the fourth flock was not possible. Oestrogenic mycotoxins were not detected in any of the feed samples taken from these flocks. The following year the management, nutrition and energy, and the protein and calcium status of ewes in 12 flocks of greyface/mule ewes with a history of a regular high (greater than 3 per cent) or low (less than 1 per cent) prevalence of vaginal prolapse were compared. A high prevalence was not associated with any particular feedstuff. A high or intermediate (1 to 3 per cent) prevalence of vaginal prolapse was found in three of the four flocks managed as a single group and these three flocks were fed on an unrestricted basis. Body condition scoring and beta-hydroxybutyrate estimation confirmed that ewes in these flocks were overfed. The prevalence of vaginal prolapse in the flocks was not related to the serum albumin, calcium or urea of the ewes. Therefore subclinical hypocalcaemia was probably a consequence of vaginal prolapse rather than a cause. 相似文献

Four experiments were conducted to examine (1) how patterns of luteinizing hormone (LH) change as lambs approach first estrus, (2) whether mimicry of these changes by exogenous LH will initiate events of the pubertal process, (3) aspects of possible roles of the ovary and estradiol in the initiation of an ovulatory surge of LH in prepubertal lambs and (4) the time requirements for the presence of the ovary in induction of that surge of LH.In 19 lambs (Exp. 1), concentrations of LH in plasma were higher at puberty than 7 weeks earlier. Increased concentrations of LH in samples taken every 20 min for 6 hr during the luteal and follicular phases preceding first estrus were attributed to increased amplitude, but not increased frequency, of episodic pulses of LH. In Exp. 2, endocrine events which are included in the natural onset of ovarian cyclic activity (i.e. surge of LH and subsequent rise in progesterone) were initiated in , , , , and lambs, receiving either injections of 7.5 μg/hr, 15 μg/hr, 30 μg/2hr, 45 μg/3hr or constant infusions of 15 μg/hr of purified ovine LH, respectively. However, these lambs did not exhibit estrus or cyclic ovarian activity. In Exp. 3, the mechanism by which hourly injections of LH prompted surges of LH was determined to be mediated through ovarian stimulation and not through a direct effect of exogenous LH on the hypothalamo-pituitary axis. All the intact (n=5) but no acutely (two weeks) ovariectomized (n=5) lambs had a preovulatory-like surge in response to exogenous LH. Further, (Exp. 4) it was shown that the ovarian signal must be maintained or evoked within 6 hr preceding the surge of LH, because lambs (n=20) ovariectomized at or 9, 18 or 27 hr after initiation of the exogenous LH with the one exception failed to show a surge of LH. It could not be demonstrated that the ovarian signal involved changes in peripheral concentrations of androstenedione, testosterone or estradiol-17β, although patterns of LH in ovariectomized lambs were responsive to both negative and positive feedback effects of estradiol (n=6, Exp. 3) and in general estradiol levels were increasing prior to the induced surge of LH. 相似文献
