Blood and faecal samples were studied from 200 Omani goats of both sexes and various age groups, within five different regions in Oman. Survey questionnaires were undertaken on each goat. The serum vitamin B12 values ranged from 105 to >2000 pg/ml, with a mean value of 723.1 pg/ml and a 95% confidence interval of 61-1507 pg/ml. Kids in the age group 1-3 months showed significantly lower levels of serum vitamin B12 than the older animals. Varying levels of serum vitamin B12 were found in different regions of Oman and on farms within the same region. Goats maintained on commercial feed showed higher levels of serum vitamin B12 than those maintained on pasture or fed on household leftover food, as commonly practised in Oman. Coccidial counts were highest in animals with lower levels of serum vitamin B12. The results of this study indicate that there is a wide range of serum vitamin B12 levels in Omani goats and suggests that younger animals, with lower levels of serum vitamin B12, might be more susceptible to vitamin B12 deficiencies than older animals.  相似文献

As a result of a marked decline in dry matter intake (DMI) prior to parturition and a slow rate of increase in DMI relative to milk production after parturition, dairy cattle experience a negative energy balance. Changes in nutritional and metabolic status during the periparturient period predispose dairy cattle to develop hepatic lipidosis and ketosis. The metabolic profile during early lactation includes low concentrations of serum insulin, plasma glucose, and liver glycogen and high concentrations of serum glucagon, adrenaline, growth hormone, plasma β-hydroxybutyrate and non-esterified fatty acids, and liver triglyceride. Moreover, during late gestation and early lactation, flow of nutrients to fetus and mammary tissues are accorded a high degree of metabolic priority. This priority coincides with lowered responsiveness and sensitivity of extrahepatic tissues to insulin, which presumably plays a key role in development of hepatic lipidosis and ketosis. Hepatic lipidosis and ketosis compromise production, immune function, and fertility. Cows with hepatic lipidosis and ketosis have low tissue responsiveness to insulin owing to ketoacidosis. Insulin has numerous roles in metabolism of carbohydrates, lipids and proteins. Insulin is an anabolic hormone and acts to preserve nutrients as well as being a potent feed intake regulator. In addition to the major replacement therapy to alleviate severity of negative energy balance, administration of insulin with concomitant delivery of dextrose increases efficiency of treatment for hepatic lipidosis and ketosis. However, data on use of insulin to prevent these lipid-related metabolic disorders are limited and it should be investigated.  相似文献

旨在探究风柜斗草提取物(Sarcopyramis napalensis Wall extract,SNE)对蛋氨酸胆碱缺乏(MCD)饮食诱导的肝脏脂质沉积的作用与机制。本研究将60只雄性C57BL/6小鼠随机分为6组,每组10只,设置MCS组(MCD饲料的对照饲料组)、MCS+风柜斗草提取物组(400 mg·(kg·bw)-1)、MCD组、MCD+风柜斗草提取物低、中、高剂量组(100、200、400 mg·(kg·bw)-1),造模及药物干预6周。进行肝组织切片HE染色、天狼猩红染色,检测血清碱性磷酸酶(AKP)、谷草转氨酶(AST)、谷丙转氨酶(ALT),肝组织谷胱甘肽过氧化物酶(GSH-Px)、超氧化物歧化酶(SOD)活力水平及丙二醛(MDA)含量以评估肝脂质沉积、肝纤维化以及肝抗氧化情况。使用Western blot技术检测腺苷酸激活蛋白激酶(AMPK)蛋白的激活情况,使用qRT-PCR技术检测固醇调节元件结合蛋白-1C (SREBP-1c)、肝型脂肪酸结合蛋白(L-FABP)、脂肪酸转运蛋白-3(FATP-3)、脂肪酸转运蛋白-4(FATP-4)、细胞间黏附分子-l (ICAM-1)、血管细胞黏附分子-l (VCAM-1)、基质金属蛋白酶-2(MMP-2)、组织金属蛋白酶抑制剂-2(TIMP-2) mRNA表达水平,同时采用LC-MS/MS技术进行肝组织非靶向脂质组学分析。结果表明,给予200、400 mg·(kg·bw)-1剂量的风柜斗草提取物时均可显著改善MCD饲料饲喂的小鼠肝脂肪变性,减少炎症簇以及肝纤维化程度。给予风柜斗草提取物100、200、400 mg·(kg·bw)-1时均可显著降低MCD饲料饲喂的小鼠血清中ALT和AKP的活力以及肝组织中MDA的含量,显著提高小鼠肝中GSH-Px的活力。Western blot及qRT-PCR检测结果显示,风柜斗草提取物显著激活MCD饲料饲喂的小鼠肝组织AMPK;显著抑制MCD饲料饲喂的小鼠肝组织SREBP-1c、L-FABPFATP-3、FATP-4、ICAM-1、VCAM-1、MMP-2、TIMP-2的mRNA表达水平。肝脂质组学检测结果显示,风柜斗草提取物显著降低MCD饲料饲喂的小鼠肝中甘油三酯(TG)、磷脂酰甘油(PG)以及神经酰胺(Cer)的相对含量;显著提高鞘磷脂(SM)以及辅酶Q9相对含量。结果提示,风柜斗草提取物可通过AMPK/SREBP-1c途径抑制TG合成,以及通过抑制FATPL-FABP表达来减少肝对脂肪酸的摄取,从而改善MCD饲料饲喂的小鼠肝脂肪变性;通过提高MCD饲料饲喂的小鼠肝组织中SM和辅酶Q9相对含量以及降低Cer相对含量,改善肝细胞氧化应激损伤情况,进而改善MCD饮食诱导的小鼠肝炎症和纤维化,最终达到改善肝脂质沉积症的作用。  相似文献

Hypophosphatemia associated with hemolytic anemia was diagnosed in five cats with diabetes mellitus and in one cat with idiopathic hepatic lipidosis. The hematocrit began decreasing within 24 to 48 hours after documented hypophosphatemia in each case. The anemia resolved in all five surviving cats. Because of the temporal relationship and lack of other detectable causes, hemolytic anemia was presumed to be caused by hypophosphatemia. There were increased Heinz bodies in three of six hypophosphate-mic cats during episodes of hemolysis. Intravenous potassium phosphate administration corrected the hypophosphatemia in four of five cats. The effective dosages of intravenous phosphate ranged from 0.011 to 0.017 mmol of phosphate/kg/h for 6 to 12 hours. Hypocalcemia (5.4 to 8.7 mg/dL) occurred in four of five cats treated with intravenous phosphate; however, only one cat developed clinical signs attributable to hypocalcemia. Based on this retrospective study, we recommend monitoring serum phosphorus concentration every 6 to 12 hours in cats likely to become hypophosphatemic. Treatment of hypophosphatemia in cats is warranted because of the apparent increased susceptibility of cats to hypophosphatemia-induced hemolysis. Cats with severe hypophosphatemia (≤1.5 mg/dL) should be given oral or parenteral phosphate if contraindications do not exist. (Journal of Veterinary Internal Medicine 1993; 7:266–271. 【目的】 研究6月龄波尔山羊与麻城黑山羊的血液生理生化指标、胴体性状、肌纤维直径、肌内脂肪含量的差异情况,以期为2个品种山羊的饲养管理、引种改良及杂交利用提供参考依据。【方法】 分别选取8只3月龄±5 d、健康的波尔山羊与麻城黑山羊在统一条件下饲养3个月,饲养结束采集血液测定白细胞数(WBC)、红细胞数(RBC)、血红蛋白浓度(HGB)、红细胞比容(HCT)、平均红细胞体积(MCV)等14项血液生理指标及肌酐(CRE)、总胆红素(TBIL)、尿素氮(BUN)、血糖(GLU)、总蛋白(TP)等14项血液生化指标;测定体重、体高、体斜长、胴体重、屠宰率、眼肌面积等12项胴体性状;屠宰后取背最长肌肌肉组织,切片后经苏木素伊红和改良油红O染色,分别检测肌纤维直径和肌内脂肪含量。【结果】 血液生理指标中,波尔山羊的红细胞比容、平均红细胞体积均显著低于麻城黑山羊(P<0.05),平均红细胞血红蛋白浓度显著高于麻城黑山羊(P<0.05),其余指标均无显著性差异(P>0.05);血液生化指标中,波尔山羊的总蛋白、球蛋白、血磷含量与淀粉酶活性均极显著低于麻城黑山羊(P<0.01),钾离子含量极显著高于麻城黑山羊(P<0.01),其余指标均无显著性差异(P>0.05);胴体性状中,波尔山羊的体重、平均日增重、背最长肌重、股二头肌重和胴体重均显著高于麻城黑山羊(P<0.05),眼肌面积极显著大于麻城黑山羊(P<0.01),其余指标均无显著性差异(P>0.05);波尔山羊的肌纤维直径极显著大于麻城黑山羊(P<0.01),肌内脂肪含量显著高于麻城黑山羊(P<0.05)。【结论】 波尔山羊与麻城黑山羊血液指标、屠宰性能均存在较大的差异,结果可为研究波尔山羊与麻城黑山羊的种质特征提供理论依据。  相似文献

绒山羊产绒量主要由遗传决定,产绒量低是制约绒山羊产业化发展的瓶颈。在同一群体中选择高产绒量(>1 000 g)和低产绒量(<480 g)的山羊母羊各3只,其年龄均为3周岁,体重约34 kg;每只实验羊全基因组测序深度为30×,每个样本大约测得8×106 SNP。对比高产组和低产组,利用选择性消除区域基因分析方法筛选出18个基因组区域可能与产绒量相关,从这些区域中筛选出选择信号较强的4个羊绒产量性状相关候选基因,分别是CUL1、FBXL3、YY1、EZH2,这些基因参与昼夜节律信号通路、Wnt/β-catenin信号通路和TGF-β信号通路,而这3个信号通路正是参与绒山羊次级毛囊发育的重要信号通路。研究结果表明,以上4个基因和相应的SNP可以作为高产绒量绒山羊的基因组辅助育种标记,有助于缩短高产绒量绒山羊品种选育进程。  相似文献

选择南江黄羊、南江黄羊×本地黄羊的F1代黄羊以及本地黄羊母羊的生长发育资料,分别对其体重、日增重和体尺等指标进行对比分析,研究南江黄羊对杂交改良效果。结果表明,F1代黄羊体重明显高于本地黄羊(P〈0.05);F1代黄羊与本地黄羊初生-6月龄日增重差异极显著(P〈0.01),初生-12月龄日增重差异显著(P〈0.05);F1代黄羊与本地黄羊6月龄体长、24月龄胸围差异显著(P〈0.05),12月龄体高差异极显著(P〈0.01)。试验结果显示,南江黄羊杂交改良本地黄羊母羊在生长早期效果明显,显著高于本地黄羊。  相似文献

Seven goats were given a single dose of an aqueous extract derived from 30 g (wet weight) of Narthecium ossifragum per kg liveweight. Their serum creatinine and urea concentrations increased to day 5 but then fell to normal by day 10. Serum magnesium increased to day 4 and decreased to normal by day 9. Their serum calcium concentration was lower than normal on days 4, 5 and 6. Histopathological examination of the kidneys of goats killed or found dead 2, 4, 6, 8, 11 or 16 days after dosing revealed tubular epithelial cell degeneration and necrosis. Regeneration of the tubular epithelium and signs of interstitial fibroplast proliferation and fibrosis could be seen in animals killed on days 8, 11, 16 and 42. No signs of liver damage were observed in 3 goats dosed with the insoluble plant material from 40 g (wet weight) Narthecium ossifragum per kg liveweight. The total dose was divided into three doses, which were given intraruminally within 7 h. The activities of aspartate aminotransferase, -glutamyl-transferase and glutamate dehydrogenase remained within the normal range in all 10 goats after dosing.  相似文献

为进一步探讨核黄素缺乏时外周神经变化的发生机理,对琥珀酸脱氢酶、酸性磷酸酶、腺苷三磷酸酶、碱性磷酸酶进行了酶细胞化学研究。结果表明,核黄素不足的鸡,雪旺氏细胞和轴突中琥珀酸脱氢酶活性下降,酸性磷酸酶活性明显升高,定位于雪旺氏细胞膜上的腺苷三磷酸酶和碱性磷酸酶活性均下降。通过对琥珀酸脱氢酶、腺苷三磷酸酶、碱性磷酸酶活性变化的动态观察,证明轻症鸡自愈的现象以及雪旺氏细胞中琥珀酸脱氢酶活性下降可能是形成脱髓鞘、外周神经水肿、导致雪旺氏细胞膜上的腺苷三磷酸酶和碱性磷酸酶活性下降的原因之一,而增生的雪旺氏细胞中琥珀酸脱氢酶活性增强则可能是修复损伤的一种代偿。腺苷三磷酸酶活性下降与神经水肿密切相关。  相似文献

Haemoglobin (Hb) types were studied in three breeds of Omani goats, Batinah (n = 22), Jebel Akhdar (n = 27) and Dhofari (n = 85). Type A was the only adult Hb observed in adult Batinah and Jebel Akhdar goats. In contrast, only 34% of the Dhofari goats were homozygous for Hb A, while 66% were heterozygous for Hb A and Hb B. Dhofari goats with type AB could further be differentiated into those with approximately 67% type A and 33% type B and those with approximately 33% type A and 67% type B. None of the goats was homozygous for type B. Seventeen kids observed from birth exhibited different levels of fetal Hb, dependent upon whether they developed the adult phenotype AA or AB. Kids that became homozygous for type A were born with approximately 90% Hb F and 10% Hb A. In contrast, goats that developed the heterozygous AB phenotype were born with only 30–60% Hb F, the remaining Hb being types A and B. These findings are not in accordance with previous reports that kids are born without any adult Hb. Hb F was no longer detectable in any of the three breeds of goats at 49 days of age. There was no evidence of Hb C, the pre-adult form of Hb.  相似文献

Freshwater turtle species are suffering from anthropocentric‐caused population declines, making preservation of professionally managed populations increasingly important. Turtles under professional care have an increased risk to develop hepatic lipidosis, potentially resulting in early death. Computed tomography can provide an antemortem screening for increased fatty liver composition. A goal of this prospective analytical cross‐sectional study was to assess the hepatic attenuation measured as Hounsfield units (HU) in a wild population of a freshwater turtle species, and then compare hepatic HU to freshwater turtles under professional care. Ninety‐five wild Blanding's turtles (BT; Emydoidea blandingii) as well as 10 Vietnamese Pond turtles (VPT; Mauremys annamensis) and six Northern Snake‐Necked turtles (NSNT; Chelodina oblonga) under professional care underwent CT with quantification of hepatic HU. Hepatic HU were correlated to serum chemistry findings and the presence of follicles was recorded in BT. The mean (±SD) hepatic attenuation for 95 wild BT was 97.5 HU ±9.6. There were significant differences in hepatic attenuation among VPT, NSNT, and BT, with median HU values (range) of 5.39 HU (–6.45 to 61.50), 71.74 HU (59.44‐94.49), and 95.43 HU (74.41‐124.56), respectively. Aspartate aminotransferase (AST) values had a weak negative correlation to HU with a coefficient of –0.85 (P < .001). The hepatic attenuation was significantly higher for individuals undergoing folliculogenesis (P = .007). The decreased HU values were significantly negatively correlated with AST values. Findings supported the use of CT as an aid for guiding future management practices in freshwater turtles.  相似文献

8~12月龄雄性山羊(n=5)人工感染肝片吸虫后,每周定时颈静脉采血1次,用硝酸还原酶法检测血清中一氧化氮(NO)的动态变化,并观察了NO与IgG抗体、嗜酸性粒细胞(EOS)之间的相关性.结果,山羊在感染后第1周,NO含量明显低于对照组,第2~6周或高或低呈波动趋势,自第6周后明显高于对照组,其中第7~8周达高峰,即(93.73±17.233)μmol/L,第7周与对照组相比P<0.01,第8、10周P<0.05;IgG抗体用间接ELISA最早在感染后第2周就可检出,并从第4周起保持高水平,第8~9周达到最高峰,血清中NO含量与IgG水平的相关系数为r=0.511(P<0.05),而对照组的NO与IgG相关系数为r=0.25(P>0.05);感染山羊在整个试验期间的EOS明显升高,其中第8周达峰值(P<0.01).本试验结果表明,NO、IgG和EOS在山羊感染肝片吸虫过程中的变化趋势基本一致并呈正相关,且与虫体在宿主体内的移行、发育密切关联,显示NO、IgG和EOS共同参与宿主抗肝片吸虫感染的作用.  相似文献

试验选用8只平均(15±2)月龄、平均体重(32.08±3.12)kg半同胞内蒙古白绒山羊羯羊,研究自然条件下绒山羊生绒期皮肤脱碘酶(MD)活性与绒毛生长的关系,从酶活性途径探讨绒山羊绒毛生长机理。结果表明:绒毛开始萌发到绒毛生长旺盛期,羊绒生长速度与粗毛生长速度呈高度负相关(r=-0.92,P<0.05);羊绒生长速度与皮肤二型脱碘酶(MDⅡ)活性呈正相关(r=0.95,P<0.05);羊绒生长速度与皮肤三型脱碘酶(MDШ)活性(r=-0.90,P<0.05)及MDШ/MDⅡ比值(r=-0.91,P<0.05)呈负相关;粗毛生长速度与MDⅡ活性存在高度负相关(r=-0.98,P<0.01),与MDШ活性(r=0.90,P<0.05)及MDШ/MDⅡ比值(r=0.97,P<0.01)呈现正相关。因此,皮肤组织中MD活性与绒山羊绒毛生长的生理过程具有相关关系。  相似文献
