This study examined the influence of adding different amounts of maternal dietary l ‐carnitine and two fat types on fatty acid (FA) composition and the expression of lipid metabolism‐related genes in piglets. The experiment was designed as a 2 × 2 factorial with two fat types (3.5% soyabean oil, SO, and 3.5% fish oil, FO) and two levels of l ‐carnitine (0 and 100 mg/kg) added to the sows' diets. A higher proportion of n‐3 polyunsaturated fatty acids (PUFA) and a lower ratio of n‐6/n‐3 PUFA in sow milk and piglet tissues were observed in the FO groups than in the SO groups. Adding l ‐carnitine increased the proportion of C16:1 in sow milk and decreased n‐3 PUFA in piglet subcutaneous fat. Hepatic peroxisome proliferator‐activated receptor α (PPAR‐α) was more abundantly expressed in piglets from the FO groups than from the SO groups (p < 0.05), whereas stearoyl‐CoA‐desaturase (SCD), sterol regulatory element binding protein‐1 (SREBP1) and ?6‐desaturase (D6D) genes were less expressed in the FO groups compared with piglets from the SO groups. The expression of fatty acid synthase (FAS) genes was decreased in the SO groups with l ‐carnitine compared to that of the other dietary treatments. No differences among dietary treatments were observed with regard to the expression of acetyl‐CoA carboxylase (ACC). In conclusion, FO and l ‐carnitine supplementation in sows affect FA composition and hepatic gene expression in piglets.  相似文献

Thirty‐six Holstein‐Friesian crossbred lactating dairy cows were used to determine the effects of linseed oil supplementation on performance and milk fatty acid (FA) profile. Three treatments were as follows: basal diet (56:44 Roughage:concentrate [R:C] ratio, dry matter basis) supplemented with 500 g of palm oil as control (PO), 500 g mixture (1:1, w/w) of palm oil and linseed oil (POLSO) and 500 g of linseed oil (LSO). The LSO supplementation had no effects on total dry matter intake (DMI), milk yield and milk composition. Compared to control cows, cows supplemented with LSO increased milk concentrations of cis‐9,trans‐11 conjugated linoleic acid (CLA) and n‐3 FA (P < 0.05), particularly C18:3n‐3, C20:5n‐3 and C22:6n‐3. Feeding LSO reduced concentrations of milk short‐ and medium‐chain saturated fatty acids (P < 0.05) while it increased concentration of milk unsaturated fatty acids (P < 0.05). Milk proportions of n‐3 FA increased, whereas n‐6/n‐3 ratio decreased in the LSO as compared with the control (P < 0.05). In conclusion, supplementing dairy cows' diet based on corn silage with LSO at 500 g/day could improve the nutritional value of milk with potential health‐beneficial FA without detrimental effect on milk composition or cow's performance.  相似文献

The aims of this study were to investigate the influence of the short‐term addition of sunflower and linseed oil and castration on fatty acid composition and desaturation indexes in chicken broilers. Forty‐eight male Ross 308 chicken broilers were supplemented with 5% of sunflower or linseed oil. The four experimental groups were linseed oil supplementation and castration (LC), linseed oil without castration (LN), sunflower oil and castration (SC) and sunflower oil without castration (SN). There was no significant influence of castration or oil supplement on live weights, weight gain, feed intake or feed conversion. Castration resulted in an increase in polyunsaturated fatty acids (PUFA), total n3, n6, measured desaturation indexes and a decrease in the saturated fatty acid (SFA) and monounsaturated fatty acid (MUFA) content of abdominal fat. In breast muscle, castration increased PUFA and 18:3n3 values, while in the liver tissue, castration did not influence the parameters measured. Linseed oil supplementation significantly increased 18:3n3, n3 long chain polyunsaturated fatty acids (LC PUFA), total n3 and decreased total n6, n6/n3 ratio, and 20:4n6 content. Values for 20:4n6 were the highest in SC and the lowest in the LC group. Linseed oil also significantly decreased ?5 and ?4 desaturation indexes in the thighs and ?5 and ?5, 6 in abdominal fat and the liver. These results suggest that short‐term supplementation of basal diet with 5% of linseed oil could significantly increase n3 LC PUFA and decrease n6/n3 ratio content in the edible tissues of chicken broilers, without adverse effects on growth performance. Meanwhile, castration only improved fatty acid profile in abdominal fat, which is not nutritionally important. The interactions observed between basal diet, supplemented oil, sex hormones and other non‐nutritional factors must be elucidated in future trials in order to correctly predict the nutritional value of linseed‐fed poultry.  相似文献

胡麻是一种富含不饱和脂肪酸尤其是α-亚麻酸的油料作物,明晰TAG合成途径中与胡麻含油量和脂肪酸组分相关的基因具有重要的理论意义。以3个含油量和脂肪酸组分有显著差异的胡麻品种(系)为材料,分析了不同品种(系)、不同组织和不同发育阶段胡麻油脂和脂肪酸组分的动态积累模式和TAG合成途径中7个关键基因的动态表达模式,以及含油量和不饱和脂肪酸与7个关键基因表达的相关性。结果表明,胡麻开花后10~20d,是种子油脂和亚麻酸的快速积累期,且不同胡麻品种(系)油脂和亚麻酸的动态积累模式差异显著。TAG合成途径中的7个关键基因(GPAT9、DGAT1、DGAT2、PDAT1、PDAT2、FAD2A和FAD3A)在胡麻不同发育阶段的不同组织中均有表达,在不同品种(系)间的表达模式各不相同。其中PDAT1、DGAT1和DGAT2基因的动态表达模式与含油量的动态积累模式显著正相关,PDAT1的动态表达模式与亚麻酸的动态积累模式显著正相关,且在高油高亚麻酸材料的种子动态发育阶段中PDAT1基因的累积表达量也显著高于低油低亚麻酸材料。因此,PDAT1可能是影响胡麻不同品种(系)中含油量和亚麻酸含量的关键基因。  相似文献

N‐3 polyunsaturated fatty acids (PUFA) are essential for foetal development. Hence, including n‐3 PUFA in the sow diet can be beneficial for reproduction. Both the amount and form (precursor fatty acids vs. long chain PUFA) of supplementation are important in this respect. Furthermore, including n‐3 PUFA in the diet can have negative effects, such as decreased arachidonic acid (ARA) concentration and increased oxidative stress. This study aimed to compare the efficacy to increase eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) concentrations in the piglet, when different concentrations of linseed oil (LO, source of precursor α‐linolenic acid) or fish oil (FO, source of EPA and DHA) were included in the maternal diet. Sows were fed a palm oil diet or a diet including 0.5% or 2% LO or FO from day 45 of gestation until weaning. Linoleic acid (LA) was kept constant in the diets to prevent a decrease in ARA, and all diets were supplemented with α‐tocopherol acetate (150 mg/kg) and organic selenium (0.4 mg/kg) to prevent oxidative stress. Feeding 0.5% LO or 0.5% FO to the sows resulted in comparable EPA concentrations in the 5‐day old piglet liver, but both diets resulted in lower EPA concentrations than when 2% LO was fed. The highest EPA concentration was obtained when 2% FO was fed. The DHA level in the piglet liver could only be increased when FO, but not LO, was fed to the sows. The 2% FO diet had no advantage over the 0.5% FO diet to increase DHA in the piglet. Despite the constant LA concentration in the sow diet, a decrease in ARA could not be avoided when LO or FO were included in the diet. Feeding 2% FO to the sows increased the malondialdehyde concentration (marker for lipid peroxidation) in sow plasma, but not in piglets.  相似文献

The transfer of passive immunity from sows to piglets is important and it is the first immune protection of the new born piglet. Improving sows immunity by adding immuno‐stimulating product in sows diet can positively affect colostrum composition and transfer of immune molecules to piglets. The aim of the current study is to evaluate the benefit of a different solution, made of specific fatty acids from marine origin that have been used in human medicine for decades, for sows and piglets. Two trials were conducted in commercial farm, involving 240 sows at different periods of the year. Sows were divided in a control group, without supplementation, and a test group, supplemented with the feed additive from the 90th day of gestation to weaning. Sows body condition, piglets viability and growth were recorded. Milk immunoglobulin content was measured, as well as Aujeszky antibodies in sows and piglets blood as marker of specific immunity, and blood bactericidal activity, complement activity and lysozyme as markers of non specific immunity. No effect of the product was observed on piglets zootechnical criteria and specific immunity parameters but significant improvement of piglet non specific immunity, was observed. No difference was observed neither in the piglets blood PRRSV and PCV2 antibodies and viruses nor in Aujeszky antibodies. Blood complement activity seems to be an accurate indicator of immuno‐stimulating additive efficiency. Giving alkyl‐glycerol fatty acids to sows in late gestation and lactation can improve the passive immunity transfer to piglets.  相似文献

Linseed as a feed ingredient contains a number of constituents, such as mucilage, gel forming polysaccharides, structural carbohydrates and ω-3 fatty acids, which could have functional properties in relation to maintaining and supporting gastro-intestinal health in post weaning piglets. Intestinal gene expression was studied in weanling piglets fed diets containing either or not linseed or linseed expeller meal. Piglets were either or not orally challenged with an enterotoxigenic strain of E. coli K88. Pairwise comparisons identified several mRNAs expressed significantly different, in particular between treatments C− (control, not challenged) and LS− (linseed, not challenged) and LS+ (linseed, challenged) and LSM+ (linseed expeller meal, challenged). The intestinal micro-array technique is a promising tool to study the relationship between nutrition and feeding and gut tissue responses under defined conditions.  相似文献

Liver metabolism is affected by nutrients. The aim of this study was to explore the effects of low‐protein diets (17% crude protein, CP) supplemented with branched‐chain amino acids (BCAAs), including leucine (Leu), isoleucine (Ile) and valine (Val), on hepatic amino acid profile and lipid metabolism in growing pigs. The ratio of Leu : Ile : Val in all groups was 1 : 0.51 : 0.63 (20% crude protein, CP), 1 : 1 : 1 (17% CP), 1 : 0.75 : 0.75 (17% CP), 1 : 0.51 : 0.63 (17% CP) and 1 : 0.25 : 0.25 (17% CP) respectively. Results revealed that compared to the positive control group (1 : 0.51 : 0.63, 20% CP), the low‐protein diets significantly augmented the concentrations of most essential amino acids and non‐essential amino acids (< .05), with the greatest values observed in the 1 : 0.25 : 0.25 group. Moreover, relative to the control, the low‐protein diets with the Leu : Ile : Val ratio ranging from 1 : 0.75 : 0.75 to 1 : 0.25 : 0.25 markedly downregulated the mRNA abundance of acetyl‐CoA carboxylase (ACC), lipoprotein lipase (LPL) and fatty acid‐binding protein 4 (FABP‐4) (< .05), and upregulated the mRNA expression of hormone‐sensitive lipase (HSL), peroxisome proliferator‐activated receptor‐g coactivator‐1α (PGC‐1α), uncoupling protein 3 (UCP3) and liver carnitine palmitoyltransferase 1 (L‐CPT‐1) (< .05). Therefore, our data suggest that protein‐restricted diets supplemented with optimal BCAA ratio, that is, 1 : 0.75 : 0.75–1 : 0.25 : 0.25, induce a shift from fatty acid synthesis to fatty acid oxidation in the liver of growing pigs. These effects may be associated with increased mitochondrial biogenesis.  相似文献

Twenty primiparous sows were allocated to two treatments to evaluate the effects of maternal 25‐hydroxycholecalciferol (25OHD3) supplementation during gestation and lactation on milk quality and serum bone status markers of sows and bone quality of piglets. Immediately after mating, sows were randomly allotted to one of two diets supplemented with 50 µg/kg 25OHD3 or basal diets without 25OHD3. Blood and milk samples were obtained. At birth and weaning, 10 piglets from each treatment were killed for bone quality analysis. 25OHD3‐fed sows provided one more piglet at farrowing and 1.17 more piglets at weaning than sows fed basal diets. The contents of solids not‐fat, protein, fat or lactose were increased in milk from days 7 and 14 of lactation in 25OHD3‐supplemented sows and 25OHD3 concentrations in milk were increased by dietary 25OHD3 supplementation. Dietary 25OHD3 supplementation increased serum alkaline phosphatase activity but had no effect on serum tartrate‐resistant acid phosphatase activity of sows. Maternal 25OHD3 supplementation improved bone strength, density and ash content of newborn piglets rather than those of weaning piglets. In conclusion, 25OHD3 supplementation in maternal diets improved reproductive performance, milk quality and bone status of sows as well as bone quality of newborn piglets.  相似文献

Maternal sodium butyrate (SB) intake has important effects on offspring growth and development. This study aimed to investigate the impacts of maternal SB supplementation during gestation and lactation on fatty acid composition and lipid metabolism in the offspring skeletal muscle of pigs. Twenty sows (Yorkshire, parity 2 to 3) were assigned to the control group (diets without SB, n = 10) and SB group (diets with 0.1% SB, n = 10). The results showed maternal SB supplementation throughout gestation and lactation increased (P < 0.05) body weight of offspring piglets at weaning. The concentrations of triglyceride in plasma and milk were enhanced (P < 0.05). Maternal SB induced (P < 0.05) lipid accumulation with increased expression of peroxisome proliferator activated receptor γ (PPARγ) by enrichment of the acetylation of H3 acetylation K27 (H3K27) in offspring skeletal muscle. Meanwhile, the concentrations of C18:2n-6, C18:3n-3, total polyunsaturated fatty acid (PUFA), n-6 PUFA and n-3 PUFA decreased (P < 0.05) in skeletal muscle of weaning piglets derived from SB sows. Together, these results showed that maternal SB supplementation could influence offspring growth performance, lipid metabolism and fatty acid composition of the skeletal muscle.  相似文献

The present study assessed the effect of feeding palm oil (PO), sunflower oil (SO) and their combination on performance, fat deposition, fatty acid composition and lipogenic gene expression of broilers reared for 42 days. A total of 144 1‐day‐old broilers (Cobb500) were randomly allotted into four treatment diets with each having six replicates of six chicks in each replicate following a completely randomized design. Live weight gain and feed efficiency was significantly (P < 0.05) higher in birds fed with a combination of oil sources compared to controls. Birds fed with the combination of oil and SO alone had higher carcass yield and lower abdominal fat. Higher (P < 0.05) concentrations of unsaturated fatty acids (UFA) and lower concentrations of palmitic acid and saturated fatty acid (SFA) was found in birds fed SO alone and combinations of SO and PO. Furthermore, the outcomes showed that birds fed diet supplemented with SO and the combination of SO and PO down‐regulated gene expression of key hepatic lipogenic enzymes of fatty acids synthase (FAS), acetyl‐CoA carboxylase (ACC) and stearoyl‐CoA desaturase (SCD). These findings suggest that the diet containing the combination of 2% PO and 4% SO may reduce hepatic lipogenesis, as well as lower abdominal fat content of broilers.  相似文献

Sterol regulatory element-binding factor 1 (SREBP1) plays an important role in the lipogenesis which affects fatty acid (FA) composition in backfat and consequently influences beef nutritional quality. This study analyzed the association of 84 bp-indel, both short (S) and long (L) alleles in intron 5 of SREBP1, with FA composition and gene expression of SREBP1 in backfat of northern Spanish beef breeds (Pirenaica, Salers and Holstein-Friesian). Phylogenetic analysis suggests that 84 bp-indel of ruminants is a highly conserved region compared with those in the full-length sequence of intron 5 or mRNA of SREBP1 among species. Overall, higher content of polyunsaturated FAs was observed in SL genotype compared to LL genotype of 84 bp-Indel (p < .05). In particular, in Pirenaica, SL genotype was associated with a higher content of stearic (18:0), α-linolenic (18:3n-3) acid, and total n-3 content (p < .05). However, the gene expression of SREBP1 did not differ among genotypes of 84 bp-Indel (p > .05).  相似文献

Antioxidants have been widely used during in vitro production to decrease the negative effect of reactive oxygen species. It was reported that the complex resveratrol–methyl β‐cyclodextrin (RV ‐CD ) improves resveratrol's stability and bioavailability and increases its antioxidant activity. This study evaluates the effect of RV ‐CD during in vitro oocyte maturation (IVM ) or in vitro embryo culture (IVC ) on developmental competence and quantitative changes in gene expression of developmental important genes. In experiment 1, RV ‐CD was added to IVM media and maturation level, embryo development and oocytes, cumulus cells, and blastocysts gene expression by RT ‐qPCR were examined. In experiment 2, presumptive zygotes were cultured in SOF supplemented with RV ‐CD and embryo development and blastocysts gene expression by RT ‐qPCR were studied. A group without RV ‐CD (control?) and a group with cyclodextrin (control+) were included. No differences were found in cleavage rate or blastocyst yield between groups. However, the expression of LIPE was higher in blastocysts derived from oocytes treated with resveratrol compared with control groups (<  .05). Blastocysts produced by IVC with resveratrol showed that RV ‐CD could modify the expression of genes related to lipid metabolism (CYP 51A1 , PNPLA 2 and MTORC 1 ) compared with control groups (p  < .05). RV ‐CD in the IVM and IVC media could reduce accumulated fat by increasing lipolysis and suppressing lipogenesis of blastocysts.  相似文献

We performed a genome‐wide association study (GWAS) and candidate gene analysis to: (i) evaluate the effectiveness of the GWAS in our small population by performing GWAS for carcass weight (CW) and fatty acid composition; (ii) detect novel candidate regions affecting non‐CW carcass traits, chemical composition and sugar; and (iii) evaluate the association of the candidate genes previously detected in CW and fatty acid composition with other economically important traits. A total of 574 Japanese Black cattle and 40 657 Single nucleotide polymorphisms were used. In addition, candidate gene analyses were performed to evaluate the association of three CW‐related genes and two fatty acid‐related genes with carcass traits, fatty acid composition, chemical composition and sugar. The significant regions with the candidate genes were detected for CW and fatty acid composition, and these results showed that a significant region would be detectable despite the small sample size. The novel candidate regions were detected on BTA23 for crude protein and on BTA19 for fructose. CW‐related genes associated with the rib‐eye area and fatty acid composition were identified, and fatty acid‐related genes had no relationship with other traits. Moreover, the favorable allele of CW‐related genes had an unfavorable effect on fatty acid composition.  相似文献

Atmospheric ammonia in animal housing is reported to have adverse effects on livestock performance and animal health. Previous experiments have found that 75 ppm ammonia reduced the production performance and altered body fat distribution quality of broilers. In this study, we examined the body fat distribution, serum metabolites and lipid metabolism gene expression of broiler exposed to ammonia. A total of 400 chickens were randomly allocated to four groups with four replicates and received ammonia treatments at 0, 25, 50 and 75 ppm, respectively, for 3 weeks. The average daily feed intake and weight gain were decreased when broiler was exposed to ammonia concentration exceeding 50 ppm (< .05). The increased abdominal fat and reduced thickness of subcutaneous adipose were found in broilers of 75 ppm group (< .05). When ammonia exceeded 50 ppm, the content of fat in breast muscle of broiler was increased, and when ammonia was higher than 25 ppm, the fat in liver was increased (< .05). It showed that the fat content in liver was a sensitive index for broilers exposed to ammonia. Furthermore, ammonia exposure had no significant effect on total cholesterol and triglyceride in serum, but significantly increased the relative mRNA expression of acetyl‐CoA carboxylase (= .046) and malic enzyme in liver (= .038), which indicated that ammonia exposure may increase the de novo fat synthesis in liver. In addition, ammonia increased the high‐density lipoprotein cholesterol (= .02) and activity of hepatic lipase in serum (< .001), which indicated that ammonia exposure may improve the transportation of cholesterol to liver. To conclude, our results indicated that ammonia exposure might increase the de novo fat synthesis in liver and increased the transportation of cholesterol to liver. In addition, the concentration of ammonia in poultry house should be limited lower than 25 ppm based on the variation of hepatic fat content.  相似文献

This study aimed to evaluate the hepatic glucose and lipid metabolism‐related parameters of adult male and female White King pigeons (Columba livia) during incubation and chick rearing. At day 4 (I4), 10 (I10) and 17 (I17) of incubation and day 1 (R1), 7 (R7), 15 (R15) and 25 (R25) of chick rearing, livers were sampled from six pigeons for each sex. Glycogen and fat contents, activities of glycolytic enzymes (hexokinase, HK; 6‐phosphofructokinase, 6‐PFK), and genes expressions of key enzymes involved in glycolysis (pyruvate kinase, PK; glucokinase, GK), gluconeogenesis (phosphoenolpyruvate carboxykinase cytosolic, PCK1; fructose‐1,6‐bisphosphatase, FBP1; glucose‐6‐phosphatase, G6Pase), fatty acid synthesis (fatty acid synthase, FAS; acetyl‐CoA carboxylase, ACC) and fatty acid β‐oxidation (carnitine palmitoyltransferase 1, CPT1; acyl‐CoA 1, ACO) were measured. In male and female pigeon livers, glycogen content and HK activity dramatically increased after I17 and after R1, respectively; expressions of FBP1 and G6Pase genes were maximized at R15; activity of 6‐PFK and expressions of PK and CPT1 genes were highest at R7; fat content and expressions of FAS and ACC genes steeply increased from I10 to R1. In females, hepatic expressions of GK and PCK1 genes were greatest at R7 and I17, respectively; however, in males, both of them were maximized at R15. Hepatic expression of ACO gene was significantly enhanced at R1 compared to I17 and R7 in males, whereas it was notably up‐regulated at I17 and R7 in females. Furthermore, expressions of PCK1, GK, FAS and ACC genes were in significant relation to fat content in the livers of female pigeons, while fat content in male pigeons was highly correlated with expression of PCK1, ACC, CPT1 and ACO genes. In conclusion, regulations of glucose and lipid metabolic processes were enhanced in parent pigeon livers from terminal phases of incubation to mid phase of chick rearing with sexual effects.  相似文献

The cryopreservation of testicular tissue is a potential method for preserving male fertility. However, the effect of cryopreservation on bovine calf testicular tissue is scarce. This study investigated the effect of different cryoprotectants on bovine calf testicular tissue at the molecular level. Testicular tissue from ten immature bovine calves (6 months) was collected after slaughter and cryopreserved in an extender containing different concentrations of the following five cryopreservation solutions (CP): bovine serum albumin (BSA) with 5% dimethyl sulfoxide (DMSO), trehalose with 5% DMSO, DMSO and glycerol and ethylene glycol (EG). After 7‐day cryopreservation, the expression levels of three spermatogonial stem cell (SSC)‐related genes, octamer‐4 (OCT4), KIT ligand (MGF/SCF) and kit oncogene (C‐KIT), were investigated by quantitative PCR (qPCR). The cell viability was highest for the tissues preserved with 30 mg/ml BSA (77.82% ± 1.22) and 40 mg/ml trehalose (74.23% ± 1.16) compared with other groups (p < 0.05), and the level of expression of the three genes was highest with 30 mg/ml BSA (p < 0.05). Compared with other CPs, the 30 mg/ml BSA and 40 mg/ml trehalose have the better cryopreserve protection. The 30 mg/ml BSA is the most viable media for the cryopreservation of testicular tissue from cattle.  相似文献

The aim of this study was to compare the influence of a high α-linolenic acid (ALA) diet (linseed diet) and a high linoleic acid (LA) diet (sunflower diet) on performances of pigs, on the dietetical quality of their tissues (adipose tissues and muscles), and on the lipogenic potential of these tissues. Growth and carcass performances, and the lipid content of the tissues were not affected by the diet. Feeding the linseed diet increased the content of n-3 PUFA and decreased the LA/ALA ratios in all the tissues, while the sunflower diet led to an increase in the n-6 PUFA contents. Neither the stearoyl-CoA-desaturase and glucose-6-phosphate-dehydrogenase activities nor the β-hydroxyacyl-CoA-dehydrogenase activity were affected by the diet. Acetyl-CoA-carboxylase and fatty acid synthase activities were enhanced with the linseed diet in respectively subcutaneous adipose tissue and intermuscular adipose tissue, while malic enzyme activity was decreased in liver and subcutaneous adipose tissue of pigs fed the linseed diet.  相似文献
