This study compared the growth of sea urchin Paracentrotus lividus larvae cultured using two different rearing methods: a variable method based on a variable amount of feed (microalgae) and seawater exchange (30% or 50%) established according to the phytoplankton concentration in the larval cultures and a fixed method characterized by a fixed amount of feed and seawater exchange. Three microalgae diets, Isochrysis sp. (Tahitian strain, T‐Iso), Chaetoceros gracilis and a 50:50 mixed diet, were tested with both rearing methods. Larval development and survival were assessed at the 6‐arm pluteus stage (P6), competence (Cp) and metamorphosis (Mt). Data showed that the variable method reduced the requirements for phytoplankton and seawater exchange. Indeed, through the optimization of feed rations, it was possible to reduce the production of debris and settled phytoplankton, minimizing the need for water exchanges. Higher larval survival resulted at Cp and Mt stages for those reared with the variable method as opposed to the fixed one. Survival and development were also influenced by the tested dietary treatments: at Mt stage, the mixed diet resulted in a higher larval survival (63.3 ± 8.9%) than T‐Iso (19.7 ± 12.1%) and C. gracilis (23.4 ± 15.1%) (P < 0.05). These results suggest that the use of the variable method improves the larval survival and development and also it reduces resource consumption (phytoplankton, seawater use and work effort), which in turn could potentially improve the hatchery production of P. lividus. 相似文献

The development of sustainable methods for sea urchin juvenile production is currently constrained by high mortality rates during larval growth and the high costs of larval rearing systems management. With the aim of developing a method for the production of juveniles of the purple sea urchin Paracentrotus lividus in a medium‐scale recirculating system, the present study focused on the effects of high stocking densities on larval growth. Plutei larvae were reared at three different densities (up to 7 ind/ml) in a semi‐static culture system. Larval survival and metamorphosis success were evaluated in order to identify the most effective density range. The highest metamorphosis rates (80%–95%) were obtained at 4 and 7 larvae/ml. These results are comparable with (and in some cases higher than) those reported for the same species at much lower larval densities. In conclusion, the rearing conditions tested here show for the first time that a significantly higher (4 ind/ml) stocking density than those of traditional P. lividus rearing methods (based on large volumes and low densities) can be adopted, thus supporting the feasibility of an increase in the final output of competent larvae with no increase in rearing volumes. 相似文献

The effects of formulated feeds on Paracentrotus lividus broodstock performances and carotenoid compositions were investigated. Performance of offspring derived from each dietary treatment was compared to determine whether maternal provisioning of nutrients affected offspring development and survival under commercial culture conditions. At the end of the experimental period (3 months), urchins were induced to spawn, relative fecundity measured, and offspring derived from each treatment were reared independently and survival up to competence assessed. Carotenoid composition of the gonads was also measured. Data showed that the highest dietary protein content significantly improved somatic growth, whereas higher lipid content increased gonadal index (GI) consistent with the use of gonads as nutrient storage in sea urchins. Nonetheless, gonad lipid concentration was not significantly different between treatments indicating that higher dietary lipid content favoured gonadal size rather than increase lipid content of the gonadal tissue. Interestingly, GI and fecundity were not correlated, although the latter may have been enhanced by dietary xanthophyll. This suggests the importance of including carotenoids in broodstock diets to enhance hatchery outputs. This study also indicated that, although broodstock diet can influence fecundity, it had no significant impact on larval survival, which could instead be influenced by rearing conditions. 相似文献

The objective of this study was to investigate the effects of extenders and storage time on motility, viability and fertilization of preserved black sharkminnow, Labeo chrysophekadion spermatozoa. Sperm were diluted 1:3 in one of five extenders: modified Cortland solution (MC); Hanks' balanced salt solution (HBSS); 0.9% sodium chloride (NaCl); Kurokura solution (KU); and modified extender, and undiluted sperm samples were used as control and stored at 4°C for 5 days. Motility, viability and fertilization rates were evaluated every day. After a storage time of three days, the highest motility, viability and fertilization rates (61.27 ± 2.26%, 58.60 ± 2.29% and 40.58 ± 0.57, respectively) were achieved with sperm diluted with modified extender. Motility, viability and fertilization rates decreased significantly (P < 0.05) with increasing storage time in all treatments. In addition, this study found that motility, viability and fertilization had a positive significant correlation (P < 0.01). The results indicate that isotonic extender is suitable for the short‐term preservation of black sharkminnow spermatozoa. 相似文献

The quality of the microalgae provided on Paracentrotus lividus larvae rearing is a primordial factor having a direct (nutritional properties) and indirect (water quality) impact on growth, competence and survival. Skeletonema costatum is a diatom commonly used in the bivalve cultivation. However, the use of this diatom in P. lividus larval cultivations is poorly known. The Rhodomonas spp. is a microalgae commonly used in sea urchin larvae culture. Three different diets were tested on P. lividus larvae and post‐larvae cultivation (D1—Rhodomonas marina, D2—S. costatum, D3—mixture of both algae). Larvae fed with the D2 diet (55.8%) and D3 (39.9%) had a survival at 15 DAH higher than D1 (5.5%). The low survival in D1 could be due to the higher microbiological load on microalgae (Vibrio alginolyticus and V. pectenicide). Larvae fed with S. costatum (D2) showed a lower development than other diets. The competency index was lower for larvae fed with the D2. These results show that microalgae diversified diets contribute to a better development of P. lividus larvae. During the settlement and post‐settlement phase, there was also a lower growth of the sea urchin fed with the D2 and a higher survival for D3. 相似文献

A pilot project aimed at testing roe enhancement strategies based on offshore Paracentrotus lividus cultures was conducted off the south‐east coast of Italy (Apulia Region). Adult sea urchins were reared in sea cages located 700 m offshore at a depth of 12 m for 3 months. The animals were fed once a week on two formulated diets, prepared mixing nutrients with agar 20 g/Kg and differing only in terms of the protein source: anchovy flour (Diet A) or krill flour (Diet K). At the end of the rearing trial, the gonad somatic index of sea urchins fed on formulated diets significantly exceeded that of wild sea urchins. Total FAA content in the gonads of wild sea urchins and Diet A‐fed sea urchins was similar, whereas in Diet K‐fed sea urchins it was significantly higher. In terms of fatty acids, the gonads contained SFAs, MUFAs and PUFAs. In visual and sensory assessment of gonads by panel test and electronic nose, the gonads of reared sea urchins were rated as being of better size, while no differences were recorded for coloration, taste and odour. This study shows that under these experimental conditions, commercial‐grade Paracentrotus lividus roe enhancement can be achieved after 3 months in sea cages. 相似文献

Knowledge on the biochemistry of proteins and amino acids (AA) of eggs and larvae of echinoids is scarce and the possibility to modify their profiles by diet manipulations is unknown. The protein content of eggs, prisms and pre‐plutei and the amino acid composition of eggs of Paracentrotus lividus from captive broodstock fed prepared diets were analysed and compared with the ones obtained from wild broodstock. Diets differed on protein source (fish or soy meals) and on protein content (10–40% dry weight – DW). Total and soluble protein content of both eggs and larvae was higher than 400 g kg?1 DW and 200 g kg?1 DW respectively. Glycine was the most abundant free AA. Very few differences were found among P. lividus eggs and larvae biochemical parameters, suggesting that their AA composition can hardly be changed by broodstock diet manipulations. 相似文献

The development of sperm cryopreservation has enabled transcendental changes to occur in the reproductive biotechnology of both mammals and fish; it has become a basic tool for animal improvement. Nevertheless, these protocols cause damage to cell structure and physiology, altering sperm functioning due to cryo‐injuries during freezing and thawing. However, studies of the effects on the structural, functional and genomic stability of the mitochondria in fish spermatozoa during cryopreservation are still lacking. The object of this review was to analyse the effect of cryopreservation on mitochondrial metabolic pathways in fish spermatozoa. This effect is related with the bioenergy mechanism for flagellar movement during the activation of sperm motility. In teleost fish, the mitochondria may be cylindrical, spherical or irregular in shape and adhere in a helicoidal or conical pattern to the middle piece. The salmonidae have only a single mitochondrion, but this may vary in other species; the mitochondria provide the flagellum with energy during sperm motility, when sperm respiration is essential. The effects of cryopreservation can induce structural damage to the mitochondria, altering the biochemical process involved in ATP production and thus causing a reduction in sperm motility. Fragmentation damage to nuclear DNA and diminution in sperm motility is mainly associated with damage to the structure and metabolic functioning of the mitochondrion. A direct correlation exists between the mitochondrial membrane potential (transmembrane integrity, ∆Ψm) and the motility and fertilizing capacity of the cryopreserved spermatozoa, confirming that this organelle is the energy nucleus of the spermatozoon and that the cessation or prolongation of motility and successful fertilization depend on the availability of ATP in the spermatozoa. Further works are need to incorporate biotechnology studies, at cell and molecular level, of the possible effects of cryopreservation on mitochondrial DNA, enzymatic or metabolic modifications of the citric acid cycle, and the oxidative phosphorylation process in the inner membrane, as well as studies of the mitochondrial ultrastructure. Thus, mitochondrial dynamics could be established as a potential target for therapeutic strategies. 相似文献

Spermatological research of the Patagonian blennie was carried, specifically biometric parameters, sperm density, sperm count and motility in different activation mediums (815, 716, 590 and 0 mOSm kg?1), at different temperatures (5, 10 and 15°C) and pH levels (5, 7 and 9). The results indicate that Patagonian blennie spermatozoa have a primitive form, with a total length of 44.09 ± 3.36 μm, with a head length of 2.15 ± 0.28 μm and head width of 2.5 ± 0.31 μm. The mid‐piece had a length of 0.72 ± 0.12 μm, and its tail measures 41.21 ± 3.21 μm long. The motility pattern indicates that the spermatozoa are found immobile in the seminal plasma and only initiates its movement in a hypertonic medium from 590 to 815 mOsm kg?1. The longest motility time that was registered at 10°C in 716 mOSm kg?1 was of 245 ± 39 s and an optimum pH of 7 was observed. 相似文献

Sea urchins are commercially harvested for their edible gonads, also known as roe or uni. Most species, particularly those from the Northern hemisphere are reported to exhibit limited variation in gonad colour, which is proposed to be due to carotenoid pigments. The commercially harvested species from New Zealand, Evechinus chloroticus, and from Australia, Heliocidaris erythrogramma, however, exhibit considerable gonad colour variation, ranging from a culinary desirable light yellow/orange to an undesirable brown. Gonads from E. chloroticus and H. erythrogramma were characterized spectrophotometrically and the carotenoid content extracted and analysed by reversed‐phase high‐performance liquid chromatography. The main carotenoid found in the gonads of both species was 9′‐cis‐echinenone, with lower levels of all‐trans‐echinenone, lutein and isozeaxanthin. Carotenoid composition contributed to the yellow/orange colour components of the gonads, but was not correlated with the darker roe coloration observed in some E. chloroticus individuals. Our results highlight important considerations relating sea urchin gonad coloration to carotenoid content and as such will provide useful information for the future aquaculture and processing of these two species. 相似文献

Under normal farming conditions, shrimp can experience starvation periods attributable to disease outbreaks or adverse environmental conditions. Starvation leads to significant morphological changes in the hepatopancreas (HP), being the main organ for absorption and storage of nutrients. In the literature, limited research has described the effect on the HP of periods of starvation followed by refeeding and none in whiteleg shrimp (Penaeus vannamei) using computer‐assisted image analysis (CAIA). This study describes the effect of starvation and starvation followed by refeeding on the HP of whiteleg shrimp using CAIA. Visiopharm® software was used to quantify the following morphological parameters, measured as ratio to the total tissue area (TLA): total lumen area (TLA:TTA), haemocytic infiltration area in the intertubular spaces (HIA:TTA), B‐cell vacuole area (VBA:TTA), lipid droplet area within R cells (LDA:TTA) and F‐cell area (FCA:TTA). Significant changes were measured for HIA:TTA and LDA:TTA during starvation (increase in HIA:TTA associated with decrease in LDA:TTA) and starvation followed by refeeding (decrease in HIA:TTA associated with increase in LDA:TTA). In the future, HIA:TTA and LDA:TTA have the potential to be used in a pre‐emptive manner to monitor the health of the HP, facilitate early diagnosis of diseases and study the pathophysiology of the organ. 相似文献

The aim of this study was to evaluate the effects of Roswell Park Memorial Institute (RPMI) 1,640 medium on chilled storage of eggs and spermatozoa of rainbow trout (Oncorhynchus mykiss). After 3 days of storage, eggs in RPMI 1,640 media (pH 8.2, 9 and 10), Cortland medium and coelomic fluid were inseminated with fresh spermatozoa (Experiment 1). Eggs in RPMI 1,640 medium at pH 8.2 shown the lowest thiobarbituric acid‐reactive substances (TBARS, 0.053 ± 0.003 nmol/ml) and pH changes (from 8.20 ± 0.01 to 8.18 ± 0.01), the highest fertilization rate (82 ± 3%). Undiluted and diluted spermatozoa at ratios of 1:2 and 1:9 with RPMI 1,640 media (pH 8.2, 9 and 10) and Cortland medium were inseminated with fresh eggs (Experiment 2). Spermatozoa in RPMI 1,640 medium at pH 9 (1:9) caused the lowest TBARS content (0.037 ± 0.002 nmol/ml) and pH changes (from 9.00 ± 0.01 to 8.98 ± 0.01), the highest fertilization rate (77 ± 2%) and motility parameters. Based on Experiments 1 and 2, eggs and spermatozoa were stored for another 3 days in RPMI 1,640 medium at pH 8.2 and 9 (1:9) respectively (Experiment 3). Fertilization rate of storage eggs and spermatozoa in Experiment 3 was 79 ± 5%, showing successfully storage of rainbow trout gametes with the same medium. 相似文献

In this study, cryopreservation feasibility of Persian sturgeon (Acipenser persicus) and the effect of different doses of 2‐hydroxypropyl‐beta‐cyclodextrin on thawed spermatozoa quality (motility duration and motility percentage) were investigated. For freezing, semen of seven male individuals was pooled in equal volumes and diluted with 4°C [Tris‐HCl (100 mM), pH = 8, DMSO 10%] extenders containing 0, 5, 10, 15 mM of HβCD in a ratio of 1:1(semen/extenders). Then semen was filled into 0.5‐mL straws, and was frozen with vapour of liquid nitrogen at 4‐cm above surface of liquid nitrogen. After 3 min, straws were plunged in to liquid nitrogen. Thawing was performed at 40°C water baths for 15 s. Motility duration of the 10 mM HβCD treated spermatozoa at days 14 (228.98 ± 16.39) and 56 (199.66 ±21.78) were longer than other treatments. In day 56, the motility percentage in treatment with 10 mM was significantly higher (16.14 ± 2.54) (P < 0.05) compared with 5 mM treatment (8.75 ± 2.47) (P < 0.05). Therefore, it is recommended that 10 mM of HβCD can be used as an additive cryoprotectant for increasing cryopreserved spermatozoa quality in this species. 相似文献

The aim of this work was to assess the potential use of different dried macroalgae as food in the rearing of Paracentrotus lividus. Growth, consumption and food conversions were compared in adult sea urchins fed with fresh or dried thalli of four macroalgae species. Six experimental diets were tested: (a) fresh Palmaria palmata; (b) fresh Saccharina latissima; (c) dry P. palmata; (d) dry S. latissima; (e) dry Laminaria digitata and (f) dry Grateloupia turuturu. Linear growth rates were similar for all treatments. Specific growth rate was higher in sea urchins fed with fresh P. palmata, but no difference was found between animals fed with fresh S. latissima and those fed with dried diets. Regarding daily food consumption (DFC), sea urchins consumed the same amount of dried macroalgae as fresh but exhibited a higher food conversion efficiency (FCE) when fed with fresh P. palmata. However, this FCE was only significantly higher when compared to sea urchins fed with dry L. digitata. Dried G. turuturu is not a suitable diet due to its rapid degradation after rehydration. The results suggest that P. lividus adults can be reared on dried macroalgae thalli without detriment to their somatic growth, especially over short periods. The low cost of feeding sea urchins with this diet could help small shellfish farmers to diversify their production into echinoculture. 相似文献

Experiments were carried out to develop an optimal cryopreservation protocol for tench sperm by testing the fertilizing capacity and motility parameters including progressive motility, curvilinear velocity (VCL) and linearity (LIN) of cryopreserved sperm. Three experiments were designed to this aim: first experiment where we tested the effects of two extenders (sugar‐based Grayling and ion‐based Kurokura 180) and two cryoprotectants (DMSO and methanol) on fertilization and hatching success; second where we tested the effect of cryoprotectant type (methanol or DMSO) in different concentrations (5%, 10% and 15%) on fertilization and hatching success; and third where we tested the effect of two cryoprotectants (methanol and DMSO) on sperm motility parameters (progressive motility, VCL and LIN) after 4 h post‐thaw storage (4°C). Sperm prepared with the sugar‐based Grayling extender displayed better fertilization and hatching rates independently of the applied cryoprotectant most likely due to glucose present which acted as an external cryoprotectant. Concerning cryoprotectant concentrations, the use of 10% methanol yielded the highest fertilization (85 ± 15%) and hatching (80 ± 13%) rates, which were significantly higher than in all other groups. During the post‐thaw storage time, 5% methanol, 10% methanol and 5% DMSO groups had significantly higher motility parameters than other groups and we observed no significant decline in any of the parameters during the storage time. Overall, we found that a sugar‐based extender in combination with methanol as cryoprotectant is suitable for the cryopreservation of tench sperm and allows storage of cryopreserved sperm for up to 4 h post thaw. 相似文献

In experiment 1, juvenile sea urchins (n = 80, 0.088 ± 0.001 g wet weight and 5.72 ± 0.04 mm diameter) were held individually and fed ad libitum one of three semi‐purified formulated diets (n = 16 individuals treatment?1). In the diets, protein was held constant (310 g kg?1 dry, as fed) and carbohydrate level varied (190, 260, or 380 g kg?1 dry, as fed). Wet weights were measured every 2 weeks. Total wet weight gain was inversely proportional to dietary carbohydrate level and energy content of the respective diet. In experiment 2, sea urchins (5.60 ± 0.48 g wet weight, n = 40) fed 190 g kg?1 carbohydrate consumed significantly more dry feed than those fed 260 g kg?1, but not more than those fed 380 g kg?1 carbohydrate. Based on differential feed intake rates, sea urchins that consumed more feed also consumed higher levels of protein and had the highest weight gain. Consequently, protein content and/or protein: energy ratio may be important in determining feed utilization and growth among sea urchins in this study. The average digestible energy intake was approximately 70 kcal kg?1 body weight day?1, suggesting daily caloric intake of juvenile Lytechinus variegatus is lower than in shrimp and fish. 相似文献
