Adipose tissue expresses adipokines, which are involved in regulation of energy expenditure, lipid metabolism, and insulin sensitivity. To adapt for the transition from pregnancy to lactation, particularly in high-yielding dairy cows, adipokines, their receptors, and particular G-protein coupled receptors (GPRs) are of potential importance. Signaling by GPR 41 stimulates leptin release via activation by short-chain fatty acids; GPR 43/109A inhibits lipolysis, and GPR 109A thereby mediates the lipid-lowering effects of nicotinic acid and β–hydroxybutyrate. The aim of this study was to compare the mRNA expression of adiponectin and visfatin, adiponectin receptors 1 and 2 (AdipoR1/2), leptin receptor (obRb), insulin receptor as of the aforementioned GPRs during the transition period in high-yielding dairy cows. Biopsies from subcutaneous fat and blood samples were obtained from 10 dairy cows 1 week before and 3 weeks after calving. For AdipoR1 and AdipoR2 mRNA abundance as well as for leptin concentrations in plasma, a reduction (P ≤ .05) was observed postpartum; for visfatin and putative GPR 109A mRNA abundance in adipose tissue, there was a trend (P < .1) for analogous changes. In contrast, the mRNA content of obRb and GPR 41 in adipose tissue was higher (P ≤ .05) in samples from early lactation than in those from late gestation. Our results indicate decreasing adiponectin sensitivity in adipose tissue after calving, which might be involved in the reduced insulin sensitivity of adipose tissue during early lactation. In addition, visfatin, GPR 41, and GPR 109A may further modulate insulin sensitivity.  相似文献

Two experiments were conducted with lactating Angus x Gelbvieh beef cows to determine the effects of postpartum lipid supplementation, BCS at parturition, and day of lactation on fatty acid profiles in plasma, adipose tissue, and milk. In Exp. 1, 36 pri-miparous cows (488 +/- 10 kg of initial BW; 5.5 +/- 0.02 initial BCS) were given ad libitum access to hay and assigned randomly to a low-fat (control) supplement or supplements with cracked, high-linoleate safflower seeds (linoleate) or cracked, high-oleate safflower seeds (oleate) from d 3 to 90 of lactation. Diets were formulated to be isonitrogenous and isocaloric; safflower seed diets provided 5% of DMI as fat. Plasma and milk samples were collected on d 30, 60, and 90 of lactation. Adipose tissue biopsies were collected near the tail-head region of cows on d 45 and 90 of lactation. In Exp. 2, 3-yr-old cows achieving a BCS of 4 +/- 0.07 (479 +/- 36 kg of BW) or 6 +/- 0.07 (580 +/- 53 kg of BW) at parturition were used in a 2-yr experiment (n = 36/yr). Beginning 3 d postpartum through d 61 of lactation, cows were fed diets similar to those of Exp. 1. Adipose tissue and milk samples were collected on d 30 and 60, and plasma was collected on d 31 and 61 of lactation. Responses to postpartum dietary treatment were comparable in both experiments. Cows fed linoleate and oleate had greater (P < 0.001) total fatty acid concentrations in plasma than cows fed control. Except for 15:1, milk fatty acids with <18 carbons were greatest (P < or = 0.01) for cows fed control, whereas milk from cows fed linoleate had the greatest (P < or = 0.02) 18:1trans-11, 18:2n-6, and cis-9, trans-11 CLA. Milk from cows fed oleate had the greatest (P < 0.001) 18:1cis-9. In Exp. 1, total fatty acid concentrations in adipose tissue samples decreased at d 90 compared with d 45 of lactation, but the fatty acid profile of cow adipose tissue was not affected (P = 0.14 to 0.80) by dietary treatment. In Exp. 2, the percentage of cis-9, trans-11 CLA in adipose tissue of cows with a BCS of 6 decreased (P = 0.001) from d 30 to 60 of lactation. Plasma and milk fatty acid composition reflected alterations in postpartum diet. Less medium-chain fatty acids and more 18-carbon fatty acids in milk were indicative of reduced de novo fatty acid synthesis in the mammary gland of beef cows fed lipid supplements; however, the metabolic demands of lactation prevented the deposition of exogenously derived fatty acids in adipose tissue through d 90 of lactation.  相似文献

Three-year-old Angus x Gelbvieh beef cows, which were nutritionally managed to achieve a BCS of 4 +/- 0.07 (479 +/- 36 kg of BW) or 6 +/- 0.07 (580 +/- 53 kg of BW) at parturition, were used in a 2-yr experiment (n = 36/yr) to determine the effects of maternal BCS at parturition and postpartum lipid supplementation on fatty acid profile of suckling calf plasma and adipose tissue. Beginning 3 d postpartum, cows within each BCS were assigned randomly to 1 of 3 treatments in which cows were all fed hay and either a low-fat (control) supplement or supplements with either high-linoleate cracked safflower seeds (linoleate) or high-oleate cracked safflower seeds (oleate) until d 61 of lactation. Diets were formulated to be isonitrogenous and isocaloric, and safflower seed supplements were provided to achieve 5% of DMI as fat. Total concentration of fatty acids in plasma did not differ (P = 0.48) due to maternal BCS at parturition. Percentage of 20:5n-3 in plasma tended (P = 0.06) to be greater for calves suckling cows with a BCS of 6 at parturition. No other differences (P = 0.12 to 0.99) were noted in calf plasma fatty acid profile due to maternal BCS at parturition. Likewise, no differences were detected for total fatty acid concentration (P = 0.88) in calf adipose tissue due to maternal BCS at parturition. Weight percentage of 14:1 (P = 0.001) was greatest in adipose tissue of calves suckling cows fed control and oleate; however, the percentages of 14:0, 15:0, 16:0, 16:1, 17:0, and 18:3n-3 were greater (P < 0.001) in adipose tissue from calves suckling cows fed control compared with calves suckling cows fed linoleate or oleate. Percentages of 18:0, 18:1trans-11, 18:2n-6, and cis-9, trans-11 CLA were greater (P < 0.001) in adipose tissue from calves suckling cows fed linoleate compared with calves suckling cows fed control and oleate. Calves suckling cows fed oleate had greater (P < 0.001) percentages of 18:1trans-9, 18:1trans-10, and 18:1cis-9 in adipose tissue than calves suckling cows fed control or linoleate. Calf plasma and adipose tissue fatty acid profiles were reflective of milk fatty acids. Because fatty acids play an important role in metabolic regulatory functions, changes in milk fatty acid profile should be considered when beef cows are fed lipid supplements.  相似文献

Beginning 3 d postpartum, cows within each BCS were randomly assigned to be fed hay and a low-fat control supplement or supplements with either cracked high-linoleate safflower seeds or cracked high-oleate safflower seeds until d 60 of lactation. Diets were formulated to be isonitrogenous and isocaloric, and safflower seed diets provided 5% DMI as fat. Adipose tissue biopsies were collected near the tail-head region of cows on d 30 and 60 of lactation. Dietary treatment did not affect (P > or = 0.43) adipose tissue lipogenesis. Body condition score at parturition did not affect acetate incorporation into lipid (P = 0.53) or activity of acetyl CoA carboxylase (P = 0.77) or fatty acid synthase (P = 0.18). Lipoprotein lipase activity and palmitate incorporation into triacyl-glycerol tended to be greater (P = 0.06), and palmitate esterification into total acylglycerols was greater (P = 0.01) in cows with a BCS of 4 at parturition. Mean activity of acetyl-CoA carboxylase (P < 0.001), lipoprotein lipase (P = 0.01), and rate of palmitate incorporation into monoacylglycerol (P = 0.02), diacylglycerol (P = 0.001), triacylglycerol (P = 0.003), and total acylglycerols (P = 0.002) were greater at d 30 than d 60, suggesting a greater proclivity for fatty acid biosynthesis and esterification by adipose tissue at d 30 of lactation. Although dietary lipid supplementation did not affect adipose tissue lipogenesis, results suggest that cows with a BCS of 4 at parturition have a greater propensity to deliver exogenously derived fatty acids to the adipocyte surface and incorporate preformed fatty acids into acylglycerols as stored adipocyte lipid. Additionally, cows in early lactation seemed to be able to synthesize and incorporate more fatty acids into stored lipid than cows during peak lactation.  相似文献

Natural antibodies (NAb) are defined as antibodies that circulate in normal healthy individuals under the absence of deliberate antigenic stimulation. Two types of NAb are distinguished: NAb towards exogenous antigens and NAb towards autoantigens (N(A)Ab). The objectives of the current study were threefold. First, we studied the relation between metabolic health and concentrations of NAb binding keyhole limpet hemocyanin (KLH) or lipopolysaccharide (LPS) in milk and plasma of dairy cows in early lactation. Second, we determined the presence of N(A)Ab binding transferrin, myosin and thyroglobulin in bovine milk. Third, we studied the relation between N(A)Ab in bovine milk and mammary health. For the first objective, dairy cows were either fed a control (C) (n = 8) or a diet where 2 kg of concentrates were replaced by an iso-energetic concentrate containing marine algae (ALG) from week −3 till 8 postpartum (experiment 1). Plasma and milk samples were analyzed weekly for NAb binding either KLH or LPS. Plasma was analyzed for glucose, non-esterified fatty acids (NEFA) and β-hydroxybutyrate (BHBA). For the second and third objective, milk samples were collected weekly from 96 dairy cows from week 2 till 9 postpartum and analyzed for milk composition and N(A)Ab binding myosin, transferrin and thyroglobulin (experiment 2). For both datasets, N(A)Ab titers are expressed as 2log values of the highest dilution giving a positive reaction. Data are expressed as means ± SEM. Repeated observations were analyzed in a mixed model. In experiment 1, no diet effect (P > 0.05) was observed on NAb binding LPS in plasma or milk, NAb binding KLH in milk was greater (P = 0.05) for cows fed the control diet. Concentration of NAb binding KLH and LPS in plasma was negatively related to plasma NEFA concentration (P < 0.05). In experiment 2, NAb binding myosin (5.66 ± 0.06), thyroglobulin (4.85 ± 0.06), and transferrin (5.76 ± 0.07) were identified in milk. Clinical mastitis incidence (9%) tended to be positively related to concentration of NAb binding myosin (P = 0.06) and negatively related to Nab binding transferrin (P = 0.08). In conclusion, NAb binding KLH and LPS in plasma and milk are related to metabolic health, as indicated by plasma NEFA concentration. Furthermore, this study demonstrates the presence of N(A)Ab in bovine milk and shows trends for a relation between N(A)Ab binding auto-antigens and mastitis. Future studies should confirm these trends and shed light on the predictive value of N(A)Ab in bovine milk for mammary health.  相似文献

Energy intake prepartum is critically important to health, milk performance, and profitability of dairy cows. The objective of this study was to determine the effect of reduced energy density of close-up diets on dry matter intake (DMI), lactation performance and energy balance (EB) in multiparous Holstein cows which were housed in a free-stall barn and fed for ad libitum intake. Thirty-nine dry cows were blocked and assigned randomly to three groups fed a high energy density diet [HD, n = 13; 6.8 MJ of net energy for lactation (NEL)/kg; 14.0% crude protein (CP) ], or a middle energy density diet (MD, n = 13; 6.2 MJ NEL/kg; 14.0% CP), or a low energy density diet (LD, n = 13; 5.4 MJ NEL/kg; 14.0% CP) from d 21 before expected day of calving. After parturition, all cows were fed the same lactation diet to d 70 in milk (DIM). The DMI and NEL intake prepartum were decreased by the reduced energy density diets (P < 0.05). The LD group consumed 1.3 kg/d (DM) more diet compared with HD group in the last 24 h before calving. The milk yield and the postpartum DMI were increased by the reduced energy density diet prepartum (P < 0.05). The changes in BCS and BW prepartum and postpartum were not affected by prepartum diets. HD group had higher milk fat content and lower lactose content compared with LD group during the first 3 wk of lactation (P < 0.05). The energy consumption for HD, MD and LD groups were 149.8%, 126.2% and 101.1% of their calculated energy requirements prepartum (P < 0.05), and 72.7%, 73.1% and 75.2% during the first 4 wk postpartum, respectively. In conclusion, the low energy density prepartum diet was effective in controlling NEL intake prepartum, and was beneficial in increasing DMI and milk yield, and alleviating negative EB postpartum.  相似文献

The objective of this study was to investigate serum lipids, metabolic parameters and activity of the anti-oxidative enzyme paraoxonase-1 (PON1). The study was conducted on non-pregnant heifers with optimal health status and on healthy dairy cows in the period of intensive lactation, assuming that the energy and metabolic demands during lactation reduce anti-oxidative protection. Total cholesterol and HDL-cholesterol concentrations were significantly higher (P < 0.05) in lactating cows than in heifers. Bilirubin concentration and γ-glutamyltransferase (GGT) activity were also significantly higher in lactating cows (P < 0.05), indicating increased hepatic efforts of cows to meet energy requirements for lactation. Significantly lower PON1 activity and PON1/HDL ratio in lactating cows compared to heifers (P < 0.05) showed that metabolic efforts during pregnancy, parturition and lactation influence PON1 activity due to oxidative stress. Concurrent increase in total and HDL-cholesterol during lactation indicated that the HDL particle is a major carrier of cholesterol in cows.  相似文献

The objectives of this study were to examine the influence of body condition of cows on metabolic and antioxidative status, as well as to investigate the relationship between metabolic indicators of lipid mobilization and oxidative stress during transition period. The study was conducted on 24 Holstein‐Friesian dairy cows divided into 2 groups according to their body condition score (BCS) as optimal (n = 12; BCS from 3.25 to 3.75) or adipose (n = 12; BCS ≥4). Metabolic status (glucose, triglycerides, total cholesterol, HDL cholesterol, NEFA and BHB), paraoxonase‐1 (PON1) and apolipoprotein A‐I (ApoA‐I) were analysed in sera taken on days ?30, ?10, ?2, 0, 5, 12, 19, 26 and 60 relative to parturition. Adipose cows had significantly higher glucose concentration at parturition being significantly decreased after parturition on days 12 and 19. Total cholesterol and HDL‐C concentrations were the lowest at parturition and significantly higher on days 26 and 60 after parturition in both groups of cows. Both investigated groups had significantly higher NEFA concentration from parturition until day 19 after parturition, indicating energy deficit and an increased lipid mobilization after calving. There were no significant differences in BHB concentration during transition period in both groups. No significant differences were found in PON1 activity and ApoA‐I concentration during transition period in both groups of cows. However, in adipose cows, although not significantly different, PON1 was decreased from calving until day 19 after parturition indicating a disturbance in antioxidative status in adipose cows. PON1 significantly positively correlated with total cholesterol and HDL‐C concentrations and negatively with NEFA indicating a strong relationship of PON1 with lipid metabolism. Significant positive correlation between NEFA and BHB in both groups of cows points out on energy deficit during transition period that cows tend to overcome by lipid mobilization providing alternative source of energy needed for parturition and lactation.  相似文献

The effects of floor heating at parturition on sow and piglet behaviour in pens were examined in a Danish production herd. A total of 50 gilts and sows were split into two groups; one (n = 25) experienced no treatment; one (n = 25) was exposed to floor heating in the area of the sow around parturition. The sows and piglets were recorded on video 24 h a day for behavioural analyses.Floor heating tended to improve piglet weight at weaning (P = 0.05) but did not affect average daily gain. Piglets in pens with floor heating were more often in proximity of the sow at posture changes than piglets from pens without floor heating on day one postpartum (P = 0.01). Floor heating did not affect duration of parturition or any of the recordings of sow behaviour significantly.  相似文献

The present study aimed to determine the effects of differing nutrient levels during the far‐off period on postpartum metabolism and milk production in lactating cows. Twenty‐six multiparous cows were assigned to three dietary treatments in the far‐off period: a low‐energy diet (L, n = 9, 80% intake of the total digestible nutrients requirement), a moderate‐energy diet (M, n = 8, 105%) and a high‐energy diet (H, n = 9, 130%). During the close‐up period, all cows were provided with 105% intake. After parturition, all cows were fed a lactation diet. The BCS recovery was slow, and low milk yield was found in the H group. In the L group, BCS recovery was favorable after parturition, and lactation persistence was increased. The L group had low rumen endotoxin activity and a high initial ovulation rate after parturition. These findings indicate that a high‐energy diet during the far‐off period has a deleterious effect on milk production. In contrast, the restricted diet in the far‐off period increased adaptability with respect to peri‐parturition metabolic changes, improved the post‐parturition nutritional state, and increased milk production. Furthermore, it suggests that the nutrient levels in the far‐off period affect rumen endotoxin activity and reproductive function after parturition.  相似文献

Synchronization of parturition in a commercial flock of sheep has many advantages including the maintenance of the efficiency of the operation as well as optimization of care for periparturient animals. In a research setting, using an ovine model for experiments involving the induction of parturition yields the results that can be applied to other mammalian species. In sheep, as well as in a variety of other farmed species, dexamethasone is the drug used to induce synchronized parturition at the end of gestation. However, considerable variations have been noticed in the time between induction and the onset of lambing. This study set out to determine the role that the breed of dam and sire, the age and reproductive history of ewes, the month of induction and the litter/lamb characteristics played in determining the time between dexamethasone treatment and parturition. The lambing was induced in 146 ewes (4 purebred Dorset, 19 purebred Rideau Arcott and 123 Rideau Arcott × Dorset ewes) using a single i.m. dose of 20 mg dexamethasone (Unidex®, Univet Pharmaceuticals, Milton, ON, Canada) on day 140 of gestation. The ewe breed was weakly correlated (r = − 0.19, P < 0.05) whereas the sire breed was moderately correlated (r = − 0.37, P < 0.001) to the time between dexamethasone treatment and the onset of parturition (the breed in this study was defined as the % of the Rideau Arcott genotype). The time between induction and the onset of labour averaged 57.3 ± 5.3 h and 64.3 ± 8.2 h in February and April, respectively, and it was significantly longer (P < 0.05) compared with the times recorded in July, September and November (42.0 ± 2.0 h, 38.7 ± 2.8 h and 36.1 ± 5.0 h, respectively). There was a negative correlation (r = − 0.41, P < 0.001) between the proportion of ram lambs and the mean birth weight of ewe lambs. These results can be interpreted to suggest that the breed and time of the year may both affect the synchrony of dexamethasone-induced lambing. The reasons for the associations found between the month of induction and the time elapsed to the onset of parturition as well as between the proportion of ram lambs and birth weights of ewe lambs are not fully understood and warrant further studies.  相似文献

To determine the effects of BCS at parturition and postpartum lipid supplementation on blood metabolite and hormone concentrations, 3-yr-old Angus x Gelbvieh beef cows, which were nutritionally managed to achieve a BCS of 4 +/- 0.07 (479.3 +/- 36.3 kg of BW) or 6 +/- 0.07 (579.6 +/- 53.1 kg of BW) at parturition, were used in a 2-yr experiment (n = 36/yr). Beginning at 3 d postpartum, cows within each BCS were assigned randomly to be fed hay and a low-fat control supplement or lipid supplements with either cracked high-linoleate or high-oleate safflower seeds until d 61 of lactation. The diets were formulated to be isonitrogenous and isocaloric, and the safflower seed supplements were formulated to achieve 5% DMI as fat. On d 31 and 61 of lactation, blood samples were collected preprandially and then hourly postprandially (at 0, 1, 2, 3, and 4 h). Serum insulin (P = 0.27) and glucose (P = 0.64) were not affected by BCS at parturition. The mean concentrations of plasma NEFA (P = 0.08) and beta-hydroxybutyrate (P = 0.08) tended to be greater, and serum IGF-I was greater (P < 0.001) in BCS 6 than BCS 4 cows. Conversely, serum GH was greater (P = 0.003) for BCS 4 cows, indicating that regulation of IGF by GH may have been uncoupled in BCS 4 cows. The postpartum diet did not affect NEFA (P = 0.94), glucose (P = 0.15), IGF-I (P = 0.33), or GH (P = 0.62) concentrations. Oleate-supplemented cows had greater (P = 0.03) serum insulin concentrations, whereas control cows had greater (P = 0.01) plasma beta-hydroxybutyrate concentrations. Concentrations of NEFA (P = 0.05) and glucose (P < 0.001) were greater, and beta-hydroxybutyrate tended (P = 0.07), to be greater at d 3, whereas serum IGF-I was greater (P = 0.003) at d 6 of lactation. Similar concentrations of NEFA, glucose, GH, and IGF-I indicate that the nutritional status of beef cows during early lactation was not influenced by lipid supplementation. However, perturbations of the somatotropic axis in BCS 4 cows indicate that the influence of energy balance and BCS of the cow at parturition on postpartum performance should be considered when making managerial decisions.  相似文献

Glucose, fatty acids, and l-carnitine are important substrates that support mammary epithelial cell metabolism, biosynthetic capacity, and milk yield and composition. Our study investigated the effects of LPS-induced inflammation on the expression of several glucose, fatty acid, and l-carnitine transporters in the lactating rat mammary gland at different lactation stages. Day 4, 11, and 18 lactating rats (n = 3/treatment) were administered LPS (1 mg/kg) or saline by intraperitoneal (IP) injection. Fold differences in the mRNA expression of glucose transporters Glut1, Glut8 and Sglt1, fatty acid transporters Fatp1, Fatp4 and Fabp3, and l-carnitine transporters Octn1, Octn2, and Octn3 were determined using the Comparative CT method. The mRNA expression levels of all transporters evaluated, except Fatp4 and Octn2 were markedly higher in mammary gland at lactation day 11 compared to lactation day 4. LPS caused a marked decrease in transporter mRNA expression at each lactation stage except for Octn3 and Fatp1, which were markedly increased with LPS administration at lactation day 4, and Sglt1, which was slightly increased at day 11 of lactation. Our results suggest LPS-induced inflammation generally downregulates glucose, fatty acid, and l-carnitine transporter expression. Whether such changes lead to reductions in transporter substrate availability to the lactating mammary epithelial cell requires investigation since decreases in the availability of these nutrients may significantly impact mammary epithelial function and milk quality and yield.  相似文献

The objectives of this study were to evaluate the effects of dietary organic sources of zinc (Zn), copper (Cu) and selenium (Se) for dairy cows on somatic cell count (SCC), occurrence of subclinical mastitis, number of clinical mastitis cases and concentration of superoxide dismutase (CuZnSOD), glutathione peroxidase (GSH-Px) and ceruloplasmin (CP). Nineteen dairy cows, with confirmed pregnancy, were selected by body weight, body condition score (BCS), number of lactation, and milk yield in previous lactation, and randomly distributed among two groups, one to receive organic (n = 9) and the other the inorganic (n = 10) source of Zn, Cu and Se. Diets were formulated to meet the nutritional requirements of animals from 60 days before the expected date of calving up to 80 days of lactation. Blood samples were collected at − 60, − 21, 1, 21, 40 and 80 days in relation to the expected date of calving for analysis of CuZnSOD, GSH-Px and CP concentration. Milk samples for SCC determination were collected weekly after 15 days of lactation, also at day 1 and 7 and when a clinical case was diagnosed for microbiological culture. The number of new and total cases of subclinical mastitis was lower for the group of cows fed with organic sources of Zn, Cu and Se compared to animals receiving inorganic sources. Average SCC during the first 80 days of lactation tended to be lower for the group fed with organic Zn, Cu and Se. Feeding organic Zn, Cu and Se did not alter the concentration of CuZnSOD, GSH-Px and CP.  相似文献

Background

Objectives were to examine the effects of selenium (Se) supply and maternal nutritional plane during gestation on mammary gland growth, cellular proliferation, and vascularity at parturition and d 20 of lactation. Rambouillet primiparous ewes (n = 84) were allocated to treatments in a 2 x 3 factorial. Factors were dietary Se (adequate Se [ASe, 11.5 μg/kg BW] or high Se [HSe, 77.0 μg/kg BW]) and nutritional plane (60% [RES], 100% [CON], or 140% [EXC]). At parturition, lambs were removed and 42 ewes (7/treatment) were necropsied. Remaining ewes were fed a common diet meeting requirements for lactation and mechanically milked twice daily until necropsy on d 20. At both necropsy periods, mammary glands were dissected and tissues harvested. Samples were analyzed for RNA, DNA, and protein content, cell proliferation, and vascularity. Where interactions were present (P ≤ 0.05), least squares means from the highest-order interaction are presented.

Results

Final body weight of ewes was least (P ≤ 0.002) in RES, intermediate for CON, and greatest for EXC, regardless of stage of the ewe at necropsy (parturition or d 20 of lactation). In ewes necropsied at parturition, mammary glands were heavier (P = 0.02) in EXC compared to RES, with CON intermediate. Concentration of RNA (mg/g) was decreased (P = 0.01) in EXC compared to CON at parturition. There was a tendency (P = 0.07) for a Se by nutrition interaction in percentage of cells proliferating where ASe-EXC ewes had greater (P ≤ 0.02) number of proliferating cells then all other treatments. Mammary vascular area tended (P = 0.08) to be affected by a Se by nutrition interaction where ASe-CON had less (P = 0.007) vascular area than HSe-CON ewes. In ewes necropsied at d 20 of lactation, the number of alveoli per area was decreased (P ≤ 0.05) in RES compared to CON and EXC-fed ewes.

Conclusions

Results of this study indicate that proper maternal nutritional plane during gestation is important for mammary gland development, even out to d 20 of lactation.  相似文献

This investigation studied the effects of intravaginal administration of a mixture of lactic acid bacteria (LAB) on the incidence of purulent vaginal discharges (PVD), plasma haptoglobin concentrations, and milk production in dairy cows. A total of 82 pregnant primiparous and multiparous Holstein dairy cows were used in this study. Half of the cows received intravaginally 1 mL of LAB at 1010–1012 cfu/mL and the other half 1 mL of reconstituted skim milk (i.e., carrier) (controls). Administration of LAB was conducted once per wk during 2 and 1 wk before the expected day of calving and at 1, 2, 3, and 4 wk postpartum. Data demonstrated that intravaginal administration of LAB decreased the occurrence of PVD at 3 wk postpartum (P < 0.05). Concentrations of plasma haptoglobin, an acute phase protein often associated with uterine infections, was lower in cows treated with the LAB mixture at 2 wk (P < 0.001) and 3 wk (P < 0.05) postpartum. Treatment with LAB did not improve overall pregnancy rate, but the treated multiparous cows produced more milk than their control counterparts (P < 0.05), whereas no difference was observed in primiparous cows regarding milk yield (P > 0.05). Overall, this is the first study demonstrating that intravaginal LAB administration lowers the incidence of PVD and enhances milk production in dairy cows. Further research is warranted to evaluate the effects of LAB on reproductive performance in a larger cohort of cows.  相似文献

Glucose delivery and uptake by the mammary gland are a rate-limiting step in milk synthesis. It is thought that insulin-independent glucose uptake decreases in tissues, except for the mammary gland, and insulin resistance in the whole body increases following the onset of lactation. To study glucose metabolism in peak-, late-, and nonlactating cows, the expression of erythrocyte-type glucose transporter (GLUT1) and the insulin-responsive glucose transporter (GLUT4) in the mammary gland, adipose tissue, and muscle were assessed by Western blotting and real-time PCR. Our results demonstrated that the mammary gland of lactating cows expressed a large amount of GLUT1, whereas the mammary gland of nonlactating cows did not (P < 0.05). On the other hand, adipose tissue of late and nonlactating cows expressed a large amount of GLUT1, whereas the adipose tissue of peak-lactating cows did not (P < 0.05). There were no significant differences in the abundance of GLUT4 mRNA in adipose tissue and muscle, whereas GLUT4 mRNA was not detected in the mammary gland. The plasma insulin concentration was greater (P < 0.05) in nonlactating cows than in peak- and late-lactating cows. The results of the present study indicate that in lactation, GLUT1 expression in the mammary gland and adipose tissue is a major factor for insulin-independent glucose metabolism, and the expression of GLUT4 in muscle and adipose tissue is not an important factor in insulin resistance in lactation; however, the plasma insulin concentration may play a role in insulin-dependent glucose metabolism. The objective of this experiment was to investigate the effect of fat supplementation during the transition period on pre and postpartum body weight (BW), body condition score (BCS), non‐esterified fatty acids (NEFA), glucose and leptin concentrations in Holstein cows. Holstein cows (n = 15) received a low fat diet (LF; 1.61 Mcal net energy for lactation (NEL)/kg of dry matter [DM]), moderate fat diet (MF; 1.68 Mcal NEL/kg DM) or a high fat diet (HF; 1.74 Mcal NEL/kg DM) for 4 weeks prior to calving. All cows were fed similar lactation diets ad libitum (1.74 Mcal NEL/kg DM) for 30 days after calving. Increasing diet energy density during transition period had no effect on prepartum DMI, BCS, BW, glucose and NEFA concentrations (P > 0.05); but leptin concentrations and energy balance (EB) were affected by treatments (P < 0.05). Animals fed HF had less plasma leptin prepartum. After parturition, BW, milk production, milk fat, protein, urea nitrogen and plasma glucose concentrations were affected by prepartum diets (P < 0.05). Fat supplementation prepartum did not affect postpartum NEFA. In conclusion, prepartum fat supplementation decreased leptin concentration prepartum.  相似文献

Effects of grain source and dietary oil supplement on dry matter intake (DMI), feeding, chewing behavior, and production performance of lactating dairy cows were evaluated using eight multiparous Holstein cows (77±22.1 days in milk; mean±SD) in a duplicated 4×4 Latin square design with a 2×2 factorial arrangement of treatments. Experimental diets contained either ground barley or ground corn supplemented with either fish oil or soybean oil at 2% of dietary dry matter (DM). Geometric mean particle size of dietary treatments was 4.1 mm. Dry matter intake tended (P=0.09) to be greater for barley- vs. corn-based diets (23.2 vs. 22.3 kg/d), but was reduced for the fish oil compared to soybean oil supplemented diets (21.1 vs. 24.3 kg/d; P<0.001). This reduction in DMI was attributed to smaller meal size (1.24 vs. 1.55 kg of DM; P=0.004) and slower eating rate (0.082 vs. 0.098 kg of DM/min; P<0.001) for fish oil compared to soybean oil supplemented diets. Main treatment effects interacted (P=0.005) for DMI of particles retained on 19 mm sieve but not for sorting index. Eating rate (0.090 kg of DM/min) was similar between barley- and corn-based diets, however, rumination time was greater for barley- compared to corn-based diets as result of longer rumination bout duration (32.5 vs. 28.5 min/bout; P=0.01). Barley- compared to corn-based diets increased total chewing time by 71 min (709 vs. 638 min) for cows fed fish oil, but not for cows fed soybean oil. Grain source did not affect milk yield or milk composition. Compared to soybean oil, fish oil negatively affected milk yield (40.4 vs. 43.4 kg/d; P=0.01), and thereby, both milk fat (0.91 vs. 1.26 kg/d; P<0.001) and protein (1.23 vs. 1.33 kg/d; P=0.007) production. However, feed efficiency (milk yield/DMI) was greater in fish oil compared to soybean oil supplemented diets (1.94 vs. 1.80; P=0.003). Results indicated that grain source and oil supplement can interact to affect feeding and chewing behavior, but not lactational performance of lactating cows.  相似文献

One hundred and ninety non-lactating, pregnant beef cows (three-fourth Bos taurus and one-fourth Bos indicus; 138 multiparous and 52 primiparous) were assigned to this experiment at 117 ± 2.2 d of gestation (day 0). Cows were ranked by parity, pregnancy type (artificial insemination = 102 and natural service = 88), body weight (BW), and body condition score (BCS) and assigned to receive a supplement containing: 1) sulfate sources of Cu, Co, Mn, and Zn (INR; n = 95) or 2) an organic-complexed source of Cu, Mn, Co, and Zn (AAC; Availa 4; Zinpro Corporation, Eden Prairie, MN; n = 95). The INR and AAC provided the same daily amount of Cu, Co, Mn, and Zn, based on 7 g of the AAC source. From day 0 to calving, cows were maintained in a single pasture and were segregated three times weekly into 1 of the 24 individual feeding pens to receive treatments. Cow BW and BCS were recorded on days −30, 97, upon calving, and at weaning (day 367). Milk production was estimated at 42 ± 0.5 d postpartum via weigh–suckle–weigh (WSW) method. Liver biopsies were performed in 30 cows per treatment on days −30, 97, upon calving, and the day after WSW. Calf BW was recorded at birth and weaning. Liver and longissimus muscle (LM) biopsies were performed in 30 calves per treatment upon calving and 24 h later, the day after WSW, and at weaning. No treatment effects were detected (P ≥ 0.49) for cow BCS during gestation, despite AAC cows having greater (P = 0.04) BW on day 97. Liver Co concentrations were greater (P < 0.01) for AAC compared with INR cows, and liver concentrations of Cu were greater (P = 0.02) for INR compared with AAC cows on day 97. Upon calving, INR cows had greater (P ≤ 0.01) liver Cu and Zn concentrations compared with AAC cows. No other treatment differences were noted (P ≥ 0.17) for cow and calf liver trace mineral concentrations. Cows receiving AAC had greater (P = 0.04) hepatic mRNA expression of metallothionein 1A at calving, and their calves had greater (P = 0.04) hepatic mRNA expression of superoxide dismutase at weaning. Milk production did not differ between AAC and INR cows (P = 0.70). No treatment effects were detected (P ≥ 0.29) for mRNA expression of LM genes associated with adipogenic or muscle development activities in calves at birth and weaning. Calf birth and weaning BW also did not differ (P ≥ 0.19) between treatments. In summary, supplementing AAC or INR to beef cows during the last 5 mo of gestation yielded similar cow–calf productive responses until weaning.  相似文献
