A field population of Plutella xylostella from Malaysia (SERD4) was divided into five sub-populations and four were selected (G2-G5) with the Bacillus thuringiensis insecticidal crystal (Cry) toxins Cry1Ac, Cry1Ab, Cry1Ca and Cry1Da. Bioassay at G6 gave resistance ratios of 88, 5, 2 and 3 for Cry1Ac, Cry1Ab, Cry1Ca and Cry1Da respectively compared with the unselected sub-population (UNSEL-SERD4). The Cry1Ac-selected population showed little cross-resistance to Cry1Ab, Cry1Ca and Cry1Da, (3-, 2- and 3-fold compared with UNSEL-SERD4), whereas the Cry1Ab-SEL sub-population showed marked cross-resistance to Cry1Ac (40-fold), much greater than Cry1Ab itself. In contrast, the Cry1Ca- and Cry1Da-SEL sub-population showed little if any cross-resistance to Cry1Ac and Cry1Ab. The mode of inheritance of resistance to Cry1Ac was examined in Cry1Ac-selected SERD4 by standard reciprocal crosses and back-crosses using a laboratory insecticide-susceptible population (ROTH). Logit regression analysis of F1 reciprocal crosses indicated that resistance to Cry1Ac was inherited as an incompletely dominant trait. At the highest dose of Cry1Ac tested, resistance was recessive, while at the lowest dose it was almost completely dominant. The F2 progeny from a back-cross of F1 progeny with ROTH were tested with a concentration of Cry1Ac that would kill 100% of ROTH. The mortality ranged between 50 and 95% in seven families of back-cross progeny, which indicated that more than one allele on separate loci were responsible for resistance to Cry1Ac.  相似文献

苏云金芽孢杆菌cry基因在大肠杆菌中表达产物和生物活性   总被引:4,自引:0,他引:4  
研究了几种Bt cry基因于大肠杆菌(Escherichia coli)中表达产物在pH 10.0的50mmol/L碳酸钠和20mmol/L乙醇胺溶解液中的溶解性 ,发现同样的Cry蛋白在碳酸钠中的溶解度大于乙醇胺。通过胰蛋白酶消化 ,明确Cry1Ca7、Cry1Ia8酶解产物为 38kD多肽 ;Cry1Ie1、Cry1Cb2、Cry2Ab4酶解产物为 41kD多肽 ;Cry1Ac酶解产物为60kD多肽。采用FPLC层析方法对 6种原毒素及其酶解后得到的毒素多肽进行了分离纯化 ,比较了原毒素和毒素的杀虫活性的差异。其结果表明 ,Cry1Ac的原毒素和毒素对棉铃虫初孵幼虫的校正死亡率均为 100% ,Cry2Ab4的原毒素的毒力高于其酶解毒素。  相似文献

对黏虫具有杀虫活性的Bt蛋白筛选及分析   总被引:1,自引:0,他引:1  
为筛选对黏虫[Mythimna separata(Walker)]具有毒杀作用的苏云金芽胞杆菌杀虫蛋白,本研究提取Cry1类、Cry2类、Cry9类及Vip3A类等11种蛋白,通过人工饲料喂毒方法对黏虫进行生物活性测定。结果表明,Cry1Ac、Cry1Ab、Cry2Ab、Cry1Be及Cry1Bb蛋白对黏虫具有杀虫活性,其致死中浓度依次为5.09、17.71、26.75、27.42及43.93μg/g;Cry9Aa、Cry9Eb、Cry9Ee蛋白对黏虫生长具有抑制作用,其浓度为10μg/g时的体重抑制率分别为78.4%、79.0%、86.9%,浓度为100μg/g时的体重抑制率分别为92.8%、95.7%、96.7%;Cry1Ba、Cry1Ca和Vip3Aa蛋白对黏虫无显著活性。本研究为黏虫的生物防治奠定了基础,并为抗虫转基因作物的研究提供优良的候选基因。  相似文献

Cry1Ba3、Cry1Ia8蛋白对Cry1Ac抗性小菜蛾的杀虫活性研究   总被引:4,自引:2,他引:2  
利用2种苏云金芽胞杆菌原毒素Cry1Ba3、Cry1Ia8及其组合,分别对Cry1Ac抗性种群小菜蛾幼虫进行生物活性测定。结果表明Cry1Ba3、Cry1Ia8对2种目标试虫均有高毒力,LC50分别为0.2175、0.6706μg/mL;Cry1Ba3毒力3倍于Cry1Ia8。2种蛋白混配的结果也表现出高毒力,LC50为0.4375μg/mL,没有显著的协同增效作用,也不存在拮抗。敏感与抗性小菜蛾种群生测结果统计分析比较,结果表明这2种蛋白及其组合与Cry1Ac并无交互抗性。  相似文献

为更好地了解苏云金芽胞杆菌Bacillus thuringiensis毒素蛋白对二点委夜蛾Athetis lepigone的毒力以及作用机理,通过饲喂含有Cry1Ac、Cry1Ab、Cry2Ab和Vip3Aa四种不同Bt毒素蛋白饲料,测定Bt毒素蛋白对二点委夜蛾幼虫的毒力,并观察取食4种毒素蛋白后幼虫中肠组织的病理学变化。结果显示,二点委夜蛾幼虫取食毒素蛋白后72 h,Cry1Ab和Cry1Ac毒素蛋白对二点委夜蛾幼虫的杀虫活性较高,校正死亡率为84.7%和76.4%;Vip3Aa和Cry2Ab毒素蛋白的毒力较弱。二点委夜蛾幼虫取食4种Bt毒素蛋白后,中肠柱状细胞微绒毛脱落,杂乱地分散在肠腔内,杯状细胞变形和腔内微绒毛脱落,线粒体和内质网等变形破裂,细胞核的核膜消失、核质凝聚和形状发生变化,经Cry1Ab和Cry1Ac毒素蛋白处理后中肠细胞的病变症状和速度明显高于Cry2Ab和Vip3Aa毒素蛋白处理。表明Cry1Ab和Cry1Ac毒素蛋白对二点委夜蛾幼虫杀虫活性较高,显著高于Cry2Ab和Vip3Aa毒素蛋白,且对其中肠细胞的破坏作用也较强。  相似文献

Cry9Ee是近年来发现的对亚洲玉米螟Ostrinia furnacalis(Guenée)等害虫具有高毒力的蛋白,具有良好的应用前景。为了明确亚洲玉米螟对Cry9Ee和Cry1Ab蛋白是否存在交互抗性,本文首先分别表达、活化、纯化了Cry9Ee和Cry1Ab蛋白,进而对敏感和Cry1Ab抗性亚洲玉米螟进行生物活性测定;随后对2种蛋白在敏感亚洲玉米螟中肠刷状缘膜囊泡(brush border membrane vesicles,简称BBMVs)上的结合进行分析比较。生物活性测定结果表明,Cry9Ee和Cry1Ab蛋白的活性片段对敏感亚洲玉米螟的LC50分别为3.04和0.89 μg/g;而Cry9Ee活性片段在5.00 μg/g的浓度下,对Cry1Ab蛋白抗性亚洲玉米螟致死率高达96%,与对敏感种群活性相当。竞争结合试验表明,Cry9Ee和Cry1Ab两种蛋白间不存在竞争结合,推测它们在亚洲玉米螟中肠上存在不同的受体。综合生物活性测定及体外结合试验两方面结果得出结论:亚洲玉米螟对Cry9Ee与Cry1Ab蛋白不存在交互抗性。cry9Ee基因可以作为理想的候选基因,用于我国新一代转基因抗虫玉米的研制,为延缓和克服亚洲玉米螟对转基因抗虫玉米抗性的产生提供重要保证。  相似文献

Bt棉有效控制了棉田主要害虫棉铃虫(Helicoverpa armigera),然而原来处于次要地位的刺吸式口器害虫盲蝽(Heteroptera:Miridae)为害逐年加重,目前对绿盲蝽(Apolygus lucorum Meyer-Dür)有效的抗虫基因未见报道。本研究以本实验室保存的Bt杀虫基因和菌株为材料,对绿盲蝽进行杀虫活性筛选。利用本实验室先前克隆的Mtx类杀虫基因cry15Aa表达产物进行绿盲蝽杀虫活性测定,研究结果显示Cry15Aa蛋白对绿盲蝽具有一定的杀虫活性。通过杀虫活性测定,获得对绿盲蝽有效的Bt菌株20株;对这些菌株表达蛋白进行质谱鉴定,LC-MS/MS质谱鉴定结果显示,这些菌株可能含有Cry1Aa、Cry1Ab、Cry1Ac、Cry1Ae、Cry1Af、Cry1Ag、Cry1Ah、Cry1Ba、Cry1Be、Cry8Ha等十余种对鳞翅目、鞘翅目害虫有杀虫活性的蛋白片段,并且有4株Bt菌株样品中检出了Mtx类杀虫蛋白Cry15Aa的片段。但是进一步基因鉴定结果表明,这4株菌株并不含有cry15Aa全长基因,推测这些菌株中含有Cry15类的新基因。上述研究结果表明这些菌株中可能存在对盲蝽有效的新型杀虫蛋白。本研究在发现了cry15Aa基因所表达的蛋白对绿盲蝽具有杀虫活性的同时,获得了对绿盲蝽具有杀虫活性的Bt新菌株,对绿盲蝽的生物防治具有重要的意义。  相似文献

Cry1Ac和Cry2Ab蛋白对大草蛉生长发育及酶活力的影响   总被引:2,自引:0,他引:2  
为明确转Cry1AcCry2Ab基因棉花对大草蛉的影响,运用Bt蛋白与人工饲料混合的方法,以大于转基因棉花叶片中蛋白含量20倍的剂量饲喂大草蛉初孵幼虫,初步研究了Cry1Ac和Cry2Ab对大草蛉生物学参数和消化酶、解毒酶活性的影响。结果表明:取食含Bt蛋白饲料的大草蛉幼虫的发育历期、体重、蛹重、成虫体重、羽化率等生物学参数与对照相比均没有显著差异;在大草蛉幼虫体内可以检测到含量较高的Cry1Ac和Cry2Ab蛋白,分别为974.92~1 282.39 ng/g鲜重和5 592.62~6 082.92 ng/g鲜重,而在大草蛉成虫体内检测到的Cry1Ac和Cry2Ab蛋白含量非常低,分别为0.29~0.39 ng/g鲜重和50.34~56.71 ng/g鲜重;取食含Bt蛋白的饲料对大草蛉幼虫的类胰蛋白酶、类胰凝乳蛋白酶、氨肽酶和谷胱甘肽-S-转移酶活性有一定的影响,但对大草蛉成虫影响与对照差异不显著。表明大草蛉取食含Bt蛋白的人工饲料后,虽然体内可以检测到一定含量的Bt蛋白,但对大草蛉的生长发育并没有显著的直接不利影响。  相似文献

为明确靶标害虫对Bt蛋白的抗性及对不同类型Bt蛋白的交互抗性,采用生物测定法进行了二化螟Chilo suppressalis(Walker)敏感品系和Cry1Ac汰选品系在Cry1Ac毒饲料上的时间-死亡率反应、对不同Bt杀虫蛋白的敏感性及不同Bt蛋白复配组合对二化螟毒力效果的研究。结果表明,二化螟Cry1Ac汰选品系已对36.67μg/mL(LC_(50))的Cry1Ac杀虫蛋白产生了一定的适应性,但对308.69μg/mL(LC90)的Cry1Ac蛋白反应仍较敏感。Cry1Ac敏感品系和汰选品系对Cry1Ab蛋白的LC_(50)分别为1.40μg/mL和3.86μg/mL,二者差异显著,但对Cry1Ca(1.63μg/mL和1.73μg/mL)或Cry2Aa(127.48μg/mL和144.50μg/mL)的LC_(50)差异不显著,即Cry1Ac汰选品系与Cry1Ab存在明显的交互抗性,但与Cry1Ca和Cry2Aa不存在交互抗性。在不同Bt蛋白复配组合中(1∶1复配),Cry1Ab+Cry1Ca、Cry1Ab+Cry2Aa和Cry1Ca+Cry2Aa增效作用最为显著。表明抗虫基因cry1Ca和cry2Aa可作为双价转基因抗虫水稻研发的候选基因。  相似文献

入侵云南草地贪夜蛾种群对5种常用Bt蛋白的敏感性评价   总被引:2,自引:0,他引:2  
草地贪夜蛾Spodoptera frugiperda(J. E. Smith)是原分布于美洲大陆热带和亚热带地区的一种重要玉米害虫。在当地,种植抗虫转基因玉米是防控草地贪夜蛾危害的主要手段。该虫于2019年1月入侵我国云南省,为明确入侵我国云南的草地贪夜蛾种群对常用Bt蛋白的敏感性水平,本文通过饲料表面涂抹法测定了瑞丽草地贪夜蛾幼虫对Cry1Ab、Cry1Ac、Cry1F、Cry2Ab以及Vip3A等5种Bt蛋白的敏感性。结果表明:几种常用Bt蛋白对瑞丽草地贪夜蛾致死作用顺序为Vip3A>Cry1Ab>Cry1F>Cry2Ab>Cry1Ac,对草地贪夜蛾抑制生长发育的顺序为Cry1Ab>Cry1F>Vip3A>Cry1Ac>Cry2Ab。此外,与美国相对敏感种群比较,云南瑞丽草地贪夜蛾种群对Cry1Ab、Cry1Ac、Cry1F、Cry2Ab和Vip3A的敏感性指标在0.28~3.76之间,表明该入侵种群对此5种Bt蛋白均未产生抗性。此研究可为将来建立以转Bt基因玉米作为防控草地贪夜蛾的技术体系提供依据。  相似文献

Evolution of resistance by pests is the greatest threat to the continuous success of theBacillus thuringiensis (Bt) toxins used in conventional sprays or in transgenic plants. The most common mechanism of insect resistance to Bt is reduced binding of toxins to target sites in the brush border membrane of the larval mid-gut. In this paper, binding experiments were performed with three 125I-Cry1A toxins and the brush border membrane vesicles from Cry1Ac resistant or susceptible strains of Helicoverpa armigera. The homologous competition test showed that there was no significant difference in Cry1Ac-binding affinity, but the concentration of Cry1Ac-binding sites dramatically decreased in the resistant strain (Rt decreased from 5.87 ± 1.40 to 2.23 ± 0.80). The heterologous competition test showed that there were three Cry1Ac-binding sites in the susceptible strain. Among them, site 1 bound with all three Cry1A toxins, site 2 bound with both Cry1Ab and Cry1Ac, and site 3 only bound with Cry1Ac. In the Cry1Ac resistant strain, the binding capability of site 1 with Cry1Ab decreased and site 2 did not bind with Cry1Ac. It is suggested that the absence of one binding site is responsible for H. armigera resistance to Cry1Ac. This result also showed that the resistance fitted the “mode 1” pattern of Bt resistance described previously.  相似文献

使用苏云金芽胞杆菌库斯塔克亚种(Btk)可湿性粉剂对小菜蛾进行继代汰选获得F80代和F100代抗性品系。通过分别测定Cry1Ab、Cry1Ac、Cry1Ah和Cry1Ca四种杀虫晶体蛋白对小菜蛾Btk抗性品系F80代和F100代的室内毒力,明确了小菜蛾Btk抗性品系对四种Bt杀虫晶体蛋白抗性发展规律。研究结果表明,汰选至F80代时,Cry1Ca对小菜蛾的毒力最高,LC50约12.1mg/L,其次为Cry1Ac,LC50约47.7mg/L;而汰选至F100代时,仍以Cry1Ca对小菜蛾的毒力最高,LC50约21.4 mg/L,其次为Cry1Ab,LC50约72.2 mg/L。与相对敏感品系相比,小菜蛾抗性品系对Cry1Ac的抗性发展较快(抗性倍数高达67.3~106.8倍),对Cry1Ab的次之(抗性倍数高达60.0~66.1倍),而对Cry1Ca和Cry1Ah的抗性发展较慢(抗性倍数分别为3.4~6.0倍和1.6~2.5倍)。以上结果说明,在主效杀虫基因为Cry1Ac的Btk药剂选择压力下,小菜蛾对四种Bt杀虫晶体蛋白的抗性发展速度差异较大,且Cry1Ac和Cry1Ab存在交互抗性风险...  相似文献

目前对转苏云金芽胞杆菌Bacillus thuringiensis(Bt)的研究发现,液泡型ATP酶(Vacuolar-type proton ATPase,V-ATPase)可能是Bt的一类新型受体。我们前期通过构建棉铃虫的中肠酵母文库筛选Cry1Ac的结合蛋白发现棉铃虫V-ATPase亚基B(V-ATPase B)可以与Cry1Ac结合。为明确V-ATPase B在Cry1Ac毒力和昆虫对Cry1Ac抗性机制中的作用,本研究首先采用实时荧光定量PCR技术分析了该基因在抗感Cry1Ac棉铃虫幼虫中及其受到Cry1Ac诱导时的基因表达情况;通过Ligand blot进一步地证实了其与Cry1Ac的结合特性;并通过在Sf9细胞中表达V-ATPase B的试验验证了其功能。结果表明V-ATPase B在抗性品系及受到Cry1Ac诱导时均下调表达,Ligand blot证实了V-ATPase B与Cry1Ac特异性结合;并且在昆虫细胞内过表达该基因,会增强Cry1Ac的细胞毒力。研究结果表明棉铃虫V-ATPase B是Cry1Ac的功能受体,并有可能通过降低基因表达来参与棉铃虫对Cry1Ac的抗性形成。  相似文献

We evaluated the ability of Cry1Aa9, Cry1Ab4, and Cry1Ac1 insecticidal toxins from Bacillus thuringiensis to destroy liposomes. Cry1A toxins are thought to form pores in midgut apical cell membranes (BBMV), thereby disrupting midgut cells. Liposomes containing fluorescent calcein were prepared using phosphatidylcholine (PC) and phosphatidylserine (PS) (PC/PS-Liposomes) or PC alone (PC-Liposomes). Cry1Ab (1.4 μM), but not Cry1Aa or Cry1Ac, disrupted PC/PS-Liposomes and PC-Liposomes. PC/PS-Liposomes containing cholesterol and oligosaccharylceramide from Plutella xylostella midgut were damaged even more extensively by Cry1Ab, but the inclusion of either lipid alone had no effect. The initial velocity of Cry1Ab-mediated liposome disruption increased 17-fold when liposomes were prepared with Triton X-100-soluble proteins from Bombyx mori BBMV and PC (PC/Proteo-Liposomes), and Cry1Aa and Cry1Ac also caused slight disruption. These data suggest that Cry1Ab achieves higher penetration into PC/PS-Liposomes, PC-Liposomes, and PC/Proteo-Liposomes compared with Cry1Aa or Cry1Ac and that Cry1Ab may interact with membrane proteins.  相似文献

为了提高Bt的杀虫活性,运用扫描电镜、SDS-PAGE电泳和生物测定技术,研究了荧光增白剂FB28对棉铃虫Cry1Ac抗性和敏感品系围食膜的影响及对Cry1Ac杀虫蛋白的增效作用。结果表明,正常棉铃虫围食膜表面光滑致密,没有空洞和缝隙。用含1%FB28的人工饲料饲喂刚蜕皮的5龄棉铃虫幼虫2 h后,抗感棉铃虫围食膜表面均出现许多空洞,且敏感棉铃虫的空洞明显大于抗性棉铃虫;随着处理时间的延长,围食膜上的空洞逐渐变小,13 h后恢复正常。用1%FB28离体或活体处理抗、感品系棉铃虫,均可造成围食膜蛋白被降解。用1%FB28与Cry1Ac杀虫蛋白混合饲喂抗、感棉铃虫初孵幼虫,可明显提高棉铃虫的死亡率,且对抗性品系的增效作用明显大于敏感品系,对敏感和抗性品系棉铃虫的增效比分别为2.23和9.34;1%FB28还可以明显增加对抗、感品系3龄棉铃虫幼虫的生长抑制作用。  相似文献

采用人工饲料喂毒法和浸叶喂毒法测定了二化螟、大螟和稻纵卷叶螟初孵幼虫对4种Bt杀虫蛋白的敏感性。二化螟对Cry1Ab、Cry1Ac、Cry1C和Cry2A的LC50分别为1.739、2.6452、.794 ng/mg和15.965 ng/mg;大螟依次为2.455、10.046、11.803 ng/mg和35.359 ng/mg;稻纵卷叶螟依次为4.000、1.611、69.031μg/mL和12.708μg/mL。作者就不同水稻螟虫对不同Cry蛋白的敏感性差异及其在转基因抗虫作物中的应用作了讨论。  相似文献

菌株FB是1株对小菜蛾Plutella xylostella幼虫具有高毒力的苏云金芽胞杆菌Bacillus thuringiensis (Bt)。本研究通过扫描电子显微镜、大质粒电泳、总蛋白SDS-PAGE及菌株生长特征观察的方式研究了菌株FB特征,克隆得到了基因cry1Ia、cry1Ea、cry1Ab、cry2Ab和vip3Aa全长,依据全基因组测序结果得到了1个cry8基因部分片段,首次在Bt菌株中同时发现基因cry1类和cry8类,这五种基因推导的氨基酸序列与已知基因序列相比,最高相似性分别为99%、98%、99%、100%、99%,而cry8半长基因与已知基因仅为63%。生测结果表明,蛋白Cry1Ia、Cry1Ea和Cry1Ab对小菜蛾幼虫具有较高杀虫活性。  相似文献

分别用对CyrlAc抗性达11.6倍的Bt棉叶和Bt粉剂(Btk)汰选棉铃虫(Heliocoverpa armigera)种群(对Btk的抗性指数为5.2倍),测定了其对不同Bt制剂,Bt毒素,化学农药的交互抗性,结果表明:停止汰选后,Bt棉叶汰选种群对CrylAc的抗性指数为5.9倍,对CrylAc 1C的交互抗性为3.7倍,对Btk的敏感性降低,对Bta,Cry2A无交互抗性,Btk汰选种群对Btk的抗性指数为4.1倍,对Bta,CrylAc,CrylAc 1C的交互抗性为2.3,3.1,2.3倍,对Cry2A无交互抗性,两汰选种群对化学农药灭多威,辛硫磷,氯氰菊酯的敏感性均有所增加。  相似文献

BACKGROUND: The pink bollworm is one of the most destructive pests of cotton. Transgenic cotton producing Bt toxin Cry1Ac or a combination of Cry1Ac and Cry2Ab2 has been used effectively against this pest. However, some other insects have evolved resistance to Bt toxins in the field. During the 2007–2008 and 2008–2009 seasons, pink bollworm populations in India were surveyed to evaluate their responses to Cry1Ac and seed powder containing Cry1Ac and Cry2Ab2. RESULTS: The results provide evidence that resistance to Cry1Ac had evolved by 2008 in a population sampled from non‐Bt cotton in the Amreli district of Gujarat in western India. The median lethal concentration of Cry1Ac for five‐day‐old larvae (LC50) was significantly higher for insects derived in 2008 from Amreli than for any of the other field populations tested from four locations in India. For Cry1Ac, the mean LC50 for the strain derived from Amreli in 2008 was 44 times higher than for the most susceptible population. However, for seed powder of Bollgard II containing primarily Cry2Ab2, the 2008 Amreli population was only slightly less susceptible than the most susceptible population. CONCLUSIONS: The data reported here constitute the first evidence of field‐evolved resistance of pink bollworm to Cry1Ac. This initial evidence spurred more extensive evaluations during the 2009–2010 growing season, which confirmed field‐evolved resistance to Cry1Ac in Amreli. The lack of cross‐resistance to Cry2Ab2 suggests that plants producing this toxin are likely to be more effective against resistant populations than plants producing only Cry1Ac. 为了明确Cry1F杀虫蛋白对亚洲玉米螟和黏虫两种重要玉米害虫的作用机制,本文通过透射电镜观察了亚洲玉米螟和黏虫4龄幼虫取食含Cry1F蛋白人工饲料后的中肠组织病理变化,并与其取食含Cry1Ab蛋白人工饲料后的组织病理变化进行了比较。亚洲玉米螟和黏虫4龄幼虫取食Cry1F蛋白48 h后中肠杯状细胞发生了明显的病变,主要表现为:细胞微绒毛肿胀、脱落;细胞核变形,质膜和核膜界限不清晰;染色质发生固缩、贴近核膜;线粒体拉伸变形数量减少,严重的发生空泡化;内质网肿胀与断裂、数量减少,与取食含Cry1Ab蛋白人工饲料相比,玉米螟和黏虫中肠组织发生的病变程度相似。本研究可为Cry1F作为转基因玉米重要的杀虫蛋白在未来亚洲玉米螟和黏虫防治中更好地发挥作用提供理论依据。  相似文献
