Phosphorus (P) deficiency in the diet of teleost fishes decreases the mineral content of bone. The present study investigated the changes in the microanatomy and metabolism of acellular-boned juvenile haddock (Melanogrammus aeglefinus L.) in relation to the P content of the diet and to determine whether bone deformities develop in association with P deficiency. Fish were fed diets containing low (0.42% diet), optimum (1.02% diet) and excess (1.42% diet) levels of P for 8 and 20 weeks. At the end of two experimental periods, fish were subjected to gross examination, x-rayed, and the mineral content of the plasma and the vertebrae was measured. The vertebrae were also subjected to histological, histomorphometric and enzyme histochemical examinations to determine the level of bone formation and resorption. A P-deficient diet caused a delay in bone mineralization. In contrast, excess P in the diet increased matrix deposition and resulted in accelerated mineralization. Counting the numbers of osteoblasts and osteoclasts, and measuring the mineralized bone and the non-mineralized bone suggest that P deficiency in haddock initially increases bone resorption and subsequently decreases bone mineralization followed by a decrease in bone formation. The increase of the number of osteoclasts in P-deficient animals suggests that osteoclasts could be involved in P homeostasis. In the same fish group low bone mineral and deformed bones observed on x-rays, could be the result of a decrease in mineralization of the bone matrix.  相似文献

为探明鲑降钙素(salmon calcitonin, sCT)对鱼类骨组织钙代谢过程的调节机制, 对虹鳟(Oncorhynchus mykiss) 幼鱼进行 sCT 腹腔注射, 并在注射后 24 h 采集鳞组织进行转录组测序, 分析其中长链非编码 RNA (long non-coding RNA, lncRNA)表达水平的变化。结果显示, 注射 sCT 后的虹鳟鳞组织中共鉴定出 847 个差异表达的 lncRNA, 其中 247 个表达上调, 600 个表达下调。GO 注释结果显示, 差异表达 lncRNA 靶基因主要被注释到转录调控、运输、信号转导、膜、细胞质、金属离子结合和核苷酸结合等功能中。KEGG 通路富集结果显示, 差异表达 lncRNA 靶基因在硫胺素代谢通路、炎症介质对 TRP 通道调节、血小板活化、谷氨酸能突触、神经营养因子通路、ARVC 通路和 NF-kappa B 信号通路等通路中显著富集。利用实时荧光定量 PCR (quantitative real-time PCR, qRT-PCR)对随机选取的 6 个差异表达 lncRNA 的表达量进行验证, 结果显示, qRT-PCR 与 RNA-Seq 结果一致。基于上述结果, 本研究筛选到 MSTRG.68909.2、MSTRG.39805.1、MSTRG.121429.1、MSTRG.9137.1 和 MSTRG.43721.1 共 5 个可能参与虹鳟钙代谢的关键 lncRNA, 这些关键基因的筛选鉴别可为探明硬骨鱼类骨代谢的调控机理提供新的切入点, 为虹鳟养殖生产实践提供参考。  相似文献

This study was performed to determine compensatory growth of juvenile olive flounder fed the extruded pellet (EP) with different feeding regimes. Seven treatments with triplicates of different feeding regimes were prepared; α fish was daily fed for 6 d a week throughout 8 wk (8WF); α fish was starved for 1 wk and then fed for 3 wk twice [(1WS + 3WF) × 2]; β fish was starved for 2 wk and then fed for 6 wk (2WS + 6WF); χ fish was starved for 5 d and then fed for 9 d four times [(5DS + 9DF) × 4]; δ fish was starved for 10 d and then fed for 18 d twice [(10DS + 18DF) × 2]; δ fish was starved for 2 d, fed for 5 d, starved for 3 d, and then fed for 4 d four times [(2DS + 5DF + 3DS + 4DF) × 4]; and φ fish was starved for 4 d, fed for 10 d, starved for 6 d, and then fed for 8 d twice [(4DS + 10DF + 6DS + 8DF) × 2], respectively. Total feeding day was all same, 36 d except for control group (48 d). Weight gain of flounder in the 8WF treatment was higher than that of fish in other treatments. And weight gain of flounder in the 2WS + 6WF treatment was higher than that of fish in the (5DS + 9DF) × 4 and (4DS + 10DF + 6DS + 8DF) × 2 treatments. Feed consumption of flounder in the 8WF treatment was higher than that of fish experienced feed deprivation. Feed efficiency ratio (FER), protein efficiency ratio (PER), and protein retention (PR) were not significantly different among treatments. Chemical composition of the whole body of fish with and without liver, except for moisture content of liver, was not different among treatments. T3 level of fish in the 8WF and 2WS + 6WF treatments was higher than that of fish in the (5DS + 9DF) × 4 treatment. It can be concluded that juvenile olive flounder achieved better compensatory growth at 6‐wk refeeding after 2‐wk feed deprivation compared with that of fish with different feeding regimes. And T3 level of fish seemed to partially play an important role in achieving compensatory growth.  相似文献

The existence of a food-entrainable circadian oscillator (FEO) was examined in goldfish Carassius auratus. Single goldfish were exposed to light–dark (LD) 12:12 (lights on 06:00–18:00 h) and fed at 12:00 h (phase I). The photoperiod regime was shifted to constant light (LL) (phase II). The fish were then fed at 06:00 or at 18:00 h (phase III), and all fish were fasted (phase IV). In both groups, three out of eight fish exhibited food-anticipatory activity (FAA) in phases II and III, the free-running period (τ) of locomotor activity was approximately 24.0 h, and circadian rhythms of locomotor activity started to free-run from the previous FAA in phase IV, indicating the existence of an FEO. Next, single goldfish were exposed to LD 12:12 and were fed twice daily at 12:00 and 00:00 h (phase I). The photoperiod regime was shifted to LL (phase II), and the fish were fasted (phase III). In phase II, FAA was observed in three out of six fish for both feeding times. In phase III, a circadian rhythm of locomotor activity was observed in five fish, and circadian rhythms started to free-run from the previous feeding time (either 12:00 or 00:00 h) in three fish. These results suggest that individual goldfish can remember two daily feeding times.  相似文献

Protein-nitrogen flux (the proportions of consumed and absorbed protein-nitrogen partitioned into protein synthesis and growth) was examined in Atlantic salmon, Salmo salar L. Salmon were held in groups and fed high or low rations or starved. Individual food consumption rates were measured using radiography. Fish varied widely in protein growth efficiency (protein growth divided by protein consumption), but this did not correlate with consumption rate, digestive capacity (as measured by absorption efficiency, trypsin levels and pyloric caecal size) or feeding hierarchy rank. Protein synthesis rates, measured in whole-animals, were linearly correlated with protein consumption and assimilation. There was a significant correlation between protein growth efficiency and the efficiency of retention of synthesised proteins. The capacity for protein synthesis and RNA activity were positively correlated with rates of food consumption and growth but were not correlated with protein growth efficiency. It was concluded that individual differences in protein growth efficiency related to differences in synthesis retention efficiency, but not to differences in the capacity for protein synthesis, RNA activity, digestive capacity or feeding hierarchy rank.  相似文献

ABSTRACT:   Effects of feeding time on approaching behavior to food odor in goldfish Carassius auratus were examined using the Y-maze flow-through system to test whether the behavior differed according to feeding time and experimental time. Fish were fed at 08.00, 12.00 or 16.00 hours once a day or three times (08.00, 12.00 and 16.00 hours) a day for at least 3 weeks under a natural photoperiod, and at 22 ± 2°C, preference reactions to food odor were tested at 08.00, 10.00, 12.00, 14.00 and 16.00 hours. The fish fed once a day showed significant positive approaching behavior only at the time when they were normally fed. The fish fed three times a day showed no significant positive approaching behavior. These results suggest that goldfish can remember when food is available and/or that there exists a mechanism to inhibit approaching behavior when food is not available.  相似文献

Portunus trituberculatus broodstock were stocked in plastic tanks to evaluate the effects of starvation and feeding on gonadal development, blood chemistry, fatty acid composition, and expression of vitellogenin (Vtg) and fatty acid-binding protein genes (FABP) in females. Two treatments (starved and fed) were randomly assigned to triplicate groups of 90 swimming crab broodstock (approximately 230 ± 45 g). In the starved treatment, crabs were starved for 30 days, whereas in the fed treatment crabs were fed once a day with clams. The gonadosomatic index decreased significantly in starved crabs (P < 0.05), as did the serum glucose and cholesterol concentrations; conversely, the total protein concentration in serum significantly increased (P < 0.05). In the ovary, there was a significant relative decline of 18:0, 16:1n-7 and 20:1n-9 fatty acids and relative increases of 20:4n-6, 22:6n-3, 18:1n-9 and 20:5n-3 in starved crabs compared to fed crabs (P < 0.05). Relative expression of Vtg in the ovary decreased significantly in starved crabs (P < 0.05), while there was no significant difference in hepatopancreas Vtg expression between starved and fed crabs (P > 0.05). Starvation suppressed gonadal development in female swimming crab broodstock.  相似文献

Two rearing trials were conducted to evaluate the effects of dietary lupin (LM) and rapeseed (RM) meals in gilthead seabream (Sparus aurata L.) and goldfish (Carassius auratus L.) juveniles. Each plant meal was incorporated at the rate of 200 g kg?1 in two distinct diets, which were compared with a fishmeal‐based diet as control. After 1 month, the two plant diets did not influence the whole body growth, but the digestive systems were affected. The splenosomatic index was reduced with two plant meals in goldfish and with RM in seabream. The hepatosomatic index was only reduced in LM‐fed seabream. Cellular characteristics were also affected. The largest liver cells were observed in RM‐fed goldfish suggesting changes in metabolic function. The LM and RM diets stimulated in seabream, especially the reaction in haematopoietic tissues with the proliferation of melano‐macrophages centres, and a tendency for elongated villus height in the anterior intestine thus that possibly compensated for a reduction in digestive function. Such adaptive structural modifications and the absence of degenerative signs allowed concluding that the integrity of the digestive system was maintained in fish fed plant meals.  相似文献

Abstract

A new demand for live black sea bass, Centropristis striata L., is emerging in the fish markets of the northeast United States and Canada. Aquaculturists need to know suitable commercial diets and feeding rates for growing black sea bass to increase profitability. In these experiments, four commercial diets and three feeding rates were compared for juvenile black sea bass (0.8-1.3 g). The diet experiment compared growth rates for fish fed four commercial diets, Rangen Trout and Salmon Starter (R), Nelson and Son's Silver Cup Trout Crumbles (SCT), Nelson and Son's Silver Cup Salmon Crumbles (SCS), and Zeigler Salmon Starter (Z). The feeding rate experiment compared growth rates for fish fed Zeigler Salmon Starter, at 2.5%, 5.0%, and 7.5% levels (dry weight of food/wet weight of fish). At the end of the diet experiment (18 weeks), Tukey's studentized range test (? = 0.05) showed that fish fed diet Z (14.5±0.92 g) were significantly larger than fish fed diet R (11.4±0.93 g). Diets Z and R both produced significantly larger fish than diets SCS (7.3±0.85 g) and SCT (4.5±0.29 g). At the end of the feeding rate experiment (10 weeks), Duncan's multiple range test (? = 0.05) showed that fish fed a 7.5% (4.61±0.24 g) rate and a 5.0% (4.49±0.24 g) rate were not significantly different in weight. Both of these treatments produced significantly larger fish than a feeding rate of 2.5% (3.59+0.21 g).  相似文献

Two experiments were conducted to evaluate the addition of astaxanthin from red yeast, Xanthophyllomyces dendrorhous, in the diets of goldfish, Carassius auratus. The first was designed to investigate the distribution of pigments in different tissues of goldfish and the effect of astaxanthin in the diet. The carotenoid concentration of tissues was not homogenous. The content of pigments in fish caudal fin was the highest followed by those of scales and head. Flesh had the least carotenoid deposition. Fish fed the diet containing 60 mg/kg astaxanthin had increased concentration of pigment in its head (22.6%), scales (45.5%), flesh (31.0%), and fin (21.2%), compared to fish fed basal diet (P < 0.05). Sixty parts per million astaxanthin had no effect on the weight gain and survival rate. High‐performance liquid chromatography analysis showed astaxanthin in its esterified form in goldfish. The second experiment was aimed at determining the dietary level of astaxanthin that improved color of goldfish. Goldfish were fed the same diet supplemented with 0, 10, 20, 40, 60, and 80 mg yeast astaxanthin/kg for 60 d. The deposition of carotenoids in goldfish fed diets supplemented with astaxanthin increased significantly (P < 0.05) after 15 d of feeding compared to that of the fish fed the diet without astaxanthin, but the effect of dosage of astaxanthin in the diets on the color of goldfish was not completely evident until Day 60 (P < 0.05). During the period of 15–45 d, the deposition of pigments in fish did not increase significantly (P > 0.05) in any treatment with the exception of the diet with 40 mg yeast astaxanthin/kg.  相似文献

Triplicate groups of gilthead sea bream, Sparus aurata (10.4 g), were distributed among 27 tanks (12 fish per tank) and reared in flow‐through seawater. A factorial experiment (3 × 3) was designed to include a continuously fed control group and two cycled starvation groups: 1 + 3 (starved 1 d, fed 3 d), 1 + 5 (starved 1 d, fed 5 d). Each of the feeding groups was subjected to one of three feeding frequencies (2, 4, and 6 times per day) over the 60‐d experiment duration. The average final weight of fish in 1 + 3 and 1 + 5 groups were significantly lower than that of the control group. Partial compensation was observed in the starved groups subjected to any of the three feeding frequencies. Regardless of the feeding frequency, control fish consumed less feed than the starved groups. The highest body protein content was found in the control group. The rate of oxygen consumption significantly increased 30 min after the feeding and the magnitude of the effect increased with the feeding frequency. These results suggest that the present cycling starvation schedules did not invoke a full compensation in gilthead sea bream.  相似文献

Intensive grow‐out of goldfish, Carassius auratus (L.), larvae and juveniles in closed systems requires the control of environmental conditions and feeding. This study investigates the use of different types of live food and combinations of live food and dry food in a series of four rearing experiments. Juvenile goldfish can be weaned from Artemia onto live food at about 24 days after the onset of feeding without causing a reduction in growth and survival. The replacement of Artemia by Daphnia at day 10 appears feasible, as growth and survival were not significantly affected. Fish fed decapsulated Artemia cysts grew better than fish fed live Artemia. Within the first 14 days, goldfish juveniles should be fed at least 155 cysts per fish per day to achieve fast growth and to minimize size variation.  相似文献

ABSTRACT: The validity of using the RNA : DNA ratio, glycogen content, and C : N ratio in the columellar muscle as indicators of the nutritional condition in the Japanese turban shell Turbo cornutus was examined. These biochemical indices were compared between fed and starved animals. The fed animals were fed brown algae to satiation for 108 days as a control, whereas the starved animals were not fed for the first 75 days and then fed for next 33 days. All three indices declined during the starvation period, and the values for the starved group were significantly lower than those for the fed group during days 20–75. At day 108, after the re-feeding period, the indices for the starved group were found to have increased. Among the indices, only the RNA : DNA ratio for the starved males responded rapidly to starved conditions and became significantly lower even at day 4, and only the RNA : DNA ratio for the starved males and females recovered to those levels of the fed animals at day 108. The results indicated that the RNA : DNA ratio is the most rapid indicator of nutritional stress among the three indices. However, it is recommended that glycogen content and C : N ratio be used in addition to the RNA : DNA ratio for monitoring the health of T. cornutus as the RNA : DNA ratio showed large variations.  相似文献

In vivo andin vitro techniques were used to examine the influence of various vertebrate peptides on growth hormone (GH) secretion in the goldfish. Tetradecapeptide somatostatin (SRIF-14) was found to inhibit GH secretionin vitro from perifused pituitary fragments, whereas similar concentrations of a salmonid SRIF peptide (sSRIF-25) did not affect GH secretion from the goldfish pituitary fragments. This indicates that SRIF receptors on the goldfish pituitary are very specific for SRIF-14-like peptides. Salmon gonadotropin (GTH)-releasing hormone (sGnRH) was found to elevate serum GH levels in male goldfish. The dopamine antagonist pimozide alone or injected in combination with sGnRH did not influence serum GH levels, although injection of pimozide alone significantly elevated serum GTH levels, in addition to potentiating the effects of sGnRH on GTH secretion. sGnRH stimulated GH secretion from goldfish pituitary fragmentsin vitro, indicating that sGnRH acts directly at the level of the pituitary to stimulate GH secretion in the goldfish. These results suggest that GnRH may also function as a GH-releasing factor in the goldfish, although the release-inhibitory factors for GH and GTH secretion do appear to be separate and distinct. Two human GH-releasing hormone (hGHRH) peptides were found to be ineffective in altering GH secretionin vitro from the perifused pituitary fragments. Consequently, a role for a mammalian GHRH-like peptide in the hypothalamic regulation of GH secretion in the goldfish remains questionable.  相似文献

The influence of sodium fluoride (NaF) on calcium metabolism was examined in nibbler fish (marine teleosts). Two days after the administration of NaF (5 μg/g of body weight) (around 10?4 M in fish), we showed that plasma calcium levels significantly decreased in NaF-treated nibbler fish. In addition, we detected fluoride in the treated scales by use of a scanning electron microscope with an energy-dispersive X-ray microanalysis, indicating that NaF directly affects their scales. Therefore, the influence of NaF on osteoblasts and osteoclasts in the scales was examined. In the scales of NaF-injected nibbler fish, tartrate-resistant acid phosphatase (TRAP) (osteoclastic marker enzyme) decreased, although alkaline phosphatase (osteoblastic marker enzyme) was activated. To confirm the effect of NaF on osteoclasts, furthermore, the mRNA expressions of osteoclastic markers (matrix metalloproteinase-9 and TRAP) were decreased significantly 2 days after incubation. In barred knifejaws, plasma calcium levels decreased as they did in nibbler fish. Therefore, NaF functions in both osteoblasts and osteoclasts and then influences calcium metabolism in marine fish. In the marine environment, high levels of fluoride (1.2–1.5 mg F?/l) (around 10?5–10?4 M) are present in seawater. It is probable that teleosts living in seawater efficiently use fluoride to regulate their blood calcium levels.  相似文献

Total lipid and fatty acid compositions were determined during embryogenesis and larval development in Eurasian perch (Perca fluviatilis). During embryonic development, perch did not catabolize lipids and fatty acids as an energy source. However, during larval development, there was an exponential relationship between the decrease in total lipids and the duration of starving (r 2=0.9957) and feeding (r 2=1). The duration of the starving period (10 days post hatching) was shorter than the feeding period (35 days post hatching). In both starved and fed larvae, there is an apparent preference in utilization of polyunsaturated fatty acids followed by monounsaturated fatty acids. Saturated fatty acids were utilized by neither fed perch larvae nor by starved perch larvae. In starved larvae, palmitoleic 16:1(n-7) and oleic 18:1(n-9) acids were the preferentially monounsaturated fatty acids catabolized and their contribution as energy source from total fatty acids catabolized over the first week was 37.6%. In fed larvae, these 2 nutrients were also the most monounsaturated fatty acids utilized as energy source and possibly also as precursors for others monounsaturated fatty acids biosynthesis. During the same period and among (n-6) polyunsaturated fatty acids, starved perch utilized less linoleic 18:2(n-6) and arachidonic 20:4(n-6) acids than fed larvae despite the fact that the starved perch were in more unfavorable nutritional conditions. In the case of (n-3) fatty acids, starved larvae utilized more linolenic acid 18:3(n-3) and less eicosapentaenoic 20:5(n-3) acid and docosahexaenoic 22:6(n-3) acid than fed perch. Starved larvae probably spared 20:5(n-3) and 22:6(n-3) for physiological functions.  相似文献

Rainbow trout, Oncorhynchus mykiss, were fed diets containing untreated or phytase-treated soybean meal and corn gluten meal with or without 50 mg supplemental zinc kg–1 to determine if dietary phytate caused a zinc (Zn) deficiency. Fish fed diets containing untreated soybean meal and corn gluten meal without supplemental Zn (basal diet) showed no signs of a Zn deficiency after 170 d of weight gain. Weight gain, carcass protein content, total bone Zn, and alkaline phosphatase and carboxypeptidase B activities were not significantly altered in fish fed the basal diet. Dietary phytate removal and supplemental dietary Zn were not required to prevent Zn deficiency in rainbow trout fed a plant-based diet. Although bone Zn concentrations were reduced in fish fed the basal diet, total bone Zn increased in all fish regardless of dietary treatment. Future assessments of Zn status of fish should be based on changes in Zn-dependent metabolism or total bone Zn rather than on bone Zn concentration.  相似文献

The present study was conducted to evaluate growth performance and color enhancement of goldfish, Carassius auratus, fed diets containing 0, 50, 100, 200, and 250 mg kg?1 diet of annatto dye (AD) for 60 days. The survival rate was significantly higher in fish fed 100, 200, and 250 mg AD kg?1 diet over than these fed control and 50 mg AD kg?1 diet (p < 0.05). AD significantly (p <0 .05) increased the pigmentation in the skin and caudal fin of goldfish in a concentration dependent manner (R2 = 0.995, 0.997). The highest amount of total carotenoid deposition in fish skin and fins were given by diets containing 200–250 mg AD kg?1 diet. The highest redness (a*) of 43.21 and yellowness (b*) of 12.53 were obtained by 250 and 50 mg AD kg?1, respectively. The present results show that AD can be successfully used as an alternative natural carotenoid source in goldfish diets at levels of 200–250 mg AD kg?1 diet.  相似文献

The aim of the present study was to elucidate if increasing levels of added vitamin K in the feed influenced fish growth, health or the incidences of bone deformities in Atlantic salmon ( Salmo salar L.) from onset of start feeding to ∼100 g in freshwater. Duplicate groups of Atlantic salmon fry (∼0.20 g) were fed eight levels (0, 2.5, 5, 7.5, 10, 15, 20 and 50 mg menadione kg−1) of the vitamin K derivate menadione nicotinamide bisulphite (MNB) in a regression design for 28 weeks. All fish maintained high growth rates throughout the experiment, and showed no significant differences in specific growth rate, condition factor, whole body proximate analysis, blood coagulation time, vertebra morphology or mechanical properties of vertebrae. We found a dose-response between whole body vitamin K concentration and the dietary MNB supplementation level. Analysis of liver γ-glutamylcarboxylase activity revealed significant dose-dependent differences between groups given the 0, 10 and 50 mg MNB kg−1 diets. In conclusion, Atlantic salmon seems to require low levels of dietary vitamin K, and the amount of vitamin K found naturally in the presently used feed ingredients may be enough to maintain optimal growth, health and bone strength in Atlantic salmon fry from start feeding.  相似文献
