Susceptibility of eight commercial European wheat cultivars to fusarium head blight (FHB) disease caused by Fusarium graminearum , F. culmorum , F. poae and Microdochium nivale (formerly known as Fusarium nivale ) was compared under controlled environment conditions (16°C). FHB did not differentially affect cultivars in terms of disease symptoms, fungal DNA content of grain or deoxynivalenol (DON) contamination. However, the Hungarian-grown cultivars GK-Othalom and Fatima 2 (of Romanian origin) showed greater type V resistance (yield tolerance) to FHB than did the others. Also, nivalenol was produced by F. poae in these two cultivars and in Italian cultivar Norba, but not in other cultivars. Overall, significant relationships were found between the FHB and seedling blight resistance in vitro of these eight cultivars, but such relationships were generally highly dependent on cultivar, and therefore it is likely that the in vitro test is at best measuring components of FHB resistance and/or genotype-specific resistance components.  相似文献

Fusarium culmorum causes head blight, produces toxins and reduces yield and quality of cereals. To prevent damage caused by fusarium head blight (FHB), azole fungicides are mainly applied. The occurrence of insensitivity to azoles is a major problem in agriculture. The present study shows that a tebuconazole insensitive strain of F. culmorum can be readily produced in the laboratory, but that the resulting strain of the fungus is of lower fitness in vitro. Insensitivity was confirmed microscopically and by cell viability and metabolic activity. The tebuconazole insensitive strain shows cross insensitivity to nine important azoles. In addition, plants inoculated with the insensitive F. culmorum strain showed no reduction of FHB symptoms and deoxynivalenol (DON) content after tebuconazole treatment, compared to an inoculation with the sensitive strain. Use of wheat cultivars carrying a high resistance level (i.e. cv. Toras) was the most effective method for reducing symptoms and decreasing DON content, independent from the level of fungicide insensitivity of the F. culmorum strain. In conclusion, resistant cultivars and a fungicide mixture which combines different mechanisms of action in fungal metabolism should be applied to avoid fungicide insensitivity of Fusarium spp. in future.  相似文献

Fusarium head blight (FHB) resistance of 50 cultivars from the National List of winter wheat cultivars approved for sale (or were undergoing trails for approval in 2003) in the UK was compared with 21 reference cultivars from continental Europe which had previously been characterized for resistance. Only three UK National List cultivars (Soissons, Spark and Vector) had stable resistance over trial sites that was significantly greater than that of the FHB susceptible cultivar Wizard. In addition, under moderate disease pressure, 21 of the National List cultivars had levels of the trichothecene mycotoxin deoxynivalenol (DON) above the proposed European Union limit of 1·25 ppm in grain. Surveys show that levels of FHB and DON in the UK crop are currently very low, however, should disease pressure increase for any reason, then an improvement in the overall levels of FHB resistance of UK winter wheat germplasm will be required. In order to infer the origin of resistance and to identify potentially novel resistance, allele sizes of microsatellite (simple sequence repeat, SSR) markers linked to quantitative trait loci (QTL) for FHB resistance were compared between the test cultivars and known, characterized resistance sources. The major FHB resistance QTL from the Chinese cv. Sumai-3 (3BS, 5A and 6B), the Romanian cv. Fundulea F201R (1B and 5A) and the French cv. Renan (5AL) were screened with 17 SSRs. No National List cultivar had haplotypes similar to any of these QTL. However, the highly resistant German reference cultivar Petrus had an identical haplotype to cv. Fundulea F201R on 1B indicating that this cultivar has an allelic FHB resistance QTL at that location.  相似文献

In the western part of Japan, two wheat cultivars, Nishinokaori and Minaminokaori, are currently cultivated for breadmaking. Breadmaking wheat requires a higher protein content compared to the Japanese noodle wheat (the major type of wheat in Japan). This high protein level in the grain is obtained by top-dressing with nitrogen (N) near anthesis. Because such N applications may increase levels of Fusarium head blight (FHB) and consequent mycotoxin [deoxynivalenol (DON) and nivalenol (NIV)] accumulation in the grain, the effect of N application (0, 4, and 8 g/m2) at anthesis on FHB and mycotoxin accumulation in Nishinokaori and Minaminokaori was tested in the greenhouse in 2004 and 2005 and in two fields in 2006. In the greenhouse, plants were spray inoculated at 3, 10, and 20 days after N treatment. In field experiments, colonized maize kernels, which generate ascospores during the testing season, served as inoculum. In all experiments for both cultivars, N application at anthesis significantly increased grain protein as expected, but had no significant effect on FHB and DON and NIV levels in grain. These results suggest that, at least in these cultivars, N can be applied close to anthesis without increasing the risk of FHB and mycotoxin (DON and NIV) accumulation.  相似文献

Wheat is affected by many diseases, in Germany eight fungal diseases are recorded during the cultivar registration process. For a commercially successful cultivar, therefore, at least moderate resistances to important diseases, like yellow rust (YR) and Fusarium head blight (FHB), are necessary. Additionally, in 2013 a regional stem rust (SR) epidemic occurred in Central Germany for the first time for decades. Our objective was to analyze the resistance of 36 commercially grown winter wheat cultivars to YR, FHB, and SR in three individual and one combined inoculation. Appreciable disease severities were achieved for YR and FHB at three to four locations in two years (= seven location × year combinations), for SR at one to two locations in two years (= three location × year combinations). Wheat cultivars showed a significant genotypic variation for all diseases with high heritabilities (0.90–0.95). Interaction between inoculation treatments (individual vs. combined) and wheat genotype was not significant for each of the three diseases. Accordingly, correlations between both inoculation treatments were very high (R2?=?0.95–0.99). Several cultivars showed multi-disease resistance (MDR) to YR, FHB, and SR. In conclusion, resistance ranking among genotypes was not changed when plants were challenged with all three pathogens together compared to factorial inoculations of only one of them. Substituting factorial inoculation trials by multi-pathogen inoculation makes it more efficient to select for MDR in practical breeding programs.  相似文献

Glasshouse studies were undertaken to determine if fungicides used for the control of Fusarium head blight (FHB) result in elevated concentrations of the trichothecene mycotoxin, deoxynivalenol (DON) in harvested wheat grain. Metconazole and azoxystrobin, at double, full, half or quarter the manufacturer's recommended dose rate, were applied to ears of wheat (cv. Cadenza), artificially inoculated with conidia of either Fusarium culmorum or F. graminearum. Metconazole demonstrated high activity against both pathogens, reducing significantly the severity of FHB and the DON concentrations at each of the four dose rates tested when compared to untreated controls. Applications of azoxystrobin significantly reduced FHB and DON compared to unsprayed controls. However, their effectiveness was significantly less than that of metconazole and no dose rate response was observed. Quantification of the amount of trichothecene-producing Fusarium present in harvested grain was determined using a competitive PCR assay based on primers derived from the trichodiene synthase gene (Tri5). Simple linear regression analyses revealed strong relationships between the amount of trichothecene-producing Fusarium present in grain and the DON concentrations (r 2=0.72–0.97). It is concluded that fungicides, applied for the control of FHB, affect DON concentrations indirectly by influencing the amount of trichothecene-producing Fusarium species present in wheat grain. There was no evidence that fungicide applications directly increase the concentration of DON in grain.  相似文献

为探明不同杀菌剂对小麦赤霉病和小麦籽粒DON毒素(包括DON、3-ADON和15-ADON)的控制效果, 采用菌丝生长速率法测定了12种药剂对禾谷镰刀菌野生型菌株PH-1的室内活性, 同时采用液相色谱-串联质谱法(LC-MS)测定了这些药剂对DON毒素的抑制效果, 并开展了小麦赤霉病及籽粒DON毒素的田间防治试验。结果表明, 12种原药对菌丝生长抑制活性强弱依次为氟唑菌酰羟胺>咪鲜胺>戊唑醇>丙硫菌唑>叶菌唑>氰烯菌酯>氟环唑>多菌灵>甲基硫菌灵>吡唑醚菌酯>嘧菌酯>井冈霉素A。氟环唑EC50和EC90离体胁迫均刺激DON毒素产生, 其他杀菌剂EC50和EC90胁迫均抑制DON毒素产生。田间试验结果表明, 200 g/L氟唑菌酰羟胺SC、30%丙硫菌唑OD和20%叶菌唑WP病指防效和DON防效为87.68%~94.77%; 430 g/L戊唑醇SC、25%氰烯菌酯SC、45%咪鲜胺EW、25%氟环唑SC、50%多菌灵WP和70%甲基硫菌灵WP病指防效和DON防效为57.63%%~85.49%; 250 g/L吡唑醚菌酯EC和250 g/L嘧菌酯SC病指防效分别为72.18%和51.98%, DON防效分别为43.06%和-7.96%; 24%井冈霉素A AS病指防效和DON防效分别为42.37%和62.87%。药剂离体和田间控毒效果不完全一致, 赤霉病有效防控是DON防控的前提, 病害防效与DON防效不完全一致, 本研究为小麦赤霉病及籽粒DON毒素防控提供了科学依据。  相似文献

 为明确不同小麦品种(系)对赤霉病的抗性和麦穗组织中DON毒素积累水平,培育和利用抗赤霉病和DON毒素积累的品种提供资源和依据,本研究采用单小花滴注接种法对河南省的106个小麦品种(系)抗赤霉病性进行鉴定分析,并用ELISA测定了病穗组织中DON毒素水平。结果表明不同小麦品种(系)对赤霉病的抗性有显著差异,106个小麦品种(系)中未发现抗病和中抗材料,中感品种(系)有华育198、郑麦103和春丰0021等14个,占13.2%;感病的有曌式2010-06、百农898和中麦63等92个,占86.8%。不同小麦品种(系)籽粒、颖壳和穗轴中DON毒素积累水平有显著差异,籽粒中DON毒素水平在(0.70~287.63)mg/kg之间,其中郑03876、豫保1号和中麦63 的DON毒素水平在2 mg/kg 以下,为抗毒素材料;其他的103个品种DON毒素水平大于2 mg/kg;颖壳和穗轴中的DON毒素水平在(51.03~392.87)mg/kg之间,普遍比籽粒中DON毒素含量高。籽粒中DON毒素水平与小麦品种(系)的平均病害严重度间呈极显著正相关。  相似文献

Fusarium head blight (FHB) is a major threat to wheat production globally, causing not only yield losses but also food and feed contamination. FHB research began at the International Maize and Wheat Improvement Center (CIMMYT) in the early 1980’s, and since then, large-scale FHB screening has been conducted to identify and incorporate new resistance genes into elite CIMMYT germplasm. Promising lines with good FHB resistance were regularly compiled as a Fusarium Head Blight Screening Nursery (FHBSN) and distributed worldwide. The first FHBSN was assembled in 1985, and the most recent two were the 13th and 14th FHBSN that were released in 2011 and 2012, respectively. Candidate lines for a FHBSN came mainly from different CIMMYT wheat breeding programs and were tested for three consecutive years before being included in an FHBSN. FHBSN screening was conducted under strictly standardized field conditions at El Batán, where CIMMYT headquarters is located, using artificial inoculation of F. graminearum strains, whose aggressiveness and DON chemotypes had been previously identified. FHB index was scored at 31 days after inoculation for all lines and DON concentration was measured only for elite lines in their 2nd and 3rd year of evaluation. Haplotyping is a new tool for genetic characterization of FHBSN entries and helps to identify new resistance sources with novel resistance genes and to better target crosses toward diversifying and/or pyramiding resistance. The 13th FHBSN was taken as an example in this paper to show the procedure and strategy for the development of new FHB resistant lines.  相似文献

Fusarium head blight (FHB) is an important disease of wheat, which can result in the contamination of grains with mycotoxins such as deoxynivalenol (DON). Artificial inoculation of flowering ears with conidial suspensions is widely used to study FHB diseases. Our goal was to compare four inoculation treatments in which a conidial suspension was sprayed on flowering ears and to study the effect of the application of moisture during kernel setting and filling with a mist-irrigation system. Ten wheat genotypes were inoculated with a DON-producing Fusarium culmorum strain. Inoculation treatments varied in time of application of the inoculum (morning or evening) and in the method of controlling humidity during inoculation (bagging or mist irrigation). A wet season was simulated with a mist-irrigation system, keeping the crop canopy wet for at least 26 days after flowering. The severity of FHB symptoms (area under disease progress curve (AUDPC)), yield loss and DON contamination in the grains were determined. AUDPC data obtained with the different inoculation treatments were highly correlated (r=0.85–0.95). Mist irrigation after inoculation resulted in a higher mean disease severity, but in a overall lower toxin contamination as compared to the non-irrigated treatments. Genotypic differences in DON accumulation were present: for one wheat line toxin contamination significantly increased when irrigated, while two genotypes accumulated significantly less toxin. The closest relationships (r=0.73–0.89) between the visual symptoms and the DON content were obtained under moderate mean infection pressure. This relation between visual symptoms and the DON content deteriorated at higher infection levels.  相似文献

 镰刀菌产生的脱氧雪腐镰刀菌烯醇(deoxynivalenol,DON)污染小麦后对人畜具有较大的毒害作用。为明确DON毒素在小麦籽粒中的积累数量及其与镰刀菌菌株、接菌数量、小麦品种和病害严重度的关系,试验采用单花滴注方法在5个抗病性不同的小麦品种上分别接种9 个禾谷镰刀菌菌株,每个菌株接种1 × 106 、1 × 105 和1 × 104 个/ mL 3 个分生孢子浓度,并利用ELISA 法测定收获麦粒中DON 毒素含量。结果表明,麦粒中DON 毒素含量差异主要是由于不同禾谷镰刀菌菌株产生毒素能力不同所致。接种菌株8003、4020 的所有麦粒中DON 毒素含量均显著高于相同条件下接种其他7 个菌株的小麦。当接种产毒素能力强的菌株时,小麦抗病品种表现出在一定程度上降低DON 毒素积累的能力。不同接菌浓度对小麦赤霉病的发病程度和麦粒中DON 毒素含量有显著影响,在相同条件下,接菌浓度越高,病害严重度越高,DON 毒素含量也越高;反之,接菌浓度越低,病害严重度越低,DON 毒素含量也越低。  相似文献

Introduction of alien genes into wheat has been proposed as a strategy to breed cultivars with improved resistance to Fusarium seedling blight (FSB) and Fusarium head blight (FHB). In this study, we co-transformed different anti-fungal peptides (AFPs) into an elite wheat cultivar Yangmai11. We identified the genetically stable transgenic wheat lines carrying single or multiple genes by PCR, qRT-PCR and Southern blot analyses. Transgenic wheat lines 451 and 513 expressing two AFPs displayed a consistent, significantly improved overall resistance to FSB and FHB, whereas only FHB resistance was observed from other lines. Furthermore, crude proteins extracted from the lines 451 and 513 showed a clear inhibitory activity against F. graminearum in vitro. Taken together, it was essential to properly combine and express AFPs in transgenic wheat in order to obtain an improved overall resistance to Fusarium pathogens.  相似文献

Fusarium head blight (FHB), caused by Fusarium graminearum sensu stricto (s.s.), causes tremendous annual yield losses in wheat worldwide. Variation of aggressiveness of isolates from individual field populations in terms of FHB infection and deoxynivalenol (DON) concentration in the host are important population parameters reflecting parasitic ability. Our main objective was to estimate the variation of both traits within three populations of F. graminearum s.s., each consisting of 30 single-spore isolates collected from small wheat fields in Germany, and to compare it with 11 isolates of a collection (F. graminearum collection) from four countries. The same isolates were characterized using 19 single-sequence repeat markers. All isolates were spray inoculated on a moderately resistant spring wheat cultivar at two field locations over 2 years (i.e., in four environments). The genotypic proportion of phenotypic variance (σ(2)(G)) within populations was significant (P < 0.01) for both traits, and the σ(2)(G) × environment interaction was even more important for mean FHB severity. Ranges in mean FHB severity and DON concentration in the host were only slightly smaller for the field populations than for the F. graminearum collection. Both traits were significantly (P < 0.05) correlated within and across populations. A further partitioning of σ(2)(G) revealed 72% of σ(2)(G) within and 28% of σ(2)(G) across populations for both traits. Molecular variance of the three populations was similarly distributed (73.6% within versus 26.4% between populations). In view of this high within-field variation for traits of parasitic ability and selection, neutral molecular markers, multiple resistance genes of different origin should be employed in wheat breeding programs to obtain a long-term stable FHB resistance.  相似文献

The data available indicate that aggressiveness of Fusarium graminearum and F. culmorum depends on their deoxynivalenol (DON) and nivalenol-producing capacity: toxin-producing ability correlated closely with the level of aggressiveness measured. This agrees well with other literature findings. However, the resistance of a cultivar influenced DON production significantly. In the most resistant genotypes, toxin contamination remained near zero, whereas the same isolates and inoculum produced very high toxin levels in susceptible cultivars. As toxin levels were correlated with the ratio of Fusarium-damaged kernels (FDK) and this ratio is very low in highly resistant cultivars, the conclusion is that the level of resistance level is more important in governing DON accumulation in a given cultivar than is the aggressiveness of an isolate. In susceptible cultivars, DON producing ability is decisive, but in highly resistant cultivars resistance is the major factor in suppressing disease development and DON accumulation. In different years, the same FDK values were associated with different DON concentrations and this depended very much on the precipitation towards the end of May, the time of inoculation.  相似文献

Wheat crops in southeast Queensland (Qld) and northern New South Wales (NSW) were infected with fusarium head blight (FHB)‐like symptoms during the 2010–11 wheat growing season. Wheat crops in this region were surveyed at soft dough or early maturity stage to determine the distribution, severity, aetiology and toxigenicity of FHB. FHB was widespread on bread wheat and durum, and Fusarium graminearum and/or F. pseudograminearum were diagnosed from 42 of the 44 sites using species‐specific PCR primers directly on spikelets or from monoconidial cultures obtained from spikelets. Stem base browning due to crown rot (CR) was also evident in some samples from both states. The overall FHB and CR severity was higher for NSW than Qld. Deoxynivalenol (DON) concentration of immature grains was more than 1 mg kg?1 in samples from 11 Qld and 14 NSW sites, but only 13 of 498 mature grain samples sourced from the affected areas had more than 1 mg kg?1 DON. DON concentration in straw also exceeded 1 mg kg?1 in eight Qld and all but one NSW sites but this was not linked to DON concentration of immature grains. The proportion of spikelets with positive diagnosis for F. graminearum and/or F. pseudograminearum and weather‐related factors influenced DON levels in immature grains. The average monthly rainfall for August–November during crop anthesis and maturation exceeded the long‐term monthly average by 10–150%. Weather played a critical role in FHB epidemics for Qld sites but this was not apparent for the NSW sites, as weather was generally favourable at all sites.  相似文献

ABSTRACT Fusarium graminearum causes Fusarium head blight (FHB) in small grains worldwide. Although primarily a pathogen of cereals, it also can infect noncereal crops such as potato and sugar beet in the United States. We used a real-time polymerase chain reaction (PCR) method based on intergenic sequences specific to the trichodiene synthase gene (Tri5) from F. graminearum. TaqMan probe and primers were designed and used to estimate DNA content of the pathogen (FgDNA) in the susceptible wheat cv. Grandin after inoculation with the 21 isolates of F. graminearum collected from potato, sugar beet, and wheat. The presence of nine mycotoxins was analyzed in the inoculated wheat heads by gas chromatography and mass spectrometry. All isolates contained the Tri5 gene and were virulent to cv. Grandin. Isolates of F. graminearum differed significantly in virulence (expressed as disease severity), FgDNA content, and mycotoxin accumulation. Potato isolates showed greater variability in producing different mycotoxins than sugar beet and wheat isolates. Correlation analysis showed a significant (P < 0.001) positive relationship between FgDNA content and FHB severity or deoxynivalenol (DON) production. Moreover, a significant (P < 0.001) positive correlation between FHB severity and DON content was observed. Our findings revealed that F. graminearum causing potato dry rot and sugar beet decay could be potential sources of inoculum for FHB epidemics in wheat. Real-time PCR assay provides sensitive and accurate quantification of F. graminearum in wheat and can be useful for monitoring the colonization of wheat grains by F. graminearum in controlled environments, and evaluating wheat germplasms for resistance to FHB.  相似文献

ABSTRACT Barley has two flowering types, chasmogamous (open-flowering) and cleistogamous (closed-flowering). We examined the effect of the timing of Fusarium graminearum infection on Fusarium head blight (FHB) and mycotoxin accumulation in barley cultivars with different flowering types using greenhouse experiments. In the first experiment, 13 cultivars were spray inoculated at two different developmental stages, and the severity of FHB was evaluated. The effect of the timing of infection differed among cultivars. Cleistogamous cultivars were resistant at anthesis but susceptible at 10 days after anthesis, whereas chasmogamous cultivars were already susceptible at anthesis. In the second experiment, five cultivars were inoculated at three different developmental stages and the concentrations of deoxynivalenol (DON) and nivalenol (NIV) in mature grain were analyzed. Cleistogamous cultivars accumulated more mycotoxins (DON and NIV) when inoculated 10 or 20 days after anthesis than when inoculated at anthesis, whereas chasmogamous cultivars accumulated more mycotoxins when inoculated at anthesis. Thus, the most critical time for F. graminearum infection and mycotoxin accumulation in barley differs with cultivar, and likely is associated with the flowering type. Late infection, even without accompanied FHB symptoms, was also significant in terms of the risk of mycotoxin contamination.  相似文献

Twenty four isolates of Fusarium graminearum, half of which were 3-acetyldeoxynivalenol (3-ADON) and half 15-acetyldeoxynivalenol (15-ADON) chemotypes, were tested for their ability to produce deoxynivalenol and to cause Fusarium head blight (FHB) in spring wheat cultivars. The objectives of this study were to determine (1) whether 3-ADON isolates differ in aggressiveness, as measured by the FHB index, and DON production from 15-ADON isolates under field conditions, and (2) whether the performance of resistant host cultivars was stable across isolates. Field tests of all isolates were conducted with three replicates at each of two locations in Canada and Germany in 2008 with three host genotypes differing in FHB resistance level. The resistant host genotype showed resistance regardless of the chemotype or location. The differences between mean FHB indices of 3-ADON and 15-ADON isolates were not significant for any wheat genotype. In contrast, average DON production by the 3-ADON isolates (10.44 mg kg−1) was significantly (P < 0.05) higher than for the 15-ADON isolates (6.95 mg kg−1) at three of the four locations where moderately resistant lines were tested, and at both locations where susceptible lines were evaluated. These results indicate that 3-ADON isolates could pose a greater risk to food safety. However, as the mean aggressiveness and DON production of 3-ADON and 15-ADON chemotypes was similar on highly resistant lines, breeding and use of highly resistant lines is still the most effective measure of reducing the risks associated with DON in wheat.  相似文献

DON detoxification, mainly through glucosylation into DON-3-O-glucose, has been correlated with resistance to FHB. A UDP-glucosyltransferase from the model cereal species Brachypodium distachyon has been shown to confer resistance both to initial infection and to spike colonization (type I and type II resistances, respectively). Here, the functional characterization of transgenic wheat lines expressing the Bradi5g03300 UGT gene are described. The results show that, following inoculation with the fungal pathogen, these lines exhibit a high level of type II resistance and a strong reduction of mycotoxin content. In contrast, type I resistance was only weakly observed, although previously seen in B. distachyon, suggesting the involvement of additional host-specific characteristics in type I resistance. This study contributes to the understanding of the functional relationship between DON glucosylation and FHB resistance in wheat.  相似文献
