Simple sequence repeats (SSRs) defined as sequence repeat units between 1 and 6 bp occur abundantly in both coding and non-coding regions in eukaryotic genomes and these repeats can affect gene expression. In this study, ESTs (expressed sequence tags) of Betula pendula (silver birch) were analyzed for in silico mining of EST-SSRs, protein annotation, open reading frames (ORFs), designing primers, and identifying codon repetitions. In B. pendula, the frequency of ESTs containing SSRs was 7.8 % with an average of 1SSR/4. 78 kb of EST sequences. A total of 188 SSRs was identified by using MISA software and di-nucleotide SSR motifs (65.9 %) were found to be the most abundant type of repeat motif followed by tri- (27.1 %), tetra- (4.8 %), and penta- (2.2 %) motifs. Based on ORF analysis, 175 of 178 sequences were predicted as ORFs and the most frequent SSRs were detected in 5′ UTR (58.43 %), followed by in ORF (31.46 %) and in 3′ UTR (8.43 %). 102 of 178 ESTs were annotated as ribosomal protein, transport protein, membrane protein, carrier protein, binding protein, and transferase protein. For a total of 102 SSRs (57.3 %) with significant matches, a set of 102 primers (100 %) with forward and reverse strands was designed by using Primer3 software. Serine (Ser, 19.6 %) was predominant in putative encoded amino acids and most of amino acids showed nonpolar (35.3 %) nature. Our data provide resources for B. pendula and can be useful for in silico comparative analyses of Betulaceae species, including SSR mining.  相似文献

Limited nutrient acquisition from soil is a key process limiting productivity in boreal forest. We investigated short-root morphological adaptations and rhizosphere effect in relation to site conditions in interaction with tree mineral nutrition. We studied seven young (8- to 14-year-old) silver birch (Betula pendula Roth.) stands on abandoned agricultural land in Estonia. Soil pH varied from 3.8 to 7.0, and soil N % from 0.07 to 0.26%. Tree nutrient (NPK) status was expressed by leaf nutrient concentrations. Leaf N correlated negatively with short-root specific length and area. Summed activity (SA) and metabolic diversity of bacteria (by BIOLOG Ecoplate™), bacterial community diversity (by DGGE) and pHKCl were determined for rhizosphere (R) and bulk soil (S) to reveal the extent of the rhizosphere effect. Bacterial activity in rhizosphere was 1.4–4.7 times higher than in bulk soil. Ratio SAR/SAS indicating root support to the rhizosphere bacterial communities decreased with increasing bulk soil pH; however, when bulk soil pH was ≥5, the decrease in SAR/SAS was insignificant, i.e. the rhizosphere effect stayed at a stable level. Diversity of bacterial community was 6% higher in bulk soil than in rhizosphere. Rhizosphere acidification occurred in studied stands when bulk soil pHKCl ≥ 5. Short-root N % correlated positively with SAR/SAS. We concluded that tree N-nutritional status was related to short-root morphological parameters but not to studied microbiological characteristics in the soil of young silver birch stands.  相似文献

Context

Liming, an ameliorative method for acidified forest soils, may modify the structure of an arthropod community by altering the soil characteristics and/or the availability of food resources.

Aims

We investigated the effect of liming on the community structure of ground-hunting spiders in a birch forest.

Methods

We established six experimental birch stand plots. Each stand was exposed to one of three experimental treatments: control, 1.5 t/ha, or 3 t/ha of dolomitic limestone. We collected spiders using pitfall traps during 5 years. We characterized the community in terms of activity density, species richness, community-weighted mean body size, and functional diversity and evenness in body size. We further investigated the potential links through which the liming might affect spiders, namely soil characteristics, effect of liming on birch, and densities of potential prey.

Results

The commonly used dosage of 3 t/ha reduced densities of functionally similar species which led to the reduced functional evenness in body size and increased functional divergence in body size. Liming increased soil pH only slightly but decreased the densities of spiders’ preferred prey.

Conclusion

The liming affected the community of ground-hunting spiders, at least partially, through reduced densities of their preferred prey.

In conventional drying, sawn birch (Betula sp.) timber darkens and reddens from the inside while the layer a few millimetres under the yellowish surface remains light in color. Lack of information concerning the chemical basis of the discoloration hinders the development of a reliable solution for this problem. In this study, the role of soluble proanthocyanidins in discoloration of birch wood was investigated because the polymerization and oxidation of these compounds are known to yield insoluble reddish compounds. Different periods of log storage affected the synthesis of soluble proanthocyanidins during conventional drying. Concentration of proanthocyanidins also correlated with changes in the color of birch wood. Discoloration appeared differently in conventionally dried and vacuum-dried wood, which indicates that the discoloration mechanism in these drying methods may differ chemically, and/or the compounds that take part in discoloration may be different at different drying temperatures.  相似文献

Abies nordmanniana and Abies lasiocarpa, established for Christmas tree production in southwestern Norway (58°44′N, 5°38′E), were treated with different weed control methods, including chemicals, use of black plastic mulch, grass or clover as ground cover, living mulch and mechanical hoeing. Ground cover with black plastic mulching resulted in the best growth and quality in A. lasiocarpa; the least favourable treatment was when grasses were allowed to grow close to the trees. Unless measures were taken to remove this competing vegetation, the ground cover grew over the trees, smothering 94% of them. A. nordmanniana plants grew to the same height on plots with no weed control as on plots with thorough weed eradication. Stem diameter decreased on plots with ground vegetation compared to seedlings grown in the weed-free environment. The use of Trifolium repens as ground cover decreased height growth by 30% compared to thorough weed control. Both grasses and clover sown as living mulch, damaged the trees more than did natural weed vegetation.  相似文献

Indonesia’s pulp and paper industry needs a large area of sustainably grown plantations to support its continued development. Acacia mangium has been the key species underpinning the pulp and paper industries in Sumatra, however increased disease pressure on A. mangium is expected to require large-scale conversion of Acacia plantations to Eucalyptus in the near future. The effect of such a large scale change in plantation species on soil moisture, for both tree production, and catchment hydrology is unknown. In this study we sought to characterize the impacts of plantation species (Acacia or Eucalyptus) and nitrogen management, on soil moisture, soil water depletion and depth to groundwater under stands of Acacia mangium and Eucalyptus pellita over the first 2–3 years after establishment. The study was conducted in experiments at four sites in Sumatra, Indonesia. Soil moisture and soil water depletion were not influenced by plantation species or fertilizer treatment. Soil moisture content and soil water depletion were strongly influenced by shallow groundwater at two of the four sites, however depth to groundwater did not influence stem growth. Results from the field trials cautiously suggest that large scale conversion of Acacia mangium to Eucalypt species in these regions is unlikely to result in increased moisture stress, nor is conversion of plantation species likely to lead to substantial differences in catchment hydrology. This study demonstrated the importance of conducting multi-site studies when investigating biophysical relationships in forest/plantation systems.  相似文献

Spatial pattern of recruitment is an important factor influencing population dynamics of plant communities. The spatial pattern is determined by seed dispersal and by the spatial variability of germination and initial survival. In the process of forest expansion following farmland abandonment, mid- and late-successional species are often dispersed in pioneer forests by birds. This could result in an aggregated spatial pattern of seeds that could influence the dynamics of these species in mixed pioneer forests. In the sub-Mediterranean area, mid- and late-successional species such as Quercus pubescens (downy oak) and Fagus sylvatica (European beech) are expected to replace pioneer Pinus species. Using a point sampling method we demonstrated that beech and oak seedlings (height <2 m) have a clumped distribution in the understorey of pine. This could result from an aggregated dispersal by jays (dispersal effect) or from preferential recruitment in particular habitats (habitat effect). To test these hypotheses we proposed a statistical analysis of spatial patterns of regeneration of beech and oak. Ground cover variables (i.e. cover by rock outcrops, herbs, box shrubs, mosses or pine) did not differ significantly around seedlings as compared with random sample plots. Likewise, clumped seedlings had growth similar to isolated seedlings, thus refuting the hypothesis of preferential location in the most favourable microsites. Aggregated dispersal seems to be involved in the process of regeneration. Since beech and oak seedlings have contrasting ecological demands, we discuss the implication of this pattern for the replacement dynamics of pine by these species.  相似文献

The problem of salinized soils has become one of the most serious constraints to agricultural and forest productivity. With the purpose of enhancing salt stress tolerance of Populus tomentosa, we transformed this tree species with spermidine synthase (SPDS) genes derived from an apple by an Agrobacterium-mediated method. The relationship between Xenorhabdus nematophilus and Photorhabdus luminescens, the bacterial symbionts of the entomopathogenic nematodes Steinernema carpocapsae and Heterorhabditis bacteriophora, against the diamondback moth, P. xylostella (L.), pupae showed the pathogenic capability of P. luminescens to be over that of X. nematophilus. They gave 60 and 40?% mortality with LC50 values of 5?×?104 and 5.5?×?105 cells/ml, respectively.The number of bacterial cells influences the rate of killing the pupae of P. xylostella and a direct correlation between mortality of the pupae, percentage of deformed adults and the spraying dose with significant differences were observed. An inverse correlation found between the no. of eggs laid/female, percentage of hatching, adult survival and the spraying dose. These bacterial effects have been attributed to different toxic bacterial enzymes which damage the haemocytes and inhibit activation of the humoral immune system of the insect.  相似文献

Young trees were harvested to explore non-destructive methodologies to estimate live branch dry weights in young fast-growing Eucalyptus species under different spacing and fertilizer treatments. Branch growth can vary with silvicultural management such as spacing, fertilizing and thinning, and over relatively short periods in response to environmental conditions. Many published regressions based on standard measurements of height and diameter are site, age and treatment specific. The aim of this study was to improve our capacity to predict woody crown dry weight, based on stem measurements, and to minimize (or eliminate) treatment effects on the resulting model. In young trees, branches are temporary support structures for foliage and are often discarded as the base of the green crown rises. As temporary structures they represent an investment of biomass and nutrient elements, and are subject to selection pressures to maximize the return on investment by the tree. Trees were harvested from existing plantation experiments located in south-eastern Queensland for E. grandis W. Hill ex Maiden (ranging from 0.28 to 15.85 m in height, to 5 years old) and south-western Australia for E. globulus Labill. (0.10–34.4 m in height, to 10.2 years) in order to examine the impact of spacing, nitrogen and phosphorus fertilization on early growth. Relationships to estimate crown woody biomass from non-destructive measurements were developed, and these relationships tended to have different slopes and intercepts for trees with predominantly juvenile foliage and those with intermediate or adult foliage. Dry weight of whole-crown live branch wood (Wbranch) was related to heights and/or diameter at breast height (DBH), but the regressions parameters were different, depending on treatment. The relationships became more generic (i.e. less dependent on treatment effects) between Wbranch and stem sectional area at the height of the base of the green crown (SACB), consistent with the pipe model theory (R2 > 0.91 for the two species for trees with intermediate/adult leaves). However, Wbranch was more closely related again to the stem volume above the base of the green crown and treatment effects were not significant (VCon,gc, R2 > 0.93). Branches exit the stem below the green crown, and for E. grandis the best relationship was on stem volume above the lowest live branch (VCon,llb, R2 0.94). Limited sampling from four other species with similar or contrasting crown characteristics indicated that the relationship could be applied quite generally. Individual E. grandis branch woody dry weight was closely related to the conical volume of the main (first order) branch (Vcon,br, R2 0.98). The whole crown equivalent, branch woody dry weight plus stem dry weight above the lowest live branch, was also closely related to the stem volume within the woody crown (VCon,llb, R2 0.97–0.99). While the slope of this relationship was still significantly different between trees with juvenile and intermediate/adult foliage, it had a similar form, suggesting that trees with juvenile foliage allocated a different proportion of their woody biomass within the crown to branches than older trees.  相似文献

We investigated the effects of two commercial diatomaceous earth based insecticides (DE), Protect-It® and SilicoSec®, the nano-structured silica product AL06, developed by the section for Urban Plant Ecophysiology at Humboldt University Berlin, and the monoterpenoids, eugenol, and cinnamaldehyde on two stored product pests, Callosobruchus maculatus and Sitophilus oryzae. Protect-It® was more effective than SilicoSec® against C. maculatus while the reverse was true for S. oryzae. Generally C. maculatus was more sensitive towards DE and silica treatment than S. oryzae. Mortality rate of both pest species increased when DE’s were applied to food commodities previously treated with a monoterpenoid. In admixture experiments, the toxicity of SilicoSec® + cinnamaldehyde (LD50 = 42.73 ppm), SilicoSec® + eugenol (LD50 = 24.30 ppm), and Protect-It® + eugenol (LD50 = 2.60 ppm) was increased over DE alone against S. oryzae. Both substances showed a synergistic effect considering their co-toxicity coefficient relative to the LD50-value. In contrast, we could not find any synergistic effects in experiments with C. maculatus. Here only Protect-It® + cinnamaldehyde (LD50 = 20.84 ppm) showed an additive effect while all other combinations of monoterpenoid and DE indicated antagonistic effects. In addition to contact insecticidal effects both monoterpenoids showed a strong fumigant action. The presented results indicate that the natural product DE has great potential to replace synthetic pesticides commonly used in stored product pest management. Efficacy of DE can be improved by adding certain monoterpenoids against certain insect pests.  相似文献

Red flour beetle Tribolium castaneum (Herbst) and rice weevil Sitophilus oryzae (L.) are considered to be the major insect pests in storage. Essential oils from aromatic plants are recognized as proper alternatives to fumigants. Thymus persicus (Ronniger ex Rech. f.) is one of these plants that have medicinal properties and is indigenous to Iran. The essential oil was obtained from aerial parts of the plant and analyzed by GC and GC–MS. Carvacrol (44.69%) and thymol (11.05%) were the major constituents of the oil extracted. In this experiment, fumigant toxicity of the essential oil was studied against T. castaneum, S. oryzae at 27 ± 1°C and 60 ± 5% RH in dark condition. The adult insects were exposed to the concentrations of 51.9, 111.1, 207.4 and 370.4 μl/l air to estimate median lethal time (LT50) values. The fumigant toxicity was increased in response to increased essential oil concentrations. The LT50 values at the lowest and the highest concentrations tested were ranged from 28.09 to 13.47 h for T. castaneum, and 3.86 to 2.30 h for S. oryzae. It was found that S. oryzae adults were much more susceptible to the oil than T. castaneum. After 24 h of exposure, the LC50 values (95% fiducial limit) for T. castaneum and S. oryzae were estimated to be 236.9 (186.27–292.81) and 3.34 (2.62–4.28) μl/l air, respectively. These results suggest that T. persicus essential oil merits further study as potential fumigant for the management of these stored-product insects.  相似文献

The efficiency of the botanical insecticide (BI) NeemAzal T/S (containing 1% Azadirachtin A), on the basis of azadirachtin applied in a dose of 20 g a.i. ha−1 against Brassica pod midge (Dasineura brassicae), has been monitored for 4 years. The biological efficiency of BI was compared with the efficiency of some synthetic insecticides. It was ascertained that BI was very efficient in decreasing the number of damaged oilseed rape pods (ranging from 56.5 to 85.9% compared to untreated plants) and its efficiency was comparable with synthetic insecticides based on Chloronicotinyl (Thiacloprid) and Neonicotinoid (Acetamiprid). BI’s efficiency was, in some years, even significantly higher compared to pyrethroid (λ-cyhalothrin). The high biological efficiency of azadirachtin that we ascertained was significantly reflected in increasing potential crop yields. The yield increase of azadirachtin ranged between 9.3 and 19.4% compared to the control sample. Azadirachtin showed the highest yield for the whole time of experimentation, and in some years the yield increase was significantly higher compared to some synthetic agents.  相似文献

We studied the effects of the amount of advanced regeneration before logging and different understory treatments on forest recovery, particularly the establishment of lucidophyllous (evergreen broadleaved) species, following the logging of conifer plantation with a focus on the sprouting ability. The establishment of lucidophyllous tree and shrub species originating from advanced regeneration was closely related to the total amount of these elements in the advanced regeneration, regardless of understory treatment. The understory-left treatment could preserve understory as surviving individuals by high proportion (60% or more of the understory) compared with the understory-cleared practice after logging. However, half of the surviving individuals were damaged to some extent by logging and skidding. Relatively large trees were likely to survive with no damage, indicating that they face a lower risk than smaller trees. In understory-cleared stands, a high proportion (60% or more) of the understory regenerated by resprouting. However, the resprouting ratio depended on tree size and species. Therefore, as well as the amount of the understory, the tree size of understory before logging would be also an important factor for early forest recovery. Further, the infrequent (low-density) species could easily disappear after logging because of their low resprouting ratio and low density. Thus, recovery from resprouting alone would risk simplification of the species composition after logging. We concluded that leaving the developed understory intact before logging can enhance the speed and biodiversity of forest recovery.  相似文献

The success of tree plantings in agricultural landscapes within dry and variable rainfall regions is often below expectations. The aims of this study were to investigate techniques for establishing a local native species (Eucalyptus pauciflora) and an exotic species (E. nitens) in the Midlands of Tasmania, a tree-denuded region that has been grazed for over 100 years in Australia. We established trials at the Sorrel Springs and Woodland Park properties. We compared six establishment techniques: control; understorey nurse planting; spot burn; mulch; tree-guard and; self-watering tree-guard. Three months, 1 and 6 years after planting we measured tree height, health index and mortality. At Sorrell Springs tree-guarded treatments had greater height than other treatments at year one, but there were no effects of treatment at year six on height, health or mortality. At Woodland Park mulch, burn and tree-guarded treatments had greater height 1 and 6 years after planting. At year six tree-guarded treatments had lower mortality than other treatments. At Sorrell Springs height of E. nitens was greater than E. pauciflora at year one but not at year six. At Woodland Park height of E. nitens was generally greater at year six but having almost double the incidence of mortality relative to E. pauciflora. We conclude that expensive tree-guarding is only warranted where high browse pressure is expected and that on such sites species with tough leaves that are resistant to browsing should be considered. Further that in environments marginal for growing trees, best practice establishment technique is generally as effective as additional treatments of understorey nurse planting, spot burning or mulching.  相似文献

In the African rim of the Western Mediterranean Basin, cork oak forests and pine plantations coexist. Under similar fire regimes, cork oak forest is more resilient in terms of habitat structure (canopy, understory, and complexity of vegetation strata) than pine plantation. By contrast, both woodland types show similar resilience in plant species composition. Resilience in habitat structure varies between the two woodland types because of the resprouting and seeding strategies of cork oak and pine species, respectively. These differences can be relevant for the conservation of biodiversity of forested ecosystems in a future scenario of increased fire frequency and scale in the Mediterranean basin.

Context

Wildfires have major impacts on ecosystems globally. In fire-prone regions, plant species have developed adaptive traits (resprouting and seeding) to survive and persist due to long evolutionary coexistence with fire. In the African rim of the Western Mediterranean Basin, cork oak forest and pine plantation are the most frequently burnt woodlands. Both species have different strategies to respond fire: cork oak is a resprouter while pines are mostly seeders.

Aims

We have examined the hypothesis that pine plantations are less resilient in habitat structure (canopy, understory, diversity of vegetation strata) and plant composition than cork oak woodlands.

Methods

The habitat structure and plant species composition were measured in 30 burnt and 30 unburnt 700-m transects at 12 burnt sites from north-western Africa, where the two forest types can coexist. Habitat structure and plant species composition were compared between burnt and unburnt transects from cork oak and pine plantation woodlands with generalized linear mixed models and general linear models.

Results

The results showed significant interaction effect of fire and forest type, since cork oak forest was more resilient to fire than was pine plantation in habitat structure. By contrast, both forest types were resilient to fire in the composition of the plant communities, i.e., plant composition prior to fire did not change afterwards.

Conclusion

The higher structural resilience of cork oak forest compared to pine plantation is related to the resprouting and seeding strategies, respectively, of the dominant tree species. Differences in the responses to fire need to be considered in conservation planning for the maintenance of the Mediterranean biodiversity in a future scenario of changes in fire regime.

Key message

A new system of additive tree biomass equations was developed for juvenile white birch plantations based on tree diameter at breast height (DBH) and tree height (HT). Compared with previous equations developed for natural white birch forests, the new system included one more biomass component and provided more accurate predictions.

Context

Accurate estimates of tree component and total biomass are necessary for evaluating alternative forest management strategies for biomass feedstock, carbon sequestration, and products. Previous biomass equations developed for white birch trees in natural stands provided substantially biased predictions for white birch plantations.

Aims

A new system of additive tree biomass equations was developed for juvenile white birch plantations in the northeastern China.

Methods

With destructive biomass sampling data from 501 trees sampled from white birch provenance and family trails at ages 7, 9, 10, and 13 in three provinces, a system of nonlinear additive tree biomass equations based on DBH and tree height was developed using the nonlinear seemingly unrelated regressions (NSUR) approach.

Results

Compared with previously published equations developed for natural white birch forests, the new system provided more accurate predictions of white birch tree component and aboveground and total biomass, especially of branch, foliage, and root biomass.

Conclusion

The new system extended the applicability of biomass equations to white birch plantations in the northeastern China.
