The medical records of 266 dogs diagnosed as having atopic dermatitis were reviewed. Statistical data were evaluated referable to breed predilections, clinical signs and positive reactions to allergens. Positive reactions were most common to house dust mites (more common with clinical signs in the fall) followed by moulds (more common with clinical signs in the fall and spring). Dogs with positive reactions to moulds, trees or cultivated plants were more likely to have skin and ear yeast infections. Dogs with positive reactions to cultivated plants were more likely to have otitis externa and pedal lesions. Positive reactions to house dust were more common in dogs with early onset of signs and in those tested early in the disease. Dogs had more positive reactions to weeds when allergy tests were performed in the summer and fall. Positive reactions to flea antigen were highly correlated with the clinical diagnosis of flea allergy dermatitis. 相似文献

The membrane sterol compositions of ten strains ofMalassezia pachydermatis were analysed by gas-liquid chromatography. Six of these were wild-type strains from clinical sources that were sensitive to polyene antibiotics, whereas the other four strains were resistant mutants that had arisen after treatment withN-methyl-N-nitrosoguanidine or UV radiation.The wild-type strains contained 838.8 to 1291.7 µg of sterol per gram of freeze-dried mycelium. The sterols were composed of 79% ergosterol, 19% fecosterol and 2% lichesterol. Combinations of EDTA-Tris and two aminoglycoside antibiotics (amikacin and neomycin) were tested for synergistic activities against the microorganisms associated with otitis externa in dogs and for the solutions' stability over time. Synergistic activity was observed when EDTA-Tris plus amikacin and EDTA-Tris plus neomycin were tested againstStaphylococcus intermedius, Proteus mirabilis, Pseudomonas aeruginosa, andEscherichia coli, but not againstCandida albicans. Stability studies over a 3-month period indicated that the test solutions were stable at room temperature and that their antimicrobial activity was maintained.Abbreviations CFU

The use of fluoroquinolones in veterinary dermatology 总被引:3,自引:1,他引:3
The fluoroquinolones are a group of antibiotics with considerable application for use in veterinary dermatology. They are rapidly bactericidal against a wide variety of clinically important organisms including Staphylococcus intermedius and gram-negative enteric bacilli by virtue of interference with the supercoiling of bacterial chromosomal material. Their favourable pharmacokinetic features make them applicable in many animal species, and in a range of dose formulations. The only major clinical contraindication is that fluoroquinolones should not be given to young, rapidly growing dogs as they can induce a noninflammatory, erosive arthropathy. For many years the only veterinary-labelled fluoroquinolone available was enrofloxacin. The selection of a fluoroquinolone has become more complex now that there are more choices available. Orbifloxacin, difloxacin and marbofloxacin now join enrofloxacin on the veterinary market, although not all of these are licensed in every country. The use of fluoroquinolones in dermatology remains controversial. The authors recommend that fluoroquinolones be considered in circumstances where canine pyoderma has been refractory to appropriate 'first line' antibiotics. They are most useful in the management of recurrent pyoderma and in chronic, deep pyoderma with extensive scar tissue. In addition, fluouroquinolones frequently are the drugs of choice for canine ear infections caused by Pseudomonas aeruginosa. 相似文献

Cytological examination using the tape-strip technique and fungal culture using contact plates with modified Dixon's medium were compared to evaluate the carriage of Malassezia yeasts on four cutaneous sites (left pinna, umbilical region, axilla and perianal area) in adult Basset Hounds. Twenty animals were included in the study. High numbers of Malassezia were isolated from at least one area in 100% of the animals. The frequencies of isolation and population sizes differed significantly according to anatomical location. They were greater on the pinna, followed by the umbilical area, axilla and perianal area. Fungal culture was more sensitive than cytology for the isolation of Malassezia yeasts. Frequencies of isolation were greater using this method, but population sizes were constantly smaller than with cytology. 相似文献

In this study, the isolation of Acinetobacter baumannii in a dog with clinical bilateral otitis externa is described. Moreover, to investigate the zoonotic potential of the isolate, microbiological examinations on the family members were performed. An A. baumanniistrain was isolated from nasal swab in one of the dog owners. The identity of bacterial strains, either from dog and owner, was confirmed by phenotypic and molecular typing (wgMLST). Furthermore, to assess the pathogenic potential of the isolates a deep characterization of virulence and antibiotic resistance genes was done by Whole Genome Sequencing (WGS). Finally, the susceptibility towards a wide panel of antimicrobials was investigated. In our knowledge, this is the first recorded case of A. baumanniiisolation from canine auricular swabs in Italy. And interestingly, this study underlines the possible spread of this microorganism from human to animal. 相似文献

Cytology is the most common and immediately informative diagnostic tool in the practice of dermatology. It is simple to perform and requires equipment usually already present in most practices such as a microscope and slides, cotton-tipped applicators and stains. Cytology interpretation is also easily self taught. With little practice the practitioner can easily become familiar with sample interpretation from inflammatory lesions; including recognition of bacteria, yeast, dermatophyte spores and hyphae, deeper fungal elements and parasites. Additionally, the practitioner should become familiar with the variety of inflammatory cells seen with these conditions, as well as keratinocyte morphology. : Over a seven-year period, chronic otitis externa was surgically managed in 43 dogs at the University Veterinary Hospital of University College Dublin. Lateral ear canal resection (LECR) was undertaken in nine of the 43 dogs: results were unsatisfactory, with a failure of the surgery in five of eight dogs and one dog lost to follow-up. Once end-stage otitis externa, with or without otitis media, is diagnosed, total ear canal ablation and lateral bulla osteotomy (TECA/LBO) is the best treatment option. In this series, 37 of 43 dogs underwent TECA/LBO and of the 29 dogs for which follow-up results were obtained 27 (93%) had an excellent or improved outcome to surgery. Complications following all procedures were most common in cases with a concurrent dermatopathy; therefore, definitive diagnosis and medical treatment for skin and ear disease is essential. 相似文献

BACKGROUND: Malassezia-type yeasts previously have been observed on cytologic examination of the intermammary region of mares that presented with tail-head pruritus; topical antiyeast treatment resolved the pruritus. Further, Malassezia dermatitis has been observed in horses in intertriginous areas such as the udder and prepuce; the species of yeast was not confirmed. It is not known whether healthy mares or male horses can be carriers of this yeast in these body areas. HYPOTHESIS: Malassezia spp. are present in the intermammary region in healthy mares and the preputial fossa in healthy geldings. ANIMALS: Eleven healthy horses (5 mares and 6 geldings). METHODS: Samples of surface material were taken digitally from the intermammary area of 5 mares and the preputial fossa region of 6 geldings. The samples were examined cytologically and were cultured on modified Sabouraud's dextrose agar. The DNA from yeast colonies grown on the agar was extracted, and samples were assayed using fungal generic polymerase chain reaction (PCR) analysis. Amplicons with positive PCR results were sequenced and compared with sequences in the BLAST database search program. RESULTS: Of 44 attempts at culture, 5 yielded a species identified as Malassezia equi, and 2 yielded M slooffiae. In contrast, of 44 cytologic examinations, yeasts with the morphology of Malassezia spp. were seen in 40 samples. CONCLUSIONS AND CLINICAL IMPORTANCE: Due to its presence in healthy horses, finding of Malassezia-type yeast on cytologic examination may not incriminate it as a pathogen. Despite difficulty in culturing, cytologic examination was an effective tool to rapidly demonstrate the organism. 相似文献

Abstract Suspensions of Malassezia pachydermatis adhered to canine corneocytes attached io adhesive tape in a dose (P < 0.001) and time-dependent (P < 0.01) manner; adherence was maximal after 2 h. M. pachydermatis cells were approximately 10 times more adherent than Saccharomyces cerevisiae (P < 0.001) cells after 2 h incubation. The adherence of formalin-treated and frozen-thawed M. pachydermatis cells was comparable with untreated controls. Stationary-phase cells adhered better (P < 0.05) than exponential-phase cells. Pretreatment of the yeasts, or corneocytes, with 0.1% trypsin for 30 min reduced (P < 0.01) the adherence of four, and two, out of five strains, respectively, whereas incubation with 300 mM solutions of D(+) mannose, sucrose and N-acetyl D-glucosamine had no consistent effect. These results suggest that trypsin-sensitive proteins or glycoproteins on the yeast cell wall, and on the corneocyte surface, play an important role in the adherence of M. pachydermatis to canine corneocytes in vitro, whereas a role for carbohydrate receptors was not demonstrated. Résumé— Des suspensions de Malassezia pachydermatis adhérant à des cornéocytes des chiens sont attachées à des rubans adhésifs de façon significative en function du nombre (P < 0,001) et de la durée (P < 0,01). L'adhérence est maximale après 2 heures. Les levures du genre Malassezia pachydermatis sont approximativement dix fois plus adhérentes que les levures du genre Saccharomyces cerevisiae (P < 0,001) après une incubation de 2 heures. L'adhérence des Malassezia pachydermatis traitées par le formol et congelées - décongelées, est comparable à celle des témoins. Les levures qui ne sont pas en phase de croissance adhérent mieux (P < 0,05) que celles qui le sont. Abstract We have previously shown that both atopic and normal dogs generate an IgG response to antigens of Malassezia pachydermatis . The aim of this study was to compare IgE responses to separated proteins of M. pachydermatis in 28 atopic dogs with Malassezia dermatitis and 22 clinically normal dogs using Western immunoblotting. Six different detection systems were evaluated in order to assess sensitivity and eliminate nonspecific binding and cross-reactivity. The protocol yielding the best results utilized a monoclonal mouse antidog IgE, an alkaline phosphatase conjugated goat antimouse IgG which had been passed through a canine IgG column 3 times, a chemiluminescent substrate and a digital imaging system. Proteins of 45, 52, 56 and 63 kDa were recognized by more than 50% of the atopic dog sera and thus represented major allergens. Only a minority of normal dogs showed faint IgE binding to these proteins. The results indicate that the majority of atopic dogs with Malassezia dermatitis have a greater IgE response than normal dogs, suggesting an IgE-mediated immune response may be clinically important in the pathogenesis of the disease. 相似文献

Meningiomas are one of the most common nervous system tumors in dogs. They are mostly solitary, benign, intracranial tumors of the transitional type. Tentative diagnosis can usually be made on historical data, clinical and neurological signs, and radiographs, with an antemortem diagnosis confirmed by exploratory surgery and histologic evaluation of biopsy. The literature contains many reports of the gross, histological and ultrastructural morphology of canine meningiomas, but little on the cytologic features of these tumors. The therapeutic value of antibiotics depends on the susceptibility of the infecting microorganism and the pharmacological
profile of the drugs. To assess the value of an antibiotic combination of polymyxin B and miconazole this study examined the
in vitro synergistic potential of the two drugs on Gram-negative and Gram-positive bacteria and yeast. Antifungal and antibacterial
activity was tested by minimum inhibitory concentration (MIC) of broth macrodilution and urea broth microdilution, by fluorescence
microscopy and flow cytometry. Synergism was calculated using the fractional inhibitory concentration index (FICi). With Staphylococcus intermedius as target we found up to an eightfold reduction of the individual MICs when both drugs were combined. However, the FICi was
0.63 suggesting no real interaction between the two drugs. With Escherichia coli, Pseudomonas aeruginosa, and Malassezia pachydermatis as targets the antimicrobial drug combination reduced the MICs of polymyxin B and miconazole from fourfold to hundredfold
resulting in FICi between 0.06 and 0.5 which defines a synergistic action. Thus, if polymyxin B and miconazole are combined
their effect is greater than the sum of the effects observed with polymyxin B and miconazole independently, revealing bactericidal
and fungicidal synergism. Our results indicate a strong therapeutic value for the combination of these antimicrobial agents
against Gram-negative bacteria and yeast and a weaker value against Gram positive bacteria for clinical situations where these

A 6-year-old female goat was presented with a seborrhoeic dermatosis of 5 months duration. The condition developed following a severe enteritis associated with weight loss. Dermatological examination showed a generalized greasy seborrhoeic dermatosis, which spared the head and extremities of the limbs. Microscopic examination of impression smears of lesional skin revealed numerous yeasts typical of Malassezia sp. Culture on Sabouraud's dextrose agar yielded Malassezia pachydermatis growth. Histopathological examination of haematoxylin/eosin and safranin (HES) stained sections of biopsies showed mild lymphocytic superficial perivascular hyperplastic dermatitis. Numerous budding yeasts were visible both on the surface, and follicular keratin, in HES and periodic acid Schiff (PAS) stained sections. A dramatic response was observed after 1 week of a topical anti-Malassezia treatment, and the resolution of the condition was complete after 4 weeks. 相似文献

Abstract Epidermal hyperplasia is one of the major histopathological features seen in dogs with Malassezia dermatitis. The aim of this study was to investigate the effects of extracts and culture supernatants from Malassezia pachydermatis on the proliferation of canine keratinocytes. Keratinocyte cultures were established from normal dog skin, and cell monolayers were co-cultured with Malassezia extracts (prepared either with or without protease inhibitors) and supernatants derived from organisms grown in liquid culture. The proliferation of keratinocytes was measured using a colourimetric assay. Neither the culture supernatants nor the Malassezia extracts had significant effects on the proliferation rate of canine keratinocytes, regardless of whether protease inhibitors were present or not. The results indicate that the epidermal hyperplasia seen in Malassezia dermatitis is unlikely to be caused directly by secretion of products from the organism. 相似文献

Otitis externa (OE) is a frequently reported disorder in dogs associated with secondary infections by Staphylococcus, Pseudomonas and yeast pathogens. The presence of biofilms may play an important role in the resistance of otic pathogens to antimicrobial agents. Biofilm production of twenty Staphylococcus pseudintermedius and twenty Pseudomonas aeruginosa canine otic isolates was determined quantitatively using a microtiter plate assay, and each isolate was classified as a strong, moderate, weak or nonbiofilm producer. Minimum biofilm eradication concentration (MBEC) of two ionophores (narasin and monensin) and three adjuvants (N‐acetylcysteine (NAC), Tris‐EDTA and disodium EDTA) were investigated spectrophotometrically (OD570nm) and quantitatively (CFU/ml) against selected Staphylococcus and Pseudomonas biofilm cultures. Concurrently, minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) of planktonic cultures were assessed. 16/20 of the S. pseudintermedius clinical isolates were weak biofilm producers. 19/20 P. aeruginosa clinical isolates produced biofilms and were distributed almost equally as weak, moderate and strong biofilm producers. While significant antibiofilm activity was observed, no MBEC was achieved with narasin or monensin. The MBEC for NAC ranged from 5,000–10,000 µg/ml and from 20,000–80,000 µg/ml against S. pseudintermedius and P. aeruginosa, respectively. Tris‐EDTA eradicated P. aeruginosa biofilms at concentrations ranging from 6,000/1,900 to 12,000/3,800 µg/ml. The MBEC was up to 16‐fold and eightfold higher than the MIC/MBC of NAC and Tris‐EDTA, respectively. Disodium EDTA reduced biofilm growth of both strains at concentrations of 470 µg/ml and higher. It can be concluded that biofilm production is common in pathogens associated with canine OE. NAC and Tris‐EDTA are effective antibiofilm agents in vitro that could be considered for the treatment of biofilm‐associated OE in dogs. 相似文献
