Potato common scab is a widespread disease in which scab-like lesions develop on tubers. The disease is caused by pathogenic Streptomyces species, which synthesize the phytotoxin thaxtomin. The txtAB operon, responsible for thaxtomin production, can be used as a marker to identify pathogenic strains of the bacterium. Screening methods to assess scab susceptibility in breeding programs are time-consuming and can produce variable results. Management practices to control the disease vary and include crop rotation, tolerant varieties, monitoring soil pH, avoiding low soil moisture at tuber initiation, and application of soil- and/or seed-applied pesticides. There is a wide range in levels of tolerance among potato varieties. Many public research programs are committed to breeding for scab-tolerant varieties and evaluating management methods. Topics reviewed target readers focused on breeding and disease management objectives to reduce the incidence and severity of potato common scab.  相似文献

The cause of deep-pitted scab of potatoes   总被引:2,自引:0,他引:2
By use of a tyrosine-casinate-nitrate medium,Streptomyces atroolivaceous, S. cinerochromogenes, S. corchorusii, S. diastatochromogenes, S. lydicus, S. malachiticus, and three unidentifiedStreptomyces spp. were isolated from deep-pitted scab lesions on potato cultivars Green Mountain, Red Pontiac, Russet Burbank, and Saco. Over 90% of the deep-pitted lesions contained one or more of theseStreptomyces spp., butS. scabies the cause of shallow scab was not isolated. Isolates ofStreptomyces spp. were also consistently isolated from aseptically excised non-necrotic tissue near the edge of lesions. TheseStreptomyces spp. appeared to be the primary colonizers of this tissue. Alternaria spp.,Fusarium spp.,Penicillium spp.,Rhizopus spp.,Trichoderma spp., and several unidentified non-sporulating fungi as well asBacillus spp.,Pseudomonas spp., andMicrococcus spp. were isolated from deep-pitted scab lesions. Fungal and bacterial isolates varied from one scab lesion to another and except for anAlternaria sp., were not pathogenic. Most of theStreptomyces spp. isolates used to infest heat pasteurized field soil caused deep and/or shallow scab on tubers of the potato cultivar Kennebec in greenhouse and outdoor plantings. Re-isolations from resulting scab lesions yieldedStreptomyces spp. similar in morphology to those used as inoculum. We concluded that a number ofStreptomyces spp. probably includingS. scabies (ATCC 3352 = S.griseus) may cause deep and shallow scab.  相似文献

The effect of chloropicrin fumigation on the soil populations of Spongospora subterranea and the development of powdery scab, formation of root galls and tuber yield was investigated in seven field trials conducted in Minnesota and North Dakota. Sixteen potato cultivars, with different levels of susceptibility to disease on roots and tubers, were planted in plots treated with chloropicrin at rates ranging from zero to 201.8 kg a.i. ha?1. The amount of S. subterranea DNA in soil was determined using qPCR. Bioassays were conducted to further assess the effect of chloropicrin fumigation on root colonization by S. subterranea in two potato cultivars with contrasting disease susceptibility. In the field, chloropicrin applied at rates between 70.1 to 201.8 kg a.i. ha?1 significantly decreased S. subterranea initial inoculum in soil but increased the amount of disease observed on roots and tubers of susceptible cultivars. The effect of increasing disease was confirmed in controlled conditions experiments. Although the amount of S. subterranea DNA in roots of bioassay plants increased with increasing chloropicrin rates, it remained similar among potato cultivars. Chloropicrin fumigation significantly increased tuber yield which in cultivars such as Shepody and Umatilla Russet were associated with the amount root galls (r = 0.30; P < 0.03). Results of these studies contradict earlier reports on the use of chloropicrin fumigation for the control of powdery scab. Factors other than inoculum level, such as environmental conditions that affect inoculum efficiency and host susceptibility, may be significant contributors to the development of powdery scab and root gall formation.  相似文献

The interaction ofStreptomyces scabies, causal agent of common scab, and its potato host was examined under scab-conducive conditions in the greenhouse. Three potato cultivars with different levels of resistance to common scab were inoculated with four levels of two rifampin-resistant strains ofS. scabies. In general, scab severity increased linearly with an increase in log10 of the inoculum, but both cultivar resistance and strain ofS. scabies affected the slope of the regression line. Rhizosphere and rhizoplane populations ofS. scabies also increased with increasing soil population. One strain ofS. scabies consistently had higher rhizosphere and rhizoplane populations than the other strain. However, rhizosphere population was not consistently related to scab severity, scab incidence, cultivar resistance, or strain virulence.  相似文献

Common scab caused by the soil-borne bacterium Streptomyces scabies is a serious disease for the potato industry. We have identified a strong source of resistance in the diploid wild relative Solanum chacoense. This resistance has been introgressed into tetraploid cultivated potato via unilateral sexual polyploidization. This paper describes three hybrid clones (M8, M16, M17) for use by potato breeding programs to enhance resistance to common scab. They were created by crossing a diploid clone (50% S. tuberosum, 50% S. chacoense) to tetraploid cultivars, producing hybrids with 75% cultivated and 25% wild germplasm. The clones are male and female fertile, and are adapted to temperate zone production environments.  相似文献

A total of 23 isolates obtained from scab infected potato tubers representative of six sampling areas in Eastern and Central Canada and five ATCCStreptomyces strains were screened for pathogenicity on the basis of their ability to initiate scab development on aseptically cultured minitubers and plant generated tubers. The results were then correlated with any associated generation of the scab inducing phytotoxin, Thaxtomin A. In all cases a positive correlation was demonstrated between the pathogenicity of variousStreptomyces scabies isolates and their ability to produce the phytotoxin.  相似文献

Candidatus Liberibacter solanacearum’ (CLso) haplotype C, a bacterial pathogen transmitted by the carrot psyllid Trioza apicalis, causes yield losses in carrot production. Due to concerns that this pathogen might also threaten potato (Solanum tuberosum) production, the occurrence of CLso in cultivated and volunteer potatoes in Tavastia Proper and Satakunta regions of Finland was studied. Volunteer potato plants were found in 13 of the 27 inspected carrot fields. Of the 148 potato samples tested by PCR, eight volunteer potato plants and one cultivated potato grown at the edge of a carrot field were found to be CLso positive. The PCR products obtained from these potatoes with primers OA2/OI2c, LpFrag4-1611F/LpFrag4-480R and CL514F/CL514R all showed 100% sequence identity to CLso haplotype C. This is the first observation of CLso haplotype C in field-grown potatoes. In addition, transmission experiments were performed. Attempts to transmit CLso into potato with carrot psyllids were not successful; however, CLso haplotype C was transmitted from infected carrots to potato plants by leaf grafting and by phloem connection formed by dodder, a parasitic plant, and found to survive in the potato plants for several weeks after transmission. However, the bacterial colonisation progressed slowly in the potato phloem and the amount of bacteria detected was low. The plants produced from the daughter tubers of the CLso-positive potato plants were all CLso negative, suggesting that CLso haplotype C was not able to pass to the daughter plants. None of the CLso-positive potatoes inoculated in greenhouse or collected from fields showed symptoms characteristic of zebra chip disease, associated with CLso haplotypes A and B.  相似文献

Summary Based on visual inspection, discrimination between common scab (Streptomyces spp.) and powdery scab (Spongospora subterranea) can be difficult. Inspections are performed on unwashed samples, incidentally supported by microscopic examination. During 1994–1996 surveys were performed in The Netherlands on tubers with symptoms resembling common scab. Under microscopic assessment nearly all samples showed the presence of structures resembling cystosori (sporeballs) ofS. subterranea. At that time confirmation using alternative techniques was not possible. In 2003 research was undertaken to clarify the situation with respect to scab on potato tubers in The Netherlands. One hundred and eighteen scab samples were extensively tested forS. subterranea. All samples were digitally photographed, microscopically examinated and tested with real-time PCR and DAS-ELISA. Use of these modern methods resulted in a clear picture of symptoms that can be attributed toS. subterranea. A lot of scab samples with structures resembling cystosori could not be confirmed as contaminated withS. subterranea.  相似文献

The importance of the potato tuber disease powdery scab, caused by the zoosporic pathogen Spongospora subterranea f. sp. subterranea, has increased worldwide, and the disease is one of the most important problems facing potato production in some regions. This soilborne pathogen produces many resting spores which can remain dormant for long periods, are highly resistant to environmental stresses and can spread the disease on seed potatoes and in contaminated soil. The enigmatic nature of this organism exacerbates the development of effective powdery scab control methods. Substantial knowledge has been gained in the last decade on the biology of the pathogen and the epidemiology of the disease, but no single effective control measure is, or is likely to be, available. An integrated approach to powdery scab management is the aim, with host resistance as a substantial and sustainable component. Further research on the epidemiology of powdery scab and population genetics of the pathogen is urgently required. All stakeholders involved in the potato industry must become aware that solution of the powdery scab problem is likely to be a long-term goal. When resistant cultivars with all the other characteristics demanded for marketing high-quality potatoes are available, and when disease risk from the pathogen on seed and/or in soil can be accurately determined, then the mission to develop effective powdery scab control will become achievable.  相似文献

Summary The effect of green manuring and soil amendments in various combinations on common scab of potato have been investigated. Soybean green manuring in combination with partially decomposed wheat straw was most effective with a decrease of 56.9% and 54.8% respectively in scab index and scab incidence. There was also a 14.2% increase in potato yield. Soil microbial population was also studied and a considerable increase in bacterial population was observed with soybean green manuring. Maximum population was recorded at planting and at harvest, microbial population was drastically decreased.  相似文献

Krantz is an oblong to slightly blocky, russet-skinned potato cultivar with medium-late maturity and a vigorous vine. It is resistant to hollow heart, has high resistance to common scab and possesses high field resistance to late blight. It is moderately resistant toVerticillium wilt, susceptible to early blight, viruses S and X, and exhibits symptoms of bacterial ring rot. Krantz is adapted to the irrigated sands, but will growth crack in heavier soils.  相似文献

In 2000 and 2004, 19 potato varieties were grown in separate plots inoculated with the scab pathogensStreptomyces scabiei andS. acidiscabies. Reaction of the varieties to these two species were highly correlated in both years, with no host variety-pathogen species interaction. These results are consistent with the central role of the bacterial toxin thaxtomin in scab development, and indicate that this and other mechanisms involved in pathogen infection and establishment are not expressed differentially with regard to variety. Likewise, there is no apparent differential response of the pathogens to those host factors determining the degrees of resistance and symptoms expressed in different varieties.  相似文献

Summary The resistance to common scab of nine cultivars and eight advanced breeding lines of potatoes, fiveSolanum andigenum Juz. et Buk. clones, and three potato breeding lines selected for resistance to common scab, was investigated in a field and a pot trial. In the pot trial potaroes were planted in inoculated coarse river sand in plastic pots (150 mm in diameter) placed on irrigated subsoil in a glassfibre house. In a field trial potatoes were planted in an artificially infested field. The harvested tubers were scored individually for scab incidence according to two scales, viz. a 1–5 point scale for percentage surface covered with scab and a 1–3 point scale for lesion type. A scab index value was calculated for each tuber. A significant correlation coefficient of 0.82 was obtained for the results of the two tests.  相似文献

Summary DiploidSolanum phureja, diploid and tetraploidS. tuberosum × S. phureja hybrid, and tetraploidS. tuberosum backcross clones were assessed for resistance to commercially important diseases. There was a reduction in the expression ofS. phureja characteristics with increasing dosage ofS. tuberosum genetic material. The generally high resistance of theS. phureja group to common scab, potato leafroll virus and potato virus Y decreased, while resistance to gangrene, foliage and tuber blight tended to increase. Resistance genes were probably well dispersed throughout theS. phureja genotype so that many were lost on hybridisation, butS. phureja could be a useful source of scab and virus resistance in the production ofS. tuberosum cultivars.  相似文献

Resistance to common scab continues to be a high priority trait for potato breeders. We have identified a source of resistance in the diploid wild potato relative Solanum chacoense and have introgressed it into cultivated potato by crossing it to a dihaploid. A clone generated by crossing two full-sib hybrids is highly resistant and produces both 2n pollen and 2n eggs. This clone, named 4–48, is homozygous for a major QTL for scab resistance derived from S. chacoense. Clone 4–48 was reciprocally crossed to three round white cultivars Megachip, Pike, and White Pearl. Common scab resistance was transferred to tetraploid offspring, with transmission through the male parent generally more effective than through the female parent. The majority of progeny (70%) exhibited stability for resistance to common scab across environments. This germplasm will be useful for breeding new resistant cultivars.  相似文献

An investigation was made to determine possible effects of gypsum and sulfur for control of common scab of potato [Streptomyces scabies (Thaxt.) Waksman and Henrici] in a highly buffered calcareous soil. This investigation included a comparison of the effectiveness of gypsum and sulfur with Terraclor and Terraclor Super-X. Both gypsum and sulfur reduced potato scab and measurements indicate that change of soil pH was slight (0.1–0.4 pH reduction). Band applications of sulfur in the sulfate (gypsum) or elemental form were effective at 600 lbs/A (672.5 Kg/ha) but not at lower rates. The weight of tubers thrown out of grade with scab was reduced by 53% with the effective sulfur rate. Treatment effects of gypsum (600 lb S/A-672.5 Kg/ha) were not significantly different from sulfur dust (600 lb/A-672.5 Kg/ha), Terraclor or Terraclor Super-X (both at 25.0 lb PCNB/A-28.0 Kg/ha). Tissue analyses of tuber peelings showed a significant reduction in calcium from treatments involving sulfur and gypsum, indicating that calcium levels in tuber peelings were positively correlated with scab susceptibility.  相似文献

In Search of Better Management of Potato Common Scab   总被引:1,自引:0,他引:1
Common scab (CS) is an important disease and quality problem in potato crops worldwide. CS degrades the appearance of the potato tubers, thereby diminishing market value. Knowledge of CS has expanded considerably over recent years, enabling improved detection of the causal pathogens and increased understanding of mechanisms of pathogenicity, and providing potential methods of modulating pathogen response for disease resistance. However, effective control of this disease remains elusive, and will require increased understanding of both the host and the pathogen. Traditional control strategies such as irrigation and reduced soil pH are not sufficient and often fail. Optimizing environmental conditions for reduction of CS can also lead to favorable conditions for other diseases. The most desirable control method would be disease-resistant potato cultivars. However, no currently available commercial potato cultivar has been shown to be completely resistant to CS. In this review, we provide an overview of potato CS caused by plant pathogenic Streptomyces species, recent research on mechanisms and management of the disease, and knowledge gaps that limit successful control of this ubiquitous and troublesome disease.  相似文献

Haploids were extracted from the tetraploid potato cultivars, Avon and Hindenburg, and subjected to tests of their resistance to the common scab disease. The distribution of Hindenburg haploids according to scab reaction was significantly different from that of Avon haploids. The former distribution was skewed strongly toward high levels of resistance in contrast with the large susceptible class among Avon haploids.  相似文献
