Summary

In a series of experiments the effect of administering KNO2 was studied, during parturition, on the capability of oxygen transport of maternal blood and on oxygen transfer to foetal blood. The following blood parameters were analysed, MHb percentage, pO2, O2‐saturation, pH, pCO2, and (NO2) in maternal arterial blood (carotid art.) and venous blood (jugular vein) and in foetal arterial blood (umbilical art.) and venous blood (umbilical vein). The relative O2‐saturation was calculated from the estimated O2‐saturation by multiplying with the factor Hb (mmol/1) minus MHb (mmol/1), divided by Hb (mmol/1). In addition, blood pressure in the carotid artery, heart rate, and respiration rate in the dam were continuously recorded for some hours.

A dosage of 9 to 12 mg of NO2/kg body weight intravenously or of 30 mg of NO2/kg body weight orally to the dam caused much higher MHb percentages and NO2 contents in the maternal blood than in the foetal blood. In maternal blood the ratio of NO2 content td MHb percentage was proportional to that in foetal blood. In the arterial blood, MHb percentages were almost as high as in the venous blood. After administering of nitrite, relative O2‐saturation dropped simultaneously with the increase in methaemoglobin.

Nitrite treatment caused a drop in the maternal blood pressure; heart rate and respiration rate increased. O2‐saturation in the blood in the umbilical vein was much lower in the animals with nitrite treatment than in those without. These experimental results show clearly that the oxygen capacity of the blood decreases after nitrite treatment. In pregnant cows the oxygen supply to the foetus will be adversely affected after nitrate intake, especially by the lower oxygen transfer via the placenta, though hardly at all by methaemoglobin formation in the foetal blood.

When the oxygen transfer to the foetal blood decreases too sharply, intra‐uterine death and ultimately abortion may result.  相似文献

The uterine and umbilical vessels of 12 pregnant ponies were catheterised to study foetal metabolism. The effects of this procedure on maternal and foetal cardiovascular, respiratory, metabolic and adrenocortical activity were monitored during and after surgery. Premedication with acepromazine-butorphanol-detomidine was followed by induction of anaesthesia with detomidine and ketamine and maintenance, using mechanical ventilation, with halothane in oxygen and nitrous oxide. Mean maternal arterial blood pressure was greater than 70 mmHg during anaesthesia and arterial oxygen tension remained over 100 mmHg. The foetuses were adequately oxygenated but were hypercapnic and lactic acidaemic. Most maternal and foetal blood gases and metabolites had returned to normal by 24 h, although foetal plasma lactate fell more slowly. The maternal adrenocortical discharge was less severe than reported previously and plasma cortisol had fallen to basal levels by 48 h after surgery. Foetal plasma cortisol remained low and did not change during or after surgery. Arterio-venous metabolite and gas tension differences across the uterine and umbilical circulations were slightly greater at operation than in the recovery period, suggesting that uteroplacental perfusion may have been impaired during surgery. Post-operative recovery of the mare and foetus was satisfactory and subsequent problems associated with the foetal catheters were not related to the anaesthesia or surgery.  相似文献

To evaluate the influence of cow breed and number of fetuses on uterine and umbilical blood flows and nutrient fluxes or uterine tissues from gravid cows, surgery was performed on Charolais or Hereford cows with single or twin fetuses at 177 +/- .2 d (mean +/- SEM) after mating. Indwelling catheters were placed in a fetal femoral artery and vein, in an umbilical vein of each fetus, and in a uterine artery and vein of each gravid horn. Deuterium oxide (D2O) was infused into a fetal femoral vein at 183 +/- .3 and 190 +/- .5 d after mating to estimate uterine and umbilical blood flows (liters/minute). Blood oxygen and plasma glucose and lactate concentrations were determined and uterine arterio-venous (A-V) and umbilical venous-fetal arterial (v-a) differences and net uterine and fetal uptakes were calculated. Net utilization by the uteroplacenta was calculated as the difference between uterine and fetal net uptakes. Uterine blood flows were lower (P less than .01) in Hereford (4.80 +/- .28) than in Charolais (7.07 +/- .33) and lower (P less than .01) per fetus in cows with twin fetuses (5.22 +/- .34) than in cows with a single (6.65 +/- .28) fetus. Umbilical blood flows were greater for single than for twin fetuses. Fetal oxygen and glucose net uptakes averaged 57 and 12%, respectively, of net uteroplacental utilization. Lactate was released from uteroplacental tissues to fetal (42%) and maternal circulations (58%). The concentrations and fatty acid composition of the plasma free fatty acid, triacylglycerol and phospholipid fractions were determined in maternal and umbilical cord vein blood samples taken at delivery from 17 mares. Maternal and umbilical vein plasma free fatty acid concentrations were of a similar order and a positive correlation was found between the two levels suggesting that the equine placenta is permeable to fatty acid. Substantial amounts of the essential fatty acids and their longer chain derivatives were seen in both umbilical vein plasma free fatty acid and phospholipid fractions supporting this view. Certain long chain polyunsaturated derivatives of the essential fatty acids found in the umbilical venous plasma phospholipid fraction were not seen in the maternal circulating lipids. The precursor fatty acids were readily available to both foetal and placental tissues and therefore must have been elongated and incorporated into phospholipid by either or both. Very small amounts of the essential fatty acids were found in adipose stores in the newborn foal and virtually no fat stores at all in the newborn foal liver.  相似文献

The aim of this study was to determine whether venous blood samples can be used as an alternative to arterial samples in calves with respiratory problems and healthy calves. Jugular vein and ventral coccygeal artery were used to compare blood gas values. Sampling of the jugular vein followed soon after sampling of the ventral coccygeal artery in healthy calves (group I) and calves with respiratory problems (group II). Mean values of arterial blood for pH, pCO2, HCO3act in healthy calves were 7.475 +/- 0.004, 4.84 +/- 0.2 kPa, 28.45 +/- 1.30 mmol/L compared with venous samples, 7.442 +/- 0.006, 6 +/- 0.3 kPa, 30.93 +/- 1.36 mmol/L, respectively. In group II, these parameters were 7.414 +/- 0.01, 5.93 +/- 0.3, 27.73 +/- 1.96 mmol/L for arterial blood and 7.398 +/- 0.008, 6.85 +/- 0.2 kPa, 29.77 +/- 1.91 mmol/L for venous blood, respectively. There were no statistically significant differences between arterial and venous pH, HCO3act, Be(ecf), ctCO2 values with the exception of pCO2 (P = 0.001) in group II. In group I, correlation (r2) between arterial and venous blood pH, pCO2, HCO3act were 84.5%, 87.5%, 95.7%, respectively compared with the same parameters in group II, 80.8%, 77.1%, 70.3%. In conclusion, venous blood gas values can predict arterial blood gas values of pH, pCO2 and HCO3ecf, Be(ecf) and ctCO2- for healthy calves but only pH values in calves with acute respiratory problems (r2 value>80%).  相似文献

The purpose of this study was to compare the effects of inhaled isoflurane and a constant infusion of propofol on maternal haemodynamics and uterine arterial and umbilical venous flows in pregnant ewes. Late term pregnant ewes ( n  = 5) were randomly assigned to receive either inhaled isoflurane or an intravenous infusion of propofol for 1 h, each on separate occasions. Maternal systemic arterial, right atrial and pulmonary arterial blood pressures, cardiac index, systemic vascular resistance index, stroke volume index, heart rate, and uterine arterial and umbilical venous flows were determined over the 1 h period of each treatment. Data were analysed using an univariate analysis of variance for repeated measures performed on the ranks of the data. Propofol anaesthetized ewes had significantly higher heart rate ( P  = 0.0040), mean arterial pressure ( P  = 0.0003) and cardiac index ( P  = 0.0475) compared to isoflurane anaesthetized ewes. There were no significant differences in uterine arterial flows, umbilical venous flows, or other measured variables. Continuous propofol infusions maintain maternal haemodynamics at significantly higher levels than does inhaled isoflurane, while uterine arterial and umbilical venous flows do not differ significantly.  相似文献

The hindlimb arteriovenous difference (AVD) model was used to determine whether 30 mg/ kg of the nitric oxide synthase (NOS) inhibitor L-NGnitroarginine methyl ester (hydrochloride; L-NAME) inhibited ovine NO synthesis and influenced muscle metabolism. Eight Border Leicester x Merino cross lambs (50 to 55 kg BW) were infused with saline (control) or saline containing L-NAME via an indwelling jugular vein catheter in a balanced randomized crossover design with 3 d between treatments. The abdominal aorta and deep femoral vein were catheterized for assessment of AVD of hind limb metabolism. Arterial hematocrit and insulin concentration and both arterial and venous concentrations of nitrate/nitrite (NOx), glucose, lactate, NEFA, and urea were determined. Infusion of L-NAME decreased arterial NOx concentrations (P = 0.049), indicating inhibition of systemic NO synthesis. Treatment had no effect on arterial (3.5 vs. 3.6 +/- 0.19 mmol/L for control and L-NAME lambs, respectively; P = 0.39) or venous (3.3 vs. 3.4 +/- 0.16 mmol/L, P = 0.55) plasma glucose concentrations or on glucose AVD (0.19 vs. 0.27 +/- 0.065 mmol/L, P = 0.20). There was an interaction (P = 0.038) between time and treatment, such that L-NAME initially increased the AVD of glucose (up to 180 m) divergent from control lambs. The response was then decreased before a possible inflection beyond 240 min. Infusion of L-NAME increased hindlimb venous NEFA (222 vs. 272 +/- 13.2 micromol/L, P = 0.007) and NEFA AVD (79.4 vs. -13.3 +/- 31.5 micromol/L, P = 0.018). These metabolic changes were independent of plasma insulin concentrations, which were not affected by L-NAME infusion (25.3 vs. 27.8 +/- 3.62 mU/L, P = 0.85). The increase in hindlimb lipolysis after L-NAME infusion does not seem to be due to increased lipolysis of plasma triacylglycerol because circulating arterial (155 vs. 142 +/- 20.8 micromol/L, P = 0.58), venous (154 vs. 140 +/- 20.5 micromol/L, P = 0.50), and AVD (1.0 vs. 2.9 +/- 3.17 micromol/L, P = 0.38) triacylglycerol concentrations were unaffected by L-NAME infusion. In conclusion, these data indicate that infusion of 30 mg of L-NAME/kg inhibits NO synthesis, which in turn influences fat and carbohydrate metabolism in the ovine hindlimb independently of plasma insulin concentrations.  相似文献

Objectives of this study were to evaluate maternal and fetal influences on uterine and umbilical blood flows and nutrient fluxes of gravid uterine tissues of cows. Brahman cows with Brahman or Charolais fetuses and Charolais cows with Brahman or Charolais fetuses were used. Indwelling catheters were placed into a uterine artery and vein, an umbilical vein, and a fetal femoral artery and vein at 220 +/- .4 d after mating. Uterine and umbilical blood flows (liters/min) and net uptakes of oxygen, glucose, lactate, alpha-amino N, urea N, ammonia N, and estrone sulfate by the gravid uterus, fetus, and uteroplacenta were determined on d 227 +/- .4. Uterine blood flows in Brahman cows with Brahman (5.01) or Charolais (4.66) fetuses were similar but less (P less than .001) than in Charolais cows with Brahman (7.14) or Charolais fetuses (9.24), which differed (P less than .01). Umbilical blood flows of Charolais (3.78) were greater (P less than .01) than those of Brahman (2.29) fetuses. Rate of placental D2O clearance as well as net fetal uptake of oxygen, glucose, and alpha-amino N, gravid uterine uptake of alpha-amino N, and uteroplacental uptake of glucose and release of estrone sulfate were greater with Charolais than with Brahman fetuses. Gravid uterine oxygen uptake and estrone sulfate release and gravid uterine and uteroplacental lactate output were influenced by the interaction between cow and fetal breed. It is suggested that fetal growth may be limited by uterine blood flow and by function of the uteroplacenta, particularly in late gestation.  相似文献

The relationship between mixed venous O2 tension and cardiac output was studied in six anesthetized horses breathing 100% O2. Cardiac output, O2 consumption, mean arterial pressure, heart rate, and arterial and venous blood gases were measured after administration of xylazine or dobutamine to horses in lateral, sternal, and dorsal recumbencies. After approximately 3 hours, Escherichia coli endotoxin was administered while horses were in dorsal recumbency, and all measurements were repeated. Relationships between cardiac index (CI) and PVO2, heart rate, mean arterial pressure, jugular PVO2, and PVO2 of blood from a superficial limb vein were evaluated by linear regression analysis. Mean arterial pressure was significantly (P less than 0.05) correlated with CI in horses in all positions and after endotoxin administration. However, data points were poorly grouped. Heart rate and CI were significantly correlated in horses in all positions, but not after endotoxin administration. Correlations between jugular PVO2 and PVO2 of blood from a superficial limb vein were not significant in horses in sternal recumbency, and PVO2 of blood from a superficial limb vein was not significantly correlated with CI in horses in lateral recumbency. There was a significant and tight correlation between PVO2 and CI in horses in all positions and after endotoxin administration.  相似文献

OBJECTIVE: To determine changes in splanchnic oxygen extraction ratio during experimentally induced portal hypertension in dogs. ANIMALS: 6 clinically normal dogs. PROCEDURE: Standard midline laparotomy and median sternotomy were performed in anesthetized dogs. Baseline measurements of arterial blood pressure, aortic blood flow, portal vein blood flow, and portal vein pressure were acquired, and arterial, venous, and portal vein blood samples were obtained to determine systemic and splanchnic oxygen extraction ratios. The portal vein was gradually occluded until a pressure of 18 cm of H2O was reached; this pressure was maintained for 30 minutes, and measurements and collection of blood samples were repeated. RESULTS: Portal vein blood flow decreased significantly from 457 +/- 136 ml/min before to 266 +/- 83 ml/min after induction of portal hypertension. Oxygen content in the portal vein significantly decreased from 12.3 +/- 1.85 to 8.2 +/- 2.31%, and splanchnic oxygen extraction ratio significantly increased from 15.8 +/- 6.2 to 37.4 +/- 10.9% during portal hypertension. There was a significant inverse correlation between portal vein blood flow and splanchnic oxygen extraction ratio at baseline and during portal hypertension. CONCLUSIONS AND CLINICAL RELEVANCE: An increase in splanchnic oxygen extraction ratio is evident with partial attenuation of the portal vein and the concurrent decrease in portal vein blood flow. Correlation of oxygen extraction ratio with portal vein blood flow may be a more important indicator for determination of an endpoint to prevent congestion and ischemia of the gastrointestinal tract and pancreas during ligation of portosystemic shunts.  相似文献

The comparative study of the acid-base balance (ABB) parameters has been performed on 20 clinically healthy mature Ma?opolski horses. An arterial blood sample from the facial artery and a sample of venous blood from the external cervical vein were colected from each animal. In the samples tested, the blood pH, pCO2, tCO2, HCO3-, concentration of Na+, K+, Cl-, and a value of the anion gap were determined. The difference among pCO2, tCO2, and HCO3- in both samples tested was statistically significant, whereas the pH of the arterial blood and the pH of the venous blood did not differ significantly. The anion gap in both types of blood did not differ significantly. Conclusions: 1) ABB parameters such as pCO2, HCO3-, and tCO2 determined in the arterial and venous blood of the Ma?opolski horses differ from each other significantly. 2) In spite of the lack of the differences between pH of the arterial and venous blood, the ABB parameters in horses should be determined in the arterial blood, because the comparative study performed proves that the analysis of the ABB parameters determined for the venous blood of a healthy horse may lead to a wrong diagnosis of the compensated respiratory acidosis. 3) The mean value of anion gap in horses aged 8-12 years amounts to 20.9 mmol/l for the arterial blood and 19.93 for the venous blood; the difference between the two values is not statistically significant.  相似文献

The pH, partial pressure of oxygen (pO(2)), partial pressure of carbon dioxide (pCO(2)), concentration of bicarbonate (HCO(3)(-)), base excess and oxygen saturation (SO(2)) were measured in venous and arterial blood from 57 newborn calves from 55 dams. Blood samples were collected immediately after birth and 30 minutes, four, 12 and 24 hours later from a jugular vein and a caudal auricular artery. The mean (sd) pO(2) and SO(2) of arterial blood increased from 45.31 (16.02) mmHg and 64.16 (20.82) per cent at birth to a maximum of 71.89 (8.32) mmHg and 92.81 (2.32) per cent 12 hours after birth, respectively. During the same period, the arterial pCO(2) decreased from 57.31 (4.98) mmHg to 43.74 (4.75) mmHg. The correlation coefficients for arterial and venous blood were r=0.86 for pH, r=0.85 for base excess and r=0.76 for HCO(3)(-). The calves with a venous blood pH of less than 7.2 immediately after birth had significantly lower base excess and HCO(3)(-) concentrations for 30 minutes after birth than the calves with a venous blood pH of 7.2 or higher. In contrast, the arterial pO(2) was higher in the calves with a blood pH of less than 7.2 than in those with a higher pH for 30 minutes after birth.  相似文献

OBJECTIVE: To measure and compare palmar digital venous plasma nitric oxide (NO) concentrations and digital arterial blood flow after application of topical nitroglycerine (NTG). STUDY DESIGN: Experimental study. ANIMALS: Healthy adult horses (n=8). METHODS: Digital blood flow was measured by an ultrasonic Doppler flow probe surgically implanted around the medial palmar digital artery. Blood was collected from a catheter placed in the medial palmar digital vein for quantification of NO. NTG patches, NTG ointment or control patches were placed over the palmar digital vessels at the level of the fetlock. Two horses had an intra-arterial infusion of an NTG solution into the medial palmar digital artery in a pilot study. RESULTS: Digital arterial blood flow did not change significantly with application of the NTG patches, NTG ointment, or control patches. There were no statistically significant or biologically important changes in digital venous NO concentrations across time or between treated and control horses. In the pilot study, digital arterial blood flow and palmar digital venous NO concentrations increased with intra-arterial infusion of NTG. CONCLUSIONS: In clinically healthy horses, digital arterial blood flow and digital venous plasma NO concentrations did not change significantly with application of the NTG patches/ointment. These treatments are unlikely to have an effect on the digital vasculature of laminitic horses, however, further investigation is warranted. CLINICAL RELEVANCE: Although NTG patches have been used as a method of decreasing vasomotor tone and improving digital blood flow in horses with laminitis, this study provides evidence in healthy conscious horses that this treatment is not effective in altering digital blood flow.  相似文献

Blood oxygen transport and tissue oxygenation were studied in 28 calves from the Belgian White and Blue breed (20 healthy and 8 hypoxaemic ones). Hypoxaemic calves were selected according to their high respiratory frequency and to their low partial oxygen pressure (PaO2) in the arterial blood. Venous and arterial blood samples were collected, and 2,3-diphosphoglycerate, adenosine triphosphate, chloride, inorganic phosphate and hemoglobin concentrations, and pH, PCO, and PO2 were determined. An oxygen equilibrium curve (OEC) was measured in standard conditions, for each animal. The arterial and venous OEC were calculated, taking body temperature, pH and PCO2 values in arterial and venous blood into account. The oxygen exchange fraction (OEF%), corresponding to the degree of blood desaturation between the arterial and the venous compartments, and the amount of oxygen released at the tissue level by 100 mL of blood (OEF Vol%) were calculated from the arterial and venous OEC combined with the PO2 and hemoglobin concentration. In hypoxaemic calves investigated in this study, the hemoglobin oxygen affinity, measured under standard conditions, was not modified. On the contrary, in vivo acidosis and hypercapnia induced a decrease in the hemoglobin oxygen affinity in arterial blood, which combined to the decrease in PaO2 led to a reduced hemoglobin saturation degree in the arterial compartment. However, this did not impair the oxygen exchange fraction (OEF%), since the hemoglobin saturation degree in venous blood was also diminished.  相似文献

The effect of two dietary electrolyte balance (dEB, Na+ + K+ - Cl-) levels on arterial and portal blood oxygen content, blood pH, and acid-base status in pigs was studied during a 9-h period after a meal, using a crossover experimental design. The dEB levels were established by changing the Cl- level in the diets. Four pigs with a mean weight of 45 kg were surgically fitted with catheters in the carotid artery and portal vein. Two dEB levels (-100 and 200 mEq/kg) were evaluated in two periods of 1 wk each. Feed was given at 2.6 times the maintenance requirement for energy in two meals per day. Water was freely available. Blood samples were taken at 0, 0.5, 1, 1.5, 2, 3, 4, 6, and 9 h after feeding. Blood hemoglobin; O2 pressure; O2 saturation; O2 content; pH; PCO2; HCO3-; base excess; and Na+, K+, and Cl- contents were measured. Oxygen contents in arterial and portal blood were lower (P < 0.008) in the -100 mEq/kg group (5.78 and 4.82 mmol/L respectively) compared to the 200 mEq/kg group (6.18 and 4.99 mmol/L respectively). This was related to the lower hemoglobin content in the blood of animals in the -100 mEq/kg group. Arterial and portal blood pH were lower (P < 0.003) at -100 mEq/kg (7.46 and 7.37) than at 200 mEq/kg (7.49 and 7.43). The difference in blood pH between the two groups was sustained throughout the sampling period. The average values of arterial and portal blood for base excess and HCO3- content were higher (P < 0.001) at high dEB (6.96 and 31.0 mmol/L, respectively, for -100 mEq/kg and 12.54 and 35.9 mmol/ L, respectively, for 200 mEq/kg). The Na+ concentration in the blood was increased and K+ and Cl- concentrations were decreased (P < 0.02) by increasing dEB from -100 mEq/kg to 200 mEq/kg. Blood electrolyte balance level was higher (P < 0.001) in the 200 mEq/kg dEB group than in the -100 mEq/kg dEB group. In conclusion, dEB changed blood oxygen content and pH, and influenced the acid-base buffer system in pigs. Also, within each group, pigs maintained a relatively constant blood pH level during the 9-h period after feeding.  相似文献

Large mongrel dogs were anesthetized, instrumented, and subjected to electrically induced ventricular fibrillation after breathing either 100% oxygen (O2) or 10% O2 and 90% nitrogen for 10 minutes before arrest. Four minutes after arrest, open chest cardiopulmonary resuscitation (CPR) or intermittent abdominal compression closed chest CPR was initiated and continued for 20 minutes, at which time defibrillation was attempted. Central arterial and mixed venous blood samples were collected serially for the measurement of pH, carbon dioxide partial pressure (PCO2), and O2 partial pressure (PO2), and calculation of bicarbonate concentration and base excess. Mixed venous blood was collected serially for the measurement of lactate concentration. Hemodynamically variable resuscitation techniques and pre-arrest hypoxia or hyperoxia did not significantly influence blood-gas values during CPR. Mixed venous lactate concentrations after 20 minutes of CPR were significantly higher when hypoxia preceded the arrest and when intermittent abdominal compression closed chest CPR was used for resuscitation. Mixed venous PCO2 was significantly higher than arterial PCO2 in all dogs during CPR but was not significantly different before arrest.  相似文献

The aim of this work is to evaluate the haemodynamic characteristics of maternal and fetal vessels during normal pregnancy in bitches, using Colour and Pulsed wave Doppler ultrasonography, in order to obtain more information about maternal and fetal circulation. The blood waveforms of the uteroplacental arteries, aorta, caudal cava vein and umbilical cord of the fetuses were recorded weekly in 16 pregnant bitches. Also, the measurements of Peak Systolic, End Diastolic Velocities, Resistance and Pulsatility Indices were carried out. Uteroplacental blood flow was biphasic while the ones of the umbilical artery and aorta were first systolic and then diastolic. The cava showed a typical waveform of venous vessels. During gestation the EDV and PSV of fetal vessels increased (alpha<0.05) while the PI and RI of all vessels examined decreased (alpha<0.05) except for the IP of the Aorta. The Doppler ultrasonography was used to study the characteristics of maternal and fetal vessel flow and their progressive changes during pregnancy. This study can be considered a further contribution in diagnosing and monitoring high-risk pregnancies in Veterinary Medicine.  相似文献

Respiratory function and acid-base variables were studied in Welsh Mountain ponies before and at predetermined times after the intravenous injection of Immobilon and Revivon.A marked depression of respiratory rate was accompanied by large reductions in arterial blood oxygen tension and saturation and the development of a mild respiratory acidosis following the injection of Immobilon. It was concluded that at least three factors contributed to the hypoxic hypoxia produced by Immobilon; the posture of lateral recumbency, the decrease in respiratory rate and the laboured character of the respiration. Arterial oxygen and carbon dioxide tensions returned towards control levels soon after administering Revivon. Mixed venous oxygen tensions were little affected by either Immobilon or Revivon, and mixed venous carbon dioxide tensions were increased to smaller degrees that those of arterial blood. Haemoglobin was increased initially by Immobilon, had returned to the control level by 30 min and fell below the control following the administration of Revivon.  相似文献

The study was carried out on healthy Friesian calves (n = 10) aged between 10 and 30 days. Hypochloremia and alkalosis were induced by intravenous administration of furosemide and isotonic sodium bicarbonate. The venous and arterial blood samples were collected repeatedly. 2,3-diphosphoglycerate (2,3-DPG), hemoglobin and plasmatic chloride concentrations were determined. The red blood cell chloride concentration was also calculated. pH, PCO2 and PO2 were measured in arterial and mixed venous blood. The oxygen equilibrium curve (OEC) was measured in standard conditions. The correspondence of the OEC to the arterial and mixed venous compartments was calculated, taking blood temperature, pH and PCO2 values into account. The oxygen exchange fraction (OEF%), corresponding to the degree of blood desaturation between the arterial and mixed venous compartments and the amount of oxygen released at the tissue level by 100 mL of blood (OEF Vol%) were calculated from the arterial and mixed venous OEC, combined with PO2 and hemoglobin concentration. Oxygen delivery (DO2) was calculated using the arterial oxygen content, the cardiac output measured by thermodilution, and the body weight of the animal. The oxygen consumption (VO2) was derived from the cardiac output, OEF Vol% and body weight values. Despite the plasma hypochloremia, the erythrocyte chloride concentration was not influenced by furosemide and sodium bicarbonate infusion. Due to the alkalosis-induced increase in the 2,3-DPG, the standard OEC was shifted to the right, allowing oxygen to dissociate from hemoglobin more rapidly. These changes opposed the increased affinity of hemoglobin for oxygen induced by alkalosis. Moreover, respiratory acidosis, hemoconcentration, and the slight decrease in the partial oxygen pressure in mixed venous blood (Pvo2) tended to improve the OEF Vol% and maintain the oxygen consumption in a physiological range while the cardiac output, and the oxygen delivery were significantly decreased. It may be concluded that, despite reduced oxygen delivery, oxygen consumption is maintained during experimentally induced hypochloremic alkalosis in healthy 10-30 day old calves.  相似文献

Free amino-acid concentrations were measured in maternal venous and fetal umbilical vein plasma, and in the allantochorion, of Thoroughbred mares at term. Concentrations in maternal and fetal plasma were similar to those reported previously in equids. The concentrations of free amino-acids in the allantochorion were higher than those in the maternal and fetal plasmas and were characterised by high levels of the nonessential amino-acids as observed in other species. Fourteen of the 20 amino-acids measured had similar allantochorion/umbilical vein concentration ratios suggesting that simple gradient diffusion might play a part in their transfer from the placenta to the fetus.  相似文献
