A previously undescribed species of human leukocyte, or alpha, interferon is present in the serum of many patients with systemic lupus erythematosus. It was shown to be alpha-interferon by neutralization with specific antiserums, affinity column chromatography, and antiviral activity on bovine cells. However, 23 of 30 interferon samples tested were inactivated by incubation at pH 2, a characteristic of human "immune," or gamma, interferon. Multiple samples of interferon from the same patient had similar biological properties, but samples from different patients were not all identical, suggesting that several variants of this species of human alpha-interferon may exist.  相似文献

The biological response modifier human beta-interferon had pronounced antigrowth effects on various histologic types of human brain tumor cells but no effects on a nontransformed cell line, MRC-5. The cultures of brain tumor cells showed severe alterations indicative of cell injury and death after exposure to beta-interferon for 2 to 6 days. Similar results were obtained with cells freshly explanted from human brain tumors. The results indicate that it may be possible to use fresh, explanted tumor tissue to identify patients who might benefit from therapy with beta-interferon.  相似文献

Chick embryo cells given prior treatment with interferon make new interferon earlier and in larger quantities upon stimulation with Chikungunya virus than cells not so treated. By the criterion of loss of sensitivity to actinomycin, the time needed for formation of messenger RNA for interferon was decreased in the primed cells. Thus interferon affects virus action within 1 hour after infection.  相似文献

Human primary skin fibroblasts trisomic for chromosome 13, 18, or 21 and diploid human skin fibroblasts were induced for an antiviral response with human interferon. The cells that were trisomnic for chromosome 21 were three to seven times more sensitive to protection by human interferon than the normal diploid or trisomic 18 or 13 fibroblasts. The differential response in trisomnic 21 cells is consistent with the known assignment of the human antiviral gene to chromosome 21.  相似文献

The lead-lead isochron age of chondrules in the CR chondrite Acfer 059 is 4564.7 +/- 0.6 million years ago (Ma), whereas the lead isotopic age of calcium-aluminum-rich inclusions (CAIs) in the CV chondrite Efremovka is 4567.2 +/- 0.6 Ma. This gives an interval of 2.5 +/- 1.2 million years (My) between formation of the CV CAIs and the CR chondrules and indicates that CAI- and chondrule-forming events lasted for at least 1.3 My. This time interval is consistent with a 2- to 3-My age difference between CR CAIs and chondrules inferred from the differences in their initial 26Al/27Al ratios and supports the chronological significance of the 26Al-26Mg systematics.  相似文献

The discovery of isotopic anomalies in the calcium- and aluminum-rich inclusions of the Allende meteorite has improved our knowledge of the origin of the solar system. Inability to find more inclusions without destroying the meteorite has hampered further study. By using a fourth-generation computed tomographic scanner with modifications to the software only, the interior of heterogeneous materials such as Allende can be nondestructively probed. The regions of material with high and low atomic numbers are displayed quickly. The object can then be cut to obtain for analysis just the areas of interest.  相似文献

Analytical transmission electron microscopy, electron microprobe analyses, and singlecrystal x-ray diffraction data support the conclusion that high potassium contents, up to 1.5 weight percent K(2)O, of some diopside and omphacite inclusions from diamonds represent valid clinopyroxene compositions with K in solid solution. This conclusion contradicts the traditional view of pyroxene crystal chemistry, which holds that K is too large to be incorporated in the pyroxene structure. These diopside and omphacite inclusions have a high degree of crystal perfection and anomalously large unit-cell volumes, and a defect-free structure is observed in K-bearing regions when imaged by transmission electron microscopy. These observations imply that clinopyroxene can be a significant host for K in the mantle and that some clinopyroxene inclusions and their diamond hosts may have grown in a highly K-enriched environment.  相似文献

Calcium- and aluminum-rich inclusions (CAIs) occur in all classes of chondritic meteorites and contain refractory minerals predicted to be the first condensates from the solar nebula. Near-infrared spectra of CAIs have strong 2-micrometer absorptions, attributed to iron oxide-bearing aluminous spinel. Similar absorptions are present in the telescopic spectra of several asteroids; modeling indicates that these contain approximately 30 +/- 10% CAIs (two to three times that of any meteorite). Survival of these undifferentiated, large (50- to 100-kilometer diameter) CAI-rich bodies suggests that they may have formed before the injection of radiogenic 26Al into the solar system. They have also experienced only modest post-accretionary alteration. Thus, these asteroids have higher concentrations of CAI material, appear less altered, and are more ancient than any known sample in our meteorite collection, making them prime candidates for sample return.  相似文献

Double-stranded (ds) RNA and many viruses are inducers of interferon (IFN), the latter presumably because they contain, or can form, dsRNA. Concomitant with the induction of IFN in chicken embryo cells was the induction of a novel double-stranded ribonuclease (dsRNase), which was released into the medium and continued to accumulate long after IFN production ceased. Only avian cells (chicken, quail, turkey, or duck) expressed high levels of this dsRNase; mammalian, turtle, or fish cells did not. Production of the nuclease was inducer dose-dependent. Optimum pH and cation requirements distinguished it from other dsRNase activities. Degradation of dsRNA was endonucleolytic. Activity resided in a molecule of an Mr of approximately 34,500. Low levels of a single-stranded (ss) RNase activity were inseparable from the dsRNase. The role for a dsRNA-inducible dsRNase released from cells is unknown.  相似文献

Intranuclear inclusions found in glial cells of two cases of subacute sclerosing leukoencephalitis have been examined in the electron microscope. The inclusions are composed of filamentous tubules 170 to 230 angstroms in diameter, which bear a general resemblance to the intranuclear structures observed in cells infected with measles virus. This finding provides further suggestive evidence that a virus may be involved in the pathogenesis of subacute sclerosing leukoencephalitis.  相似文献

Two distinct suites of minerals included in natural diamond occur and probably represent different physical and chemical conditions during diamond growth. Minerals of the ultramafic suite appear to have equilibrated in the range 1000 degrees to 1300 degrees C between 45 and 65 kilobars, whereas the temperature range for minerals of the eclogitic suite is 850 degrees to 1250 degrees C. At present, models relating the partitioning of magnesium and iron between coexisting phases are not sufficiently rigorous to determine a value for the pressure of equilibration of these eclogitic suite inclusions.  相似文献
