通过分析中国对虾人工选育两个群体WSSV感染相关免疫与生化因子的变化,研究选育的两个中国对虾群体对WSSV的敏感性和抵抗力。结果表明,两个群体感染WSSV后,总细胞数(THC)在24h达到最大,随后呈下降趋势;两个群体血淋巴蛋白在感染初期和中期变化不同,但在后期均呈下降趋势;相比而言2'对虾群体比6'对虾群体血蛋白含量和THC下降幅度稍慢;2'对虾群体和6'对虾群体感染24h后酸性磷酸酶(ACP)稍有下降,随后保持较高的活性,而碱性磷酸酶(AKP)的变化除2'对虾群体在感染48h有显著增大外,6'对虾群体变化不明显;过氧化氢酶(CAT)、活性氧(ROS)、超氧化物歧化酶(SOD)与WSSV感染有密切的联系,两个群体抗氧化酶活性随WSSV增殖的变化略有不同;细胞内酚氧化酶(proPO)原变化趋势也表现出差异,斑点杂交结果显示6'对虾群体比2'对虾群体阳性反应较早,揭示不同群体间对WSSV感染的敏感性存在一些差异。 相似文献

The defence ability of Fenneropenaeus chinensis and Marsupenaeus japonicus against the intrusion of the white spot syndrome virus (WSSV) was compared after injecting WSSV intramuscularly by recording cumulative mortality, diagnosing the virus and examining variations in immunological parameters including the total haemocyte counts (THCs), phagocytic percentage (PP), plasma protein concentration (PPC), phenoloxidase (PO) and nitric oxide synthase (NOS) activity. The results showed that the variations in the immunological parameters of F. chinensis and M. japonicus showed similar trends. The THCs of the two species decreased significantly postchallenge of WSSV. The virus was detected at 78 h in M. japonicus and at 42 h in F. chinensis after infection, which was in correlation with the accumulative mortality, and the variations in PO, PP, NOS and PPC in the two species. All shrimps of F. chinensis in the mortality experiment died within 66 h, much more quickly than M. japonicus, whose cumulative mortality reached 100% after 198 h. In conclusion, the immune system of M. japonicus has a stronger resistant ability to antagonize and endure the invasion of WSSV than that of F. chinensis. 相似文献

一个包括42个全同胞家系的中国对虾选育群体,荧光标记后每个家系各取40尾健康个体混养,自然发病并确认感染白斑综合征病毒(white spot-syndrome virus,WSSV)。发病1周后将所有个体取出,统计各家系的存活率,并对活体利用巢式聚合酶链式反应(polymerase chain reaction,PCR)进行WSSV检测。对存活率进行分析,结果显示:4个家系的存活率在60%以上,7个家系的存活率在20%以下;存活率与第一次扩增阳性率、第二次扩增阳性率呈不显著负相关。第一次扩增阳性率与第二次扩增阳性率呈极显著正相关。结合两次扩增的阳性率分析得出:青岛野生群体内自交产生的一个家系和韩国群体养殖家系与乳山野生群体杂交产生的一个家系在抗病力上显著优于其它家系;经独立样本t检验表明,群体间交配组合的存活率差异不显著,因此家系选育可能是进行抗WSSV品种选育的重要途径。多重比较结果发现:韩国群体养殖家系(♂)与乳山野生群体(♀)杂交后代的存活率(56.20%±8.75%) 显著高于韩国群体养殖家系自交后代(22.50%±2.50%) (P<0.05),说明该杂交后代在抗WSSV方面有一定程度的杂种优势。而韩国群体养殖家系 (♂)和乳山野生群体(♀)的杂交后代所表现的杂种优势(26.95)与其反交后代(3.85)差异显著。图3表5参15

Two kinds of specific chicken egg yolk immunoglobulins (IgYs), IgY‐WSSV and IgY‐VP28, were, respectively, raised against the 2 mM binary ethylenimine (BEI)‐inactivated white spot syndrome virus (WSSV) and a principal envelope protein VP28. The activity of purified specific IgYs was stable under the conditions of 20–70 °C, pH 3.0–10.0 and 0–700 g L?1 sucrose solution. In the neutralization assay, these high‐affinity IgY antibodies can specifically bind with the virus particles to protect shrimp (Fenneropenaeus chinensis) against WSSV infection. After oral delivery for 20 days, the IgY‐WSSV exerted a higher protection effect (RPS: 71.5%) than IgY‐VP28 (RPS: 63.7%). Moreover, an increase in RPS (79.2%) was found on addition of IgY‐WSSV:VP28 (0.1% IgY‐VP28 plus 0.2% IgY‐WSSV). This may indicate that neutralization of WSSV refers to the multiple‐hit model. By time‐course study of the levels of the specific IgYs in vivo, the data showed that the titre was enhanced to a relatively high level (P/N=8.35±0.45) at 3 days post administration, declined slightly (P/N=7.13±1.01) at 7 days post administration and then remained stable for further investigation. The stable antibody level potentially contributes towards blocking a large number of WSSV particles from entering and infecting on the major tissues at the early and late stages after challenge in shrimp. 相似文献

蚯蚓与蝇蛆对中国对虾生长及抗白斑综合征病毒感染的研究 总被引:2,自引:0,他引:2
以中国对虾(Fenneropenaeus chinensis)为实验材料,分别投喂配合饲料、蚯蚓、蛤蜊、蝇蛆等四种饵料,利用生长和抗病性指标综合评价蚯蚓和蝇蛆作为中国对虾饵料的可行性。生长实验前测定每个实验组的初始体长和体重,养殖40d后再次测定生长指标,之后,分别投喂不同毒饵量进行人工感染实验。方差分析表明:投喂四种饵料后,蛤蜊组生长最快,其次是蚯蚓,再次是蝇蛆,最后是配合饲料,各组之间对虾体长增长差异显著(P<0.05),体重增长差异极显著(P<0.01) 。利用线性固定模型分析不同饵料及不同病毒量感染下对中国对虾存活率的影响,其中蚯蚓组存活率最高,配合饲料组存活率最低,并且蚯蚓组存活率显著高于蛤蜊组和配合饲料组(P<0.05),蝇蛆组显著高于配合饲料组(P<0.05)。研究结果表明:蚯蚓和蝇蛆可显著提高中国对虾的生长速度,明显提高中国对虾的抗病性,是很好的对虾饵料。 相似文献

In the present study, we used TaqMan real‐time polymerase chain reaction to quantify and compare infection of white spot syndrome virus (WSSV) with shrimp production of Fenneropenaeus chinensis cultured in outdoor ponds along the west coast of the South Korea. In 2007, a total of 60 specimens in summer and 116 specimens in autumn were collected from 12 growing‐out ponds and 12 harvest ponds respectively. Pond harvest data were obtained from farmers. Of the summer samples, all specimens were WSSV positive, with a wide range of 12.4–7.0 × 107 (mean 7.5 × 106) copies ng?1 DNA; shrimp production was 1.7 metric tonnes per hectare (mt ha?1). Of the 116 autumn‐sample specimens, 81 (69.8%) were WSSV positive; WSSV infection had been decreased dramatically, to 0–7.2 (mean 3.5) copies ng?1 DNA. Shrimp production of autumn ponds was 2.1 mt ha?1. Statistical analysis indicated that the difference in WSSV infections detected in summer and autumn was highly significant (P<0.01). In summer, seven ponds (58.3%) with low‐WSSV infection loads (0–1000 WSSV copies ng?1 DNA) had shrimp production of 2.7 mt ha?1; the others had shrimp production of only 0.2 mt ha?1. 对虾WSSV病是亚洲对虾养殖业中的一个棘手问题。本研究采用Kimura引物 ,用PCR技术对不同生长期的中国对虾 (Penaeuschinensis)进行了WSSV的检测 ,同时也检测了对虾发病时养殖池中多见的野生厚蟹 (Helicesp .)和矛尾刺虎鱼 (Acanthogobiushasta)。检测结果表明 :分别在检测的 5尾亲虾中的 1尾 ,6尾仔虾中的 1尾 ,5尾稚虾中的 3尾及所检测的 5尾病虾和 2只厚蟹中获得到 982bp的PCR扩增产物 ,说明为WSSV感染阳性。在检测的 2尾矛尾刺虎鱼中均未获得PCR扩增产物 ,说明为WSSV感染阴性。在亲虾、虾苗以及虾池内的野生厚蟹中检测到WSSV感染的阳性结果表明 :WSSV感染的亲虾有可能是病毒的储主 ,WSSV感染的野生厚蟹有可能是病毒中间宿主或病毒的携带者 ,它们在对虾WSSV病的感染、传播中起了重要的作用 相似文献

Bantam能够调控细胞增殖、细胞凋亡等过程,影响生物的免疫过程。本研究利用实时荧光定量PCR(qRT-PCR)技术对感染WSSV的中国明对虾(Fenneropenaeus chinensis)肝胰腺和鳃组织内bantam表达水平进行检测,发现感染WSSV后6、12、24和48 h,中国明对虾肝胰腺中的bantam表达水平分别是对照组的(0.16±0.03)(P<0.05)、(0.63±0.26)、(0.32±0.06)(P<0.05)和(0.41±0.13)倍;中国明对虾鳃中的bantam表达水平分别是对照组的(0.30±0.17)(P<0.05)、(1.88±0.26)(P<0.01)、(0.84±0.36)和(0.51±0.25)倍。利用miRanda软件进一步对中国明对虾bantam靶基因进行预测分析,评分最高的靶基因是泛素缀合酶E2。中国明对虾泛素缀合酶E2包含UBCc功能域。多序列比对显示,UBCc功能域氨基酸残基序列在不同物种间保守性较高。进化树分析显示,分类学地位相近的物种的泛素缀合酶E2聚为一类。qRT-PCR检测感染WSSV的中国明对虾肝胰腺和鳃中的泛素缀合酶E2表达水平,结果显示,在感染WSSV后6、12、24和48 h,中国明对虾肝胰腺中泛素缀合酶E2的表达水平分别是对照组的(0.54±0.10)、(1.19±0.62)、(3.69±0.51) (P<0.01)和(1.94±0.07)(P<0.05)倍;中国明对虾鳃中泛素缀合酶E2的表达水平分别是对照组的(0.22±0.05)、(1.34±0.38)、(4.29±0.52)(P<0.01)和(1.28±0.79)倍。研究表明,bantam和泛素缀合酶E2的表达都受WSSV侵染的影响,可能与中国明对虾和WSSV之间的互作相关。但bantam和泛素缀合酶E2表达水平的变化是对虾抵抗WSSV侵染过程的免疫反应,还是宿主基因被病毒胁迫后的结果,需要进一步验证。 相似文献

This study aimed at evaluating the ploidy effects on growth performances of Chinese shrimp ( Fenneropenaeus chinensis Osbeck, 1765) reared in different salinities under laboratory conditions. In the acute salinity experiment, there was no difference ( P >0.05) in tolerance observed in triploid and diploid shrimp due to abrupt salinity changes. The lethal salinity for 50% of the individuals in 96 h at 23–25 °C was about 2 g L−1 in both triploids and diploids. While for the chronic salinity experiment, statistical analyses confirmed that the differences in growth performances including the specific growth rate (SGR), the feeding rate (FR), feed conversion efficiency (FCE) and intermoult period (IP) between triploid and diploid were related to salinity. Diploid shrimp reared in 20 g L−1 exhibited highest SGR ( P <0.05), while triploids performed well in 20 and 30 g L−1 salinities ( P <0.05). Based on the survival and growth data, the optimal salinity for the culture of diploid F. chinensis should be 20 g L−1 and for triploids it should be between 20 and 30 g L−1 . 相似文献

基于1982年和2014~2015年渤海渔业资源与环境调查数据,采用Ecopath模型,分析了渤海生态系统的营养关系、结构及功能参数,评估了中国对虾(Fenneropenaeus chinensis)在渤海的生态容量变化。结果显示,渤海生态系统中底栖甲壳类、软体动物等功能群处于重要的营养位置,但中国对虾不是渤海生态系统的关键种,其生物量的增加对口虾蛄(Oratosquilla oratoria)、三疣梭子蟹(Portunus trituberculatus)、多毛类、底栖甲壳类有负影响,花鲈(Lateolabrax japonicus)、虾虎鱼类等生物量的增加将对中国对虾产生负影响。渤海生态系统2个时期均处于发育的不稳定期,仍有较高的剩余生产量有待利用,2014~2015年渤海生态系统成熟度和稳定性较1982年有所降低,系统出现一定程度的退化。中国对虾1982年和2014~2015年在渤海的生态容量为0.810和0.702 t/km~2;与当年依据调查数据评估的生物量相比较,中国对虾有较大的增殖潜力,当生物量增长至71.68倍和585倍时,仍不会超过生态容量。 相似文献

A total of 36 gravid female shrimp Fenneropenaeus chinensis, captured off the coast of Qingdao, Peoples Republic of China, from April 20 through April 24, 2001, were examined by PCR for the presence of WSSV and HPV, the two most prevalent viral diseases affecting this species of shrimp in this region of China. Eight spawns, derived from 8 different females, were selected from shrimp where no WSSV or HPV had been detected. Nauplii obtained from 7 individual spawns and unhatched eggs from one other shrimp were transported to the University of Arizona, in Tucson, Arizona, USA, for larval rearing and production of postlarvae under quarantine protocols. The resultant postlarvae were kept in Primary Quarantine for approximately 2 months, while additional tests were performed to determine their specific pathogen-free (SPF) status according to criteria set by the United States Marine Shrimp Farming Program (USMSFP). By the time the shrimp had reached an average weight of approximately 0.4 g, no USMSFP specific listed pathogens had been detected in any of the 7 hatches and the shrimp were released into Secondary Quarantine at the Oceanic Institute in Kailua-Kona, Hawaii for production of a F1 generation. Serpin家族成员作为主要的丝氨酸蛋白酶抑制剂,通过调节一系列丝氨酸蛋白酶和半胱氨酸蛋白酶的活性来调节蛋白酶级联反应,进而参与了包括血液凝结、补体激活、纤维蛋白溶解、炎症反应、肿瘤抑制和激素转运等大量的基本生物过程,在调节机体免疫及其它重要生理功能方面发挥着重要作用。我们在前期研究中,从中国明对虾血细胞中克隆得到一个serpin型丝氨酸蛋白酶抑制剂基因(Fc-serpin, GenBank注册号:DQ318857),初步研究显示它在转录水平参与了病原菌和白斑杆状病毒(WSSV)引发的对虾先天免疫应答反应;本研究利用原核重组表达系统,成功获得了重组的对虾serpin型蛋白酶抑制剂(rFc-serpin),纯化复性后得率为0.3g/L;活性分析显示,重组目的蛋白(rFc-serpin)对多种病原菌的生长均有一定抑制作用,研究结果进一步证实它参与病原微生物引发的对虾防御应答过程,为探讨甲壳动物先天免疫防御应答机制提供了有价值的参考。 相似文献

根据已测序的WSSV-VAP1全基因序列设计1对引物,经PCR扩增及琼脂糖凝胶电泳分析,扩增出约838bp的特异条带,将其回收后克隆入pGAPZα-A载体中,并进行序列测定与分析。结果表明,扩增序列与GeneBank登录序列(GenBank No.AF41634)核苷酸同源性100%。序列分析表明,WSSV-VAP1基因编码蛋白无跨膜区域,有多个蛋白激酶C、酪蛋白激酶Ⅱ磷酸化位点和糖基化位点,并含有RGD序列,预示其为WSSV浸染中的一个重要蛋白。 相似文献

SUMMARY: Growth and reproduction of hatchery-reared Chinese white prawn Penaeus chinensis released in the Ariake Sea, Japan, were examined. Chinese white prawn grew rapidly, reaching a body length of 154 mm in males and 198 mm in females by November (219–229 days after hatching). Maximum body length of sampled individuals was 164 mm in males and 223 mm in females. Growth curve of the Chinese white prawn was fitted to the Pitcher and MacDonald's formula, Lt = 155.0{1 − e 2.925sin[2π(t − 16.151)/365] − 0.0623( t − 10.712) } for males and the logistic curve, Lt = 200.3/[1 + e (1.985–0.034 t ) ] for females (where Lt is the body length t days after release and t is the number of days after release). Females reached sexual maturity in late February and spawning occurred until April. Minimum size at ripe and spawned stages was 189 mm and 193 mm body length, respectively. 相似文献

为探讨特异性卵黄抗体对凡纳滨对虾(Litopenaeus vannamei)抗白斑综合征病毒(white spot syndrome virus, WSSV)的免疫保护机制及效果,本研究以添加不同剂量WSSV卵黄抗体制剂(0、0.2%和0.5%)的饲料投喂凡纳滨对虾幼虾,免疫28 d后使用WSSV进行人工感染,测定感染对虾的肝胰腺免疫酶活力和免疫基因表达水平,以及感染后14 d内对虾的存活率。结果显示,WSSV感染3 d后,与未添加卵黄抗体制剂的对照组相比,0.2%免疫组对虾肝胰腺的超氧化物歧化酶(SOD)和酚氧化酶(PO)活力显著升高,酸性磷酸酶(ACP)和碱性磷酸酶(AKP)活力显著降低,热休克蛋白70基因(Hsp70)表达水平显著升高,凝集素基因(lectin)和β-1,3-葡聚糖结合蛋白–脂蛋白基因(β-GBP-HDL)表达水平显著降低;0.5%免疫组对虾肝胰腺的SOD活力显著升高,ACP和AKP活力显著降低,Hsp70基因表达水平显著升高,β-GBP-HDL基因表达水平显著降低。人工感染实验结果显示,WSSV感染14 d后,0.2%和0.5%免疫组对虾的存活率分别为48.89%和87.78%,均显著高于对照组(存活率为0),且0.5%免疫组对虾存活率显著高于0.2%免疫组。特异性卵黄抗体制剂能在一定程度上改变发病的进程,延迟对虾的发病和死亡时间,提高同期存活率。研究表明,口服特异性卵黄抗体制剂可以调节对虾肝胰腺免疫酶活力和免疫基因表达水平,显著提高凡纳滨对虾抗WSSV感染的能力。本研究为卵黄抗体抗WSSV感染机制的研究提供了参考,也为在生产上使用卵黄抗体防控WSSV感染提供了科学依据。 相似文献

Studies were conducted by injecting/feeding white spot syndrome virus (WSSV) derived from infected shrimp, Penaeus monodon (Fabricius), to different life-stages, namely post-larvae, juveniles, sub-adults and adults of Macrobrachium rosenbergii (de Man). The disease was also induced in brood stock, and the eggs and larvae derived from these animals were subsequently tested for WSSV infection. All the stages except egg used for the experiment were found WSSV positive in histopathology, cross infection bioassay and polymerase chain reaction (PCR) analysis. Experimentally infected post-larvae and juveniles showed a high percentage of mortality and an increased rate of cannibalism. The cumulative mortality in post-larvae was up to 28%; with 28–40% cannibalism resulting in a maximum loss of up to 68%. In juveniles, observed mortality and cannibalism were 10–20% and 6.7–30.0%, respectively, and the maximum loss recorded was 50%. In sub-adults, mortality ranged from 2.8 to 6.7%, cannibalism was up to 20% and the total loss was up to 26.7%. Sub-adults and adults were found to be more tolerant to the infection as evidenced by the mortality pattern. A nested (two-step) PCR resulted in a 570-bp product specific to WSSV in all stages, except the eggs. 相似文献

以中国明对虾为试验对象,优化了拥挤应激条件下中国明对虾肝胰脏蛋白质双向电泳的条件,并分析了关联的差异蛋白质。试验结果表明,中国明对虾肝胰脏蛋白双向电泳优化条件为裂解液中添加三丁基膦,采用17 cm的固相p H值(p H 4~7)梯度干胶条,上样量为120μg ,等电聚焦时间为65000 V/h。蛋白图谱分析结果显示,得到的700多个蛋白点中差异蛋白点11个,其中8个差异蛋白点丰度显著上调(P <0.05),3个差异蛋白点丰度显著下调(P <0.05)。研究表明,在拥挤应激条件下中国明对虾肝胰脏蛋白表达发生显著变化。 相似文献
