Summary A survey to detect antibodies againstAnaplasma marginale, using the Card Test, was conducted on serum samples collected from cattle of Cordoba Department and north-eastern region of the Department of Antioquia in the Republic of Colombia. The samples were from native cattle of Spanish descent(Bos taurus), Zebu(Bos indicus) and various Zebu-native crosses. The age range was between three and eight year olds. The results revealed that90·3 per cent of the samples were positive. All breeds showed a high prevalence of antibodies.

Resumen Se llevó a cabo un reconocimiento epidemiológico para detectar anticuerpos deAnaplasma marginale, mediante la utilización de la prueba de la tarjeta. Los sueros fueron colectados en el Departamento de Córdoba y la región nororiental del Departamento de Antioquia. El ganado examinado fue nativo (Bos taurus), Cebú (Bos indicus), y varios cruces de Cebú y nativo. La edad de los animales osciló entre tres y ocho años. Los resultados dieron una positividad del 90.3 por ciento en el total de sueros colectados. Todas las razas tuvieron gran prevalencia de anticuerpos.

Les échantillons de sang provenaient de taurins locaux de descendance espagnole (Bos taurus) de zébus (Bos indicus) et de sujet à divers degré de croisement. Leur âge se situant entre trois et neuf ans. Les résultats obtenus ont montré que 90.3 p. 100 des échantillons étaient positifs avec une forte proportion d'anticorps.

Two experiments evaluated the effects of conventional and natural feedlot management systems (MS) on ractopamine-HCl (RAC) response in yearling steers. Feedlot performance, carcass characteristics, skeletal muscle gene expression, and circulating IGF-I concentrations were measured. The conventional system included a combined trenbolone acetate and estradiol implant, Revalor-S (IMP), as well as monensin-tylosin feed additives (IA). Treatments were arranged in a 2 x 2 factorial and included: 1) natural (NAT): no IMP-no IA, no RAC; 2) natural plus (NAT+): no IMP-no IA, RAC; 3) conventional (CON): IMP-IA, no RAC; and 4) conventional plus (CON+): IMP-IA, RAC. In Exp. 1, one hundred twenty crossbred steers (initial BW = 400 +/- 26 kg) were allotted randomly to treatment in a randomized complete block design (BW was blocking criteria); pen was the experimental unit. In Exp. 2, twenty-four individually fed crossbred steers (initial BW = 452 +/- 25 kg) were used in a randomized complete block design (BW was blocking criteria) and assigned to the same treatments as Exp. 1, with 6 steers/treatment. In Exp. 2, serum was harvested on d 0 and 31 and within the 28-d RAC feeding period, at d 0, 14, and 28. Longissimus biopsy samples were taken on d 0, 14, and 28 of the RAC feeding period for mRNA analysis of beta-adrenergic receptors and steady-state IGF-I mRNA. In Exp. 1, ADG, G:F, final BW, and HCW were greatest for CON+ (P < 0.01). During the final 37 d, RAC increased ADG (P = 0.05) and increased overall G:F (P = 0.02). Marbling score was reduced (P = 0.02), and yield grade was improved with RAC (P = 0.02), but RAC did not affect dressing percentage (P = 0.96) or HCW (P = 0.31). In Exp. 2, MS x RAC interactions were detected in ADG and G:F the last 28 d, overall ADG and overall G:F, final BW, and HCW (P < 0.01). Dressing percentage, yield grade, and marbling score were not altered by MS or RAC (P > 0.10). Circulating IGF-I concentration was increased on d 31 by the conventional MS, and concentration was greater throughout the study than NAT steers (P < 0.01). Circulating IGF-I concentrations were not changed by RAC (P = 0.49). Abundance of beta(1)-AR mRNA tended to increase (P = 0.09) with RAC, but RAC did not affect beta(2)-AR, beta(3)-AR, or IGF-I mRNA (P > 0.40). Management system did not affect beta(1)-AR, beta(2)-AR, beta(3)-AR, or IGF-I mRNA (P > 0.18), yet a trend (P = 0.06) for MS x RAC for beta(2)-AR mRNA was detected. These results indicate that response to RAC is affected by feedlot management practices.  相似文献

Studies were made of physicochemical and immunochemical characteristics of Brucella abortus agglutinating and non-agglutinating antibodies in the sera of cattle repeatedly injected with living B. abortus (Strain 1119). Both agglutinating and non-agglutinating antibody were shown to be IgG1, and by immunodiffusion against rabbit anti-cattle gamma-globulin, agglutinating antibody gave a precipitation line of identity with that given by non-agglutinating antibody. Whilst agglutinating antibody increased clearance of antigen from the blood of passively protected mice, non-agglutinating antibody did not enhance clearance. Determination of the spleen infection index in mice pre-treated with agglutinating and non-agglutinating antibody showed that in animals passively immunized with non-agglutinating antibody the number of living (infecting) bacteria was approximately 4 times higher than in the case of agglutinating antibody. The possible potentiation of chronic B. abortus infection by non-agglutinating antibody is discussed.  相似文献

OBJECTIVE: To determine the effect of maternally derived antibodies on induction of protective immune responses against bovine viral diarrhea virus (BVDV) type II in young calves vaccinated with a modified-live bovine viral diarrhea virus (BVDV) type I vaccine. DESIGN: Blinded controlled challenge study. ANIMALS: 24 neonatal Holstein and Holstein-cross calves that were deprived of maternal colostrum and fed pooled colostrum that contained a high concentration of (n = 6) or no (18) antibodies to BVDV. PROCEDURE: At 10 to 14 days of age, 6 seropositive and 6 seronegative calves were given a combination vaccine containing modified-live BVDV type I. All calves were kept in isolation for 4.5 months. Six calves of the remaining 12 untreated calves were vaccinated with the same combination vaccine at approximately 4 months of age. Three weeks later, all calves were challenged intranasally with a virulent BVDV type II. RESULTS: Seronegative unvaccinated calves and seropositive calves that were vaccinated at 2 weeks of age developed severe disease, and 4 calves in each of these groups required euthanasia. Seronegative calves that were vaccinated at 2 weeks or 4 months of age developed only mild or no clinical signs of disease. CONCLUSIONS AND CLINICAL RELEVANCE: Results indicate that a single dose of a modified-live BVDV type-I vaccine given at 10 to 14 days of age can protect susceptible young calves from virulent BVDV type II infection for at least 4 months, but high concentrations of BVDV-specific maternally derived antibodies can block the induction of the response.  相似文献

Sugarcane molasses is a widely used animal feed by-product, but is concentrated in S (approximately 1%, DM basis) and has been shown to reduce the Cu status of cattle. Dietary S may also antagonize Se; therefore, two 90-d studies were conducted with forage-fed, yearling steers (12 pens; 2 steers/pen for each study) to investigate the impact of molasses supplementation on measures of Se status. In Exp. 1, steers were assigned isonitrogenous supplements with equivalent amounts of TDN from 2 sources (molasses or corn). Supplemental Se was provided (3.0 mg of Se/d; Na selenite) to both treatments. After 90 d of supplementation, steers provided corn diets had greater (P = 0.02) liver Se concentrations and tended (P = 0.07) to have greater ADG compared with steers supplemented with molasses. Irrespective of treatment (P >/= 0.54), plasma Se concentrations decreased (P < 0.001) and plasma glutathione peroxidase activity increased (P < 0.001) from d 0 to 90. In Exp. 2, sources of supplemental Se (2.5 mg/ d), fed within molasses supplements, were compared. A panel of 8 monoclonal antibodies to rabies glycoprotein antigen was used to characterize the modified-live virus vaccines marketed in the United States during the last 10 years. Thirteen of 14 rabies virus isolates from 11 dogs, 2 cats, and 1 fox suspected of developing vaccine-induced rabies were shown to have reactivity patterns that were identical to the vaccine administered. Reactivity patterns for 20 rabies isolates from human beings, wild animals, or domestic animals with no history of recent vaccination with modified-live virus rabies vaccine were different from those obtained for vaccines.  相似文献

Neonate progeny from 3 Anaplasma-free (clean) and 5 Anaplasma-carrier cows were splenectomized and each was challenge exposed with 5 ml of carrier blood. Prepatent times were between 18 and 25 days in calves born of clean cows and between 21 and 36 days in progeny from carrier dams. The lowest packed cell volume values in the clean group occurred at 25 to 39 days after the challenge inoculation and at 29 to 47 days in the carrier group. Highest parasitemias in the clean-calves ranged between 13% and 51% in 25 to 35 days and between 35% to 64% in 38 to 43 days in the carrier calves. Seven splenectomized calves were inoculated with 60 ml of whole blood from progeny of known Anaplasma-free or Anaplasma-carrier cows. After 183 days, all but 1 calf remained free of anaplasmosis. A 1% parasitemia was first observed in that calf 12 days after inoculation with blood from a calf which showed signs of acute anaplasmosis at birth. The infected neonate's dam had recovered from acute anaplasmosis infection during the middle of the second trimester of the gestation. Although not statistically significant, colostral antibodies and/or other maternal factors did not seem to completely protect progeny, but lengthened the prepatent period and delayed anemia and the climax of parasitemia. Further, it was determined that it was possible for an animal affected with acute anaplasmosis before the 190th day of the gestation to transmit anaplasmosis in utero.  相似文献

Longissimus muscle area and fat thickness were measured following weaning, at yearling, and prior to harvest using real-time ultrasound, and corresponding carcass measurements were recorded 3 to 7 d following the preharvest scan in composite steers (n = 116, 447 +/- 19 d), bulls (n = 224, 521 +/- 11 d), and heifers (n = 257,532 +/- 12 d). Although fat deposition was limited in bulls and heifers from weaning to yearling, coefficients of variation ranged from 8.46 to 13.46% for muscle area, and from 27.55 to 38.95% for fat thickness, indicating that significant phenotypic variance exists across genders. Residual correlations, adjusted for the effects of year of birth, gender, and age at measurement, were high and ranged from 0.79 to 0.87 among ultrasound and carcass measures of muscle area. Residual correlations among ultrasound and carcass measures of fat thickness were also high, ranging from 0.64 to 0.86. Weaning and/or yearling ultrasound muscle area yielded similarly accurate predictions of carcass muscle area. Yearling ultrasound fat thickness accounted for 13% more of the observed variance in carcass fat thickness than the weaning ultrasound measure in single-trait prediction models. When both weaning and yearling ultrasound measures were used to predict carcass fat thickness, partial R2 values were 0.15 and 0.61 for weaning and yearling ultrasound fat thickness, respectively. The difference between predicted and carcass measures with respect to muscle area (fat thickness) was less than 6.45 cm2 (2.5 mm) for 80.2 to 88.9% (90.3 to 95%) of animals. Preharvest ultrasound measures yielded standard errors of prediction of less than 4.95 cm2 for muscle area and 1.51 mm or less for fat thickness. These results indicate that ultrasound measures taken between weaning and yearling provide accurate predictors of corresponding carcass traits in steers, bulls, and heifers.  相似文献

Eight rumen-cannulated steers (initial wt 330kg) were adapted to a mixed alfalfa-grass hay diet for 30 d and abruptly changed to complete mixed diets of corn silage, snapped ear corn and a corn-based supplement to determine the effects of buffers on diet adaptation, digestion and ruminal metabolism. The diets contained: 1) no buffer, 2) .5% magnesium oxide (MgO), 3) 1.0% sodium bicarbonate (NaHCO3) and 4) .5% MgO and 1.0% NaHCO3, as a percentage of diet dry matter (DM). Digestion, metabolism and ruminal characteristics were measured in each of 2 wk immediately after the diet change. The animals were then adapted to the mixed alfalfa-grass hay diet, re-randomized and assigned to the four diets, thus four steers consumed each diet. Intakes and digestibilities of DM were generally greater for the diets containing buffers. The most notable differences were a greater DM intake with added NaHCO3 and an improved DM digestibility with added MgO. The higher DM digestibility with MgO was apparently related to improved neutral detergent fiber (NDF) and starch digestion. Fecal pH was significantly increased with MgO addition. Because of the greater intake and digestibilities, the amount of DM, NDF, and starch digested tended to be greater for the buffered diets. There were no diet X week interactions for intake and digestibilities, thus the responses observed existed during both wk 1 and 2 after the change in diets. Equine protozoal myeloencephalitis (EPM) is the most important protozoal disease of horses in North America and it is caused by Sarcocystis neurona. Natural cases of encephalitis due to S. neurona have been reported in raccoons, Procyon lotor. We examined 99 raccoons for agglutinating antibodies to S. neurona using the S. neurona agglutination test (SAT) employing formalin-fixed merozoites as antigen. Raccoons originated in Florida (N=24, collected in 1996), New Jersey (N=25, collected in 1993), Pennsylvania (N=25, collected in 1999), and Massachusetts (N=25, collected in 1993 and 1994). We found that 58 (58.6%) of the 99 raccoons were positive for antibodies to S. neurona using the SAT; 44 of 99 raccoons (44%) had titers of ≥1:500. This prevalence is similar to the reported seroprevalence of 33–60% for S. neurona antibodies in horses from the United States using the Western blot test.  相似文献

The influence of age and maternal immunity on the development and duration of postvaccinal humoral response against swine influenza viruses (SIV) were investigated under experimental conditions. Piglets born to immune and non-immune sows were vaccinated twice with bivalent inactivated vaccine. Vaccination was done according to 5 different schedules: 1+4, 1+8, 4+8, 8+10 or 8+12 weeks of age. Antibodies to the haemagglutinin type 1 and 3 were determined using the haemagglutination inhibition (HI) test. Maternally derived antibodies (MDA) against H1N1 and H3N2 in the serum of unvaccinated piglets born to immune sows were above the positive level until about 13-14 and 9-10 weeks of life, respectively. No serological responses were seen in any of the groups after the first vaccination. After the second dose of vaccine production of antibodies was observed even before the complete disappearance of maternal antibodies. MDA, however, were associated with reduced antibody response. In MDA-negative piglets, an active humoral postvaccinal response was developed in all vaccinated pigs. The age at which the vaccine was given was associated with the differences in the magnitude of antibody response to SIV. In general those pigs that were vaccinated for the first time at the age of 1 week, developed lower maximum titres after the second vaccination, and become seronegative earlier than pigs that were vaccinated for the first time at 4 or 8 weeks of age.  相似文献

A serologic survey was conducted in yearling cattle imported into Alberta feedlots from Montana during October 2001 to estimate the prevalence of antibodies to bluetongue virus (BTV) and Anaplasma marginale in Montana yearling cattle. The apparent prevalence of antibodies to BTV when the competitive enzyme-linked immunosorbent assay (cELISA) was used was 0.37% (21/5608). Test positive cELISA samples were also all positive when tested by virus neutralization (VN) and they reacted to 1 or more BTV serotypes, including 2, 10, 11, 13, and 17. The apparent prevalence of antibodies to A. marginale when a recombinant cELISA (rcELISA) was used with a positive cutoff at 30% inhibition was 1.93% (108/5608). When the rcELISA positive cutoff was at 42% inhibition, the apparent prevalence was 0.73% (41/5608). After the reported sensitivity and specificity of the test had been accounted for, the A. marginale antibody results were consistent with a population that was either free of exposure or had a very low prevalence for A. marginale.  相似文献

Soluble extracts were prepared from sporulated oocysts, unsporulated oocysts and merozoites of Eimeria crandallis, E faurei and E ovinoidalis. They were assessed for antigenicity and specificity by ELISA using rabbit antisera to sporulated oocysts or merozoites. Antibody levels were examined in sera from colostrum-deprived coccidia-free lambs, conventionally reared lambs and lambs which had received experimental infections. Maternal antibody was demonstrated in colostrum and in serum taken at 24 hours from all conventionally reared animals but not colostrum-deprived animals. Antibody levels in conventional animals dropped over the first five weeks of life and rose again during the next five weeks. Antibody was not detected in coccidia-free animals. Monospecific infections of E faurei or E ovinoidalis demonstrated antibody responses to primary and secondary infections. Some specificity of response was suggested with E faurei infection. The antigen preparations showed considerable cross-reactions between species. These serum antibody responses, although appearing too late for individual diagnosis, may assist diagnosis on a flock basis.  相似文献

OBJECTIVE: To determine the prevalence and effect of Neospora caninum infection and persistent infection (PI) with bovine viral diarrhea virus (BVDV) on weight gain, morbidity, and mortality rate in dairy-breed steer calves located on a feedlot in California. DESIGN: Prospective cohort observational study. ANIMALS: 900 dairy-breed steer calves in 2 pens. PROCEDURES: The 3- to 4-month-old calves were evaluated for serum antibodies against N caninum and infection with BVDV at entry to the feedlot. Five months later, sera were again analyzed for anti-N caninum antibodies; calves that were determined to have BVDV infection initially were retested to evaluate PI status. Average daily gain, morbidity, and deaths were recorded for all calves. RESULTS: Among 900 calves, prevalence of N caninum infection was 16.7% (95% confidence interval, 14.3% to 19.3%); prevalence of BVDV-associated PI was 0.2% (95% confidence interval, 0.03% to 0.9%). Morbidity rate and time to first illness were not significantly different between calves that were seropositive or seronegative for N caninum. At the second sample collection, weight and average daily gain of calves that were seropositive for N caninum was less than that of seronegative steers in 1 pen, whereas these measures did not differ between groups in the other pen. Statistical power was insufficient to evaluate the effect of BVDV PI on any outcome measurement. CONCLUSIONS AND CLINICAL RELEVANCE: Although N caninum serostatus had no significant effect on morbidity rate, some seropositive calves had reduced growth, compared with seronegative calves, 5 months after entry to the feedlot.  相似文献

Due to increased production of ethanol, abundance of distillers grains (DG) is increasing. Steers (n = 176) were assigned to 1 of 5 treatment groups: steam-flaked corn (SFC), 10% dry DG (DDG), 10% wet DG (WDG), 20% WDG, or 30% WDG. The objectives were to determine the effects of feeding greater amounts of WDG, or DDG on meat quality. Steaks, 2.54 cm, were cut from strip loins and identified for simulated retail display, Warner-Bratzler shear force analysis, palatability, and fatty acid composition. Steaks from cattle fed 10% WDG and 30% WDG had smaller (P < 0.05) Warner-Bratzler shear force values than steaks from cattle fed 20% WDG. Trained sensory panelists found no differences (P > 0.05) in overall tenderness and off-flavors. No differences were found in total SFA and MUFA composition among treatments; however, 20% and 30% WDG had a greater proportion of PUFA and n-6 fatty acids than 10% WDG. No differences were found during simulated retail display between various amounts of WDG. Further research needs to be conducted to evaluate methods that aid in increasing shelf life of steaks from cattle fed greater rates of WDG.  相似文献

A seroepidemiological survey of Toxoplasma gondii infection in sheep and goats was conducted in the Madrid region of Spain. Sera were collected from 60 herds, for which farming management information and other relevant data for their characterization were also obtained through a questionnaire. The seroprevalence was 11.8% (64 out of 541), using the modified (2-mercaptoethanol) direct agglutination technique with a 1:64 cut-off titre. The relationship between seropositivity and the variables in the questionnaire was assessed by multivariate analysis. Four variables were found to be significantly associated with seroprevalence. Two of them, the presence of cats and a previous history of abortion outbreaks in the farm, were factors known to be linked with toxoplasmosis, indicating the validity of the serological data. Seropositivity was also related to a lack of replacements in the preceding year.
