Swine were vaccinated with adsorbate bacterin made from Erysipelothrix rhusiopathiae of serotype 2 and were subsequently allotted to 4 exposure groups, each of which was exposed to 1 of the strains of E rhusiopathiae of serotypes 1, 2, 9, or 10. Mice were vaccinated with the same bacterin and were subsequently allotted to 60 exposure groups which were exposed to 60 strains of E rhusiopathiae, comprising 10 strains each of serotypes 1, 2, 4, 9, 10, and 11. Response to challenge of immunity in swine was determined by the presence of clinical signs of acute generalized erysipelas; response in mice was determined by the quantal (live-dead) method. Vaccinated swine were as susceptible to the strain of serotype 10 as were nonvaccinated control swine, whereas vaccinated swine were immune and control swine were susceptible to the strains of serotypes 1 and 2. The strain of serotype 9 was not sufficiently virulent to induce acute generalized erysipelas, even in control swine. Arthritis was not prevented by vaccination, but its frequency and severity were less in vaccinated swine exposed to strains of serotype 1 or 2 than in those exposed to strains of serotype 9 or 10. Vaccinated mice were significantly (P less than 0.05) more susceptible to the strains of serotype 10 than to those of any other serotype tested.  相似文献

To investigate the influence of antibiotics used as feed additives on the immune response to erysipelas live vaccine, the pig inoculation test was applied. Avilamycin, oxytetracycline quaternary salt, enramycin, virginiamycin and tylosin phosphate were selected as test antibiotics. Five experimental feeds containing each antibiotic at the highest concentration permitted for feed additives in Japan, and the basal diet lacking antibiotics were examined. Twenty-nine pigs were divided into six groups. At first all the groups were fed with the antibiotic-free basal diet for 7 days, and then each group received the experimental feeds. On the 14th day after feeding with test feeds all the pigs, except for one control pig in each group, were immunized with the vaccine and all the pigs were then challenge-exposed to a virulent strain of Erysipelothrix rhusiopathiae 14 days after vaccination. The clinical response was observed every day for 14 days. In all the groups, most of the vaccinated pigs did not develop any clinical signs of acute erysipelas after the challenge exposure, whereas non-vaccinated control pigs died or showed severe generalized erythema with profound depression and anorexia. No differences in the protection against the challenge exposure were observed among the groups. Therefore, the present results suggest that these selected antibiotics would not interfere with the immune effect of the vaccine if given at the usual concentrations used for feed additives.  相似文献

Swine and mice were vaccinated with standard erysipelas adsorbate bacterins made from Erysipelothrix rhusiopathiae of serotype 2 and were subsequently exposed to pathogenic strains of E rhusiopathiae, serotypes 1, 2, 4, 9, 10, and 11. Response to challenge of immunity in swine was determined by presence of urticarial lesions at the sites of intradermal injection of culture; response in mice was determined by the quantal (live-dead) method. After vaccination with standard bacterins, swine and mice were significantly more susceptible (P less than of equal to 0.01) to infection with strains of serotypes 9 and 10 than with strains of serotypes 1, 2, 4, or 11. An adsorbate bacterin made from the challenge strain of serotype 10 induced specific immunity to homologous challenge exposure in swine but not in mice. In order to prove the effects of mass application of oral erysipelas vaccine via drinking water, in a farrow-to-finish production unit in Croatia, the growing-finishing animals were divided into 3 groups and treated as follows:--Group 1 (n=199) was vaccinated intramuscularly against swine erysipelas at 1 week and 3 weeks after arrival in the growing-finishing facility with a swine erysipelas bacterin.--Group 2 (n=199) were vaccinated at the same time with an avirulent culture of Erysipelothrix rhusiopathiae oral vaccine through drinking water.--Group 3 (n=200) was not vaccinated. Animals with clinical signs of swine erysipelas, chronic progressive arthritis at slaughter, mortality, average daily weight gain during the growing-finishing phase were evaluated. None of the pigs in the groups 1 and 2 showed clinical signs typical for acute swine erysipelas. Twenty-four of the pigs (12 %) in group 3 had pyrexia and skin lesions typical for swine erysipelas. Fifteen pigs in group 1, 13 pigs in group 2, and 63 pigs in group 3 had chronic progressive arthritis (group 1 and 2 vs. group 3: P < 0.01). No significant differences in mortality were recorded between the groups. Group 1 and 2 had higher (P < 0.05) average daily weight gains compared with the group 3.  相似文献

We compared 3 modified-live pseudorabies virus (PRV) vaccine strains, administered by the intranasal (IN) or IM routes to 4- to 6-week-old pigs, to determine the effect of high- and low-challenge doses in these vaccinated pigs. At the time of vaccination, all pigs had passively acquired antibodies to PRV. Four experiments were conducted. Four weeks after vaccination, pigs were challenge-exposed IN with virulent virus strain Iowa S62. In experiments 1 and 2, a high challenge exposure dose (10(5.3) TCID50) was used, whereas in experiments 3 and 4, a lower challenge exposure dose (10(2.8) TCID50) was used. This low dose was believed to better simulate field conditions. After challenge exposure, pigs were evaluated for clinical signs of disease, weight gain, serologic response, and viral shedding. When vaccinated pigs were challenge-exposed with a high dose of PRV, the duration of viral shedding was significantly (P less than 0.05) lower, and body weight gain was greater in vaccinated pigs, compared with nonvaccinated challenge-exposed pigs. Pigs vaccinated IN shed PRV for fewer days than pigs vaccinated IM, but this difference was not significant. When vaccinated pigs were challenge-exposed with a low dose, significantly (P less than 0.05) fewer pigs vaccinated IN (51%) shed PRV, compared with pigs vaccinated IM (77%), or nonvaccinated pigs (94%). Additionally, the duration of viral shedding was significantly (P less than 0.05) shorter in pigs vaccinated IN, compared with pigs vaccinated IM or nonvaccinated pigs. 应用本实验室构建的嵌合型猪圆环病毒(PCV1—2)及真核表达质粒pcDNA3.1/V5-His-ORF2作为免疫原免疫母源抗体ELISA效价在0.07~0.60不等的商品猪,9头猪随机分为4组,1组(3头)肌肉注射免疫10^3.5 TCID50的PCV1-2/头,2组(2头)肌肉注射真核表达质粒200μg/头,3组(2头)肌肉注射空载体(pcDNA3.1)200μg/头,4组(2头)不免疫作为攻毒对照组。于免疫后42d,PCV1—2组及真核表达质粒组产生了PCV2抗体。免疫后42d所有组攻毒PCV2和PRRSV,剂量分别为2×10^4.5 TCID50/头和10^6 TCID50/头。攻毒后21d,攻毒对照组猪淋巴结比免疫组显著肿大,免疫组猪血清、淋巴结中PCV2病毒栽量低于对照组,攻毒对照组猪淋巴结中PCV2抗原含量高于免疫组。这些结果表明,嵌合型PCV1-2及真核表达质粒肌肉注射免疫商品猪后,对PCV2感染能产生保护性免疫应答,有可能成为候选疫苗。  相似文献

Resistance to renal leptospirosis was demonstrated in cattle vaccinated with Leptospira interrogans serotype pomona bacterin. Fewer vaccinated cattle given challenge inoculum of virulent serotype pomona leptospires 12 months after vaccination had kidneys with gross focal lesions in the cortex than did nonvaccinated controls. Histopathologic changes characteristically associated with renal leptospirosis occurred less frequently and renal tissue damage was less severe in vaccinated cattle than in nonvaccinated controls. The isolation of serotype pomona from only 1 of 29 vaccinated cattle, compared to 7 isolations from 11 nonvaccinated cattle, at 26 days after challenge inoculum was given, indicated that mild renal infection occurred only infrequently in vaccinated cattle. It appeared that challenge inoculation of vaccinated cattle with virulent serotype pomona leptospires stimulated an accelerated secondary immune response in which immunity limited the multiplication of leptospires in the kidney.  相似文献

The effects of aflatoxin B1 on the development of the immune response to oil-adjuvanted Bordetella bronchiseptica vaccine and on acquired resistance to bacterial challenge were studied in rabbits. The doses of aflatoxin used were insufficient to produce clinical intoxication. Rabbits were randomly assigned to three groups, each having six animals: control (T), vaccinated (V), and vaccinated plus aflatoxin (VA) at 0.05 mg/kg daily per os. Groups V and VA were vaccinated twice, and the three groups were subsequently challenged with virulent B. bronchiseptica. The average weight gain at weekly intervals was significantly reduced in group VA, and no statistically significant differences were found in the titers of agglutinating antibodies between groups V and VA. There were significant differences between groups V and VA in the extent and severity of the pneumonic process, group VA being most affected. Results indicated that agglutinating antibody titers were not related to the level of protection in the latter group. Other mechanisms, such as alveolar macrophage activity and cell-mediated immunity, were implicated in the impairment of the acquired resistance in rabbits subclinically intoxicated with aflatoxin.  相似文献

Groups of pigs vaccinated with an inactivated bivalent vaccine containing porcine parvovirus (PPV) and pseudorabies virus (PRV) developed geometric mean titers (GMT) of humoral antibody for each of the viruses as high or slightly higher than those of other groups of pigs that were vaccinated with inactivated monovalent vaccines containing one or the other of the same viruses. An increase in GMT after challenge exposure of vaccinated pigs to live virus indicated that vaccination did not prevent virus replication. However, an indication that replication was less extensive in vaccinated pigs was provided by the following. Although neither vaccinated nor nonvaccinated (control) pigs had clinical signs after exposure to the live PPV, the effect of vaccination was evident by the fact that GMT were higher in nonvaccinated pigs after exposure than they were in vaccinated pigs. Conversely, all pigs exposed to live PRV had clinical signs, but these signs varied between mild-to-moderate and transient for vaccinated pigs to severe and fatal for nonvaccinated pigs.  相似文献

In a large Slovakian growing-finishing pig production unit, the effects of oral vaccination against swine erysipelas (SE) were investigated in three groups of pigs of 10 weeks of age. In group 1, the pigs were vaccinated intramuscularly at 1 and 3 weeks after arrival in the growing-finishing barn using an Erysipelothrix rhusiopathiae bacterin. Group 2 pigs were vaccinated at the same time as group 1 using an oral avirulent live SE vaccine administered through drinking water; the pigs in the third group were placebo treated. Clinical signs of acute SE, arthritic changes, average daily weight gain (ADG), feed conversion ratio, and mortality were evaluated. None of the pigs in groups 1 and 2 but 31.7% of the control animals (group 3) showed typical clinical signs of acute SE. More (P<0.01) non-vaccinated pigs had chronic arthritic changes compared with groups 1 and 2. No significant differences in mortality were recorded between the groups. Groups 1 and 2 had higher (P<0.05) ADG and lower feed conversion ratios compared with group 3 pigs. The results demonstrated that the oral avirulent live culture was efficacious in significantly reducing the clinical symptoms caused by E. rhusiopathiae infection, so enhancing the pigs' performance.  相似文献

OBJECTIVE: To evaluate immune responses induced by administration of Mycoplasma hyopneumoniae bacterin to pigs. Animals-60 healthy 7- to 10-day-old cross-bred boars. PROCEDURE: Pigs were assigned to 1 of 4 pig groups (15 pigs/group): vaccinated, challenged; vaccinated, nonchallenged; nonvaccinated, challenged; nonvaccinated, nonchallenged. Vaccinated pigs received IM injections of a mycoplasma bacterin on days 0 and 14, whereas nonvaccinated pigs received saline (0.9% NaCl) solution. Pigs in the challenged groups were inoculated intratracheally with M hyopneumoniae on day 42. Pigs were euthanatized and necropsied 41, 44, 48, and 70 days after the first vaccination, and proportion of lung surface with pneumonic lesions was determined. Percentage of lymphocyte subpopulations and number of interferon-gamma (IFN-gamma) secreting lymphocytes in blood and tissues, cytokine and antibody concentrations in bronchoalveolar lavage (BAL) fluid, and serum antibody concentrations were determined. RESULTS: Vaccination against and infection with M hyopneumoniae induced a local mucosal immune response in the respiratory tract of pigs. Proportion of lung surface with pneumonic lesions in vaccinated challenged pigs was reduced on day 70, compared with nonvaccinated challenged pigs. Vaccination stimulated the production of M hyopneumoniae-specific IFN-gamma secreting blood lymphocytes. Tumor necrosis factor-alpha concentration in BAL fluid on day 70 was increased in nonvaccinated challenged pigs, compared with vaccinated challenged pigs. CONCLUSIONS AND CLINICAL RELEVANCE: Vaccination against M hyopneumoniae induced local, mucosal, humoral, and cellular immune responses. Moreover, vaccination reduced the severity of lung lesions in challenged pigs, suggesting that mucosal antibodies, mediation of the inflammatory response, and cell-mediated immune responses are important for control of mycoplasmal pneumonia in pigs.  相似文献

The efficacies of attenuated live- and inactivated-virus vaccines against pseudorabies (PR) in fattening pigs were compared. Pigs born from vaccinated or nonvaccinated sows were vaccinated with one or the other vaccine and were challenge exposed at the end of the fattening period. The particular form of PR observed in fattening units in the field could be reproduced. A marked difference was seen between the control lot and the lots of the pigs vaccinated with the attenuated live- and inactivated-virus vaccines. The protection was real, but not absolute, in the vaccinated pigs. The inactivated-virus vaccine conferred a strong passive immunity to the young pigs of vaccinated dams which interfered with the development of an active immunity. The titer of the colostral antibodies in the sera of pigs born from the sows vaccinated with the attenuated-live virus vaccine was low and decreased rapidly. The active protection obtained with this vaccine was similar to that observed with the inactivated-virus vaccine. Thermal curves, weight losses, and time necessary for recovering the weight of pigs at the time of challenge exposure seemed to be good criteria for measuring the protection of fattening pigs against this particular form of PR. The conditions of the outcome of respiratory tract disorders in these pigs are discussed.  相似文献

The purpose of this study was to determine whether intranasal/oral administration of probiotics can assist vaccination efficacy against an important swine pathogen, porcine reproductive and respiratory syndrome (PRRS) virus (PRRSV). A controlled challenge trial was performed employing: (a) pigs vaccinated against PRRS and treated with a Lactobacillus casei, (b) pigs vaccinated against PRRS only, (c) pigs treated with L. casei only, and (d) pigs neither vaccinated against PRRS nor treated with L. casei. All pigs were challenged intranasally with a wild PRRSV strain. There was no difference in clinical signs or rectal temperature among the four groups. However, pigs that received L. casei gained significantly more weight than pigs that did not. Vaccinated pigs did not gain more weight than nonvaccinated pigs. Vaccinated groups had significantly fewer viraemic pigs on days post-challenge 4, 11 and 17 than nonvaccinated groups of pigs. There was no effect of probiotic on prevalence or duration of viraemia. Among viraemic pigs, there was no significant difference in mean log base(10) titer of PRRS virus among groups. These results suggest that orally administered L. casei does not affect immune response in such a way as to affect PRRS viraemia or nasal shedding. However, it still appears to provide significant benefit when administered during vaccination as indicated by the higher bodyweight gain following PRRS virus infection.  相似文献

The effect of dextran sulfate (DS), known to be cytotoxic to macrophages, on the cell-mediated and humoral immune response to nonviable Mycoplasma hyopneumoniae in pigs was investigated. The cell-mediated immune response was determined by means of lymphocyte transformation a test, using uptake of [3H]thymidine in a microculture system and the humoral immune response by means of a microplate complement-fixation test. Peripheral blood lymphocytes from pigs vaccinated with nonviable M hyopneumoniae and DS incorporated substantially more [3H]thymidine than did those from pigs given Mycoplasma or DS alone. The transformation of lymphocytes from M hyopneumoniae-DS vaccinated pigs was enhanced when M hyopneumoniae cells used in the assay system were heated at 60 C for 30 minutes. Similarly prepared M flocculare and M hyorhinis cells also stimulated lymphocytes from M hyopneumoniae-DS vaccinated pigs, but not nearly as great as when M hyopneumoniae cells were used. The humoral antibody response and the cell-mediated immune response to nonviable M hyopneumoniae was markedly enhanced by DS. Pigs were vaccinated with nonviable M hyopneumoniae and/or DS 4 times and challenge exposed intratracheally with viable M hyopneumoniae. Pigs vaccinated with M hyopneumoniae and DS had less severe pneumonia than did nonvaccinated pigs.  相似文献

The effects of challenge exposure on the humoral and cellular immune responses in pseudorabies vaccinated swine were studied in 84 barrows. The pigs were divided into seven groups and challenge exposed to a virulent strain of pseudorabies virus on months 1, 3, 5, 8, 10, 12 and 14 after vaccination. The pigs were vaccinated with commercial attenuated and inactivated pseudorabies virus vaccines. The protection conferred by vaccination was equally effective with both types of vaccines. The levels of cellular and humoral immunity after challenge exposure in pigs vaccinated with either type of vaccine were similar. The cell-mediated immune response can be effectively used for the early detection of pigs exposed to pseudorabies virus. Virus isolation attempts from the brain and spleen in most of the vaccinated pigs were unsuccessful.  相似文献

Kittens vaccinated with an avirulent biotype of the Black strain of feline infectious peritonitis virus (FIPV; given oronasally) developed both indirect fluorescent and virus-neutralizing antibodies, but were not protected against oronasal challenge exposure with virulent virus. In fact, kittens vaccinated with avirulent virus were more readily infected than were nonvaccinated cats. A proportion of kittens could be immunized to FIPV by giving sublethal amounts of virulent virus. This technique, however, was too inconsistent and hazardous to have clinical relevance. The results of these studies indicated that humoral immunity was not protective in FIPV infection. There was no correlation between fluorescent and virus-neutralizing antibodies and either disease or immunity. Immune serum from FIPV-resistant cats failed to passively protect susceptible animals against virulent virus given intraperitoneally or oronasally, and as expected, actually sensitized them to infection. It was concluded that cell-mediated immunity was probably responsible for protection.  相似文献

The interaction of the morantel sustained release bolus with the development of immunity in calves vaccinated with two doses of gamma irradiated (40 Kr) Dictyocaulus viviparus larvae was investigated under laboratory conditions. A total of 37 helminth-naive calves were used. Eight calves were used in the first part of the study to test the efficacy of a larval vaccine prepared by using gamma rays delivered from a cobalt source. In the second part of the study, four groups of four groups of four calves each were vaccinated and of these, all the animals in two groups each received a bolus. The remaining three groups (two groups of four and one group of five calves each) remained nonvaccinated with each animal in one group receiving a bolus. All the calves were challenged with approximately 2000 lungworm larvae four months postvaccination. In order to simulate possible field conditions, two of the vaccinated groups and two of the nonvaccinated groups were given a trickle infection of 800 lungworm larvae over a four-week period, three months prior to challenge. Based on a comparison of clinical signs, pathology and lungworm burdens at necropsy, the vaccination of the calves conferred a significant degree of protection (P less than 0.001) to a subsequent challenge compared with controls. The introduction of a morantel sustained release bolus and/or a trickle infection had no effect on the high degree of protection engendered by the vaccination. Nonvaccinated calves given a trickle infection, with or without a bolus, were also highly immune to challenge.  相似文献

Mice and swine vaccinated (subcutaneous inoculation) with live acriflavine-fast attenuated Erysipelothrix rhusiopathiae, strain Koganei 65-0.15 (serovar 2), were challenge exposed with 10 strains of E rhusiopathiae pathogenic for swine; the latter strains comprised serovars 9 and 10 and other previously undetermined. Vaccinated mice did not die after they were challenge exposed (subcutaneous inoculation) with serovars 4, 6, 7, 8, 9, 10, 15, 16, or N, but vaccinated mice challenge exposed with strain 2553 (serovar 20) had 30% mortality. Nonvaccinated control mice died after they were challenge exposed with all serovars tested. One of 2 vaccinated swine challenge exposed (intradermal inoculation) with each of strains 911 (serovar 8), 2179 (serovar 10), or 2553 developed localized urticarial lesion at the site of intradermal inoculation. Vaccinated swine challenge exposed with serovars 4, 6, 7, 9, 15, 16, or N did not have clinical signs of acute swine erysipelas. Nonvaccinated control swine developed localized lesions at the site of intradermal challenge inoculation.  相似文献

The objective of this study was to compare the efficacy and safety of single-strain and multi-strain vaccines for the prevention of the respiratory facet of porcine reproductive and respiratory syndrome. The study comprised six groups of pigs (A through F, eight pigs per group). At the beginning of the study (Day 0) Groups C and D were vaccinated with a single-strain vaccine, and Groups E and F were vaccinated with a multi-strain vaccine. The multi-strain vaccine contained five attenuated strains of PRRSV including the strain used as the single-strain vaccine. On Day 28 Groups B (nonvaccinated/challenged control), D, and F were challenged with a highly virulent field strain of PRRSV that was unrelated to any of the strains used for vaccination. Group A was kept as a nonvaccinated/nonchallenged control. On Day 42 all pigs were necropsied. Their lungs and lymph nodes were examined for virus-induced changes. Serum samples obtained at weekly intervals during the study and lung lavage fluids obtained at necropsy were tested for the presence and titer of PRRSV. Serum samples were also tested for antibody. The presence and severity of clinical signs and lesions were the primary means by which vaccine efficacy and safety were evaluated. Both vaccines provided a high level of protective immunity to challenge. However, appreciable lymph node enlargement in pigs vaccinated with multi-strain vaccine, with or without subsequent challenge, raised a question as to its safety. Collectively these results indicate that both single-strain and multi-strain attenuated PRRSV vaccines can be effective immunogens, but additional studies in regard to safety are needed before multi-strain vaccines can be recommended for routine field use.  相似文献

The antibody response to pseudorabies virus nucleocapsid proteins (NCP) was evaluated by the western immunoblot analysis before and after challenge of immunity by nasal inoculation of 10(2.3) plaque-forming units of virus in 10 pigs that had been vaccinated with pseudorabies virus envelope glycoproteins. Antibody to 5 NCP with molecular mass of 140, 63, 41, 34, and 23 kD was first detected in vaccinated and nonvaccinated pigs on day 14 after challenge of immunity. Antibody to 2 of the 5 NCP continued to be detected through day 113 in 9 of 10 vaccinated pigs. Beyond day 32, antibody to NCP was not detected in 1 vaccinated pig. The 23-, 34-, and 41-kD proteins were the most immunogenic. Antibody to each of these proteins was first detected on day 14 in 10, 10, and 8 pigs, respectively. Seven, 6, and 8 pigs, respectively, were antibody-positive for these proteins on day 113. The 140- and 63-kD proteins were the least immunogenic. Antibody to these proteins was detected in 8 and 9 pigs, respectively, on day 14, and in 4 and 5 pigs, respectively, on day 113. Chi-square analysis for dependency indicated that the antibody response to the 140- and 63-kD proteins was interdependent. These results suggested that combinations of NCP may be useful as nonvaccine diagnostic antigens.  相似文献
