A method has been devised to produce eosinophilia in birds experimentally. Multiple intraperitoneal injections of bovine serum albumin (BSA) produced a greater response than horse serum in young adult fowls, and injections of 2 ml BSA were more effective than injections of 0.2 ml. The highest eosinophilic responses were obtained with injections of BSA together with aluminium hydroxide. Thymectomy had little effect on eosinophil numbers following horse serum injections but irradiation produced a depressed response. Horse serum that had been heated to 56 degrees C for 30 minutes to inactivate complement and to denature proteins, including possible horse IgE, induced an eosinophilic response in 50 per cent of birds compared with only 14 per cent with unheated horse serum. Furthermore, when the former birds were reinjected after a rest period of 13 weeks, the eosinophilic response followed a similar curve as before but a higher proportion of birds responded. Birds aged over two years did not give a good eosinophilic response after horse serum stimulation. There was no consistent correlation between raised IgE levels and eosinophil numbers. Tolerance to horse serum was detected in some birds using a radial immunodiffusion technique. Analyses of blood plasma from birds with eosinophilia showed a significant increase in total amino acids compared with normal birds. 相似文献

Intraneural perineurioma is an extremely rare condition characterized by perineurial cell proliferation within peripheral nerve (PN) sheaths. In the veterinary field, this entity has been reported only in a dog. We examined multiple enlargements of PNs in 11 chickens (Gallus gallus domesticus) (9 Japanese bantams and 2 specific pathogen-free White Leghorn), which were inoculated with an avian leukosis virus (ALV) causing so-called fowl glioma. All chickens clinically exhibited progressive leg paralysis. Lumbosacral plexus, brachial plexus, and/or spinal ganglion were commonly affected, and these nerves contained a diffuse proliferation of spindle cells arranged concentrically in characteristic onion bulb-like structures surrounded by residual axons and myelin sheaths. The spindle cells were immunohistochemically negative for S-100alpha/beta protein. Electron microscopy revealed that these cells were characterized by short bipolar cytoplasmic processes, occasional cytoplasmic pinocytotic vesicles, and discontinuous basal laminae. These features are consistent with those of intraneural perineurioma. Furthermore, the specific sequence of the ALV was detected in the PN lesions of 8/11 (73%) birds by polymerase chain reaction. These results indicate that the multiple intraneural perineuriomas of chicken may be associated with the ALV-A causing fowl glioma. 相似文献

In this study we examined some histologic and histochemical characteristics of the embryonic sheep dental epithelium in early odontogenesis. During the first trimester of development, a short-lived dental lamina was observed. Four young broiler chickens affected by multiple melanotic tumors are described. Grossly, there were multiple tumors composed of melanocytes within the skin, skeletal muscle, and multiple visceral organs. Tumors ranged from flattened macules to masses that extensively replaced viscera. Microscopically, melanocytes were often well pigmented, and while there was moderate nuclear anisokaryosis, mitotic rates were low. Immunohistochemical staining of some melanomas with antibodies to S100 proteins, Melan-A, vimentin, or neuron-specific enolase after bleaching of tumor cells with potassium permanganate revealed lack of immunostaining of tumor cells with antibodies to S100, strong positive staining of tumor cells for neuron-specific enolase, moderate staining with antibodies to vimentin, and faint staining for Melan-A. Only neuron-specific enolase staining was evident in unbleached tumor cells. Attempts to identify exogenous avian leukosis viruses in these tumors were unsuccessful. 相似文献

1. Development of brain temperature regulation was studied in the domestic fowl (Gallus gallus domesticus) from hatching to 21 d of age. 2. Body and brain temperature at hatching were relatively low compared with adult levels. However, both were effectively regulated at ambient temperatures of 30 and 35 degrees C, with only a minor difference between the 2 temperatures. 3. Body and brain temperature increased as a power function of age at significantly different rates, approaching adult levels at around 10 d of age and resulting in a linear increase in body-to-brain temperature difference with age. 4. The results indicated the existence of different patterns of post-hatching development of brain temperature regulation in relation to the degree of precocity of a species. 相似文献

After a preliminary finding of ultrasonographically detectable ring structure within the yolk of avian eggs, a study was initiated to investigate these properties. Ultrasonographically detectable alternating hyperechoic and hypoechoic rings within the egg yolk were found to be consistent but variability occurred in ring numbers, thickness, and clarity. Rings were best seen at two days post oviposition and were not affected by refrigeration or heating of the yolks. The usefulness of rings in experimental studies of yolk content of lipoprotein is postulated. 相似文献

An 8-month-old white feathered, black skinned Moroseta hen was presented for examination because of numerous 2 mm- to 30 mm-diameter irregularly shaped, hard nodules in the skin of the head, wings, back, and abdomen. The nodules were confined to the skin and did not involve subcutaneous tissues. Nodules consisted of dilated feather follicles packed with a caseous tan-to-pale-yellow material admixed with feather remnants. Histologically, affected feather follicles were markedly dilated and filled with laminated keratin debris. Necrosis of the epidermis and perifollicular lymphocyte infiltration was also present. Bacteriologic investigation of internal organs was negative, while secondary bacteria, Proteus spp. and Bacillus spp., were isolated from skin nodules. A concomitant lice infestation of Menopon spp., as well as leg mange caused by Cnemidocoptes spp., were also present. These bacterial isolates and parasites were not related to the disease condition. The condition observed was differentiated from benign feather follicle tumors, and a diagnosis of multiple feather follicle cysts was made. In addition, a breed predisposition was hypothesized. 相似文献

The finer innervation of the tissues of the lung was studied with several methods of silver impregnation, focussing especially on the fibers and endings of the nerves. The richest terminal innervation could be found in the epithelial and subepithelial tissue, and was sometimes similar to the situation in the smooth muscles which might indicate that, somehow, secretory and motor innervation are alike morphologically. On the other hand, there were many nerve fibers in the perichondrium of the small nodules and plaques of cartilage too, although they did not enter the cartilaginous tissue. Atmospheric ammonia in animal housing is reported to have adverse effects on livestock performance and animal health. Previous experiments have found that 75 ppm ammonia reduced the production performance and altered body fat distribution quality of broilers. In this study, we examined the body fat distribution, serum metabolites and lipid metabolism gene expression of broiler exposed to ammonia. A total of 400 chickens were randomly allocated to four groups with four replicates and received ammonia treatments at 0, 25, 50 and 75 ppm, respectively, for 3 weeks. The average daily feed intake and weight gain were decreased when broiler was exposed to ammonia concentration exceeding 50 ppm (p < .05). The increased abdominal fat and reduced thickness of subcutaneous adipose were found in broilers of 75 ppm group (p < .05). When ammonia exceeded 50 ppm, the content of fat in breast muscle of broiler was increased, and when ammonia was higher than 25 ppm, the fat in liver was increased (p < .05). It showed that the fat content in liver was a sensitive index for broilers exposed to ammonia. Furthermore, ammonia exposure had no significant effect on total cholesterol and triglyceride in serum, but significantly increased the relative mRNA expression of acetyl‐CoA carboxylase (p = .046) and malic enzyme in liver (p = .038), which indicated that ammonia exposure may increase the de novo fat synthesis in liver. In addition, ammonia increased the high‐density lipoprotein cholesterol (p = .02) and activity of hepatic lipase in serum (p < .001), which indicated that ammonia exposure may improve the transportation of cholesterol to liver. To conclude, our results indicated that ammonia exposure might increase the de novo fat synthesis in liver and increased the transportation of cholesterol to liver. The rhythmic locomotor behavior of flies and mice provides a phenotype for the identification of clock genes, and the underlying molecular mechanism is well studied. However, interestingly, when examining locomotor rhythm in the wild, several key laboratory‐based assumptions on circadian behavior are not supported in natural conditions. The rooster crowing ‘cock‐a‐doodle‐doo’ is a symbol of the break of dawn in many countries. Previously, we used domestic inbred roosters and showed that the timing of roosters' crowing is regulated by the circadian clock under laboratory conditions. However, it is still unknown whether the regulation of crowing by circadian clock is observed under natural conditions. Therefore, here we used red jungle fowls and first confirmed that similar crowing rhythms with domesticated chickens are observed in red jungle fowls under the laboratory conditions. Red jungle fowls show predawn crowing before light onset under 12:12 light : dim light conditions and the free‐running rhythm of crowing under total dim light conditions. We next examined the crowing rhythms under semi‐wild conditions. Although the crowing of red jungle fowls changed seasonally under semi‐wild conditions, predawn crowing was observed before sunrise in all seasons. This evidence suggests that seasonally changed crowing of red jungle fowls is under the control of a circadian clock. 相似文献

A radiologic and tomographic study was made of the cranioencephalic topography of canary goats of medium weight and size. In a first step, the exocranialis references corresponding to an axis designed at the level of the meatus acusticus externus and parallel to the basicranial axis were anatomically and radiologically defined. Conventional stereotaxic techniques were then used to reach the standard position of the head in order to obtain macroscopical cross sections at different levels perpendicular to the base of the cranium. Campylobacter jejuni, a Gram-negative rod, is a zoonotic pathogen associated with human acute bacterial gastroenteritis worldwide. The flagellum, composed of more than 35 proteins, is responsible for colonization of C. jejuni in the host gastrointestinal tract as well as inducing protective antibodies against the homologous serotype. In our previous study, we demonstrated that the flagellar capping protein (FliD) is an immunodominant protein that reacted strongly to sera from field chickens. In this communication, we mapped linear immunoreactive epitopes on FliD using a set of 158 synthetic peptides of 15-mer overlapping with 11 amino acid residues on peptide microarrays with sera from field chickens. The results from peptide microarrays showed (1) no cross-reactivity of the immobilized peptides with the secondary anti-chicken antibody in the control incubation, and (2) heterogeneous patterns of sera reacting to the immobilized peptides. The peptides that reacted to more than three chicken sera and had higher averages of fluorescence units were selected for further validation by the peptide ELISA. The results showed peptides 24, 91 and 92 had relatively high reactivity and less variation among 64 individual serum samples, indicating these peptides represented the shared immunodominant epitopes on the C. jejuni FliD protein. These peptides were also recognized by sera from chickens immunized with the purified recombinant FliD protein. The findings of the specific shared linear immunodominant epitopes on FliD in this study provide a rationale for further evaluation to determine their utility as epitope vaccines covering multiple serotypes for chicken immunization, and subsequently, for providing safer poultry products for human consumption. 相似文献
