Hematologic parameters of 23 foals were studied at weeks 0, 2, 4, 6, and 10 following the onset of a strangles epizootic. The epizootic was initiated by group exposure to a foal experimentaUy infected with S, equi. The group consisted of 12 foals previously exposed to an S. equi epizootic 6 months prior, and 11 previously unexposed foals. Seventeen percent of the previously exposed foals and 91% of the unexposed foals developed clinical signs of strangles. Significant increases in mean white blood cell count, neutrophil cell count, fibrinogen concentration, and plasma protein concentration were seen in strangles cases, compared to foals not classified as cases, and were associated with clinical signs. Decreases in packed cell volume, hemoglobin concentration, and red blood cell count, although statistically insignificant, were observed in strangles cases compared to noncases during weeks 4, 6, and 10 and may have biological significance. Similar, but more pronounced, changes were observed in the hematologic parameters of the foal experimentally inoculated with S. equi. The effect of Streptococcus equi infection on the hematology of foals should be considered in their convalescent care.  相似文献

Newborn foals, deprived of colostrum and its rich supply of immunoglobulin G (IgG), were supplemented both orally and intravenously with purified equine immunoglobulin G (Lyphomune®). Data were obtained from 18 foals given oral administration of IgG at Colorado State University and 26 foals given IgG intravenously at the Jockey Club de Sao Paulo in Brazil.Oral administration of 10-gm doses of Lyphomune® in 18 colostrum-deprived Arabian foals, at various intervals within the first 24 hours after birth, resulted in increased serum concentrations of IgG. Administration of one 10-gm dose of Lyphomune® immediately following birth provided a mean serum IgG level of 125 mg/dl after two hours. The recommended dosage of two 10-gm doses per 15 kg of body weight produced mean IgG serum concentrations of approximately 400 mg/dl by 14 hours. It was determined that an early bolus of IgG was most effective, although administration at any period during the first 24 hours would increase IgG levels significantly and in direct relationship to grams of Lyphomune® administered.After the 24-hour study period, colostrum from each respective mare was provided by bottle feeding (200 ml) to 10 of the foals that were then allowed to nurse their dams normally. Significant increases in circulating IgG were observed in nine of these ten animals at four and eight hours after colostrum administration. No interfering effect was noted when colostrum and Lyphomune® were given to the same foal.Intravenous administration of 10-gm doses of Lyphomune® in Thoroughbred foals, immediately after birth, resulted in serum concentrations of IgG of 200–300 mg/dl six hours later. A second intravenous dose, at six hours after the initial dose, resulted in an additional average increase of 184 mg/dl. Four of six foals administered 10 gm of Lyphomune® for each 15 kg of body weight reached serum concentrations greater than 400 mg/dl. It was demonstrated that Lyphomune® was able to increase circulating levels of IgG, by either oral or intravenous administration, to levels considered protective in the newborn foal.  相似文献

The postnatal iron status of 20 trotter foals (7 female, 13 male) was studied by analyzing different parameters in blood drawn on the day of birth, and 14 +/- 2 and 28 +/- 2 days later. Hemoglobin hematocrit, plasma iron and transferrin saturation averaged 14.9 g/dl, 41.8%, 300 micrograms/dl and 59.4%, respectively, on the day of birth. These values were significantly reduced after 14 days, but had approached again the initial levels by day 28. There was a slight, statistically nonsignificant rise in the mean total iron-binding capacity over the four-week period. Iron status was not affecting by the sex of the foals. On the day of parturition, the mares had much lower levels of plasma iron and transferrin saturation than their foals. In the discussion, the importance of checking the iron status of each individual foal during the critical period of the first 14 days post partum is stressed.  相似文献

Rhodococcus equi causes severe pyogranulomatous pneumonia in foals and in immunocompromised people. In mice, both CD4+ and CD8+ T lymphocytes contribute to host defense against R. equi, but CD4+ T lymphocytes are required for pulmonary clearance of the bacteria. In this prospective study of 208 foals at two equine breeding farms with endemic R. equi infections, we collected peripheral blood samples at 2 and 4 weeks of age and at the time of diagnosis of R. equi pneumonia. Samples were analyzed for concentrations of total and differential leukocytes, EqCD4+ and EqCD8+ T lymphocytes, and B lymphocytes. Thirty (14.4%) foals developed R. equi pneumonia. At the 2nd week of life, affected foals had significantly lower concentrations of white blood cells (WBC) and segmented neutrophils, significantly lower proportions of EqCD4+ T lymphocytes, and significantly higher proportions of EqCD8+ T lymphocytes. The EqCD4:EqCD8 ratio was significantly lower for affected foals. At the 4th week of life, affected foals had significantly lower concentrations of segmented neutrophils and EqCD4+ T lymphocytes than did unaffected foals. The ratio of EqCD4:EqCD8 was significantly lower for affected foals. Two- and 4-week-old foals with ratios of EqCD4:EqCD8<3 were significantly more likely to develop R. equi pneumonia. There was a significant farm effect which diluted our statistical power to detect differences; however; after adjusting for the farm effect, 2-week-old foals with ratios of EqCD4:EqCD8<3 remained significantly more likely to develop R. equi pneumonia. There were no significant differences in immunophenotypic variables between affected foals (at the time of diagnosis) and age-matched control foals. These data suggest that there are hematologic and immunophenotypic differences between affected and unaffected foals during the first 2-4 weeks of life, prior to onset of clinical signs of R. equi pneumonia. These differences may represent important immunologic mechanisms associated with increased susceptibility of individual foals to infection with R. equi. Because there was considerable overlap between values for affected and unaffected foals, we cannot yet recommend immunophenotyping of foals at endemically-infected farms as a clinically useful screening tool to identify foals at increased risk of developing R. equi pneumonia.  相似文献

Iron deficiency anemia was identified and characterized in three 14 to 29-month-old male llamas (llama Nos. 1-3) from separate herds in Colorado. The identification of iron deficiency anemia was based on hypoferremia (serum iron = 20-60 micrograms/dl), erythrocytic features, and hematologic response to iron therapy. The anemia was moderate and nonregenerative and characterized by erythrocyte hypochromia, microcytosis (mean cell volume = 15-18 fl), and decreased mean corpuscular hemoglobin concentration (36.0-41.0 g/dl). Morphologic features unique to llamas with iron deficiency anemia included irregular distribution of hypochromia within erythrocytes and increased folded cells and dacryocytes. The cause of iron deficiency was not determined. The llamas were treated with various doses and schedules of parenteral iron dextran. Two of the llamas were monitored for up to 14 months after the start of iron therapy and experienced increases in hematocrit and mean cell volume values. In one llama, progressive replacement of microcytic cells with normal cells was visualized on sequential erythrocyte volume distribution histograms following iron therapy.  相似文献

Hematologic abnormalities consistent with iron deficiency anemia were experimentally induced in two healthy llamas by repeated phlebotomy. Hematologic abnormalities included erythrocyte microcytosis and hypochromia, decreased hemoglobin concentration, hypoferremia, and decreased transferrin saturation. Erythrocyte volume distribution histograms were more sensitive than mean corpuscular volume values for detection of microcytosis. Hypochromia, which was often eccentric, was morphologically observed on Wright-Giemsa-stained blood films. Frequent folded erythrocytes and dacryocytes were also noted on the blood films. Hematologic abnormalities resolved rapidly after cessation of blood removal, without parenteral iron supplementation.  相似文献

A 5-year-old, spayed, female husky-Labrador retriever cross was diagnosed with primary hypoadrenocorticism, an uncommon endocrine disorder caused by a deficiency of glucocorticoid and mineralocorticoid hormones. The long‐acting azalide antibiotic gamithromycin is marketed for intramuscular treatment of bovine and swine infections. Off‐label use in foals leads to severe local lesions likely caused by hyperosmolality of the injected solution. We provide evidence from a pharmacokinetic study in 10 warm‐blooded healthy foals for intravenous bolus injection of gamithromycin diluted in distilled water to be a safe and well tolerated alternative. By intravenous dosing, markedly higher plasma exposure and better penetration into bronchoalveolar lavage cells but lower distribution into epithelial lining fluid are achieved as after intramuscular or subcutaneous administration. Intravenously injected gamithromycin was tolerated without any adverse drug reactions. The protocols for treatment of equine pulmonary infections caused by Rhodococcus equi should be revised accordingly.  相似文献

Six healthy two-day-old foals (3 pony foals and 3 horse foals) were given a single intravenous (iv) injection of trimethoprim (TMP)--sulphamethoxazole (SMZ) at a dosage of 2.5 mg of TMP/kg bodyweight (bwt) and 12.5 mg of SMZ/kg bwt. Serum TMP and SMZ concentrations were measured serially during a 24 hour period. The overall elimination rate constant (K) for TMP in the pony and horse foals was 0.45/h, whereas the K values for SMZ for the pony and horse foals were 0.12/h and 0.07/h, respectively (no significant difference; P greater than 0.05). Based on published minimum inhibitory concentration values for equine pathogens (Adamson et al 1985), the primary indication for the use of TMP/SMZ in foals may be in the treatment of infections caused by gram-positive bacteria. A dosage of 2.5 mg of TMP/kg bwt and 12.5 mg of SMZ/kg bwt, given iv at 12 h intervals would be appropriate.  相似文献

The potential role of dietary copper in the development of cartilage defects in foals was investigated. Twenty-one mares were fed rations containing 13 ppm copper (CuC, control) or 32 ppm copper (CuS, supplemented) during the last three to six months of gestation and first three months of lactation. Their foals were fed pelleted concentrate containing 15 or 55 ppm Cu and were destroyed at 90 (5 CuC and 5 CuS foals) or 180 (6 CuC and 5 CuS foals) days. Focal cartilage lesions were found at multiple sites on necropsy. In foals killed at 90 days, there were over twice (9 versus 4) as many lesions of osteochondrosis and more than four times (9 versus 2) as many articular lesions of osteophyte formation or thinning in CuC foals compared with CuS foals. These differences were due predominantly to a higher number of lesions in one CuC foal. Two 90-day CuC foals had osteochondrosis of articular-epiphyseal (A-E) complex, one with thickenings and separation from subchondral bone and one with subchondral fibrosis. One 90-day CuS foal had a cartilage thickening of the A-E complex in the tibiotarsal joint with separation from subchondral bone. In foals killed at 180 days, there were seven times more articular lesions (21 versus 3) of osteophyte formation or thinning, nearly twice as many lesions of osteochondrosis (13 versus 8) [corrected] in the physis and over five times as many involving the A-E complex (11 versus 2) in six CuC foals compared with five CuS foals.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Twenty-one healthy Thoroughbred and Quarter Horse foals were studied from birth until 1 year of age. Foals had access to an iron-supplemented creep feed before weaning and were fed an iron-supplemented concentrate as part of their diet after weaning at 4 months of age. Initial blood samples were taken before foals were allowed to nurse. Serum iron concentration, total iron-binding capacity, and PCV decreased during the foal's first 24 hours of life. Serum iron concentration decreased rapidly from 446 +/- 16 micrograms/dl (mean +/- SE) at birth to 105 +/- 11 micrograms/dl at 3 days of age. Serum ferritin concentration increased from a mean of 85 +/- 8 ng/ml at birth to 159 +/- 11 ng/ml at 1 day of age. Thereafter, ferritin concentration decreased gradually to a minimum of 61 +/- 6 ng/ml at 3 weeks of age, and then at 6 months increased to values similar to those from reference adult horses. The ferritin concentration in colostrum at birth was 354 +/- 42 ng/ml, compared with 25 +/- 2 ng/ml in milk 1 day later. The decrease and then increase in serum ferritin concentration occurred concomitantly with opposite changes in serum total iron-binding capacity. The mean PCV decreased gradually to a minimum at 3 months of age. This decrease was associated with an increasing number of microcytes, as determined with a cell-size distribution analyzer.  相似文献

Diets traditionally used in the horse industry are very often imbalanced in mineral composition, mainly in what concerns Ca, P, Mg and Ca/P ratio. The main objective of this study was to evaluate the effect of calcium, phosphorus and magnesium supplementation of traditional diets, on bone formation and growth in Lusitano breed foals, from weaning to 1 year old. Two homogenous groups of eight colts each received for 140-day iso-energetic and iso-proteic diets, which differed only on Ca, P and Mg levels (respectively, 0.26%, 0.27% and 0.12% vs. 0.56%, 0.43% and 0.18%). Throughout the experiment, blood samples were periodically collected for evaluation of total calcium, inorganic phosphorus, magnesium, copper, zinc, intact parathyroid hormone, bone alkaline phosphatase and osteocalcin concentrations. Animals were weighed and measured (wither height, hip height, girth and cannon circumference) for body growth evaluation. No significant differences were observed between treatments, with respect to growth, plasmatic concentrations of analysed minerals or biochemical markers. The linearity of growth in both groups is in agreement with studies reported for other light breeds. The observed results could be explained by the fact that calcium, phosphorus and magnesium levels in the base diet were close to supply the animal needs for a moderate growth. Further studies concerning the relationship between bone biochemical markers and mineral nutrition, in the growing horse, should consider the evaluation of bone mineral density and content.  相似文献

The objectives of this study were to determine pharmacokinetics of intravenous (i.v.) ceftiofur in foals, to compare ultra-high performance liquid chromatography tandem mass spectometry (UPLC-MS/MS) and microbiologic assay for the measurement of ceftiofur concentrations, and to determine the minimum inhibitory concentration ( MIC ) of ceftiofur against common equine bacterial pathogens. In a cross-over design, ceftiofur sodium was administered i.v. to six foals (1–2 days-of-age and 4–5 weeks-of-age) at dosages of 5 and 10 mg/kg. Subsequently, five doses of ceftiofur were administered i.v. to six additional foals between 1 and 5 days of age at a dose of 5 mg/kg q 12 h. Concentrations of desfuroylceftiofur acetamide (DCA), the acetamide derivative of ceftiofur and desfuroylceftiofur-related metabolites were measured in plasma, synovial fluid, urine, and CSF by use of UPLC-MS/MS. A microbiologic assay was used to measure ceftiofur activity for a subset of plasma samples. Following i.v. administration of ceftiofur at a dose of 5 mg/kg to 1–2 day-old foals, DCA had a t ½ of 7.8 ± 0.1 h, a body clearance of 74.4 ± 8.4 mL/h/kg, and an apparent volume of distribution of 0.83 ± 0.09 L/kg. After multiple i.v. doses at 5 mg/kg, DCA concentrations in CSF were significantly lower than concurrent plasma concentrations. Ceftiofur activity using a microbiologic assay significantly underestimated plasma concentrations of DCA. The MIC of ceftiofur required to inhibit growth of 90% of isolates of Escherichia coli , Pasteurella spp, Klebsiella spp, and β-hemolytic streptococci was <0.5 μg/mL. Intravenous administration of ceftiofur sodium at the rate of 5 mg/kg every 12 h would provide sufficient coverage for the treatment of susceptible bacterial isolates.  相似文献

The purpose of this study was to determine the pharmacokinetic profile of intravenous firocoxib in neonatal foals. Six healthy foals were administered 0.09 mg/kg firocoxib intravenously once a day for 7 days. Blood was collected for plasma firocoxib analysis using high‐performance liquid chromatography with fluorescence detection at times 0 (day 1 of study only) and 0.08, 0.25, 1, 2, 4, 6, 8, 16 and 24 hr on dose numbers 1, 5 and 7. Blood was also collected immediately prior to doses 3, 4, 5 and 7. Final samples were collected at 36, 48, 72 and 96 hr following the final dose. Noncompartmental analysis using the trapezoidal method with linear interpolation revealed a moderate half‐life (15.9 ± 9.1 hr) with a large volume of distribution at steady state (1.79 ± 0.57 L/kg) and a clearance (96.0 ± 59.2 ml h?1 kg?1) that was more rapid than that observed in adult horses.  相似文献

Equine thyroid disorders pose a diagnostic challenge in clinical practice because of the effects of nonthyroidal factors on the hypothalamic–pituitary–thyroid axis, and the horse's ability to tolerate wide fluctuations in thyroid hormone concentrations and survive without a thyroid gland. While benign thyroid tumours are common in older horses, other disorders like primary hypothyroidism or hyperthyroidism in adult horses and congenital hypothyroidism in foals are rare. There is a common misunderstanding regarding hypothyroidism in adult horses, especially when associated with the clinical profile of obesity, lethargy, and poor performance observed in dogs and humans. Low blood thyroid hormone concentrations are often detected in horses as a secondary response to metabolic and disease states, including with the nonthyroidal illness syndrome; however, it is important to note that low thyroid hormone concentrations in these cases do not necessarily indicate hypothyroidism. Assessing equine thyroid function involves measuring thyroid hormone concentrations, including total and free fractions of thyroxine (T4) and triiodothyronine (T3); however, interpreting these results can be challenging due to the pulsatile secretion of thyroid hormones and the many factors that can affect their concentrations. Dynamic testing, such as the thyrotropin-releasing hormone stimulation test, can help assess the thyroid gland response to stimulation. Although true hypothyroidism is extremely rare, thyroid hormone supplementation is commonly used in equine practice to help manage obesity and poor performance. This review focuses on thyroid gland pathophysiology in adult horses and foals, interpretation of blood thyroid hormone concentrations, and evaluation of horses with thyroid disorders. It also discusses the use of T4 supplementation in equine practice.  相似文献
